外源脯氨酸对盐胁迫下甜瓜脯氨酸代谢的影响   总被引:2,自引:0,他引:2  
为探明外源脯氨酸对盐胁迫下甜瓜脯氨酸代谢的影响,以甜瓜品种‘雪美’为材料采用营养液栽培,对盐胁迫(100mmol·L-1 NaCl)、盐胁迫下添加外源脯氨酸(100mmol·L-1 NaCl+0.2mmol·L-1 Proline)以及对照3种处理后甜瓜幼苗叶片脯氨酸(Pro)含量、吡咯啉-5-羧酸合成酶(P5CS)、鸟氨酸转氨酶(OAT)和脯氨酸脱氢酶(ProDH)活性进行测定,并对OAT和ProDH基因进行克隆及半定量表达分析。结果显示:与对照相比较,盐胁迫条件下甜瓜幼苗叶片内Pro含量显著增加,P5CS活性增幅大于OAT活性,OAT基因表达量大部分时段内没有增加,ProDH活性下降,ProDH基因表达量减少;盐胁迫下添加外源脯氨酸进一步使幼苗叶片内Pro含量增加、OAT、ProDH活性提高、P5CS活性降低,并且使OAT基因表达量迅速增加、ProDH基因表达量先增加后回落。研究表明,盐胁迫条件下,甜瓜幼苗体内脯氨酸积累主要是通过增强脯氨酸的谷氨酸合成途径和抑制脯氨酸降解来实现;适量外源脯氨酸可以增强盐胁迫幼苗脯氨酸的鸟氨酸合成途径,但对谷氨酸合成途径有一定的抑制作用;通过调节合成和降解2种代谢途径进一步提高了脯氨酸含量,从而增强甜瓜幼苗耐盐胁迫能力。  相似文献

研究黑果枸杞在不同浓度盐胁迫下基因表达谱变化情况,为进一步研究黑果枸杞抗盐分子机制奠定研究基础。对0(CK)、50、250 mmol/L NaCl溶液胁迫的黑果枸杞组培苗的根和叶在胁迫时间为0、1、12 h时分别取样,采用转录组测序(RNASeq)技术进行测序分析。结果表明,转录组测序共产生222.49 Gb原始数据,拼接出Unigenes 86 037条,注释到7大功能数据库(GO、KEGG、KOG、NR、Pfam、Swiss-Prot和egg NOG)上的Unigenes总数为46 594个,占总Unigenes的54.76%,还有38 929个Unigenes在这些数据库中没有得到注释。通过GO分类和KEGG Pathway富集性分析,分别归于51个GO类别和211条代谢途径。差异表达基因分析显示,黑果枸杞叶片和根的上调基因和下调基因数随着NaCl浓度的增大和处理时间的延长均呈增加趋势,叶片中的上调基因数(7 514)小于下调基因数(9 032),根中的上调基因数(12 347)大于下调基因数(11 559)。在黑果枸杞盐胁迫下转录组中发现28 325个SSR位点,最多的为单核苷酸SSR,占70.47%。综合分析表明,黑果枸杞对盐胁迫的反应是一个多基因参与、多个生物过程协同调控的过程,基因表达量的变化可能是基因调控的主要方式。  相似文献

通过转录组测序技术对扎龙湿地野大麦幼苗在200 mmol/L NaCl胁迫2 d后的叶片进行分析。对测序结果进行De novo拼接后,将差异表达基因在GO、KEGG数据库中进行比对注释。结果表明:NaCl胁迫2 d后,对照组与处理组分别获得Unigene序列61038个和46754个,检测到差异表达基因25465个,差异基因GO功能注释到3个大类的55个功能组。差异表达基因被注释到135个Pathway上,直观地显示出NaCl胁迫下野大麦幼苗体内发生调节及改变的代谢过程和信号通路,其中主要涉及光合作用、脯氨酸代谢、叶绿素代谢等途径。通过对野大麦幼苗叶片转录组分析,发掘相关耐盐基因,有助于培育野大麦及其近缘作物新品种,并对揭示植物耐盐性分子机制及相关代谢途径具有重要意义。  相似文献

