本文探讨介质中ATP浓度和急,慢性缺氧暴露对大鼠脑线粒体内RNA和蛋白质合成的影响。用差速离心法分离正常和低压舱模拟4000m高原急性连续缺氧暴露3d和慢性连续缺氧暴露40d大鼠脑线粒体,用体外无细胞(cell-free in vitro)^3H-UTP和^3H-Leucine掺入法分别测定线粒体RNA和蛋白质合成活性,结果显示,大鼠急性缺氧暴露后大脑皮质线粒体RNA体外合成活性降低40%,蛋白质合成活性降低60%;慢性缺氧暴露后线粒体RNA和蛋白质合成活性分别为对照的72%和76%;ATP对正常大鼠脑线粒体RNA以及蛋白质的体外合成活性的影响均呈双相性,大于或小于1mmol/L均可产生不同程度的抑制效应,结果提示,缺氧可在转录和翻译两个水平上影响脑线粒体mtDNA的表达,而慢性缺氧暴露时,线粒体半自主性功能的改善可能是机体对缺氧适应的细胞机制之一;ATP对脑线粒体内转录和释放活性的调节是一种经济有效的反馈调节方式。  相似文献

本文旨在通过观察棕榈酸对模拟高原低氧大鼠离体脑线粒体解耦联蛋白(uncoupling proteins,UCPs)活性的影响及脑线粒体质子漏与膜电位的改变,探讨UCPs在介导游离脂肪酸对低氧时线粒体氧化磷酸化功能改变中的作用.将SpragueDawley大鼠随机分为对照组、急性低氧组和慢性低氧组.低氧大鼠于低压舱内模拟海拔5 000 m高原23 h/d作低氧暴露,分别连续低氧3 d和30 d.用差速密度梯度离心法提取脑线粒体,[3H-GTP法测定UCPs含量与活性,TPMP 电极与Clark氧电极结合法测量线粒体质子漏,罗丹明123荧光法测定线粒体膜电位.结果显示,低氧使脑线粒体内UCPs含量与活性升高、质子漏增加、线粒体膜电位降低;同时,低氧暴露降低脑线粒体对棕榈酸的反应性,UCPs活性的改变率低于对照组,且线粒体UCPs含量、质子漏、膜电位变化率亦出现相同趋势.线粒体质子漏与反映UCPs活性的Kd值呈线性负相关(P<0.01 r=-0.906),与反映UCPs含量的Bmax呈线性正相关(P<0.01,r=0.856),与膜电位呈线性负相关(P<0.01,r=-0.880).以上结果提示,低氧导致的脑线粒体质子漏增加及膜电位降低与线粒体内UCPs活性升高有关,同时低氧暴露能降低脑线粒体对棕榈酸的反应性,提示在高原低氧环境下,游离脂肪酸升高在维持线粒体能量代谢中起着自身保护和调节机制.  相似文献

目的:探讨低氧对大鼠脑线粒体DNA表达的影响及其与能量生成的关系。方法:雄性Wistar大鼠随机分为3组:急性低氧组(AH)、慢性低氧组(CH)和对照组,其中急、慢性低氧组动物分别连续暴露于模拟海拔4000m高原3d(AH)和40d(CH)。分离脑线粒体,分别测定线粒体体外转录活性、F0F1-ATP酶活性以及ATP对线粒体体外转录的影响。结果:急性低氧大鼠脑线粒体体外转录活性及F0F1-ATP酶活性显著降低,慢性低氧时有所回升,两者呈线性相关。ATP对大鼠脑线粒体体外转录活性呈双相效应。结论:低氧时脑线粒体转录活性改变可能参与低氧抑制线粒体能量代谢的机制,ATP可能通过反馈作用对线粒体转录进行微调。  相似文献

目的:观察模拟高原低氧对成年大鼠脑内胶质细胞的影响。方法:大鼠在模拟海拔4000m高原低压舱内连续暴露1、3、7d,胶质原纤维酸性蛋白(GFAP)与Griffoniasimplicifolia同功凝集素(GSA-IB4)组织细胞化学分别显示星形胶质细胞与小胶质细胞。结果:GFAP与GSA-IB4阳性细胞在低氧后3.7d两个时相显著增多,主要分布于新皮层、海马、纹状体及室管膜下层等区域。结论:模拟高原低氧能引起大鼠脑内星形胶质细胞与小胶质细胞的显著活化。  相似文献

