In the present study, we investigated whether growth and main nutrient ion concentrations of cabbage (Brassica campestris L.) could be increased when plants were subjected to different NH4^+/NO3- ratios. Cabbage seedlings were grown in a greenhouse in nutrient solutions with five NH4^+/NO3- ratios (1:0; 0.75:0.25; 0.5:0.5; 0.25:0.75; and 0:1). The results showed that cabbage growth was reduced by 87% when the proportion of NH4^+-N in the nutrient solution was more than 75% compared with a ratio NH4^+/NO3- of 0.5:0.5 35 d after transplanting, suggesting a possible toxicity due to the accumulation of a large amount of free ammonia in the leaves. When the NH4+/NO3- ratio was 0.5:0.5, fresh seedling weight, root length, and H2PO4- (P), K^+, Ca^2+, and Mg^2+ concentrations were all higher than those in plants grown under other NH4^+/NO3- ratios. The nitrate concentration in the leaves was the lowest in plants grown at 0.5: 0.5 NH4^+/NO3-. The present results indicate that an appropriate NH4^+/NO3- ratio improves the absorption of other nutrients and maintains a suitable proportion of N assimilation and storage that should benefit plant growth and the quality of cabbage as a vegetable.  相似文献

Carbon isotope discrimination in C3–C4 intermediates is determined by fractionations during diffusion and the biochemical fractionations occurring during CO2 fixation. These biochemical fractionations in turn depend on the fractionation by Rubisco in the mesophyll, the amount of CO2 fixation. These biochemical fractionations in turn depend on the fractionation by Rubisco in the mesophyll, the amount of CO2 fixation occurring in the bundle sheath, the extent of bundle-sheath leakiness and the contribution which C4-cycle activity makes to the CO2 pool there. In most instances, carbon isotope discrimination in C3–C4 intermediates is C3-like because only a small fraction of the total carbon fixed is fixed in the bundle sheath. In particular, this must be the case for Flaveria intermediates which initially fix substantial amounts of CO2 into C4-acids. In C3–C4 intermediates that refix photorespiratory CO2 alone, it is possible for carbon isotope discrimination to be greater than in C3-species, particularly at low CO2 pressures or at high leaf temperatures. Short-term measurements of carbon isotope discrimination and gas exchange of leaves can be used to study the photosynthetic pathways of C3-C4 intermediates and their hybrids as has recently been done for C3 and C4 species.  相似文献

Plants with the C4 photosynthetic pathway have predominantly one of three decarboxylation enzymes in their bundle sheath cells. Within the grass family (Poaceae) bundle sheath leakiness to CO2 is purported to be lowest in the nicotinamide adenine dinucleotide phosphate-malic enzyme (NADP-ME, EC 1.1.1.40) group, highest in the NAD-ME (EC 1.1.1.39) group and intermediate in the phosphoenolpyruvate carboxykinase (PCK, EC 4.1.1.32) group. We investigated the hypothesis that growth and photosynthesis of NAD-ME C4 grasses would respond more to elevated CO2 treatment than NADP-ME grasses. Plants were grown in 8-1 pots in growth chambers with ample water and fertilizer for 39 days at a continuous CO2 concentration of either 350 or 700 µl l?1. NAD-ME species included Bouteloua gracilis Lag. ex Steud (Blue grama), Buchloe dactyloides (Nutt.) Engelm. (Buffalo grass) and Panicum virgatum L. (Switchgrass) and the NADP-ME species were Andropogon gerardii Vittman (Big bluestem), Schizachyrium scoparium (Michx.) Nash (Little bluestem), and Sorghastrum nutans (L.) Nash (Indian grass). Contrary to our hypothesis, growth of the NADP-ME grasses was generally greater under elevated CO2 (significant for A. gerardii and S. nutans), while none of the NAD-ME grasses had a significant growth response. Increased leaf total non-structural carbohydrate (TNC) was associated with greater growth responses of NADP-ME grasses. Decreased leaf nitrogen in NADP-ME species grown at elevated CO2 was found to be an artifact of TNC dilution. Assimilation (A) vs intercellular CO2 (Ci) curves revealed that leaf photosynthesis was not saturated at 350 µl l?1 CO2 in any of these C4 grasses. Assimilation of elevated CO2-grown A. gerardii was higher than in plants grown in ambient CO2. In contrast, B. gracilis grown in elevated CO2 displayed lower A, a trait more commonly reported in C3 plants. Photosynthetic acclimation in B. gracilis was not related to leaf TNC or nitrogen concentrations, but A:Ci curves suggest a reduction in activity of both phosphoenolpyruvate (PEP) carboxylase (EC 4.1.1.31) and ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco, EC 4.1.1.39). Some adaptation of stomatal functioning was also seen in B. gracilis and A. gerardii leaves grown in elevated CO2. Our study shows that C4 grasses have the capacity for increased growth and photosynthesis under elevated CO2 even when water and nutrients are non-limiting. While it was the NADP-ME species which had significant responses in the present study, we have previously reported significant growth increases in elevated CO2 for B. gracilis.  相似文献

