The stable isotopes of nitrogen (δ15N) and carbon (δ13C) provide powerful tools for quantifying trophic relationships and carbon flow to consumers in food webs; however, the isotopic signatures of organisms vary within a lake. Assessment of carbon and nitrogen isotopic signatures in a suite of plants, invertebrates, and fishes in Lake Kyoga, indicated significant variation between two sites for δ13C (paired t = 6.305; df = 14, P < 0.001 and δ15N paired t = 1.292; df = 14; P < 0.05). The fish fauna in Bukungu was generally more 13C enriched (mean δ13C = –16.37 ± 1.64‰) than in Iyingo (mean δ13C = –20.80 ± 2.41‰) but more δ15N depleted (mean δ15N = 5.57 ± 0.71‰) than in Iyingo (mean δ15N = 6.92 ± 0.83‰). The simultaneous shifts in phytoplankton and consumer signatures confirmed phytoplankton as the major source of carbon for the food chain leading to fish. Limited sampling coverage within lakes may affect lake wide stable isotope signatures, and the same error is transferred into trophic position estimation. Consideration of potential intra‐lake spatial variability in isotope ratios and size is essential in evaluating the spatial and trophic structure of fish assemblages.  相似文献

Stable isotopes of carbon (C) and nitrogen (N) are commonly used to track resource flows through lake food webs. However, there remains a weak understanding of the spatial variation in isotopic composition of benthic resources and how this variation affects inference about energy flows among species. Boundary layers at the interface between benthic substrates and the overlying water column restrict diffusion of nutrients and carbon from the water column to benthic algae and may affect the isotopic composition of benthic algae as nutrient and CO2 concentrations can become locally depleted in the benthic boundary layer. We quantified the variation in C and N stable isotope composition of benthic resources along a depth gradient in a large oligotrophic lake to assess the magnitude of change in stable isotope composition. Snails were increasingly depleted in 13C with depth, by about 10 ‰ from 0 to 20 m, while 15N in snails showed only subtle enrichment over this depth range. Sculpin (Cottas aleuticus) δ 13C and δ 15N signatures did not significantly change with depth and were more enriched in 15N than would be expected from consumption of snails alone. A comparison of δ 13C and δ 15N values from sculpins relative to shallow and deep snails, and alternative prey (marine-derived salmon resources), within a mixing model suggested sculpins feed selectively on deep grazers in this system in addition to marine-derived resources provided by migrating sockeye salmon. This study illustrates the importance of accounting for depth-related variation in isotope patterns when assessing benthic resource contributions to food webs using stable isotope data.  相似文献

Evaluating tissue fractionation between mothers and their offspring is fundamental for informing our interpretation of stable isotope values in young individuals and can provide insight into the dynamics of maternal provisioning. The objectives of this study were to investigate the isotopic relationships between maternal reproductive (i.e., yolk, yolk-sac placenta) and somatic tissues (i.e., muscle and liver) relative to embryos in the Bonnethead Shark Sphyrna tiburo, to evaluate the fractionation of stable carbon (δ13C) and nitrogen (δ15N) isotopes between these tissues. Additionally, we examined intra-uterine variability in the isotopic relationships to ascertain whether this species may exhibit variable nutrient allocation. Embryos showed similar magnitudes of enrichment in 13C (i.e., Δδ13C, difference between adult and embryo) relative to adult tissues (Δδ13C?=?~1.0‰). However, embryos were depleted in 15N relative to adult muscle tissues (Δδ15N?=??1.0‰), a finding that contrasts Δδ15N values reported for other placentotrophic sharks. Embryo-muscle Δδ15N was correlated with length, supporting the contention that the magnitude of enrichment between embryonic and maternal tissues results from the shift from yolk to placental feeding. Embryo δ15N and Δδ15N values showed significant intra-uterine variability; a result not observed for δ13C and Δδ13C values. The contrasting patterns in fractionation among placentotrophic sharks highlight the importance of evaluating these relationships across elasmobranch taxa with consideration for different tissues, reproductive strategies and stages of gestation. The divergent findings support future evaluation of stable isotope relationships between mothers and offspring for purposes of estimating inherent isotopic variability and how this variability may inform physiological and dietary mechanisms.  相似文献

