The fungi isolated from 100 samples of flue-cured tobacco from 12 markets in 2 tobacco belts comprised 11 genera, including 10 species of Aspergillus. The mean percentage per sample isolated from 62 samples of tobacco from Middle Belt markets was Alternaria, 40.6%; Aspergillus niger, 47.8%; Aspergillus repens, 38.0%; and Penicillium, 25.8%. The mean percentage per sample isolated from 38 samples of tobacco from Old Belt markets was Alternaria, 74.0%; Penicillium, 52.5%; Aspergillus repens, 38.0%; and Aspergillus ruber, 36.2%. Damaged (74 samples) and nondamaged (26 samples) stored tobacco yielded species of six genera of fungi, including eight species of Aspergillus. Species of Aspergillus and Penicillium were commonly isolated from both damaged and nondamaged tobacco, whereas species of Alternaria, Cladosporium, Fusarium, and Rhizopus were isoalted more frequently from nondamaged tobacco. The fungi that occurred in the highest population in damaged tobacco were Aspergillus repens, A. niger, A. ruber, and Penicillium species.  相似文献

The management of fungal contaminants inside libraries and archives has become a big challenge for librarians, restorers and scientists. Several disinfection treatments have been developed in recent years, using both chemical and physical approaches on book collections and indoor environments. However, there is a lack of knowledge about the temporal efficiency of these cleanings, especially in relation with the preservation environments. The aim of this study was to determine the long-term effect of a chemical disinfection that interested a previous-contaminated book collection inside a University library. The monitoring after 6 months and 1 year from the cleaning confirmed any fungal growth on the disinfected books and the reduction of 90% of the airborne fungal load, highlighting anyway the presence of high fungal diversity. Sixty-eight different airborne fungal entities were isolated, in particular Aspergillus vitricola, Bulleromyces albus, Cladosporium cladosporioides, Cladosporium pseudocladosporioides, Cladosporium sphaerospermum, Penicillium brevicompactum, Rodothorula mugillaginosa and Sporodiobolus pararoseus. Several fungal species were sampled from the disinfected books, in particular Aspergillus penicillioides and Penicillium chrysogenum. The presence of these fungi both as airborne and as settled particles highlights the importance to maintain clean the preservation environments in order to prevent further microbial contaminations.  相似文献

The aim of this study was to analyze the viable fungi in the air of indoor environments of the National Archive of the Republic of Cuba (NARC). The samples were taken in fifteen areas (five points for the three floors) at 2 days (rainy and dry seasons) using a biocolector SAS in 3 h: 10:00 a.m., 1:00 p.m. and 4:00 p.m. The fungal concentration was indicated as colony forming units per cubic meter of air (cfu/m3 of air). It was concluded that indoor air of the NARC was not polluted due to the average fungal concentration that was obtained (59 and 327 cfu/m3 of air in rainy and dry seasons, respectively). In front of the electric transformers, the fungal concentration was significantly higher than those obtained along the corridors of the lower-ground floor. A trend to the increasing of fungal concentration was observed: when the floors were increased, in dry season was higher than rainy season, where also there was biggest fungal diversity. Among the fungal genera isolated, Aspergillus had the highest relative frequency of appearance, and in dry season, it was isolated Actinobacillus sp. (Actinomycetes). Six fungal species were isolated in both samplings: Aspergillus alliaceus, Aspergillus niger, Cladosporium cladosporioides, Fusarium oxysporum, Penicillium chrysogenum and Mucor racemosus. All of fungal species isolated have high biodeteriogenic capacity, and the 61 % showed pathogenic attributes.  相似文献