Epileptic foci are associated with locally reduced taurine (2-aminoethanesulfonic acid) concentration and Na+, K+-ATPase (EC 3.6.1.3) specific activity. Topically applied and intraperitoneally administered taurine can prevent the development and/or spread of foci in many animal models. Taurine has been implicated as a possible cytosolic modulator of monovalent ion distribution, cytosolic “free” calcium activity, and neuronal excitability. Taurine may act in part by modulating Na+, K+-ATPase activity of neuronal and glial cells. We characterized the requirements for in vitro modulation of Na+, K+-ATPase by taurine. Normal whole brain homogenate Na+, K+-ATPase activity is 5.1 ± 0.4 (4) μmol Pi± h?1± mg?1 Lowry protein. Partial purification of the plasma membrane fraction to remove cytosolic proteins and extrinsic proteins and to uncouple cholinergic receptors yields a membrane-bound Na+, K+-ATPase activity of 204.6 ± 5.8 (4) mol Pi± h?1± mg?1 Lowry protein. Taurine activates the Na+, K+-ATPase at all levels of purification. The concentration dependence of activation follows normal saturation kinetics (K1/2= 39 mM taurine, activation maximum =+87%). The activation exhibits chemical specificity among the taurine analogues and metabolites: taurine = isethionic acid > hypotaurine > no activation =β-alanine = methionine = choline = leucine. Taurine can act as an endogenous activator/modulator of Na+, K+-ATPase. Its action is mediated by a membrane-bound protein.  相似文献

The stimulation of dicotyledonous leaf growth by light depends on increased H+ efflux, to acidify and loosen the cell walls, and is enhanced by K+ uptake. The role of K+ is generally considered to be osmotic for turgor maintenance. In coleoptiles, auxin‐induced cell elongation and wall acidification depend on K+ uptake through tetraethylammonium (TEA)‐sensitive channels (Claussen et al., Planta 201, 227–234, 1997), and auxin stimulates the expression of inward‐rectifying K+ channels ( Philippar et al. 1999) . The role of K+ in growing, leaf mesophyll cells has been investigated in the present study by measuring the consequences of blocking K+ uptake on several growth‐related processes, including solute accumulation, apoplast acidification, and membrane polarization. The results show that light‐stimulated growth and wall acidification of young tobacco leaves is dependent on K+ uptake. Light‐stimulated growth is enhanced three‐fold over dark levels with increasing external K+, and this effect is blocked by the K+ channel blockers, TEA, Ba++ and Cs+. Incubation in 10 mm TEA reduced light‐stimulated growth and K+ uptake by 85%, and completely inhibited light‐stimulated wall acidification and membrane polarization. Although K+ uptake is significantly reduced in the presence of TEA, solute accumulation is increased. We suggest that the primary role of K+ in light‐stimulated leaf growth is to provide electrical counterbalance to H+ efflux, rather than to contribute to solute accumulation and turgor maintenance.  相似文献

We evaluated the influences of CO2[Control, ~ 370 µ mol mol ? 1; 200 µ mol mol ? 1 above ambient applied by free‐air CO2 enrichment (FACE)] and soil water (Wet, Dry) on above‐ and below‐ground responses of C3 (cotton, Gossypium hirsutum) and C4 (sorghum, Sorghum bicolor) plants in monocultures and two density mixtures. In monocultures, CO2 enrichment increased height, leaf area, above‐ground biomass and reproductive output of cotton, but not sorghum, and was independent of soil water treatment. In mixtures, cotton, but not sorghum, above‐ground biomass and height were generally reduced compared to monocultures, across both CO2 and soil water treatments. Density did not affect individual plant responses of either cotton or sorghum across the other treatments. Total (cotton + sorghum) leaf area and above‐ground biomass in low‐density mixtures were similar between CO2 treatments, but increased by 17–21% with FACE in high‐density mixtures, due to a 121% enhancement of cotton leaf area and a 276% increase in biomass under the FACE treatment. Total root biomass in the upper 1.2 m of the soil was not influenced by CO2 or by soil water in monoculture or mixtures; however, under dry conditions we observed significantly more roots at lower soil depths ( > 45 cm). Sorghum roots comprised 81–85% of the total roots in the low‐density mixture and 58–73% in the high‐density mixture. CO2‐enrichment partly offset negative effects of interspecific competition on cotton in both low‐ and high‐density mixtures by increasing above‐ground biomass, with a greater relative increase in the high‐density mixture. As a consequence, CO2‐enrichment increased total above‐ground yield of the mixture at high density. Individual plant responses to CO2 enrichment in global change models that evaluate mixed plant communities should be adjusted to incorporate feedbacks for interspecific competition. Future field studies in natural ecosystems should address the role that a CO2‐mediated increase in C3 growth may have on subsequent vegetation change.  相似文献

