以58个长白落叶松无性系1年生枝条为材料,对其进行低温胁迫处理,探讨不同无性系抗寒差异,结果表明:不同温度和时间的处理条件下,4个落叶松无性系电导率不同,随着低温胁迫时间的延长和温度的降低,不同无性系电导率呈上升趋势。在相同温度及时间处理条件下(-40℃、12 h),58个长白落叶松无性系相对电导率差异达极显著水平(P<0.01)。以-40℃、12 h处理下各无性系电导率与对照的差值为指标,利用聚类法进行分析,初步筛选出9个抗寒性较强的无性系(L5、L16、L21、L23、L27、L40、L73、L78和L90),这些无性系可作为抗寒优良无性系进行考察,本研究为落叶松抗寒良种选育提供基础。  相似文献

以‘左山1号’、101-14、3309C、SO4、140R和‘黑比诺’等葡萄砧木和品种直径为0.5~0.7 cm的一年生枝条为材料,经低温梯度(0℃、-14℃、-19℃、-24℃、-29℃和-34℃)处理后,对TTC染色的温度和时间进行优化并观察统计葡萄枝条不同组织的存活情况,测定枝条的相对电导率,以及韧皮部和木质部的可溶性蛋白、游离脯氨酸、可溶性糖、淀粉含量和束缚水自由水比例(束自比)5个生理指标,分别用枝条纵切面染色面积、相对电导率拟合Logistic方程计算枝条的半致死温度(LT_(50))来评价枝条的抗寒性,同时通过隶属函数法综合评价枝条韧皮部和木质部抗寒性,并将3种方法的评价结果进行比较,以建立一种直观高效鉴定葡萄品种抗寒性的方法。结果表明:(1)依据低温胁迫下各品种葡萄砧木扦插枝条的萌芽率和生根率表现,其抗寒性由强到弱依次为‘左山1号’、101-14、3309C、SO4、140R和‘黑比诺’。(2)葡萄枝条TTC活力染色的最佳条件为pH=7.0,0.5%TTC-0.1 mol/L磷酸缓冲液在35℃下避光染色36 h。随着胁迫温度的降低,各品种枝条纵切面染色面积均逐渐降低,根据优化TTC法获得‘左山1号’、101-14、3309C、SO4、140R和‘黑比诺’枝条的LT_(50)分别为-31.38℃、-26.51℃、-26.10℃、-23.60℃、-23.33℃和-19.26℃。(3)随着胁迫温度的降低,各品种枝条的相对电导率逐渐增加,且‘黑比诺’的相对电导率基本保持最高、增幅最大(51.93%),而‘左山1号’的相对电导率始终最低、增幅最小(44.07%);根据相对电导率获得‘左山1号’、101-14、3309C、SO4、140R和‘黑比诺’枝条的LT_(50)分别为-30.02℃、-26.40℃、-25.75℃、-23.16℃、-21.13℃和-17.72℃。(4)通过5种生理指标进行枝条抗寒性隶属函数综合性评价结果显示,同一品种中韧皮部抗寒性强于木质部,同一部位不同品种的抗寒性表现为:‘左山1号’>101-14>3309C>SO4>140R>‘黑比诺’。研究发现,TTC染色法与电导法和综合隶属函数法获得的葡萄枝条抗寒力评价结果一致,也与枝条生长恢复鉴定结果一致;但与其他两种方法相比,TTC染色法能更加直观和有效地预测评估葡萄枝条的抗寒性。  相似文献

选用7个白杨杂种无性系1年生苗为材料,采用P-V技术于秋季落叶前测定各无性系枝叶水分参数Ψ0π、Ψ1π00、RWC0、ROWC0、mεax,并于冬季不同时期测定了各无性系相对电导率、钾离子渗漏率、SOD酶活性和POD酶活性的动态变化,对无性系抗寒性进行了综合评定。无性系各项生理指标存在一定变化模式,相对电导率、钾离子渗漏率在12月份降至最低,以后逐步提高,无性系间在2月份变异最大;SOD酶活性和POD酶活性在各时期无性系间差异均较大;无性系抗寒能力评价结果表明,从落叶开始,抗寒能力逐渐提高,12月至翌年1月达到最大,2月份又迅速下降,此时无性系间抗寒能力差异也达到最大。将水分参数与抗寒性指标进行相关分析表明,大部分水分参数与冬季不同时期的抗寒性指标存在不同程度的相关性,特别是与春季指标相关更加显著,其中Ψ0π是相关性最为显著的指标,Ψ1π00相关不明显。  相似文献

