Effects of MgCl2 and free fatty acids (FFA) on galactolipid:galactolipid galactosyltransferase (GGGT) and UDP-galactose: 1,2-diacylglycerol galactosyltransferase (UDGT) in chloroplast envelope membranes isolated from spinach (Spinacia oleracea L.) leaves were examined. GGGT activity was sigmoidally stimulated by MgCl2 with a saturated concentration of more than 5 millimolar. Free α-linolenic acid (18:3) caused a drastic increase in GGGT activity under limiting concentrations of MgCl2, without affecting its maximum activity at higher MgCl2 concentrations. Free 18:3 alone did not affect the GGGT activity. The effective species of FFA for the stimulation of GGGT activity in the presence of MgCl2 were unsaturated 16- and 18-carbon fatty acids. GGGT activity was also stimulated by 18:3 in the presence of MnCl2, CaCl2 and a high concentration of KCl in place of MgCl2. UDGT activity was hyperbolically enhanced by MgCl2 with a saturated concentration of 1 to 2 millimolar. In contrast to GGGT, UDGT was severely inhibited by 18:3, and MgCl2-induced stimulation was completely abolished by 18:3. Unsaturated 16- and 18-carbon fatty acids were more inhibitory to UDGT than the saturated acids. The dependence of GGGT activity on monogalactosyldiacylglycerol (MGDG) and MgCl2 concentrations was identical in the envelope membranes isolated from non- and ozone (0.5 microliter/liter)-fumigated spinach leaves, indicating that GGGT remained active in the leaves during ozone fumigation. The results are discussed in relation to the regulation of galactolipid biosynthesis by the endogenous FFA in the envelopes and to the involvement of GGGT in the triacylglycerol synthesis from MGDG in ozone-fumigated leaves.  相似文献

Molecular species and fatty acid distribution of triacylglycerol (TG) accumulated in spinach (Spinacia oleracea L.) leaves fumigated with ozone (0.5 microliter per liter) were compared with those of monogalactosyldiacylglycerol (MGDG). Analysis of positional distribution of the fatty acids in MGDG and the accumulated TG by the enzymatic digestion method showed that hexadecatrienoate (16:3) was restricted to sn-2 position of the glycerol backbone in both MGDG and TG, whereas α-linolenate (18:3) was preferentially located at sn-1 position in MGDG, and sn-1 and/or sn-3 positions in TG, suggesting that 1,2-diacylglycerol moieties of MGDG are the direct precursor of TG in ozonefumigated leaves. Further analysis of TG molecular species by argentation chromatography and mass spectrometry showed that TG increased with ozone fumigation consisted of approximately an equal molar ratio of sn-1,3-18:3-2-16:3 and sn-1,2,3-18:3. Because the molecular species of MGDG in spinach leaves is composed of a similar molar ratio of sn-1-18:3-2-16:3 and sn-1,2-18:3, we concluded that MGDG was converted to 1,2-diacylglycerol and acylated with 18:3 to TG in ozone-fumigated spinach leaves.  相似文献

Purified, intact chloroplasts of Spinacia oleracea L. synthesize galactose-labeled mono- and digalactosyldiacylglycerol (MGDG and DGDG) from UDP-[U-14C]galactose. In the presence of high concentrations of unchelated divalent cations they also synthesize tri- and tetra-galactosyldiacylglycerol. The acyl chains of galactose-labeled MGDG are strongly desaturated and such MGDG is a good precursor for DGDG and higher oligogalactolipids. The synthesis of MGDG is catalyzed by UDP-Gal:sn-1,2-diacylglycerol galactosyltransferase, and synthesis of DGDG and the oligogalactolipids is exclusively catalyzed by galactolipid:galactolipid galactosyltransferase. The content of diacylglycerol in chloroplasts remains low during UDP-Gal incorporation. This indicates that formation of diacylglycerol by galactolipid:galactolipid galactosyltransferase is balanced with diacylglycerol consumption by UDP-Gal:diacylglycerol galactosyltransferase for MGDG synthesis. Incubation of intact spinach chloroplasts with [2-14C]acetate or sn-[U-14C]glycerol-3-P in the presence of Mg2+ and unlabeled UDP-Gal resulted in high 14C incorporation into MGDG, while DGDG labeling was low. This de novo made MGDG is mainly oligoene. Its conversion into DGDG is also catalyzed, at least in part, by galactolipid:galactolipid galactosyltransferase.  相似文献

