Twenty eight microsatellite markers were used to analyze genetic divergence in tandem dinucleotide repeated DNA regions between two edaphic subpopulations of Triticum dicoccoides growing on the contrasting terra rossa and basalt soilsfrom a microsite at Tabigha, north of the Sea of Galilee, Israel. The terra rossa soil niche was drier and more stressful than the basalt throughout the growing season (November to May). Significant microsatellite divergence in allele distribution, repeat length, genetic diversity, and linkage disequilibria were found between emmer wheat from the two soil types over two short transects of 100 m each. Soil-specific and -unique alleles and linkage disequilibria were observed in the terra rossa and basalt subpopulations. A permutation test showed that the effects of random genetic drift were very low for the significant genetic diversity at microsatellite loci between the two subpopulations, suggesting that an adaptive molecular pattern derived by edaphic selection may act upon variation of the microsatellites. Summary Allozymic variation in proteins encoded by 47 loci was analyzed electrophoretically in 1983/4 and 1984/5 in 356 individual plants of wild emmer wheat, Triticum dicoccoides, from a microsite at Tabigha, north of the Sea of Galilee, Israel. Each year the test involved two 100-meter transects, each equally subdivided into basalt and terra rossa soil types, and comparisons were based on 16 common polymorphic loci. Significant genetic differentiation, genetic phase disequilibria, and genome organization according to soil type were found over very short distances. Our results suggest that allozyme polymorphisms in wild emmer wheat are partly adaptive, and that they differentiate at both single and multilocus structures primarily from environmental stress of such ecological factors as soil type, topography, and temporal changes, probably through aridity stress.  相似文献

Allozymic variation in proteins encoded by 22 loci was analyzed electrophoretically in 278 individual plants of wild barley,Hordeum spontaneum, the progenitor of cultivated barley, in four 100 meter transects, in Israel, each equally subdivided into basalt and terra rossa soil types. Significant differentiation according to soil was found in 9 alleles. Our results suggest that allozyme polymorphisms in wild barley are at least partly adaptive, and differentiate by edaphic natural selection rather than by stochastic processes, and/or neutrality of allozymic variants.  相似文献

Genomic diversity of anonymous regions across the genome, most probably including coding and noncoding amplified fragment length polymorphisms (AFLPs), was examined in 20 individuals of the blind mole-rat, Spalax galili, one of four allospecies of the Spalax ehrenbergi superspecies of blind subterranean mole-rats in Israel. We compared 10 individuals from two nearby populations in Upper Galilee, separated by only a few dozen to hundreds of metres and living in two sharply contrasting ecologies: white chalk and rendzina soil with Sarcopterium spinosum and Majorana syriaca versus black volcanic basalt soil with Carlina hispanica-Psorelea bitominosa and Alhagi graecorum plant formations. The microsite tested ranged in an area of less than 10000 m2. Out of 729 AFLP loci, 433 (59.4%) were polymorphic, with 211 soil unique alleles. Genetic polymorphism was significantly higher on the ecologically more xeric and stressful chalky rendzina soil than on the neighbouring mesic basalt soil. This is a remarkable pattern for a mammal that can disperse each generation between tens to hundreds of metres. These results cannot be explained by migration (which causes homogenization) or by chance (which will exclude sharp genomic soil divergence). Natural selection is the only evolutionary adaptive force that can cause genetic divergence across the genome matching the sharp microscale ecological contrast.  相似文献

