This study investigates the effects of lifelong exposure to reversed geomagnetic and zero geomagnetic fields (the latter means absence of geomagnetic field) on the life history of Daphnia carinata King from Australia and Daphnia magna Straus from Europe. Considerable deviation in the geomagnetic field from the usual strength, leads to a decrease in daphnia size and life span. Reduced brood sizes and increased body length of neonates are observed in D. magna exposed to unusual magnetic background. The most apparent effects are induced by zero geomagnetic field in both species of Daphnia. A delay in the first reproduction in zero geomagnetic field is observed only in D. magna. No adaptive maternal effects to reversed geomagnetic field are found in a line of D. magna maintained in these magnetic conditions for eight generations. Integrally, the responses of D. magna to unusual geomagnetic conditions are more extensive than that in D. carinata. We suggest that the mechanism of the effects of geomagnetic field reversal on Daphnia may be related to differences in the pattern of distribution of the particles that have a magnetic moment, or to moving charged organic molecules owing to a change in combined outcome and orientation of the geomagnetic field and Earth's gravitational field. The possibility of modulation of self-oscillating processes with changes in geomagnetic field is also discussed.  相似文献

Background: Low Helicobacter pylori eradication rates are common in pediatric trials especially in developing countries. The aim of the study was to investigate the role of antibiotic resistance, drug dosage, and administration frequency on treatment outcome for children in Vietnam. Materials and Methods: Antibiotics resistance of H. pylori was analyzed by the Etest in 222 pretreatment isolates from children 3–15 years of age who were originally recruited in a randomized trial with two treatment regiments: lansoprazole with amoxicillin and either clarithromycin (LAC) or metronidazole (LAM) in two weight groups with once‐ or twice‐daily administration. The study design was an observational study embedded in a randomized trial. Results: The overall resistance to clarithromycin, metronidazole, and amoxicillin was 50.9%, 65.3%, and 0.5%, respectively. In LAC, eradication was linked to the strains being susceptible to clarithromycin (78.2% vs 29.3%, p = .0001). Twice‐daily dosage of proton‐pump inhibitor (PPI) and clarithromycin was more effective for eradication than once‐daily dosage for resistant strains (50.0% vs 14.7%, p = .004) and tended to be so also for sensitive strains (87.5% vs 65.2%, p = .051). Exact antibiotic dose per body weight resulted in more eradication for resistant strains (45.3% vs 8.0%, p = .006). These differences were less pronounced for the LAM regimen, with twice‐daily PPI versus once daily for resistant strains resulting in 69.2% and 50.0% eradication (p = .096), respectively. Conclusions: Helicobacter pylori clarithromycin resistance was unexpectedly high in young children in Vietnam. Clarithromycin resistance was an important cause for eradication treatment failure. Twice‐daily administration and exact antibiotic dosing resulted in more eradicated infections when the strains were antibiotic resistant, which has implications for the study design in pediatric H. pylori eradication trials.  相似文献

Background: The prevalence of antibiotic resistance varies in geographic areas. The information on the antibiotic susceptibility patterns of Helicobacter pylori (H. pylori) in our local setting is therefore relevant as a guide for the treatment options. Objective: This study was conducted to determine the primary resistance rates among H. pylori isolated from Malaysian patients. Materials and methods: Biopsy samples were obtained from the stomach antrum and corpus of 777 patients from September 2004 until 2007. H. pylori isolated from these patients were then subjected to minimum inhibitory concentration (MICs) determination using E‐test method, against metronidazole, clarithromycin, levofloxacin, ciprofloxacin, amoxicillin, and tetracycline. Results: From 777 patients, 119 were positive for H. pylori where a total of 187 strains were isolated. The resistance rates were noted to be 37.4% (metronidazole), 2.1% (clarithromycin), 1% (levofloxacin and ciprofloxacin), and 0% (amoxicillin and tetracycline). Different resistance profiles were observed among isolates from the antrum and corpus of 13 patients. Resistance to one type of antibiotic was observed in 36.4% of the strains where mono‐resistance to metronidazole was the most common. Resistance to ≥2 antibiotics was noted in 3.3% of isolates. High metronidazole MICs of ≥256 μg/mL were observed among the resistant strains. Conclusions: The resistance rates of the antibiotics used in primary treatment of H. pylori infections in Malaysia are low, and multi‐antibiotic‐resistant strains are uncommon. Infections with mixed populations of metronidazole‐sensitive and ‐resistant strains were also observed. However, the high metronidazole MIC values seen among the metronidazole‐resistant strains are a cause for concern.  相似文献

