The incidence of coiled-sprout was determined in Scotch and local-grown Arran Pilot and Duke of York seed tubers which had been stored at 10°, 4° and 15° C. and in a farm store with no temperature control. All four tuber types were planted in the field and, in addition, the two types of Duke of York were planted in Perlite at 7°, 10° and 15° C. In the field, and when maintained at 7° and 10° C, the percentage of sprouts coiling and the intensity of coiling was greater in tubers stored at 10° and 15° C. than at 4° C. There was no coiling when the Duke of York tubers were planted at 15° C. In a further experiment tubers were stored at 20° C. in the light and dark and samples were planted monthly for 3 months at temperatures of 7°, 10° and 15° C. During the following 3-month period only light-stored were planted because of the excessive amount of tip-death in the tubers stored in the dark. There was very little coiling in the dark-stored tubers. In the first two plantings of the light-stored tubers there was virtually no coiling of those planted at 15° C. There was some, however, at 7° and 10° C. In subsequent plantings there was more coiling and no effect of planting temperature. Attempts to isolate Verticiculum nubilum from sprouts were successful in only a small percentage of attempts and it was not possible to demonstrate any difference between its distribution on coiled and normal sprouts. It was not possible to induce coiling by infection of sprouts with spores of V. nubilum. Over a wide range of sprout sizes the amount of coiling was a function of sprout size at planting. However, the parts which coiled were those in the apical bud of the sprout at the time of planting and hence contributed only a small amount to the total sprout size. It is likely, therefore, that the correlation between coiling and sprout size reflects the changing metabolism of the elongating regions of sprouts with their increase in length, these regions developing in such a way as to produce a greater tendency to coiling. The internal reactions concerned in these changes, however, are not known.  相似文献

The relationships between rain and blight (Phytophthora infestans) were studied in unsprayed crops of cultivars differing widely in foliage and tuber susceptibility. The occasions when tubers were infected depended on rain and not cultivar, but numbers of tubers infected after rain was affected by the blight susceptibility of the cultivar. Infected tubers were first found when less than 5 % (BMS key) of the potato foliage was infected but few fresh infections occurred when 50–75% of the foliage had been destroyed. Some tubers were infected after 8 mm rain (tubers near the surface with even less) but large increases in numbers of tubers infected usually occurred only after 25 mm or more had increased soil moisture to above ‘field capacity’ around the tuber for at least 24 h. The most susceptible cultivars Ulster Ensign and Arran Banner had all plants with some tuber blight, and some plants with all tubers affected and often many lesions per tuber. Cultivars of intermediate susceptibility, King Edward and Up-to-Date, had some plants without blighted tubers, many with a few and very few with all. The more resistant cultivars Majestic and Arran Viking had many plants without infected tubers and many lesions that aborted while still necrotic threads, so that the fungus did not spread. Most infections occurred through tuber eyes, lenticels or sometimes growth cracks. The distribution of blight lesions on tubers differed in the different seasons, for example, lenticels were most commonly infected on Arran Banner and Ulster Ensign and eyes on King Edward, Majestic and Arran Viking. In late or slowly developing attacks, lesions on stems became more numerous and larger than in fast, early attacks and were prolific sources of spores on King Edward and Up-to-Date but not on Majestic and Arran Viking. Because much rain water runs down the stems of Up-to-Date and King Edward, stem lesions can provide an important source of inoculum for tubers.  相似文献