不同油橄榄品种对低温胁迫的生理响应及抗寒性综合评价   总被引:5,自引:0,他引:5  
为了明确油橄榄的抗寒机理,对甘肃陇南地区主栽的6个油橄榄品种离体叶片进行低温胁迫(5℃、0℃、-5℃、-10℃)处理,并以18℃常温培养为对照,测定与抗寒性相关的生理生化指标,并采用隶属函数法对不同品种抗寒性进行综合评价。结果显示:(1)各油橄榄品种离体叶片相对电导率随处理温度的降低而呈"S"型曲线变化,结合Logistic方程得出不同品种油橄榄半致死温度介于-6.687℃~-3.706℃之间,且各品种的半致死温度顺序为‘皮肖利’<‘配多灵’<‘莱星’<‘阿斯’<‘鄂植8号’<‘佛奥’,其抗寒性大小依次为‘皮肖利’>‘配多灵’>‘莱星’>‘阿斯’>‘鄂植8号’>‘佛奥’。(2)随着胁迫温度降低,各油橄榄品种离体叶片叶绿素含量逐渐降低,可溶性糖含量基本呈逐渐增加趋势,而可溶性蛋白质含量、脯氨酸含量、丙二醛含量、SOD活性、POD活性和CAT活性则呈先升后降的趋势。(3)在-10℃低温胁迫下,油橄榄品种‘皮肖利’和‘配多灵’的叶片叶绿素含量比常温对照降幅较小,渗透调节物质含量和保护酶活性相对增幅较大,丙二醛含量增幅较小,而此时品种‘佛奥’和‘鄂植8号’各指标的变化趋势与之相反。(4)根据不同油橄榄品种的平均隶属度(抗寒性综合评价值)确定的品种抗寒能力大小顺序(‘皮肖利’>‘配多灵’>‘莱星’>‘阿斯’>‘鄂植8号’>‘佛奥’)与品种半致死温度的抗寒性表现顺序完全一致。研究表明,在低温胁迫条件下,‘皮肖利’和‘配多灵’等油橄榄品种能通过提高渗透调节物质含量和增强保护酶活性来缓解胁迫造成的过氧化伤害,从而表现出较强的抗寒能力;基于相对电导率的半致死温度和隶属函数法的综合评价均能准确鉴定油橄榄的抗寒性。  相似文献

以7个葡萄砧木品种('SO4'、'5A2'、'225'、'通化-3'、'420'、'5BB'、'山河系')和2个葡萄栽培品种('红地球'、'赤霞珠')1年生成熟枝条为试材,考察了它们枝条在0~-33℃低温处理后的萌芽率、生根率、产生愈伤组织的比率、电导率、以及可溶性糖、可溶性蛋白、游离脯氨酸、丙二醛含量的变化,并通过聚类分析、相关分析和主成分分析对其抗寒性进行综合评价.结果表明:(1)随着处理温度的降低,各品种枝条的萌芽率、生根率、愈伤组织比率呈逐渐降低的趋势,而同时它们枝条的电导率、可溶性糖、可溶性蛋白、游离脯氨酸和丙二醛含量却逐渐升高,并且各品种升降的速度和幅度有明显差异,其中品种'红地球'升降最快、幅度最大,品种'5BB'升降最慢、幅度最小.(2)各品种几个参数的升降幅度大小顺序与其抗寒性强弱顺序一致;各品种综合抗寒性由强到弱依次为'5BB'>'山河系'>'通化-3'>'5A2'>'SO4'>'420'>'225' >'赤霞珠'>'红地球'.研究发现,枝条的萌芽率、生根率、产生愈伤组织的比率、电导率以及可溶性蛋白、脯氨酸、丙二醛含量可以作为评价葡萄抗寒性的重要指标.  相似文献

以北京市平原造林常见的8种观叶树种为试验材料,‘测定不同低温胁迫下叶片相对电导率及伤害率,并通过Logistic方程估算出各树种叶片半致死温度,据此对被测树种的抗寒性进行初步评价。结果表明:不同树种叶片相对电导率及伤害率均随着处理温度的降低而升高,且二者趋势一致;通过不同树种抗寒性的聚类分析,可将8个检测树种分为4类。综合测定结果显示,各树种抗寒性顺序为:栾树〉元宝枫〉金焰绣线菊〉珍珠梅〉黄栌〉红宝石海棠〉金叶榆〉紫叶小檗。  相似文献

依据闽北建阳市1971~2005年的极端低温资料构建极值分布数学模型, 用电导率指标确定邓恩桉(Eucalyptus dunnii)优株的半致死温度,预测0~5年、5~10年和10年以上可能出现的极端临界低温分别是-7.47℃、-7.47~-8.5℃和-8.68℃。首次提出利用极端低温分布模型结合电导率指标的方法定量划分林木的抗寒性, 称为“抗寒性极端低温分布法”, 该方法可广泛应用于不同区域、树种的抗寒适应性定量评价和林木个体的抗寒适应性筛选。  相似文献

以电导法检测低温胁迫、低温持续时间以及渗透时间对不同火龙果幼苗及二年生成熟火龙果抗寒性的影响。结果表明,用蒸馏水渗透6h的火龙果,其电解质渗出率基本上达到稳定。温度越低,低温时间越长,电解质渗出率受到的影响越大。在0℃低温下持续处理20d后,不同品系的火龙果幼苗全部死亡。室内低温处理火龙果的抗寒性与低温后田间调查、生长恢复的结果基本上一致。  相似文献