Abstract: The Na+ sensitivity of whole brain membrane Na+,K+-ATPase isoenzymes was studied using the differential inhibitory effect of ouabain (α1, low affinity for ouabain; α2, high affinity; and α3, very high affinity). At 100 m M Na+, we found that the proportion of isoforms with low, high, and very high ouabain affinity was 21, 38, and 41%, respectively. Using two ouabain concentrations (10−5 and 10−7 M ), we were able to discriminate Na+ sensitivity of Na+, K+-ATPase isoenzymes using nonlinear regression. The ouabain low-affinity isoform, α1, exhibited high Na+ sensitivity [ K a of 3.88 ± 0.25 m M Na+ and a Hill coefficient ( n ) of 1.98 ± 0.13]; the ouabain high-affinity isoform, α2, had two Na+ sensitivities, a high ( K a of 4.98 ± 0.2 m M Na+ and n of 1.34 ± 0.10) and a low ( K a of 28 ± 0.5 m M Na+ and an n of 1.92 ± 0.18) Na+ sensitivity activated above a thresh old (22 ± 0.3 m M Na+); and the ouabain very-high-affinity isoform, α3, was resolved by two processes and appears to have two Na+ sensitivities (apparent K a values of 3.5 and 20 m M Na+). We show that Na+ dependence in the absence of ouabain is the result of at least of five Na+ reactivities. This molecular functional characteristic of isoenzymes in membranes could explain the diversity of physiological roles attributed to isoenzymes.  相似文献

Abstract Shifts in ?13C of the graminaceous C3 halophyte Puccinellia nuttalliana (Schultes) Hitch. can be induced by salinization. To investigate this phenomenon, three approaches were taken: assay of carboxylases, CO2-enrichment studies, and gas exchange analysis. Although ribulose-1,5-bisphosphate carboxylase activity decreased with salinity, phosphoenolpyruvate carboxylase activity did not increase and its levels were not atypical of C3 plants. When plants were grown at four NaCl concentrations under atmospheres of 310 and 1300 cm3 m?3 CO2, the CO2-enrichment enhanced the effects of salinity on ?13C. This is consistent with a biophysical explanation for salt-induced shifts in ?13C, whereby there is a steepening of the CO2 diffusion gradient into the leaf. Gas exchange analysis indicated that intercellular CO2 concentrations were depressed in the leaves of salt-affected plants. This resulted from a greatly decreased stomatal conductance coupled with only small effects on intrinsic photosynthetic capacity. Water-use efficiency was enhanced.  相似文献

Various Ionic and molecular activities in the extraceUular environment are vital to plant cell physiological processes. A noninvasive microsensing system (NMS) based on either the scanning ion-selective electrode technique (SIET) or the scanning polarographlc electrode technique (SPET) is able to obtain information regarding the transportation of various Ions/molecules in Intact samples under normal physiological conditions. The two-probe simultaneous test system (2STS) Is an Integrated system composed of SIET, SPET, and a Xu-Kunkel sampling protocol. In the present study, 2STS was able to simultaneously measure fluxes of H^+ and O2 of the Uly (Lillum Iongiflorum Thunb. cv. Ace) pollen tube while avoiding interference between the two probes. The results Indicate that the proton fluxes were effluxes, whereas the oxygen fluxes were Influxes, and they were closely correlated to each other surrounding the constitutive alkaline band region. Specifically, when the proton effluxes increased, the oxygen Influxes also increased. Therefore, the hypothesis of condensed active mitochondria existing in the alkalized area of the pollen tube proposed by Hepler's group is supported.  相似文献

