Dimethylsulfoniopropionate (DMSP) is produced by many species of marine phytoplankton and has been reported to provide a variety of beneficial functions including osmoregulation. Dinoflagellates are recognized as major DMSP producers; however, accumulation has been shown to be highly variable in this group. We explored the effect of hyposaline transfer in Gambierdiscus belizeanus between ecologically relevant salinities (36 and 31) on DMSP accumulation, Chl a, cell growth, and cell volume, over 12 d. Our results showed that G. belizeanus maintained an intracellular DMSP content of 16.3 pmol cell?1 and concentration of 139 mM in both salinities. Although this intracellular concentration was near the median reported for other dinoflagellates, the cellular content achieved by G. belizeanus was the highest reported of any dinoflagellate thus far, owing mainly to its large size. DMSP levels were not significantly affected by salinity treatment but did change over time during the experiment. Salinity, however, did have a significant effect on the ratio of DMSP:Chl a, suggesting that salinity transfer of G. belizeanus induced a physiological response other than DMSP adjustment. A survey of DMSP content in a variety of Gambierdiscus species and strains revealed relatively high DMSP concentrations (1.0–16.4 pmol cell?1) as well as high intrageneric and intraspecific variation. We conclude that, although DMSP may not be involved in long‐term (3–12 d) osmoregulation in this species, G. belizeanus and other Gambierdiscus species may be important contributors to DMSP production in tropical benthic microalgal communities due to their large size and high cellular content.  相似文献

The ubiquitous, biogenic trace gas dimethylsulfide (DMS) represents the largest natural source of atmospheric sulfur. Given DMS involvement in cloud formation and climate, understanding and parameterizing the oceanic DMS source and cycling processes is a necessary challenge. We report DMS cycling rates from microzooplankton dilution grazing experiments conducted monthly during 1 year in coastal northwestern Mediterranean waters. Concentrations of DMS, its algal precursor dimethylsulfoniopropionate (DMSPt) and chlorophyll a (Chla) ranged 0.9–11 nmol L?1, 10–71 nmol L?1, and 0.2–1.5 µg L?1, respectively. By comparing the growth and stock production rates of the DMSP-producing algae to those of total phytoplankton, we estimated that 3?±?4% (range 0.4–12%) of the carbon primary production was invested in DMSP biosynthesis. Microzooplankton grazing rates on DMSP-producing phytoplankton (0.46–1.45 day?1) were generally higher than those on the bulk assemblage (0.08–0.99 day?1), except in midsummer months. This could have been due to the smaller size of most DMSP producers. There was no indication of micrograzer selection against DMSP-containing phytoplankton, since they were not grazed at lower rates than the bulk phytoplankton assemblage. A proportion of 6–20% of the grazed DMSP was converted into DMS, and this grazing-derived production accounted for 32–96% of dark gross DMS production by the total community. Bacteria consumed daily?≤?14–100% of the gross DMS production, which resulted in biological DMS turnover times of 1 to?≥?10 days. Throughout the year, grazing-mediated DMS production explained 73% of the variance in the DMS concentration, implying that microzooplankton grazing plays a major role in controlling DMS concentration in surface waters across a broad range of environmental and productivity conditions in the Mediterranean Sea. These findings should help improve the representation of herbivore grazing in prognostic models to predict the distribution and dynamics of the global DMS emission and its feedback response to changing climate.  相似文献

Abstract. The β-dimethylsulphoniopropionate (DMSP) concentrations of 5 Antarctic green algae grown at 2, 30 and 55 μmol photons m?2 s?1 were determined during a period of one year, cultivated under fluctuating daylengths mimicking the conditions of the natural habitat at King George Island. Antarctica. The intracellular DMSP content of all species decreased simultaneously with decreasing daylengths and vice versa. Additionally, the DMSP level was affected by the light intensity: the higher the photon fluence rate the greater the algal DMSP concentration. Under conditions of darkness, there was a degradation of the DMSP pool in members of the Acrosiphoniales, while the DMSP content in members of the Ulvales did not change. The results indicate a light-dependent DMSP accumulation in algae. Therefore, they may help to explain the seasonal variability of DMSP and its cleavage product dimethylsulphide (DMS) in coastal waters.  相似文献

