Barley (Hordeum vulgare L., cvs Golf, Mette, and Laevigatum)was grown under nitrogen limitation in solution culture untilnear maturity. Three different nitrogen addition regimes wereused: in the ‘HN’ culture the relative rate of nitrate-Naddition (RA) was 0·08 d–1 until day 48 and thendecreased stepwise to, finally, 0·005 d–1 duringgrain-filling; the ‘LN’ culture received 45% ofthe nitrogen added in HN; the ‘CN’ culture was maintainedat RA 0·0375 d–1 throughout. Kinetics of net nitrateuptake were measured during ontogeny at 30 to 150 mmol m–3external nitrate. Vmax (which is argued to reflect the maximuminflux rate in these plants) declined with age in both HN andLN cultures. A pronounced transient drop was observed just beforeanthesis, which correlated in time with a peak in root nitrateconcentration. Similar, but less pronounced, trends were observedin CN. The relative Vmax (unit nitrogen taken up per unit nitrogenin plants and day) in all three cultures declined from 1·3–2·3d–1 during vegetative growth to 0·1–0·7d–1 during generative growth. These values are in HN andLN cultures 15- to more than 100-fold in excess of the demandset by growth rates throughout ontogeny. Predicted balancingnitrate concentrations (defined as the nitrate concentrationrequired to support the observed rate of growth) were below6·0 mmol m–3 in HN and LN cultures before anthesisand then decreased during ontogeny. In CN cultures the balancingnitrate concentration increased during grain-filling. Apartfrom the transient decline during anthesis, most of the effectof ageing on relative Vmax can be explained in terms of reducedcontribution of roots to total biomass (R:T). The loss in uptakeper unit root weight is largely compensated for by the declinewith time in average tissue nitrogen concentrations. The quantitativerelationships between relative Vmax and R:T in ageing plantsare similar to those observed for vegetative plants culturedat different RAs. The data support the contention that the capacity for nitrateacquisition in N-limited plants is under general growth control, rather than controlled by specific regulation of the biochemicalpathway of nitrate assimilation. We examined changes in nitrogen content of the first leavesin relation to growth and nitrogen status of sunflower (Helianthusannuus L.) plants that were raised hydroponically at two irradiancelevels (high and low light, HL and LL) and at two nitrogen concentrations(high and low nitrogen, HN and LN). Initial increases in totaldry mass and total nitrogen of the whole plant were faster inHL-plants than in LL-plants irrespective of nitrogen supply, but in LN-plants the increase in total nitrogen was soon blunted. When plants grown under the same irradiance were compared, nitrogencontent of the first leaves (leaf N) decreased faster in LN-plantsthan in HN-plants, while for the plants grown at the same nitrogenconcentrations, it decreased faster in HL-plants than in LL-plants.Since these changes in leaf N were not explained solely by thechanges in plant dry mass or plant nitrogen, we introduced anindex, ‘nitrogen deficit (ND*)’, to quantify nitrogendeficit of the whole plant. ND* was expressed as ND*(t)=[Nmax–N(t)]xDM(t),where Nmax and N(t) were nitrogen contents in the young, expandingleaves that had just unfolded to expand, at the initial stagewith sufficient nitrogen and at time t, respectively, and DM(t),plant dry mass at t. The decrease in leaf N was expressed asa liner function of ND* irrespective of the growth conditions, which indicates validity of this index. Limitation of the useof ND*, and mechanisms by which leaves sense nitrogen demandare also discussed. (Received June 17, 1996; Accepted August 30, 1996)  相似文献

