A Genetic linkage map of Pinyon pine (Pinus edulis) based on amplified fragment length polymorphisms 总被引:1,自引:0,他引:1

Amplified fragment length polymorphisms (AFLP) were used to rapidly generate a dense linkage map for pinyon pine (Pinus edulis). The map population consisted of 40 megagametophytes derived from one tree at Sunset Crater, Arizona. A total of 78 primer
combinations, each with three to five selective nucleotides, amplified 542 polymorphic markers. Of these, 33 markers showed
significant deviation from the expected Mendelian genotypic segregation ratio of 1 : 1, and 164 showed complete linkage with
another marker. This resulted in 338 unique markers mapping to 25 linkage groups, each of which ranged from 2 to 22 markers,
averaging 80 centiMorgans (cM) in size and covering 2,012 cM (2,200 cM with the inclusion of 25 cM for each of 7 unlinked
markers). Pairwise linkage values gave a genome size estimate of 2,390 cM, suggesting comprehensive coverage of the genome.
A search for subsets of primer combinations giving the best map coverage found 10 primer combinations which together marked
72% of the linkage map to within 10 cM; an additional 10 primer combinations increased this percentage to 85%. Our map represents
an initial step towards the identification of quantitative trait loci associated with pest resistance and water stress in
pinyons and will further allow us to examine introgression rates between P. edulis and P. californiarum.

Genetic maps facilitate the study of genome structure and evolution, and the identification of monogenic traits or Mendelian
components of quantitative traits. We evaluated 228 RAPD, microsatellite and AFLP markers for linkage analysis in melon (Cucumis melo L.) varieties MR-1 (resistant to Fusarium wilt, powdery and downy mildews) and Ananas Yokneum (AY; susceptible to these diseases) and constructed a detailed genetic
map. The mapping population consisted of 66 backcross progenies derived from AY×(MR-1×AY). Despite a relatively low level
of polymorphism in the species, AFLP markers were found to be more efficient in mapping the melon genome than RAPD or microsatellite
markers. The map contains 197 AFLPs, six RAPDs and one microsatellite marker assigned to 14 major and six minor linkage groups,
and covers 1942 cM with the average distance between adjacent markers of approximately 10 cM. The maximum distance allowed
between markers is 27.5 cM. About 11% of the intervals (20 out of 173) are over 20 cM (but less than 27.5 cM). The map has
immediate utility for identifying markers linked to disease resistance genes that are suitable for marker-assisted breeding.
The use of microsatellite markers for integration with other maps is also discussed.

Haploid linkage analysis of eastern white pine, Pinus strobus L., was carried out using mainly RAPD markers and microsatellite, or simple-sequence-repeat, markers. Ninety one loci mapped
to 12 linkage groups of three or more markers. The resulting framework genome map, the first for a soft pine species, contained
69 markers. The map covered 58% of the estimated genome length of 2071 cM(K), with a 95% confidence interval of 1828–2242 cM(K).
A systematic comparison of linkage data from eastern white pine, longleaf pine (P. palustris Mill.) and maritime pine (P. pinaster Ait.), gave genome-length estimates for all three species very close to either 2000 cM(K) or 2600 cM(H), depending on whether
the Kosambi(K) or Haldane(H) map functions, respectively, were employed. Differences among previous pine genome-length estimates
were attributed to the divergent criteria used in the methods of estimation, and indicate the need for the adoption of uniform
criteria when performing genome-length estimates. Current data suggest that members of the two pine subgenera, which diverged
during the late Mesozoic era, have highly conserved rates of recombination.

A high-density genetic map of the rice blast fungus Magnaporthe grisea (Guy11×2539) was constructed by adding 87 cosmid-derived RFLP markers to previously generated maps. The new map consists
of 203 markers representing 132 independently segregating loci and spans approximately 900 cM with an average resolution of
4.5 cM. Mapping of 33 cosmid probes from the genetic map generated by Sweigard et al. has allowed the integration of two M. grisea maps. The integrated map showed that the linear order of markers along all seven chromosomes in both maps is in good agreement.
Thirty of eighty seven markers were derived from cosmid clones that contained the retrotransposon MAGGY (M. grisea gypsy element). Mapping of single-copy DNA sequences associated with the MAGGY cosmids indicated that MAGGY elements are
scattered throughout the fungal genome. In eight cases, the probes associated with MAGGY elements showed abnormal segregation
patterns. This suggests that MAGGY may be involved in genomic rearrangements. Two RFLP probes linked to MAGGY elements, and
another flanking other repetitive DNA elements, identified sequences that were duplicated in the Guy11 genome. Most of the
MAGGY cosmids also contained other classes of repetitive DNA suggesting that repetitive DNA sequences tend to cluster in the
M. grisea genome.