Compared with other developed countries, vancomycin‐resistant enterococci (VRE) are not widespread in clinical environments in Japan. There have been no VRE outbreaks and only a few VRE strains have sporadically been isolated in our university hospital in Gunma, Japan. To examine the drug susceptibility of Enterococcus faecalis and nosocomial infection caused by non‐VRE strains, a retrospective surveillance was conducted in our university hospital. Molecular epidemiological analyses were performed on 1711 E. faecalis clinical isolates collected in our hospital over a 6‐year period [1998–2003]. Of these isolates, 1241 (72.5%) were antibiotic resistant and 881 (51.5%) were resistant to two or more drugs. The incidence of multidrug resistant E. faecalis (MDR‐Ef) isolates in the intensive care unit increased after enlargement and restructuring of the hospital. The major group of MDR‐Ef strains consisted of 209 isolates (12.2%) resistant to the five drug combination tetracycline/erythromycin/kanamycin/streptomycin/gentamicin. Pulsed‐field gel electrophoresis analysis of the major MDR‐Ef isolates showed that nosocomial infections have been caused by MDR‐Ef over a long period (more than 3 years). Multilocus sequence typing showed that these strains were mainly grouped into ST16 (CC58) or ST64 (CC8). Mating experiments suggested that the drug resistances were encoded on two conjugative transposons (integrative conjugative elements), one encoded tetracycline‐resistance and the other erythromycin/kanamycin/streptomycin/gentamicin‐resistance. To our knowledge, this is the first report of nosocomial infection caused by vancomycin‐susceptible MDR‐Ef strains over a long period in Japan.  相似文献

It is known that magnetic fields affect ants behavior. It has been shown that Solenopsis ants are sensitive to magnetic fields but there is no experimental evidence for magnetic orientation. In this paper experiments were done to verify the magnetic orientation of Solenopsis sp. ants. The spontaneous orientation of ants in a circular arena was studied in two different magnetic conditions: in the natural geomagnetic field and under an altered magnetic field, with the horizontal geomagnetic axis shifted in 90?o. Our results show that ants consistently change their orientation direction when the magnetic field was altered. Axial circular statistics analysis showed that, in the absence of other cues, ants orient spontaneously to the horizontal geomagnetic field axis. The present paper shows for the first time magnetic orientation in Solenopsis sp. ants.  相似文献

The purpose of the work was to further investigate the effect of zero magnetic field (ZMF) on the concentration of ions in the human blood compared to the effect of the normal geomagnetic field (GMF). We have investigated the total Zn and Cu concentrations in the blood serum during in vitro aging of blood. The investigation was carried out both on blood from healthy donors as well as from chronic bronchial asthma (BA) patients. Blood samples were kept for 48 hours in a Helmholtz coil compensating system to remove the static component of the geomagnetic field, at room temperature. We found that zinc concentrations in the plasma were not significantly influenced by the exposure to ZMF compared to GMF for both healthy and pathological samples. In contrast, copper concentration was found to be significantly sensitive to the magnetic environment. Healthy blood showed a slight loss of copper from the blood serum in GMF, which further increased in ZMF. BA pathology is characterized by four distinct types of disease, which showed both qualitative and quantitative distinctive sensitivity to the magnetic environment, as compared to healthy blood. The aging effect appeared to be slowed down for most of the BA types of pathologies. These results point to the sensitivity of ion binding to serum proteins and/or transport through cell membranes in the magnetic environment, in our case in the absence of the normal geomagnetic field.  相似文献

The spread of bacterial antibiotic resistance mutations is thought to be constrained by their pleiotropic fitness costs. Here we investigate the fitness costs of resistance in the context of the evolution of multiple drug resistance (MDR), by measuring the cost of acquiring streptomycin resistance mutations (StrepR) in independent strains of the bacterium Pseudomonas aeruginosa carrying different rifampicin resistance (RifR) mutations. In the absence of antibiotics, StrepR mutations are associated with similar fitness costs in different RifR genetic backgrounds. The cost of StrepR mutations is greater in a rifampicin‐sensitive (RifS) background, directly demonstrating antagonistic epistasis between resistance mutations. In the presence of rifampicin, StrepR mutations have contrasting effects in different RifR backgrounds: StrepR mutations have no detectable costs in some RifR backgrounds and massive fitness costs in others. Our results clearly demonstrate the importance of epistasis and genotype‐by‐environment interactions for the evolution of MDR.  相似文献