A microsomal flavonoid 3'-hydroxylase (F3'H) catalyzing themetabolism of naringenin to eriodictyol in Citrus sinensis (L.)Osbeck cv. ‘Hamlin’ cell suspension cultures wasshown to be a cytochrome P450 enzyme. This reaction requiredO2 and NADPH and was inhibited by CO, with partial reversalof CO-inhibition by light at 450 nm. Cytochrome P450 contentranged from 10–20 pmol (mg microsomal protein)–1.The F3'H reaction was shown to be linear in regard to proteinconcentration between 2.5 and 25 µg of microsomal protein. The optimum pH for the reaction was 7.4–7.6 and the temperatureoptimum was between 30 and 37°C. The apparent Km and Vmaxfor naringenin were 24 µM±3.2 and 81.4±7.9pmol eriodictyol min–1 (mg protein)–1, respectively. The microsomal F3'H was also capable of forming dihydroquercetinfrom dihydrokaempferol (40 pmol min–1 (mg protein)–1)and of quercetin from kaempferol (3.25 pmol min–1 (mgprotein–1). Cytochrome c and ketoconazole were the bestinhibitors of WH activity followed by piperonyl butoxide anda-naphthoflavone. Light was shown to be an inducer of the F3'Halmost doubling the specific activity and increasing the microsomalcytochrome P450 content by 30% over that of dark grown cells.F3'H activity was also confirmed in microsomal preparationsof young (new flush) leaves from ‘Hamlin’ treesand flavedo of ‘Hamlin’ oranges, ‘Marsh’grapefruit, and ‘Lisbon’ lemon. No activity wasobserved in older, hardened leaves and albedo of all the fruittested. Initiation of embryogenesis in the ‘Hamlin’cell suspension cultures by switching from a sucrose mediumto a glycerol-based medium resulted in the down-regulation ofF3'H. 1Mention of a trademark, warranty, proprietary product, or vendordoes not constitute a guarantee by the U.S. Department of Agricultureand does not imply its approval to the exclusion of other productsor vendors that may also be suitable.  相似文献

Techniques are described for the culture of developing barley(Hordeum vulgare L.) caryopses. Over a 7 d period in culturethe dry weights and the amounts of starch and protein increasedby at least twofold. Growth was sustained for at least 20 d.The effects of glutamine and cysteine on the amount and compositionof the hordein storage proteins were also studied. Glutaminestimulated total hordein accumulation but caused a disproportionateincrease in the amount of the S-poor ‘C’ hordeinswhen supplied at 100 mol m–3. Addition of cysteine at1·0 mol m–3 did not increase the amount of Srich‘B’ hordeins. The results suggest that althoughisolated caryopses are able to take up sucrose and glutamineand convert them to starch and protein there is some limitationin their ability to convert externally supplied cysteine intoproteins. Nutrient Dilution by Starch in CO2-enriched Chrysanthemum   总被引:1,自引:0,他引:1
Increasing growth irradiance and CO2 generally decreases foliarnutrient concentration on a dry weight basis and increases foliarstarch concentration. However, the extent to which starch concentrations‘dilute’ foliar nutrient concentrations when thelatter are expressed on a dry weight basis is not known. Todetermine the importance of differential starch accumulationin calculating nutrient concentrations on a dry weight basis, leaf nutrient and starch concentrations were measured in Chrysanthemum? morifolium ‘Fiesta’ (Ramat.) cuttings grown atthree irradiance levels and two CO2 levels for eight weeks inboth winter and spring. On a dry weight basis, foliar concentrationsof most nutrients were lower in both seasons as a result ofthe elevated CO2 and irradiance levels, and total dry weightswere higher. Per cent starch was greater at the high CO2 levelin both seasons but was only greater at higher irradiances inthe winter experiment. When starch was subtracted from the leafdry weights, the differences between CO2 and irradiance treatmentsdisappeared with respect to N, P, K, Ca, Mg, S, and B but notfor Fe, Mn, Zn, and Cu. The effects of exposure of up to 2 h with sulphur dioxide ona range of plant species was observed by measuring changes inthe rate of net photosynthesis under closely controlled environmentalconditions. Ryegrass, Lolium perenne ‘S23’ was thespecies most sensitive to SO2; significant inhibition was detectedat 200 nl l–1. Fumigations at 300 nl l–1 also inhibitedphotosynthesis in field bean (Vicia faba cv. ‘Three FoldWhite’ and ‘Blaze’) and in barley (Hordeumvulgare cv. ‘Sonja’). No effect was detected inwheat (Triticum aestivum cv. ‘Virtue’) at concentrationsup to 600 