We studied the effects of light generated by LEDs on the growth of Tsururindo (Tripterospermum japonicum) shoots. Apical shoots (2–3 cm long) were cultured on MS basal media supplemented with 3% sucrose, and were maintained for four weeks under five different light qualities: F (fluorescent lamp), red LED (R), 70% red + 30% blue LED (R7B3), 50% red + 50% blue (R5B5), or blue LED (B). Rooting was promoted by red light (100%) but was inhibited by blue light. Plant growth, as defined by root number, fresh weight, and chlorophyll content, was generally healthier for cultures irradiated with mixed LEDs, and was the best under R7B3. Ventilation resulted in more rapid apical shoot growth and rooting compared with control plants, when both were treated with the R7B3 system. We demonstrated here that plant growth can be controlled by using LEDs to adjust for the most effective irradiation conditions, compared with the performance observed when conventional fluorescent lamps are utilized.

Recombinant Rhodopseudomonas palustris, harboring the carotenoid-metabolizing gene crtI (CrtIBS), and whose color changes from greenish yellow to red in response to inorganic As(III), was cultured in transparent microplate wells illuminated with a light emitting diode (LED) array. The cells were seen to grow better under near-infrared light, when compared with cells illuminated with blue or green LEDs. The absorbance ratio of 525 to 425 nm after cultivation for 24 h, which reflects red carotenoid accumulation, increased with an increase in As(III) concentrations. The detection limit of cultures illuminated with near-infrared LED was 5 microgram/l, which was equivalent to that of cultures in test tubes illuminated with an incandescent lamp. A near-infrared LED array, in combination with a microplate, enabled the simultaneous handling of multiple cultures, including CrtIBS and a control strain, for normalization by the illumination of those with equal photon flux densities. Thus, the introduction of a near-infrared LED array to the assay is advantageous for the monitoring of arsenic in natural water samples that may contain a number of unknown factors and, therefore, need normalization of the reporter event.

The effects of light generated by monochromic blue, red or mixed radiation from a fluorescent lamp (FL) with light emitting diodes (LEDs) (blue, red, or far-red) on growth and morphogenesis of marigold and salvia seedlings were investigated and the responses compared with those of plantlets grown under a broad spectrum conventional fluorescent lamp (a 16 h photoperiod per day). Dry weight of marigold seedlings was significantly increased in monochromic red light (R), fluorescent light plus red LED (FLR) or fluorescent light (FL) but reduced when monochromic blue light (B) was used, whereas in salvia dry weight was significantly greater under fluorescent light plus blue LED (FLB), fluorescent light plus red LED (FLR) and fluorescent light plus far-red LED (FLFr) as compared to other treatments. Stem length in marigold was greatest in monochromic blue light, being three times greater than in FLR or FL treatments. In salvia, FLFr increased stem length but this was significantly decreased by R as compared to other treatments. The number of visible flower buds in marigold was much higher in FLR as well as in the control (FL), and it was about five times greater than in B or R. However, the number of open flowers in salvia varied slightly in all the treatments. Different light qualities also influenced the duration of the blooming period in both the species. No flower buds were formed when monochromic B or R was used in salvia and FLFr inhibited flower bud formation in marigold. In comparison with monochromic blue or red light, the number of stomata was greater in mixed radiation of FL with LEDs in both the plants. Our study demonstrates the effectiveness of a LED system for plantlet growth and morphogenesis in space-based plant research chambers.

Flatbed scanners are the most frequently used reading instrument for radiochromic film dosimetry because its low cost, high spatial resolution, among other advantages. These scanners use a fluorescent lamp and a CCD array as light source and detector, respectively. Recently, manufacturers of flatbed scanners replaced the fluorescent lamp by light emission diodes (LED) as a light source. The goal of this work is to evaluate the performance of a commercial flatbed scanner with LED based source light for radiochromic film dosimetry. Film read out consistency, response uniformity, film-scanner sensitivity, long term stability and total dose uncertainty was evaluated. In overall, the performance of the LED flatbed scanner is comparable to that of a cold cathode fluorescent lamp (CCFL). There are important spectral differences between LED and CCFL lamps that results in a higher sensitivity of the LED scanner in the green channel. Total dose uncertainty, film response reproducibility and long-term stability of LED scanner are slightly better than those of the CCFL. However, the LED based scanner has a strong non-uniform response, up to 9%, that must be adequately corrected for radiotherapy dosimetry QA. The differences in light emission spectra between LED and CCFL lamps and its potential impact on film-scanner sensitivity suggest that the design of a dedicated flat-bed scanner with LEDs may improve sensitivity and dose uncertainty in radiochromic film dosimetry.

