Drosophila suzukii is an invasive pest causing severe damages to a large panel of cultivated crops.To facili tate its biocontrol with stratcgies such as sterile or incompatible insect techniques,D.suzukid must be mass-produced and then stored and transported under low temperature.Prolonged cold exposure induces chill injuries that can be mitigated if the cold period is interrupted with short warming intervals,referred to as fluctuating thermal regimes(FTR).In this study,we tested how to optimally use FTR to extend the shelf life of D.suzukii under cold storage.Several FTR parameters were asessed:temperature(15,20,25℃),duration(0.5,1,2,3 h),and frequency(every 12,24,36,48 h)of warming intervals,in two wild-type lines and in two developmental stages(pupac and adults).Generally,FTR improved cold storage tolerance with respect to constant low temperatures(CLT).Cold mortality was lower when recovery temperature was 20℃ or higher,when duration was 2 h per day or longer,and when warming interruptions occurred frequently(every 12 or 24 h).Applying an optimized FTR protocol to adults greatly reduced cold mortality over long-term storage(up to 130 d).Consequences of FTR on fitness-related traits were also investigated.For adults,poststorage survival was unaffected by FTR,as was the case for female fecundity and male mating capacity.On the other hand,when cold storage occurred at pupal stage,postorage survival and male mating capacity were altered under CLT,but not under FTR.After storage of pupae,female fecundity was lower under FTR compared to CLT,suggesting an energy trade-off between repair of chill damages and C22 production.This study provides detailed information on the application and optimization of an FTR-based protocol for cold storage of D.suzuki that could be useful for the biocontrol of this pest.  相似文献

Cryptosporidium parvum is an important zoonotic parasite that causes significant economic loss in the animal husbandry industry,especially the cattle industry.As there is no specific vaccine or drug against Cryptosporidium,a rapid and accurate method for the detection of C.parvum is of great significance.In this study,colloidal gold strips were developed based on Cryptosporidium parvum virus 1 (CSpV1) for the detection of C.parvum infection in cattle fecal samples.The colloidal gold solution was prepared by reducing trisodium citrate and the CSpV1 #5 monoclonal antibody was labeled with colloidal gold.A polyclonal antibody against the CSpV1 capsid protein and an anti-mouse IgG antibody were coated on the colloidal gold strips for use in the test and control lines,respectively.Our results showed that the detection sensitivity in fecal samples was up to a 1:64 dilution.There was no cross-reaction with Cryptosporidium andersoni or Giardia in the fecal samples.The different preservation conditions (room temperature,4℃,and 37℃) and preservation time (7,30,60,and 90 days) were analyzed.The data showed that the strips could be preserved for 90 days at 4℃ and for 60 days at room temperature or 37℃.The colloidal gold strips were used to detect the samples of 120 clinical fecal in Changchun,China.The results indicated that the rate of a positive test was 5%(6/120).This study provides a rapid and accurate method for detecting C.parvum infection in cattle and humans.  相似文献

The effect of temperature on the rate of development of Xyleborus fornicatus (Eichh.) was determined by rearing individuals under a range of constant temperatures (15 - 32℃). Rates of development changed in a linear fashion over a wide range of temperatures. Estimates of lower development thresholds were obtained for eggs (15.7±0.5℃), larvae (15.8±0.8℃) and pupae (14.3±1.4℃) and the degree days (DD) for development were 70±4.4, 95±8.5 and 72±5.1 DD, respectively. Optimum temperature for development was around 30~C for all stages. Temperature fluctuation in cooler High Country areas (above 1400 m) with a mean temperature around 15℃ seems to be critical for the development of the pest, which may be responsible for the near absence of pest in those areas. Temperature fluctuations (18- 30℃) in the Mid Country region (600- 1200 m) favor the development of the pest compared to development under constant conditions. The altitudinal distribution of the shot-hole borer across tea growing areas in Sri Lanka is, therefore, mainly governed by temperature.  相似文献

In order to understand the composition and quantitative variation of ecdysteriods in the adults and eggs of Opogona sacchari (Bojer), an invasive alien pest, we analyzed the ecdysteroid composition and titers in the adult and egg of this pest. On day 4 after eclosion, the titer of ecdysteriods in the male adult was 0.080 ng/adult, much lower than 5.978 ng/adult in the female adult. During the development of ovaries, the titer of ecdysteriods was low on the first two days, and high in the late period, with the peak (10.48 ng/ovary) appearing on day 3. During the development of eggs, the titer of ecdysteriods was about 0.010 ng/egg from day 1 to day 3, and then decreased to 0.006 ng/egg on day 4. In both adults and eggs, three main components of ecdysteriods were found by identification of HPLC/RIA. They were 20-hydroxyecdysone, 26-hydroxyecdysone, and an unidentified component.  相似文献

