根据已克隆的鲢鱼(Hypophthalmichthysmolitrix)微囊藻毒素去毒酶cDNA全序列设计特异引物,利用PCR方法获得鲢鱼微囊藻毒素去毒酶基因编码区,将该编码区与绿色荧光蛋白连接,分别构建融合表达载体pEGFP-N1-sGST和双顺反子表达载体pIRES2-EGFP-sGST。利用脂质体转染法将融合表达载体pEGFP-N1-sGST转染Hela细胞,60h后检测到绿色荧光基因表达;通过显微注射,将双顺反子表达载体pIRES2-EGFP-sGST注入斑马鱼(Daniorerio)受精卵,获得了转鲢鱼微囊藻毒素去毒酶基因斑马鱼,从而构建了微囊藻毒素去毒酶转基因模型。上述2种转基因模型的成功构建为进一步研究鲢鱼、鳙鱼(Aristichthysnobilis)、罗非鱼(Oreochromisnilotica)等淡水鱼类微囊藻毒素去毒酶基因调控元件、去毒分子机理及研发转基因鲢鱼、鳙鱼、罗非鱼等微囊藻毒素高效生物去毒器奠定了基础。  相似文献

地黄自毒物质提取及其生物指标测定   总被引:5,自引:0,他引:5  
连作障碍在药用植物地黄(Rehmannia glutinosa L.)栽培中尤其明显,同一地块上种植一茬地黄后,须经8-10a后方可再种。从怀地黄主产区河南省焦作地区采集种植1a地黄的茬后土壤,采用不同溶剂提取地黄土壤自毒物质并利用生物测试确定抑制率最强的土壤提取液作温室栽培实验以确定对植物生理指标的影响。利用高效液相色谱法(HPLC)对土壤提取液和根系分泌物进行比较,并利用高效液相色谱、傅立叶转换红外光谱(FTIR)和电喷雾离子阱质谱(ESI-MS)技术鉴定地黄连作自毒物质的化学成分。生物测试结果表明水和甲醇提取的地黄茬后土壤自毒物质对地黄生长具有最强的抑制作用;在培养器中加入浓度0.5g/mL的水和甲醇提取液对地黄胚根的抑制率分别达17%和26%,当浓度增至5.0g/mL时,抑制率增加到70%以上。盆栽结果显示甲醇提取的自毒物质导致地黄地上部叶绿素含量减少,根系活力下降,明显抑制了地黄体内超氧化物歧化酶、过氧化物歧化酶、过氧化物酶的活性,加剧了地黄膜质过氧化,以及引起地黄植体内生长素类激素含量下降。高效液相色谱法显示土壤提取液和根系分泌物具有相似图谱。电喷雾离子阱质谱分析共检测到6个特征物质:香草酸、D-甘露醇、二十六烷酸苯羟基乙酯、毛蕊花糖苷、β-谷甾醇和胡萝卜苷。  相似文献

将B型肉毒毒素在毒素粗提阶段用胰蛋白酶处理,再经浓缩、柱层析和结晶得到纯化的B型肉毒毒素复合物。结果表明:B型肉毒神经毒素经胰蛋白酶处理后单链裂解为双链,在非还原条件下SDS-PAGE显示神经毒素条带,在还原条件下SDS-PAGE只显示轻(L)、重(H)二链条带,而不显示神经毒素条带;纯化后毒素复合物的比活性提高了5.9倍,达到1.60×108LD50/mgPr;HPLC显示活性成分峰面积所占比例增加了9.83%。  相似文献

对肉毒中毒和肉毒毒蛋白的研究已有近百年历史,肉毒杆菌神经毒素(BoNT)作用机制研究的进展已使它成分为分析神经递质释放的胞吐分子机制和用于临床治疗的有用工具。近年发现,肉毒梭状芽胞杆菌C和D型菌株除分泌BoNT外,尚合成另外两类与BoNT结构和生物活必完全不同的蛋白质,肉毒二元毒素和肉毒细胞外酶,它们分别影响细胞骨架和细胞的生长发育,也成为分析相关生命过程的有效工具药,本文综述了这两类蛋白质的有关  相似文献

本文对梨胶锈菌性子期和锈子期菌丝吸器的形成方式、吸器及其与寄主细胞界面的超微结构进行了研究。观察结果表明:梨胶锈菌性子期和锈子期寄主胞间菌丝吸器的形成方式有两种:一种是由寄主胞间菌丝直接形成吸器;另一种是由寄主胞间菌丝先形成吸器母细胞,然后由吸器母细胞形成吸器。吸器在开始形成时只是一个乳头状的侵入楔,以后逐渐形成囊状、镰刀状、指状及其它不规则形状的吸器。多数吸器分化为颈和吸器主体两部分,在颈部及部分吸器主体外有一个由类似寄主细胞壁物质形成的领圈。吸器内部的超微结构与寄主胞间菌丝基本相同,但吸器壁比胞间菌丝或吸器母细胞的壁薄。吸器鞘的厚度随着吸器伸长膨大 而逐渐增厚。  相似文献