nl l–1 SO2, or in oil-seed rape (Brassica napuscv. ‘Rafal’) except at 800 nl l–1 SO2). Recoverycommenced immediately after the fumigation was terminated andwas complete within 2 h when inhibition had not exceeded 20%during the SO2 treatment. The effects of different NaCl concentrations on the growth andnitrogen fixation activity of white lupin (Lupinus albus [L.])was studied over a 6 d period. Plant growth parameters, photosynthesisand shoot respiration were unaffected by NaCl concentrationsup to 150 mol m–3. However, nitrogenase activity decreasedwith increased NaCl concentration up to 100 mol m–3, whilstthe O2 diffusion resistance increased with 100 mol m–3NaCl, but showed no further change when 150 mol m–3 NaClwas applied for 6 d. Increases in NaCl concentration decreasednodular starch content while increasing sucrose content, suggestingan osmotic regulation. These changes were associated with a77% decrease in sucrose synthase activity. The effect on theO2 diffusion resistance was paralleled by changes in glycoproteincontent of the nodules, as determined by immunogold localizationand ELISA. X-ray microanalysis studies of nodules showed that, following a 6 d exposure to 150 mol m–3 NaCl, Na+ ionswere largely excluded from the infected zone, whilst only lowlevels of Cl- ions penetrated into this region. Na+ entry intoroots and leaves was also at a low level. Leghaemoglobin contentdecreased with saline stress, as did superoxide dismutase; whichdecreased by 36% following exposure to 100 mol m–3 saltfor 6 d. These results are discussed in relation to the relativesalt tolerance of the Multolupa/ISLU-16 symbiosis. Differences in premature leaf abscission and in visible steminjury in genetic lines of poplar followed continuous fumigationswith air pollutant levels of SO2 (90–100 nl l–1)and O3 (70–80 nl l–1) either separately or together. The clones used were: Populus deltoides var. missiouriensisMarsh., P. nigra cv. ‘italicd’ L., and the hybridsP. nigra cv. ‘italica’ xP. deltoides (He-X/3) andP. nigra cv.‘italica’ x P. nigra cv. ‘Serres’(He-K/7). While most leaf abscission occurred within 20 d fromthe start of fumigation, stem lesions (intumescences), appearedonly after 72 d. Their anatomical characteristics include theformation of lysigenous aerenchyma in the lower parts of theintumescence, the sloughing of superficial cells from the injuredarea, and the development of crystalline formations on the surfaceof the lesions. P. deltoides exhibited the least morphologicalresponse to the gases. Ethylene released from fumigated leaves was determined at thesame gas concentration of SO2 (100 nl l–1), O3 (75 nll–1) and their mixture. Leaves of P. deltoides consistentlyshowed the lowest ethylene production after the gas treatments.P. ‘italica’ production was higher but was littlealtered by the treatments. The two hybrids He-X/3 and He-K/7showed the greatest increases in ethylene evolution with time. With He-K/7 exposed to the gas mixture the production of ethylenedecreased after the initial sharp rise during days 1–2,and reflected the considerable leaf damage observed after day3. The results suggest that sensitivity to air pollution, (as shownby leaf abscission and the formation of stem intumescences)can be correlated with the level of pollutant-induced ethyleneevolution from leaves. Initially high levels could induce abscission, whilst prolonged production could be responsible for intumescenceinitiation. The discussion proposes a series of events fromSO2 and/or O3 entry into the leaf and the physiological reasonsfor the clonal differences. Nitrate provision has been found to regulate the capacity forChara corallina cells to take up nitrate. When nitrate was suppliedto N sufficient cells maximum nitrate uptake was reached after8 h. Prolonged treatment of the cells in the absence of N alsoresulted in the apparent ability of these cells to take up nitrate.Chlorate was found to substitute partially for nitrate in the‘induction’ step. The effects on nitrate reductionwere separated from those on nitrate uptake by experiments usingtungstate. Tungstate