Fungus diseases on potato seed tubers planted in England and Wales, 1963-76   总被引:4,自引:0,他引:4  
During 1963-76 samples of potato tubers from commercial seed stocks of cvs King Edward (14 yr), Pentland Crown (9 yr), Majestic (7 yr), Pentland Dell (3 yr), Record and Arran Pilot (2 yr) were received from farms in England and Wales. Fifty tubers from each sample were examined macroscopically for fungus diseases and eyes were excised from a 20-tuber sub-sample, incubated and examined for pathogenic fungi; 50 tubers were stored on trays to sprout and examined for diseases and sprouting in May and in most years samples of 50 tubers were wounded by dropping onto expanded metal, stored at 5° C and examined for gangrene and dry rot after 12 wk. Amounts of disease varied between years and during 14 yr black scurf and powdery scab on King Edward tended to increase and skin spot and late blight decrease. On average 44% of King Edward tubers were affected with skin spot, 25% with black scurf and 16% with powdery scab. Gangrene affected 5% of tubers and 97% of the isolates from rots were identified as Phoma exigua var. foveata. Wounding tubers increased the incidence of gangrene three-fold. During 1963-69 late blight affected 2% of King Edward tubers but fewer in later years and in other cultivars. Majestic had most common scab (44% tubers) and Arran Pilot most dry rot (9% tubers) and this disease was increased by wounding tubers. Conidiophores of Helminthosporium solani (silver scurf) were more common on excised eyes of Pentland Crown, Record and Arran Pilot than of other cultivars, and isolations from verticillate conidiophores that developed on the side of incubated eye plugs of King Edward and Majestic stocks gave pure cultures of Verticillium tricorpus (78%), V. nigrescens (9%) and V. nubilum (3%). Proportions of tubers with different diseases were affected by their country of origin; Scottish seed had most skin spot and gangrene, Irish seed most powdery scab and English seed most common scab, late blight and H. solani. There was also evidence of differing disease incidence in seed from different geographical areas in Scotland and England. Up to half the King Edward and Pentland Crown stocks examined in 1975 and 1976 were derived from stem cuttings and average amounts of diseased tubers were similar to those in stocks not derived from stem cuttings. Annual and cultivar differences in disease incidence and effects of date of receipt of seed on farms are discussed.  相似文献

Large seed tubers (mean 134 g) and small seed tubers (50 g) of three early and three maincrop potato cultivars, spaced respectively 48 and 30 cm apart within rows, were inoculated with Rhizoctonia solani at planting in 1985 and 1986. All seed of early cultivars was sprouted and maincrop seed was either sprouted or not sprouted. In all cultivars, plant emergence was slower from small than large seed and with both was delayed by inoculation. From 11 wk after planting numbers of stems and tubers and weights of foliage and tubers/m2 were usually similar from small and large seed when not inoculated, but inoculating delayed plant growth and decreased tuber numbers and yield more from small than large seed. Inoculating decreased mean yields from large and small seed of early cultivars at 11 wk by respectively 24% and 31% (Arran Comet), 12% and 18% (Estima) and by 10% and 28% (Wilja) and losses were greatest with saleable sized tubers (3–4 cm). When grown to maturity inoculating decreased yields by 7% (Arran Comet), 5% (Estima) and 14% (Wilja). With maincrop cultivars, yields in October from large and small seed were decreased by respectively 4% and 10% (DCsirke), 9% and 12% (Maris Piper) and by 14% and 22% (Pentland Squire). In all cultivars yields of tubers < 44 mm and 44–70 mm were decreased and, with Pentland Squire, tubers > 82 mm were increased. The incidence of stem canker and of black scurf on progeny tubers was not affected by seed size but in all cultivars the percentage of greened tubers was slightly increased by inoculation.  相似文献

Treatments reported to affect the incidence of ‘coiled sprout’ were studied in 1968 with the cultivar Arran Pilot in experiments near Falmouth, Cornwall (F), Rothamsted (R) and near Edinburgh (E). Most coiled stems occurred at E (37%) with similar proportions at R (19%) and F (16%). Seed tubers with the longest sprouts at planting produced most coils at all sites, those with shorter sprouts less and unsprouted seed least. When coiling was assessed on the number of plants, long- and short-sprouted seed tubers were about equally affected at R and E. Sprouts emerged within 45 days at E and 30 days at R. Longer sprouts did not emerge more quickly, but emergence was more than 1 wk later from unsprouted seed at R and only a few days later at E. Harrowing ridges after planting to decrease the depth of soil covering seed tubers hastened emergence by about 1 wk at R and decreased the incidence of coiling; it had little effect at E but tended to increase coiling at F. Seed tubers inoculated with Verticillium nubilum produced more coiled sprouts but this increase was not significant. The highest final yields were achieved at R (43-2 t/ha) where short-sprouted seed outyielded long-sprouted and unsprouted seed, but cultivation treatments and fungus inoculation had no effect. At E (21-2 t/ha) only herbicide treated plots gave a significant effect, outyielding harrowed plots by 8 t/ha. At F (18-2 t/ha) short-sprouted seed outyielded long-sprouted and unsprouted seed, and herbicide treated plots outyielded harrowed plots. When ware yields from individual plants with and without coils from long-sprouted seed were compared at R on 18 June, coiled plants gave about 30% and on 2 July 60% of the yield of non-coiled, but by 16 July 120% yield (20% more); similar trends were shown by short- and by long-sprouted seed at E.  相似文献