Indoor fungi are a major cause of cosmetic and structural damage of buildings worldwide and prolonged exposure of these fungi poses a health risk. Aspergillus, Penicillium and Cladosporium species are the most predominant fungi in indoor environments. Cladosporium species predominate under ambient conditions. A total of 123 Cladosporium isolates originating from indoor air and indoor surfaces of archives, industrial factories, laboratories, and other buildings from four continents were identified by sequencing the internal transcribed spacer (ITS), and a part of the translation elongation factor 1α gene (TEF) and actin gene (ACT). Species from the Cladosporium sphaerospermum species complex were most predominant representing 44.7% of all isolates, while the Cladosporium cladosporioides and Cladosporium herbarum species complexes represented 33.3% and 22.0%, respectively. The contribution of the C. sphaerospermum species complex was 23.1% and 58.2% in the indoor air and isolates from indoor surfaces, respectively. Isolates from this species complex showed growth at lower water activity (≥ 0.82) when compared to species from the C. cladosporioides and C. herbarum species complexes (≥ 0.85). Together, these data indicate that xerotolerance provide the C. sphaerospermum species complex advantage in colonizing indoor surfaces. As a consequence, C. sphaerospermum are proposed to be the most predominant fungus at these locations under ambient conditions. Findings are discussed in relation to the specificity of allergy test, as the current species of Cladosporium used to develop these tests are not the predominant indoor species.  相似文献

Background

Data on mould sensitization in the general population are scarce and mostly on Aspergillus fumigatus, Alternaria alternata and Cladosporium herbarum.

Objectives

To validate a dot-blot assay for the detection of specific IgE and evaluate the prevalence of mould sensitization in a healthy population.

Methods

The dot-blot assay was validated against the CAP test. Sensitization rate to ten common indoor and outdoor mould species in 344 serum samples was calculated. For each serum with more than one reactivity, the “major sensitization” defined as the strongest response against a single mould species was calculated.

Results

Intra- and inter-assay variations were both below 20%, and the positivity threshold of the test was of 0.418 kU/L for A. fumigatus. Correlation with CAP results was strong. The overall prevalence of sensitization was 32.8%, and the commonest sensitizations were against A. alternaria, A. flavus and A. niger (around 15%). The most frequent “major reactivities” were against A. niger and A. alternata (20–30%). In 25.1% of the samples, “major reactivities” were directed against a group of moulds commonly found indoor (Penicillium spp., Aspergillus versicolor, Cladosporium sphaerospermum and Cladosporium cladosporioides).

Conclusions

The dot-blot assay was validated for the detection of mould-specific IgE. In the general population, sensitization to indoor species was common and accounted for 25% of overall mould sensitizations.

Health problems in people who are in indoor environments with poor ventilation have resulted in an increase in the number of studies regarding air quality. Microorganisms and inferior indoor air-climatic conditions not only affect human health but also cause decay of invaluable materials present in libraries. Therefore, this study aimed to assess the culturable bioaerosol composition and concentration in the library of Istanbul University. The culturable fungal flora, a biodeterioration agent, of the damaged archival materials was also examined. The air was sampled for a year, as were the surfaces of 207 biologically damaged books. The fungal colonies’ range was between 235 and 1850 CFU/m3, and the bacterial colonies range between 270 and 1920 CFU/m3. This is the first volumetric record in Turkey. Although the microbial contamination was not very high, molds such as Aspergillus versicolor, Cladosporium sphaerospermum, Penicillium chrysogenum, Alternaria alternata, Chaetomium globosum, Aspergillus flavus, and Aspergillus ochraceus might cause harm to human beings or to books as biodeterioration agents. The highest amount of fungus was determined in Archive 3, which contained damaged books. The fungal species isolated from the air and books were essentially the same. Therefore, it is important to determine not only the numeric values but also the microbiological composition of fungal colonies because the variety of fungal species is indicative of a deterioration process in effect over a long period. This study considers the taxonomic composition of filamentous fungi in the epibiosis on the shell of the Yesso scallop Mizuhopecten yessoensis (Jay, 1856) (Bivalvia), which were collected in open waters of Peter the Great Bay, Sea of Japan. The taxonomic composition was found to include ten species of filamentous fungi, nine of which were identified. The revealed species belong to six genera, viz., Aspergillus, Aphanocladium, Cladosporium, Penicillium, Phialophorophoma, and Eurotium.  相似文献