pretreatment had no effect on NO3uptake ‘induced’ by N starvation, but inhibitedNO3 uptake associated with NO3 pretreatment. Chloridepretreatment similarly had no effect on NO3 uptake ‘induced’by N deprivation, but inhibited NO3 uptake followingNO3 pretreatment. The data suggest that there are atleast two mechanisms responsible for the ‘induction’of nitrate uptake by Chara cells, one associated with NO3reduction and ‘induced’ by CIO3 or NO3and one associated with N deprivation. The present study explores the potential contribution of theenergy requirements associated with nocturnal carbohydrate exportto (1) the fraction of dark respiration correlating with leafnitrogen concentration and (2) the dark respiration of maturesource leaves. To this end, we determined the nocturnal carbohydrate-exportrates from leaves with an optimal nitrogen supply, and the correlationbetween the nitrogen concentration and the dark respirationof leaves. The specific energy costs of carbohydrate exportfrom starch-storing source leaves were determined both experimentallyand theoretically. The present estimate of the specific energycost involved in carbohydrate export as obtained by linear regression(0.70 mol CO2 [mol sucrose]–1), agrees well with bothliterature data obtained by different methods (0.47 to 1.26mol CO2 [mol sucrose]–1) and the theoretically calculatedrange for starch-storing species (0.40 to 1.20 mol CO2 [molsucrose]–1). The conversion of starch in the chloroplastto sucrose in the cytosol is a major energy-requiring process.Maximally 42 to ‘107’% of the slope of the relationshipbetween respiration rate and organic nitrogen concentrationof primary bean leaves, may be ascribed to the energy costsassociated with nocturnal export of carbohydrates. Total energycosts associated with export were derived from the product ofthe specific costs of carbohydrate export and the export rates, either measured on full-grown (primary) leaves of potato andbean or derived from the literature. These export costs account,on average, for 29% of the dark respiration rate in starch-storingspecies. We conclude that nocturnal carbohydrate export is amajor energy-requiring process in starch-storing species Key words: Carbohydrate export, leaf dark respiration, nitrogen concentration, respiratory costs, specific energy cost  相似文献

Plants of four isolines of soyabean [Glycine max(L.) Merrill]‘Clark’, viz‘L71-920’ (maturity genecomplemente1e2e3 ), ‘L80-5914’ (E1e2e3), ‘Clark’(e1E2E3), and ‘L65-3366’ (E1E2E3), were grown inshort (12.25 h d - 1natural light) and long days (12.25 h d- 1natural light supplemented with 2.75 h d - 1low-irradianceartificial light) from first flowering to maturity in a polythenetunnel maintained at 30/24°C (day/night). Whereas therewere few differences among the isolines grown in short days,in long days the dominant alleles increased crop duration, biomassand seed yield substantially. Increases in biological and economicyield were not solely a consequence of longer crop duration:the dominant alleles also increased crop growth rate and radiationuse efficiency in long days (from 1.3 g MJ - 1total radiationine1e2e3 to 2.8 g MJ - 1inE1E2E3 ). Greater radiation use efficiencyresulted from a relatively longer leaf area duration, betterdistribution and orientation of a larger mass of leaves withinthe canopy, and smaller partitioning of assimilates to reproductivestructures. The work reveals the substantial effects of thethree lociE1 / e1, E2/ e2and E3/e3 on the response of plantgrowth, as well as development, to environment. Their relevanceto crop adaptation is discussed. Copyright 2000 Annals of BotanyCompany Glycine max(L.) Merrill, soyabean, maturity genes, flowering, phenology, growth, yield  相似文献

