A novel UV-induced mutant strain of recombinant Bacillus subtilis MUR1 was used for the production of l-LA in continuous cultures with a variety of culture conditions. The maximal productivity of 17.6 g/L/h was obtained with a l-LA concentration of 44.1 g/L at the dilution rate of 0.4 h−1. The highest concentration of l-LA (77.1 g/L) was produced at the dilution rate of 0.05 h−1. This study showed that the maximum l-LA productivity of B. subtilis MUR1 which can only last for a very short period of time during the exponential phase in fed-batch cultures, can be extended indefinitely at steady state in continuous cultures. l-LA production increased with the increase of yeast extract concentrations in the medium. Moreover, temperature, agitation rate and various glucose concentrations in the feed were compared in continuous cultures. Different nitrogen sources (lysine, glutamine, ammonium sulphate and corn steep liquor) were studied to partly or completely replace yeast extract in the medium, most of them showed positive effects on l-LA production and cell growth. The l-LA productivities from continuous cultures in this study are higher than the productivity of current microbial industrial processes which use Lactobacillus to produce l-LA.  相似文献

We compared the growth responses of the floating-leaved species Nymphoides peltata to gradual and rapid rising water levels under two nutrient concentrations (1 g and 12 g of slow released fertilizer (N-P-K: 16-8-12) per container filled with 8 kg washed sand), and predicted the population expansion after these floods. The results showed that the capacity for petiole elongation was dependent on leaf age, and only leaves that were no more than five days old had the capability to reach the water surface when the water level increased rapidly from 50 cm to 300 cm. Plants subjected to a gradual rising water level tracked the increase in water depth whose petioles elongated at 3.96 ± 1.70 cm per day and 4.80 ± 0.16 cm per day under low and high nutrient concentrations respectively throughout the experiment period. When water levels were rapidly raised, leaf petioles elongated rapidly at 25.48 ± 1.51 cm per day and 26.64 ± 2.24 cm per day under low and high nutrient concentrations respectively during the first ten days. Plants under a constant water level maintained highest mean leaf recruitment (mean 3.0 ± 0.33 leaves and 24.4 ± 5.87 leaves every ten days under low and high nutrient concentrations, respectively). Therefore, more young leaves existed in the canopy ensuring that when the water level increases, young leaves can rapidly emerge after submergence. Gradual water level rise did not significantly affect biomass and ramet production (4.75 ± 1.41 g and 5.50 ± 1.22 ramets in low nutrient; 48.49 ± 21.45 g and 35.67 ± 11.78 ramets in high nutrient), but rapid water level rise negatively affected ramet production in both nutrient concentrations (3.00 ± 1.26 ramets and 11.25 ± 4.19 ramets in low and high nutrients, respectively). The results indicated that continual leaf recruitment and rapid petiole elongation were both important ways in which N. peltata adapted to increasing water levels. Extreme flooding may be a disturbance factor that affects plant growth and the population expansion of N. peltata, while small gradual water level rise should not harm this species.  相似文献

Annual wild rice (Zizania aquatica L.), a species of conservation concern, is an ecologically and culturally important aquatic grass found in stands in the near shore habitats of lakes and rivers in the Midwest and along the eastern coast of North America. This study examined the effects of water depth and seed provenance on the early growth of three Indiana wild rice stands (collected from two lakes) under greenhouse conditions in 2009. Plants were grown at water depths of 46 cm, 23 cm, 0 cm, or −15 cm and harvested either at the first floating leaf stage or at 48 days after transplanting. Wild rice growth was affected by both water depth and seed provenance. The dry weight of roots, stems, leaves, and inflorescences, total biomass, number of tillers, number of leaves, and total leaf area were the lowest in the −15 cm treatment. These vegetative growth parameters also decreased with increasing water depth from the 0 cm treatment. Differences in growth between seed sources were found, supporting the hypothesis that genetic differences among relatively isolated wild rice stands may influence the success of efforts to conserve this species.  相似文献