以牡竹属(Dendrocalamus Nees)的清甜竹(D.sapidus Q.H.Dai et D.Y.Huang)、花吊丝竹[D.minor var.amoenus(Q.H.Dai et C.F.Huang) Hsueh et D.Z.Li]和勃氏甜龙竹[D.brandisii (Munro) Kurz]盆栽苗为实验材料,测定了模拟低温(0℃、-5℃、-10℃、-15℃、-20℃和-25℃)条件下3个竹种离体叶片的相对电导率和伤害率,在此基础上根据Logistic方程推算出3个竹种的低温半致死温度(LT50);对自然低温(15℃、10℃、5℃、3℃、0℃和-2 C)条件下3个竹种叶片的形态变化以及相对电导率、叶绿素和MDA含量及SOD活性的变化也进行了比较分析.结果表明:在模拟低温条件下,3个竹种离体叶片的相对电导率和伤害率均随温度降低逐渐增大,且总体上与对照(25℃)有显著差异(P<0.05);根据Logistic方程推算出花吊丝竹、勃氏甜龙竹和清甜竹的LT50分别为-3.07℃、-1.83℃和-1.40 C.在日最低温度大于5℃的自然低温条件下3个竹种的叶片均能够正常生长,而随温度降低,叶片逐渐出现水浸斑点、失绿等症状直至干枯脱落,其中清甜竹叶片伤害症状最重;在自然低温条件下,随日最低气温的降低,3个竹种叶片的叶绿素含量总体呈下降趋势、相对电导率则逐渐升高、MDA含量和SOD活性总体上均呈波动的趋势;与日最低气温15℃相比,在日最低气温-2℃条件下清甜竹叶片叶绿素含量和SOD活性降幅最大而花吊丝竹降幅最小,清甜竹叶片相对电导率最高而花吊丝竹最低.根据LT50和叶片形态及生理指标的变化,可确定3个竹种的抗寒性均较差,其中清甜竹的耐寒能力最弱.  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

Seven strains of aerobic carbon monoxide-oxidizing bacteria (carboxydebacteria) when growing on CO as sole source of carbon and energy had doubling times which ranged from 12–42 h. The activity profiles obtained after discontinuous sucrose density gradient centrifugation indicated that the CO-oxidizing enzymes are soluble and the hydrogenases are membrane-bound in all strains examined. The CO-oxidizing enzymes of Pseudomonas carboxydohydrogena, Pseudomonas carboxydoflava, Comamonas compransoris, and the so far unidentified strains OM2, OM3, and OM4 had a molecular weight of 230,000; that of Achromobacter carboxydus amounted to 170,000. The molecular weights of the CO-oxidizing and H2-oxidizing enzymes turned out to be identical. The cell sonicates were shown to catalyze the oxidation of both CO and H2 with methylene blue, thionine, phenazine methosulfate, toluylene blue, dichlorophenolindophenol, cytochrome c or ferricyanide as electron acceptors. Methyl viologen, benzyl viologen, FAD+, FMN+, and NAD(P)+ were not reduced. The spectrum of electron acceptors was identical for all strains tested. Neither free formate, hydrogen nor oxygen gas were involved in the CO-oxidation reaction. Methylene blue was reduced by CO at a 1:1 molar ratio. The results indicate that CO-oxidation by carboxydobacteria is catalyzed by identical or similar enzymes and that the reaction obeys the equation CO+H2OCO2+2H++2e- as previously shown for Pseudomonas carboxydovorans.Dedicated to Otto Kandler remembering almost three decades of enjoyable cooperation  相似文献

Since the introduction of the concepts of allostery about four decades ago, much advancement has been made in elucidating the structure-function correlation in allostery. However, there are still a number of issues that remain unresolved. In this review we used mammalian pyruvate kinase (PK) as a model system to understand the role of protein dynamics in modulating cooperativity. PK has a triosephosphate isomerase (TIM)(α/β)8 barrel structural motif. PK is an ideal system to address basic questions regarding regulatory mechanisms about this common (α/β)8 structural motif. The simplest model accounting for all of the solution thermodynamic and kinetic data on ligand-enzyme interactions involves two conformational states, inactive ET and active ER. These conformational states are represented by domain movements. Further studies provide the first evidence for a differential effect of ligand binding on the dynamics of the structural elements, not major secondary structural changes. These data are consistent with our model that allosteric regulation of PK is the consequence of perturbation of the distribution of an ensemble of states in which the inactive ET and active ER represent the two extreme end states. Sequence differences and ligands can modulate the distribution of states leading to alterations of functions. The future work includes: defining the network of functionally connected residues; elucidating the chemical principles governing the sequence differences which affect functions; and probing the nature of mutations on the stability of the secondary structural elements, which in turn modulate allostery.  相似文献