An RFLP linkage map was constructed for cultivated sunflower Helianthus annuus L., based on 271 loci detected by 232 cDNA probes. Ninety-three F2 plants of a cross between inbred lines RHA 271 and HA 234 were used as the mapping population. These genetic markers plus
a fertility restoration gene, Rf
1, defined 20 linkage groups, covering 1164 cM of the sunflower genome. Of the 71 loci 202 had codominant genotypic segregation,
with the rest showing dominant segregation. Thirty-two of the 232 probes gave multiple locus segregation. There were 39 clusters
of tightly linked markers with 0 cM distance among loci. This map has an average marker-to-marker distance of 4.6 cM, with
11 markerless regions exceeding 20 cM.

The influence of ”shinrin-yoku” (forest-air bathing and walking) on blood glucose levels in diabetic patients was examined.
Eighty-seven (29 male and 58 female) non-insulin-dependent diabetic patients [61 (SEM 1) years old] participated in the present
study. Shinrin-yoku was performed nine times over a period of 6 years. The patients were divided into two parties. They then
walked in the forest for 3 km or 6 km according to their physical ability and/or the existence of diabetic complications.
The mean blood glucose level after forest walking changed from 179 (SEM 4) mg · 100 ml–1 to 108 (SEM 2) mg · 100 ml–1 (P<0.0001). The level of glycated haemoglobin A1c also decreased from 6.9 (SEM 0.2)% (before the first shinrin-yoku) to 6.5 (SEM 0.1)% (after the last shinrin-yoku; P<0.05). Blood glucose values declined by 74 (SEM 9) mg · 100 ml–1 and 70 (SEM 4) mg · 100 ml–1 after short- and long-distance walking respectively. There was no significant difference between these values. Since the
forest environment causes changes in hormonal secretion and autonomic nervous functions, it is presumed that, in addition
to the increased calorie consumption and improved insulin sensitivity, walking in a forest environment has other beneficial
effects in decreasing blood glucose levels.

A map with 246 markers (11 isozymes and 235 RFLPs) was constructed using an interspecific F2 population between almond (cv Texas) and peach (cv Earlygold). RFLPs were obtained using 213 probes from the genomic and
cDNA libraries of different species (almond, peach, P. ferganensis, cherry, plum and apple), including 16 almond probes which correspond to known genes. All markers were distributed in eight
linkage groups, the same as the basic chromosome number of the genus, covering a total distance of 491 cM. The average map
density was 2.0 cM/marker and only four gaps of 10 cM or more were found; the two largest gaps were 12cM each. This map was
compared with one constructed previously with an intraspecific almond population sharing 67 anchor loci. Locus order was nearly
identical and distances were not significantly different. A large proportion of the mapped loci (46%) had skewed segregations;
in approximately half of them, the distortion was due to an excess of heterozygotes. One of the distorted regions could be
associated with the position of the self-incompatibility gene of almond.

Bulked segregant analysis and high-resolution mapping were used to pinpoint the position of the Lv gene for resistance to Leveillula taurica in tomato. Mapping in an F2, corresponding to more than 3800 gametes, indicates that Lv is positioned within the 0.84-cM interval defined by the RFLP markers CT121 and CT129, with the closest marker, CT121, being
only 0.16 cM from the gene. The tight linkage of these markers demonstrates their usefulness in marker-assisted breeding for
Lv, and the high-resolution map provides a starting a starting point for positional cloning of this resistance gene.

