Conclusion and summary As theI σ-index is neither affected by the mean per sample unit except for regular distribution nor standing on the assumption of any definite type of contagious distribution, it may have the most wide range of application among the ones hitherto deviced for measuring the dispersion of individuals in a population. The relations of theI σ-index to ,k of negative binomial distribution andN of binomial distribution as well as the new dispersion index,I B , given in this paper may serve, if necessary, for the analysis of data in the ecological works. The bulk of the secretion of the subcommissural organ is formed by glycoproteins that appear to be derived from two precursor forms of 540 and 320 kDa. Upon release into the ventricle, these glycoproteins aggregate to form Reissner’s fiber. We report the isolation of three cDNA clones from a cDNA library prepared from bovine subcommissural organ RNA, by using an anti-Reissner’s fiber serum for immunoscreening. Inserts of 0.7, 1.2, and 2.5 kb were amplified by the polymerase chain reaction, subcloned into pUC18 vector, and sequenced. Although restriction mapping of the three inserts initially suggested that all of them were derived from the same mRNA, sequence analysis showed that a short non-homologous region was present in the 0.7-kb insert when compared with the 1.2-kb and 2.5-kb inserts, suggesting that they corresponded to two different, although highly homologous, mRNAs. Northern analyses showed a single mRNA species of approximately 9.5 kb present in the subcommissural organ and missing in the choroid plexus, brain cortex, and liver. In situ hybridization confirmed that the expression of the RNA was restricted to cells of the bovine subcommissural organ. Polyclonal antibodies raised against a synthetic peptide, whose amino-acid sequence was deduced from the 2.5-kb cDNA, reacted specifically with the bovine and rat subcommissural organ-Reissner’s fiber complex. In immunoblots of bovine subcommissural organ, this antibody revealed the precursor 540-kDa form and its putative processed form of 450 kDa. It is concluded that the cloned cDNA encodes for the major constitutive glycoprotein of Reissner’s fiber, here designated as RF-Gly I. The sequenced region of RF-Gly I displays a high degree of homology with some regions of the von Willebrand factor and certain mucins; it also displays two motifs homologous with repeats present in proteins of the spondin family and other proteins. A core sequence of the RF-Gly I repeats suggests that this molecule displays protein-binding properties.  相似文献

Tanshinone I and its analogue dihydrotanshinone I are the major active components isolated from Salvia miltiorrhiza Bunge and Salvia Przewalskii Maxim. These compounds have been found to possess significant antibacterial, anti-dermatophytic, antioxidant, anti-inflammatory and anticancer activities. Fifteen phase I metabolites and two phase II metabolites of tanshinone I and dihydrotanshinone I in rat bile were elucidated and identified by a sensitive HPLC–ESI-MSn method. The molecular structures of the metabolites are presented on the basis of the characteristics of their precursor ions, product ions and chromatographic retention times. The results indicate that the phase I metabolites are biotransformed through four main pathways: dehydrogenation, hydroxylation, furan ring cleavage and oxidation metabolism. Phase II metabolites were mainly identified as the sulfated conjugates which showed a characteristic neutral loss of 80 Da. The biotransformed pathways of tanshinone I and dihydrotanshinone I were proposed on the basis of the investigation.  相似文献

Summary A thoroughly documented account of the ultrastructure of the meiotic spindle pole body (SPB) cycle in a rust (Basidiomycota, Uredinales) is presented for the first time. The three-dimensional structure of the SPB and spindle during meiosis in the hollyhock rust fungusPuccinia malvacearum is analyzed from serial sections of preselected stages. This paper covers prophase I to prometaphase I. At late prophase I, the nucleolus disperses and does not reappear until the end of meiosis. The SPB at late prophase I consists of two, 4-layered discs, 0.8–1.0 m in diameter, connected by a middle piece (MP). The SPB is associated with a differentiated region of the nuclear envelope and nucleoplasm. At late diplotene to diakinesis, each disc generates a half spindle as it inserts into an otherwise intact nuclear envelope. The MP connecting the interdigitating half spindles elongates and eventually splits transversely during subsequent spindle elongation. Each half MP, which is attached to a SPB disc, becomes inserted in a sheath-like extension of the nuclear envelope. The intranuclear late prometaphase I spindle always becomes oriented perpendicularly to the longitudinal axis and sagittal plane of the metabasidium. There are 200–290 spindle microtubules (MTs) at each SPB at late prometaphase. The nonkinetochore MTs form a coherent central spindle around which the kinetochore MTs and bivalents are spread. A metaphase plate is absent. The results are compared with SPB behavior and spindle structure in early meiosis of other basidiomycetes and ascomycetes.  相似文献

It has been shown that the competence-inducing factor of Bacillus subtilis 168I? exhibits lytic activity toward isolated cell walls and nuclease activity toward transforming DNA. It has been shown that the competence factor covalently bound to CNBr-Sepharose exhibits the same enzymatic properties. A mechanism for the transformation process is proposed which advances the mechanism previously proposed by this laboratory.  相似文献

