通过体外模拟体内条件,选取主要消化酶胃蛋白酶、胰蛋白酶对猪血脱色液进行酶解,以高效酶木瓜蛋白酶和1394酶作为对照,结合酶解后多肽总含量以及<3 kDa的多肽得率进行酶解效果的综合评价,得到胃蛋白酶为最佳酶解酶。  相似文献

为了提高沙柳原料的丁醇发酵效果,考察沙柳原料经过蒸爆、超微粉碎+稀酸和超微粉碎+稀碱预处理后补料酶解的效果,优化了沙柳酶解液活性炭脱毒工艺参数,并对经过脱毒处理的酶解液进行了丁醇发酵研究,结果表明:预处理沙柳原料酶解底物质量浓度为200 g/m L时,3种预处理方法中蒸爆处理法水解效果最好,每克底物的滤纸酶酶加量15 U,酶解96 h后,酶解液总糖质量浓度达到57 g/L。活性炭脱毒处理的最优条件:p H 4.8,碳加量4%(质量分数)、温度70℃、1 h,该条件下的沙柳水解液脱色率达到97.4%、糖损失率3.1%。3种预处理沙柳原料的酶解液经活性炭脱毒后都可以被丁醇梭菌正常利用发酵产丁醇,发酵液总溶剂(ABE)质量浓度约为14 g/L。  相似文献

以AS1.398中性蛋白酶、胰蛋白酶、木瓜蛋白酶和菠萝蛋白酶对猪血蛋白进行水解,AS1.398中性蛋白酶对猪血蛋白的水解能力最强。采用活性炭对酶水解液进行脱色。脱色后的酶水解液中含有18种氨基酸,必需氨基酸占总氨基酸的39.18%。  相似文献

为了提高L-鸟氨酸发酵液的脱色效率,对其脱色工艺开展研究。首先通过树脂的选择性吸附确定L-鸟氨酸发酵液中色素的化学性质,随后结合颗粒活性炭的物理结构和零电荷点分析,优选出在接近中性条件下具有优良脱色性能的颗粒活性炭。在静态条件下考察pH、温度、时间和活性炭用量等因素对其脱色性能的影响。在此基础上,考察活性炭层析柱对L-鸟氨酸发酵液的动态脱色工艺和基于两步解吸法的活性炭再生工艺,单柱可动态脱色处理45倍床层体积(BV)的发酵液,脱色率达97%以上,脱色液呈无色透明状,L-鸟氨酸损失率低于1%,活性炭再生效果保持稳定。  相似文献

塔拉多糖是一种半乳甘露聚糖胶,对于我们具有非常重要的应用价值。本实验主要对塔拉提取物中的塔拉多糖进行脱色工艺化研究;在单因素实验的基础上,对活性炭颗粒质量、脱色时间、脱色温度以及脱色次数这四种因素进行正交优化实验,其最佳脱色实验参数为:活性炭颗粒0.6 g,脱色45 min,脱色温度45℃脱色次数3次,脱色率可以达到50.21%,同时多糖类保留率为90.39%。  相似文献

本研究旨在探讨活性炭对熟地多糖提取液脱色的工艺条件。在单因素试验基础上,采用正交试验,以多糖脱色率(%)和多糖剩余率(%)为指标,用紫外-可见分光光度法测定,确定了熟地多糖活性炭脱色的最优工艺的参数。结果发现:活性炭添加量对脱色效果的影响最大,其次为温度,再次为时间。最佳脱色条件为脱色温度60℃,活性碳添加量5%,吸附时间30 min,在此条件下,脱色率为98.20%,多糖剩余率为84.89%。从而表明活性炭对熟地多糖脱色工艺可行,简便,有效。  相似文献

用活性炭对菜籽降压肽双酶水解液进行脱色处理,采用单因素试验以及单因素试验基础上的正交试验法。考察活性炭用量、pH、温度、时间对水解液脱色率、肽损失率、活性及ACE抑制率的影响。正交试验优化结果表明,菜籽降压肽双酶水解液脱色最佳工艺为活性炭的用量为1%、脱色pH值为3.0、脱色温度为80℃、脱色时间为50 min,在此条件下脱色率为85.52%、ACE抑制率为57.83%、活性损失率为5.69%。  相似文献