采用营养液砂培方法,研究了不同浓度NaCl胁迫(0、50、100、150、200和250mmol·L-1)对“定莜6号”燕麦幼苗生长、活性氧代谢和渗透调节物质含量的影响.结果表明:NaCl胁迫显著抑制燕麦幼苗的生长,抑制程度随NaCl浓度提高而增强,燕麦可耐受的最高NaCl浓度约为150 mmol·L-1;随着NaCl浓度的增加,叶片O2-产生速率、H2O2和丙二醛含量明显增加,超氧化物歧化酶、过氧化物酶和抗坏血酸过氧化物酶活性先升后降,过氧化氢酶活性迅速下降后逐渐升高,NaCl胁迫明显降低了谷胱甘肽含量,而抗坏血酸含量变化不大;NaCl胁迫显著提高了叶片脯氨酸含量,Na+含量随着NaCl浓度增加不断提高,K+含量和K+/Na+逐渐下降,质膜H+-ATP酶活性、总可溶性蛋白、热稳定蛋白和热不稳定蛋白含量先升后降,游离氨基酸含量先降后升,可溶性糖含量呈降-升-降趋势变化;盐胁迫下活性氧代谢失调和Na+、K+平衡破坏及积累有机溶质进行渗透调节时更多能量的消耗可能是燕麦生长受抑的重要因素.  相似文献

以小桐子幼苗为材料,设置盐胁迫(200mmol·L-1 NaCl)和外源水杨酸处理(0~2.0mmol·L-1 SA)水培试验,通过检测幼苗叶片脯氨酸含量、脯氨酸代谢关键酶活性及相关代谢酶基因的表达水平,研究了外源水杨酸对盐胁迫下小桐子幼苗脯氨酸代谢机理的影响。结果显示:(1)外源0.9mmol·L-1 SA处理可显著提高盐胁迫下小桐子幼苗的脯氨酸含量,上调脯氨酸合成关键酶Δ1-吡咯琳-5-羧酸合成酶(P5CS)和鸟氨酸转氨酶(OAT)活性,以及上调JcP5CS和JcOAT基因的表达水平。(2)SA也显著抑制了脯氨酸降解酶ProDH的活性及JcProDH基因的表达水平。(3)SA处理还显著提高了盐胁迫下小桐子幼苗的组织活力,降低了叶片电解质渗漏率和丙二醛(MDA)含量。研究发现,外源SA可通过活化脯氨酸合成的谷氨酸途径和鸟氨酸途径,以及抑制脯氨酸的降解途径来促进盐胁迫下小桐子幼苗脯氨酸的积累;外源SA处理也可提高小桐子幼苗的耐盐性,且这种提高可能与SA诱导脯氨酸的积累密切相关。  相似文献

为了探讨‘定莜6号’燕麦对盐胁迫的生理生化响应及H2O2的调节作用,采用水培方法,研究外源H2O2对盐胁迫下燕麦活性氧代谢、渗透溶质积累和Na+、K+平衡的影响。结果表明:小于100 mmol·L-1Na Cl未对‘定莜6号’幼苗的生长造成明显影响,150 mmol·L-1及以上浓度Na Cl使幼苗干重和叶片K+/Na+显著降低,O2-·产生速率、H2O2、丙二醛(MDA)、可溶性蛋白质和脯氨酸含量及过氧化氢酶(CAT)、质膜H+-ATP酶活性明显提高,但抗坏血酸(ASA)和谷胱甘肽(GSH)含量变化不大;外施5μmol·L-1H2O2可显著缓解150 mmol·L-1Na Cl胁迫对燕麦幼苗生长的抑制作用,使燕麦叶片超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶(APX)和CAT活性及H2O2、GSH含量明显提高,O2-·产生速率和MDA含量显著降低;5μmol·L-1H2O2还提高了150 mmol·L-1Na Cl胁迫下燕麦叶片可溶性蛋白质、可溶性糖、有机酸和脯氨酸含量及质膜H+-ATP酶活性和K+/Na+,降低了游离氨基酸含量;表明外源H2O2可调控燕麦幼苗活性氧代谢和渗透溶质积累,维持K+、Na+平衡,从而增强耐盐性。  相似文献