The purpose of this study was to investigate the combined influence of three-level, three-factor variables on the formulation of dacarbazine (a water-soluble drug) loaded cubosomes. Box–Behnken design was used to obtain a second-order polynomial equation with interaction terms to predict response values. In this study, the selected and coded variables X1, X2, and X3 representing the amount of monoolein, polymer, and drug as the independent variables, respectively. Fifteen runs of experiments were conducted, and the particle size (Y1) and encapsulation efficiency (Y2) were evaluated as dependent variables. We performed multiple regression to establish a full-model second-order polynomial equation relating independent and dependent variables. A second-order polynomial regression model was constructed for Y1 and confirmed by performing checkpoint analysis. The optimization process and Pareto charts were obtained automatically, and they predicted the levels of independent coded variables X1, X2, and X3 (−1, 0.53485, and −1, respectively) and minimized Y1 while maximizing Y2. These corresponded to a cubosome formulation made from 100 mg of monoolein, 107 mg of polymer, and 2 mg with average diameter of 104.7 nm and an encapsulation efficiency of 6.9%. The Box–Behnken design proved to be a useful tool to optimize the particle size of these drug-loaded cubosomes. Photosystem I (PSI) is one of the two photosystems in photosynthesis, and performs a series of electron transfer reactions leading to the reduction of ferredoxin. In higher plants, PSI is surrounded by four light-harvesting complex I (LHCI) subunits, which harvest and transfer energy efficiently to the PSI core. The crystal structure of PSI-LHCI supercomplex has been analyzed up to 2.6 Å resolution, providing much information on the arrangement of proteins and cofactors in this complicated supercomplex. Here we have optimized crystallization conditions, and analyzed the crystal structure of PSI-LHCI at 2.4 Å resolution. Our structure showed some shift of the LHCI, especially the Lhca4 subunit, away from the PSI core, suggesting the indirect connection and inefficiency of energy transfer from this Lhca subunit to the PSI core. We identified five new lipids in the structure, most of them are located in the gap region between the Lhca subunits and the PSI core. These lipid molecules may play important roles in binding of the Lhca subunits to the core, as well as in the assembly of the supercomplex. The present results thus provide novel information for the elucidation of the mechanisms for the light-energy harvesting, transfer and assembly of this supercomplex.  相似文献

The presence of complex I, or NADH dehydrogenase, in Trypanosomatidae is debated. Several subunits of complex I have been identified by biochemical studies, but the overall composition of the complex has remained elusive. Here, the authors review the present literature related to this mitochondrial activity and carry out a bioinformatic analysis to allow the prediction of the composition of a putative trypanosomatid complex I. The complex comprises at least 19 subunits and has a minimum mass of 660 kDa. It is larger than the corresponding bacterial enzyme but smaller than the typical mitochondrial enzyme of eukaryotes. All subunits known to be involved in electron transport are present, but the complex does not seem to be involved in energy transduction because four membrane subunits, normally encoded by the mitochondrial genome and supposed to be involved in proton extrusion, are missing.  相似文献

(1) A mathematical investigation has been made of the progress of an epidemic in a homogeneous population. It has been assumed that complete immunity is conferred by a single attack, and that an individual is not infective at the moment at which he receives infection. With these reservations the problem has been investigated in its most general aspects, and the following conclusions have been arrived at. (2) In general a threshold density of population is found to exist, which depends upon the infectivity, recovery and death rates peculiar to the epidemic. No epidemic can occur if the population density is below this threshold value. (3) Small increases of the infectivity rate may lead to large epidemics; also, if the population density slightly exceeds its threshold value the effect of an epidemic will be to reduce the density as far below the threshold value as initially it was above it. (4) An epidemic, in general, comes to an end, before the susceptible population has been exhausted. (5) Similar results are indicated for the case in which transmission is through an intermediate host.  相似文献

NF1 (neurofibromatosis type?I) is a common genetic disease that affects one in 3500 individuals. The disease is completely penetrant but shows variable phenotypic expression in patients. NF1 is a large gene, and its pre-mRNA undergoes alternative splicing. The NF1 protein, neurofibromin, is involved in diverse signalling cascades. One of the best characterized functions of NF1 is its function as a Ras-GAP (GTPase-activating protein). NF1 exon 23a is an alternative exon that lies within the GAP-related domain of neurofibromin. This exon is predominantly included in most tissues, and it is skipped in CNS (central nervous system) neurons. The isoform in which exon 23a is skipped has 10?times higher Ras-GAP activity than the isoform in which exon 23a is included. Exon 23a inclusion is tightly regulated by at least three different families of RNA-binding proteins: CELF {CUG-BP (cytosine-uridine-guanine-binding protein) and ETR-3 [ELAV (embryonic lethal abnormal vision)-type RNA-binding protein]-like factor}, Hu and TIA-1 (T-cell intracellular antigen 1)/TIAR (T-cell intracellular antigen 1-related protein). The CELF and Hu proteins promote exon 23a skipping, while the TIA-1/TIAR proteins promote its inclusion. The widespread clinical variability that is observed among NF1 patients cannot be explained by NF1 mutations alone and it is believed that modifier genes may have a role in the variability. We suggest that the regulation of alternative splicing may act as a modifier to contribute to the variable expression in NF1 patients.  相似文献