将TiO2光催化剂负载于活性炭,然后将活性炭装填于冷电弧-光催化-吸附集成反应器中,用该反应器对豆粕氨基酸进行脱色。以豆粕氨基酸的脱色率和氨基酸保留率为指标,探讨处理电压、处理时间、溶液pH和活性炭填充量4个因素对脱色效果的影响。在单因素试验的基础上,利用二次回归正交旋转组合设计建立回归模型方程确定最佳工艺条件,并对方程进行回归分析和系数检验。结果表明:回归方程在#=0.01水平上高度显著。各因素对脱色效果的影响程度大小依次为:溶液pH>活性炭填充量>处理时间>处理电压。最佳脱色条件为处理电压20kV、处理时间11min、溶液pH 3.6和活性炭填充量63g,此条件下氨基酸溶液脱色率为94.8%,保留率为83.1%。  相似文献

为了探究鱼溶浆、酶解鱼溶浆和酶解鱼浆完全替代鱼粉后对黄颡鱼(Pelteobagrus fulvidraco)生长的影响, 在黄颡鱼实用型日粮配方模式下, 以28%的日本级蒸汽鱼粉为对照(FM), 按照FM组鱼粉蛋白质量的15%、30%和45%三个梯度, 分别添加鱼溶浆(SW15、SW30和SW45)、酶解鱼溶浆(SWH15、SWH30和SWH45)、酶解鱼浆(FPH15、FPH30和FPH45)完全替代鱼粉, 设计共10组等氮等能日粮, 每组设定3个重复, 在池塘网箱中养殖初始体重(18.68±0.10) g /尾黄颡鱼60d。结果显示: (1)以FM为对照, SW的3个试验组黄颡鱼的SGR降低9.89%—20.33%(P<0.05), FCR增加18.79%—44.85%(P<0.05); SWH试验组中SWH15和SWH45的SGR相对FM组分别降低8.79%和14.84%, FCR相对FM组增加16.97%和27.88%, 差异显著(P<0.05); FPH试验组中FPH15和FPH45的SGR相对FM组降低13.18%和15.38%, FCR相对FM增加30.30%和29.70%, 差异显著(P<0.05); (2)SWH30和FPH30组的SGR、FCR与FM组无显著差异(P>0.05); (3)相对于SW, 同等添加量SWH的SGR增加4.40%—9.39%, FCR降低9.81%—15.31%(P<0.05); (4)与FM组的FIFO值相比, SW30、SWH30和FPH30的FIFO值仅为0.67、0.61和0.60, 降低67.67%—70.15%(P<0.05)。结果表明: 在黄颡鱼日粮中, 8.5%添加量的酶解鱼溶浆(干物质)、8.2%添加量的酶解鱼浆(干物质)可以完全替代鱼粉(28%), 而相应的FIFO值分别比鱼粉组降低了69.95%和70.15%; 酶解鱼溶浆对黄颡鱼的生长效果优于鱼溶浆。日粮中过高或过低添加量的鱼溶浆、酶解鱼溶浆和酶解鱼浆都会导致黄颡鱼的生长速度和饲料效率下降。  相似文献

以八种常见吸附剂作为脱色剂,对蛋白质(猪血为原料)水解液进行了脱色实验并对脱色剂对水解产物——氨基酸的吸附影响进行了研究。结果表明:从脱色和氨基酸吸附两方面考虑,石墨粉是一种优于活性炭的脱色剂。本文还对各种吸附剂在不同pH值下氨基酸的吸附以及石墨粉的反复使用进行了讨论。  相似文献

An NAD(P)H-dependent H2O2 forming activity has been evidenced in thyroid tissue from patients with Grave's disease. Its biochemical properties were compared to those of the NADPH oxidase previously described in pig thyroid gland. Both were Ca2+-dependent and activated by inorganic phosphate anions in the same range of concentrations. Both are flavoproteins using FAD as cofactor, but the human enzyme was also able to utilize FMN. The apparent Km for NADPH of the human enzyme (100 microM) was 5-10 times higher than that of porcine enzyme. Vm was 3 to 10 times higher in pig (150 nmol x h(-1) x mg(-1)) than in man (14 to 45). Total content in human tissue was 7 to 9% of that in porcine tissue. An unidentified inhibitor has been detected in the 3000 g particulate fraction from most patients, which could account for this apparently low enzyme content. An NADH-dependent H2O2 production has also been observed in porcine and human thyroid tissues. This activity was only partly Ca2+-dependent (man, 50-70%; pig, 80-90%) and presented similar apparent Km values for NADH (man, 100 microM; pig, 200 microM). In pig thyrocytes, the expression of the Ca2+-dependent part of the NADH-oxidase activity was induced by TSH and down-regulated by TGFbeta, as was the NADPH oxidase activity. Furthermore, NADPH and NADH-dependent activities were not additive. We conclude that a single, inducible, NAD(P)H-oxidase can use NADPH or NADH as substrate to catalyse H2O2 formation, and that human and porcine NAD(P)H-oxidases are highly similar. Differences observed could be attributed to minor differences in enzyme structure and/or in membrane microenvironment. The NADH-dependent Ca2+-independent activity observed in human and porcine thyroid fractions could be attributed to a distinct and constitutive enzyme.  相似文献