Summary The resistance of 8 strains of Bacillus sphaericus and of 2 strains of Bacillus thuringiensis var. israelensis (B.t.i.) to various antibiotics and antibiotic combinations were tested. All B. sphaericus strains were resistant to streptomycin, lincomycin and bacitracin, and six strains were resistant to combinations of these antibiotics. This antibiotic resistance could be utilised to establish selective media to identify and follow the fate of B. sphaericus and of B.t.i. in the field.  相似文献

A total of 813 Salmonella strains isolated from raw wastewater and stool specimens of inpatient children, living in the wastewater-spreading field of Marrakesh city, were examined for their susceptibility to 15 antimicrobial agents. All the isolates were susceptible to cefotaxime, and almost of them showed susceptibility to gentamicin (99.88%), trimethoprim-sulphamethoxazole (98.04%), nalidixic acid (98.04%), kanamycin (97.30%), trimethoprim (97.18%), and chloramphenicol (96.07%). The highest levels of antibiotic resistance were obtained for cephalothin (29.27%), amoxicillin (26.44%), sulphamethoxazole (26.07%), and ampicillin (25.21%). The strains from the serogroup B showed the highest antibiotic resistance frequencies. The percentage of polyresistant strains (36.09%) was significantly higher than that of monoresistant isolates (15.49%). The incidence of drug resistance in Salmonella isolates from stools was significantly higher than in those isolated from wastewater.  相似文献

Magnetotactic bacteria produce nanometer‐size intracellular magnetic crystals. The superior crystalline and magnetic properties of magnetosomes have been attracting much interest in medical applications. To investigate effects of intense static magnetic field on magnetosome formation in Magnetospirillum magneticum AMB‐1, cultures inoculated with either magnetic or non‐magnetic pre‐cultures were incubated under 0.2 T static magnetic field or geomagnetic field. The results showed that static magnetic field could impair the cellular growth and raise Cmag values of the cultures, which means that the percentage of magnetosome‐containing bacteria was increased. Static magnetic field exposure also caused an increased number of magnetic particles per cell, which could contribute to the increased cellular magnetism. The iron depletion in medium was slightly increased after static magnetic field exposure. The linearity of magnetosome chain was also affected by static magnetic field. Moreover, the applied intense magnetic field up‐regulated mamA, mms13, magA expression when cultures were inoculated with magnetic cells, and mms13 expression in cultures inoculated with non‐magnetic cells. The results implied that the interaction of the magnetic field created by magnetosomes in AMB‐1 was affected by the imposed magnetic field. The applied static magnetic field could affect the formation of magnetic crystals and the arrangement of the neighboring magnetosome. There is an expectation that in a near future, there may be "zero tolerance" towards antibiotic-based selection and production systems. Besides the antibiotic itself, the antibiotic resistance gene is an important consideration. Non-O1/non-O139 nontoxigenic Vibrio cholerae associated with cholera-like diarrhea has been reported in Kolkata, India. However, the property involved in the pathogenicity of these strains has remained unclear. The character of 25 non-O1/non-O139 nontoxigenic V. cholerae isolated during 8 years from 2007 to 2014 in Kolkata was examined. Determination of the serogroup showed that the serogroups O6, O10, O35, O36, O39, and O70 were represented by two strains in each serogroup, and the remaining isolates belonged to different serogroups. To clarify the character of antibiotic resistance of these isolates, an antibiotic resistance test and the gene analysis were performed. According to antimicrobial drug susceptibility testing, 13 strains were classified as drug resistant. Among them, 10 strains were quinolone resistant and 6 of the 13 strains were resistant to more than three antibiotics. To define the genetic background of the antibiotic character of these strains, whole-genome sequences of these strains were determined. From the analysis of these sequences, it becomes clear that all quinolone resistance isolates have mutations in quinolone resistance-determining regions. Further research on the genome sequence showed that four strains possess Class 1 integrons in their genomes, and that three of the four integrons are found to be located in their genomic islands. These genomic islands are novel types. This indicates that various integrons containing drug resistance genes are spreading among V. cholerae non-O1/non-O139 strains through the action of newly generated genomic islands.  相似文献

Aims

To evaluate the phenotypic and genotypic profiles of Campylobacter spp. from poultry faecal samples from free range or intensively raised meat chickens and free range egg layers. In addition, a case‐comparison study of antibiotic resistance genes from different groups of poultry and some pig strains previously collected was carried out.