Summary Air and surface sampling to quantify and identify fungi were conducted over a 10-year period in 130 Belgian homes, including flats and houses. Homes were selected based on the medical files of allergic, mostly asthmatic patients to assess possible mould responsibility in the aetiology of the disease. Air sampling was done with Reuter centrifugal air sampler using different sampling times and incubation temperatures to detect mesophilic and thermotolerant or thermophilic fungi. More than 50 genera were detected, withCladosporium, Penicillium andAspergillus being the most common. The main species detected wereAspergillus versicolor, several species ofPenicillium, Cladosporium sphaerospermum andCladosporium herbarum. Surface sampling was conducted concomitant with air sampling. Surfaces with and without visible mould growth were tested equally by Rodac contact plates or swabs. AlthoughCladosporium herbarum was shown to be a common species in indoor environments by air sampling, it was not detected by surface sampling from visible mould growth. This finding suggests that its presence in dwellings is from an outdoor origin and not indicative of indoor proliferation.Aspergillus versicolor, Cladosporium sphaerospermun, andPenicillium chrysogenum were the most common species isolated from surfaces showing visible mould growth.  相似文献

Seven non-basidiomycetic fungi, Aspergillus, Candida, Cladosporium, Fusarium, Monicillium, Trichoderma and Penicillium, and two basidiomycetic fungi, Pleurotus and Phanerochaete were isolated from a creosote-contaminated soil by using mineral salts medium and soil extract broth containing antibiotics. Soil contaminated with phenol, o-cresol, m-cresol and p-cresol was collected from the yard of a wood treatment plant in South Africa and inoculated with the strains of Aspergillus, Cladosporium, Fusarium, Monicillium, Penicillium and Phanerochaete, selected from the isolate. The soil in some of the treatment reactors was amended with nutrient supplements to give a C:N:P ratio of 25:5:1. A total of 18 duplicate treatments were established and incubated in the dark at 25°C for 70days. The soil in all the reactors was tilled weekly and moisture was maintained at 70% field capacity. Soil samples were collected every 2weeks for analysis of residual concentrations of the phenols tested, pH measurement and moisture content determination. The nutrient-supplemented treatments were more effective in degrading the phenols (between 84 and 100%) than those that were not supplemented. Barley, which was used as bulking agent enhanced the growth of the fungi and subsequently the degradation of the phenols. Inoculation with a mixture of the six fungal isolates promoted more phenol degradation than with single isolates.  相似文献

Endophytic fungi live within the healthy tissues of plants and can promote host species tolerance to different environmental stresses. However, most studies have been of plants in humid environments, and there are few reports of the benefits of such associations for plants of extreme environments. Our aims were to identify endophytic fungi using morphological taxonomy, to explore richness and estimate species frequency in a cactus, C. jamacaru, in Brazilian tropical dry forest (Caatinga). We thus identified 59 taxa, corresponding to 69.7 % of the total number of isolates; the other 30.3 % were sterile mycelia. Cladosporium cladosporioides and Fusarium oxysporum were the species most commonly isolated, followed by Acremonium implicatum, Aureobasidium pullulans, Trichoderma viride, Chrysonilia sitophila, and Aspergillus flavus. Forty-seven species are recorded for the first time as endophytic fungi of cacti, and 18 others as endophytes for Brazil; this suggests that C. jamacaru harbors a highly diverse fungal community as measured by a diversity index. However, species accumulation curves suggest that our study still underestimates endophyte diversity because it does not provide an exhaustive sample. To our knowledge, this is the first report of endophytic fungi from C. jamacaru in tropical dry forests.  相似文献