When mature spinach (Spinacia oleracea L. cv. New Asia) plantswere continuously fumigated with 0.5 ppm (v/v) ozone, the totalfatty acids in the leaf lipids remained unchanged in both contentand composition during the initial 8 h, but thereafter they,especially hexadecatrienoic acid (16:3) and -linolenic acid(18:3), began to decrease with marked accumulation of malondialdehyde,indicating fatty acid peroxidation. During the first 6 h, fattyacids in polar lipids decreased to 68% of their initial level,and most were recovered in the neutral lipid fraction. Amongthe polar lipids, monogalactosyl diacylglycerol (MGDG), digalactosyldiacyiglycerol (DGDG) and phosphatidylcholine (PC) decreasedwhereas sulfoquinovosyl diacylglycerol, phosphatidyl glycerol(PG), phosphatidylethanolamine and phosphatidylinositol werestable during 6 h of ozone fumigation. Phosphatidic acid (PA)increased 3.4 times during the same period. Among the neutrallipids, triacyiglycerol (TG) and 1,2-diacylglycerol (1,2-DG)increased markedly with ozone fumigation. The constituent fatty acids of 16:3, oleic acid (18:1), linoleicacid (18:2) and 18:3 increased markedly in TG and 1,2-DG within6 h of ozone fumigation, whereas 3-trans-hexadecenoic acid (16:1),18:1, 18:2 and 18:3 increased in PA. Since 16:3 is specificto MGDG and DGDG, and 18:1 and 18:2 to PC among the glycerolipidsmainly reduced by ozone, the results suggest that the acyl moietiesof the galactolipids were converted to TG via 1,2-DG, and thoseof PC to TG through PA and 1,2-DG. The appearance of 16:1 inPA indicates that a small amount of PG, specifically acylatedwith 16:1, was degraded to PA by ozone fumigation. Mono- and digalactosyldiacylglycerol (MGDG and DGDG) were isolated from the leaves of sixteen 16:3 plants. In all of these plant species, the sn-2 position of MGDG was more enriched in C16 fatty acids than sn-2 of DGDG. The molar ratios of prokaryotic MGDG to prokaryotic DGDG ranged from 4 to 10. This suggests that 16:3 plants synthesize more prokaryotic MGDG than prokaryotic DGDG. In the 16:3 plant Spinacia oleracea L. (spinach), the formation of prokaryotic galactolipids was studied both in vivo and in vitro. In intact spinach leaves as well as in chloroplasts isolated from these leaves, radioactivity from [1-14C]acetate accumulated 10 times faster in MGDG than in DGDG. After 2 hours of incorporation, most labeled galactolipids from leaves and all labeled galactolipids from isolated chloroplasts were in the prokaryotic configuration. Both in vivo and in vitro, the desaturation of labeled palmitate and oleate to trienoic fatty acids was higher in MGDG than in DGDG. In leaves, palmitate at the sn-2 position was desaturated in MGDG but not in DGDG. In isolated chloroplasts, palmitate at sn-2 similarly was desaturated only in MGDG, but palmitate and oleate at the sn-1 position were desaturated in MGDG as well as in DGDG. Apparently, palmitate desaturase reacts with sn-1 palmitate in either galactolipid, but does not react with the sn-2 fatty acid of DGDG. These results demonstrate that isolated spinach chloroplasts can synthesize and desaturate prokaryotic MGDG and DGDG. The finally accumulating molecular species, MGDG(18:3/16:3) and DGDG(18:3/16:0), are made by the chloroplasts in proportions similar to those found in leaves.  相似文献