The effect of light quality on protocorm-like bodies (PLBs) of Dendrobium officinale was investigated. PLBs of D. officinale were incubated under a number of different light conditions in vitro, namely: dark conditions; fluorescent white light (Fw); red light-emitting diodes (LEDs); blue LEDs; half red plus half blue [RB (1:1)] LEDs; 67% red plus 33% blue [RB (2:1)] LEDs; and 33% red plus 67% blue [RB (1:2)] LEDs. Growth parameters, number of shoots produced per PLB, chlorophyll concentration and carotenoid concentration were measured after 90 days culture. The percentage of PLBs producing shoots was 85% under blue LEDs. In contrast, the percentage of PLBs producing shoots was less than 60% under dark conditions, fluorescent white light and red LEDs. The number of shoots produced per PLB was more than 1.5 times greater under blue LEDs, RB (1:1) LEDs and RB (1:2) LEDs than those cultured under other light treatments [dark, Fw, red LEDs and RB (2:1)]. Chlorophyll and carotenoid concentrations were significantly higher under blue LEDs and different red plus blue LED ratios, compared to other light treatments (dark, Fw and red LEDs). Blue LEDs, Fw, and RB (1:2) LEDs produced higher dry matter accumulations of PLBs and shoots. This study suggests that blue LEDs or RB (1:2) LEDs could significantly promote the production of shoots by protocorm-like bodies of D. officinale and increase the dry matter of PLBs and the accumulation of shoot dry matter in vitro.

In the modern view of synaptic transmission, astrocytes are no longer confined to the role of merely supportive cells. Although they do not generate action potentials, they nonetheless exhibit electrical activity and can influence surrounding neurons through gliotransmitter release. In this work, we explored whether optogenetic activation of glial cells could act as an amplification mechanism to optical neural stimulation via gliotransmission to the neural network. We studied the modulation of gliotransmission by selective photo-activation of channelrhodopsin-2 (ChR2) and by means of a matrix of individually addressable super-bright microLEDs (μLEDs) with an excitation peak at 470 nm. We combined Ca2+ imaging techniques and concurrent patch-clamp electrophysiology to obtain subsequent glia/neural activity. First, we tested the μLEDs efficacy in stimulating ChR2-transfected astrocyte. ChR2-induced astrocytic current did not desensitize overtime, and was linearly increased and prolonged by increasing μLED irradiance in terms of intensity and surface illumination. Subsequently, ChR2 astrocytic stimulation by broad-field LED illumination with the same spectral profile, increased both glial cells and neuronal calcium transient frequency and sEPSCs suggesting that few ChR2-transfected astrocytes were able to excite surrounding not-ChR2-transfected astrocytes and neurons. Finally, by using the μLEDs array to selectively light stimulate ChR2 positive astrocytes we were able to increase the synaptic activity of single neurons surrounding it. In conclusion, ChR2-transfected astrocytes and μLEDs system were shown to be an amplifier of synaptic activity in mixed corticalneuronal and glial cells culture.

The influence of light quality on growth and development of in vitro grown Doritaenopsis hort. (Orchidaceae) plants was investigated. Growth parameters like leaf and root fresh/dry mass and leaf area were highest with plants grown under red plus blue light emitting diodes (LEDs). Leaf length was greater with the plants grown under red LED. Carbohydrate (starch, sucrose, glucose and fructose) and leaf pigment (chlorophylls and carotenoids) biosynthesis of the plants was significantly increased in plants grown under red plus blue LEDs compared to red or blue LED and fluorescent light treatments. This study suggests that the production of quality Doritaenopsis plants is possible by culturing the plants in vitro under a mixture of blue plus red light sources.