This study focuses on the characteristics displayed by mesophilous and thermophilous microfungal populations occurring in two tropical monodominant plant communities, a Cocos nucifera grove and a Casuarina equisetifolia forest, that provide distinctly different edaphic conditions. The mesophilous population sampled at 25°C by the dilution plate method and the thermophilous population that developed on soil plates incubated at 45°C consisted of 1693 isolates representing 60 species and 29 genera and 8887 isolates representing 20 species and 10 genera, respectively. The mesophilous propagules averaged 9,990 per gram dry soil in the coconut grove that lacks a litter layer, is low in moisture and organic matter and is subjected to high solar irradiation. The population was characterized by the prevalence of aspergilli and dematiaceous-sphaeropsidaceous forms and the near absence of mucoraceous isolates. Ascomycetes were common. The only widespread taxa were the three species, Aspergillus niger, Penicillium chrysogenum, and Cladosporium cladosporioides. Species diversity was high and 73% of the isolates were cellulolytic. In the casuarina forest, adequate moisture and organic matter and a protecting litter layer provide a mesic environment. The mean number of mesophilous fungi per gram dry soil was 32,800. This figure is considerably lower than ones reported for mesic temperate communities and may be due to more rapid propagule removal through accelerated microfaunal and microbial activity. An abundance of mucoraceous and moniliaceous isolates and penicillia, and the rarity of aspergilli, dematiaceous-sphaeropsidaceous forms and ascomycetes characterize the population. The infrequency of aspergilli is thought to be due to their poor competitive ability. Eight species, Absidia cylindrospora, Penicillium notatum, Pestalotia cocculi?, Cylindrocarpon heteronema, Gliocladium roseum, Trichoderma viride, Paecilomyces marquandii, and Penicillium funiculosum were widespread in the area. Species diversity equaled that observed in mesic temperate communities. Less than one third of the isolates were cellulolytic. Phytopathogens were common, a feature characteristic of tropical populations. Thermophilous fungi averaged 33 per gram dry soil in the casuarina forest and increased to 943 per gram in the insolated soil of the coconut grove. Thermotolerant forms (94% of the isolates) were abundant and were principally species of Aspergillus and Chaetomium. Thermophilic fungi were rare and of the six species isolated only Chaetomiun britannicum was widespread. Four species, Ch. osmaniae, Ch. medusarum, Ch. sulphureum, and Thielavia arenaria, appear to be new records for western hemisphere soils.  相似文献

Cladosporium musae, a widespread leaf-spotting hyphomycete on Musa spp., is genetically and morphologically distinct from Cladosporium s. str. (Davidiella anamorphs, Mycosphaerellaceae, Dothideales). DNA sequence data derived from the ITS and LSU gene regions of C. musae isolates show that this species is part of a large group of hyphomycetes in the Chaetothyriales with dematiaceous blastoconidia in acropetal chains. Cladosporium adianticola, a foliicolous hyphomycete known from leaf litter in Cuba is also a member of this clade and is closely related to C. musae. A comparison with other genera in the Cladosporium complex revealed that C. musae belongs to a lineage for which no generic name is currently available, and for which the genus Metulocladosporiella gen. nov. is proposed. Two species of Metulocladosporiella are currently known, namely M. musae, which is widely distributed, and M. musicola sp. nov., which is currently known from Africa.  相似文献

The aerosolization process of fungal propagules of three species (Aspergillus versicolor, Penicillium melinii, and Cladosporium cladosporioides) was studied by using a newly designed and constructed aerosolization chamber. We discovered that fungal fragments are aerosolized simultaneously with spores from contaminated agar and ceiling tile surfaces. Concentration measurements with an optical particle counter showed that the fragments are released in higher numbers (up to 320 times) than the spores. The release of fungal propagules varied depending on the fungal species, the air velocity above the contaminated surface, and the texture and vibration of the contaminated material. In contrast to spores, the release of fragments from smooth surfaces was not affected by air velocity, indicating a different release mechanism. Correlation analysis showed that the number of released fragments cannot be predicted on the basis of the number of spores. Enzyme-linked immunosorbent assays with monoclonal antibodies produced against Aspergillus and Penicillium fungal species showed that fragments and spores share common antigens, which not only confirmed the fungal origin of the fragments but also established their potential biological relevance. The considerable immunological reactivity, the high number, and the small particle size of the fungal fragments may contribute to human health effects that have been detected in buildings with mold problems but had no scientific explanation until now. This study suggests that future fungal spore investigations in buildings with mold problems should include the quantitation of fungal fragments.  相似文献