Temperature-dependent sex determination(TSD) is a type of environmental sex determination in which the sex of the embryos depends on the ambient temperature; however,the molecular mechanisms governing this process remain unknown.Aromatase,encoded by the cyp19a1 gene,which converts androgens into estrogens in animals,was considered to be the key gene for TSD.In this study,the 5'-flanking region of the cyp19a1 gene in Reeves' turtle(Mauremys reevesii) was cloned,and the promoter region was identified using the luciferase reporter assay.Then the eggs of Reeves' turtle were incubated at different temperatures(26°C: male-biased temperature; 29°C: non-sex-biased temperature and 32°C: female-biased temperature).During the thermosensitive period,the adrenal kidney gonad complexes(AKG) were sampled.DNA methylation analysis of the AKG samples showed that the promoter region of the cyp19a1 gene was significantly de-methylated in the female-biased temperature regime(P0.01).Quantitative analysis of the cyp19a1 gene and estrogen by q PCR and Elisa assay showed that the expression level of the cyp19a1 gene and estrogen content were both upregulated significantly at the female-biased temperature(P0.01).These results indicated that the de-methylation response of the cyp19a1 gene to incubation temperature,especially at the female-biased temperature,could lead to temperature-specific expression of aromatase and increased estrogen levels,which may further determine gonadal development in Reeves' turtle.These findings provide insights into the genetic mechanisms underlying TSD.  相似文献

The functional response of adult Nabis kinbergii (Hemiptera: Nabidae) to density of diamondback moth Plutella xylostella (Lepidoptera: Plutellidae) was investigated under laboratory conditions. Holling' s (1959) type Ⅱ model was found to be a good fit for the observed functional response of this predator. The numbers of P. xylostella consumed increased with temperature from 15℃ to 35℃. The maximum number of prey killed was observed at 35℃, with average of 10.3 and 8.3 forth instar larvae consumed by adult females and males of N. kinbergii, respectively. The predation of N. kinbergii on P. xylostella increased with successive immature stages. The number of prey consumed by predators decreased as the body size of prey increased. An average of 131 eggs or 95 larvae of P. xylostella were killed by a single of female adult in 24 hours at 24"C. The pupae of P. xylostella were observed to be eaten by fifth instar nymphs and adults N. kinbergiiin numbers of less than an average of 0.7 pupae per predator in 24 hours at 24"C. Predation preference by N. kinbergii was also investigated. The number of P. xylostella and Myzus persicae killed by female N. kinbergii was not significantly different, but males killed significantly more P. xylostella than M. persicae. Both eggs and larvae of P. xylosteUa were killed in significantly greater number than those of Pieris rapae in the same feeding arena.  相似文献

When investigating insecticide resistance of pest insects, for example, the pollen beetle Meligethes aeneus, it is relevant to differentiate toxicological and molecular genetic data between male and female specimens. A molecular sex determination method would allow resistance testing to be run without prior sorting of the samples. A one-step quantitative RT-PCR method for quantification of the yolk protein vitellogenin expression in the pollen beetle was established. The expression level of vitellogenin relative to tubulin was determined. Pollen beetles were tested at different time points during their development to determine if vitellogenin is a reliable molecular marker for detection of sexually mature females. The differentiation between females and males by relative expression of vitellogenin to tubulin is conditional regarding the life cycle. Sexually mature females and males could easily be distinguished, whereas immature specimens could not be seperated. Vitellogenin expression is a successful marker for identification of sexually mature pollen beetles. Females from the spring populations showed vitellogenin expression when the temperature was above 10.2°C. Further, detailed observations of vitellogenin throughout the spring indicated a strong relationship between daily temperatures and vitellogenin expression, which is an indicator of oviposition ability.  相似文献

