目的用不同方法观察家蝇3龄幼虫血细胞的形态,并对血细胞进行分类和免疫功能研究,为昆虫血细胞形态、分类及免疫研究提供实验依据。方法 (1)应用姬氏染色结合相差显微镜及荧光染色方法观察家蝇3龄幼虫血细胞形态,并对家蝇3龄幼虫血细胞进行分类;(2)观察家蝇3龄幼虫感染大肠杆菌后不同时间血细胞总数(THC)、各类血细胞数量(DHC)及形态的变化;(3)应用倒置显微镜观察家蝇3龄幼虫离体血细胞感染大肠杆菌后的形态变化;(4)采用酶细胞化学技术测定感染前后家蝇3龄幼虫血细胞中ACP、POD活性的变化。结果 (1)家蝇3龄幼虫血细胞可分为原血胞、浆血胞、粒血胞、珠血胞、类绛血胞5类,其中浆血胞又分为大核浆血胞和小核浆血胞两种;(2)感染后各时间组血细胞总数、浆血胞和粒血胞数量均显著升高,且浆血胞和粒血胞聚集成堆,出现细胞变形、空泡等形态变化;感染后16h、24h组的珠血胞数显著升高;原血胞和类绛血胞数量和形态无明显变化;(3)家蝇幼虫离体血细胞感染大肠杆菌后粒血胞周围见大量细菌聚集,浆血胞、粒血胞聚集成团将细菌包裹形成包囊,未见原血胞、珠血胞、类绛血胞形态的变化;(4)感染后浆血胞和粒血胞中ACP、POD的活性增强,感染前后原血胞、珠血胞、类绛血胞中均未见ACP、POD的阳性反应物。结论通过3种方法能很好地将家蝇3龄幼虫血细胞分为5类,其中浆血胞和粒血胞是家蝇幼虫参与免疫反应的主要细胞类型,珠血胞不参与感染后的早期细胞免疫反应。  相似文献

血细胞是昆虫免疫防御、代谢和分泌等生理活动的重要参与者。本文通过Gimesa-Wright试剂盒染色法研究了意大利蝗成虫血细胞类型。结果表明,意大利蝗存在原血胞、浆血胞、粒血胞、脂血胞、囊血胞、巨核细胞6种细胞。粒血胞、脂血胞和浆血胞是意大利蝗主要的血细胞类型,分别占细胞总数的42.04%、29.60%和23.32%。研究结果为深入研究意大利蝗血细胞的功能及其对意大利蝗适应环境的机制提供了基础数据。  相似文献

【目的】蝗虫血细胞的研究以及不同血细胞类型间的演化关系,是蝗虫细胞免疫学的依据,也是防蝗治蝗的重要依据。【方法】本研究通过Wright-Giemsa对黑翅痂蝗Bryodema nigroptera Zheng血细胞进行了混合染色,并使用光学显微镜进行了观察。【结果】发现黑翅痂蝗血细胞包括原血胞、浆血胞、粒血胞、类绛色血细胞、囊血胞、脂血胞、蠕形细胞和巨核血细胞8种类型。【结论】在成虫体内,原血胞占细胞总数的1%,浆血胞占32.2%,粒血胞占57.3%,类绛色血细胞占1%,囊血胞占6%,脂血胞占1%,蠕形细胞占1%,巨核细胞占细胞总数的0.5%,粒血胞数量最多,巨核细胞最少。  相似文献

本研究采用Wright-Giemsa染色方法,利用光学显微镜观察阿尔泰蝠蛾Hepialus altaicola Wang幼虫血细胞.根据鳞翅目昆虫血细胞形态特征对阿尔泰蝠蛾幼虫血细胞进行鉴定,计数并计算各类血细胞所占的比例.发现阿尔泰蝠蛾幼虫血淋巴中存在原血胞、浆血胞、粒血胞、类绛色细胞、珠血胞和囊血胞6种细胞类型,所占比例分别为3.7%,29.9%,55.4%,2.3%,5.8%和2.9%.阿尔泰蝠蛾幼虫血细胞组成及其形态与其他鳞翅目昆虫基本一致,但是与其它种属相比,同种类型的血细胞大小和占比略有不同.粒血胞和浆血胞是阿尔泰蝠蛾幼虫血淋巴中的主要类型,其中粒血胞占比最高.该研究为深入研究阿尔泰蝠蛾血细胞形态和功能提供理论依据和基础数据.  相似文献