In 1983 and 1984, potato seed tubers of five early and seven maincrop cultivars were inoculated with cultures of Rhizoctonia soluni during planting to simulate severe seed infection. Shoot and stolon infection was assessed in June-August and black scurf on tubers recorded after harvest in October. Almost all shoots of all cultivars had stem canker in both years and disease on shoots, stolons and tubers was more severe in 1984 than in 1983. In 1983 similar amounts of disease developed on all early cultivars and between 11% (Ulster Sceptre) and 32% (Maris Peer) shoots were pruned off. Maris Peer had a stem canker score lower than other cultivars in 1984 but more than half the shoots were pruned off. Shoot pruning on Estima, Ulster Prince and Ulster Sceptre was more common on plants from sprouted than non-sprouted seed. Between 30 and 50% of stolons were pruned off. After harvest in 1985, black scurf was least prevalent on Arran Comet and Maris Peer tubers and in 1984 on Arran Comet and Estima tubers from non-sprouted seed. Of the maincrop cultivars, King Edward plants from sprouted seed had many shoots pruned off in both years. Shoot pruning was also prevalent on Maris Piper and Pentland Squire plants from non-sprouted seed. Record had fewest pruned shoots and stolons and the lowest stem canker score. The disease was more severe on Pentland Crown and Maris Piper plants from non-sprouted than sprouted seed. Black scurf was most common on Cara and King Edward tubers in 1983 and on King Edward and Record tubers in 1984. In both years few shoots but many tubers were infected on plants from non-inoculated seed and the significance of this is discussed.  相似文献

Strains of Phytophthora infestans of the same race (P4) isolated from a range of R0 potato varieties differed markedly in their rate of growth on agar but differences in growth rate were not related to variety of origin. Two isolates from each of the varieties Duke of York, Majestic and Arran Consul were grown for six successive subcultures on tubers of these three varieties. The six isolates differed in aggressiveness as shown by their growth on individual varieties. There was a marked difference in growth rate between alternate subcultures which is attributed to the use of too long an incubation period. Analysis of the data for subcultures 1, 3 and 5 revealed a significant interaction between isolates and varieties which reflected the enhanced growth of five of the isolates on the variety from which they had been obtained. Serial subculture on potato tubers of the same variety had no general or specific effect on the aggressiveness of the isolates. It is concluded that strains of P. infestans of the same race may be differentially adapted to the variety on which they are growing in the field. Differential interactions between pathogen biotypes and non-immune hosts are considered to be widespread in host-pathogen systems.  相似文献

Methods of measuring the prevalence of Phoma exigua on potatoes and in soil   总被引:5,自引:0,他引:5  
Methods are described for assessing inoculum of Phoma exigua by directly wounding potato test tubers and by inoculating soil into Arran Banner test tubers or slices. Large samples of tubers were wounded by dropping onto perforated metal sheet or grading on farm riddles, but standardization was achieved by inflicting four uniform wounds with shaped brass teeth. Tubers were then incubated for 12 wk at 5 oC. Soil samples to be tested were inoculated into crush wounds on surface-sterilized test tubers or tuber slices which were incubated for 8 wk at 5 oC. Assessments were speeded by immersing wounded slices in a mixture of Agrimycin and isopropylphenyl carbamate (IPC) solutions before inoculating, and then incubating for 2–3 wk at 10 oC. Although the sensitivity of test tubers is limited by their latent infection, comparisons of the tests in 3–5 years showed that the test tuber methods were frequently more sensitive than direct wounding, especially when amounts of inoculum were small.  相似文献