We documented the floristic composition of pteridophytes (ferns and fern allies) and Melastomataceae in Yasuní National Park, Amazonian Ecuador. Our main questions were: (1) Are the density of individuals, species richness, and/or species diversity (measured with Shannon's H′) of the two plant groups related to edaphic differences? and (2) How many of the pteridophyte and Melastomataceae species are non–randomly distributed in relation to a soil base content gradient within terra firme (non–inundated forest). To answer these questions, we sampled 27 line transects of 500 × 5 m distributed in an area of ca 20 × 25 km. The study area included a permanent 50 ha plot established to monitor forest dynamics; thus, our results also provide information on landscape–scale floristic variability to which results from within the plot can be compared. A total of 45,608 individuals and 140 species of pteridophytes, and 4893 individuals and 89 species of the Melastomataceae, were counted in the transects. Both with pteridophytes and with Melastomataceae, a clear negative correlation was found between the amount of extractable bases in the soil and the number of plant individuals encountered in a transect. With Melastomataceae, species richness and species diversity also were negatively correlated with soil base content, but with pteridophytes they were not. More than 50 percent of the common species of both pteridophytes and Melastomataceae were nonrandomly distributed along the soil cation content gradient within terra firme. We conclude that while the species richness patterns observed in one plant group are not indicative of similar patterns in other plant groups, it seems likely that a substantial (but unknown) proportion of species belonging to other plant groups will be found to show distribution patterns that reflect edaphic preferences within terra firme forests.  相似文献

Genetic diversity at 28 microsatellite loci was studied in a natural population of Triticum dicoccoides at the Ammiad microsite, north of the Sea of Galilee, Israel. This microsite was subdivided into four major habitats, North, Valley, Ridge and Karst, and further subdivided into nine subhabitats. The units thus defined showed strong and highly significant differentiation in ecological factors; in particular with respect to cover, proximity and height of rocks, and surface soil moisture after early rains. The results showed that allele distributions at microsatellite loci were nonrandom and associated with habitats. Significant genetic differentiation and variation in repeat number were found among subpopulations in the four major habitats and nine subhabitats. Habitat-specific and -unique alleles and linkage disequilibria were observed in the Karst subpopulation. The subpopulations dwelling in drier habitats and subhabitats showed higher genetic diversities at microsatellite loci. These results suggest that natural selection, presumably through aridity stress, acts upon microsatellite divergence predominantly on noncoding sequences, thereby contributing to differences in fitness. Genetic diversity was studied in six subpopulations (a total of 60 individuals) of wild barley, Hordeum spontaneum , the progenitor of cultivated barley, sampled from six stations located along a transect of 300 m across the two opposing slopes of 'Evolution Canyon', a Mediterranean microsite at Lower Nahal Oren, Mt Carmel. The two opposing slopes are separated by between 100 and 400 m and designated SFS (South-Facing Slope) and NFS (North-Facing Slope) with each having three equidistant test stations. The SFS, which receives up to 300% more solar radiation, is drier, ecologically more heterogeneous, fluctuating, and more stressful than the NFS. Analysis of 12 RAPD primers, representing a total of 51 putative loci, revealed a significant inter- and intraslope variation in RAPD band polymorphism. A significantly higher proportion of polymorphic RAPD loci was found amongst the subpopulations on the SFS (mean P = 0.909) than on the NFS (mean P = 0.682), on the basis of the presence and absence of 22 strong bands. Polymorphism generally increased upwards from the bottom to the top of the SFS (0.636, 0.773, 0.955) and NFS (0.409, 0.500, 0.545), respectively. Gametic phase disequilibria estimates, D, revealed SFS and NFS unique predominant combinations which sharply differentiated the two slopes and indicated that there is differential interslope selection favouring slope-specific multilocus combinations of alleles, or blocks of genes over tens to hundreds of meters. This suggests that selection overrides migration. RAPD polymorphism appears to parallel allozyme diversity which is climatically adaptive and driven by natural selection in the same subpopulations at the microsite.  相似文献