Abstract: With a partially purified, membrane-bound (Ca + Mg)-activated ATPase preparation from rat brain, the K0.5 for activation by Ca2+ was 0.8 p μm in the presence of 3 mm -ATP, 6 mm -MgCl2, 100 mM-KCI, and a calcium EGTA buffer system. Optimal ATPase activity under these circumstances was with 6-100 μm -Ca2+, but marked inhibition occurred at higher concentrations. Free Mg2+ increased ATPase activity, with an estimated K0.5, in the presence of 100 μm -CaCl2, of 2.5 mm ; raising the MgCl2 concentration diminished the inhibition due to millimolar concentrations of CaCl2, but antagonized activation by submicromolar concentrations of Ca2+. Dimethylsulfoxide (10%, v/v) had no effect on the K0.5 for activation by Ca2+, but decreased activation by free Mg2+ and increased the inhibition by millimolar CaCl2. The monovalent cations K+, Na+, and TI+ stimulated ATPase activity; for K+ the K0.5 was 8 mm , which was increased to 15 mm in the presence of dimethylsulfoxide. KCI did not affect the apparent affinity for Ca2+ as either activator or inhibitor. The preparation can be phosphorylated at 0°C by [γ-32P]-ATP; on subsequent addition of a large excess of unlabeled ATP the calcium dependent level of phosphorylation declined, with a first-order rate constant of 0.12 s?1. Adding 10 mm -KCI with the unlabeled ATP increased the rate constant to 0.20 s?1, whereas adding 10 mm -NaCl did not affect it measurably. On the other hand, adding dimethyl-sulfoxide slowed the rate of loss, the constant decreasing to 0.06 s?1. Orthovanadate was a potent inhibitor of this enzyme, and inhibition with 1 μm -vanadate was increased by both KCI and dimethylsulfoxide. Properties of the enzyme are thus reminiscent of the plasma membrane (Na + K)-ATPase and the sarcoplasmic reticulum (Ca + Mg)-ATPase, most notably in the K+ stimulation of both dephosphorylation and inhibition by vanadate.  相似文献

The H3 histamine receptor is a high-affinity receptor reported to mediate inhibition of CNS histidine decarboxylase activity and depolarization-induced histamine release. We have used (R)-alpha-[3H]methylhistamine, a specific, high-affinity agonist, to characterize ligand binding to this receptor. Saturation binding studies with rat brain membranes disclosed a single class of sites (KD = 0.68 nM; Bmax = 78 fmol/mg of protein). Competition binding assays also yielded an apparently single class of sites with a rank order of potency for ligands characteristic of an H3 histamine receptor: N alpha-methylhistamine, (R)-alpha-methylhistamine greater than histamine, thioperamide greater than impromidine greater than burimamide greater than dimaprit. In contrast, kinetic studies disclosed two classes of sites, one with fast, the other with slow on-and-off rates. Density of (R)-alpha-[3H]methylhistamine binding followed the order: caudate, midbrain (thalamus and hippocampus), cortex greater than hypothalamus greater than brainstem greater than cerebellum. These data are consistent with an H3 histamine receptor, distinct from H1 and H2 receptors, that occurs in two conformations with respect to agonist association and dissociation or with multiple H3 receptor subtypes that are at present pharmacologically undifferentiated.  相似文献

Two Na^+ and Cl^- Hyperaccumulators of the Chenopodiaceae   总被引:1,自引:0,他引:1  
The authors found five sodium (Na^ ) and chloride (Cl^-) hyperaccumulating halophytes in the Temperate Desert of Xinjiang, China and studied two of them (Suaeda salsa (L.) Pall. and Kalidium folium (Pall.) Moq.). K. folium and S. salsa had a NaCl content of 32.1% and 29.8%, respectively, on a dry weight basis. X-ray microanalysis of the Na in the vacuole, apoplasts and cytoplasm of the two plants indicated a ratio of 7.3:5.6:1.0 in K. folium and 7.3:6.6:1.0 in S. salsa. These data show that K. folium and S. salsa both have a high Na and Cl^- accumulating capacity, which is related to high activity of tonoplast H^ -ATPase and H^ -PPase.  相似文献

In this study, the response of N2 fixation to elevated CO2 was measured in Scirpus olneyi, a C3 sedge, and Spartina patens, a C4 grass, using acetylene reduction assay and 15N2 gas feeding. Field plants grown in PVC tubes (25 cm long, 10 cm internal diameter) were used. Exposure to elevated CO2 significantly (P < 0·05) caused a 35% increase in nitrogenase activity and 73% increase in 15N incorporated by Scirpus olneyi. In Spartina patens, elevated CO2 (660 ± 1 μ mol mol 1) increased nitrogenase activity and 15N incorporation by 13 and 23%, respectively. Estimates showed that the rate of N2 fixation in Scirpus olneyi under elevated CO2 was 611 ± 75 ng 15N fixed plant 1 h 1 compared with 367 ± 46 ng 15N fixed plant 1 h 1 in ambient CO2 plants. In Spartina patens, however, the rate of N2 fixation was 12·5 ± 1·1 versus 9·8 ± 1·3 ng 15N fixed plant 1 h 1 for elevated and ambient CO2, respectively. Heterotrophic non-symbiotic N2 fixation in plant-free marsh sediment also increased significantly (P < 0·05) with elevated CO2. The proportional increase in 15N2 fixation correlated with the relative stimulation of photosynthesis, in that N2 fixation was high in the C3 plant in which photosynthesis was also high, and lower in the C4 plant in which photosynthesis was relatively less stimulated by growth in elevated CO2. These results are consistent with the hypothesis that carbon fixation in C3 species, stimulated by rising CO2, is likely to provide additional carbon to endophytic and below-ground microbial processes.  相似文献