Vitamins of the B group and vitamin C were applied to purple nutsedge (Cyperus rotundus L.) tubers to study their effect on the sprouting rate, initiation and establishment of sprouts, growth of plantlets and development of orthotropic rhizomes in comparison with the corresponding effects of kinetin. Ascorbic acid up to 100 mg I?1 hastened sprouting, whereas vitamins of the B group and kinetin retarded sprouting; 100% tubers sprouted in all treatments within 10 days. Unlike kinetin, none of the vitamins resulted in the establishment of more than one sprout per tuber. Riboflavin and pyridoxine promoted root and shoot growth of the plantlets, whereas kinetin produced short, thick shoots and inhibited root growth, with increasing concentration. Ascorbic acid was only second to kinetin in the induction of orthotropic rhizomes, but the former resulted in an increase in rhizome length.  相似文献

The potato R locus codes for dihydroflavonol 4-reductase   总被引:1,自引:0,他引:1  
The potato R locus is required for the production of red pelargonidin-based anthocyanin pigments in potato (Solanum tuberosum L.). Red color also requires tissue-specific regulatory genes, such as D (for expression in tuber skin) and F (expression in flowers). A related locus, P, is required for production of blue/purple anthocyanins; P is epistatic to R. We have previously reported that the dihydroflavonol 4-reductase gene (dfr) co-segregates with R. To test directly whether R corresponds to dfr, we placed the allele of dfr associated with red color under the control of the CaMV 35S promoter and introduced it into the potato cultivar Prince Hairy (genotype dddd rrrr P-), which has white tubers and pale blue flowers. Transgenic Prince Hairy tubers remained white, but flower color changed to purple. Three independent transgenic lines, as well as a vector-transformed line, were then crossed with the red-skinned variety Chieftain (genotype D-R-pppp), to establish populations that segregated for D, R, P, and the dfr transgene or empty vector. Markers were used to genotype progeny at D and R. Progeny carrying the empty vector in the genetic background D-rrrr produced white or purple tubers, while progeny with the same genotype and the dfr transgene produced red or purple tubers. HPLC and LC–MS/MS analyses of anthocyanins present in Chieftain and in a red-skinned progeny clone with the dfr transgene in a D-rrrr background revealed no qualitative differences. Thus, dfr can fully complement R, both in terms of tuber color and anthocyanin composition.  相似文献

By growing the excised apical meristems of sprouts from the potato varieties King Edward and Arran Victory, infected respectively with potato paracrinkle virus and potato virus S , virus-free plants were obtained. Although the method failed to produce virus-free plants from varieties infected with potato virus X , this virus also seems not to be present in apical meristems, for no virus could be demonstrated in callus tissue that developed from excised meristems less than 200 μ across. The concentration of tobacco mosaic virus in tomato roots and tobacco stems is also much less near the growing point than in older cells, but there is no evidence that the meristematic region is virus-free.  相似文献

Virus-free clones of some British potato varieties, of which all commercial stocks are infected, were obtained by culturing apical meristems of sprouts on tubers. The variety Golden Wonder was freed from potato virus A, Arran Comet from potato virus X, and Epicure, Orion and Sharpe's Express from potato virus S. Of the 196 meristems cultured, forty-one (21%) grew and of these twenty developed into plants, of which nineteen were virus-free. More of the excised meristems grew when the first leaf primordium was included than when not, and its inclusion seemed not to increase the chances of the progeny being infected.  相似文献

  • Floral colour is a key reproductive character, often associated with environmental adaptation, and subject to human intervention. A large number of Rhododendron species differ widely in flower colour, providing a good model for flower colouration. The chromatic features and anthocyanin compositions of 30 species from seven subgenera were systematically analysed.
  • The Royal Horticultural Society Colour Chart and CIE L*a*b* system were employed to describe and investigate flower colours. The UPLC‐PDA/ESI‐MSn system was used to identify and quantify anthocyanins in petal extracts.
  • The flower colours of 30 Rhododendron species were categorised into four groups – red, purplish pink, purple and white. Seven anthocyanins were identified and quantified in petals: delphinidin, cyanidin and malvidin 3‐O‐arabinoside‐5‐O‐glucosides, cyanidin 3,5‐di‐O‐glucoside, 3‐O‐galactoside and 3‐O‐arabinoside, and delphinidin 3‐O‐glucoside. The red‐flowered species mainly contained cyanidin monoglycosides and had much higher total anthocyanin content than purplish pink‐ and purple‐flowered species. Purplish pink‐ and purple‐flowered species had similar anthocyanin types and content. The chromatic differences were significant among groups, except the purplish pink and purple groups. Statistical analysis showed that Cy3Gal and Cy3Arb are characteristic for red‐flowered species, and Mv3Arb5G and Dp3Arb5G play important roles in purple colouration; their contents were major components that greatly affected the chromatic parameters. In total, 21 flavonol derivates were identified. However, total flavonol content and co‐pigmentation index showed no significant difference or correlation among/with colour groups, suggesting that flavonols might not play a major role in colouration.
  • These results enhance our knowledge of the biochemical basis of flower colouration in Rhododendron species, and provide a foundation for genetic variation studies and aid in breeding cultivars with novel flower colours.