研究盐胁迫对千层金生理指标、精油含量以及成分的影响,以期为盐渍地种植千层金提供参考依据。以千层金扦插苗为试验材料,探究不同NaCl浓度(0、100、200、300、400、500 mmol/L)下千层金扦插苗光合作用、脯氨酸含量、丙二醛含量、POD酶活性、CAT酶活性以及精油含量和成分的影响。结果表明,盐胁迫环境下,千层金表现出较强的耐盐性以及抗氧化性。在100、200、300 mmol/L的NaCl处理下,其精油含量与对照组(0 mmol/L)相比分别增加22.34%、22.14%和57.83%。因此,适当的盐胁迫不会对千层金的生理指标产生影响,且能增加千层金叶片中的精油含量。  相似文献

以两种菊属野生植物'菊花脑'(Dendranthema nankingense)和'乙立寒菊'(D.indicum var.maruyamanum)幼苗为材料,通过Hogland营养液水培试验研究了120和180 mmol·L NaCl的渐进和非渐进胁迫处理对幼苗叶片受害程度及植株生物量、根系活力、叶片叶绿素含量、相对电导率和丙二醛含量的影响,以探讨它们对盐冲击的生理响应规律.结果显示:(1)在两种浓度盐胁迫下,两种菊属植物的整株鲜重、根系活力、叶片叶绿素含量大多显著下降,而同期的叶片丙二醛(MDA)含量、相对电导率显著增加;渐进盐胁迫下材料受害程度均轻于非渐进胁迫;盐胁迫浓度越大,各指标变化幅度越大,且两种胁迫方式下各指标的差异亦越大;(2)'菊花脑'对盐胁迫敏感,而'乙立寒菊'对盐胁迫的耐性较强;相同盐胁迫浓度下,非渐进胁迫与渐进胁迫间的差异在盐敏感材料上表现较明显.研究表明,两种菊属植物均存在盐冲击现象,并以耐盐性弱的材料表现更加突出,且高盐胁迫下植物所受的冲击伤害更明显.  相似文献

采用温室盆栽试验研究不同NaCl浓度(0、50 和85 mmol/L)持续胁迫接种摩西球囊霉和地表球囊霉 2种AM真菌对加工番茄耐盐性的影响。结果显示:(1)在0 mmol/L NaCl处理条件下,2种菌的番茄菌根化苗的根系活力、叶片中可溶性糖、可溶性蛋白、根系脯氨酸含量以及超氧化物歧化酶和过氧化物酶活性均高于非菌根植株,且丙二醛含量低于非菌根植株,但差异不显著。(2)在50、85 mmol/L NaCl浓度胁迫下,接种2种菌根真菌可显著提高番茄植株根系活力,促进叶片中可溶性糖、可溶性蛋白及根系脯氨酸含量的积累,显著提高叶片中与抗逆相关的超氧化物歧化酶和过氧化物酶的活性,减少丙二醛在根系中的积累;随着NaCl浓度的增加,效果更为明显。(3)RT-PCR分析显示,AM真菌和盐胁迫共同调控H+转运无机焦磷酸酶H+- PPase的表达,随NaCl浓度的增加,AVP1基因表达量下降,但菌根化番茄植株的AVP1基因表达量显著高于非菌根植株。研究表明,接种AM真菌后,菌根化植株可通过显著促进幼苗体内渗透调节物质积累和抗氧化酶活性的提高,有效降低体内膜脂过氧化水平,同时过量表达AVP1基因增加了番茄植株中离子向液泡膜的转运,从而缓解盐胁迫对植株的伤害,增强番茄幼苗对盐胁迫的耐性。  相似文献

