线粒体DNA(mtDNA)作为分子标记已被广泛应用于各物种系统发生的研究。mtDNA控制区序列(D-Loop)以其较高的突变积累对于研究物种种内的遗传分化具有重要价值。鱼类是脊椎动物中最原始但在种属数量上又最占优势的类群,其物种繁多,分布广泛,起源复杂,研究其系统发生历来是令人饶有兴趣的课题。D-Loop在研究鱼类种内遗传分化中具有多方面的重要意义。近年来,已有越来越多的研究工作将D-Loop作为分子标记来探讨各种鱼类的种内遗传分化,并且获得了许多有启发性的结果。青海湖是我国内陆最大的咸水湖,湖中主要鱼类为青海湖裸鲤(Gymnocypris przewalskii),D-Loop分析初步结果显示青海湖及其周围河流中的裸鲤似乎没有新的种内遗传分化现象。 相似文献

The vaquita, Phocoena sinus, is one of two critically endangered cetacean species, and is listed as an endangered species in both the United States and Mexico. These listings result from a small population size, estimated to be 224 animals, and a rapid decline in abundance (18% per annum) thought to be caused by human activities. To characterize the genetic composition of the vaquita, we have sequenced a portion of the mitochondrial DNA control region from 43 individuals collected between 1985 and 1993. All animals had identical sequences. While low genetic variability has been reported for cetacean species, this complete lack of polymorphism in the control region is unique. This result is concordant with the hypothesis that the evolutionary history of the species includes a bottleneck or founder event, possibly at species inception, followed by a small long-term effective population size. We recommend parallel studies of nuclear variability, which could provide understanding of the basic biology of the vaquita for use in conservation efforts. 相似文献

为全面了解青海湖裸鲤遗传多样性现状,采用水平淀粉凝胶电泳技术分析了青海湖裸鲤[样品采自青海湖的五条支流,分别为泉吉河(QJ)、沙柳河(SL)、哈尔盖河(HEG)、布哈河(BH)以及甘子河(GZ)]的13种同工酶,共记录26个位点,统计了基因频率,计算了其遗传多样性。结果表明:26个基因位点中多态位点6个,分别为Pgm、Hk、Cat、Gdh、Es-t1、S-Mdh,多态位点比例(P0.99)为23.08%,表明青海湖裸鲤的五个群体皆具有较高的遗传多样性水平;分别计算了五个群体的平均杂合度(H)为0.1161(HEG)、0.1178(GZ)、0.1304(SL)、0.1305(BH)、0.1287(QJ),多态位点比例(Ae)分别为1.3112(BH)、1.3069(SL)、1.2542(GZ)、1.2492(HEG)、1.2453(QJ);除泉吉河群体外,其它四个群体的遗传偏离指数均为负数,表明多数群体处于一定程度的杂合子缺失状态;计算了五个群体间的遗传距离以及遗传相似度,结果显示群体间遗传距离与遗传相似度与各个河流的地理位置具有很强的相关性;根据五个群体的遗传距离进行了聚类分析,通过构建系统树图将其基本分为两大种群,甘子河与哈尔盖河两群体为一类群,另外三个群体分为一个类群;计算群体间的基因流(Nm)为7.2013,说明河道之间具有较为广泛的基因交流,并计算了两两群体间的基因流,其中布哈河群体与其它群体间的基因交流都较为广泛。 相似文献

对盲囊线虫在青海湖中以锯缘真剑水蚤作为第一中间宿主,舌状绦虫和裂头绦虫都以咸水化镖水蚤和锯缘真剑水蚤作为第一中间宿主,青海湖裸鲤对咸水北镖水蚤的摄食量随体长增加而增加,对锯缘真剑水蚤摄食量随体长增加的变化则不明显。因而提出:舌状绦虫在不同大小的青海湖裸鲤中的种群数量变化不是由于宿主的摄食习性改变引起的。裂头绦虫和对盲囊线虫在青海湖裸鲤中的种群数量随宿主生长而增加的变化与较大宿主对中间宿主的较大摄取量以及它们在宿主体内的积累有关。 相似文献