A series of ureido and bis-ureido derivatives were prepared by reacting histamine with alkyl/aryl-isocyanates or di-isocyanates. The obtained derivatives were assayed as activators of the enzyme carbonic anhydrase (CA, EC 4.2.1.1), due to the fact that histamine itself has this biological activity. Although inhibition of CAs has pharmacological applications in the field of antiglaucoma, anticonvulsant, anticancer, and anti-infective agents, activation of these enzymes is not yet properly exploited pharmacologically for cognitive enhancement or Alzheimer’s disease treatment, conditions in which a diminished CA activity was reported. The ureido/bis-ureido histamine derivatives investigated here showed activating effects only against the cytosolic human (h) isoform hCA I, having no effect on the widespread, physiologically dominant isoform hCA II. This is the first report in which CA I-selective activators were identified. Such compounds may constitute interesting tools for better understanding the physiological/pharmacological effects connected to activation of this widespread CA isoform, whose physiological function is not fully understood.  相似文献

The aim of this study is to analyze the community structure, ecology and distribution of deep Weddell Sea sponge faunas. Analysis was performed on the basis of sponges sampled during ANDEEP I–III and SYSTCO I expeditions (2002–2008) by RV Polarstern. The material obtained comprises about 800 sponge specimens, representing 129 species, within these are 95 species of demosponges (including 15 new to science), 25 hexactinellid species (7 new) and 9 calcarean species (5 new). Sponges were sampled at 51 stations in depths between 500 and 5,500 m. At most stations, sponge densities were very low, and many species are represented by one or two specimens only. Community structure by Bray–Curtis similarity was analyzed as well as depth range and spatial distribution of the most common species. Zoogeographic affinities of sampled faunas are analyzed. Three associations of sponges are found in the deep Weddell Sea: (1) The Polymastia/Tentorium community, (including Rossella associations) distributed on the lower shelf and continental slope. (2) The Bathydorus community, distributed on the continental slope and upper abyssal. (3) The Caulophacus community, associated with Cladorhizidae, is characteristic for the abyssal plains. The associations follow each other successively both bathymetrically and geographically, from shallow to deep, from shelf and ridge structures into the open abyssal. A distinct faunistic boundary between shelf and deep sea is not present. In general, the sponge fauna of the deep Weddell Sea is regionally restricted and shows stronger affinities only to the sponge fauna of the subantarctic islands.  相似文献

Summary 2-glycoprotein I concentrations have been determined in human sera by the radial immunodiffusion technique. In 260 healthy individuals a bimodal distribution was observed. In 94% of sera concentrations ranged from 16 to 30 mg/100 ml with a mean value of 21.3±3.6 mg/100 ml. In 6% of sera lower concentrations were found: x=10.0±1.3 per 100 ml. The results of a study of 38 families with 79 children suggest that 2-glycoprotein I concentrations are controlled by a pair of autosomal co-dominant alleles called BgN and BgD. Individuals homozygous for the common allele BgN have levels between 16 and 30 mg/100 ml, heterozygous individuals have lower concentrations of 6 to 14 mg/100 ml. Individuals homozygous for the rate allele BgD have deficiency of 2-glycoprotein I. The results in one family, however, did not conform to this simple genetic hypothesis. The concentration of 2-glycoprotein I in serum is also influenced to some extent by non-genetic factors such as sex, age, and various pathologic conditions. During the biosynthesis of N-glycans in multicellular eukaryotes, glycans with the compositions Man(5)GlcNAc(2-3) are key intermediates. However, to reach this 'decision point', these N-glycans are first processed from Glc(3)Man(9)GlcNAc(2) through to Man(5)GlcNAc(2) by a number of glycosidases, whereby up to four α1-2-linked mannose residues are removed by class I mannosidases (glycohydrolase family 47). Whereas in the yeast Saccharomyces cerevisiae there are maximally three members of this protein family, in higher organisms there are multiple class I mannosidases residing in the endoplasmic reticulum and Golgi apparatus. The genome of the model nematode Caenorhabditis elegans encodes seven members of this protein family, whereby four are predicted to be classical processing mannosidases and three are related proteins with roles in quality control. In this study, cDNAs encoding the four predicted mannosidases were cloned and expressed in Pichia pastoris and the activity of these enzymes, designated MANS-1, MANS-2, MANS-3 and MANS-4, was verified. The first two can, dependent on the incubation time, remove three to four residues from Man(9)GlcNAc(2), whereas the action of the other two results in the appearance of the B isomer of Man(8)GlcNAc(2); together the complementary activities of these enzymes result in processing to Man(5)GlcNAc(2). With these data, another gap is closed in our understanding of the N-glycan biosynthesis pathway of the nematode worm.  相似文献

The dye eriochromblack T (erio T), added to an aqueous suspension of bovine retinal outer segments solubilized by digitonin, shows a light-induced absorption-increase at =645 nm. Erio T is shown to directly interact with micellar metarhodopsm I and metarhodopsin II.