Fourier transform infrared spectroscopy has been used to investigate the secondary structure of porcine and bovine pancreatic phospholipase A2 (PLA2) and the zymogen of porcine PLA2, prophospholipase A2 (proPLA2), in both H2O and D2O media. Detailed qualitative analysis was made of these proteins using second derivative and deconvolution techniques. Quantitative studies of the proteins in solution made using Factor Analysis gave average values of 54% alpha-helix, 15% beta-sheet and 23% beta-turns. These values agree well with the secondary structures deduced from previous studies of single crystals using X-ray techniques. No significant differences in secondary structure were observed for porcine pancreatic (pro)phospholipase A2 in the presence or absence of Ca2+ ions, or in the temperature range 10-45 degrees C. The binding of the non-degradable phospholipid analogue, n-alkylphosphocholine, in monomeric form produced no significant difference in the secondary structure of either enzyme. Conformational differences were, however, observed between the enzyme lyophilised in a solid film and in aqueous solution. The change is probably due to the formation of beta-sheet upon hydration, coupled with a loss of random structures. Conformational differences in both porcine and bovine pancreatic PLA2 were observed on binding to n-alkylphosphocholine micelles. This change may be due to a small increase in the alpha-helical structure and a decrease in the beta-sheet, and/or possibly beta-turn content. Similar conformational changes were observed for the interaction of porcine and bovine PLA2 with the substrate analogue inhibitor 1-heptanoyl-2-heptanoylamino-2-deoxy-sn-glycero-3-phospho glycol in micellar form.  相似文献

Phosphatidylcholine (PC) was synthesized from lyso-PC and long poly-unsaturated (n-3) fatty acids (PUFA) using immobilized phospholipase A2. The esterification was performed using the fatty acids as the main solvent and isooctane or ethanol (99.5%) at low concentrations (7–45%) as additional solvents. The temperature was kept constant at 45d`C and the water concentrations were carefully controlled.

The best yield of PC (22%) was found in the isooctane system at a low water content (22% of the dry immobilized enzyme). In the ethanol system, the yield of PC was only half. The best yields were attained when the concentrations of both isooctane and ethanol were below 7% and the reaction time was very long (9 days). Improved contact between the enzyme and the substrates would probably increase the reaction rate.  相似文献

Deoxyribonuclease I (DNase I) was purified 26500-fold in 39% yield from porcine pancreas to electrophoretic homogeneity using three-step column chromatography. The purified enzyme was inhibited by an antibody specific to the purified enzyme but not by G-actin. A 1303 bp cDNA encoding porcine DNase I was constructed from total RNA from porcine small intestine using a rapid amplification of cDNA ends method, followed by sequencing. Mature porcine DNase I protein was found to consist of 262 amino acids. Unlike all other mammalian DNase I enzymes that are inhibited by G-actin, porcine DNase I has H65 and S114 instead of Y65 and A114, which presumably results in the lack of inhibition. Porcine DNase I was more sensitive to low pH than rat or bovine enzymes. Compared with their primary structures, the amino acid at position 110 was N in porcine enzyme, but S in rat and bovine enzymes. A porcine mutant enzyme in which N was substituted by S alone at position 110 (N110S) became resistant to low pH to a similar extent as the rat and bovine enzymes.  相似文献