The Mariana crow (Corvus kubaryi) is an endangered species that is restricted to the islands of Guam and Rota in the Mariana archipelago. Predation by the introduced brown tree snake (Boiga irregularis) has decimated bird populations on Guam, and the crow population there is the last wild remnant of the endemic forest avifauna. The population on Guam is critically endangered and, despite intensive management, the population has continued to decline. Additional management options include intermixing the Guam and Rota populations, but such options are best evaluated within a population genetics framework. We used three types of molecular markers to assay genetic variation in the Mariana crow: mitochondrial DNA (mtDNA) sequences, minisatellites and microsatellites. The two populations could be differentiated by mtDNA sequencing and they differed in allele frequencies at nuclear markers. Thus, the populations could be designated as evolutionarily significant units. However, the Guam population is genetically more diverse than the Rota population, and its survival probability if managed separately is very low. All markers did indicate that the two populations are closely related and separated by a shallow genealogical division. Intermixing the populations is justified by two rationales. First, the apparent population differences may result from recent human activities. Second, a greater amount of genetic information may be preserved by joint management. The translocation of birds from Rota to Guam has begun, but strategies that will ensure maintenance of the variation in the Guam population warrant further exploration. 相似文献

Cedrela lilloi C. DC. (cedro coya, Meliaceae), an important south American timber species, has been historically overexploited through selective logging in Argentine Yungas Rainforest. Management and conservation programs of the species require knowledge of its genetic variation patterns; however, no information is available. Molecular genetic variability of the species was characterized to identify high‐priority populations for conservation and domestication purposes. Fourteen native populations (160 individuals) along a latitudinal gradient and with different logging's intensities were assessed by 293 polymorphic AFLP (amplified fragment length polymorphism) markers. Genetic diversity was low (Ht = 0.135), according to marginal location of the species in Argentina. Most of the diversity was distributed within populations (87%). Northern populations showed significant higher genetic diversity (R2= 0.69) that agreed with latitudinal pattern of distribution of taxonomic diversity in the Yungas. Three clusters were identified by Bayesian analysis in correspondence with northern, central, and southern Yungas. An analysis of molecular variance (AMOVA) revealed significant genetic differences among latitudinal clusters even when logging (ΦRT = 0.07) and unlogging populations (ΦPT = 0.10) were separately analyzed. Loss of genetic diversity with increasing logging intensity was observed between neighboring populations with different disturbance (ΦPT = 0.03–0.10). Bottlenecks in disturbed populations are suggested as the main cause. Our results emphasize both: the necessity of maintaining the genetic diversity in protected areas that appear as possible long‐term refuges of the species; and to rescue for the national system of protected areas some high genetic diversity populations that are on private fields. 相似文献

Insertions and deletions (indels) result in sequences of various lengths when homologous gene regions are compared among individuals or species. Although indels are typically phylogenetically informative, occurrence and incorporation of these characters as gaps in intraspecific population genetic data sets are rarely discussed. Moreover, the impact of gaps on estimates of fixation indices, such as F(ST), has not been reviewed. Here, I summarize the occurrence and population genetic signal of indels among 60 published studies that involved alignments of multiple sequences from the mitochondrial DNA (mtDNA) control region of vertebrate taxa. Among 30 studies observing indels, an average of 12% of both variable and parsimony-informative sites were composed of these sites. There was no consistent trend between levels of population differentiation and the number of gap characters in a data block. Across all studies, the average influence on estimates of PhiST was small, explaining only an additional 1.8% of among population variance (range 0.0-8.0%). Studies most likely to observe an increase in PhiST with the inclusion of gap characters were those with < 20 variable sites, but a near equal number of studies with few variable sites did not show an increase. In contrast to studies at interspecific levels, the influence of indels for intraspecific population genetic analyses of control region DNA appears small, dependent upon total number of variable sites in the data block, and related to species-specific characteristics and the spatial distribution of mtDNA lineages that contain indels. 相似文献