The effects of N and P enrichment were investigated on growth and physiological responses of dwarf Avicennia marina mangroves in a hypersaline (58 ± 8 psu) field site in Richards Bay, South Africa. It was hypothesized that at high salinities mangroves allocate more resources to roots than shoots, and that nutrient enrichment with N and P will shift resource allocation to shoots and enhance growth and productivity. In unvegetated areas of the dwarf zone, 1-year-old A. marina seedlings were planted in pots and enriched bimonthly with N, P, N + P, or remained unfertilized (control-C), and growth and morphology of plants were monitored for 2 years. Enrichment with N and N + P shifted resource allocation to shoots from 38% to 55%, and increased dry biomass accumulation by over 500%, compared to the control treatment. In the N and N + P treatments, plant height, number of leaves, leaf chlorophyll content and photosynthesis increased by over 50%, 330%, 30% and 30%, respectively, compared to the C and P treatments. Enrichment with N and N + P increased N concentrations in roots by over 60% (from 1.0 ± 0.1% to 1.6 ± 0.2% of dry mass) and in shoots by over 100% (from 1.3 ± 0.1% to 2.7 ± 02% of dry mass). Plants enriched with P alone were similar to those of the control. This study has demonstrated that dwarf A. marina in Richards Bay is N limited, and that N enrichment shifts resource allocation from roots to shoots and increases growth and productivity.  相似文献

Two dense meadows of the seagrass Thalassodendron ciliatum (Forssk.) den Hartog were sampled during the Indonesian—Dutch Snellius II expedition to Eastern Indonesia. Production data were obtained from one of these meadows. The production of leaf biomass was measured by the leaf marking technique of Zieman and by the plastochrone interval method. The two methods reached comparable results. The production of leaf tissue was 4.2 mg ADW shoot?1 day?1. The production of rhizome biomass was calculated in a similar way, based on the plastochrone interval of rhizome nodes. The production of the meadow, exclusive of the production of roots and fruits, amounted to 4.5 g ADW m?2 day?1. A significant correlation between the growth rates of rhizomes and leaves was observed. Gametophyte strains originating from indigenous sporophytes of Undaria pinnatifida (Harvey) Suringar in Iwate Prefecture, Northeast Japan, were maintained for 9–10 months at 45 μmol photons m−2 s−1. Before cryopreservation in liquid nitrogen for more than 12 h (1–14 days) using a two-step cooling method with a mixture of cryoprotectants (10% l-proline and 10% glycerol), these were pre-incubated for 2, 4 and 8 months at 15 μmol photons m−2 s−1. After 1 week of thawing, no surviving gametophytes were detected in the strains without pre-incubation, but both the female and male gametophytes, pre-incubated for more than 4 months, showed high survival rates (43–60% for females and 64–100% for males). This revealed the induction of freezing tolerance by incubation at low irradiance. Thereafter, sporophytes derived from cryopreserved gametophytes and subcultured gametophytes, stored under pre-incubation conditions, were formed from the strain, and a morphological comparison was conducted with 10 characters (stipe length, stipe wet weight, blade length, blade wet weight, blade width, incision depth, blade thickness, sporophyll length, sporophyll wet weight, and sporophyll width). The morphology of the sporophytes formed from the cryopreserved gametophytes corresponded well with that of the subcultured gametophytes from the same strain. The results suggest that the cryopreservation method is applicable for preserving culture stocks of U. pinnatifida to be used in mariculture.  相似文献

An analysis of leek leaf development and expansion was carried out over three seasons using field-grown plants of three varieties which were directly sown at different dates or transplanted from controlled conditions. In all cases, successive leaves appeared (tip visible) at equal intervals of accumulated temperature. Detailed analysis of a single sowing in 1985 showed that the regularity of leaf appearance was a consequence of the coordinated response to accumulated temperature of leaf initiation (plastochron 100°C days > 0°C) and leaf blade and sheath extension. For each successive leaf, an additional 32°C days were required between initiation and appearance to allow for the linear increase in ‘sheath’ height, giving a phyllochron of 132°C days. Direct measurement of leaf extension before and after leaf appearance, and of the length of the leaf extension zone, confirmed that the rate of leaf extension, in terms of accumulated tempeature, was constant, and independent of leaf number. However, there were differences between seasons and between varieties in the responses of leaf appearance, leaf extension and ‘sheath’ length to accumulated temperature. It was concluded that the simple ontogenetic increase in leaf dimensions, which was a feature of all the crops studied, was a consequence of the progressive increase in the duration of leaf expansion.  相似文献