Methods

Resistance to different antibiotics was assessed using the agar dilution method. In addition, all the strains were tested for ampicillin (blaOXA‐61), erythromycin (aph‐3‐1), tetracycline tet(O), streptomycin (aadE), and the energy‐dependent multi‐drug efflux pump (cmeB) resistance genes using multiplex polymerase chain reaction.

Results

The evaluation of phenotypic resistance revealed all of the strains from poultry were sensitive to ciprofloxacin, gentamicin, erythromycin or tylosin. But, widespread resistance to lincomycin (51–100%), extensive resistance to ampicillin (33·3–60·2%) and less resistance to tetracycline (5·6–40·7%) were observed in the different groups of chickens. Antibiotic resistance genes blaOXA‐61, cmeB and tet(O) were found in 82·6–92·7%, 80·3–89% and 22·3–30·9% Camp. coli isolates from pigs, whilst 59–65·4% and 19·2–40·7% Camp. jejuni from chickens were found to encode blaOXA‐61 and tet(O), respectively.

Conclusion

No significant difference between isolates from free range egg layers and meat chickens (P < 0·05) was found. However, there were significant differences between the pig strains and all the groups of poultry strains (P < 0·05) with regard to carriage of resistance genes. In addition, pulsed field gel electrophoresis of selected resistant isolates from the poultry and pig revealed closely related clonal groups.

Significance and Impact of the study

Our results suggest the resistant strains are persisting environmental isolates that have been acquired by the different livestock species. Furthermore, the different treatment practices in poultry and pigs have resulted in differences in resistance profiles in Campylobacter isolates.  相似文献

Plasmid transformation of three thermotolerant strains of methylotrophic bacterium Methylophilus spp. was investigated using three different methods. The plasmids could be transformed by electroporation and were stabily maintained in these methylotrophic bacteria. The calcium chloride (competent cell) method was successful only in the case of one of the three strains studied. Protoplast transformation of methylotrophic bacteria was not successful. The antibiotic resistance markers conferred resistance on the transformants, whereas the wild-type bacteria were sensitive to the antibiotics.  相似文献

Abstract Uptake of [3H]dihydrostreptomycin in sensitive and resistant strains of cowpea rhizobia was examined to explain one of the possible mechanisms of intrinsic antibiotic resistance. The rate of accumulation of labelled streptomycin in resistant strains was drastically reduced when compared to that of sensitive strains. Our results indicated that high levels of intrinsic antibiotic resistance was due to decreased permeability causing failure to accumulate the drug.  相似文献

Aims: Aim of this study is to characterize clinical isolates of Salmonella Typhimurium that occurred in Portuguese children on the basis of their virulence and antimicrobial resistance profiles and pulsed‐field gel electrophoresis typing and to analyse possible strain relatedness. Methods and Results: Different Salmonella serotypes were isolated from clinical cases of salmonellosis that had occurred in two Portuguese hospitals (a total of 259 isolates). All Salm. Typhimurium strains, with the age of the patients known, (total of 26 isolates) were selected for this study. These isolates were characterized for their virulence gene profiles (agfA, iroB, slyA, hin/H2, spv), antimicrobial resistance profiles and investigated for the occurrence of multidrug‐resistant Salm. Typhimurium DT 104 by PCR. Salmonella isolates showed high rates of resistance to four or more antibiotics, 100% resistance to sulfadiazine and a high percentage of strains with the resistance profile of Salm. Typhimurium DT 104, two of them with this phage type (determined by PCR). A relationship between some clusters and their resistance and virulence profiles was detected, each cluster having the same profile. Conclusions: This study showed high‐antibiotic resistance of the Salmonella strains investigated, and the presence of multidrug‐resistant Salm. Typhimurium DT104 in infections of Portuguese children. Significance and Impact of the Study: Study is based on regarding the increase in antibiotic resistance by Salmonella strains isolated from infections in Portuguese children and on the presence of Salm. Typhimurium DT 104 circulating in Portugal.  相似文献

The fruitfly (Drosophila melanogaster) visual system was investigated electrophysiologically in vivo after exposure to a zero magnetic field (ZMF). Electroretinographic (ERG) recording of fly eye electrical activity was performed on adult insects raised from pupae maintained for 20 hr in zero magnetic field. A flickering excitation regime was applied to excite the visual system, since in this way, a quasistable hyperpolarization component of the electroretinogram can be obtained, containing information from the neural cells, which are the most sensitive to the action of external factors during early ontogenetic stages. Results of the investigation of two D. melanogaster populations, sample and control, were statistically compared.

We found a significant statistical increase of sensitivity in neural cells from the first optic ganglion in the fly population developed from pupae exposed to ZMF.  相似文献