Marbled lungfish Protopterus aethiopicus in Lake Victoria and two nearby smaller lakes were found to have high levels of DNA sequence variation in their mitochondrial control regions (35 haplotypes in 61 fish) but no population genetic structure (ΦST = 0·00). In contrast, marbled lungfish in Lake Baringo, Kenya, appeared to be fixed for a single control region haplotype, which occurred at low frequency in the other lakes. Using FLUCTUATE software, the female effective population size in Lake Victoria during the late Pleistocene was estimated to be c. 500 000, similar to the value estimated for the present-day population. These observations suggest that, during the late Pleistocene dry period, a large marbled lungfish population survived either in wet refugial areas within the lake basin or in surrounding areas. Marbled lungfish were reported to have been introduced into Lake Baringo 30 years ago with a founding population of only three individuals. The lack of control region variation in the Lake Baringo population is consistent with that situation. 相似文献

Habitat loss, fragmentation, overharvest, and other anthropogenic factors have resulted in population and distribution declines for North American wolverines (Gulo gulo). Currently, wolverines east of the Hudson Bay are endangered and possibly extinct, whereas the status of wolverines throughout the remaining Holarctic is vulnerable. Three previous studies using nuclear loci have detected little to no significant structuring among wolverines sampled across northern Canada. Based on these results it has been suggested that wolverines in northern Canada represent a single, panmictic population. However, as has been shown in numerous studies, in cases of female site fidelity, it is possible to have demographically autonomous populations even with male-biased gene flow. To better assess the genetic structure of wolverines in northern Canada, we examined nine microsatellite loci and DNA sequence variation from a 200 bp fragment of the mitochondrial (mtDNA) control region for 270 wolverines from nine collecting areas representing three regions of northern Canada. In agreement with previous studies, microsatellite analyses revealed a lack of significant population substructure (F
ST=0.0004). However, analysis of molecular variance, comparisons of pairwise F
ST values and nested-clade analysis of the mtDNA data revealed considerable genetic structuring among samples of wolverines from these three regions of northern Canada. These mitochondrial data provide evidence that wolverines in Canada are genetically structured due to female philopatry. The contrasting patterns of genetic differentiation based on nuclear and mitochondrial data highlight the importance of examining both nuclear and mitochondrial loci when attempting to elucidate patterns of genetic structure. 相似文献

The numbat has been reduced to two populations in Western Australia. To better understand the effects of range reduction on gene flow and genetic variation, and to address questions crucial for the species' management, we analysed mitochondrial DNA (mtDNA) sequences of free-ranging individuals and museum specimens. The results suggest recent connectivity between the remnant populations, although one of those may have lost significant amounts of genetic diversity during the recent population size reduction. We propose that for management purposes the remnant populations should be treated as a single historical lineage and that, subject to certain caveats, consideration should be given to population augmentation by translocation. 相似文献

to stocking programs as one way to combat the negative effect that human influence has had on population densities. The present
study surveyed the genetic structure of two Danube drainage populations at the Inn and Drau rivers, in Austria, on the basis
of mtDNA sequences. Data were placed in the context of the phylogeographic structure of European Thymallus and thus could reveal unexpected geographical mixing due to stocking with allochthonous individuals. Our analyses revealed
that regular stocking of fish not originating from their natal Rivers has left genetic traces in both systems surveyed. These
traces may be classified as marginal for the Inn river and its tributaries in which 97% of the graylings investigated carried
haplotypes belonging to the northern alpine lineage, corresponding to the region through which the Inn flows. In contrast,
the genetic composition of the Drau population, situated in the southern Alps, has been seriously altered through the stocking
of fish belonging to the northern alpine mtDNA lineage as only 62% of the fishes sampled carried haplotypes representing the

