Shortnose sturgeon Acipenser brevirostrum is federally listed as ‘‘an endangered species threatened with extinction’’ in the U.S. but its listing status is currently
under review. As part of this process, the U.S. National Marine Fisheries Service will determine if shortnose sturgeon are
divided into Distinct Population Segments (DPS) across its distribution. In this regard, we sought to determine if shortnose
sturgeon occur in genetically “discrete population segments,” and if so, the boundaries of each. We used mitochondrial DNA
(mtDNA) control region sequence analysis to assess the genetic discreteness of 14 of 19 river populations that were recommended
as DPS in the 1998 Final Recovery Plan for Shortnose Sturgeon. Nine of the 14 proposed DPS proved significantly discrete (P < 0.05 after Bonferoni correction) from both of their bracketing populations, the exceptions being those in the Penobscot
River, Chesapeake Bay, Cooper River, and Ogeechee River (our sample from the Cape Fear River was insufficient to statistically
analyze). Haplotype frequencies in the newly “rediscovered” Penobscot River collection were almost identical to those in the
proximal Kennebec River system. Genetic data in combination with tagging results suggest that shortnose sturgeon in the Penobscot
River are probably migrants from the Kennebec. Likewise, shortnose sturgeon found today within the Chesapeake Bay appear to
be migrants from the Delaware River. While haplotype frequencies in the remnant Santee River population in Lake Marion differed
significantly from those in nearby Winyah Bay, they did not differ significantly from those in the Cooper River. This suggests
that the Cooper River harbors descendants of the Santee River population that are unable to access their historical spawning
locales. The Ogeechee River collection was not genetically distinct from that in the nearby Savannah River, suggesting that
it may host descendants of hatchery-reared individuals of Savannah River ancestry. Our genetic results indicate that most,
but not all, rivers with shortnose sturgeon host genetically discrete populations, constituting important information in the
consideration of DPS designations. However, shortnose sturgeon migrations through coastal waters to proximal rivers and release
of hatchery-reared fish may confound results from genetic studies such as ours and lead to the possible misidentification

The paper reports the first reliable capture since the last 150 yr of two sturgeons in the basin of the River Pechora. Both specimens had the systematic traits of the Siberian sturgeon Acipenser baerii but differed significantly in external morphology. Data on biological traits and morphological parameters of the fish are presented. It is suggested that Pechora Basin should be included into the range of the Siberian sturgeon. Occurrences of the Siberian sturgeon in Pechora are discussed. 相似文献

The influence of body size on the consumption of live zooplankton (Daphnia spp.) by freshwater crayfish was examined using yabbies (Cherax destructor) ranging from 5 to 45 g. Food preference between live zooplankton and inert pellets was also assessed under experimental
conditions. In experimental tanks, yabbies of four size classes (<15, 15–24.9, 25–34.9 and 35–45 g) were presented with live
Daphnia. All yabbies were held in separate tanks with five animals per size class. In yabbies less than 15 g, the feeding mode on
zooplankton involved rapid searching and probing with the first two pairs of walking legs. Once a prey was located, the chelae
on the end of these walking legs would grasp the zooplankton and then rapidly move it towards the mouthparts. Yabbies larger
than 25 g tended to use their walking legs to push the Daphnia nearer to their third maxilliped which would then force or scoop the zooplankton towards the mouthparts. A short-term feeding
trial showed that there was no significant difference between size classes in regards to zooplankton consumption (P > 0.05). Capture efficiency of live Daphnia by yabbies less than 15 g was significantly lower (76%, P = 0.008) than the three larger size classes (93.6%). Yabbies less than 15 g consumed a significantly (P < 0.001) higher percentage (5.2%) of their body weight than the other size classes (1.1%, 0.8%, and 0.6%, respectively).
In the presence of both live zooplankton and a pellet diet, yabbies spent significantly (P = 0.005) more time feeding on zooplankton (85%) than on inert pellets (15%). This was the first study to quantify zooplankton
consumption by yabbies and the results provide insights into understanding the trophic role of freshwater crayfish in structuring
zooplankton communities and the husbandry management of crayfish farming.