The galactose, glycerol, and fatty acids of mono- and digalactosyl diglycerides (MGDG and DGDG) have been separated and analyzed for 14C activity after 14CO2 feeding of Vicia faba leaf discs. Fully expanded and developing leaves were analyzed at time intervals following feeding during continuous illumination. In addition, fully expanded leaves were analyzed after similar times in complete darkness. In all cases, 14C was incorporated very rapidly into galactose, whereas glycerol and fatty acids were labeled much more slowly and over a longer period of time. The data are consistent with the galactosylation of a diglyceride to MGDG which is in turn galactosylated to DGDG. The data suggest that the formation of diglycerides suitable for galactosylation to MGDG is slow in comparison to the galactosylation process. It is also suggested that DGDG may be formed from more than one pool of MGDG. The complete analysis of the 14C incorporation into galactose appears to represent the only satisfactory method of comparing galactolipid synthesis by 14C incorporation. Estimates of comparative rates of synthesis of MGDG and DGDG have been made on this basis.  相似文献

This research focuses on galactolipid depletion in blast fungus‐infected rice leaves. Two major galactolipids, monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG), from rice leaves were isolated and purified. The chemical structure of MGDG was identified as 1,2‐dilinolenyl‐3‐O‐β‐d ‐galactopyranosyl‐sn‐glycerol, and that of DGDG as 1,2‐dilinolenyl‐3‐O‐[α‐d ‐galactopyranosyl‐(1→6)‐O‐β‐d ‐galactopyranosyl]‐sn‐glycerol. Both the MGDG and DGDG content in the incompatible blast fungus race‐infected leaves decreased more than those in the compatible blast fungus race‐infected leaves during the infection process. Active oxygen species had the ability to peroxygenate and de‐esterify MGDG or DGDG in vitro, suggesting that active oxygen species play an important role in galactolipid depletion during the process of rice blast fungus invasion. Other possible functions of rice galactolipids during disease resistance are also discussed.  相似文献

The changes of galactolipids (MGDG and DGDG, largely 18:3/18:3), free fatty acids (FFA), and phosphatidylcholine (PC) taking place during senescence of primary barley leaves were analysed employing HPLC and GLC. Upon induction of senescence MGDG and, with some delay, DGDG began to disappear and were largely broken down at the end of the senescence period. A concomitant appearance of a pool of FFA could not be observed. However, PC accumulated during the main period of galactolipid breakdown. This change was due to the marked increase of the 18:3/18:3 molecular species of PC. An inverse correlation between the changes of galactolipids and PC could be established. A hypothesis featuring the conversion of galactolipids via diacylglycerol to PC is presented as the principal route of galactolipid breakdown.  相似文献

Acyl lipids and pigments were analyzed in young plants of garden pea, spring wheat and spinach exposed to < 5 or 65 nl l?1 ozone 12 h per day for 6 days. In one set of experiments, the plants were exposed to 14CO2 for 2 h 3 days prior to ozone exposure. The plants responded differently to the moderately enhanced level of ozone used Spinach was not at all sensitive while in both pea and wheat, leaves of different ages differed in ozone sensitivity. In pea, ozone sensitivity increased with leaf age. In the second and third oldest leaves, the amounts of galactolipids per leaf area and the proportions of 18:3 of the total lipid extract and of phosphatidylglycerol decreased. In the second oldest leaf, ozone also caused a decreased proportion of 18:3 of monogalactosyldiacylglycerol. In the fourth oldest leaf, lipid composition and galactolipid unsaturation was unaffected, but ozone caused decreased leaf expansion resulting in increased acyl lipid content per leaf area. In both the first and second leaves of wheat, ozone fumigation caused a marked decrease in the content of monogalactosyldiacylglycerol and in the first leaf, the contents of phosphatidylcholine and phosphatidylethanolamine increased. The proportion of 18:3 in phosphatidylcholine was larger in ozone-fumigated than in control plants, while the reverse applied for phosphatidylglycerol. In the oldest sampled leaves of pea and wheat, ozone caused an increase in the radioactivity associated with β-carotene, indicating increased turnover. Thus, while spinach was unaffected, in both pea and wheat ozone caused a decrease in the proportion of chloroplast membrane lipids to non-chloroplast membrane lipids in older leaves while younger leaves were less sensitive.  相似文献