Initiation and development of grass leaves and tillers are oftendescribed individually with little attention to possible interrelationshipsamong organs. In order to better understand these interrelationships,this research examined epidermal cell division during developmentaltransitions at the apical meristem of tall fescue (Festuca arundinaceaSchreb.). Ten seedlings were harvested each day for a 9-d period,and lengths of main shoot leaves and primary tillers were measured.In addition, numbers and lengths of epidermal cells were determinedfor 0·5 mm segments along the basal 3 mm of each leafand tiller. Primordia development and onset of rapid leaf elongationwere characterized by an increase in the number of cells perepidermal file with mean cell length remaining near 20 µmper cell. After the leaf had lengthened to 1-1·5 mm,cells near the leaf tip ceased dividing and increased in length,at which time leaf elongation rate increased rapidly. Liguleformation, marking the boundary between blade and sheath cells,occurred prior to leaf tip emergence above the whorl of oldersheaths, while the earliest differentiation between blade andsheath cells probably began when leaves were < 1 mm long.Major transitions in leaf and tiller development appeared tobe synchronized among at least three adjacent nodes. At theoldest node, cessation of cell division in the leaf sheath wasaccompanied by initiation of cell division and elongation inthe associated tiller bud. At the next younger node the ligulewas being initiated, while at the youngest node cell divisioncommenced in the leaf primordium, as elongation of a new leafblade began. A double-blind, randomized, parallel and active-controlled clinical study was conducted to evaluate the anti-hypertensive effect as well as the tolerability of Olive leaf extract in comparison with Captopril in patients with stage-1 hypertension. Additionally, this study also investigated the hypolipidemic effects of Olive leaf extract in such patients. It consisted of a run-in period of 4 weeks continued subsequently by an 8-week treatment period. Olive (Olea europaea L.) leaf extract (EFLA®943) was given orally at the dose of 500 mg twice daily in a flat-dose manner throughout the 8 weeks. Captopril was given at the dosage regimen of 12.5 mg twice daily at start. After 2 weeks, if necessary, the dose of Captopril would be titrated to 25 mg twice daily, based on subject's response to treatment. The primary efficacy endpoint was reduction in systolic blood pressure (SBP) from baseline to week-8 of treatment. The secondary efficacy endpoints were SBP as well as diastolic blood pressure (DBP) changes at every time-point evaluation and lipid profile improvement. Evaluation of BP was performed every week for 8 weeks of treatment; while of lipid profile at a 4-week interval. Mean SBP at baseline was 149.3 ± 5.58 mm Hg in Olive group and 148.4 ± 5.56 mm Hg in Captopril group; and mean DBPs were 93.9 ± 4.51 and 93.8 ± 4.88 mm Hg, respectively. After 8 weeks of treatment, both groups experienced a significant reduction of SBP as well as DBP from baseline; while such reductions were not significantly different between groups. Means of SBP reduction from baseline to the end of study were −11.5 ± 8.5 and −13.7 ± 7.6 mm Hg in Olive and Captopril groups, respectively; and those of DBP were −4.8 ± 5.5 and −6.4 ± 5.2 mm Hg, respectively. A significant reduction of triglyceride level was observed in Olive group, but not in Captopril group. In conclusion, Olive (Olea europaea) leaf extract, at the dosage regimen of 500 mg twice daily, was similarly effective in lowering systolic and diastolic blood pressures in subjects with stage-1 hypertension as Captopril, given at its effective dose of 12.5-25 mg twice daily.  相似文献

