Despite several studies on the attachment ability of different insect taxa, little is known about this phenomenon in adult Lepidoptera. In this study we combined morphological and experimental analyses of tarsal adhesive devices and the attachment ability of the codling moth Cydia pomonella (L.) (Lepidoptera, Tortricidae) to smooth surfaces. Pretarsi of C. pomonella attach to smooth substrates by means of their smooth, flexible and well developed arolia. Using the centrifugal force measurement technique, friction forces of males and females were assessed on hydrophobic and hydrophilic glass surfaces. Adults of both sexes generated similar forces in spite of the noticeable difference in their body masses. That is why males showed significantly higher safety factors (attachment force divided by body weight) compared to those of females. Hydrophobicity of the substrate had no considerable effect on friction forces. For females, friction forces (sliding parallel to the substrate plane) were compared with adhesive forces (pulling off perpendicularly from the substrate plane) measured on Plexiglas surfaces. It can be concluded that the attachment system of C. pomonella is rather robust against physico-chemical properties of the substrate and is able to achieve a very good attachment on vertical and horizontal substrata.  相似文献

Host plant surfaces of the codling moth, Cydia pomonella L. (Lepidoptera, Tortricidae), vary in microtopography, which can affect its attachment, locomotion, and oviposition behaviour. This study was performed to investigate the effect of surface roughness on the attachment ability of adult insects. Using a centrifugal force device, friction forces of both sexes were assessed on six epoxy resin substrates differing only in the dimensions of their surface asperities, ranging from 0 μm to 12 μm. Surface topography significantly affected friction forces. Maximal force was measured on the smooth substrate whereas minimal force was assessed on microrough substrates with 0.3 μm and 1.0 μm size of asperities. On the remaining rough substrates, friction forces were significantly higher but still lower than on the smooth substrate. Both sexes generated similar forces on the same substrate, in spite of the considerable difference in their body mass. Thus, it is expected that both sexes can attach effectively to differently structured plant substrates in their habitat. However, since smooth surfaces have been reported previously to be the most favorable substrates for ovipositing females of C. pomonella, it is possible that they use their attachment system to sense the substrate texture and prefer those substrates to which their arolia attach the best.  相似文献

The attachment of a marine Pseudomonas sp. to a variety of surfaces was investigated, and the number of bacteria which became attached was related to the surface charge and degree of hydrophobicity of the substratum. Large numbers of bacteria attached to hydrophobic plastics with little or no surface charge [Teflon, polyethylene, polystyrene, poly(ethylene terephthalate)]; moderate numbers attached to hydrophilic metals with a positive (platinum) or neutral (germanium) surface charge; and very few attached to hydrophilic, negatively charged substrata (glass, mica, oxidized plastics). The results suggest that both electrostatic and hydrophobic interactions are involved in bacterial attachment.  相似文献

Many representatives of the beetle family Chrysomelidae exhibit a distinctive sexual dimorphism in the structure of adhesive tarsal setae. The present study demonstrates the influence of surface roughness on the friction force of Leptinotarsa decemlineata males and females. The maximum friction force of individual beetles was measured on epoxy resin surfaces (smooth and with asperities ranging from 0.3 to 12.0 microm) using a centrifugal force tester. On the smooth surface, no considerable differences between males and females were found, whereas on rough surfaces, females attached significantly (up to two times) stronger than males. Clawless beetles generated lower forces than intact ones, but demonstrated similar differences between males and females. The results indicate that the female adhesive system has its main functional trait in a stronger specialisation to rough plant surfaces whereas the adhesive system of males possess a certain trade-off between attachment to rough plant surfaces during locomotion on vegetation and to the smooth surface of the female elytra, while mating.  相似文献