An outbreak of the sweetpotato whitefly, Bemisia tabaci (Gennadius), biotype B occurred in the Imperial Valley, California in 1991. The insects destroyed melon crops and seriously damaged other vegetables, ornamentals and row crops. As a result of the need for sampling technology, we developed a whitefly trap (named the CC trap) that could be left in the field for extended time periods. We used the traps to monitor populations ofB. tabaci adults during year-round samplings from 1996 to 2002 to study variations in the weekly trap catches of the insect. The greatest number ofB. tabaci adults was recorded in 1996, followed by a continuing annual decrease in trap catches each year through 2002. The overall decline of B. tabaci is attributed in part to the adoption of an integrated pest management (IPM) program initiated in 1992 and reduced melon hectares from 1996 to 2002. Other factors may also have contributed to the population reductions. Seasonally, B. tabaci trap catches decreased during the late summer and fall concurrent with decreasing minimum tempera- tares that are suggested to be a significant factor affecting seasonal activity and reproduction.  相似文献

Nucleopolyhedrovirus (NPV) has become an integral part of integrated pest management (IPM) in many Australian agricultural and horticultural crops. This is the culmination of years of work conducted by researchers at the Queensland Department of Primary Industries and Fisheries (QDPI&F) and Ag Biotech Australia Pty Ltd. In the early 1970’s researchers at QDPI&F identified and isolated a virus in Helicoverpa armigera populations in the field. This NPV was extensively studied and shown to be highly specific to Helicoverpa and Heliothis species. Further work showed that when used appropriately the virus could be used effectively to manage these insects in crops such as sorghum, cotton, chickpea and sweet corn. A similar virus was first commercially produced in the USA in the 1970’s. This product, Elcar?, was introduced into Australia in the late 1970’s by Shell Chemicals with limited success. A major factor contributing to the poor adoption of Elcar was the concurrent enormous success of the synthetic pyrethroids. The importance of integrated pest management was probably also not widely accepted at that time. Gradual development of insect resistance to synthetic pyrethroids and other synthetic insecticides in Australia and the increased awareness of the importance of IPM meant that researchers once again turned their attentions to environmentally friendly pest management tools such NPV and beneficial insects. In the 1990’s a company called Rhone-Poulenc registered an NPV for use in Australian sorghum, chickpea and cotton. This product, Gemstar?, was imported from the USA. In 2000 Ag Biotech Australia established an in-vivo production facility in Australia to produce commercial volumes of a product similar to the imported product. This product was branded, ViVUS?, and was first registered and sold commercially in Australia in 2003. The initial production of ViVUS used a virus identical to the American product but replicating it in an Australian Helicoverpa species, H. armigera. Subsequent research collaboration between QDPI&F and Ag Biotech reinvigorated interest in the local virus strain. This was purified and the production system adapted to produce it on a commercial scale. This new version of ViVUS, which was branded ViVUS Gold?, was first registered and sold commercially in 2004. Widespread insect resistance to insecticides and a greater understanding of integrated pest management is leading to increased adoption of technologies such NPV in Australian agriculture.  相似文献

探讨了人工扩繁异色瓢虫时卵和成虫的最适冷藏条件。卵的最适保存温度是 1 0℃ ,冷藏 1 5d内孵化率平均在 60 %以上。成虫的最适保存温度是 1 0℃ ,如将刚羽化的成虫直接在该温度下冷藏 ,冷藏 40d时存活率能保持在 5 0 %左右 ;如将刚羽化成虫先在 1 5℃、0L∶2 4D条件下处理 1 6d,然后置于 1 0℃下冷藏 ,经冷藏 70d后存活率接近 1 0 0 % ,冷藏 90d后存活率仍高于 70 %。表明经一定条件下预处理后再冷藏成虫能保持较高的存活率和较长的存活期 ,可满足人工扩繁时对成虫的冷藏要求。  相似文献

大猿叶虫Colaphellus bowringiBaly是我国十字花科蔬菜上的一种重要害虫,以成虫在土壤中越冬和越夏,滞育发生主要受温度和光周期调节。本文就光周期和温度对滞育后成虫生物学特性的影响进行了研究。结果表明,在25℃下,光周期(L14∶D10和L12∶D12)对成虫滞育后的存活率、寿命、日平均产卵量、总产卵量均无显著影响。在长光照(L14∶D10)下,温度18℃、22℃和25℃)对滞育后成虫的生物学特性具有明显的影响;随温度的降低,总产卵量呈下降趋势,平均产卵量显著降低,雌虫滞育后寿命显著延长,18℃下少数个体能被诱导再次滞育。  相似文献