We investigated the substrate specificity of farnesyl diphosphate (FPP) synthase derived from Bacillus stearothermophilus and porcine liver by examining the reactivities of two cyclic substrate homologs, cyclohexylideneethyl diphosphate and cyclohexenylethyl diphosphate.Reaction of geranyl diphosphate with 2-cyclohexenylethyl diphosphate using bacterial or porcine liver FPP synthase produced (S)-geranylcyclohexylideneethyl diphosphate, with relative yields of 13.6% for the bacterial enzyme and 42.2% for the porcine liver enzyme. Reaction of cyclohexylideneethyl diphosphate with isopentenyl diphosphate produced 10-cyclohexyliden-3,7-dimethyldeca-2,6-dien-1-ol as a double condensation product, with relative yields of 23.1% (bacterial enzyme) and 3.0% (porcine liver enzyme). Reaction of cyclohexylideneethyl diphosphate with 2-cyclohexenylethyl diphosphate using bacterial enzyme produced (cyclohexylideneethyl)-cyclohexylideneethyl diphosphate (0.8% yield).  相似文献

The capacity of six basal media to support the growth of thermophilic campylobacters was tested. The most successful was Nutrient Broth No. 2 (Oxoid) solidified with New Zealand agar but it gave at best only a 9% recovery rate. Various blood products, iron compounds, detoxifying agents, reducing agents, growth stimulants and an antimetabolite were added to the selected basal medium and counts of inoculated organisms were compared with counts on basal medium containing 5% lysed horse blood. Of 22 supplements tried only blood, Fildes' peptic digest of blood, haematin, iron salts, charcoal, sodium metabisulphite and sodium pyruvate greatly improved the basal medium. The ability of these supplements used singly and in combinations to facilitate aertolerance of campylobacters was investigated. Two aspects of aerotolerance were tested; (a) the ability of the supplements to sustain the viability of campylobacters seeded onto culture plates left on the bench for up to 6 h before microaerobic incubation; and (b) the ability of the supplements to facilitate the growth of campylobacters at increasing oxygen tension (6, 10 and 17% oxygen). A combination of 0.4% charcoal, 0.025% ferrous sulphate and 0.025% sodium pyruvate was found to be as effective as blood in both tests.  相似文献

The aim of the study was to estimate occurrence of ochratoxin A (OA) in feeds and the metabolite residues in porcine blood serum in Poland. Samples were collected in the period from February to May, 1999, in the southern Wielkopolska region. Altogether 40 and 45 samples of feed and porcine blood serum, respectively, were analyzed for OA. Percentage of samples contaminated with OA, both in case of feeds and blood, collected in the winter season was considerably higher than that for the spring season. The percentages for feeds were as follows: 47.6 and 26.3 %, while for porcine serum: 66.7 and 50.0 %, respectively winter and spring. In 25 % of cases ochratoxin A was present in both types of investigated material (feed, blood), whereas in 27.5 % of samples this metabolite was detected in blood only, or in 7.5 % only in the feed. The presence of OA was found neither in the feed nor in the serum in 40 % of all cases. In subgroups (feed, blood) the concentration in the whole collective of positive samples were in the range 0.3–13.5 ng/g and 0.3–69.5 ng/ml, respectively, while median values were 2.3 ng/g and 6.0 ng/ml. Only one feed and three porcine serum samples, were found to be contaminated at concentration levels higher than 10 ng/g or 10 ng/ml.  相似文献

12-Lipoxygenases oxygenate arachidonic acid producing its 12S-hydroperoxy derivative and are well known as platelet and leukocyte enzymes. When a peroxidase-linked immunoassay of the enzyme according to the avidin-biotin method was applied to the cytosol fractions from various parts of porcine brain, a considerable amount of the enzyme was found in the anterior pituitary. The enzyme level (about 200 ng/mg cytosol protein) corresponded to about 6% of the enzyme content in porcine peripheral leukocytes. Posterior and intermediate lobes showed about one-tenth of the enzyme level of anterior pituitary. Other parts of porcine brain contained the 12-lipoxygenase in amounts below 7 ng/mg cytosol protein. The cytosol fraction (0.7 mg of protein) of anterior pituitary produced 12S-hydroxy-5,8,10,14-eicosatetraenoic acid from 25 microM arachidonic acid in about 34% conversion at 24 degrees C for 5 min, giving a specific enzyme activity about 3 nmol/min/mg protein. Furthermore, various octadecapolyenoic acids were oxygenated almost as fast as the arachidonate 12-oxygenation. When anterior pituitary was investigated immunohistochemically with anti-12-lipoxygenase antibody, most of the immunostained cells were certain parenchymal cells with granules, which were not blood cells. These biochemical and immunohistochemical results provide a good reason for considering that 12-lipoxygenase does play an important role in pituitary function.  相似文献