目的:观察共轭亚油酸(CLA)对急性缺氧大鼠肝脏线粒体呼吸链酶活性及肝脏内氧化应激(OS)的影响。方法:将60只SD大鼠随机分为正常对照组,模拟海拔5000米高原环境连续缺氧暴露3d的急性缺氧组,急性缺氧CLA干预组。生化法测定肝脏组织中丙二醛(MDA)、还原型谷胱甘肽(GSH)含量及超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性,Clark氧电极法观察大鼠肝脏线粒体呼吸链酶活性。结果:成功诱导大鼠急性缺氧模型。与对照组比,急性缺氧组大鼠肝脏MDA含量明显升高(P<0.05),同时肝脏线粒体呼吸链酶活性以及抗氧化酶活性(SOD,CAT)及GSH显著降低(P<0.05),与急性缺氧相比,CLA治疗组大鼠以上各项指标均有改善,并存在一定的剂量效应关系。结论:CLA通过抑制氧化应激增强大鼠肝脏线粒体呼吸链酶活性改善了急性缺氧大鼠肝脏的能量代谢障碍,对急性缺氧引起的氧化损伤有保护作用。  相似文献

高原冻伤大鼠骨骼肌腺苷酸含量的变化   总被引:2,自引:1,他引:1  
本文以HPLC测定了冷冻前后平原冻伤(FN)、急性缺氧冻伤(FAH)和缺氧习服缺氧冻伤(FHAC)大鼠冻肢腓肠肌中三种腺苷酸含量的变化。模拟6000m间断性缺氧习服4周大鼠腓肠肌ATP含量明显减少,ADP和AMP含量明显增高;而急性缺氧4h大鼠与FN组大鼠腓肠肌三种腺苷酸含量无明显差异。冻后24h各组大鼠冻肢腓肠肌三种腺苷酸含量均明显减少。冻后120h三种腺苷酸含量均有不同程度的增高:FN组腓肠肌ATP、ADP含量恢复较快,FHAC组AMP含量明显高于FN和FAH组。除冻前ATP/TA、AMP/TA及冻后AMP含量的变化与组织损伤程度一致外,其余未见一致性的变化。讨论了冻伤后腺苷酸减少的可能原因。作者认为能量代谢变化在平原冻伤与高原冻伤发病中的作用不尽相同;AMP降解过程中生成的氧自由基在高原冻伤组织损伤中的作用需进一步研究证实。  相似文献

腺苷酸转移酶(ANT)是线粒体内膜上负责能量分子传导的转运蛋白, 在细胞凋亡调控网络中有重要作用。本研究以棉铃虫幼虫组织的mRNA为模板, 根据鳞翅目昆虫ant基因编码区保守序列设计引物, 进行RT-PCR分析, 同时结合5′、3′ RACE方法扩增出棉铃虫ant基因的全长cDNA序列, cDNA全长为1 190 bp (GenBank登录号AY253868), 具有完整的开放阅读框架(ORF, 133~1 033 bp), 编码蛋白为300个氨基酸, 其中N端22个氨基酸为信号肽, 引导ANT蛋白定位于线粒体内膜。该蛋白具有3个保守的线粒体穿膜功能结构域, 形成能量分子传导的转运通道, 催化细胞质中ADP和线粒体内ATP间进行跨膜交换。通过与其他昆虫的腺苷酸转移酶蛋白序列比较, 发现该基因具有高度的保守性, 氨基酸序列同源性都在90%左右。  相似文献

目的:研究慢性间断低氧暴露对大鼠心肌线粒体Na 、K -ATPase和Ca2 、Mg2 -ATPase以及呼吸链酶复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ活性的影响.方法:经慢性间断低氧暴露(模拟海拔3 000 m、5 000 m分别低氧,每天4 h,共2周,最后8 000 m低氧4 h)和急性低氧(模拟海拔8 000 m低氧4 h)的大鼠,断头处死,迅速取出心脏,分离心肌线粒体,用水解磷酸根法测定ATP酶活性,用Clark氧电极法测定呼吸链酶复合物的活性.结果:①慢性间断低氧暴露对大鼠心肌线粒体Na 、K -ATPase的活性无明显影响.②急性低氧大鼠心肌线粒体Ca2 、Mg2 -ATPase的活性较正常大鼠显著降低,而慢性间断低氧暴露大鼠心肌线粒体Ca2 、Mg2 -ATPase的活性则明显升高,接近正常水平.③急性低氧大鼠心肌线粒体呼吸链酶复合物I(NADH-CoQ还原酶)、复合物Ⅱ(琥珀酸-CoQ还原酶)、复合物IV(细胞色素氧化酶)活性较正常大鼠显著降低,而经慢性间断低氧暴露后,三者的活性均显著提高.相同实验条件下,低氧对复合物Ⅲ(CoQ-细胞色素C还原酶)活性无明显影响.结论:慢性间断低氧暴露可以显著提高心肌线粒体Ca2 、Mg2 -ATPase和呼吸链酶复合物Ⅰ、Ⅱ、Ⅳ的活性,从而改善低氧时心肌线粒体呼吸链的功能,维持心肌正常能量代谢,最终提高心肌收缩和舒张功能.  相似文献