Products from phototrophic dinoflagellates such as toxins or pigments are potentially important for applications in the biomedical sciences, especially in drug development. However, the technical cultivation of these organisms is often problematic due to their sensitivity to hydrodynamic (shear) stress that is a characteristic of suspension-based closed photobioreactors (PBRs). It is thus often thought that most species of dinoflagellates are non-cultivable at a technical scale. Recent advances in the development of biofilm PBRs that rely on immobilization of microalgae may hold potential to circumvent this major technical problem in dinoflagellate cultivation. In the present study, the dinoflagellate Symbiodinium voratum was grown immobilized on a Twin-Layer PBR for isolation of the carotenoid peridinin, an anti-cancerogenic compound. Biomass productivities ranged from 1.0 to 11.0 g m?2 day?1 dry matter per vertical growth surface and a maximal biomass yield of 114.5 g m?2, depending on light intensity, supplementary CO2, and type of substrate (paper or polycarbonate membrane) used. Compared to a suspension culture, the performance of the Twin-Layer PBRs exhibited significantly higher growth rates and maximal biomass yield. In the Twin-Layer PBR a maximal peridinin productivity of 24 mg m?2 day?1 was determined at a light intensity of 74 μmol m?2 s?1, although the highest peridinin content per dry weight (1.7 % w/w) was attained at lower light intensities. The results demonstrate that a biofilm-based PBR that minimizes hydrodynamic shear forces is applicable to technical-scale cultivation of dinoflagellates and may foster biotechnological applications of these abundant marine protists.  相似文献

Bacterial species associated with the dimethylsulfoniopropionate (DMSP)-producing phytoplankton Scrippsiella trochoidea were cultured and identified, with the aim of establishing their ability to metabolise DMSP, dimethylsulfide (DMS) and dimethylsulfoxide (DMSO). Results demonstrate that of the cultivable bacteria only α-Proteobacteria were capable of producing DMS from DMSP. The concentration of DMSP was shown to affect the amount of DMS produced. Lower DMSP concentrations (1.5?μmol?dm?3) were completely assimilated, whereas higher concentrations (10?μmol?dm?3) resulted in increasing amounts of DMS being produced. By contrast to the restricted set of bacteria that metabolised DMSP,?~?70% of the bacterial isolates were able to ‘consume’ DMS. However, 98-100% of the DMS removed was accounted for as DMSO. Notably, a number of these bacteria would only oxidise DMS in the presence of glucose, including members of the γ-Proteobacteria and Bacteroidetes. The observations from this study, coupled with published field data, identify DMS oxidation to DMSO as a major transformation pathway for DMS, and we speculate that the fate of DMS and DMSP in the field are tightly coupled to the available carbon produced by phytoplankton.  相似文献

The Roseobacter clade of marine bacteria is often found associated with dinoflagellates, one of the major producers of dimethylsulfoniopropionate (DMSP). In this study, we tested the hypothesis that Roseobacter species have developed a physiological relationship with DMSP-producing dinoflagellates mediated by the metabolism of DMSP. DMSP was measured in Pfiesteria and Pfiesteria-like (Cryptoperidiniopsis) dinoflagellates, and the identities and metabolic potentials of the associated Roseobacter species to degrade DMSP were determined. Both Pfiesteria piscicida and Pfiesteria shumwayae produce DMSP with an average intracellular concentration of 3.8 μM. Cultures of P. piscicida or Cryptoperidiniopsis sp. that included both the dinoflagellates and their associated bacteria rapidly catabolized 200 μM DMSP (within 30 h), and the rate of catabolism was much higher for P. piscicida cultures than for P. shumwayae cultures. The community of bacteria from P. piscicida and Cryptoperidiniopsis cultures degraded DMSP with the production of dimethylsulfide (DMS) and acrylate, followed by 3-methylmercaptopropionate (MMPA) and methanethiol (MeSH). Four DMSP-degrading bacteria were isolated from the P. piscicida cultures and found to be taxonomically related to Roseobacter species. All four isolates produced MMPA from DMSP. Two of the strains also produced MeSH and DMS, indicating that they are capable of utilizing both the lyase and demethylation pathways. The diverse metabolism of DMSP by the dinoflagellate-associated Roseobacter spp. offers evidence consistent with a hypothesis that these bacteria benefit from association with DMSP-producing dinoflagellates.  相似文献