Light-emitting diodes (LEDs) are a promising technology with a potential to improve the irradiance efficiency, light quality, and the light spectrum for increasing plant yield and quality. In this experiment, we investigated the impacts of various LED light qualities, including 100% red, 100% blue, 70% red + 30% blue, and 100% white, on the growth and photosynthesis, phytochemical contents, and mineral element concentrations in lettuce (Lactuca sativa L. cv. ‘Grizzly’) in comparison to normal greenhouse conditions. Photon flux of 300 µmol m?2 s?1 was provided for 14 h by 120 LEDs set on a 60 cm × 60 cm sheet of aluminum platform in the growth chambers, where plants were grown for 60 d. Fresh mass per plant was significantly higher when grown under 100% blue and 70% red + 30% blue LEDs compared to the other environments including greenhouse conditions. Phytochemical concentrations and a nutritive value of lettuce were also significantly affected by the light treatments. Chlorophyll and carotenoid concentrations increased in the plants grown under 70% red + 30% blue LEDs compared to those grown in the greenhouse. Vitamin C content was 2.25-fold higher in the plants grown under 100% blue LEDs compared to those grown in the greenhouse. Higher photosynthesis and maximal quantum yield of PSII photochemistry were also observed in the plants treated with LED lights. The application of LED light led to the elevated concentrations of macro-and micronutrients in lettuce possibly because of the direct effect of LED light and lower stress conditions in the growth chambers compared to the greenhouse. Although the mechanism of the changes in lettuce grown under LED is not well understood, the results of this study demonstrated that LED light could be used to enhance the growth and nutritional value of lettuce in indoor plant production facilities.

Visual traps like yellow sticky card traps are used for monitoring and control of the greenhouse whitefly (Trialeurodes vaporariorum). However, reflected intensity (brightness) and hence, attractiveness depend on the ambient light conditions, and the colour (wavelength) might not fit with the sensitivity of whitefly photoreceptors. The use of light emitting diodes (LEDs) is a promising approach to increase the attractiveness, specificity and adaptability of visual traps. We constructed LED‐based visual traps equipped with blue and green high‐power LEDs and ultraviolet (UV) standard LEDs according to the putative spectral sensitivities of the insects' photoreceptors. In a series of small‐scale choice and no‐choice recapture experiments, the factors time of day as well as light intensity and light quality (colour) of LED traps were studied in terms of attractiveness compared to yellow traps without LEDs. Green LED traps (517 nm peak wavelength) were comparably attractive in no‐choice experiments but clearly preferred over yellow traps in all choice experiments. The time of day had a clear effect on the flight activity of the whiteflies and thereby on the trapping success. Blue LEDs (474 nm) suppressed the attractiveness of the light traps when combined with green LEDs suggesting that a yet undetected photoreceptor, sensitive for blue light, and an inhibiting interaction with the green receptor, might exist in T. vaporariorum. In choice experiments between LED traps emitting green light only or in combination with UV (368 nm), the green‐UV combination was preferred. In no‐choice night‐time experiments, UV LEDs considerably increased whitefly flight activity and efficacy of trapping. Most likely, the reason for the modifying effect of UV is the stimulating influence on flight activity. In conclusion, it seems that the use of green LEDs alone or in combination with UV LEDs could be an innovative option for improving attractiveness of visual traps.

Current biodetection illumination technologies (laser, LED, tungsten lamp, etc.) are based on spot illumination with additional optics required when spatial excitation is required. Herein we describe a new approach of spatial illumination based on electroluminescence (EL) semiconductor strips available in several wavelengths, greatly simplifying the biosensor design by eliminating the need for additional optics. This work combines EL excitation with charge-coupled device (CCD) based detection (EL-CCD detector) of fluorescence for developing a simple portable detector for botulinum neurotoxin A (BoTN-A) activity analysis. A F?rster Resonance Energy Transfer (FRET) activity assay for BoTN-A was used to both characterize and optimize the EL-CCD detector. The system consists of two modules: (1) the detection module which houses the CCD camera and emission filters, and (2) the excitation and sample module, containing the EL strip, the excitation filter and the 9-well sample chip. The FRET activity assay used in this study utilized a FITC/DABCYL-SNAP-25 peptide substrate in which cleavage of the substrate by BoTN-A, or its light chain derivative (LcA), produced an increase in fluorescence emission. EL-CCD detector measured limits of detection (LODs) were similar to those measured using a standard fluorescent plate reader with valves between 0.625 and 1.25 nM (31-62 ng/ml) for LcA and 0.313 nM (45 ng/ml) for the full toxin, BoTN-A. As far as the authors are aware this is the first demonstration of phosphor-based EL strips being used for the spatial illumination/excitation of a surface, coupled with CCD for point of care detection.