BackgroundAspergillus is a group of opportunistic fungi that cause infections, with high morbimortality in immunosuppressed patients. Aspergillus fumigatus is the most frequent species in these infections, although the incidence of other species has increased in the last few years.AimsTo evaluate the air fungal load and the diversity of Aspergillus species in hospitals with pediatric patients in critical condition.MethodsThe Intensive Care Unit and Burns Unit of a pediatric hospital were sampled every 15 days during the autumn and spring seasons. The air samples were collected with SAS Super 100® and the surface samples were collected by swab method.ResultsThe UFC/m3 counts found exceeded the acceptable levels. The UFC/m3 and the diversity of Aspergillus species found in the Intensive Care Unit were higher than those found in the Burns Unit. The fungal load and the diversity of species within the units were higher than those in control environments. The use of both methods –SAS and swab– allowed the detection of a higher diversity of species, with 96 strains of Aspergillus being isolated and 12 species identified. The outstanding findings were Aspergillus sydowii, Aspergillus niger, Aspergillus flavus, Aspergillus terreus and Aspergillus parasiticus, due to their high frequency. Aspergillus fumigatus, considered unacceptable in indoor environments, was isolated in both units.ConclusionsAspergillus was present with high frequency in these units. Several species are of interest in public health for being potential pathogenic agents. Air control and monitoring are essential in the prevention of these infections.  相似文献

The Havana aeromycobiota diversity was studied from November 2010 to October 2011 using two complementary volumetric methods. A total of 35 fungal genera were characterised, 26 of them were recognised only by non-viable methods, six with viable methodology and the other three with both sampling methods. Furthermore, 47 species were identified by cultivation and the spores collected with the non-viable methodology. These could not be included in a specific genus, and thus, were categorised into five fungal types. In general, the main, spread worldwide, mitosporic fungi also predominated the Havana atmosphere. The predominant species were Cladosporium cladosporioides, Aspergillus flavus and Penicillium citrinum. Moreover, several Zygomycetes (Syncephalastrum racemosum, Rhizopus stolonifer and Rhizopus oryzae), Ascomycetes (Chaetomium globosum) and Basidiomycetes such as Coprinus or Ganoderma were isolated. In the present paper, the review of the airborne fungi conducted in previous studies in Cuba was completed by the detection of two new genera and the first isolation of ten new records in the Cuban atmosphere. Most of the fungi detected showed a diurnal pattern with high spore peak at 11.00–12.00.  相似文献

We examined 12,026 fungal air samples (9,619 indoor samples and 2,407 outdoor samples) from 1,717 buildings located across the United States; these samples were collected during indoor air quality investigations performed from 1996 to 1998. For all buildings, both indoor and outdoor air samples were collected with an Andersen N6 sampler. The culturable airborne fungal concentrations in indoor air were lower than those in outdoor air. The fungal levels were highest in the fall and summer and lowest in the winter and spring. Geographically, the highest fungal levels were found in the Southwest, Far West, and Southeast. The most common culturable airborne fungi, both indoors and outdoors and in all seasons and regions, were Cladosporium, Penicillium, nonsporulating fungi, and Aspergillus. Stachybotrys chartarum was identified in the indoor air in 6% of the buildings studied and in the outdoor air of 1% of the buildings studied. This study provides industrial hygienists, allergists, and other public health practitioners with comparative information on common culturable airborne fungi in the United States. This is the largest study of airborne indoor and outdoor fungal species and concentrations conducted with a standardized protocol to date.  相似文献