Elevated pCO2 and warming may promote algal growth and toxin production, and thereby possibly support the proliferation and toxicity of harmful algal blooms (HABs). Here, we tested whether empirical data support this hypothesis using a meta‐analytic approach and investigated the responses of growth rate and toxin content or toxicity of numerous marine and estuarine HAB species to elevated pCO2 and warming. Most of the available data on HAB responses towards the two tested climate change variables concern dinoflagellates, as many members of this phytoplankton group are known to cause HAB outbreaks. Toxin content and toxicity did not reveal a consistent response towards both tested climate change variables, while growth rate increased consistently with elevated pCO2. Warming also led to higher growth rates, but only for species isolated at higher latitudes. The observed gradient in temperature growth responses shows the potential for enhanced development of HABs at higher latitudes. Increases in growth rates with more CO2 may present an additional competitive advantage for HAB species, particularly as CO2 was not shown to enhance growth rate of other non‐HAB phytoplankton species. However, this may also be related to the difference in representation of dinoflagellate and diatom species in the respective HAB and non‐HAB phytoplankton groups. Since the proliferation of HAB species may strongly depend on their growth rates, our results warn for a greater potential of dinoflagellate HAB development in future coastal waters, particularly in temperate regions.  相似文献

Short-term experiments showed that ingestion rates of the copepods Acartia grani and Euterpina acutifrons on different concentrations of the dinoflagellate Gyrodinium corsicum were not usually different from those obtained with the non-toxic and similar sized Prorocentrum triestinum. Long-term experiments showed that no A. grani survived for more than 288 h on concentrated cultures (1500 μg C l−1) of G. corsicum compared with survival rates of 86.7% on the non-toxic P. triestinum. In contrast high and similar survival rates were found for E. acutifrons exposed for the same time to similar concentrations of both dinoflagellates. These results demonstrated that G. corsicum produce toxins which have significant adverse effects on the long-term survival rates of some copepods like A. grani but not on other copepods like E. acutifrons. The possibility of PSP-toxin production by G. corsicum was analysed by chromatographic analysis (HPLC-FD) and mouse bioassay of extracts obtained from cultures maintained in exponential growth phase in f/2-Si and L1 culture mediums. These analyses and the bioassay ruled out the presence of PSP toxins in this dinoflagellate. However, mouse tests provided the first evidence for the presence of some type of NSP toxin in G. corsicum. Haemolytic assays also gave the first positive results for the methanol extract of this dinoflagellate species.  相似文献

Experiments were conducted to determine how much grazing pressure adult females of Calanus pacificus Brodsky, a dominant planktonic herbivore in local waters, exerted upon a bloom of the dino- flagellate, Gymnodinium flavum Kofoid & Swezy, in the waters off La Jolla in July, 1980. One set of females was presented with water collected from the chlorophyll maximum, and a second set was presented with Thalassiosira weissflogii Grunow, which is readily ingested by Calanus. Filtration rates upon the diatom were significantly higher (8.4 ml · copepod?1 · h?1) than upon the dinoflagellate (0.30 ml · copepod?1 · h?1). Calanus did not exert a significant grazing pressure upon the dinoflagellate bloom. Gut content analyses support this conclusion. The persistence of the bloom was probably due, in part, to the avoidance of Gymnodinium flavum by copepod grazers and to the consequent lack of grazing pressure.  相似文献

The spatial distributions of dimethylsulfide (DMS), dimethylsulfoniopropionate (DMSP), DMSP-lyase activity (DLA) and their controlling factors including nutrients, phytoplankton community and bacterial abundance were investigated in the East China Sea (ECS) during fall from October 19 to November 2, 2015. Diatoms and dinoflagellates dominated the phytoplankton community, while other taxonomic groups were rare and mainly found in the oligotrophic open sea. Affected by the high nutrients concentrations, Chl a, DMS, DMSP and DLA showed high values in eutrophic inshore waters, and decreased from the costal zones to the open sea. Statistical analysis suggested that diatoms and dinoflagellates were the main controlling factors of DMS, DMSP, and DLA in ECS. For size-fractionated samples, a reduced contribution of the microplankton from inshore stations to offshore stations affected by the trophic conditions was noted. Meanwhile, this decrease in microplankton led to an increase in the ratio of DLA contributed by picoplankton and free-living bacteria from the estuary area to the offshore region. The DMS sea-to-air flux was calculated using the equation of Nightingale et al. (Glob Biogeochem Cy 14(1):373–387, 2000), and approximately 2.88 × 10?2 Tg of sulfur was transferred from the sea into the atmosphere in the form of DMS in ECS during fall.  相似文献