The effects of exposure to SO2 (55 m–3) on the growthand sulphur nutrition of perennial ryegrass (Lolium perenneL.) cv. S23 were examined using a system of specially constructedgrowth chambers. The plants were grown in soil with two ratesof added nitrogen, either with or without added sulphate. Theywere harvested twice during the exposure period of 85 d. The high rate of added nitrogen gave a four-fold increase inthe yield of plants given added sulphate but resulted in sulphurdeficiency, with reduced yield and number of tillers, when nosulphate was given. Exposure to SO2 alleviated the effects ofdeficiency but was without effect on the yield of plants givensulphate. With the low rate of added nitrogen, the yield ofplants and the number of tillers were unaffected by the additionof sulphate to the soil or of SO2 to the air. The only indicationof possible injury through exposure of these plants to SO2 wasa small increase in the proportion of dead leaves at the secondharvest. Analysis of the leaves for total S and sulphate-S gave valuesconsistent with sulphur deficiency and its alleviation throughexposure to SO2; the contents of 0?045% S and 42 parts 10–6sulphate-S in deficient plants were raised to 0?102% and 153parts 10–6 respectively. The organic-S content of theleaves was always increased through exposure to SO2. There wasno evidence that exposure to SO2 increased the ‘transpirationcoefficient’ for plants having an adequate supply of sulphurfrom the soil.  相似文献

A new bioassay which employs disbudded epicotyl cuttings takenfrom light grown Azukia seedlings (A. angularis) was devisedfor testing rooting promotion activity. By use of this method, the rooting promoting principle in leaves of Portulaca grandiflorawas isolated and identified with portulal which had been previouslyobtained. Portulal was reported as an inhibitor of rooting inetiolated Raphanus cuttings and has recently been determinedto be a bicyclic diterpene containing a perhydroazulene nucleus.Portulal promoted the adventitious root formation in severalkinds of plants, i.e. Azukia angularis, Vigna Catiang var. sinensis,Phaseolus Mungo and Raphanus sativus var. acanthiformis ‘Risodaikon’. The rooting process in Azukia and Raphanus cuttings seems toinclude at least two phases; a "preparatory phase" or a portulal-and gibberellin-sensitive phase and a "main phase" or an auxin-sensitiveand portulal-insensitive phase. 1Contribution No. 16 from the Botanical Gardens, Faculty ofScience, University of Tokyo, Tokyo, Japan. The levels of purine and pyrimidine nucleotides in suspensioncultures of Catharanthus roseus were determined 24 h after stationary-phasecells were transferred to fresh complete (‘+Pi’)or phosphate-deficient (‘–Pi’) Murashige-Skoogmedium. The levels of ATP, GTP, UTP and CTP were from approx.3 to 5-fold greater in the cells grown in ‘+Pi’medium than in the cells grown in ‘–Pi’ medium. The levels of almost all other nucleotides were slightly higherin the cells in ‘+Pi’ medium. The rates of de novoand salvage biosynthesis of purine and pyrimidine nucleotideswere estimated from the rates of incorporation of radioactivityfrom [14C]formate, [2–14C]glycine, NaH14CO3, [6–14C]orotate,[8–14C]adenine, [8–14C]adenosine, [2–14C]uraciland [2–14C]uridine. The results indicated that the activityof both the de novo and the salvage pathway was higher in thecells in ‘+Pi’ medium than in the cells in ‘–Pi’medium. The rate of degradation estimated from the rate of releaseof 14CO2 from labelled purines and pyrimidines indicated thatdegradation of uridine was significantly reduced in the cellsin ‘+Pi’ medium, but no significant difference wasfound in the degradation of adenine, adenosine and uracil. Thepossible role of Pi in the control of the biosynthesis of nucleotidesand in the degradation of uridine is discussed. Pascopyrum smithii (C3) andBouteloua gracilis (C4) are importantforage grasses native to the Colorado shortgrass steppe. Thisstudy investigated photosynthetic responses of these grassesto long-term CO2enrichment and temperature in relation to leafnonstructural carbohydrate (TNC) and [N]. Glasshouse-grown seedlingswere transferred to growth chambers and grown for 49 d at twoCO2concentrations (380 and 750 µmol mol-1) at 20 and 35°C, and two additional temperatures (25 and 30 °C) at750 µmol mol-1CO2. Leaf CO2exchange rate (CER) was measuredat a plant's respective growth temperature and at two CO2concentrationsof approx. 