Adhesion of Chlorella vulgaris(chlorophyceae), Nitzschia amphibia(bacillariophceae) and Chroococcus minutus(cyanobacteria) to hydrophobic (perspex, titanium and stainless steel 316-L), hydrophilic (glass) and toxic (copper, aluminium brass and admiralty brass) substrata were studied in the laboratory. The influence of surface wettability, surface roughness, pH of the medium, culture age, culture density, cell viability and presence of organic and bacterial films on the adhesion of Nitzschia amphibia was also studied using titanium, stainless steel and glass surfaces. All three organisms attached more on titanium and stainless steel and less on copper and its alloys. The attachment varied significantly with respect to exposure time and different materials. The attachment was higher on rough surfaces when compared to smooth surfaces. Attachment was higher on pH 7 and above. The presence of organic film increased the attachment significantly when compared to control. The number of attached cells was found to be directly proportional to the culture density. Attachment by log phase cells was significantly higher when compared to stationary phase cells. Live cells attached more when compared to heat killed and formalin killed cells. Bacterial films of Pseudomonas putida increased the algal attachment significantly. %  相似文献

Insects live in a three-dimensional space, and need to be able to attach to different types of surfaces in a variety of environmental and behavioral contexts. Adult leaf beetles possess great attachment ability due to their hairy attachment pads. In contrast, their larvae depend on smooth pads to attach to the same host plant. We tested friction forces generated by larvae and adults of dock leaf beetles Gastrophysa viridula on different rough surfaces, and found that adults generate much higher attachment to various substrates than larvae, but are more susceptible to completely losing attachment ability on surfaces with “critical” roughness. Furthermore, sex-specific setal morphology has the effect that attachment forces of male adults are generally higher than those of females when adjusted for body weight. The results are discussed in the context of development, ecology, and changing behavioral strategies of successive life stages.  相似文献

The proteolytic enzymes pronase, trypsin, and chymotrypsin and the surfactant Triton X-100 inhibited attachment of Vibrio proteolytica to the hydrophobic substratum polystyrene by >97%. These treatments had no effect on attachment to hydrophilic substrata such as glass or tissue culture dishes. Both pronase and Triton X-100 effected the removal of previously attached cells from polystyrene but not from hydrophilic surfaces. Removal of cells from polystyrene by pronase left material (which we have termed footprints) that stained with the protein-specific stain Hoechst 2495 but not with the DNA-specific stain Hoechst 33342. Pronase treatment also caused a significant decrease in cell surface hydrophobicity as determined by phase partitioning in hexane or petroleum ether. Collectively, these results imply the existence of separate mechanisms for the adhesion of V. proteolytica to hydrophilic and hydrophobic substrata and suggest a role for protein in the latter mechanism.  相似文献

Several studies have shown that the initial surface wettability, is of importance in the settlement of macrofouling larvae such as barnacles, bryozoans and hydroids in the field as well as in laboratory assays. In this study we present results from laboratory assays using hydrophilic and hydrophobic polystyrene (PS) and cyprid larvae of Balanus improvisus (Darwin). The results obtained differ markedly from those reported for the barnacle Balanus amphitrite (Darwin), where a high surface wettability seemed to be preferred for settlement. Our results show that a surface with intermediary wettability (hydrophilic PS) reduced settlement by 38% as compared to surfaces of low wettability (hydrophobic PS) during an 8-day period. During the experiment, the wettability in the hydrophilic PS dishes was not significantly changed as measured by advancing contact angle with mQ water. Over an 8-day period wettability of the hydrophobic PS dishes approached that of the hydrophilic PS surfaces. We further conducted experiments with highly hydrophilic and highly hydrophobic methylsilane-treated glass surfaces with known chemistry. In this experiment, the settlement of cyprid larvae was completely inhibited by the high wettability surfaces. Contact angle measurements revealed that the wettability during the length of the experiment of the hydrophilic glass surfaces was not significantly altered. We conclude by these experiments that even an intermediate wettability can significantly affect the overall settlement success of the barnacle B. improvisus. The mechanism by which the settlement is impeded might be biologically mediated through the recognition by cyprid larvae of the molecular composition of the surface when the cyprid reverts to the settlement phase, i.e. when swimming behaviour is abandoned in favour of surface exploration, or it is mediated by physicochemical forces acting between the surface and the larval body or the larval antennules.  相似文献