Two repeated DNA sequences isolated from a partial genomic DNA library of Helianthus annuus, p HaS13 and p HaS211, were shown to represent portions of the int gene of a Ty3 /gypsy retroelement and of the RNase-Hgene of a Ty1 /copia retroelement, respectively. Southern blotting patterns obtained by hybridizing the two probes to BglII- or DraI-digested genomic DNA from different Helianthus species showed p HaS13 and p HaS211 were parts of dispersed repeats at least 8 and 7 kb in length, respectively, that were conserved in all species studied. Comparable hybridization patterns were obtained in all species with p HaS13. By contrast, the patterns obtained by hybridizing p HaS211 clearly differentiated annual species from perennials. The frequencies of p HaS13- and p HaS211-related sequences in different species were 4.3x10(4)-1.3x10(5) copies and 9.9x10(2)-8.1x10(3) copies per picogram of DNA, respectively. The frequency of p HaS13-related sequences varied widely within annual species, while no significant difference was observed among perennial species. Conversely, the frequency variation of p HaS211-related sequences was as large within annual species as within perennials. Sequences of both families were found to be dispersed along the length of all chromosomes in all species studied. However, Ty3 /gypsy-like sequences were localized preferentially at the centromeric regions, whereas Ty1/ copia-like sequences were less represented or absent around the centromeres and plentiful at the chromosome ends. These findings suggest that the two sequence families played a role in Helianthusgenome evolution and species divergence, evolved independently in the same genomic backgrounds and in annual or perennial species, and acquired different possible functions in the host genomes. 相似文献

Background
Gonadotropin releasing hormone (GnRH) is responsible for stimulation of gonadotropic hormone (GtH) in the hypothalamus-pituitary-gonadal axis (HPG). The regulatory mechanisms responsible for brain specificity make the promoter attractive for in silico analysis and reporter gene studies in zebrafish (Danio rerio).Results
We have characterized a zebrafish [Trp7, Leu8] or salmon (s) GnRH variant, gnrh 3. The gene includes a 1.6 Kb upstream regulatory region and displays the conserved structure of 4 exons and 3 introns, as seen in other species. An in silico defined enhancer at -976 in the zebrafish promoter, containing adjacent binding sites for Oct-1, CREB and Sp1, was predicted in 2 mammalian and 5 teleost GnRH promoters. Reporter gene studies confirmed the importance of this enhancer for cell specific expression in zebrafish. Interestingly the promoter of human GnRH-I, known as mammalian GnRH (mGnRH), was shown capable of driving cell specific reporter gene expression in transgenic zebrafish.Conclusions
The characterized zebrafish Gnrh3 decapeptide exhibits complete homology to the Atlantic salmon (Salmo salar) GnRH-III variant. In silico analysis of mammalian and teleost GnRH promoters revealed a conserved enhancer possessing binding sites for Oct-1, CREB and Sp1. Transgenic and transient reporter gene expression in zebrafish larvae, confirmed the importance of the in silico defined zebrafish enhancer at -976. The capability of the human GnRH-I promoter of directing cell specific reporter gene expression in zebrafish supports orthology between GnRH-I and GnRH-III.7.

Anthropogenic water management projects and facilities that alter the local and regional hydrology of riverine environments greatly influence the behavior, physiology, and survival of native fishes. To mitigate for losses of native fishes at these structures, many are outfitted with fish-exclusion screens to reduce entrainment. The effect of fish size and age on behavior near fish screens, however, is largely unknown. Therefore, we tested two size classes of juvenile green sturgeon (Acipenser medirostris; small, early juveniles: 9.2 ± 0.2 cm fork length [FL], 6.9 ± 0.3 g; intermediate juveniles: 18.8 ± 0.2 cm FL, 36.9 ± 0.8 g) near fish-exclusion screens in a laboratory swimming flume. Although size was a significant factor influencing the way in which fish contacted the screens (i.e., proportion of body contacts, p = 2.5 × 10?9), it did not significantly influence the number of times fish contacted screens or the amount of time fish spent near screens. We also compared the performance of these two size classes to that of older and larger sturgeon that were tested previously (29.6 ± 0.2 cm FL, 147.1 ± 3.1 g), and documented a clear difference in the behavior of the fish that resulted in disparities in how the large fish contacted screens relative to small- or intermediate-sized juveniles (p = 0.005, 5.4 × 10?4, respectively). Our results further our understanding of how ontogeny affects fish behavior near anthropogenic devices, and are informative for managers seeking to identify the most susceptible size and age class of juvenile green sturgeon to water-diversion structures to potentially develop size-specific conservation strategies. 相似文献

A genetic transformation system has been developed for callus cells of Crataegus
aronia using Agrobacterium
tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with
5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different
types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red
colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. were obtained 4 weeks after incubation of the co-cultivated explants onto maintenance medium supplemented with 50 mg l−1 hygromycin. Molecular analysis confirmed the integration of the transgenes in transformed callus. To our knowledge, this