Primary leaves of Phaseolus vulgaris show concomitant changes in phospholipid, galactolipid, chlorophyll and fresh weight during leaf development from 3 to 32 days after planting. Phosphatidyl choline, phosphatidyl ethanolamine, and phosphatidyl inositol show only small changes on a mole per cent lipid phosphate basis during leaf development. The chloroplast lipids, phosphatidyl glycerol, monogalactosyl diglyceride (MGDG) and digalactosyl diglyceride (DGDG) all show marked increases and decreases which are coincident with chloroplast development. The decline in the leaf content of chloroplast polar lipids and chlorophyll become evident upon reaching maximal leaf size. The molar ratio of galactolipids (MGDG/DGDG), reaches a maximum value of 2.3 in expanding leaves, but steadily declines during senescence to a minimum value of 1.5 at abscission. The declining ratio is caused by a preferential loss of MGDG in the senescing leaves.  相似文献

The thylakoid membranes of the chloroplast harbor the photosynthetic machinery that converts light into chemical energy. Chloroplast membranes are unique in their lipid makeup, which is dominated by the galactolipids mono‐ and digalactosyldiacylglycerol (MGDG and DGDG). The most abundant galactolipid, MGDG, is assembled through both plastid and endoplasmic reticulum (ER) pathways in Arabidopsis, resulting in distinguishable molecular lipid species. Phosphatidic acid (PA) is the first glycerolipid formed by the plastid galactolipid biosynthetic pathway. It is converted to substrate diacylglycerol (DAG) for MGDG Synthase (MGD1) which adds to it a galactose from UDP‐Gal. The enzymatic reactions yielding these galactolipids have been well established. However, auxiliary or regulatory factors are largely unknown. We identified a predicted rhomboid‐like protease 10 (RBL10), located in plastids of Arabidopsis thaliana, that affects galactolipid biosynthesis likely through intramembrane proteolysis. Plants with T‐DNA disruptions in RBL10 have greatly decreased 16:3 (acyl carbons:double bonds) and increased 18:3 acyl chain abundance in MGDG of leaves. Additionally, rbl10‐1 mutants show reduced [14C]–acetate incorporation into MGDG during pulse?chase labeling, indicating a reduced flux through the plastid galactolipid biosynthesis pathway. While plastid MGDG biosynthesis is blocked in rbl10‐1 mutants, they are capable of synthesizing PA, as well as producing normal amounts of MGDG by compensating with ER‐derived lipid precursors. These findings link this predicted protease to the utilization of PA for plastid galactolipid biosynthesis potentially revealing a regulatory mechanism in chloroplasts.  相似文献

Metabolic Turnover of Fatty Acids and Acylglycerols in Rat Sciatic Nerve   总被引:3,自引:3,他引:0  
To explain the discrepancy between the low level and high metabolic activity of endoneurial free fatty acids (FFAs) and triacylglycerol (TG), levels of de novo synthesized FFA and acylglycerols were measured in rat sciatic endoneurium at various intervals after endoneurial microinjection of [14C]acetate. Soon after injection (less than 10 min), the [14C]acetate was metabolized to FFA and incorporated into diacylglycerol (DG), TG, sterols, ceramides, and various phospholipids. The proportions of 14C-labeled FFA, DG, TG, and ceramides to total 14C-labeled lipids decreased, whereas those of phospholipids and cerebrosides increased with time after injection. These findings suggest that rapid turnover of FFA and TG may contribute to their low level in sciatic endoneurium. The de novo synthesized fatty acids were largely incorporated into phosphatidylcholine (approximately 50% of total 14C-labeled phospholipids), probably via the cytidine nucleotide pathway using 1,2-DG as a metabolic intermediate. Hydrolysis of [14C]phosphatidylcholine revealed that fatty acids were labeled at both the C-1 (approximately 43%) and C-2 (approximately 57%) positions. On the other hand, a temporal association between decreased amounts of 14C-label in ceramides and increased amounts of 14C-label in sphingomyelin and galactocerebrosides supports the hypothesis that peripheral nerve galactocerebroside is derived, in vivo, from ceramide via acylation of sphingosine. This exclusive labeling of endoneurial lipids by endoneurial microinjection of labeled precursor provides a unique model for studying synthesis and metabolic turnover of membrane lipids in experimental neuropathies.  相似文献

Soybean seedlings were grown at 28°C under dark or light conditions for 12 days. Non-polar lipids (NPL) were separated by silicic acid column chromatography from total lipids in epicotyl containing young leaves, hypocotyl and root. The glyceride (TG, DG, and MG), free fatty acid (FFA) and sterol lipid (SE) components in NPL were analyzed mainly by thin-layer and gas-liquid chromatographies (TLC and GLC).