Island radiations can offer challenging systemsfor the implementation of conservation policiesbecause descendent populations may exhibitdifferent levels of adaptive divergence,reproductive isolation, and phylogeneticdistinctiveness. This seems particularly truefor the endangered Galápagos gianttortoises (Geochelone nigra), whichcomprise a lineage that radiated rapidly andconcomitantly with the evolution of thearchipelago. We used mitochondrial DNAsequences and microsatellite markers toinvestigate the genetic structure, and toreconstruct genealogical relationships and thehistory of population colonization of gianttortoises from the Islands of Santa Cruz andPinzón, including samples of a basal taxonfrom the Island of San Cristóbal.Populations displayed marked geneticdivergence, contrasting demographic histories,and deep phylogeographic structure. The patternof diversification among populations wasconsistent with geological and biogeographichistory, and to some extent, with adaptive andmorphological divergence. Results stronglyindicate the presence of a minimum of fourconservation units with long-standingevolutionary separation: two in Santa Cruz, onein Pinzón, and one in San Cristóbal. Wepropose that these findings be effectivelyintegrated with other existing data by theappropriate environmental agencies to evaluatecurrent conservation efforts and implement newstrategies aimed at protecting the integrityand diversity of giant tortoise populations. 相似文献

Polyphyly of mtDNA lineages in the Russian sturgeon, Acipenser gueldenstaedtii: forensic and evolutionary implications 总被引:1,自引:0,他引:1
Molecular species identification methods are an important component of CITES monitoring programs for trade in sturgeon and caviar. To date, obtaining molecular evidence for distinguishing caviar from four closely related Eurasian sturgeon species Acipenser baerii (Siberian sturgeon), A. gueldenstaedtii (osetra), A. persicus (Persian sturgeon), A. naccarii (Italian sturgeon) remains problematic. Using approximately 2.3 kb of mtDNA sequence data (cytochrome b, NADH5, control region), we find this to be attributable to the polyphyletic nature of these mitochondrial DNA markers in the Russian sturgeon, A. gueldenstaedtii. Two mitochondrial lineages are present within this species: one is phylogenetically affiliated with A. persicus and A. naccarii, while the other clusters with A. baerii. These findings have a direct impact on molecular testing of commercial caviar and demonstrate the necessity of using large sample sizes when constructing forensic databases. Furthermore, the results affect current taxonomic designations for these species as well as hypotheses concerning their evolutionary origins. 相似文献

通过分析81尾采自华南西部12条水系的中间黄颡鱼(Pelteobagrus intermedius)mtDNA控制区435 bp的序列,研究其群体遗传变异及亲缘生物地理格局。结果显示,中间黄颡鱼群体间的遗传分化较小,核苷酸变异只有0.54%。12条水系的群体共有7个单倍型,其中一些现已相互隔离的水系中的群体共享同一个单倍型,提示这些水系曾经有非常密切的联系。根据嵌套进化支序分析,中间黄颡鱼可能起源于峒中河、北仑河、防城河所在的广西与越南交界地区,并通过两条途径向华南沿海西部诸独立水系和海南岛扩散,在演化过程中,曾发生片断化事件,长距离建群和持续的分布区扩张。 相似文献

We used mitochondrial DNA control region sequences to examine phylogeography and population differentiation of the endangered Amazonian manatee Trichechus inunguis. We observe lack of molecular differentiation among localities and we find weak association between geographical and genetic distances. However, nested clade analysis supports restricted gene flow and/or dispersal with some long-distance dispersal. Although this species has a history of extensive hunting, genetic diversity and effective population sizes are relatively high when compared to the West Indian manatee Trichechus manatus. Patterns of mtDNA haplotype diversity in T. inunguis suggest a genetic disequilibrium most likely explained by demographic expansion resulting from secession of hunting and enforcement of conservation and protective measures. Phylogenetic analysis of T. manatus and T. inunguis haplotypes suggests that T. inunguis is nested within T. manatus, effectively making T. manatus a paraphyletic entity. Paraphyly of T. manatus and recent divergence times of T. inunguis and the three main T. manatus lineages suggest a possible need for a taxonomic re-evaluation of the western Atlantic Trichechus. 相似文献