A comparative genetic map of Aegilops umbellulata with wheat was constructed using RFLP probes that detect homoeoloci previously mapped in hexaploid bread wheat. All seven
Ae. umbellulata chromosomes display one or more rearrangements relative to wheat. These structural changes are consistent with the sub-terminal
morphology of chromosomes 2 U, 3 U, 6 U and 7 U. Comparison of the chromosomal locations assigned by mapping and those obtained
by hybridization to wheat/Ae. umbellulata single chromosome addition lines verified the composition of the added Ae. umbellulata chromosomes and indicated that no further cytological rearrangements had taken place during the production of the alien-wheat
aneuploid lines. Relationships between Ae. umbellulata and wheat chromosomes were confirmed, based on homoeology of the centromeric regions, for 1 U, 2 U, 3 U, 5 U and 7 U. However,
homoeology of the centromeric regions of 4 U with wheat group-6 chromosomes and of 6 U with wheat group-4 chromosomes was
also confirmed, suggesting that a re-naming of these chromosomes may be pertinent. The consequences of the rearrangements
of the Ae. umbellulata genome relative to wheat for gene introgression are discussed.

An avocado genomic library was screened with various microsatellite repeats. (A/T)n and (TC/AG)n sequences were found to be the most frequent repeats. One hundred and seventy-two positive clones were sequenced successfully
of which 113 were found to contain simple sequence repeats (SSR). Polymerase chain reaction primers were designed to the regions
flanking the SSR in 62 clones. A GenBank search of avocado DNA sequences revealed 1 sequence containing a (CT)10 repeat. A total of 92 avocado-specific SSR markers were screened for polymorphism using 50 offspring of a cross between the
avocado cultivars ‘Pinkerton’ and ‘Ettinger’. Both are standard avocado cultivars which are normally outcrossed and highly
heterozygous. Fifty polymorphic SSR loci, 17 random amplified polymorphic DNA (RAPD) and 23 minisatellite DNA Fingerprint
(DFP) bands were used to construct the avocado genetic map. The resulting data were analyzed with various mapping programs
in order to assess which program best accommodated data from progeny of heterozygous parents. The analyses resulted in 12
linkage groups with 34 markers (25 SSRs, 3 RAPDs and 6 DFP bands) covering 352.6 cM. This initial map can serve as a basis
for developing a detailed genomic map and for detection of linkage between markers and quantitative trait loci.

Soil salinity markedly suppresses the growth of rice (Oryza sativa L.). We established rice anther culture to select for rice callus lines adapted to NaCl stress and regenerated plant progenies
resistant to a NaCl stress of E.C. 16–18 mS. When exposed to NaCl, NaCl-adapted rice calli lost K+ and accumulated little Na+. Conversely, plant cells lost relatively little K+ and accumulated Na+. It is plausible that, NaCl-resistant mechanisms are different at callus and plant level. The stable NaCl-resistant lines
produced have potential use in elucidating the molecular mechanisms behind NaCl resistance in rice and in rice breeding.

We have constructed a genetic linkage map within the cultivated gene pool of cowpea (2n=2x=22) from an F8 recombinant inbred population (94 individuals) derived from a cross between the inbreds IT84S-2049 and 524B. These breeding
lines, developed in Nigeria and California, show contrasting reactions against several pests and diseases and differ in several
morphological traits. Parental lines were screened with 332 random RAPD decamers, 74 RFLP probes (bean, cowpea and mung bean
genomic DNA clones), and 17 AFLP primer combinations. RAPD primers were twice as efficient as AFLP primers and RFLP probes
in detecting polymorphisms in this cross. The map consists of 181 loci, comprising 133 RAPDs, 19 RFLPs, 25 AFLPs, three morphological/classical
markers, and a biochemical marker (dehydrin). These markers identified 12 linkage groups spanning 972 cM with an average distance
of 6.4 cM between markers. Linkage groups ranged from 3 to 257 cM in length and included from 2 to 41 markers, respectively.
A gene for earliness was mapped on linkage group 2. Seed weight showed a significant association with a RAPD marker on linkage
group 5. This map should facilitate the identification of markers that “tag” genes for pest and disease resistance and other
traits in the cultivated gene pool of cowpea.