Coral reefs provide habitat for an array of marine invertebrates that host symbiotic microbiomes. Photosynthetic symbionts including Symbiodinium dinoflagellates and diatoms potentially influence the diversity of their host-associated microbiomes by releasing carbon-containing photosynthates and other organic compounds that fuel microbial metabolism. Here we used 16S ribosomal RNA (rRNA) gene amplicon pyrosequencing to characterise the microbiomes of 11 common Great Barrier Reef marine invertebrate species that host photosynthetic symbionts and five taxa in which they are absent. The presence of photosynthetic symbionts influenced the composition but not the species richness, evenness and phylogenetic diversity of invertebrate-associated microbiomes. Invertebrates without photosynthetic symbionts were dominated by Alphaproteobacteria, whereas those hosting photosynthetic symbionts were dominated by Gammaproteobacteria. Interestingly, many microbial species from photosymbiont-bearing invertebrates, including Oceanospirillales spp., Alteromonas spp., Pseudomonas spp., Halomonas spp., are implicated in the metabolism of dimethylsulfoniopropionate (DMSP). DMSP is produced in high concentrations by photosynthetic dinoflagellates and is involved in climate regulation by facilitating cloud formation. Microbiomes correlated with host taxa and replicate individuals from most sampled species grouped in distance-based redundancy analysis of retrieved 16S rRNA gene sequences. This study highlights the complex nature of invertebrate holobionts and confirms the importance of photosynthetic symbionts in structuring marine invertebrate bacterial communities.  相似文献

The southern Barents Sea is considered to be the most productive area in the Arctic Ocean; however, there are no assessments of daily production rates in the coastal waters. During the summer and autumn of 2007, we investigated the variation of mesozooplankton community structure relative to environmental conditions at 12 coastal stations. Copepods dominated the total zooplankton biomass and abundance during both periods. Diversity indices and the total biomass of zooplankton communities differed significantly between the two seasons. Cluster analyses revealed two distinct groups of stations which were associated with Ura Bay and the adjacent open sea, respectively. Daily production rates of the copepod species examined were calculated using three methods based on: (1) a temperature-dependent equation and (2) two multiple regressions that consider temperature, body weight, and chlorophyll a concentration. Significant seasonal differences for daily production rates were found using all three model equations (p?<?0.05): 358?±?188–1,775?±?791 versus 198?±?85–1,584?±?559?μg?dry?mass?m?3?day?1. Results of principal components analyses demonstrated that the abundance and biomass of herbivorous species were related to variation in chlorophyll a concentration while the abundance and biomass of other species (omnivorous copepods and Ctenophora) were related mainly with water temperature and salinity. Mesozooplankton biomass was higher during this study relative to previous studies. Computed copepod production rates were higher compared with other Arctic seas confirming a high productive potential of the coastal southern Barents Sea.  相似文献