The antifungal activities of cinnamon oil, clove oil, anise oil, and peppermint oil, and their main components (cinnamaldehyde, eugenol, trans-anethole, and menthol, respectively) against molds identified from areca palm leaf sheath (Mucor dimorphosporus, Penicillium sp., Aspergillus niger, and Rhizopus sp.) were investigated. An agar dilution method was employed to determine the minimum inhibitory concentration (MIC) of essential oils and their main components. Zone inhibition tests and the inhibitory effect of the leaf sheath dip-treated with essential oils against those molds were examined. Major components of essential oils on the leaf sheath during storage were quantified by gas chromatography analysis. The MIC values of essential oils on agar and on the leaf sheath were identical. With an MIC of 50 ??g ml−1, cinnamon oil had the strongest inhibitory effect. At their MICs the oils were capable of providing protection against mold growth on the leaf sheath for at least 12 weeks during storage at 25 °C and 100% RH. Scanning electron microscope examination showed that essential oils prevented spore germination. Except for menthol in peppermint oil, the main components of the essential oils, which were fairly stable over the storage period, largely contributed to the antifungal activity.  相似文献

Females generally avoid selecting sites for oviposition which have a high predation risk to increase offspring survival. Previous studies have focused on costs to ovipositing females. However, although offspring may also incur costs by being oviposited at low predation risk sites, no studies have focused on costs to offspring. Such costs to offspring were examined by using Aquarius paludum insularis, females of which avoid eggs parasitism by ovipositing at deep sites. Deep sites are safe from egg parasitism but may be unsuitable for hatching due to environmental factors. We examined the costs to offspring at deep sites by comparing the hatching rate, the duration to hatching and the proportion of drowned larvae between eggs that were set at three levels of water depth (0 cm, 25 cm and 50 cm depth). While the hatching rate at 50 cm was lower than that at 0 cm, the rate at 25 cm did not differ from that at 0 cm. Duration to hatching and the proportion of drowned larvae did not differ between the three depths. It is suggested that the declining survival rate of A. paludum eggs was due to increased water pressure at greater depth. Such a cost may exist in other species and such an observation may aid in understanding oviposition site selection.  相似文献

Hypericum perforatum L. has become one of the leading plant-based dietary supplements worldwide and its biological activities have been mainly attributed to hypericin and phenolic contents. The present study was conducted to determine chemical and morphological variability of H. perforatum sampled from different locations of Northern Turkey. The populations were investigated according to the hypericin, chlorogenic acid and the flavonoids, namely rutin, hyperoside, apigenin-7-O-glucoside, quercitrin and quercetin contents and morphological characters including leaf dark gland density, leaf area, leaf length/width ratio and plant height. Significant chemical and morphological variations were detected among the populations. Hypericin content among populations ranged between 0.44 and 2.82 mg/g dry weight, chlorogenic acid between 0.0 and 1.86 mg/g dry weight, rutin between 0.0 and 8.77 mg/g dry weight, hyperoside between 5.41 and 22.28 mg/g dry weight, quercitrin between 1.64 and 3.98 mg/g dry weight and quercetin between 1.01 and 1.76 mg/g dry weight. Hypericin content was found to be correlated positively with leaf dark gland density, however, negatively with leaf area and no correlation was detected between the other morphological traits and bioactive substances examined.  相似文献

Green turtles Chelonia mydas of immature and adult size (n = 19, curved carapace length 49 to 118 cm) were equipped with time-depth recorders for short periods (≤ 7 d) to investigate diel and seasonal variation in diving behaviour. Research sessions were distributed over 2 years to cover seasonal variation in sea temperature from 14 °C to 30 °C. Diurnal dives were shallower and shorter than nocturnal dives, with diel patterns also evident in dawn and dusk peaks in occupation of depths within 1 m of the surface, elevated diurnal occupation of depths 1 to 2 m below the surface and elevated nocturnal occupation of depths > 2 m. Dive duration increased as sea temperature decreased, showing strong negative correlation by day and by night. Study turtles made resting dives that were 3 to 4 times longer in median duration, and six times longer in maximum duration, at cool temperatures than they were at warm temperatures, but there was no evidence of winter diapause or location shift to avoid cold water. The large majority of turtles spent 89 to 100% of their time at depths ≤ 5 m below the surface, three individuals did not exceed 3 m and the maximum depth recorded by any turtle was 7.9 m, although deeper water was available. Furthermore, the dive data indicated that study turtles collectively spent more than 80% of their time at charted (low tide) depths of 3 m or less, indicating that they consistently used the shallow margins of the bay where human activities tend to be concentrated, thereby potentially increasing their exposure to anthropogenic threats.  相似文献