The yellowfin seabream Acanthopagrus latus is widely distributed throughout the Indo‐West Pacific. The genetic analyses of mtDNA control region sequence variation in samples from Chinese waters revealed a pattern of genetic structure between southern and northern samples (P < 0·05) and high levels of genetic diversity in this species. Significant isolation by distance between southern and northern locations (Mantel test r = 0·35, P = 0·04) and among northern populations (r = 0·55, P = 0·01) indicated that ocean straits and other barriers substantially limit gene flow. Phylogeographical analysis also revealed two major divergent mtDNA lineages across Chinese waters. For conservation, A. latus populations in Chinese waters should be divided into at least two management units for protection. 相似文献

Hypochaeris salzmanniana DC. (Asteraceae, Lactuceae) is an endangered species on the Iberian Peninsula, known from only eight coastal populations. Most authors have treated it as a variety, subspecies or simply as a synonym of H. glabra L. On the basis of morphological and cytological characters, Talavera recently separated H. salzmanniana (2n = 8) from H. glabra (2n = 10). Material of H. salzmanniana, H. glabra and H. radicata was collected from Spain, Italy, Sicily and Tunisia in order to assess taxonomic status and population relationships. Amplified Fragment Length Polymorphism (AFLP) analysis revealed three well-differentiated species. A close relationship between H. salzmanniana and H. radicata is also confirmed by AFLP analysis and chromosome number (2n = 8), morphology, and rDNA localization (FISH, fluorescence in situ hybridization). Hypochaeris salzmanniana and H. radicata share three fixed diagnostic AFLP fragments out of 348 fragments scored. The population structure of H. salzmanniana reveals distinct groups in southern Spain that are separated geographically. High differentiation among a western (Conil to Zahara), an intermediate (Punta Paloma and Los Algarbes) and an eastern (Algeciras and La Línea) group may reflect ancient separation. Population sizes and genetic compatibility differ greatly among populations and can be used to explain levels of within-population genetic diversity, together with recent documented loss of habitats resulting from tourist developments. Population structures of H. radicata and H. glabra show a similar geographical patterning: strongly differentiated populations from the Betic Cordillera and from the Iberian Massif, which are separated at present by the Guadalquivir river. Geological events at the end of the Tertiary (Tortonian–Messinian Miocene) might help explain patterns of differentiation in these three species of sect. MtDNA sequence data ( c . 2100 bp of complete D‐loop + cytochrome b gene regions) showed a principal phylogenetic split within common carp Cyprinus carpio between the Lake Biwa wild form ( n = 3) and 'Eurasian' wild ( n = 18) and domesticated ( n = 35) forms. This supports the idea of the ancient origin of the Lake Biwa wild form, as well as the East Asian origin of wild common carp, previously proposed on the basis of fossil data. In addition, a possible multiple origin of koi carp was indicated by the polyphyletic distribution of five mtDNA haplotypes of koi carp within the 'Eurasian' clade. 相似文献

Phylogeographic studies from western Palaearctic have generally focused on species able to disperse and track their emerging suitable habitats after the last ice age. However, data on species whose biogeographical histories differ from this bulk of Palaearctic fauna are scarce. This is clearly the case of some specialized organisms inhabiting inland hypersaline environments, which are likely to have had a wider distribution range during the late Tertiary and may have persisted through the Pleistocene to the present day only constituting relict populations. In this study, we use partial sequences from two mitochondrial genes [16S rRNA (16S) and cytochrome oxidase subunit II (COII)] to investigate the phylogeography of the Iberian populations of Mioscirtus wagneri (Orthoptera: Acrididae), a specialized grasshopper exclusively inhabiting hypersaline low grounds. Our results show that M. wagneri exhibits a marked phylogeographical structure, forming three main clades which correspond with populations located in north-east, central–south-east and south-west Iberia. These geographical areas did not share any haplotype, indicating that gene flow between them is absent. Nested clade analyses revealed that these lineages have probably evolved in allopatry and data on sequence divergence suggest population fragmentation from the Early Pleistocene. Overall, these results provide a broader perspective on the contribution of historical climate/geological events to biogeographical patterns of organisms currently forming relict populations of great conservation concern. © 2009 The Linnean Society of London, Biological Journal of the Linnean Society , 2009, 97 , 623–633. 相似文献