During germination, the amounts of polar lipids (PL) markedly increased in the tissues of soybean seedlings, especially in light-grown seedlings, whereas these of NPL increased slightly or maintained constant values. The features of the compositions and changing patterns of NPL in the tissues were more clarified in light-grown seedlings than in dark-grown ones. The pattern of change in fatty acid composition was similar in TG and 1,2-DG, which showed higher proportions of linoleic and linolenic acids, whereas FFA, 1,3-DG or MG had high proportions of saturated fatty acids. These results indicate that the compositions and changing patterns of NPL and their fatty acids in the tissues depend on the differences under two germinating conditions tested.  相似文献

Green and white leaves of the barley mutant line `albostrians' were compared for their polar lipid content and fatty acid composition. The mutant plastids of the white leaves have a double-layered envelope, but in contrast with the normal chloroplasts, lack 70 S ribosomes and thylakoids. In the green leaves, the amount of monogalactosyldiacylglycerol (MGDG) consistently exceeds the amount of digalactosyldiacylglycerol (DGDG) and the amount of galactolipids exceeds the amount of phospholipids. In contrast, in white leaves the amount of DGDG exceeds the amount of MGDG and the amount of phospholipids exceeds the amount of galactolipids. In white leaves, the galactolipid composition reflects the plastid envelope composition which is rich in DGDG, whereas in green leaves the galactolipid composition reflects the thylakoid composition which is rich in MGDG. These results demonstrate the likelihood that all the enzymes involved in galactolipid, sulfolipid and fatty acid synthesis are coded by the nuclear genome.  相似文献

The effect of light intensity upon galactolipid synthesis in Vicia faba leaf tissue was studied at two CO2 concentrations, 0.03 and 1%. The rates of galactolipid synthesis were estimated by determining the amount of radioactivity in each of the two galactoses of digalactosyl diacylglycerol (DGDG) and the single galactose of monogalactosyl diacylglycerol (MGDG), a technique based upon the accepted pathway for galactolipid synthesis in which galactosylation is the terminal step in biosynthesis. The results suggest that the rates of MGDG and DGDG synthesis were similar under all conditions and that galactolipid synthesis was not directly affected by light intensity. The quantity of radioactivity incorporated into the galactoses of individual molecular species of MGDG and DGDG were similar under the light conditions used.  相似文献

A study is reported on the incorporation of14C-acetate into lipid classes from three different growth stages ofLaminaria japonica, a species long used for food in Japan. This was done because of the possible utilization of its lipids.Radioactivity incorporated into whole lipids in the three growth stages under the same experimental conditions (10 °C, 500 lux) increased with maturity of the thalli. The radioactivity was found mainly in PC, TG and 1,2-DG and subsequently distributed into other lipid classes (PG,PI,PE,MGDG,SQDG and DGDG) to a lesser extent. The incorporation patterns of the former group were similar at all stages, but those of the latter group differed slightly according to growth stage.In juvenile thalli,14C was incorporated to a much higher extent into PG, MGDG, PI and fucosterol than PE, SQDG, DGDG and MG, while the14C-incorporation into MG, SQDG, DGDG and PS in the mature growth stage was higher than into the other lipid classes. The absolute level of incorporation was higher for all these compounds in mature thalli than the thalli of other growth stages.  相似文献