One of the largest contiguous seagrass ecosystems in the world is located on the shallow continental shelf adjacent to the west coast of Florida, USA and is comprised of seasonally ephemeral Halophila decipiens meadows. Little is known about the demography of the west Florida shelf H. decipiens, which may produce 4.56 × 108 g C day−1 or more during the peak growing season. We documented seagrass distribution, biomass, and productivity, and density of sediment seed reserves, seedlings, flowers and fruits on the southeastern portion of the west Florida shelf by sampling along a transect at three stations in 10, 15, and 20 m water depth. Biomass, flower, fruit, seedling, and seed bank densities tended to be highest at stations in 10–15 m water depth and lowest at 20 m. Flowers and fruit were most prevalent during summer cruises (June and August 1999, July 2000). Seedling germination occurred during summer, fall (October 1999), and winter (January 2000) sampling events, with the highest seedling densities present during the winter. Seed bank density remained consistent through time. A Category I hurricane with sustained winds of 120 km h−1 passed over the stations, but only limited impact on H. decipiens biomass was observed. The presence of a persistent seed bank provides for recovery after storm disturbance, annual reestablishment of populations, and continual maintenance of the 20,000 km2 of deep water seagrass habitat present on the west Florida shelf.  相似文献

The brown alga Laminaria japonica is distributed from southern Hokkaido to the northeastern Honshu in Japan. Recently, aquaculture of L. japonica has expanded to the southern coast of Japan and to China along the East China Sea. In order to elucidate the growth, biomass and productivity of L. japonica in a subtropical area, we cultivated and examined it in the Uwa Sea, in southwestern Japan over a period of 2 years. The seawater temperature ranged from 13.8 to 26.8 °C in 2001/2002 and from 13.1 to 27.2 °C in 2002/2003. In 2001/2002, the maximum density, maximum mean length and maximum mean wet wt. of L. japonica were 59.7 ± 28.0 ind. 50 cm− 1 (mean ± S.D.), 187.5 ± 82.7 cm (360 cm in the largest individual) and 130.1 ± 94.6 g wet wt., respectively. In 2002/2003, these values were 94.7 ± 22.2 ind. 50 cm− 1, 159.3 ± 74.4 cm (300 cm in the largest individual) and 95.2 ± 69.5 g wet wt., respectively. Thus, the length and weight increased when the density was low (2001/2002), and the length and weight decreased when the density was high (2002/2003). The maximum biomass was estimated to be 7200 ± 3400 g wet wt. 50 cm− 1 in 2001/2002 and 7300 ± 2000 g wet wt. 50 cm− 1 in 2002/2003. Annual production was estimated to be 33.3 kg wet wt. m− 1 year− 1 in 2001/2002 and 34.0 kg wet wt. m− 1 year− 1 in 2002/2003. The present study indicates that the annual production of L. japonica per rope of 1 m at Uwajima Bay, the Uwa Sea corresponded to 1.1-2.2 m2 of that of Hokkaido in their native area. Thus, the present study indicates that L. japonica is highly adaptable because it is able to keep a high level of productivity when grown in water with a high temperature.  相似文献