Dinoflagellate algae of the genus Symbiodinium occur as endosymbionts in a variety of hosts including coral. The response of Symbiodinium spp. to environmental changes could dictate survival of their hosts and the ecological success of coral reef ecosystems. Oxidative stress has been linked to a breakdown in this symbiotic relationship, known as bleaching. Increased temperature is one of the primary environmental changes linked to this phenomenon. Preliminary studies have established high concentrations of the sulfur compound dimethylsulfoniopropionate (DMSP) in Symbiodinium spp., with increased temperature. To examine the potential use of DMSP as an antioxidant, a 5?day incubation experiment was conducted at two temperatures with the algae S. microadriaticum (CCMP1633) isolated from the cnidarian host Aiptasia pulchella. An HPLC assay for the activity of the enzyme B12-dependent methionine synthase was modified and used to determine the link between de novo production of methionine, a precursor to DMSP, and temperature induced oxidative stress. DMSP concentrations per cell increased approximately 38?% in the 33?°C treatment cultures over 120?h. However, these cells also increased more than 2-fold in biovolume (127?±?43?%), and SYTO-BC stain indicated increased DNA content (approximately 4-fold), suggesting arrested cell division. Normalization of DMSP to biovolume revealed that the concentrations actually decreased approximately 49?% after 2?days in cultures exposed to elevated temperature (33?°C), but were not significantly different from the control treatment at 120?h (27?°C). Concomitant changes in the 33?°C treatment relative to the control (after 120?h) resulted in an approximately 8-fold increase in reactive oxygen species, a 37?% (±7?%) decrease in photosynthetic efficiency of photosystem II, and a 5-fold increase in xanthophyll cycling. Methionine synthase activity (MSA) correlated to the decrease in DMSP concentration (R 2?=?0.778), with decreasing activity at the high temperature. Given this decrease in MSA, the increase in DMSP per cell may be due to DMSP production utilizing methionine from protein turnover, and not de novo synthesis via MSA. The findings of this study provide insight into the responses of algal symbionts to environmental changes, shed light on the potential use of DMSP and other known photo-protective mechanisms such as xanthophyll cycling under temperature induced oxidative stress, and support the suspected cessation of cell division under these conditions. This information could be crucial to understanding cellular responses to environmental changes and the ability of these organisms to survive under elevated sea surface temperatures projected for the near future.  相似文献

Diatoms and dinoflagellates not only have extensive distribution and a huge biomass in marine ecosystems, but also have high lipid accumulation in nature or after physiological and genetic modification, which indicates that these organisms may be optimal candidate algal strains for biodiesel production. In this study, we determined the content of intracellular neutral lipids (triacylglycerol [TAG]) in the dinoflagellate Prorocentrum micans and in the diatom Phaeodactylum tricornutum using NR and BODIPY 505/515 staining. The freshwater green alga Scenedesmus obliquus was used as a control. Optimum concentrations of 1.000 and 1.500 μg mL?1 were determined for neutral lipid Nile red (NR) staining in P. micans and P. tricornutum. Unlike NR staining, the optimal concentrations of BODIPY 505/515 staining in P. micans and P. tricornutum were lower, at 0.100 and 0.075 μg mL?1, respectively. High correlation coefficients of R 2?=?0.990 and R 2?=?0.989 were obtained for P. micans and P. tricornutum intracellular neutral lipid content and the relative fluorescence intensity with NR staining, while the reference alga, S. obliquus, had a relatively low correlation coefficient of R 2?=?0.908 when stained with NR. The neutral lipid content determined by thin-layer chromatography-flame ionization detector matched the analytical data from fluorescence measurements. These results indicated that NR and BODIPY 505/515 staining can be used as an excellent high-throughput approach to screen marine diatoms and dinoflagellates.  相似文献

Dinoflagellates are characterized by low maximum photosynthetic rates and high respiratory costs. Recent evidence also suggests that dinoflagellates are disproportionately abundant in the diets of many copepods. This suggests that at least some species are preferred prey types. This begs the question: 'How do dinoflagellates co-exist with other, seemingly competitively superior, algal taxa'? Their motility may enable them to maintain position better in the light-rich surface waters, and more successfully make the return journey into deeper waters in order to replenish their internal nutrient stores when surface nutrients become depleted. This theory is examined using a depth-resolved model of algal dynamics. Nutrients and organic detrital matter are represented on a Eulerian grid, whilst a Lagrangian approach is used to represent dinoflagellates and diatoms. The model indicates that strictly autotrophic dinoflagellates have difficulty balancing their metabolic budgets. Even in the absence of competition from diatoms, motility is almost essential to dinoflagellate persistence. Only extremely motile dinoflagellates are able to compete successfully with diatoms. In reality, an increasing number of dinoflagellate species are being found to be mixotrophic. This model suggests that mixotrophy may often be an obligatory, rather than optional, behaviour.   相似文献