为探究盐胁迫条件下宁夏枸杞苯丙烷代谢相关基因差异表达规律,以不同浓度NaCl(0,100,200,300 mmol/L)处理的水培宁夏枸杞幼苗为研究材料,利用高通量测序技术和qRT-PCR对盐胁迫下宁夏枸杞苯丙烷代谢相关基因差异表达进行分析,同时对该途径中关键酶活性及产物含量进行测定。结果表明,(1)宁夏枸杞在不同浓度NaCl处理下共有58个苯丙烷代谢相关基因差异表达,且随着盐胁迫程度的增加大部分基因表达水平上调或不变;(2)随着NaCl浓度的增加,宁夏枸杞叶片抗氧化酶SOD、POD、CAT的活性均下降,而酚类物质、类黄酮和木质素的含量在100 mmol/L NaCl处理下均显著积累。研究发现,宁夏枸杞可能通过调控苯丙烷代谢相关基因上调表达,增加酚类物质、类黄酮和木质素的合成,来清除过多活性氧和提升细胞壁强度以适应盐胁迫;宁夏枸杞可耐受的NaCl浓度在100~200 mmol/L之间。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献

Seven strains of aerobic carbon monoxide-oxidizing bacteria (carboxydebacteria) when growing on CO as sole source of carbon and energy had doubling times which ranged from 12–42 h. The activity profiles obtained after discontinuous sucrose density gradient centrifugation indicated that the CO-oxidizing enzymes are soluble and the hydrogenases are membrane-bound in all strains examined. The CO-oxidizing enzymes of Pseudomonas carboxydohydrogena, Pseudomonas carboxydoflava, Comamonas compransoris, and the so far unidentified strains OM2, OM3, and OM4 had a molecular weight of 230,000; that of Achromobacter carboxydus amounted to 170,000. The molecular weights of the CO-oxidizing and H2-oxidizing enzymes turned out to be identical. The cell sonicates were shown to catalyze the oxidation of both CO and H2 with methylene blue, thionine, phenazine methosulfate, toluylene blue, dichlorophenolindophenol, cytochrome c or ferricyanide as electron acceptors. Methyl viologen, benzyl viologen, FAD+, FMN+, and NAD(P)+ were not reduced. The spectrum of electron acceptors was identical for all strains tested. Neither free formate, hydrogen nor oxygen gas were involved in the CO-oxidation reaction. Methylene blue was reduced by CO at a 1:1 molar ratio. The results indicate that CO-oxidation by carboxydobacteria is catalyzed by identical or similar enzymes and that the reaction obeys the equation CO+H2OCO2+2H++2e- as previously shown for Pseudomonas carboxydovorans.Dedicated to Otto Kandler remembering almost three decades of enjoyable cooperation  相似文献

Since the introduction of the concepts of allostery about four decades ago, much advancement has been made in elucidating the structure-function correlation in allostery. However, there are still a number of issues that remain unresolved. In this review we used mammalian pyruvate kinase (PK) as a model system to understand the role of protein dynamics in modulating cooperativity. PK has a triosephosphate isomerase (TIM)(α/β)8 barrel structural motif. PK is an ideal system to address basic questions regarding regulatory mechanisms about this common (α/β)8 structural motif. The simplest model accounting for all of the solution thermodynamic and kinetic data on ligand-enzyme interactions involves two conformational states, inactive ET and active ER. These conformational states are represented by domain movements. Further studies provide the first evidence for a differential effect of ligand binding on the dynamics of the structural elements, not major secondary structural changes. These data are consistent with our model that allosteric regulation of PK is the consequence of perturbation of the distribution of an ensemble of states in which the inactive ET and active ER represent the two extreme end states. Sequence differences and ligands can modulate the distribution of states leading to alterations of functions. The future work includes: defining the network of functionally connected residues; elucidating the chemical principles governing the sequence differences which affect functions; and probing the nature of mutations on the stability of the secondary structural elements, which in turn modulate allostery.  相似文献