The partitioning of photosynthetically fixed carbon betweencarbohydrate fractions and the processes of export and storagewere compared in mature leaf blades and sheaths of the grassPoa pratensis L. Most of the fixed carbon was destined for exportfrom the leaf blade with only 1% of the carbon fixed duringthe photoperiod being stored after 24 h. Although most of theassimilates imported to the sheath from the blade were subsequentlyexported, there was some unloading and storage of assimilates.Autoradiography was used to compare the translocation of 14C-labelledassimilates through non-fed areas of leaf blade and sheath andrevealed that the veins in the sheath showed a greater capacityfor storage of assimilates compared to the leaf blade. Biphasickinetics of sucrose and glucose uptake were observed in segmentsof leaf blade and sheath. Although similar carriers for eachof the sugars appear to exist in the blade and sheath, the rateof uptake via these carriers was significantly lower in thesheath compared to the blade. Assuming that unloading proceedsvia a symplastic pathway, it would appear that the conversionof sucrose to starch in the sheath could be an important meansof regulating unloading and in determining sink strength ofthe sheath. It is concluded that although the net amount ofsugars unloaded in the sheath is small, the storage of assimilatesin the vein network could be an important means of bufferingchanges in sucrose concentration in the translocate during periodsof fluctuating assimilation. The immediate precursor of L-ascorbic acid, or vitamin C, is 2-keto-l-gulonic acid (2-KLG). This is commonly produced commercially by Ketogulonicigenium vulgare and Bacillus megaterium, using corn steep liquor powder (CSLP) as an organic nitrogen source. In this study, the effects of the individual CSLP components (amino acids, vitamins, and metal elements) on 2-KLG production were evaluated, with the aim of developing a complete, chemically defined medium for 2-KLG production. Forty components of CSLP were analyzed, and key components were correlated to biomass, 2-KLG productivity, and consumption rate of L-sorbose. Glycine had the greatest effect, followed by serine, biotin, proline, nicotinic acid, and threonine. The combination of 0.28 g L−1 serine, 0.36 g L−1glycine, 0.18 g L−1 threonine, 0.28 g L−1proline, 0.19 g L−1nicotinic acid, and 0.62 mg L−1biotin in a chemically defined medium produced the highest maximum biomass concentration (4.2 × 109 cfu mL−1), 2-KLG concentration (58 g L−1), and yield (0.76 g g−1) after culturing for 28 h.  相似文献

The effects of temperature, water level and burial depth on seed germination of two submerged species, Myriophyllum spicatum and Potamogeton malaianus, were investigated under controlled laboratory conditions. There was no significant difference in final germination of M. spicatum among water level treatments, but P. malaianus germinations at 1 cm and 12 cm water levels were better than at 0 cm water level at temperatures of 20 °C and 30 °C. Little to no germination was observed for either species at the temperature of 10 °C. At 15 °C, however, germination increased significantly to 66.3-70.6% for M. spicatum and to 29.4-48.1% for P. malaianus under all three water level treatments. Increased temperature from 15 °C to 30 °C had no significant effect on the final germination of M. spicatum except at the 1 cm water level, but enhanced significantly the germination of P. malaianus. Analysis of the mean time to germination revealed that M. spicatum was a faster germinator relative to P. malaianus. The two species’ germination differed markedly in response to burial depth. Germination percentage of M. spicatum was 71.3% at 0 cm burial depth, but decreased to 5.0% and to 2.5% at depths of 1 cm and 2 cm, respectively; whereas germination percentages of P. malaianus were 40.0%, 23.8%, 12.5%, 7.5% and 1.3% at depths of 0 cm, 1 cm, 2 cm, 3 cm and 5 cm, respectively. We concluded that the two species respond differently to germination strategies. The findings provided further insight into how germination strategy contributes to the seed bank formation and species invasion.  相似文献

Coral catsharks (Atelomycterus marmoratus) are kept at the Aquarium of Cologne Zoo in a public exhibit tank measuring 300 x 160 x 110 cm (length x width x water height) with a water volume of ca. 5000 l. The 1.3 adults have been producing fertilized eggs since early 2010. Deposited eggs are longish-oval in shape, with a length of about 10 cm. Juveniles hatched in our rearing tanks (measuring 100-210 x 45-80 x 30-50 cm, with 280-430 l water volume) after periods of 4-6 months. The freshly hatched coral catsharks measured from 10-13 cm in total length. After 3 months, each juvenile had grown about 4-5 cm in length and the distinct, bright color pattern of the juveniles had faded towards the more subtle adult colour pattern. The keeping and breeding of coral catsharks can be classified as uncomplicated, given that sufficient swimming space and hiding places are provided.