LED lighting is predicted to constitute 70% of the outdoor and residential lighting markets by 2020. While the use of LEDs promotes energy and cost savings relative to traditional lighting technologies, little is known about the effects these broad‐spectrum “white” lights will have on wildlife, human health, animal welfare, and disease transmission. We conducted field experiments to compare the relative attractiveness of four commercially available “domestic” lights, one traditional (tungsten filament) and three modern (compact fluorescent, “cool‐white” LED and “warm‐white” LED), to aerial insects, particularly Diptera. We found that LEDs attracted significantly fewer insects than other light sources, but found no significant difference in attraction between the “cool‐” and “warm‐white” LEDs. Fewer flies were attracted to LEDs than alternate light sources, including fewer Culicoides midges (Diptera: Ceratopogonidae). Use of LEDs has the potential to mitigate disturbances to wildlife and occurrences of insect‐borne diseases relative to competing lighting technologies. However, we discuss the risks associated with broad‐spectrum lighting and net increases in lighting resulting from reduced costs of LED technology.

The effect of light quality (spectral quality) and photoperiod (day length) were studied on flowering of Cyclamen persicum cv. Dixie White. Light generated from light emitting diodes (LED) i.e. monochromatic blue (10 or 12 h per day), monochromatic red (10 or 12 h per day), blue plus red (10 or 12 h per day) and fluorescent lights were used in these studies. It was found that blue plus red LEDs improved flower induction in cyclamen, the number flower buds and open flowers being highest in the plants grown under blue plus red LED (10 h per day). Blue and red LEDs alone reduced the flowering response. Peduncle length (flower stalk length) and blooming period of flowers were also influenced by light qualities and photoperiod treatments. Peduncle length was 23.8 cm on plants grown under red LED (12 h per day) treatment but 14 cm on plants grown under fluorescent light. Blooming period of flowers grown under fluorescent light was 20 d, whereas it was 40 d with the plants grown under red LEDs (10 h per day). The results indicate that flowering and subsequent growth of cyclamen can be controlled by manipulating light quality and lighting period.

结果表明:不同光照下顶芽培养的植株高生长顺序为H_(FL)﹥H_(1RB)﹥H_S﹥H_(2RB)﹥H_(3RB)﹥H_D,茎段培养的腋芽高生长顺序为h_(FL)﹥h_S﹥h_D﹥h_(1RB)﹥h_(2RB)﹥h_(3RB)。

CdTe quantum dots, 3.28 nm in size, were synthesized using a one‐step method in an aqueous medium. The CdTe quantum dots were successfully employed as hybrid phosphors for white light‐emitting diode (LED) devices by combining them with yellow‐emitting YAG:Ce phosphor. The color‐rendering index value and International Commission on illumination coordinates for hybrid phosphor white LEDs were 75 and (x = 0.30, y = 0.29), respectively. Compared with conventional phosphors, semiconductor quantum dots have larger band gap energy and broader absorption features, and can be excited more efficiently by optical pumping sources. The results confirmed that the high color‐rendering index value of the white LED was due to the CdTe quantum dots introduced in the hybrid phosphor system. Proper illumination is essential for light microscopy. Whereas in early years incandescent light was the only illumination, today, more and more specialized light sources, such as lasers or arc lamps are used. Because of the high efficiency and brightness that light-emitting diodes (LED) have reached today, they have become a serious alternative for almost all kinds of illumination in light microscopy. LED have a high durability, do not need expensive electronics, and they can be switched in nanoseconds. Besides this, they are available throughout the UV/Vis/NIR-spectrum with a narrow bandwidth. This makes them ideal light sources for fluorescence microscopy. The white LED, with a color temperature ranging from 2,600 up to 5,000 K is an excellent choice for bright-field illumination with the additional advantage of simple brightness adjustments without changing the spectrum. This review discusses the different LED types, their use in the fluorescence microscope, and discusses LED as specialized illumination sources for F?rster resonance energy transfer and fluorescent lifetime imaging microscopy.