 Genetic diversity was examined by RAPD-PCR analysis in 118 registered individuals of wild emmer wheat, Triticum dicoccoides, from a microsite at Yehudiyya, northeast of the Sea of Galilee, Israel. The test involved two climatic microniches in the open oak-park forest of Quercus ithaburensis: (1) sunny between trees and (2) shady under the trees’ canopies. Comparisons were based on 97 loci amplified by 20 oligonucleotide primers. Significant genetic differentiations were found at single-, two- and multilocus structures between the neighbouring shady and sunny niches. These DNA polymorphisms appear to be associated with microclimatic stresses. The pronounced niche-effect on the significance of linkage disequilibrium and niche-specific linkage disequilibrium may suggest that natural selection directed the two-locus associations. The structure of the multilocus associations also mainly results from natural selection, and not by chance from population subdivision, or founder effects. These findings are largely parallel to the previous allozymic results at single-locus and multilocus levels. Both the DNA and the allozymic results suggest that microclimatic selection appears to play an important role in DNA differentiation as well as in protein polymophism. Question: How is the diversity of woody species in a seasonally dry savanna related to plant available water (PAW)? Location: Savannas in central Brazil. Methods: Two‐dimensional soil resistivity profiles to 10‐m depth previously measured along three 10 m × 275 m replicate transects revealed differences in belowground water resources among and within transects: (1) driest/most heterogeneous; (2) wettest/least heterogeneous; and (3) PAW‐intermediate. All woody plants along these transects were identified to species, and height and basal circumference measured. Species diversity was evaluated for the whole transect (total diversity), 100‐m2 plots (alpha‐diversity) and dissimilarity among 100‐m2 plots within transects (beta‐diversity). Correlation analyses were conducted between PAW and vegetation variables at the 100‐m2 scale. Results: The driest/most heterogeneous transect had the lowest total species diversity, while the wettest/least heterogeneous transect showed the lowest beta‐diversity. Floristic variation was correlated with PAW in all transects. In the most heterogeneous transect, species density was positively correlated with PAW in the 0‐400 cm soil layer. Evenness and Simpson's diversity were negatively correlated with PAW in the 700‐1000 cm soil layer. Conclusion: Woody species diversity was related to PAW at a fine spatial scale. Abundant PAW in the top 4 m of soil may favour many species and increase species total diversity. Conversely, abundant PAW at depth may result in lower evenness and total diversity, probably because the few species adapted to obtaining deep soil water can become dominant. Environmental changes altering soil water availability and partitioning in soil layers could affect the diversity of woody plants in this savanna.  相似文献

Summary Color notations of dorsal pelage were analyzed in 451 adult subterranean mole rats, comprising 19 populations and 4 chromosomal species (2n=52, 54, 58 and 60) of the Spalax ehrenbergi superspecies in Israel. In addition, the color notations of soil samples from the collection sites were measured. In an attempt to evaluate the degree of correlation between pelage color of mole rats and the local soil color, each color was compared both macro- and microgeographically. The macrogeographic comparisons were among populations of the 4 chromosomal species, and the microgeographic comparisons were among pairs of geographically neighboring subpopulations which occur under similar climatic conditions but differ in soil type and color. The results indicate that 1. no differences in pelage color notation exist between the sexes; 2. pelage of mole rats is generally characterized by an over-all grayish color, but its shades vary regionally and locally in accordance with varying soil color; 3. a positive correlation was found between pelage and local soil colors in the macrogeographic analysis and this correlation was verified in a microgeographic comparison; 4. specimens from populations of the northern chromosomal species (2n=52, 54) mainly inhabit terra rossa and basalt soils, respectively, with reddish brown tones, and they tend to have a more reddish shade, whereas animals from the central species (2n=58), which inhabits mainly dark soils including alluvial clays, terra rossa and brown rendzina and the souther species 2n=60, which occurs mainly in light soil types such as pale rendzina, sandy loams and loess, tend to have a more yellowish shade. The general appearance of specimens from 2n=58 was dark gray while that of 2n=60 specimens was light gray; 5. mole rats living in xeric environments (particularly 2n=60, but also 2n=54) are lighter than those which live in mesic environments (2n=52, 58). The results support the conclusion that pelage color of strictly subterranean mole rats is subject to selection pressures of overground predation of disharmonious types with their background soil color. The possibility that the pelage color variation and patterns also contributes to better thermoregulation while mole rats are above ground is discussed.  相似文献

