对金沙江干热河谷地区麻疯树(Jatropha curcas L.)种子中具有抗肿瘤活性的核糖体失活蛋白curcin含量及其生物学性状进行研究。结果表明,用酶联免疫吸附法测定麻疯树单颗种子的curcin含量为0.275~3.183 mg g-1,平均(1.369±0.055)mg g-1。Pearson相关性分析表明:单粒麻疯树种子的curcin含量与种子其他性状的相关性均未达显著水平,但与种子质量、种仁质量及含油量存在一定程度的负相关,而与出仁率和可溶性蛋白含量存在一定的正相关,且与可溶性蛋白含量的正相关系数(0.190)和与含油率的负相关系数(–0.177)较高。这表明可在金沙江干热河谷地区开展高curcin含量麻疯树种质资源收集和选育工作,以及培育高含油量、高curcin含量的双高品种,以提高麻疯树生物燃油产业的综合效益。  相似文献

核糖体失活蛋白是一类可以使核糖体28S rRNA的保守α茎环结构域脱嘌呤的蛋白质。麻疯树毒蛋白(curcin)前体基因编码麻疯树胚乳I型核糖体失活蛋白。从麻疯树基因组中克隆得到其5'侧翼区0.6 kb长的片段,将该片段插入pBI121载体置换其中的CaMV 35S启动子并在相应的转基因烟草中检测报告基因GUS的表达情况。经过GUS活性检测分析发现,该0.6 kb长的片段能够启动报告基因在种子中的表达,并且其在种子不同发育阶段的表达活性存在差异。同时,GUS组织化学染色定位分析表明,在双子叶植物中该启动子片段是胚乳特异性表达的,它从心形胚时期开始持续地在烟草的胚乳中发挥启动活性。  相似文献

油质蛋白基因对种子中油体的形成至关重要,该研究通过实时荧光定量PCR,对麻疯树的两个油质蛋白基因JcOle14.3和JcOle16.6在种子不同发育时期的表达模式进行了分析。结果表明:两个基因在种子发育初期(10~30 d)表达量逐渐升高,但表达水平均较低;40 d时表达量急剧增加并达到最高,而种子发育后期(50~55 d)两个基因表达水平均逐渐降低。由此可初步推测,JcOle14.3和JcOle16.6基因的表达量可能与种子油脂积累量存在正相关。该研究结果为麻疯树油体形成机理和油质蛋白的深入研究提供了理论基础。  相似文献

首次从麻疯树胚乳cDNA丈库中克隆得到一个RJNG型锌指蛋白基(GenBank登录号为JF920726),命名为JcRFP1。该cDNA长度为728bp,包含编码JcRFP1蛋白的完整开放阅读框(516bp)。脚,基因在麻疯树各器官中均检测到表达且表达量依次为:叶〉茎〉花〉果实〉种子〉根。将克隆到的JcRFP1基因的cDNA序列连接到表达载体pET32a(+)上,导入BL21(DE3)pLysS菌株,成功诱导表达相对分子质量为33.2kDa的可溶性融合蛋白。该融合蛋白免疫新西兰大白兔,得到效价为1:6500的抗血清。研究表明,JcRFP1蛋白具有体外泛素连接酶E3活性,在麻疯树体内可能参与油菜素甾醇信号转导途径。  相似文献

以海南近海岸陆地上正常生长、结实的3个不同基因型麻疯树(南油1、2和3号)种子为材料,分析比较3种种子及其种子油的理化特性.结果表明:南油1、2和3号麻疯树种子长、宽、厚度和表面积差异均不显著,但南油2号种子千粒重显著大于1、3号、不完善粒率显著小于1、3号,其种子饱满度好,结实性状优.南油3号种子种仁含油量显著高于1、2号,而1、2号间无显著差异.3种麻疯树种子油的过氧化值、折射率和皂化值无显著差异,而南油2号的酸值显著低于1、3号;三者碘值皆低于100,且南油2号的碘值显著小于1、3号.3种麻疯树油脂肪酸成分主要是油酸、棕榈酸、亚油酸、硬脂酸和十七碳酸,且皆以不饱和脂肪酸为主,南油2号的饱和脂肪酸含量相对较高.与南油1号相比, 南油2、3号的应用潜力相对大.  相似文献

麻疯树(Jatropha curcas)为大戟科麻疯树属植物,是一种多年生木本油料植物。原产美洲,广泛分布于热带亚热带地区。因其种子含油量高达 60%,可作生物燃料之用,麻疯树是目前正在被开发利用的重要能源植物之一。除此之外,麻疯树的种子含有多种活性成分,有着重要的农药和医药价值;其生长能耐干旱贫瘠,可用于荒山造林。因此,麻疯树是一种具有多种用途、重要经济价值和学术研究价值的植物。目前对其研究多停留在植物组织培养、植物化学、毒理学、种植业方面等。  相似文献