In the current work we demonstrate the relevance of monochromatic light conditions in moss plant cell culture. Light intensity and illumination wavelength are important cultivation parameters due to their impact on growth and chlorophyll formation kinetics of the moss Physcomitrella patens. This moss was chosen as a model organism due to its capability to produce complex recombinant pharmaceutical proteins. Filamentous moss cells were cultivated in mineral medium in shaking flasks. The flasks were illuminated by light emitting diodes (LED) providing nearly monochromatic red and blue light as well as white light as a reference. A maximum growth rate of 0.78 day((1) was achieved under additional CO(2) aeration and no growth inhibition was observed under high light illumination. The application of dual red and blue light is the most effective way to reach high growth and chlorophyll formation rates while minimizing energy consumption of the LEDs. These observations are discussed as effects of photo sensory pigments in the moss. The combination of monochromatic red and blue light should be considered when a large scale process is set up.

The cell‐cycle progression of Ulva compressa is diurnally gated at the G1 phase in accordance with light–dark cycles. The present study was designed to examine the spectral sensitivity of the G1 gating system. When blue, red, and green light‐emitting diodes (LEDs) were used for illumination either alone or in combination, the cells divided under all illumination conditions, suggesting that all colors of light were able to open the G1 gate. Although blue light was most effective to open the G1 gate, red light alone or green light alone was also able to open the G1 gate even at irradiance levels lower than the light compensation point of each color. Occurrence of a period of no cell division in the course of a day suggested that the G1 gating system normally functioned as under ordinary illumination by cool‐white fluorescent lamps. The rise of the proportion of blue light to green light resulted in increased growth rate. On the other hand, the growth rates did not vary regardless of the proportion of blue light to red light. These results indicate that the difference in growth rate due to light color resulted from the difference in photosynthetic efficiency of the colors of light. However, the growth rates significantly decreased under conditions without blue light. This result suggests that blue light mediates cell elongation and because the spectral sensitivity of the cell elongation regulating system was different from that of the G1 gating system, distinct photoreceptors are likely to mediate the two systems.

Light traps equipped with light emitting diodes (LEDs) have been applied to manage some phototactic insect pests. The diamondback moth, Plutella xylostella, is a cosmopolitan insect pest to be seriously harmful to many cruciferous plants. The present research focused on evaluating the phototactic behavior responses of the moths to several wavelengths and photon flux densities of LED lights under laboratory and field conditions. The results from the laboratory showed that the highest phototactic behavior responses of P. xylostella moths were recorded for UV (380 nm) LED light under 1.5 µmol m−2 s−1 and 2.5 µmol m−2 s−1. The moths were more attracted to light traps equipped with 4 LEDs and 6 LEDs of 380 nm, respectively, between 20:00 and 22:00 than the other groups and night times in the field. The finding from the field was consistent with the results from the laboratory. We found that the 380 nm LED light results in the strongest attraction rate of the moths by 92.4 % and the moths caught in light trap with the UV LEDs was 1.62 times more than that with a black light. These data clearly demonstrate that P. xylostella moths have a high sensitivity to 380 nm, therefore, a 380 nm LED light trap could be useful for monitoring and controlling the moths.

We have measured the critical flicker detection frequency (CFDF) and double pulse recognition threshold (DPT) using three LEDs with power peaks at 460, 525 and 625 nm for target illumination. Brightness equalization was performed by customized heterochromatic flicker photometry (cHFP). Reference luminance levels were 170 cd/m2 (blue LED, 60 subjects), 4 cd/m2 (green LED, 20 subjects), and 1 cd/m2 (green LED, 20 subjects). The measurement at 1 cd/m2 was preceded by 15 min of dark adaptation. The angle of view for the target was 3°, and the duration of stimuli was 1 ms. An experimental pulse generator with three channels and a projector was used. No differences in CFDF at different spectral properties of stimulus were observed at all three levels of luminance. Thus, it is concluded that temporal vision resolution does not depend on the spectral properties of visual stimuli.