The Random Amplified Polymorphic DNA technique was used to assess the level of genetic diversity in Bryum argenteum from Ross Island and southern Victoria Land, Antarctica. Samples were collected from three separate transects, and from other geographically distinct populations within 150 km of Ross Island. Moss growth in two transects, sampled down small exposed meltstream channels at Cape Royds and Cape Chocolate, was very sparse with no other moss colonies found within 0.4 or 4 km, respectively. However, samples from these channels showed similar levels of genetic variation to those from a transect at Granite Harbour, where moss colonies were large, luxuriant and turf-like between boulders. In all transects, high levels of genetic diversity were apparent both within and between colonies, and some spatial relationships were observed down the length of the channels, with more extensive variation at the top than the bottom of two transects. Samples from other sites in the region showed varying but high levels of genetic diversity; overall, the majority showed some clustering according to site of collection, with short-distance dispersal of propagules by water and transmission between sites presumably by wind. The extensive genetic diversity observed appears mainly due to somatic mutation within colonies, with some contribution by immigration of propagules from elsewhere into established colonies. The RAPD technique (Random Amplified Polymorphic DNA) was used to assess the level of genetic diversity in Bryum pseudotriquetrum from Southern Victoria Land, Antarctica. Isolates were collected from two transects, and from several other geographically distinct populations within 150 km of Ross Island. Moss growth in one transect, sampled down a small exposed meltstream channel at Cape Chocolate, was very sparse with no other moss colonies found within several hundred meters. Isolates from this transect showed low levels of genetic variation, with many moss clumps appearing identical; these were probably dispersed by water along the channel. In another transect analysed from Granite Harbour, the moss colonies were large, luxuriant and protected by boulders. These isolates showed considerably higher levels of genetic variability than the Cape Chocolate samples, and both within- and between-clump variation was observed. Samples from other sites showed varying levels of genetic diversity. It appears from these results that the degree of protection from the harsh Antarctic environment can influence not only the type of moss growth, but also the level of genetic diversity. The diversity and composition of soil bacterial communities in three topographic sites (summit, foot slope, and lakeshore) from subtropical montane forest ecosystem in Taiwan were examined by using 16S rRNA gene clone library analysis. This locality is temperate, perhumid, and has low soil acidity (pH < 4), which is an uncommon ecosystem in a monsoonal part of Southeast Asia. A total of 481 clones were sequenced and placed into ten phylogenetic groups according to their similarities to type strains of described organisms. Toposequence of the transect was investigated from summit to foot slope and at the lakeshore. More than 86% of the clones were affiliated with members of the Proteobacteria, Acidobacteria, and Actinobacteria. Within the Proteobacteria, the β-Proteobacteria was the most abundant, then α-Proteobacteria and γ-Proteobacteria. Based on the Shannon diversity index (H) analysis, the bacterial community in the foot slope was the most diverse (H = 0.86) and that in summit was the least diverse (H = 0.68). The composition and diversity of soil bacterial communities in the three sites suggested no trend with topographic change. Less than 20% of the sequences were Acidobacteria-affiliated clones. The low proportion of Acidobacteria observed may be related to the high soil moisture and anaerobic microhabitats. Moreover, Shannon diversity indices revealed these bacterial communities to have lower diversity than that of other temperate (H = 0.90) and tropical forest (H = 0.82) ecosystems. The extreme acidity of soil pH and high soil moisture of this forest may explain composition and reduced the diversity of these soil bacterial communities.  相似文献