The results of experiments on the sprouting and growth of purple nutsedge (Cyperus rotundus L.) in relation to pH and aeration are described. All pH levels between 2.2 to 9.0 allowed high sprouting percentage but the sprouted tubers did not survive at pH below 3 and survival beyond pH 7 was very poor. Post-sprouting growth of nutsedge was optimal at pH 4. Pre-sprouting aeration of tubers stimulated early sprouting and produced vigorous shoots, whereas submersion of the tubers was inhibitory. Continuous aeration of the submerged tubers substantially removed the inhibitory influence on sprouting but the shoots remained weak. Seedlings grown in constantly aerated nutrient solution showed significantly better growth than in a non-aerated one.  相似文献

Summary T-even type phages recognize their cellular receptors with the tip of their long tail fibres. The gene products involved in receptor recognition are proteins 37 and 38. While screening libraries of phage K3 with a probe of gene38 from phage T2, a class of weakly hybridizing clones was found in addition to the expected clones of gene38 of K3. One of these clones was identified as being from gene23 of the phage which codes for the major head subunit; another clone originated from gene34, which codes for the proximal half of the long tail fibres. Neither gene product 23 nor 34 is involved in receptor recognition. Phages can recombine with the DNA of the gene23 and gene34 clones and change the host range. Potato mop-top virus (PMTV) was best detected in field soils by air-drying them for more than a week before remoistening and growing seedlings of Nicotiana tabacum or N. debneyi for a 6–10 week period. Infection of N. tabacum was assessed by inoculating sap from roots and shoots to Chenopodium amaranticolor. Similar inoculations from N. debneyi were far less convenient for detecting PMTV than recording leaf symptoms, but slightly more efficient. Air-dry soil retained PMTV infectivity for 9 months, when passed through a 50 μ sieve or when diluted with 103 but not 104 parts of steamed soil. Tobacco seedlings were not infected when their roots were steeped in PMTV-containing tobacco sap. Infective soils contained Spongospora subterranea, spore balls of which resisted air-drying for more than a year and passed a 50 μ sieve. Roots of susceptible seedlings were infected with PMTV when exposed to spore balls of S. subterranea taken from powdery scabs on PMTV-infected potato tubers, or to suspensions obtained by steeping, in nutrient solution, roots infected with virus-carrying cultures of S. subterranea. Plants in several families were hosts of S. subterranea, but probabilities of infection when exposed to spore balls differed greatly between families and only species of Solanaceae were good hosts. The ten species infected with PMTV when grown in infective soil or when exposed to spore balls of S. subterranea taken from PMTV-infected potato tubers are all members of this family. PMTV seems to be carried internally in S. subterranea spore balls and survived in them for at least a year. PMTV was transmitted by S. subterranea to Arran Pilot potato, causing yellow blotches in some leaves and spraing in many tubers. However, when newly infected with PMTV in the field, not all Arran Pilot tubers developed spraing. Also, although many spraing-affected or symptomless but PMTV-infected tubers carried PMTV-containing spore balls of S. subterranea, powdery scabs were rarely found near the centres of the rings of primary spraing. PMTV became established in virus-free soil when PMTV-infected tubers carrying S. subterranea were planted as ‘seed’ but not when virus-free tubers bearing powdery scabs were used. 5. subterranea seems the main, and possibly the only, vector of PMTV in the soils examined. S. subterranea did not transmit potato aucuba mosaic virus from potato to N. debneyi or Capsicum annuum.  相似文献