1. The variability in the stable isotope signatures of carbon and nitrogen (δ13C and δ15N) in different phytoplankton taxa was studied in one mesotrophic and three eutrophic lakes in south‐west Finland. The lakes were sampled on nine to 16 occasions over 2–4 years and most of the time were dominated by cyanobacteria and diatoms. A total of 151 taxon‐specific subsamples covering 18 different phytoplankton taxa could be isolated by filtration through a series of sieves and by flotation/sedimentation, followed by microscopical identification and screening for purity. 2. Substantial and systematic differences between phytoplankton taxa, seasons and lakes were observed for both δ13C and δ15N. The values of δ13C ranged from ?34.4‰ to ?5.9‰ and were lowest in chrysophytes (?34.4‰ to ?31.3‰) and diatoms (?30.6‰ to ?26.6‰). Cyanobacteria were most variable (?32.4‰ to ?5.9‰), including particularly high values in the nostocalean cyanobacterium Gloeotrichia echinulata (?14.4‰ to ?5.9‰). For δ13C, the taxon‐specific amplitude of temporal changes within a lake was usually <1–8‰ (<1–4‰ for microalgae alone and <1–8‰ for cyanobacteria alone), whereas the amplitude among taxa within a water sample was up to 31‰. 3. The values of δ15N ranged from ?2.1‰ to 12.8‰ and were high in chrysophytes, dinophytes and diatoms, but low in the nitrogen‐fixing cyanobacteria Anabaena spp., Aphanizomenon spp. and G. echinulata (?2.1‰ to 1.6‰). Chroococcalean cyanobacteria ranged from ?1.4‰ to 8.9‰. For δ15N, the taxon‐specific amplitude of temporal changes within a lake was 2–6‰, (2–6‰ for microalgae alone and 2–4‰ for cyanobacteria alone) and the amplitude among taxa within a water sample was up to 11‰. 4. The isotopic signatures of phytoplankton changed systematically with their physical and chemical environment, most notably with the concentrations of nutrients, but correlations were non‐systematic and site‐specific. 5. The substantial variability in the isotopic signatures of phytoplankton among taxa, seasons and lakes complicates the interpretation of isotopic signatures in lacustrine food webs. However, taxon‐specific values and seasonal patterns showed some consistency among years and may eventually be predictable.  相似文献

Stable isotope analysis (SIA) in combination with growth analysis using scales collected from adult chum salmon Oncorhynchus keta migrating back to Hokkaido, Japan, was performed to describe the variation of isotopic composition of carbon (δ13C) and nitrogen (δ15N) in scales and to examine relationships with growth traits [age, fork length (FL), and relative growth ratio in the last growth period [(RGRlast)]. Scale stable isotope (SI) values in 3‐ to 6‐year‐old fish were highly variable, ranging from ?17.6‰ to ?14.3‰ for δ13C and 9.5‰ to 13.4‰ for δ15N. The δ15N was positively correlated with FL, and this tendency may indicate changes in trophic level with growth. Significant effect was not detected between δ15N and RGRlast, it can be inferred that factors potentially yielding high δ15N may not necessarily result in higher growth rates. No trend found between FL and δ13C may imply that there is no clear segregation in feeding locations between the 3‐ to 6‐year groups. This study provided basic information for scale SI values of chum salmon adults and indicated that SIA using scales could be a new approach to elucidating the trophic ecology of chum salmon.  相似文献

Stable carbon, nitrogen, hydrogen and oxygen isotopes have been used to infer aspects of species ecology and environment in both modern ecosystems and the fossil record. Compared to large mammals, stable isotopic studies of small‐mammal ecology are limited; however, high species and ecological diversity within small mammals presents several advantages for quantifying resource use and organism–environment interactions using stable isotopes over various spatial and temporal scales. We analyzed the isotopic composition of hair from two heteromyid rodent species, Dipodomys ordii and Perognathus parvus, from localities across western North America in order to characterize dietary variation in relation to vegetation and climatic gradients. Significant correlations between the carbon isotopic composition (δ13C) of these species and several climatic variables imply that seasonal temperature and precipitation control the composition and distribution of dietary resources (grass seeds). Our results also suggest a moisture influence on the nitrogen isotopic composition (δ15N) of heteromyid diets. Population‐ and species‐level variation in δ13C and δ15N values record fine‐scale habitat heterogeneity and significant differences in resource use between species. Using classification and regression‐tree techniques, we modeled the geographic variation in heteromyid δ13Cdiet values based on 10 climatic variables and generated an isotope landscape model (‘isoscape’). The isoscape predictions for δ13Cdiet differ from expectations based on observed C4 distributions and instead indicate that D. ordii and P. parvus record seasonally abundant grass resources, with additional model deviations potentially attributed to geographic variation in dietary selection. The oxygen and hydrogen isotopic composition of D. ordii is enriched relative to local meteoric water and suggests that individuals rely on highly evaporated water sources, such as seed moisture. Based on the climatic influences on vegetation and diet documented in this study, the isotopic composition of small mammals has high potential for recording ecological responses to environmental changes over short and long time scales.  相似文献