Valuation of Forests and Plant Species in Indigenous Territory and National Park Isiboro-Sécure, Bolivia. A quantitative ethnobotanical study was conducted in Indigenous Territory and National Park Isiboro-Sécure (TIPNIS), Bolivia, to assess the usefulness assigned by local Yuracaré and Trinitario ethnic groups to different terra firme and floodplain forests. Furthermore, we investigated which variables are good predictors for the use value attributed to plant species in the research area. Plants were collected during transect, walk-in-the-woods and homegarden sampling. Ethnobotanical and ethnoecological data of the inventoried plants were obtained from 12 Yuracaré and 14 Trinitario participants through semistructured interviews. On average, 84% of species in transects were claimed to be useful to people. The understorey (2.5 cm ≤ dbh < 10 cm) of the sampled forest types contained more useful species than the overstorey (dbh ≥ 10 cm), particularly for species with a medicinal and/or social use function. The local use value of plant species can be predicted, in part, from their botanical family, growth form, density, frequency, mean and maximum dbh, and ecological importance value. Our data confirm the hypothesis that density and frequency of plants in the landscape are both related to perceived plant accessibility. Accessibility of plants partly seems to guide their usefulness in TIPNIS. Indigenous assessment of accessibility and abundance of plants also covaried with their perceived usefulness and therefore has a potential for uncovering patterns in the perceived utility of plants.  相似文献

Abstract. Patterns of β‐diversity in a highly diverse tropical dry forest tree community are described; the contribution of environmental heterogeneity and distance to β‐diversity was assessed. Significant differences in elevation, insolation, slope and soil water holding capacity (p < 0.01), variables related to water availability, were found among 830 m × 100 m transects laid along contrasting slopes of a system of three parallel microbasins. A gradient in elevation and insolation was found within north‐facing transects, among 10 m × 10 m sites; south‐facing transects showed an elevation gradient while crest transects showed a gradient in water holding capacity. In total 119 species were registered, with 27 to 64 species per transect, and 4 to 16 species per site. A large β‐diversity was found among and within transects; two indices of β‐diversity consistently showed a higher β‐diversity within transects than among them. Among transects, 64% of the variance in species composition could be attributed to the environmental variables; an additional 22% to the spatial distribution of sites. Within transects, 42% of the deviance in β‐diversity values was explained by insolation, and 19% by distance. β‐diversity increased with distance and with difference in insolation among sites; north‐facing transects, those with most contrasting insolation conditions, had the steepest increase in β‐diversity with distance. Such increase was clearly associated with changes in species composition, not with changes in species richness.  相似文献

During the austral summer of 1995, distributions of phytoplankton biomass (as chlorophyll a), primary production, and nutrient concentrations along two north-south transects in the marginal ice zone of the northwestern Weddell Sea were examined as part of the 8th Korean Antarctic Research Program. An extensive phytoplankton bloom, ranging from 1.6 to 11.2 mg m−3 in surface chlorophyll a concentration, was encountered along the eastern transect and extended ca. 180 km north of the ice edge. The spatial extent of the bloom was closely related to the density field induced by the input of meltwater from the retreating sea ice. However, the extent (ca. 200 km) of the phytoplankton bloom along the western transect exceeded the meltwater-influenced zone (ca. 18 km). The extensive bloom along the western transect was more closely related to local hydrography than to the proximity of the ice edge and the resulting meltwater-induced stability of the upper water column. In addition, the marginal ice zone on the western transect was characterized by a deep, high phytoplankton biomass (up to 8 mg Chl a m−3) extending to 100-m depth, and the decreased nutrient concentration, which was probably caused by passive sinking from the upper euphotic zone and in situ growth. Despite the low bloom intensity relative to the marginal ice zone in both of the transects, mean primary productivity (2.6 g C m−2 day−1) in shelf waters corresponding to the northern side of the western transect was as high as in the marginal ice zone (2.1 g C m−2 day−1), and was 4.8 times greater than that in open waters, suggesting that shelf waters are as highly productive as the marginal ice zone. A comparison between the historical productivity data and our data also shows that the most productive regions in the Southern Ocean are shelf waters and the marginal ice zone, with emerging evidence of frontal regions as another major productive site. Dandelions (Taraxacum officinale Weber(sensu lato); Asteraceae) have been introduced to NorthandSouth America with human migration from Europe. While potential sourcepopulations have both sexually and obligate asexually (agamospermous)reproducing lineages, apparently only the latter have successfully colonizedtheAmericas. The consequences of obligate agamospermy on dandelion populationgenetic diversity in North America remain little explored. Here we use fourdifferent synthetic DNA probes that reveal genetic markers at multiplevariable-number-tandem-repeat (VNTR) loci to examine patterns of geneticvariation among plants collected along three different central North Americantransects with plants (21 to 22 individuals per transect) separated by: 1) >2 m and < 60 m (short transect); 2) > 5km and < 30 km (medium transect); and 3) > 30km and < 340 km (long transect). The mean numberofVNTR markers revealed per plant was 59.3. Co-clonal individuals (proportion ofbands shared exceeding 90%) were found in each transect, with the indexof clonality (the percent of co-clonal individuals detected in a transect)ranging from 34.12% for the short transect to 18.65% for the longtransect. Co-clonal individuals were separated by up to 200 km.With redundant examples of co-clonal individuals removed, mean similarity(proportion of band sharing) of distinct genotypes within transects was 0.426,and no statistical differences in level of similarity between transects, norindication of genetic differentiation between transects, was detected (meanFst between transect levels with all individuals included =0.05). These results indicate: 1) that dandelion genetic diversity ofcolonizinglineages in central North America is moderately high and does not reflectextreme bottleneck effects shown by some colonizing species; and 2) thatdandelion seed dispersal can be very effective in maintaining similar levels ofgenetic diversity at the different scales of sampling in this study, withcertain clones maintaining numerous, widespread individuals. Evidence that VNTRmutation is detectable within dandelion clonal lineages is presented,demonstrating that clonal families with lines increasinglydifferentiated from one another will continually evolve, and that Muller'srachet is, in all likelihood, turning for asexual lines.  相似文献