To examine possible calcium (Ca2+)-mediated prefertilization events in male gametes of higher plants, we studied protein phosphorylation and the Ca2+-binding proteins, calmodulin and calreticulin, in sperm cells isolated from maize (Zea mays L.) pollen in the presence and absence of Ca2+. Using immunoblotting, we detected calmodulin and calreticulin and Ca2+-induced variations. Exposure of sperm cells to 1 mM Ca2+ for 1 h increased calmodulin content by 136% compared with the control. Ca2+ had little effect on calreticulin at 1 h, but induced a 34% increase after 3 h. Phosphorylation of proteins was low in 1 h-control
and Ca2+-treated cells. However, a 13-fold increase in phosphorylation of a 18-kDa protein was found at 12 h in the presence of Ca2+. Ca2+-induced changes in calmodulin, calreticulin and protein phosphorylation observed in maize sperm cells may reflect prefertilization
changes in vivo that facilitate sperm cell fusion with egg and central cells.

Human thermal physiological and comfort models will soon be able to simulate both transient and spatial inhomogeneities in
the thermal environment. With this increasing detail comes the need for anatomically specific convective and radiative heat
transfer coefficients for the human body. The present study used an articulated thermal manikin with 16 body segments (head,
chest, back, upper arms, forearms, hands, pelvis, upper legs, lower legs, feet) to generate radiative heat transfer coefficients
as well as natural- and forced-mode convective coefficients. The tests were conducted across a range of wind speeds from still
air to 5.0 m/s, representing atmospheric conditions typical of both indoors and outdoors. Both standing and seated postures
were investigated, as were eight different wind azimuth angles. The radiative heat transfer coefficient measured for the whole-body
was 4.5 W/m2 per K for both the seated and standing cases, closely matching the generally accepted whole-body value of 4.7 W/m2 per K. Similarly, the whole-body natural convection coefficient for the manikin fell within the mid-range of previously published
values at 3.4 and 3.3 W/m2 per K when standing and seated respectively. In the forced convective regime, heat transfer coefficients were higher for hands,
feet and peripheral limbs compared to the central torso region. Wind direction had little effect on convective heat transfers
from individual body segments. A general-purpose forced convection equation suitable for application to both seated and standing
postures indoors was h

0.6 for the whole-body. Similar equations were generated for individual body segments in both seated and standing postures.

In vitro pollen germination and tube length studies are valuable in elucidating mechanisms (germination capacity and rate, tube growth
rate) possibly associated with genetic differences in male transmission. On each of two collection dates, the percentage germination
and tube length of the binucleate pollen grains from five diverse sesame (Sesamum indicum L.) genotypes were determined at eight times (30, 60, 90, 120, 150, 180, 240, 300 min) after inoculation on a semisolid medium
containing 10% (100 g l-1) sucrose (C12H22O11), 0.4% (4 g l-1) purified agar (Fisher Lot 914409), 0.1% (1 g l-1) calcium nitrate [Ca(NO3)2 ⋅ 4H2O] and 0.01% (100 mg l-1) boric acid (H3BO3). Before heating, the pH of the medium was adjusted to 7.0 with a 0.1 N potassium hydroxide (KOH) solution. Over the five genotypes, 5% germination was found 30 min after inoculation and a maximum
of 37% germination 120 min after inoculation with no significant changes thereafter. As indicated by the highly significant
genotype×time after inoculation interaction, the genotypes differed in the time at which germination was initiated and maximum
germination attained. Over all five genotypes, the tube length was 91 μm 30 min after inoculation, reaching a maximum of 1000 μm
300 min after inoculation. As shown by the highly significant genotype×time after inoculation interaction, the genotypes differed
in the time at which tube length was observed and the maximum tube length was attained. Little or no relationship between
percent germination and tube length was observed among the genotypes. For both percent germination and tube length, the statistical
significance of collection date and its interactions with genotype and time after inoculation indicated that environment in
the form of collection date was also an influencing factor. These results indicated that genetic differences among genotypes
were present for in vitro germination capacity, germination rate and tube growth rate and that these factors singly or in combination could alter male
transmission of genetic elements.

To improve the quality of cytoplasmic labelling of GAL4-expressing cells in Drosophila enhancer-trap and transgenic strains, a new GAL4-responsive reporter UAS-tau, which features a bovine tau cDNA under control of a yeast upstream activation sequence (UAS), was tested. Tau, a microtubule-associated protein, is distributed
actively and evenly into all cellular processes. Monoclonal anti-bovine Tau antibody reveals the axonal structure of the labelled
cells with detail similar to that of Golgi impregnation. We demonstrate that the UAS-tau system is especially useful for studying processes of differentiation and reorganisation of identified neurones during postembryonic
development.