1. Despite the ubiquity and abundance of water striders (Hemiptera: Gerridae) in temperate streams and rivers and their potential usefulness as sentinels in contaminant studies, little is known about their feeding ecology and lipid dynamics. 2. In this study we used stable isotopes of carbon (δ13C) and nitrogen (δ15N) and elemental carbon to nitrogen ratios (C/N) to assess dietary habits and lipid content, respectively, for water striders. 3. To determine diet‐tissue fractionation factors, nymphs of the most common species in New Brunswick, Canada, Aquarius remigis were reared in the laboratory for 73 days and exhibited rapid isotopic turnover in response to a switch in diet (C half‐life = 1.5 days, N half‐life = 7.8 days). Their lipid content increased towards the end of the growing season and resulted in lower δ13C values. Diet‐tissue fractionation factors were established after correction of δ13C data for the confounding effect of de novo lipid synthesis (strider δ13Cadj– diet δ13Cadj = 0.1‰, strider δ15N – diet δ15N = 2.7‰). 4. Water striders from the majority of 45 stream sites (83%) in New Brunswick had less than 50% contribution of aquatic carbon to their diets but showed a gradual increase in the contribution of this carbon source to their diet with increasing stream size. 5. These data indicate that striders exhibit a strong connection to terrestrial carbon sources, making them important users of energy subsidies to streams from the surrounding catchment. However, this dependence on terrestrial organic matter may limit their utility as indicators of contamination of aquatic systems by heavy metals and other pollutants.  相似文献

We compared the δ13C and δ15N of forest material with an extensive sporocarp collection to elucidate the role of litter, wood and soil as fungal carbon and nitrogen sources in Finnish boreal Picea abies-dominated forests. Ectomycorrhizal Hydnum and Cortinarius had higher δ15N than other ectomycorrhizal fungi, suggesting use of 15N-enriched, deeper nitrogen. Russula had lower δ15N than other ectomycorrhizal fungi and resembled some litter decay genera, suggesting use of litter-derived nitrogen. There was little variation in δ15N among other genera of ectomycorrhizal fungi, indicating limited functional diversity in nitrogen use. Saprotrophic Leotia, Gymnopus, Hypholoma, Pholiota, Rhodocollybia and Calocera had δ15N values similar to ectomycorrhizal fungi, indicating overlap in use of older nitrogen from soil or roots or use of newly fixed nitrogen. Genera of litter and wood decay fungi varied up to 6‰ in δ13C and 10‰ in δ15N, suggesting large differences in carbon and nitrogen sources and processing. Similar δ13C between white and brown rot wood decay fungi also suggest that white rot fungi do not use lignin-derived carbon. Together, these δ13C and δ15N patterns of fungi from Finnish boreal forests enhance our knowledge of fungal functional diversity and indicate broad use of litter, wood and soil resources.  相似文献

Southern African forests are naturally fragmented yet hold a disproportionately high number of bird species. Carbon and nitrogen stable isotopes were measured in feathers from birds captured at Woodbush (n = 27 species), a large afromontane forest in the eastern escarpment of Limpopo province, South Africa. The δ13C signatures of a range of forest plants were measured to categorise the food base. Most plants sampled, including two of five grass species, had δ13C signatures typical of a C3 photosynthetic pathway (?29.5 ± 1.9‰). Three grass species had a C4 signature (?12.0 ± 0.6‰). Most bird species had δ13C values representing a predominantly C3‐based diet (?24.8‰ to ?20.7‰). δ15N values were as expected, with higher levels of enrichment associated with a greater proportion of dietary animal matter. The cohesive isotopic niche defining most species (n = 22), where the ranges for δ13C and δ15N were 2.4‰ and 3.4‰, respectively, highlight the difficulties in understanding diets of birds in a predominantly C3‐based ecosystem using carbon and nitrogen stable isotopes. However, variation in isotopic values between and within species provides insight into possible niche width and the use of resources by different birds within a forest environment.  相似文献