本研究在37℃条件下,以硝酸纤维素滤膜(NCM)为载体:3%酪蛋白为封闭剂、辣根过氧化物酶标记的A蛋白为酶标结合物(SPA-HRP)和以稀释100倍兔抗PRV-Ys的IgG为其工作浓度,成功地建立了检测PRV的间接Dot-ELISA。其检测灵敏度达到了较满意的ng水平。同时,本研究以PRV-Ys株系RNA作模板、oligo(dT)~(12-18)作引物、pUC19作cDNA克隆载体和以E.coli JM107作受体,采用缺口翻译(Nick Translation)方法成功地制备了pYs6 cDNA分子探针(插入片段约300bp)。其检测PRV-RNA的灵敏度达到了较理想的pg水平。  相似文献

Aims:  To elucidate and characterize the metabolic putrescine synthesis pathway from agmatine by Lactobacillus hilgardii X1B.
Methods and Results:  The putrescine formation from agmatine by resting cells (the normal physiological state in wine) of lactic acid bacteria isolated from wine has been determined for the first time. Agmatine deiminase and N -carbamoylputrescine hydrolase enzymes, determined by HPLC and LC-Ion Trap Mass Spectrometry, carried out the putrescine synthesis from agmatine. The influence of pH, temperature, organic acids, amino acids, sugars and ethanol on the putrescine formation in wine was determined.
Conclusions:  Resting cells of Lact. hilgardii X1B produce putrescine in wine. The putrescine production was carried out from agmatine through the agmatine deiminase system.
Significance and Impact of the Study:  These results have significance from two points of view, wine quality and toxicological and microbiological aspects, taking account that putrescine, which origin is still controversial, is quantitatively the main biogenic amine found in wine.  相似文献

Edwardsiella tarda was isolated from the intestinal tract of a 2-month-old pig. This is the first reported isolation of Edwardsiella tarda from swine in the United States. Swine have been reported as potential carriers of Edwardsiella tarda, but pathogenicity of this organism for swine has not been determined. Although the pig had access to several farm ponds, the exact source of infection was not determined.  相似文献

Two anodic isoperoxidases (A1 and A2) from tobacco tissue culture W-38 and two cathodic isoperoxidases (C3 and C4) from tobacco suspension culture WR-132 have been separated and characterized. Molecular weights for each of the isoperoxidases have been determined by two different methods. Only C4 contained a carbohydrate component. The substrate specificity and the pH optima for the four enzymes with each of five substrates were determined.  相似文献

An alpha-glucosidase maximally active at acid pH has been purified from human heart some 2,600-fold and its properties compared to a purified alpha-glucosidase from human liver. Molecular weight was evaluated using three different analytical procedures. The effect of various cations was determined. Thermal lability was evaluated using three different substrates. Affinity and hydrolysis velocity constants for maltose, glycogen and 4-methylumbelliferyl-alpha-D-glucose were determined for both preparations at optimal hydrogen ion concentration. Inhibition studies were carried out using the disaccharide turanose. From this study, we conclude there are no significant differences in molecular weight or kinetic properties between the cardiac and hepatic alpha-glucosidase enzymes.  相似文献

Spices and herbs are recognized as sources of natural antioxidants and thus play an important role in the chemoprevention of diseases resulting from lipid peroxidation. Our studies on spices and herbs have given us over a hundred compounds, known and new, having high antioxidant activity. From the Labiatae family, Rosmarinus officinalis, Thymus vulgaris, Origanum vulgare and O. majorana gave 26 active comopounds. Over 40 antioxidative compounds from Zingiber officinale, 26 compounds from Curcuma domestica = C. longa, C. xanthorrhiza and Z. cassumunar were determined, these belonging to the family Zingiberaceae. From the family Myrtaceae, 25 compounds from the berries of Pimenta dioica were determined and 3 carbazoles were isolated from Murraya koenigii. Structure-activity relationships of some of the isolated compounds were also discussed.  相似文献

Trehalose, a non-reducing disaccharide, was separated for the first time from Ganoderma applanatum fruit body. The structure of trehalose was determined by electrospray ionization mass spectrometric and NMR data. The content of trehalose was determined by improved anthrone-sulphuric acid colorimetric method and it was 0.48% of dried weight of G. applanatum fruit body. Mannitol was simultaneously obtained during separation of trehalose from G. applanatum.  相似文献

The percentage of tannins in leaves, bark, wood, and immature fruits of several species of Acacia and related mimosoid legumes from the southwestern U.S. and Mexico, along with a few from Costa Rica and Argentina, was determined by a modified hide powder procedure and by precipitation with casein. The relative percentages of hydrolyzable and condensed tannins were determined by the iodate and the vanillin-HCl methods, respectively. Gallotannins of selected samples were also determined by the rhodanine method. Although the amount of total tannins was similar for the first two methods, values for condensed tannins by the vanillin-HCl method were frequently two to four times greater than the total tannin values.  相似文献