Pearl millet [Pennisetum glaucum (L.) R.Br.] is a warm-season grass used for food, feed, fodder and forage, primarily in countries of Africa and India but
grown around the world. The two most-destructive diseases to pearl millet in the United States are rust (caused by Puccinia substriata var. indica) and pyricularia leaf spot (caused by Pyricularia grisea). Genes for disease resistance to both pathogens have been transferred into agronomically acceptable forage and grain cultivars.
A study was undertaken to identify molecular markers for three rust loci and one pyricularia resistance locus. Three segregating
populations were screened for RAPDs using random decamer primers and for RFLPs using a core set of probes detecting single-copy
markers on the pearl millet map. The rust resistance gene Rr
1
from the pearl millet subspecies P. glaucum ssp. monodii was linked 8.5 cM from the RAPD OP-G8350. The linkage of two RFLP markers, Xpsm108 (15.5 cM) and Xpsm174 (17.7 cM), placed the Rr
1
gene on linkage-group 3 of the pearl millet map. Rust resistance genes from both Tift 89D2 and ICMP 83506 were placed on linkage-group 4 by determining genetic linkage to the RFLP marker Xpsm716 (4.9 and 0.0 cM, respectively). Resistance in ICMP 83506 was also linked to the RFLP marker Xpsm306 (10.0 cM), while resistance in Tift 89D2 was linked to RAPD markers OP-K19350 (8.8 cM) and OP-O8350 (19.6 cM). Fragments from OP-K19 and OP-O8 in the ICMP 83506 population, and Xpsm306 in the Tift 89D2 population, were monomorphic. Only one RAPD marker (OP-D11700, 5.6 cM) was linked to pyricularia leaf spot resistance. Attempts to detect polymorphisms with rice RFLP probes linked to
rice blast resistance (Pyricularia oryzae; syn=P. grisea) were unsuccessful.

Disposable sensor for biochemical oxygen demand 总被引:6,自引:0,他引:6
Disposable-type microbial sensors were prepared for the determination of biochemical oxygen demand (BOD). The yeast, Trichosporon cutaneum, was directly immobilized on the surface of miniature oxygen electrodes using an ultraviolet crosslinking resin (ENT-3400).
The oxygen electrodes (15 mm× 2 mm×0.4 mm) were made on silicon substrates using micromachining techniques. They were Clark-type
two-electrode systems with−1021 mV applied to the working electrode. Typical response times of the BOD sensors were in the
range of 7–20 min. At 20°C, the sensors’ dynamic range was from 0 to 18 mg/l BOD when a glucose/glutamate BOD standard solution
was used. The lower limit of detection was 0.2 mg/l BOD. This value was one order of magnitude lower than that of sensors
previously reported. The sensors’ operational lifetime of 3 days was satisfactory for a disposable type. The sensors’ responses
were reproducible to within 8% relative standard deviation. The BOD sensors’ were applied to untreated and treated waste waters
from industrial effluents and municipal sewage. BOD values determined using these sensors correlated well with those determined
by the conventional 5-day BOD determination method.

Tropical zooplankton in the highly-enclosed lagoon of Taiaro Atoll (Tuamotu Archipelago, French Polynesia) 总被引:1,自引:0,他引:1
Nocturnal zooplankton assemblages around Taiaro Atoll were sampled over six nights during February 1994. Replicate zooplankton
samples were collected at windward and leeward locations in the enclosed lagoon and adjacent ocean with a metered net (85 cm
diameter, 500 μm mesh) towed for 15 min at 5 m depth. The zooplankton community in the lagoon was very different from that
in the ocean. Oceanic samples contained 50 mostly holoplanktonic taxa (diversity index, H′=2.62; evenness index, J′=0.67).
Lagoonal samples contained 19 mostly meroplanktonic taxa (H′=1.54, J′=0.52) with three taxa (decapod larvae; an ostracod,
Cypridina sp.; a copepod, Acartia fossae) contributing >90% of the individuals. Unlike the ocean, zooplankton distributions in the lagoon were not homogenous; instead
spatial patterns were apparently formed by the interaction between hydrodynamic processes and species-specific behaviour.