为研究蚧虫血淋巴中血细胞的种类及其结构特征,采用相差显微镜、荧光显微镜和扫描电镜与透射电镜技术,观察了草履蚧雌成虫血细胞的显微形态与超微结构特征.结果显示,在荧光显微镜和扫描电镜下观察识别出草履蚧血淋巴中的5种血细胞,即:原血胞、浆血胞、粒血胞、囊血胞和类绛色血细胞.在透射电镜下可见,原血胞的细胞核明显,表现出高的细胞质密度;浆血胞最典型的特征是细胞质中有大量的囊泡;粒血胞的细胞质中有发达的粗面内质网和许多玫瑰形的细颗粒.类绛色血细胞最典型的特征是有许多结晶,并在细胞质的空泡区内分布的更多.囊血胞透明性强,具有围核空间,膨大成潴泡状.  相似文献

利用相差显微镜,并辅以涂片染色、化学反应方法,将粘虫各虫态虫龄的血细胞分为8个类型。他们是:原血胞、浆血胞、粒血胞、珠血胞、类绛血胞、囊血胞、梭血胞及脂血胞。并统计出各类血细胞在粘虫一生中的数量变化。本文认为足血胞与粒血胞不同之处在于前者的伸延部分含有细胞质。符合这一标准的足血胞在此次工作中看到的不多,因此对粘虫足血胞的存在问题暂作保留。 苏芸金杆菌处理后,粘虫血细胞产生了明显的病态变化,随着病症的进展,非正常血胞自2%上升为30%,50%,83%左右。直到死亡,血淋巴内未发现杆菌出现。  相似文献

利用光学显微镜和显微数码拍照系统,对红褐斑腿蝗Catantops pinguis(Stal)血细胞的形态进行观察和分类。结果在红褐斑腿蝗血淋巴中观察到5种血细胞,分别是原血胞、浆血胞、粒血胞、珠血胞和囊血胞。原血胞为小型圆形细胞,边缘圆滑、清晰,核质比例很大。粒血胞多为中型,形状不规则,边缘凹突不平,内含较大的异质性溶酶体颗粒。浆血胞多为中型,刚离体时形状较规则,常呈圆形、卵圆形。浆血胞内缺乏大的颗粒,细胞核大而圆形,细胞质内具许多小型颗粒状物质。浆血胞离体后形状变化较多,常发展出伪足,呈丝状、短芒状、钩状或片状伪足。珠血胞多为大、中型,外形大体呈圆形,但边缘由于大小不等的珠形内含物突出,呈花瓣状。囊血胞多为中型,圆形或椭圆形,细胞质内具有大小不一的带有折光性的颗粒或块状物,细胞边缘比较光滑。  相似文献

光学显微镜下观察苏云金杆菌感染印度谷螟后幼虫血细胞的病态变化证明:幼虫感病后,原血胞、粒血胞、浆血胞、珠血胞、类绛血胞、囊血胞均受到破坏,多数血胞出现空泡和变形,珠血胞珠粒之间互相脱离,细胞质外流,细胞膜破裂,囊血胞则有细胞外溢现象。这些病态变化均可在感病8小时以后观察到。  相似文献

Abstract Parasitism by the endoparasitic wasp Pteromalus puparum (Hymenoptera: Pteromalidae) by using only its associated venom, can suppress the immunal responses of Pieris rapae (Lepidoptera: Pieridae). However, up to now, current knowledge of the mechanisms has been limited. The response of host hemocytes to parasitism was investigated using a combination of light and transmission electron microscopy (TEM). Five hemocyte types, prohemocytes (PRs), granulocytes (GRs), plasmatocytes (PLs), oenocytoids (OEs) and coagulocytes (COs), were observed and characterized from both unparasitized and parasitized Pieris rapae pupae. Light microscopy showed that both GRs and PLs became more round and spread abnormally after parasitism, whereas the shape of other types of hemocytes remained unaffected. In addition, the size of PRs and PLs became larger while OEs became smaller. The proportion of PRs significantly increased after parasitism and that of PLs decreased by 43.9%, but there was no significant increase of GRs and OEs. TEM showed that all types of hemocytes except COs were damaged to various degrees after parasitism, especially resulting in electron opaque cytoplasm and nucleus, fewer cell organelles of rough endoplasmic reticulum, mitochondria and vesicles. Our results indicate that parasitism by P. puparum affects differential hemocyte counts and structures of host hemocytes, particularly for GRs and PLs, which may be the main cause of the parasitoid suppressing host cellular immune responses.  相似文献