Investigations were made on the effect of species of fungi isolated from therhizosphere of broad bean (Vicia faba Linn.) variety Giza I, and cotton (Gossypium barbadense Linn.) variety Giza 47, on plant growth. Broad bean rhizosphere fungi differently affected plant growth.Fusarium moniliforme, Penicillium martensii, Rhizopus stolonifer, andCladosporium sphaerospermum stimulated both root and shoot growth.Aspergillus niger andAlternaria tenuis have an inhibitory effect on plant growth. The objectives of this study were to evaluate the microbial prevalence inside six repositories of the National Archive of the Republic of Cuba in 2?months of the year and to examine some of the physiological features of fungi isolated in order to evaluate their potential for biodeterioration. The microbiological sampling was conducted in February and September using a slit impactor as air sampler. Appropriate selective culture media were used to isolate fungi and bacteria. Temperature and relative humidity were measured during the samplings. The cellulolytic activity and the production of acids and pigments of the fungi isolated were qualitatively determined. Total viable microbiota and bacteria concentrations were greater in February while the fungal concentration was higher in September. Aspergillus, Cladosporium, Penicillium, Curvularia and Alternaria were the predominant fungal genera in February while Cladosporium prevailed in September, although Fusarium, Mucor and Neurospora genera were also isolated in this month. The fungi isolated were capable of degrading cellulose and excreting pigments and acids. The Gram-positive bacteria group prevailed in the air and Corynebacterium, Streptomyces, Bacillus, Streptococcus, Staphylococcus, Enterobacter and Serratia were some of the genera identified.  相似文献

100 species and 2 varieties of A. nidulans which belong to 35 genera were identified from 40 samples of barley collected from different places in Upper Egypt. Two methods of isolation were used: the grain-and the dilution plate. The spectrum of fungal genera and species collected by the former (35 genera, 96 species+1 variety) was broader than by the latter method (28 genera, 76 species+1 variety). Nine genera were of high occurrence (represented in more than 50% of the samples) namely, Aspergillus, Penicillium, Fusarium, Alternaria, Chaetomium, Cladosporium, Humicola, Drechslera and Myrothecium. The most frequent species were A. fumigatus, A. niger, A. flavus, A. sydowii, F. oxysporum, F. moniliforme, A. alternata, M. verrucaria, P. funiculosum, P. corylophilum, P. duclauxi, C. herbarum, C. globosum, D. spicifera and H. grisea. Several fungi such as Chaetomium, Humicola, Altemaria, Drechslera, Stachybotrys, Myrothecium, Pénicillium corylophilum and P. funiculosum were far more frequently recovered on cellulose than on glucose agar.  相似文献

Original data on the survival of fungal spores exposed to space conditions are presented. The experiment was carried out on the Earth-orbiting Russian satellite Foton-M4. The flight duration of the satellite was 45 days. Thirteen fungal species (hyaline as well as pigmented) from 10 genera recovered from destructed stone materials were studied. Sterile quartz sand was inoculated by the fungal spores and was placed into Eppendorf tubes. During the space flight, the Eppendorf tubes with fungal spores were kept inside the Foton descent capsule in the “Biokont” containers and on the external surface of the capsule in the “Exobiofrost” containers exposed to the open space as well. Spores of ten species (77% of all tested species), i.e. Acremonium charticola, Aspergillus niger, Aspergillus versicolor, Chaetomium globosum, Cladosporium sphaerospermum, Penicillium chrysogenum, Penicillium verrucosum, Purpureocillium lilacinum, Sarocladium kiliense, and Trichoderma harzianum, survived after the flight both inside and outside the descent capsule. Only three species (23% of all tested species), i.e. Acremonium furcatum, Engyodontium album and Verticillium zaregamsianum, failed to survive outside as well as inside the capsule. Spore viability differed depending on the fungal species. Thus, spores of some fungal species are able to survive under the complex of stress factors such as low temperature values, radiation, etc. We have shown that micromycetes can be used as a model group for study of eukaryotic organisms’ resistance to stress factors, due to their high tolerance not only to extreme terrestrial environments, but to the extraterrestrial ones as well.  相似文献
