The locust is well known for its ability to jump large distances to avoid predation. This class sets out a series of investigations into the mechanisms underlying the jump enabling students to bring together information from biomechanics, muscle physiology, and anatomy. The nature of the investigation allows it to be undertaken at a number of levels of complexity from relatively simple comparative observations to detailed analysis of the properties of the muscles and the energy storage systems involved in powering the jump. The relative size and robustness of the locust make it simple to handle and ideal for such investigations. 相似文献

The laboratory rat, Rattus novegicus, is a major model system for physiological and pathophysiological studies, and since 1966 more than 422,000 publications describe
biological studies on the rat (NCBI/Medline). The rat is becoming an increasingly important genetic model for the study of
specific diseases, as well as retaining its role as a major preclinical model system for pharmaceutical development. The initial
genetic linkage map of the rat contained 432 genetic markers (Jacob et al. 1995) out of 1171 developed due to the relatively
low polymorphism rate of the mapping cross used (SHR × BN) when compared to the interspecific crosses in the mouse. While
the rat genome project continues to localize additional markers on the linkage map, and as of 11/97 more than 3,200 loci have
been mapped. Current map construction is using two different crosses (SHRSP × BN and FHH × ACI) rather than the initial mapping
cross. Consequently there is a need to provide integration among the different maps. We set out to develop an integrated map,
as well as increase the number of markers on the rat genetic map.
The crosses available for this analysis included the original mapping cross SHR × BN reciprocal F2 intercross (448 markers),
a GH × BN intercross (205 markers), a SS/Mcw × BN intercross (235 markers), and a FHH/Eur × ACI/Hsd intercross (276 markers),
which is also one of the new mapping crosses. Forty-six animals from each cross were genotyped with markers polymorphic for
that cross. The maps appear to cover the vast majority of the rat genome. The availability of these additional markers should
facilitate more complete whole genome scans in a greater number of strains and provide additional markers in specific genomic
regions of interest.

The effects of Rose Bengal (RB) on plate counts of the bacteria Escherichia coli and Pseudomonas aeruginosa, and the yeast Saccharomyces cerevisiae, were studied under natural sunlight and artificial fluorescent lighting. While RB was not inherently toxic in darkness at concentrations found in mycological media, the illumination of media containing RB caused a decrease in colony counts in all cases, and especially for surface spread plates. A negative synergy was observed between chloramphenicol, RB and illumination using a spring water sample containing substantial numbers of Gram-negative bacteria. Exposure of media containing RB to moderate amounts of light during standard laboratory procedures may inhibit microbial growth, with positive benefits in relation to the suppression of contaminant bacteria, or negative implications where fungi are inhibited. 相似文献

MOTIVATION: The development of an integrated genetic and physical map for the maize genome involves the generation of an enormous amount of data. Managing this data requires a system to aid in genotype scoring for different types of markers coming from both local and remote users. In addition, researchers need an efficient way to interact with genetic mapping software and with data files from automated DNA sequencing. They also need ways to manage primer data for mapping and sequencing and provide views of the integrated physical and genetic map and views of genetic map comparisons. RESULTS: The MMP-LIMS system has been used successfully in a high-throughput mapping environment. The genotypes from 957 SSR, 1023 RFLP, 189 SNP, and 177 InDel markers have been entered and verified via MMP-LIMS. The system is flexible, and can be easily modified to manage data for other species. We describe an integrated proteome database, termed Yonsei Proteome Research Center Proteome Database (YPRC-PDB) which can store, retrieve and analyze various information including two-dimensional electrophoresis (2-DE) images and associated spot information that were obtained during studies of hepatocellular carcinoma (HCC). YPRC-PDB is also designed to perform as a laboratory information management system that manages sample information, clinical background, conditions of both sample preparation and 2-DE, and entire sets of experimental results. It also features query system and data-mining applications, which are amenable to automatically analyze expression level changes of a specific protein and directly link to clinical information. The user interface is web-based, so that the results from other laboratories can be shared effectively. In particular, the master gel image query is equipped with a graphic tool that can easily identify the relationship between the specific pathological stage of HCC and expression levels of a potential marker protein on the master gel image. Thus, YPRC-PDB is a versatile integrated database suitable for subsequent analyses. The unique bio-analytical properties of the amino acid tyrosine (Tyr) are the focus of this experiment from the research oriented biochemistry laboratory course at our university. In the present study pK(a(1)), pK(a(2)), and pK(a(3)) values for free Tyr were estimated to be 2.30, 9.40, and 9.97, respectively, when free Tyr was titrated with 1mM NaOH and 1mM HCl using a pH meter. Spectrophotometric analysis of the phenolic side chain pK(a(3)) revealed a value of 10.14, which was consistent with the pK(a)s estimated from the pH meter. The results from this experiment will allow students to compare the free Tyr properties with those present in a protein. 相似文献