麻疯树(Jatropha curcas)种子含油率高,种子中的油类物质可作为生物柴油被开发和利用,是极具潜力的生物质能源树种之一。麻疯树雌雄异花,在自然条件下雄花数量通常远远大于雌花,这大大限制了种子和油的产量,因此开展麻疯树性别分化与花发育分子机理的研究具有重要意义。该研究选取10个麻疯树的MADS-BOX基因(JcAGL1,JcAGL6,JcAGL9,JcAGL11,JcAGL15,JcAGL61-3,JcAGL62-1,JcAGL62-6,JcAGL62-7,JcAGL80-2),提取麻疯树早期发育各个阶段的雌雄花总RNA,并反转录成cDNA,采用实时荧光定量方法,探索早期发育不同阶段的麻疯树雌雄花目的基因的表达情况。结果表明:目的基因在发育起始的雌雄花中的表达具有差异,比如JcAGL6和JcAGL15在雄花中表达量要高于雌花,而JcAGL1,JcAGL9和JcAGL11在雌花中的表达量要高于雄花,这说明花原基中目的基因表达会直接或间接决定性别分化的方向;在之后的发育过程中,目的基因的表达情况在雌雄花中有所不同:随着花的发育,目的基因在雌雄花中的表达量变化存在差别,这反应出麻疯树雌雄花发育中目的基因表达模式上的差异;另外,也能看出在此过程中各个目的基因又发挥着不同的功能。该研究结果为进一步探究麻疯树雌雄花发育相关基因的表达提供了理论依据,为了解麻疯树性别分化和花发育的分子机理奠定了基础。  相似文献

麻疯树(Jatropha curcas L. )为大戟科(Euphorbiaceae)麻疯树属(Jatropha L. )植物,果实含油量高,种仁含油量在60%左右,可用于加工提炼优质生物柴油,以替代日益紧缺的石油能源~([1-2]).近年来,由国家科学技术部和联合国开发计划署共同开展的绿色扶贫项目中,麻疯树已经在中国石漠化最为严重的贵州、四川及云南等省区试种成功.麻疯树全株有毒,其种子、树皮、叶、根和乳汁中含有多种化学成分,可作为医药和生物农药资源;其种子加工后残留的油饼中蛋白质含量较高,可作为优质肥料,经脱毒后也可作为饲料~([3]).  相似文献

小桐子(Jatropha curcas)种子含油率高,油脂组成适合于生产生物燃油,是制备航空生物燃油和生物柴油的理想原料。小桐子是一种雌雄同株植物,雌雄花着生于同一花序,其花序中雌花比例很低,可能是其种子产量低的主要原因之一。本文研究用不同浓度的赤霉素外源喷施处理小桐子花序芽对其花和种子发育的影响。结果表明:外源赤霉素处理能够诱导小桐子产生两性花,且处理浓度越高,两性花数量越多;随着两性花数量的增加,雌花数量相应越少,但雌花与两性花数量之和在各处理和对照之间差异不显著,这表明赤霉素诱导出的两性花可能来源于雌花。另外,高浓度(500~1500mg·L-1)的赤霉素处理会导致小桐子的种子不能正常发育, 表现为每个果实中的种子数量、大小、单粒种子重量、每个果序的种子重量以及种子的含油量都显著减少。这些结果有助于深入理解赤霉素在小桐子花器官形成及种子发育过程中所发挥的生理作用,为从分子水平上对小桐子进行遗传改良、提高其种子产量奠定基础。  相似文献

Plant terpenes constitute a large class of compounds and have numerous biological roles as either primary or secondary metabolites. Terpene synthases (TPSs) play key roles on catalyzing the formation of different terpenes; they are divided into seven subfamilies based on sequence relatedness (TPSa–h). TPS-a proteins catalyze the formation of sesquiterpenes and diterpenes in plants. Physic nut (Jatropha curcas L.) is an attractive biofuel tree, but its seeds contain diterpene derivatives, which make them inedible for animals. In this study, 59 putative TPS genes (JcTPS01 to JcTPS59) were identified in the physic nut genome, and 26 belong to the TPS-a subfamily. Eight among the 26 TPS-a genes showed expression in developing seeds of physic nut in the present study. After heterologous expression of these eight genes in Escherichia coli and in vitro enzyme assays, six were shown to have TPS activities. Two (JcTPS09 and JcTPS11) catalyzed the production of diterpene casbene, which was consistent with earlier findings. The other four (JcTPS02, 23, 55, and 56) catalyzed the production of sesquiterpenes. These results may facilitate the efforts for identifying TPS genes involving the physic nut terpene synthesize.  相似文献