The maT clade of transposons is a group of transposable elements intermediate in sequence and predicted protein structure to mariner and Tc transposons, with a distribution thus far limited to a few invertebrate species. We present evidence, based on searches of publicly available databases, that the nematode Caenorhabditis briggsae has several maT-like transposons, which we have designated as CbmaT elements, dispersed throughout its genome. We also describe two additional transposon sequences that probably share their evolutionary history with the CbmaT transposons. One resembles a fold back variant of a CbmaT element, with long (380-bp) inverted terminal repeats (ITRs) that show a high degree (71%) of identity to CbmaT1. The other, which shares only the 26-bp ITR sequences with one of the CbmaT variants, is present in eight nearly identical copies, but does not have a transposase gene and may therefore be cross mobilised by a CbmaT transposase. Using PCR-based mobility assays, we show that CbmaT1 transposons are capable of excising from the C. briggsae genome. CbmaT1 excised approximately 500 times less frequently than Tcb1 in the reference strain AF16, but both CbmaT1 and Tcb1 excised at extremely high frequencies in the HK105 strain. The HK105 strain also exhibited a high frequency of spontaneous induction of unc-22 mutants, suggesting that it may be a mutator strain of C. briggsae. 相似文献

We have investigated the floral ontogeny of Arillastrum, Allosyncarpia, Stockwellia and Eucalyptopsis (of the eucalypt group, Myrtaceae) using scanning electron microscopy and light microscopy. Several critical characters for establishing relationships between these genera and to the eucalypts have been determined. The absence of compound petaline primordia in Arillastrum, Allosyncarpia, Stockwellia and Eucalyptopsis excludes these taxa from the eucalypt clade. Post-anthesis circumscissile abscission of the hypanthium above the ovary in Stockwellia, Eucalyptopsis and Allosyncarpia is evidence that these three taxa form a monophyletic group; undifferentiated perianth parts and elongated fusiform buds are characters that unite Stockwellia and Eucalyptopsis as sister taxa. No floral characters clearly associate Arillastrum with either the eucalypt clade or the clade of Stockwellia, Eucalyptopsis and Allosyncarpia.We gratefully acknowledge Clyde Dunlop and Bob Harwood (Northern Territory Herbarium) for collecting specimens of Allosyncarpia, and Bruce Gray (Atherton) for collecting specimens of Stockwellia. Data on distribution of digestive enzymes along the intestinal tract of mature specimens of the beluga Huso huso and Russian sturgeon Acipenser gueldenstaedtii are presented. The absence of a distinct proximo-distal gradient of the enzyme activity can be explained by short length of the intestine of sturgeons. Considerable differences are found in the distribution pattern of enzymes along the intestinal tract of beluga and Russian sturgeon. Notocactus scopa cv. Soonjung was subjected to in planta Agrobacterium tumefaciens-mediated transformation with vacuum infiltration, pin-pricking, and a combination of the two methods. The pin-pricking combined with vacuum infiltration (20-30 cmHg for 15 min) resulted in a transformation efficiency of 67-100%, and the expression of the uidA and nptII genes was detected in transformed cactus. The established in planta transformation technique generated a transgenic cactus with higher transformation efficiency, shortened selection process, and stable gene expression via asexual reproduction. All of the results showed that the in planta transformation method utilized in the current study provided an efficient and time-saving procedure for the delivery of genes into the cactus genome, and that this technique can be applied to other asexually reproducing succulent plant species. 相似文献

The former is characterized by having lageniform caulocystidia with a slightly thick-walled broadened base and no cheilocystidia. Lao leaf monkeys (Trachypithecus (francoisi) laotum) are endemic to a small area of central and, marginally, north Lao. They are known from a few, mostly vague, historical records.
We here present a detailed examination of the distribution of this little-known taxon and discuss its conservation status.
Surveys since 1992 show its range to be centered upon the karst-dominated Phou Hin Poun National Protected Area (NPA), Nam
Sanam Provincial Protected Area, and the southern part of Nam Kading NPA. The known range encompasses <2000 km2, within which
occurrence is patchy, reflecting habitat availability. The taxonomic identity of leaf monkeys plausibly of this group reported
to the north of this area is not known. In the south of Phou Hin Poun NPA, village reports that the monkeys have black heads
are corroborated by the few sightings; their taxonomic relationship with typical Trachypithecus laotum is unknown. In the mid–late 1990s large populations remained and individuals were easily seen. There is no apparent large-scale
threat to their habitat. There has been no significant reassessment of status since the late 1990s, nor is there any active
conservation action in place. Although the monkeys are to a significant extent protected by the arduous terrain, this cannot
be relied upon indefinitely: Trade-directed hunting, although apparently limited in the 1990s, is a potential threat that
could cause rapid population declines. Local traditions offer significant starting points for conserving these monkeys. 相似文献

A newly isolated Bacillus megaterium with epoxide hydrolase activity resolved racemic glycidyl (o, m, p)-methylphenyl ethers to give enantiopure epoxides in 84–99% enantiomeric excess and with 21–73 enantiomeric ratios. The (S)-enantiomer was obtained from rac-glycidyl (o or m)-methylphenyl ether while the (R)-epoxides was obtained from glycidyl p-methylphenyl ether.