In crayfish, phenoloxidase was located in the hemocytes. The plasma had infinitesimal enzyme activity. A phenoloxidase preparation from hemocytes precipitated spontaneously after approximately 1.5 hr at 22°C, which became attached spontaneously to glass, Plexiglas, and polystyrene plastic. The enzyme preparation could also become attached to Saccharomyces cerevisiae cell walls. Attachment was mediated by a proteinaceous substance, since trypsin significantly decreased the degree of attachment. Calcium ions were also necessary for attachment. A β-1,3-glucan, laminaran, partially prevented attachment to the fungal cell walls. Heparin caused precipitation of the phenoloxidase preparation from hemocytes. In crayfish cuticle, proteins with associated phenoloxidase activity were attached to cell walls of Aphanomyces astaci as well as to those of S. cerevisiae.  相似文献

The southern green stink bug Nezara viridula L. (Heteroptera, Pentatomidae) is highly polyphagous, preferring apically situated seeds and fruits on more than 150 plant species belonging to over 30 plant families all over the world. This forces them to move over highly variable terrains, including plant stems, leaves, pods and buds, which requires efficient attachment. Stink bugs have long slender legs and feet (tarsi) equipped with paired curved claws, paired soft adhesive pads (pulvilli), and flattened lanceolate hairs (setae), which arise ventrally on the first and second foot segments (tarsomeres). To characterize their attachment abilities on well‐defined test substrates, here we comparatively measured and analyzed the traction forces of bugs walking horizontally and vertically on hydrophilic (water attractive) and hydrophobic (water repellent) glass plates and rods. The latter correspond to the geometry of preferred feeding sites of stink bugs in the field. The results show a clear contribution of tarsal flattened lanceolate hairs to the stink bug's attachment. Higher traction forces are generated on a glass rod than on a glass plate, corresponding to up to individual maximum of 43 times the stink bug's body weight. Substrate hydrophobicity promotes the attachment, while the measured forces are up to eight times lower when tarsal hairs are disabled. The combination of smooth and hairy tarsal pads results in a remarkable attachment ability, which enables N. viridula to climb unstable apical plant parts, and supports their invasive behavior and global dispersion.  相似文献

Giardia lamblia cell populations show 90% detachment from glass under normal forces of 2.43+/-0.33 nN applied by centrifugation. Detachment forces were not significantly different for cells attached to positively charged, hydrophobic, or inert surfaces than for cells attached to plain glass. Escherichia coli O157 are an important group of foodborne pathogens with the ability to attach to materials commonly used in food processing environments such as slightly hydrophilic stainless steel. The aim of this study was to characterise six E. coli isolates, including five E. coli O157, for curli production, autoaggregation, hydrophobicity and attachment to highly hydrophilic glass and hydrophobic Teflon®. Curli production and autoaggregation were determined using absorbance assays; hydrophobicity by bacterial adherence to hydrocarbons, hydrophobic interaction chromatography and contact angle measurements; and attachment using epifluorescence microscopy. Curli production varied between strains and for some strains correlated with autoaggregation. Curli production correlated with decreased hydrophobicity for two strains. Four of the six isolates increased attachment to glass, but decreased attachment to Teflon® with increased curli production. In contrast, one of the six isolates decreased attachment to glass, but increased attachment to Teflon® with increasing curli production. Curli production by the remaining isolate did not correlate with hydrophobicity or attachment. Attachment of some E. coli, including E. coli O157, to abiotic surfaces may be influenced by curli production, autoaggregation and hydrophobicity. However, for other strains, a variety of factors may be of greater influence on these properties and ability to attach to abiotic surfaces. This study highlights the complexity of bacterial surface properties and their relationship with bacterial attachment.  相似文献

The relationships among surface energy, adsorbed organic matter, and attached bacterial growth were examined by measuring the degradation of adsorbed ribulose-1,5-bisphosphate carboxylase (a common algal protein) by attached bacteria (Pseudomonas strain S9). We found that surface energy (work of adhesion of water) determined the amount and availability of adsorbed protein and, consequently, the growth of attached bacteria. Percent degradation of adsorbed ribulose-1,5-bisphosphate carboxylase decreased with increasing hydrophobicity of the surface (decreasing work of adhesion). As a result, growth rates of attached bacteria were initially higher on hydrophilic glass than on hydrophobic polyethylene. However, during long (6-h) incubations, growth rates increased with surface hydrophobicity because of increasing amounts of adsorbed protein. Together with previous studies, these results suggest that the number of attached bacteria over time will be a complex function of surface energy. Whereas both protein adsorption and bacterial attachment decrease with increasing surface energy, availability of adsorbed protein and consequently initial bacterial growth rates increase with surface energy.  相似文献