380 and 700 µmol mol-1. Long-term CO2enrichmentstimulated CER in both species, although the response was greaterin the C3,P. smithii . Doubling the [CO2] from 380 to 750 µmolmol-1stimulated CER ofP. smithii slightly more in plants grownand measured at 30 °C compared to plants grown at 20, 25or 35 °C. CO2-enriched plants sometimes exhibited lowerCER when compared to ambient-grown controls measured at thesame [CO2], indicating photosynthetic acclimation to CO2growthregime. InP. smithii , such reductions in CER were associatedwith increases in TNC and specific leaf mass, reductions inleaf [N] and, in one instance, a reduction in leaf conductancecompared to controls. InB. gracilis , photosynthetic acclimationwas observed more often, but significant changes in leaf metabolitelevels from growth at different [CO2] were generally less evident.Temperatures considered optimal for growth (C3: 20 °C; C4:35 °C) sometimes led to CO2-induced accumulations of TNCin both species, with starch accumulating in the leaves of bothspecies, and fructans accumulating only inP. smithii. Photosynthesisof both species is likely to be enhanced in future CO2-enrichedand warmer environments, although responses will sometimes beattenuated by acclimation. The effect of tetraploidy on leaf characteristics and net gasexchange was studied in diploid (2x ) and autotetraploid (4x) ‘Valencia’ sweet orange (Citrus sinensis (L.)Osb.) and ‘Femminello’ lemon (Citrus limon (L.)Burm. f.) leaves. Comparisons between ploidy levels were madeunder high irradiance (I) in a growth chamber or low total Iin a glasshouse. Tetraploids of both species had thicker leaves, larger mesophyll cell volume and lower light transmittance thandiploids regardless of growth I. Mesophyll surface area perunit leaf area of 2x leaves was 5–15% greater than on4x leaves. Leaf thickness and mesophyll cell volume were greaterin high I leaves than low I leaves. In high I, average leafarea was similar for 2x and 4x leaves, whereas in low I it was30% greater in 4x than in 2x leaves. Nitrogen and chlorophyllconcentration per cell increased with ploidy level in both growthconditions. The ratio of chlorophyll a:b was 25% greater in2x than in 4x leaves. When net CO2assimilation rate (ACO2) wasbased on leaf area, 4x orange leaves had 24–35% lowerACO2than their diploids. There were no significant differencesin ACO2between 2x and 4x orange or lemon leaves when expressedon a per cell basis. Overall, lower ACO2per unit leaf area oftetraploids was related to increase in leaf thickness, largermesophyll cell volume, the decrease in mesophyll area exposedto internal air spaces, and the lower ratio between cell surfaceto cell volume. Such changes probably increased the resistanceto CO2diffusion to the site of carboyxlation in the chloroplasts. The theory and practice of applying the thermodynamics of irreversibleprocesses to mass-flow theories is presented. Onsager coefficientswere measured on cut and uncut phloem and cut xylem strandsof Heracleum muntegazzimum. In 0.3 N sucrose + 1 mN KC1 theyare as follows. In phloem, LEE = 5 ? 10–4 mho cm–1,LpE = 9 ? 10–6 cm3 s–1 cm–2 volt–1 cm,and LPP = 0.16 cm3 s–1 cm–2 (J cm–3)–1cm. In uncut phloem strands LEE is about 1 ? 10–3 mhocm–1. In xylem in 2 x 10–3 N KCI, Lpp = 50 to 225,LPE = 2 ? 10–4, and LEE = 4 ? 10–3. The measurementsare tentative since the blockage of the sieve plates is an interferingfactor, but if they are valid they lead to the conclusion thatneither a pressure-flow nor an electro-kinetic mechanism envisaginga ‘long distance’ current pathway can be the majormotive ‘force’ for transport in mature phloem. Measurementsof biopotentials along conducting but laterally detached phloembundles of Heracleum suggest, nevertheless, that there may bea small electro-osmotic component of at least 0.1 mV cm–1endogenous in the phloem.  相似文献