目的:探讨睡眠剥夺对大鼠心肌和抗氧化指标的影响。方法:大鼠随机分为睡眠剥夺2 d组(SD 2 d)、睡眠剥夺4 d组(SD 4 d)、睡眠剥夺6 d组(SD 6 d)、大平台对照组(TC)、正常对照组(CC),利用"小平台水环境法"建立大鼠睡眠剥夺模型,利用BL-410机能实验系统描记体表心电图,采用光镜及透射电镜观察心肌形态学改变,测定心肌线粒体丙二醛含量、超氧化物歧化酶活性。结果:睡眠剥夺组大鼠心率加快,心电图出现缺血性改变;心肌细胞染色质、肌浆网、线粒体、闰盘等亚细胞结构出现损伤性改变,部分心肌纤维溶解、坏死,心肌间质水肿、出血、炎性细胞浸润;心肌线粒体MDA水平升高,SOD活性随睡眠剥夺时间延长有降低趋势。结论:睡眠剥夺能够引起大鼠心肌损伤,其引起的氧化应激可能是心肌损伤的机制。  相似文献

Carboxyatractylate (CAT) and atractylate inhibit the mitochondrial adenine nucleotide translocator (ANT) and stimulate the opening of permeability transition pore (PTP). Following pretreatment of mouse liver mitochondria with 5 microM CAT and 75 microM Ca2+, the activity of PTP increased, but addition of 2 mM ADP inhibited the swelling of mitochondria. Extramitochondrial Ca2+ concentration measured with Calcium-Green 5N evidenced that 2 mM ADP did not remarkably decrease the free Ca2+ but the release of Ca2+ from loaded mitochondria was stopped effectively after addition of 2 mM ADP. CAT caused a remarkable decrease of the maximum amount of calcium ions, which can be accumulated by mitochondria. Addition of 2 mM ADP after 5 microM CAT did not change the respiration, but increased the mitochondrial capacity for Ca2+ at more than five times. Bongkrekic acid (BA) had a biphasic effect on PT. In the first minutes 5 microM BA increased the stability of mitochondrial membrane followed by a pronounced opening of PTP too. BA abolished the action about of 1 mM ADP, but was not able to induce swelling of mitochondria in the presence of 2 mM ADP. We conclude that the outer side of inner mitochondrial membrane has a low affinity sensor for ADP, modifying the activity of PTP. The pathophysiological importance of this process could be an endogenous prevention of PT at conditions of energetic depression.  相似文献

The adenine nucleotide translocase (ANT) is a key component in maintaining cellular energy homeostasis, and has also been implicated in formation of the mitochondrial permeability transition pore. Human ANT-3 was cloned from a human heart cDNA library and expressed as a histidine-tagged fusion protein in the mitochondria of the Trichoplusia ni. cell line. Overexpression resulted in a concomitant decrease in the endogenous ANT content, allowing for the characterization of binding of known ANT ligands to the human protein. Binding affinities for bongkrekic acid (BKA), ADP, and atractyloside (ATR) were measured in mitochondria from the human ANT-3 expressing cell line, and compared to similar preparations from bovine heart mitochondria by use of a novel radioiodinated derivative of ATR. Binding to ANT-3 by the high affinity inhibitors BKA and ATR, as well as the lower affinity natural ligand ADP, was similar to that measured in bovine heart mitochondria, and to that previously reported for mammalian heart mitochondria. Characterizations such as these of human ANT isoforms may lead to drug development for enhanced mitochondrial function and cellular viability.  相似文献