1. Adult Pacific salmon (Oncorhynchus spp.) transport marine nutrients to fresh waters and disturb sediments during spawning. The relative importance of nutrient fertilisation and benthic disturbance by salmon spawners can be modulated by environmental conditions (e.g. biological, chemical and physical conditions in the catchment, including human land use). 2. To determine the importance of the environmental context in modifying the uptake and incorporation of salmon‐derived material into stream biota, we measured the nitrogen (δ15N) and carbon (δ13C) isotopic composition of benthic algae (i.e. epilithon) and juvenile coho salmon (Oncorhynchus kisutch) in seven streams across a timber‐harvest gradient (8–69% catchment area harvested), both before and during the salmon run. Conditional bootstrap modelling simulations were used to assess variability in the response of epilithon and juvenile coho salmon to spawning salmon. 3. In response to spawning salmon, epilithon exhibited enrichment in both δ15N (mean: 1.5‰) and δ13C (2.3‰). Juvenile coho were also enriched in both δ15N (0.7‰) and δ13C (1.4‰). Conditional bootstrap models indicate decreased variation in data as spatial replication increases, suggesting that the number of study sites can influence the results of Pacific salmon isotope studies. 4. Epilithon isotopic enrichment was predicted by environmental conditions, with δ15N enrichment predicted by stream temperature and timber harvest (R2 = 0.87) and δ13C enrichment by discharge, sediment size, timber harvest and spawner density (R2 = 0.96). Furthermore, we found evidence for a legacy effect of salmon spawners, with pre‐spawner δ15N and δ13C of both epilithon and juvenile coho predicted by salmon run size in the previous year. 5. Our results show that the degree of incorporation of salmon‐derived nitrogen and carbon differs among streams. Furthermore, the environmental context, including putative legacy effects of spawning salmon, can influence background isotopic concentrations and utilisation of salmon‐derived materials in southeast Alaska salmon streams. Future studies should consider the variation in isotopic composition of stream biota when deciding on the number of study sites and samples needed to generate meaningful results.  相似文献

Temporal changes in fatty acid composition and δ15N, δ13C stable isotope values of the phytobenthos growing on artificial clay substrates under natural conditions over a 28-day period at an upstream and a downstream site in the Kowie River near Grahamstown were investigated in 2012. High concentrations of diatom markers 16:1ω7 and 20:5ω3 fatty acids were recorded, especially at the downstream site, reflecting the importance of diatoms in contributing to the phytobenthos communities at that station. After day 7 at the downstream site the average δ15N value of the phytobenthos was lighter, gradually increasing by ~2‰ and ~5‰ overall to heavier values on day 28. At the upstream site there were no significant changes (<1‰ increase) in δ15N values of the phytobenthos over time. Stable nitrogen (δ15N) and carbon (δ13C) signatures in the phytobenthos communities were significantly different between sites (one-way ANOVA; p < 0.001). The stable isotope values and fatty acid concentrations of phytobenthos at the downstream site were different to those of the phytobenthos at the upstream site, and they changed concurrently with changes in the phytobenthos community structure. At the downstream site there was a strong correlation of the δ15N of phytobenthos with nitrates (R = 0.56) and time (weeks; R = 0.81). However, the fatty acids were not specific enough to characterise the composition of phytobenthos communities. Other biomarker methods, such as stable isotopes and microscopic examination of the communities, were found to be useful. The results from this relatively small-scale tile experiment indicate the complexity of changes in fatty acid composition and δ15N, δ13C stable isotope values of a phytobenthos community. Stable isotope and fatty acid composition can be successfully used to map changes in phytobenthos composition and carbon and nitrogen flow patterns along a river continuum.  相似文献

Stable carbon and nitrogen isotope ratios (δ13C and δ15N) of serum, red blood cells (RBC), muscle, and blubber were measured in captive and wild northeast Pacific harbor seals (Phoca vitulina richardii) at three coastal California sites (San Francisco Bay, Tomales Bay, and Channel Islands). Trophic discrimination factors (ΔTissue‐Diet) were calculated for captive seals and then applied in wild counterparts in each habitat to estimate trophic position and feeding behavior. Trophic discrimination factors for δ15N of serum (+3.8‰), lipid‐extracted muscle (+1.6‰), and lipid‐blubber (+6.5‰) are proposed to determine trophic position. An offset between RBC and serum of +0.3‰ for δ13C and ?0.6‰ for δ15N was observed, which is consistent with previous research. Specifically, weaner seals (<1 yr) had large offsets, suggesting strong trophic position shifts during this life stage. Isotopic values indicated an average trophic position of 3.6 at both San Francisco Bay and Tomales Bay and 4.2 at Channel Islands. Isotopic means were strongly dependent on age class and also suggested that mean diet composition varies considerably between all locations. Together, these data indicate that isotopic composition of blood fractions can be an effective approach to estimate trophic position and dietary behavior in wild pinnipeds.  相似文献