The effects of the foliar application of phytocidal concentrationsof 2-methyl-4-chlorophenoxyacetic acid (MCPA) on change in totaldry weight, and in ‘available carbohydrate’ (starch,‘total’ and ‘reducing’ sugars), totalnitrogen, phosphorus, potassium, calcium, and magnesium of ‘tops’and roots of tomato plants have been followed over a periodof 14 days following spraying. There were two main treatments—‘nutrient’(nutrient supply to roots continued after spraying) and ‘water’(distilled water only supplied to roots after spraying) and‘water’ (distilled water only supplied to rootsafter spraying)—the sub-treatments consisting of ‘MCPA’versus ‘no-MCPA’ for each of the main treatments.Twelve different times of sampling were used. In analysing the present data, the quantity ‘residualdry weight’ (total dry weight less ‘available carbohydrate’),which was originally introduced by Mason and Maskell as a basisof reference for analyses of plant organs in short-period experimentsnot involving appreciable growth, has been used as an estimateof the permanent structure of plant growth. This new use ofthe ‘residual dry weight’ basis has brought outimportant features which were obscured when the data were leftin their primary form (as percentages of total dry weight oramounts per plant). Growth, as measured by increase in ‘residual dry weight’,was greatly inhibited by 2-methyl-4-chlorophenoxyacetic acidshortly after spraying, in both the presence and the absenceof nutrient. In the presence of 2-methyl-4-chlorophenoxyacetic acid, netassimilation rate (estimated as rate of increase in total dryweight per gram ‘residual dry weight’ of the ‘tops’)was greatly diminished while uptake of total nitrogen and ofP2O5 (estimated as increase in total nitrogen or of P2O5 ofthe whole plant per day per 1 g. ‘residual dry weight’of the roots) appeared to undergo a similar but much smallerdiminution. It seemed probable, however, that in the presenceof MCPA a larger proportion of the carbohydrate actually formedwas utilized for synthesis of aminoacids and protein. In the plant as a whole there was no evidence of actual depletionof ‘available carbohydrate’ as a result of MCPAtreatment, this fraction showing a steady increase in all treatmentsthroughout the experiment. The rate of increase was, however, much reduced by MCPA treatment. The ‘tops’ presentedmuch the same picture as the whole plant, but for the rootsthe situation was quite different. While the roots of the ‘no-MCPA’plants and also of the ‘MCPA-water’ plants showeda steady increase in available carbohydrate, those of the ‘MCPA-nutrient’plants rose only very slightly (from the initial value of 8mg. per plant to about 10 mg.) during the first 2 days, andthen in the next 2 days declined to a value (about 6 mg.) belowthe initial and remained at this low level for the rest of theexperiment. It is suggested that the phytocidal effect of 2-methyl-4-chlorophenoxyaceticacid in the presence of nutrient may be due to depletion ofthe ‘available carbohydrate’ supplies in the roots, which is shown to be brought about, in part, by reduced transportfrom the tops, and partly by the relatively greater utilizationof the carbohydrate present. These results offer an explanationfor the facts that plants showing vigorous growth are more easilykilled by MCPA and that perennial plants, particularly thosewith storage tissues in their roots, are more resistant. Further, they suggest the useful practical application that MCPA treatmentshould be given when the carbohydrate reserves of the rootsare at a minimum. For perennial plants, conditions might beexpected to be optimal for the application of MCPA in late spring,at a time when the first ‘flush’ of growth is slowingdown and before any appreciable new reserves of carbohydratehave been accumulated. It was also shown that 2-methyl-4-chlorophenoxyacetic acid preventedthe net synthesis of starch, but still permitted an appreciablenet formation of sucrose. 2-methyl-4-chlorophenoxyacetic acid appeared to have no effecton the uptake of potassium, calcium, or of magnesium. The lackof effect on potassium is contrasted with the previous observationby Rhodes, Templeman, and Thruston (1950) that sub-lethal concentrationsof MCPA, applied over a relatively long period to the rootsof tomato plants, specifically depressed the uptake of potassium.  相似文献