Uncoupling proteins (UCPs) mediate fatty acid-induced proton cycling in mitochondria, which is stimulated by superoxide and inhibited by GDP. Fatty acid anions can also be transported by adenine nucleotide translocase (ANT), thus resulting in the uncoupling of oxidative phosphorylation. In the present work, an attempt was made to distinguish between the protonophoric activity of UCP3 and that of ANT using inhibition analysis. This study was carried out using mitochondria from skeletal muscles of hibernating Yakut ground squirrel, which have a significant level of UCP3 mRNA. We found that millimolar concentrations of GDP, which is considered to be a specific inhibitor of UCPs, slightly recoupled the mitochondrial respiration and restored the membrane potential. Addition of the specific ANT inhibitor CAT (carboxyatractylate), in micromolar concentration, prior to GDP prevented its recoupling effect. Moreover, GDP and ADP exhibited a competitive kinetic behavior with respect to ANT. In brown adipose tissue, CAT did not prevent the UCP1-iduced increase in chloride permeability and the inhibitory effect of GDP, thus confirming the inability of CAT to affect UCP1. These results allow us to conclude that the recoupling effect of purine nucleotides on skeletal muscle mitochondria of hibernating ground squirrels can be explained by interaction of the nucleotides with ANT, whereas UCP3 is not involved in the process.  相似文献

Morphological observations in some tissues indicate that dietary copper deficiency results in structural damage to mitochondria. The purpose of this study was to determine whether mitochondrial function is impaired as well. Male, weanling Sprague-Dawley rats were fed diets deficient or sufficient in copper for 4 weeks. Mitochondria were isolated from heart, liver, kidney cortex, and kidney medulla. P/O ratio, state 3 and state 4 respiration rates (oxygen consumed in the presence and absence of ADP, respectively), and acceptor control index (ratio of state 3:state 4) were determined using succinate or pyruvate/malate as substrate. State 3 respiration rate in mitochondria from copper-deficient hearts and livers was lower than in mitochondria from copper-sufficient hearts. Copper deficiency reduced the state 4 respiration rate only in cardiac mitochondria. Neither respiration rate was affected by copper deficiency in mitochondria from kidney medulla or cortex. P/O ratio was not significantly affected by copper deficiency in any tissue examined. Acceptor control index was reduced only in liver mitochondria. The observed decreases in respiration rates are consistent with decreased cytochrome c oxidase activity, shown by others to occur in mitochondria isolated from hearts and livers of copper-deficient rats.  相似文献

The effect of acute hypoxia on adenine nucleotides, glutamate, aspartate, alanine and respiration of heart mitochondria was studied in rats. The losses of intramitochondrial adenine nucleotides (ATP+ADP+AMP) during hypoxia were related to depression of state 3 respiration supported by glutamate and malate, as well as decrease in uncoupled respiration. Hypoxia had less prominent effect on succinate-dependent state 3 respiration. Non-phosphorylating (state 4) respiratory rates and ADP/O ratios were slightly affected by oxygen deprivation. Glutamate fall in tissue and mitochondria of hypoxic hearts was concomitant with significant increase in tissue alanine and mitochondrial aspartate. The losses of intramitochondrial ATP and respiratory activity with NAD-dependent substrates during hypoxia were related to a decrease in mitochondrial glutamate. The results suggest that hypoxia-induced impairment of complex I of respiratory chain and a loss of glutamate from the matrix may limit energy-producing capacity of heart mitochondria.  相似文献

Mitochondria were isolated from etiolated corn shoots (Zea mays L.) that were stressed to a measured water potential. The rates of mitochondrial respiration in state III, state IV, and without phosphate or ADP on a milligram protein basis decreased as water stress increased with succinate, malatepyruvate, or reduced nicotinamide adenine dinucleotide as substrates. Coupling (as determined by respiratory control and ADP/O ratios) did not decrease with increasing water stress. At water potentials greater than −35 bars all respiration had ceased.  相似文献

It has been shown that KM values for ADP when rat liver mitochondria oxidized succinate were strictly dependent on the values of the respiratory control ratios. The Ki values for palmitoyl-CoA inhibition of the ADP-stimulated succinate oxidation and the inhibition of the uncoupler-stimulated ATPase activity were equal to 0.5 muM. Mitochondria from livers of starved rats showed 30% inhibition of the state 3 respiratory rate (compared to the uncoupled respiratory rate) which was abolished by addition of carnitine. It was supposed that this inhibition was due to the influence of acyl-CoAs bound to the inner mitochondrial membrane on the adeninenucleotide translocase. Mitochondria from livers of fed rats showed a strong inhibition of succinate oxidation both in state 4 and state 3, although the rate of uncoupled respiration was normal. It was assumed that in this case the changes in mitochondrial behaviour was caused by the decrease in the concentration of ADP and ATP in the matrix space of mitochondria.  相似文献