Stable nitrogen (δ15N) and carbon (δ13C) isotopes of Atlantic sharpnose shark Rhizoprionodon terraenovae embryos and mothers were analysed. Embryos were generally enriched in 15N in all studied tissue relative to their mothers' tissue, with mean differences between mother and embryo δ15N (i.e. Δδ15N) being 1·4‰ for muscle, 1·7‰ for liver and 1·1‰ for cartilage. Embryo muscle and liver were enriched in 13C (both Δδ13C means = 1·5‰) and embryo cartilage was depleted (Δδ13C mean = ?1·01‰) relative to corresponding maternal tissues. While differences in δ15N and δ13C between mothers and their embryos were significant, muscle δ15N values indicated embryos to be within the range of values expected if they occupied a similar trophic position as their respective mothers. Positive linear relationships existed between embryo total length (LT) and Δδ15N for muscle and liver and embryo LT and Δδ13C for muscle, with those associations possibly resulting from physiological differences between smaller and larger embryos or differences associated with the known embryonic nutrition shift (yolk feeding to placental feeding) that occurs during the gestation of this placentatrophic species. Together these results suggest that at birth, the δ15N and δ13C values of R. terraenovae are likely higher than somewhat older neonates whose postpartum feeding habits have restructured their isotope profiles to reflect their postembryonic diet.  相似文献

Within-lake variability in carbon and nitrogen stable isotope signatures   总被引:3,自引:0,他引:3
1. We assessed spatial and temporal variation in carbon and nitrogen isotopic signatures in different compartments of a single lake ecosystem. Stable isotope analyses were made on samples of particulate organic matter (POM), zooplankton, periphyton, macrophytes, macroinvertebrates and fish collected from several locations throughout the ice‐free period. 2. No spatial variation in δ13C or δ15N values was found for pelagic samples of POM and zooplankton. However, pelagic δ15N signatures increased steadily through the summer resulting in an almost 6‰ average increase in POM and zooplankton. A concurrent decrease in epilimnetic nitrate concentrations suggested that the increase in δ15N of POM and zooplankton could have resulted from a progressive 15N‐enrichment of the available inorganic nitrogen pool as the size of this pool was reduced. 3. Significant spatial variation in isotopic ratios was observed within littoral and profundal communities. Some spatial differences were likely related to lake‐specific characteristics, such as a major inlet and a small harbour area and some were interconnected with temporal events. 4. Marked differences between spring and autumn δ15N and δ13C values of fish at one site probably reflected a spring spawning immigration from a larger downstream lake and also indicated limited dispersal of these immigrants. 5. Our results indicate that restricted sampling of ecosystem components from lakes may provide misleading single values for the isotope end members needed for quantitative uses of stable isotopes in mixing models and for estimating trophic position. Hence we strongly advise that studies of individual lakes, or multiple lake comparisons, that utilise stable isotope analyses should pay more attention to potential within lake spatial and temporal variability of isotope ratios.  相似文献

Interspecific competition is considered a major determinant of ecological niche. It is hypothesized that increased competition should reduce niche breadth. However, there are scarce field tests on this hypothesis. Here, we test this central hypothesis in ecology by using the Atlas day gecko Quedenfeldtia trachyblepharus. This alpine gecko faces fewer competitors as altitude increases, and thereby, we predict that this species should increase niche breadth and relevant fitness parameters with altitude. We tested this prediction by analysing the isotopic signature of carbon (δ13C) and nitrogen (δ15N). Our results reveal that specimens from higher altitudes showed higher values for both carbon (δ13C) and nitrogen (δ15N) isotopes, had better body condition and a greater isotopic breadth when compared to specimens from lower altitudes. Altitudinal variation in carbon values was not explained by variation in isotopic concentration in the baseline of the trophic chain. Therefore, our findings support the prediction that relaxed interspecific competition favours increased trophic niche breadth. These results also suggest that global warming may represent an important threat for this species, as it may provoke the ascent in altitude of competitors, with negative consequences for the conservation of this endemism.  相似文献