Spatial heterogeneity in the plant species composition of tropical forests is expected to influence animal species abundance and composition because vegetation constitutes the primary habitat feature for forest animals. Floristic variation is tied to variation in soils, so edaphic properties should ultimately influence animal species composition as well. The study of covariation in floristic and faunistic turnover has been hindered by the difficulty of completing coordinated surveys in hyperdiverse tropical communities, but this can be overcome with the use of a few plant taxa that function as surrogates for general floristic turnover. We used avian and plant transect surveys and soil sampling in a western Amazonian upland (terra firme) forest landscape to test whether spatial variation in bird community composition is associated with floristic turnover and corresponding edaphic gradients. Partial Mantel tests and Non‐metric Multidimensional Scaling showed floristic distinctiveness between two forest types closely associated with differences in soil cation concentrations, and differences in both floristic composition and cation concentrations were further linked to compositional differences in avian species, independent of geographic distances among sites. Ten percent of bird species included in Indicator Species Analyses showed significant associations with one of the two forest types. The upland forest types that we sampled, each corresponding to a different geological formation, are intermediate relative to edaphically extreme environments in the region. Models of avian diversification should take into account this environmental heterogeneity, as should conservation planning approaches that aim to represent faunal diversity. This study was conducted to test the effects of internal (genetic) and external factors on allelic diversity at 27 dinucleotide microsatellite (simple sequence repeat [SSR]) loci in three Israeli natural populations of Triticum dicoccoides from Ammiad, Tabigha, and Yehudiyya, north of the Sea of Galilee. The results demonstrated that SSR diversity is correlated with the interaction of ecological and genetic factors. Genetic factors, including genome (A vs. B), chromosome, motif, and locus, affected average repeat number (ARN), variance in repeat number (sigma), and number of alleles (NA) of SSRs, but the significance of some factors varied among populations. Genome effect on SSR variation may result from different motif types, particularly compound (or imperfect) versus perfect motifs, which may be related to different evolutionary histories of genomes A and B. Ecological factors significantly affected SSR variation. Soil-unique and soil-specific alleles were found in two edaphic groups dwelling on terra rossa and basalt soils across macro- and microgeographical scales. The largest contributions of genetic and ecological effects were found for diversity of ARN and NA, respectively. Multiple regression indicated that replication slippage and unequal crossing over could be important mutational mechanisms, but their significance varied among motifs. Edaphic stresses may affect the probability of replication errors and recombination intermediates and thus control diversity level and divergence of SSRs.