The model for export of fatty acids from plastids proposes that the acyl-ACP (acyl carrier protein) product of de novo fatty acid synthesis is hydrolyzed in the stroma by acyl-ACP thioesterases and the free fatty acid (FFA) released is then transferred to the outer envelope of the plastid where it is reactivated to acyl-CoA for utilization in cytosolic glycerolipid synthesis. Experiments were performed to assess whether the delivery of nascent FFA from the stroma for long chain acyl-CoA synthesis (LACS) occurs via simple diffusion or a more complex mechanism. The flux through the in vivo FFA pool was estimated using kinetic labeling experiments with spinach and pea leaves. The maximum half-life for FFA in the export pool was < or =1 s. Isolated pea chloroplasts incubated in the light with [14C]acetate gave a linear accumulation of FFA. When CoASH and ATP were present there was also a linear accumulation of acyl-CoA thioesters (plus derived polar lipids), with no measurable lag phase (<30 s), indicating that the FFA pool supplying LACS rapidly reached steady state. The LACS reaction was also measured independently in the dark after in situ generated FFA had accumulated yielding estimates of LACS substrate-velocity relationships. Based on these experiments the LACS reaction with in situ generated FFA as substrate is only about 3% of the LACS activity required in vivo at the very low concentrations of the FFA export pool calculated from the in vivo experiment. Furthermore, bovine serum albumin rapidly removed in situ generated FFA from chloroplasts, but could not compete effectively for "nascent" FFA substrates of LACS. Together the data suggest a locally channeled pool of exported FFA that is closely linked to LACS.  相似文献

Light-induced changes in the membrane lipid compositions were studied in pea leaves and in protoplasts and a plasmamembrane-enriched fraction (PMEF)* of pea leaves. PC, PE, PI, PG, PA, MGDG, DGDG and SL were identified as the glycerolipids. The relative levels of various membrane lipids changed due to light-induced greening. There was an increase in the galactolipids of leaves and leaf protoplasts. The galactolipid constituent of the PMEF was very low and showed no change. Among the plasmamembrane phospholipids, PI increased with a concomitant decrease in PC.  相似文献

Abstract: Protein kinase C (PKC) is activated by 1,2- sn -diacylglycerol (DAG), the source of which can either be phosphatidylinositol bisphosphate or phosphatidylcholine. Here, we show that monogalactosyl diglyceride (MGDG), a minor galactolipid present in oligodendrocytes (OLs) and myelin, which is designated as a marker for myelination, can enhance OL PKC activity. Based on different calcium and substrate requirements we conclude that MGDG and DAG activate different isoforms of PKC group A: MGDG primarily stimulates PKC-α, and DAG primarily activates PKC-γ. The presence of these PKC isoforms in OLs was confirmed by western blotting, whereas PKC-β was only weakly stained, if at all. Addition of MGDG to the culture medium provided a higher density of regenerating OL fibers, which was not observed when membrane-permeable DAG was used. These findings indicate that MGDG can modulate the OL PKC activity and that PKC-α is the major PKC isoform involved in OL process formation.  相似文献

Acetone powders of a 20,000g pellet fraction from spinach leaves (Spinacia oleracea L.) synthesized [4-(14)C]cholesteryl esters when incubated with [4-(14)C]cholesterol. The reaction was inhibited by digitonin. There was a reciprocal relationship between the decline of label in cholesterol and its incorporation into cholesteryl ester, indicating that free cholesterol was the direct precursor for cholesteryl ester biosynthesis. The hydrolysis of cholesteryl [1-(14)C]palmitate into free cholesterol and [1-(14)C]palmitate was not detected in these acetone powder preparations. Exogenous cholesteryl palmitate had no effect on the esterification of [4-(14)C]cholesterol. The data indicate that an esterase-type mechanism was not involved in the biosynthesis of these steryl esters. Label from [1-(14)C]palmitoyl-CoA was incorporated into steryl esters when incubated with spinach leaf acetone powder preparations. The optimal buffer for steryl ester biosynthesis was 2-(N-morpholino)ethanesulfonate and the optimal pH was 6. Iodoacetamide, N-ethylmaleimide, and dithiothreitol had no effect on the esterification reaction. Ethylenediaminetetraacetate, MgCl(2), CaCl(2), MnCl(2), and ZnSO(4) inhibited at concentrations of 10 to 30 mm.  相似文献