Anthracnose caused by Colletotrichum gloeosporioides is an important physic nut (Jatropha curcas) disease causing damages on leaves, stems and fruits and consequently a decrease in seed quantity and quality. Physic nut is a native plant of Central America that has gained international attention due to its potential as biodiesel crop. However despite its highest relevance, studies concerning the response of different Jatropha genotypes to infection with this disease are very limited. The main objective of this study is the evaluation of susceptibility of 38 Jatropha curcas genotypes from a worldwide germplasm collection to Colletotrichum gloeosporioides. A simple method using wounded detached leaves inoculated with a 5-mm mycelia plug was used. Observations (lesion diameter and % of infection) were made after 3 and 7 days under controlled conditions (25 degrees C and 85% RH). Results showed significantly differences (P < or = 0.001) in both percentage of infection and lesion diameter among the tested accessions. Two genotypes from Brazil and Cape Verde, presented no lesions after 7 days of treatment, while the others presented between 67 and 100% of infection. Lesion diameter showed an increase with time of incubation and proved to be a useful tool for evaluation of plant susceptibility.  相似文献

Evaluation of damage to physic nut (Jatropha curcas) by true bugs   总被引:4,自引:0,他引:4  
The type of injury and amount of damage to the fruits and seeds of physic nut (Jatropha curcas L., Euphorbiaceae) caused by two species of fruit feeding true bugs, Pachycoris klugii Burmeister (Heteroptera: Scutelleridae) and Leptoglossus zonatus (Dallas) (Heteroptera: Coreidae), and the flower feeding true bug, Hypselonotus intermedius Distant (Heteroptera: Coreidae), were assessed using field cages. The parameters analysed were infructescence, fruit, seed and seed kernel weight, seed length, fruit and seed abortion, seed malformation and oil and protein content of the seed kernels. All three species reduced overall yield. The two members of the fruit feeding guild led to premature fruit abortion and malformed seeds. Reduction of fruit, seed and seed kernel weight as well as seed length could be expressed as linear regressions of adult population densities. The oil content of the seeds was slightly reduced by the bugs, but protein content remained unchanged. Damage increased with the developmental stage of the larvae. Whereas adult L. zonatus of both sexes produced more damage than the nymphs, female P. klugii caused less damage than nymphs, and male P. klugii no significant damage at all. H. intermedius is a pollinator of this entomogamous plant. At high densities, however, it reduced the number of fruits maturing.  相似文献

The leaf-footed bug Leptoglossus zonatus (Dallas) (Het., Coreidae) was reared in Nicaragua on a diet consisting of unripe physic nut ( Jatropha curcas L., Euphorbiaceae) fruit only. The suitability of this host plant is described by the mortalities in the developmental stages from egg to adult, which are presented in a life table, and the durations of the different stages. Pre-oviposition time, number of ovipositions and total number of eggs per female are given. Biometric measurements of all instars were made. Protein, oil and water content of fresh physic nut fruit were measured throughout a whole year. The results show that physic nut is a highly suitable host plant, which can maintain populations of this insect pest throughout the whole year as fruit are constantly present with only little fluctuation in their nutritional quality.  相似文献

Jatropha curcas, internationally and locally known, respectively, as physic nut and pinh?o manso, is a highly promising species for biodiesel production in Brazil and other countries in the tropics. It is rustic, grows in warm regions and is easily cultivated. These characteristics and high-quality oil yields from the seeds have made this plant a priority for biodiesel programs in Brazil. Consequently, this species merits genetic investigations aimed at improving yields. Some studies have detected genetic variability in accessions in Africa and Asia. We have made the first genetic evaluation of J. curcas collected from Brazil. Our objective was to quantify genetic diversity and to estimate genetic parameters for growth and production traits and seed oil content. We evaluated 75 J. curcas progenies collected from Brazil and three from Cambodia. The mean oil content in the seeds was 31%, ranging from 16 to 45%. No genetic correlation between growth traits and seed oil content was found. However, high coefficients of genetic variation were found for plant height, number of branches, height of branches, and stem diameter. The highest individual narrow-sense heritabilities were found for leaf length (0.35) and width (0.34), stem diameter (0.24) and height of branches (0.21). We used a clustering algorithm to genetically identify the closest and most distant progenies, to assist in the development of new cultivars. Geographical diversity did not necessarily represent the genetic diversity among the accessions collected. These results are important for the continuity of breeding programs, aimed at obtaining cultivars with high grain yield and high oil content in seeds.  相似文献

本文以小桐子的叶片、叶柄、茎段及下胚轴和子叶作为外植体,研究不同外植体类型对愈伤组织诱导的影响,结果表明叶柄的诱导率最高,其次为茎段的诱导率。同时以小桐子的下胚轴作为外植体,研究植物生长调节剂种类及浓度配比对愈伤组织诱导的影响,结果显示6-BA与2,4-D的组合更适宜小桐子愈伤组织的诱导,MS+6-BA0.5mg/L+2,4-D0.1mg/L为小桐子愈伤组织诱导的最佳培养基,其愈伤组织诱导率高达96.7%。本研究为小桐子愈伤组织的分化、植株再生及相关的遗传转化研究提供了参考。  相似文献