Laboratory studies were conducted on certain aspects of biology ofDiadegma semiclausum Hellén, a larval parasite of a crucifer pest,Plutella xylostella (L.). Within the range of 15°C to 35°C, the higher temperature, the shorter was the duration of larval and adult stages. Egg hatching and adult emergence were high at 15°C to 30°C but were significantly reduced at 35°C. The higher the temperature, the higher was the proportion of males produced. Temperature threshold was 5.74°C for eggs, 3.80°C for larvae, 5.91°C for pupae and 6.60°C for adults.D. semiclausum oviposition in the first threeP. xylostella larval instars produced more parasite males than females but oviposition in the fourth instar produced significantly more females than males. Parasite adults tended to emerge from their pupae from 06∶00 to 09∶00 hours although some emerged at other hours during the photophase. Adult longevity and production of eggs increased when adults were provided with a food source (honey) compared with no food or provision of water alone. Parasite adults survived and laid eggs for 28 days when provided with food but for only three days when deprived of food.  相似文献

莲草直胸跳甲是恶性入侵杂草喜旱莲子草的有效天敌。为探明莲草直胸跳甲成虫繁育的适宜温度条件和卵的低温冷藏适期,本文研究了莲草直胸跳甲成虫在25、27、30、32、35℃下的存活、取食和产卵特性及卵在15℃下冷藏不同时期的孵化率。结果表明:在25~35℃,莲草直胸跳甲成虫的生存曲线差异显著;随着温度的升高,雌雄成虫寿命呈缩短趋势,表现为25℃>32℃>35℃;但27和30℃的生存曲线与25和32℃差异均不显著。温度对莲草直胸跳甲成虫的取食量和产卵量均有显著影响,在25~35℃,随着温度的升高,跳甲的取食量和产卵量呈下降趋势;成虫期取食量、日均产卵量和世代产卵量表现为25℃>27℃、30℃>32℃>35℃;成虫期取食量和世代产卵量呈显著正相关,相关系数为0.960。将莲草直胸跳甲卵在15℃下冷藏1~10d后转移至25℃,其孵化率均可达100%,但孵化进度则有差异,孵化高峰期随着冷藏时间的延长而提前。与未经冷藏的卵(对照)相比,在15℃下冷藏1d,莲草直胸跳甲卵的始孵化日提前1d,但高峰期不变,均在第5天;在15℃下冷藏4、7和10d后,其孵化高峰期分别较对照提前1、2和3d。可见,在25~35℃时,25℃是莲草直胸...  相似文献

【目的】桃小食心虫Carposina sasakii是我国北方落叶果树的重要害虫。本研究旨在探索短时高温对桃小食心虫生长发育和繁殖的影响。【方法】在室内23±1℃、 相对湿度80%±7%和15L∶9D条件下, 测定了桃小食心虫卵、 幼虫、 蛹、 成虫在经历35, 38和41℃高温处理1~4 h后各阶段的发育历期、 存活率和产卵量。【结果】 短时高温对卵的孵化率无明显影响; 经41℃处理后, 初蛀果幼虫(1日龄)的发育历期明显延长, 且存活率显著降低, 3日龄以上的幼虫受到的影响不明显; 11日龄蛹的羽化率在38℃和41℃处理中明显降低, 畸形率也显著升高; 经38℃和41℃处理的成虫存活率降低, 寿命缩短, 产卵量也减少。【结论】短时高温处理对桃小食心虫卵的影响较小, 而对成虫的影响较大。这些结果有助于深入了解该虫在高温季节种群数量变动机制。  相似文献

研究微小花蝽Orius minutus(L.)对温室白粉虱Trialeurodes vaporariorum(Westwood)的捕食作用。结果表明,微小花蝽成虫对温室白粉虱各虫态的功能反应呈HollingⅡ型。微小花蝽成虫对温室白粉虱卵、1龄和2龄混合若虫及其3龄若虫的理论最大捕食量分别为123,74和52头/d。微小花蝽成虫对温室白粉虱卵的捕食效应随捕食者个体间干扰作用的增加而下降,符合Hassel-Varley方程,捕食作用率(E)随着微小花蝽数(P)增加而呈幂指数下降,模拟模型E=0.1021P-0.3189,干扰系数为0.3189。在15~40℃的温度范围内,随着温度的升高微小花蝽成虫对温室白粉虱卵的寻找效率提高,最高达1.1990,处置时间缩短,最低达到0.0035d。  相似文献