Using a surface force apparatus, we have measured the normal and friction forces between layers of the human glycoprotein lubricin, the major boundary lubricant in articular joints, adsorbed from buffered saline solution on various hydrophilic and hydrophobic surfaces: i), negatively charged mica, ii), positively charged poly-lysine and aminothiol, and iii), hydrophobic alkanethiol monolayers. On all these surfaces lubricin forms dense adsorbed layers of thickness 60–100 nm. The normal force between two surfaces is always repulsive and resembles the steric entropic force measured between layers of end-grafted polymer brushes. This is the microscopic mechanism behind the antiadhesive properties showed by lubricin in clinical tests. For pressures up to ~6 atm, lubricin lubricates hydrophilic surfaces, in particular negatively charged mica (friction coefficient μ = 0.02–0.04), much better than hydrophobic surfaces (μ > 0.3). At higher pressures, the friction coefficient is higher (μ > 0.2) for all surfaces considered and the lubricin layers rearrange under shear. However, the glycoprotein still protects the underlying substrate from damage up to much higher pressures. These results support recent suggestions that boundary lubrication and wear protection in articular joints are due to the presence of a biological polyelectrolyte on the cartilage surfaces.  相似文献

Broad-leaved dock, Rumex obtusifolius L. (Polygonaceae), is the main host plant of the green dock beetle, Gastrophysa viridula Degeer (Coleoptera: Chrysomelidae). Adult beetles are able to attach and walk on leaves of this plant. Leaf surface is rather uneven, because of irregularly shaped prominent epidermal cells with a maximum height of about 9 µm. The surface is covered with a smooth epicuticular wax layer having relatively low free surface energy (FSE). The aim of this study was to measure beetle attachment force applying a 'centrifugal technique' on adult insects on the leaf surface and other substrates, in order to understand the effect of surface architecture and its physicochemical properties on insect attachment. We compared forces on an adaxial leaf surface with forces on a smooth silanized glass plate having low FSE, and on a polishing paper having slightly lower FSE and similar surface roughness (asperities' size = 9 µm). Smooth plate made of normal untreated clean glass with high FSE was used as a reference substrate. On the leaf surface and the polishing paper, attachment forces were lower compared to both glass samples. There was no significant difference between force values on leaves and the polishing paper, but on both glass surfaces they were significantly higher than on the other substrates. Hydrophobicity alone explains a decrease in attachment force of the beetle, but when combined with roughness the decrease in force is four times greater.  相似文献

Intraspecific interactions among ectosymbionts may occur when suitable attachment sites are limited to specific parts of their hosts. In this study, we examined attachment site selection by the ectosymbiotic chironomid Nanocladius (Plecopteracoluthus) asiaticus. Larvae and pupae of this chironomid attach to larvae of the megalopteran Protohermes grandis: they feed on algae and detrital materials caught on the chironomid's silken nets and on the body surface of the host. Sampling at approximately monthly intervals in central Japan revealed that the proportion of hosts with chironomids was relatively high and stable during the 2 year study period. When attached singly, larvae were usually found on the ventral side of the host's mesothorax. However, when two or more larvae were attached to a host, the mesothorax was occupied by only one larva (usually a large individual) and the other(s) were attached to the ventral side of the abdomen. Density dependent shifts in attachment sites were confirmed by a reattachment experiment in which a larva was removed from its host and released onto a new host with or without symbiosis by another individual. All of the larvae released singly on a host devoid of symbionts attached to the thoracic region and survived, whereas those released in pairs and not occupying this site were frequently lost. Thus, the thorax (particularly the mesothorax) is the most suitable attachment site, and there is a possibility that chironomid larvae compete for this site.  相似文献