Abstract The hemocytes of the pericardial body of Ciona intestinalis were studied by electron microscopy. Our findings showed that stem cells, clear vesicular granulocytes, microgranulocytes, unilocular granulocytes and globular granulocytes are present at the periphery of the smaller-sized pericardial bodies. The stem cells are small round cells with a large nucleus, with or without nucleolus, and homogeneous cytoplasm containing numerous ribosomes. The clear vesicular granulocytes are characterized by an ameboid shape and cytoplasm containing several large electron-lucent vacuoles and small electron-dense granules. The microgranulocytes are variable in shape and contain numerous large electron-dense granules. The unilocular granulocytes show a single large vacuole with an electron-dense or electron-lucent content and a thin layer of peripheral cytoplasm that contains the flattened nucleus. The globular granulocytes are characterized by the presence of large vacuoles containing either fibrogranular material or electron-dense aggregates.  相似文献

四种淡水鱼的血细胞形态基本相似。红血球形态与其他低等脊椎动物基本相似。淋巴球绝大部分是小淋巴球:单核球数量较少;四种鱼的嗜中性白血球形态结构差不多,胞核多为蚕豆形,很少见分叶核,分叶一般也只有二叶,这与哺乳类显然不同;嗜酸性白血球的形态结构与其他脊椎动物基本相似;在少数血涂片中看到了嗜碱性白血球。    相似文献

作者观察了鸡胚生殖新月区的原生殖细胞(PGC)的超微结构。PGC为圆形或椭圆形,13—16μm,有丰富的伪足和微绒毛,尚可见到相邻PGC存在桥粒样结构。细胞核为圆形、椭圆形及分叶状,并呈多处凹陷。与同期胚的其它细胞相比,胞质内细胞器相当丰富且较成熟。观察到有大量微丝。上述PGC的形态,除了细胞桥粒样结构及微丝很少见到报道外,其它特征与鸟类PGC的超微记载相一致。 作者首次观察到PGC中有一种特殊颗粒(即电子致密小体),它自核内产生,进入核周池,并借核膜破裂的方式进入胞质。这种颗粒可能就是生殖颗粒,而由该颗粒在胞质中聚集所构成的特殊高电子致密区可能就是生殖质。从而从形态学上提供了鸟类具有生殖质的证据。  相似文献

Series of experiments were undertaken to investigate whether application of Precocene II (PII) to parasitized S. littoralis larvae can influence their THCs and DHCs. PII was applied in single dose treatments (70μg/5 μl/dose). Haemolymph samples were taken at daily intervals from day 3 to day 8 post‐treatments. Two distinct alterations were noted; first, PII‐treatments caused significant decrease in THCs. Once the parasitoid larvae emerge from either treated or untreated hosts, a sharp decline in THCs was observed. The second effect was changes in DHCs of S. littoralis larvae. In controls, haemocyte types could be arranged in descending order due to their relative occurrence into plasmatocytes (PLs), prohaemocytes (PRs), spherule cells (SPs), granular cells (GRs) and oenocytoids (OEs). This order changed in treated hosts to PLs, GRs, PRs, SPs and OEs, i.e., GRs that were less prevalent in the haemolymph of control larvae increased significantly in PII‐treated hosts. In the latters, sharp rise in GRs levels followed parasitoid emergence. OEs cells were scarce in both treated and untreated hosts. No significant difference was observed between the overall means of PLs in both types of host larvae. But GRs‐PRs was markedly altered in treated larvae.  相似文献

Ultrastructural analyses revealed the presence of six hemocyte types in the hemolymph of Panstrongylus megistus, partially confirming our previous results obtained through light microscopy. Prohemocytes: small, round hemocytes with a thin cytoplasm layer, especially rich in free ribosomes and poor in membranous systems. Plasmatocytes: polymorphic cells, whose cytoplasm contains many lysosomes and a well developed rough endoplasmic reticulum (RER). They are extremely phagocytic. Sometimes, they show a large vacuolation. Granulocytes: granular hemocytes whose granules show different degrees of electrodensity. Most of them, have an internal structuration. Coagulocytes: oval or elongated hemocytes, which show pronounced perinuclear cisternae as normally observed in coagulocytes. The cytoplasm is usually electrodense, poor in membranous systems and contains many labile granules. Oenocytoids: large and very stable hemocytes, whose homogeneous cytoplasm is rich in loose ribosomes and poor in membranous systems. Adipohemocytes: large cells, containing several characteristic lipid droplets. The cytoplasm is also rich in glycogen, RER and large mitochondria. The total and differential hemocyte count (THC and DHC) were also calculated for this reduviid. THC increases from 2,900 hemocytes/mm3 of hemolymph in the 4th instar to 4,350 in the 5th and then, decreases to 1,950 in the adults. Plasmatocytes and coagulocytes are the predominant hemocyte types.  相似文献