Using Percoll density gradient centrifugation, free (nonsynaptosomal) mitochondria were isolated from the dorsal-lateral striatum and paramedian neocortex of rats during complete forebrain ischemia and reperfusion. Mitochondria prepared from either region after 30 min of ischemia showed decreased state 3 (ADP and substrate present) and uncoupled respiration rates (19-45% reductions) with pyruvate plus malate as substrates, whereas state 4 respiration (no ADP present) was preserved. At 6 h of recirculation, state 3 and uncoupled respiration rates for mitochondria from the paramedian neocortex (a region resistant to ischemic damage) were similar to or even increased compared with control values. By contrast, in mitochondria from the dorsal-lateral striatum (a region containing neurons susceptible to global ischemia), decreases in state 3 and uncoupled respiration rates (25 and 30% less than control values) were again observed after 6 h of recirculation. With succinate as respiratory substrate, however, no significant differences from control values were found in either region at this time point. By 24 h of recirculation, respiratory activity with either pyruvate plus malate or succinate was greatly reduced in samples from the dorsal-lateral striatum, probably reflecting complete loss of function in some organelles. In contrast with these marked changes in free mitochondria, the respiratory properties of synaptosomal mitochondria, assessed from measurements in unfractionated homogenates, were unchanged from controls in the dorsal-lateral striatum at each of the time points studied, but showed reductions (19-22%) during ischemia and after 24 h of recirculation in the paramedian neocortex.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The effects of fluoride on respiration of plant tissue and mitochondria were investigated. Fumigation of young soybean plants (Glycine max Merr. cv. Hawkeye) with 9–12 μg × m?3 HF caused a stimulation of respiration at about 2 days of treatment followed by inhibition 2 days later. Mitochondria isolated from the stimulated tissue had higher respiration rates, greater ATPase activity, and lower P/O ratios, while in mitochondria from inhibited tissue, all three were reduced. Treatment of etiolated soybean hypocotyl sections in Hoagland's solution containing KF for 3 to 10 h only resulted in inhibition of respiration. Mitochondria isolated from this tissue elicited increased respiration rates with malate as substrate and inhibited respiration with succinate. With both substrates respiratory control and ADP/O ratios were decreased. Direct treatment of mitochondria from the etiolated soybean hypocotyl tissue with fluoride resulted in inhibition of state 3 respiration and lower ADP/O ratios with the substrates succinate, malate, and NADH. Fluoride was also found to increase the amount of osmotically induced swelling and cause a more rapid leakage of protein with mitochondria isolated from etiolated corn shoots (Zea mays L. cv. Golden Cross Bantam). The results are discussed with respect to possible effects of fluoride on mitochondrial membranes.  相似文献

Increased O(2) metabolism resulting in chronic hypoxia is common in models of endstage renal disease. Mitochondrial uncoupling increases O(2) consumption but the ensuing reduction in mitochondrial membrane potential may limit excessive oxidative stress. The present study addressed the hypothesis that mitochondrial uncoupling regulates mitochondria function and oxidative stress in the diabetic kidney. Isolated mitochondria from kidney cortex of control and streptozotocin-induced diabetic rats were studied before and after siRNA knockdown of uncoupling protein-2 (UCP-2). Diabetes resulted in increased UCP-2 protein expression and UCP-2-mediated uncoupling, but normal mitochondria membrane potential. This uncoupling was inhibited by GDP, which also increased the membrane potential. siRNA reduced UCP-2 protein expression in controls and diabetics (-30-50%), but paradoxically further increased uncoupling and markedly reduced the membrane potential. This siRNA mediated uncoupling was unaffected by GDP but was blocked by ADP and carboxyatractylate (CAT). Mitochondria membrane potential after UCP-2 siRNA was unaffected by GDP but increased by CAT. This demonstrated that further increased mitochondria uncoupling after siRNA towards UCP-2 is mediated through the adenine nucleotide transporter (ANT). The increased oxidative stress in the diabetic kidney, manifested as increased thiobarbituric acids, was reduced by knocking down UCP-2 whereas whole-body oxidative stress, manifested as increased circulating malondialdehyde, remained unaffected. All parameters investigated were unaffected by scrambled siRNA. In conclusion, mitochondrial uncoupling via UCP-2 regulates mitochondria membrane potential in diabetes. However, blockade of the diabetes-induced upregulation of UCP- 2 results in excessive uncoupling and reduced oxidative stress in the kidney via activation of ANT.  相似文献