In 1983 and 1984, potato seed tubers of five early and seven maincrop cultivars were inoculated with cultures of Rhizoctonia solani during planting in field experiments to simulate severe seed infection. The size of foliage was assessed during June-August and tuber yields recorded during growth and at harvest in October. Stem canker delayed shoot emergence, decreased the number and length of stems and caused increased variation in stem length; these effects were greatest with Maris Peer and Arran Comet (early cultivars) and King Edward and Pentland Squire (maincrop cultivars). Total weight of foliage was decreased, especially with earlies, dry matter of stems increased and the proportion of foliage on lateral stems increased. With the early cultivars, tuber yield from sprouted Maris Peer seed 11 wk after planting in 1983 was decreased by 24%, and 13 wk after planting in 1984 yields were decreased by 42% (Maris Peer), 40% (Ulster Sceptre), 34% (Estima), 30% (Arran Comet) and 17% (Ulster Prince) with sprouted seed and by 20, 29, 53, 39 and 28% respectively with non-sprouted seed. Decrease in total yield at harvest in October averaged 13% with sprouted seed and 10% with non-sprouted seed. In all cultivars the weight of small tubers was decreased and with Estima the weight of large tubers was increased. Tuber bulking was also delayed with all maincrop cultivars and at harvest yields from sprouted King Edward seed were decreased by 13% in 1983 and by 16% (sprouted seed) and 23% (non-sprouted seed) in 1984; yields of Pentland Squire were decreased by 5, 16 and 21% respectively. Yield losses with other cultivars ranged from 5–13% with sprouted seed and 0–16% with non-sprouted seed. The yields of small tubers were decreased with all cultivars and yields of large tubers were increased with Pentland Squire, Pentland Crown and Cara.  相似文献

Thiabendazole, iprodione and benodanil were tested for control of stem canker and black scurf disease of potatoes using seed tubers with or without black scurf planted in soil infested or not with Rhizoctonia solani . Dormant seed tubers were treated with fungicide dusts, dips or sprays and before planting dusts were applied to soil or to sprouts on seed tubers.
Plants grown from seed tubers with black scurf had more stem canker, stolon infection and Thanatephorus cucumeris and progeny tubers had more black scurf than from seed without black scurf. Infesting soil with R. solani increased black scurf but not stem canker or stolon infection.
Fungicide treatment of seed tubers controlled all phases of the disease and sprout and soil treatments also had significant effects. Numbers of tubers in July were usually increased by treatment although by harvest many small tubers (>3 cm) had been resorbed. Fungicide treatment substantially increased yield only when shoot tips were pruned by R. solani .
Regression analyses showed that stolon pruning, Thanatephorus and black scurf were significantly related to stem canker, and numbers of tubers in July but not in September were inversely related to stolon pruning.
The significance of sources of inoculum and the effects of controlling disease on tuber production are discussed in relation to methods of fungicide use.  相似文献

Summary Gynandric basitarsi possessing areas heterozygous for the sexlinked mutant sexcombless (sx) and hemizygous for it, and mosaic basitarsi possessingsx/+,sx/sx and +/+ areas have been studied for the developmental expression ofsx andsx/sx patches in the sex comb region. The mosaic basitarsi were obtained as the result of somatic crossing over following X-ray exposure of larvae. In order to observe the sex comb-forming potencies in these XX tissues the flies had been made homozygous for the genetra which transforms females into phenotypic males. The development ofsx/+;tra/tra females is greatly prolonged over that of normal females.Five of the 63 gynandric and two of the 39 radiation-induced mosaics showed a single typical tooth, a sixth gynandric basitarsus had two teeth. In all other cases intermediate bristles, or macrochaetae, or both, have been found iny sn sx hemizygous or homozygous patches irrespective of their size.Thesx-initiated distortion of bristle arrangement found in non-mosaicsx male basitarsi has also been seen in many mosaic patches.The mutant genesx acts autonomously and interferes with the process of seta-cell to tooth determination at a relatively early stage in development of a bristle organ.With 38 Figures in the Text  相似文献