The non-photosynthetic, phagotrophic dinoflagellate, Gymnodinium fungiforme Anissimova is attracted to a variety of amino acids and other organic compounds. Glycine, taurine and serine attracted the dinoflagellates at a threshold detection level of 10?8 M fallowed by dextrose (10?7 M) and alanine, proline and threonine (10?6 M). Glycine, taurine and alanine are three of the most abundant free amino acids found in invertebrates and protozoa which are major food sources of this dinoflagellate. Three additional species of cultured heterotrophic dinoflagellates were exposed to the water soluble fraction of a shrimp extract known to attract G. fungiforme. All three species responded to the extract, but one species, Oxyrrhis marina, did so only after changing its food source. It is suggested that chemosensory behavior may be suppressed or expressed depending on culture conditions.  相似文献

Dimethyl sulfide (DMS) and dimethylsulfoniopropionate (DMSP) are sulfur compounds that may function as antioxidants in algae. Symbiotic dinoflagellates of the genus Symbiodinium show strain‐specific differences in their susceptibility to temperature‐induced oxidative stress and have been shown to contain high concentrations of DMSP. We investigated continuous cultures of four strains from distinct phylotypes (A1, A13, A2, and B1) that can be characterized by differential thermal tolerances. We hypothesized that strains with high thermal tolerance have higher concentrations of DMSP and DMS in comparison to strains with low thermal tolerance. DMSP concentrations were strain‐specific with highest concentrations occurring in A1 (225 ± 3.5 mmol · L?1 cell volume [CV]) and lowest in A2 (158 ± 3.8 mmol · L?1 CV). Both strains have high thermal tolerance. Strains with low thermal tolerance (A13 and B1) showed DMSP concentrations in between these extremes (194 ± 19.0 and 160 ± 6.1 mmol · L?1 CV, respectively). DMS data further confirmed this general pattern with high DMS concentrations in A1 and A13 (4.1 ± 1.22 and 2.1 ± 0.37 mmol · L?1 CV, respectively) and low DMS concentrations in A2 and B1 (0.3 ± 0.06 and 0.5 ± 0.22 mmol · L?1 CV, respectively). Hence, the strain‐specific differences in DMSP and DMS concentrations did not match the different abilities of the four phylotypes to withstand thermal stress. Future work should quantify the possible dynamics in DMSP and DMS concentrations during periods of high oxidative stress in Symbiodinium sp. and address the role of these antioxidants in zooxanthellate cnidarians.  相似文献

综述了甲藻的异养类型。目前已知异养营养型在甲藻中广泛存在,只有很少几种甲藻营严格自养营养方式。有近一半的甲藻物种是没有色素体的,还有很多甲藻即使具有色素体也会有异养营养需求,称为兼养营养类型。这些兼养类群不一定主要以有机物作为其获取碳的来源,而仅仅是补充一些生长必需的有机物如维生素、生物素等。兼养类群以渗透营养和腐食营养方式进行,同时也可以寄生方式和共生方式进行兼养生活。无色素体的甲藻以有机物作为碳的唯一来源,仅仅依靠异养方式生存,属于严格异养营养方式,又称有机营养型。它们是甲藻异养营养型的主体,其主要类型有寄生、渗透营养和吞噬营养。由于吞噬营养是甲藻异养的主要类型,因此论述了3种吞噬营养型:吞噬营养方式、捕食茎营养方式和捕食笼营养方式。吞噬营养方式在无甲类和具甲类甲藻中都有存在,主要通过甲藻细胞的纵沟或底部对猎物进行吞噬,也有研究发现吞噬部位为顶孔或片间带。捕食茎营养方式是通过捕食茎刺穿猎物细胞膜并吸食其细胞质来获取营养,在异养甲藻中也较常见。捕食笼营养方式只在原多甲藻属(Protoperidinium)和翼藻属(Diplopsalis)里发现,是甲藻通过鞭毛孔分泌细胞质到胞外形成捕食笼将猎物包裹并进行消化来摄食的。甲藻摄食对象尺寸范围变化较大,小至几微米,大至几百微米。有些甲藻具有摄食选择性,通过感应猎物释放的化学物质来判断猎物的位置并进行摄食,摄食完成后由于体积的增加经常会发生细胞分裂和蜕鞘。对于甲藻异养的其他形式如拦截摄食营养方式、伪足摄食营养方式、口足摄食营养方式、触手摄食营养方式等只作简单介绍。还就甲藻异养的研究方法、其生态学意义和进化学意义进行简要论述,并对相关研究进行展望。  相似文献