The most effective and important strategy in the insect immune response is based on cellular reactions incorporating haemocytes. The present study uses Galleria mellonella (Lepidoptera: Pyralidae) as a host to study the pathogenesis caused by the entomopthoralean fungus Conidiobolus coronatus (Entomophthorales). Five types of haemocytes with different morphologies and behaviour are observed in the haemolymph of G. mellonella: granulocytes (GRs), plasmatocytes (PLs), spherulocytes (SPs), oenocytes (OEs) and prohaemocytes (PRs). During in vitro cultivation, three morphological subtypes of PLs are distinguished: flattened PLs, sun‐like PLs and oval PLs. In fresh smears of haemolymph observed under phase‐contrast microscopy, only flattened PLs are identified. No morphological changes are observed between fresh smears and in vitro cultures for GR, OE, SP and PR. Haemocytes cultured in vitro form a cellular network composed of PLs and GRs. Changes in the numbers, morphology and behaviour of haemocytes induced by fungal infection are compared with those observed in normally‐developing untreated larvae. Infection results in a significant drop in the number of haemocyte types. Fresh smears of haemocytes from mycosed larvae reveal malformed OEs, vacuolized PLs and GRs, as well as PLs with apoptotic blebs. Haemocytes from mycosed larvae incubated in vitro look similar, with degranulated GRs and vacuolized PLs forming microaggregations, as well as deformed OEs; only the SPs remain unharmed. Fungal infection impairs the ability of haemocytes to attach and spread on the culture dish. The actin cytoskeleton of haemocytes from mycosed larvae appear disorganized.  相似文献

Five hemocyte types were identified in the hemolymph of Panstrongylus megistus by phase contrast and common light microscopy using some histochemical methods. These are: Prohemocytes, small cells presenting a great nucleus/cytoplasm ratio; Plasmatocytes, the most numerous hemocytes, are polymorphic cells mainly characterized by a large amount of lysosomes; Granulocytes, hemocytes very similar to plasmatocytes which contain cytoplasmic granules and are especially rich in polysaccharides; Oenocytoids, cells presenting a small nucleus and a thick cytoplasm; they show many small round vacuoles when observed in Giemsa smears and many cytoplasmic granules under phase microscopy; Adipohemocytes, very large hemocytes, presenting many fat droplet inclusions which could correspond to free fat bodies which entered the hemolymph. Only prohemocytes and plasmatocytes can be clearly classified; all the other hemocyte types have a more ambiguous classification.  相似文献

采用常规瑞氏染色和细胞化学染色方法对团头鲂(Megalobrama amblycephala)外周血细胞的显微结构及细胞化学特征进行了观察。在团头鲂外周血细胞中可区分出六类细胞: 红细胞、淋巴细胞、单核细胞、嗜中性粒细胞、嗜酸性粒细胞和血栓细胞。其中淋巴细胞是除红细胞外含量最多的细胞, 其次分别为血栓细胞、单核细胞、嗜中性粒细胞、嗜酸性粒细胞。成熟红细胞多为卵圆形, 表面光滑, 胞核呈椭圆形或圆形, 染色质较为致密; 淋巴细胞多呈圆形, 胞质较少, 胞核常偏位; 单核细胞多为圆形, 胞核呈圆形或椭圆形, 胞质内可见空泡状结构; 嗜中性粒细胞近似圆形, 胞核常偏于细胞一侧, 呈分叶状、肾形或椭圆形, 核质界限清晰; 嗜酸性粒细胞一般为圆形, 胞核为肾形或椭圆形, 胞质中充满紫红色颗粒; 血栓细胞形态多样, 主要有椭圆形、纺锤形、长杆状和泪滴形, 核质比较大。淋巴细胞呈α-醋酸萘酚酯酶(ANAE)阳性, 呈过碘酸-雪夫(PAS)、氯乙酸AS-D萘酚酯酶(AS-DCE)弱阳性, 呈苏丹黑B(SBB)、酸性磷酸酶(ACP)、碱性磷酸酶(AKP)及过氧化物酶(POX)阴性; 单核细胞呈POX、ACP强阳性, PAS、SBB、AS-DCE和ANAE为阳性, 呈AKP阴性; 嗜中性粒细胞除PAS和ANAE为弱阳性外, 其他染色结果和单核细胞相同; 嗜酸性粒细胞呈POX、ANAE强阳性, SBB、ACP阳性, PAS及AS-DCE则为弱阳性, 呈AKP阴性; 血栓细胞呈PAS、AS-DCE及ANAE弱阳性, 呈SBB、ACP、AKP及POX阴性。团头鲂外周血细胞的显微结构及细胞化学特征与其他鱼类具有相似之处, 但亦有其明显的物种特异性。该研究结果可作为监测团头鲂健康状态的依据, 为其养殖及病理诊断提供基础资料。  相似文献