The rose sawfly, Arge ochropus (Gmelin), is one of the most destructive pests attacking rose bushes in northern Iran. In the present study, the haemocyte reactions of A. ochropus larvae were assessed against two entomopathogenic nematodes, Steinernema carpocapsae (Weiser) and Heterorhabditis bacteriophora Poinar. With injection of infective juveniles of H. bactriophora into A. ochropus larvae, the total haemocyte count showed alternating fluctuations at all time intervals. Encapsulation occurred 22 hours post injection (hpi) of H. bacteriophora infective juveniles into the rose sawfly larvae, while melanization was observed after 24 h. In the case of S. carpocapsae, initial attachment of the haemocytes was detected at 18 hpi and complete encapsulation at 24 hpi; no melanization was observed. In general, findings showed strong immune responses of the rose sawfly larvae against H. bacteriophora, while these reactions were weakened for S. carpocapsae. Therefore, it is concluded that S. carpocapsae has more ability to overcome the host defence system, and this research provides a new window on its potential as a biocontrol agent.  相似文献

A simple method for rendering atomic force microscope tips and cantilevers hydrophilic or hydrophobic through glow discharge in an appropriate gas atmosphere is introduced. Force curves at different humidities of these modified cantilevers were taken on freshly cleaved mica (hydrophilic surface) and on a monolayer of dipalmitoylphosphatidylethanolamine transferred onto mica (hydrophobic surface) to characterize the behavior of the cantilevers on hydrophilic and hydrophobic surfaces. Furthermore, Langmuir-Blodgett bilayers, with a dipalmitoylphosphatidylethanolamine bottom layer and a dipalmitoylphosphatidylcholine top layer, were imaged in the constant force mode in a multimode atomic force microscope in air under controlled humidity conditions. The friction and elasticity signal were recorded parallel to the topography. By varying the force exerted by the tip on the sample, different layers of the Langmuir-Blodgett system could be removed or flattened. Removal exposed underlying layers that exhibited a different friction and elasticity behavior. Furthermore, force scans with tips rendered hydrophobic were taken on the different layers of the sample to characterize the hydrophilic/hydrophobic nature of the layers. Only by combining the results obtained by the different methods can the structure of the lipid layer systems be identified.  相似文献

Conidia of the plant pathogenic fungus Botrytis cinerea adhered to tomato cuticle and to certain other substrata immediately upon hydration. This immediate adhesion occurred with both living and nonliving conidia. Adhesion was not consistently influenced by several lectins, sugars, or salts or by protease treatment, but it was strongly inhibited by ionic or nonionic detergents. With glass and oxidized polyethylene, substrata whose surface hydrophobicities could be conveniently varied, there was a direct relationship between water contact angle and percent adhesion. Immediate adhesion did not involve specific conidial attachment structures, although the surfaces of attached conidia were altered by contact with a substratum. Freshly harvested conidia were very hydrophobic, with more than 97% partitioning into the organic layer when subjected to a phase distribution test. Percent adhesion of germinated conidia was larger than that of nongerminated conidia. Evidence suggests that immediate adhesion of conidia of B. cinerea depends, at least in part, on hydrophobic interactions between the conidia and substratum.  相似文献

Many animals that locomote by legs possess adhesive pads. Suchorgans are rapidly releasable and adhesive forces can be controlledduring walking and running. This capacity results from the interactionof adhesive with complex mechanical systems. Here we presentan integrative study of the mechanics and adhesion of smoothattachment pads (arolia) in Asian Weaver ants (Oecophylla smaragdina).Arolia can be unfolded and folded back with each step. Theyare extended either actively by contraction of the claw flexormuscle or passively when legs are pulled toward the body. Regulationof arolium use and surface attachment includes purely mechanicalcontrol inherent in the arrangement of the claw flexor system. Predictions derived from a ‘wet’ adhesion mechanismwere tested by measuring attachment forces on a smooth surfaceusing a centrifuge technique. Consistent with the behavior ofa viscid secretion, frictional forces per unit contact arealinearly increased with sliding velocity and the increment stronglydecreased with temperature. We studied the nature and dimensions of the adhesive liquidfilm using Interference Reflection Microscopy (IRM). Analysisof ‘footprint’ droplets showed that they are hydrophobicand form low contact angles. In vivo IRM of insect pads in contactwith glass, however, revealed that the adhesive liquid filmnot only consists of a hydrophobic fluid, but also of a volatile,hydrophilic phase. IRM allows estimation of the height of theliquid film and its viscosity. Preliminary data indicate thatthe adhesive secretion alone is insufficient to explain theobserved friction and that rubbery deformation of the pad cuticleis involved.  相似文献