Sub-fossil wood is often affected by the decaying process that introduces uncertainties in the measurement of oxygen and carbon stable isotope composition in cellulose. Although the cellulose stable isotopes are widely used as climatic proxies, our understanding of processes controlling their behavior is very limited. We present here a comparative study of stable oxygen and carbon isotope ratios in tree ring cellulose in decayed and non-decayed wood samples of Swiss stone pine (Pinus cembra) trees. The intra-ring stable isotope variability (around the circumference of a single ring) was between 0.1 and 0.5‰ for δ18O values and between 0.5 and 1.6‰ for δ13C values for both decayed and non-decayed wood. Observed intra-tree δ18O variability is less than that reported in the literature (0.5–1.5‰), however, for δ13C it is larger than the reported values (0.7–1.2‰). The inter-tree variability for non-decayed wood ranges between 1.1 and 2.3‰ for δ18O values, and between 2 and 4.7‰ for δ13C values. The inter-tree differences for δ18O values are similar to those reported in the literature (1–2‰ for oxygen and 1–3‰ for carbon) but are larger for δ13C values. We have found that the differences for δ18O and δ13C values between decayed and non-decayed wood are smaller than the variation among different trees from the same site, suggesting that the decayed wood can be used for isotopic paleoclimate research.  相似文献

The trophodynamics of pelagic and benthic animals of the North Sea, North Atlantic shelf, were assessed using stable isotope analysis (SIA) of natural abundance carbon and nitrogen isotopes, lipid fingerprinting and compound-specific SIA (CSIA) of phospholipid-derived fatty acids (PLFAs). Zooplankton (z), epi- and supra-benthic macrofauna were collected in the Southern Bight, at the Oyster Grounds and at North Dogger, 111 km north of the Dogger Bank. The study included 22 taxonomic groups with particular reference to Mollusca (Bivalvia and Gastropoda) and Crustacea. Primary consumers (Bivalvia) were overall most 15N enriched in the southern North Sea (6.1‰) and more depleted in the Oyster Grounds (5.5‰) and at North Dogger (2.8‰) demonstrating differences in isotopic baselines for bivalve fauna between the study sites. Higher trophic levels also followed this trend. Over an annual cycle, consumers tended to exhibit 15N depletion during spring followed by 15N enriched signatures in autumn and winter. The observed seasonal changes of δ 15N were more pronounced for suspension feeders and deposit feeders (dfs) than for filter feeders (ffs). The position of animals in plots of δ 13C and δ 15N largely concurred with the expected position according to literature-based functional feeding groups. PLFA fingerprints of groups such as z were distinct from benthic groups, e.g. benthic ffs and dfs, and predatory macrobenthos. δ 13CPLFA signatures indicated similarities in 13C moiety sources that constituted δ 13CPLFA. Although functional groups of pelagic zooplankton and (supra-) benthic animals represented phylogenetically distinct consumer groups, δ 13CPLFA demonstrated that both groups were supported by pelagic primary production and relied on the same macronutrients such as PLFAs. Errors related to the static categorization of small invertebrates into fixed trophic positions defined by phylogenetic groupings rather than by functional feeding groups, and information on seasonal trophodynamic variability, may have implications for the reliability of numerical marine ecosystem models.  相似文献

Nitrogen isotope composition (δ15N) in plant organic matter is currently used as a natural tracer of nitrogen acquisition efficiency. However, the δ15N value of whole leaf material does not properly reflect the way in which N is assimilated because isotope fractionations along metabolic reactions may cause substantial differences among leaf compounds. In other words, any change in metabolic composition or allocation pattern may cause undesirable variability in leaf δ15N. Here, we investigated the δ15N in different leaf fractions and individual metabolites from rapeseed (Brassica napus) leaves. We show that there were substantial differences in δ15N between nitrogenous compounds (up to 30‰) and the content in (15N enriched) nitrate had a clear influence on leaf δ15N. Using a simple steady‐state model of day metabolism, we suggest that the δ15N value in major amino acids was mostly explained by isotope fractionation associated with isotope effects on enzyme‐catalysed reactions in primary nitrogen metabolism. δ15N values were further influenced by light versus dark conditions and the probable occurrence of alternative biosynthetic pathways. We conclude that both biochemical pathways (that fractionate between isotopes) and nitrogen sources (used for amino acid production) should be considered when interpreting the δ15N value of leaf nitrogenous compounds.