为了全面评价冷贮对米蛾Corcyra cephalonica Stainton卵繁育螟黄赤眼蜂Trichogramma chilonis Ishii的影响, 本试验采用米蛾卵为寄主, 以冷贮温度(3和10℃)、 冷贮虫龄(以接蜂开始时间为起点, 接蜂后1, 2, 3, 4, 5, 6, 7, 8和9日龄)和冷贮时间(1, 2, 3和4周)为试验因子, 研究了冷贮对螟黄赤眼蜂活蛹率、 羽化率和畸形率的影响。结果表明: 3个因素均能单独或互作显著影响活蛹率、 羽化率和畸形率(P<0.05)。综合各冷贮虫龄对低温的反应, 螟黄赤眼蜂对3℃较敏感, 此温度下1-3日龄的活蛹率和各虫龄的羽化率均显著下降(P<0.05), 不适宜长期冷贮; 10℃下各虫龄的活蛹率受冷贮时间的影响不显著, 1-3日龄的羽化率受冷贮时间的影响也不显著, 冷贮4周后仍有74%以上的羽化率, 10℃适宜冷贮。不同冷贮温度下冷贮适宜虫龄不同。在10℃下, 最适冷贮虫龄是3日龄的赤眼蜂, 此虫龄不同冷贮时间的活蛹率(74.09%~77.59%)和羽化率(74.26%~90.37%)均与对照无显著差异(P>0.05), 但显著高于其他虫龄(P<0.05)。此虫龄在10℃下冷贮1~3周, 畸形率在27%以内。结果表明米蛾卵内螟黄赤眼蜂不同冷贮温度下适宜冷贮虫龄不同。  相似文献

大猿叶虫Colaphellus bowringi Baly是十字花科蔬菜上的重要害虫。在田间,该虫的繁殖期出现在春季和秋季,以成虫在土中越夏和越冬。本试验在光周期L∶D=16∶8条件下,系统调查了不同温度(20、25和28℃)对大猿叶虫非滞育成虫繁殖的影响。结果表明:每雌平均日产卵量在28℃最大(51.24±2.90)粒,显著高于25℃和20℃的(39.57±2.86)粒和(31.53±2.02)粒,但由于28℃下的产卵持续时间明显短,导致了总产卵量在这3种温度间没有显著差异;28℃下的卵孵化率低于20℃和25℃,但没有显著差异;随着温度的升高,成虫寿命逐渐缩短,20℃与28℃间的雌虫寿命存在显著差异,20℃与25℃,28℃的雄虫寿命存在显著差异;在20、25、28℃下,均有30%的个体经历一段时间繁殖后进入滞育,其滞育前的产卵持续时间随着温度的升高而明显缩短,总产卵量也随着温度的升高而下降。  相似文献

在17~29℃、RH80%±7%和光周期L:D=15:9的条件下,测定了桃小食心虫Carposina sasakii Matsumura各虫态的发育历期,计算了桃小食心虫卵、幼虫、蛹和全世代的发育起点温度和有效积温。结果表明,各虫态的发育历期随着温度的升高而缩短,各虫态的发育速率与温度之间具有良好的线性关系。桃小食心虫卵、幼虫、结茧前期、蛹及生殖前期的发育起点温度分别为10.02、9.44、10.58、10.09和9.51℃,有效积温分别为87.3、238.6、10.7、156.9和31.6日·度,全世代的发育起点温度和有效积温分别为9.22℃和543.2日·度。  相似文献

大猿叶虫Colaphellus bowringi是江西山区十字花科蔬菜上的重要食叶害虫,以成虫在土中越夏和越冬。由于成虫滞育期的差异,该虫的化性显示了明显的种内变异。有些个体隔年繁殖;有些个体是一化性的,仅在春季或秋季繁殖1代;有些个体是二化性的,春季和秋季各繁殖1代;有些个体是多化性的,春季1代,秋季2~3代。因此,大猿叶虫在田间一年可发生4代。在春季,滞育成虫于2月底至4月初陆续出土繁殖;在秋季,滞育成虫于8月中旬至10月初陆续出土繁殖。春季羽化的成虫于4月底至6月上旬陆续入土越夏,秋季羽化的成虫于9月中旬至12月底陆续入土越冬。大猿叶虫一生能交配多次,大多数雌虫的产卵期超过1个月,最长达67天。平均每雌产卵量:春季世代为644粒,秋季世代为963粒,最高达1.950粒。各虫态的发育历期:在15~30℃间,卵为13.78~3.14天,幼虫为22.83~6.95天,蛹为12.10~3.18天。发育阈值温度:卵为10.7℃,幼虫为8.8℃,蛹为9.6℃。非滞育成虫的寿命约为1~2个月,滞育成虫的寿命为5~38个月。滞育成虫均入土蛰伏,在土中的蛰伏深度为9~31 cm 。  相似文献
