The inky cap, Coprinellus congregatus, produces mushrooms which become autolyzed rapidly to generate black liquid droplets, in which no cell wall is detected by microscopy. A chitinase (Chi2) which is synthesized during the autolytic phase of C. congregatus inhibits the growths of Candida albicans and Cryptococcus neoformans up to 10% at the concentration of 10 μg/ml, about 50% at concentration of 20 μg/ml, and up to 95% at the concentration of 70 μg/ml. Upon treatment these yeast cells are observed to be severely deformed, with the formation of large holes in the cell wall. The two yeast species show no growth inhibition at the concentration of 5 μg/ml, which means the minimum inhibitory concentrations for both yeast species are 10 μg/ml under these experimental conditions.  相似文献

Summary This paper deals with the application of the osmium tetroxide fixation followed by p-phenylenediamine treatment to salivary gland cells from Chironomus larvae. After this procedure, cytoplasm, nucleoli and Balbiani rings show a high degree of staining both in light and electron microscopy, while chromatin remains unstained. Ethanol fixation followed by osmium tetroxide/p-phenylenediamine does not modify the above mentioned staining pattern. Under these conditions, extractive procedures for lipids do not affect the osmiophilia of nucleoli and Balbiani rings, while RNase or trichloroacetic acid treatment decreaes the staining degree of these structures. In osmium tetroxide/p-phenylenediamine treated salivary glands, the highest contrast within nuclei is seen to occur in the pars granulosa from normal or segregated nucleoli, as well as in Balbiani ring granules, which appear either as hollow granules or with a bipartite or horseshoe-like structure.  相似文献

RNA synthesis in a Balbiani ring in Chironomus tentans salivary gland cells   总被引:3,自引:0,他引:3  
Rapidly labelled RNA in Balbiani ring 2 on chromosome IV in the salivary glands of Chironomus tentans was investigated. This RNA is likely to be transcribed from only one chromosomal band, supposed to be a single operational unit in these polytenic cells (Beermann, 1966).Salivary glands were incubated in larval haemolymph, supplemented with tritiated RNA precursors and fixed afterwards. Balbiani rings 2 (in some experiments also Balbiani ring 1 and 3) were isolated with micromanipulation. The labelled RNA was extracted with SDS-pronase and analysed with electrophoresis in agarose.The rapidly labelled RNA in Balbiani ring 2 was as heterogeneous as RNA from the remainder of the chromosome set (10–90 S) but the peak of the distribution of label in BR 2 corresponded to molecules of about 50 S as compared to that of RNA from the rest of the chromosome set which was about 35 S. When the synthetic activity in Balbiani ring 2 was very high, relatively more molecules with very high molecular weights were produced compared with the state when the synthetic activity was moderate or low. The synthetic activity in Balbiani ring 2 compared to that in Balbiani ring 1 was well correlated to the relative sizes of the two Balbiani rings. The results on Balbiani ring 2 are discussed in relation to the size and structure of the chromomere.  相似文献

Drosophila auraria and its sibling species, D. biauraria, D. triauraria, and D. quadraria are unique among Drosophila species in that their salivary gland chromosomes exhibit Balbiani rings. In this report we present a cytological map of D. auraria and information on the developmental profiles of its puffs and Balbiani rings. Information is presented on the existence of tandem inverted duplications involving the Balbiani ring regions and other regions of the chromosomes, and data are given concerning the puffing patterns of the duplicated bands. Possible homologies between puffs of D. melanogaster and D. auraria and certain differences between the two species in the developmental sequences of the active loci are discussed.  相似文献

The fine structure and cytochemistry of the extremely large RNA puffs, or Balbiani rings, in salivary gland nuclei of midge, Chironomus thummi, larvae have been investigated. The Balbiani rings are composed of a diffuse mass of electron-opaque 400 to 500 A granules, short threads about 180 to 220 A in diameter and associated fine chromatin fibrils. These components appear to be organized into brushlike elements which form the ring. Electron microscope cytochemistry has shown that the granules and short threads contain RNA. After ribonuclease digestion, only 50 to 100 A chromatin fibrils were apparent in the Balbiani ring, and the granules were no longer demonstrable. Deoxyribonuclease digestion had no apparent effect on these structures. Observations indicate that the granules are formed from the short threads and released into the nucleoplasm in which they are evenly distributed. At the nuclear envelope, many granules have been observed partially or completely within the nuclear pores. These granules become elongated and are shown to penetrate the center of the pore in a rodlike form, about 200 A in diameter. The Balbiani ring granules are not normally visible within the cytoplasm adjacent to the nuclear envelope, but have been rarely found in this region. It is suggested that the granules represent the product of the Balbiani ring, possibly a messenger RNA bound to protein, and that they regularly pass into the cytoplasm through a narrow central channel in the pores of the nuclear envelope.  相似文献

The concentration and molecular weight of hyaluronan often dictates its physiological function. Consequently full characterisation of the anabolic products and turnover rates of HA could facilitate understanding of the role that HA metabolism plays in disease processes. In order to achieve this it is necessary to interrupt the dynamic balance between concurrent HA synthesis and degradation, achievable through the inhibition of the hyaluronidases, a group of enzymes which degrade HA. The sulphated polysaccharide, dextran sulphate has been demonstrated to competitively inhibit testicular hyaluronidase in a non-biological system, but its application to in vitro biological systems had yet to be developed and evaluated. This study determined the inhibitory concentrations of dextran sulphate against both testicular and Streptomyces hyaluronidase in a cell-free and breast cancer model followed by characterisation of the effect that hyaluronidase inhibition exerted on HA synthesis and degradation. The IC100 of dextran sulphate for both hyaluronidases in a cell-free and biological system was determined to be ≥400 μg/ml. At concentrations up to 10 mg/ml the dextran sulphate did not effect breast cancer cell proliferation or morphology, while at 400 μg/ml HA degradation was totally inhibited, enabling an accurate quantitation of HA production as well as characterisation of the cell-associated and liberated HA. FACS quantitation of the HA receptor CD44, HA synthase and the hyaluronidases HYAL 1 and HYAL 2 demonstrated that dextran sulphate down-regulated CD44 and HA synthase while upregulating the hyaluronidases. These results suggest dynamic feedback signalling and complex mechanisms occur in the net deposition of HA in vivo. Published in 2004.  相似文献

A 5 minutes exposure of Chironomus larvae to near-lethal temperatures (39–40° C) produces a characteristic sequence of puffing changes in the Balbiani rings of the 4th salivary gland chromosomes: An initial phase of complete puff regression is followed, after a variable time lag, by a phase of rapid recovery to overnormal puff sizes. This is accompanied by RNP droplet formation. RNA synthesis at Balbiani rings during the initial puff regression still occurs. Regression is inhibited by 2,4-dinitrophenol, while recovery can be prevented by both 2.4dinitrophenol and actinomycin D. Regression and recovery are insensitive to cycloheximide. RNP droplets, as observed in Balbiani rings, in the nuclear sap and at the nucleolar rim, are composed of a fine fibrillar matrix which is covered by Balbiani ring granules in various phases of assembly. The results are discussed in terms of a model of puffing based on an equilibrium between RNA synthesis, RNA processing and RNP release from the puff.

The switchover from a larval to a pupal epidermal commitment was studied on integument tissue fragments from early last-instar larvae (1–2 days after ecdysis) of Chilo suppressalis cultured in Grace's medium containing 0.01–0.5 μg/ml 20-hydroxyecdysone for 24–72 hr. Fragments were subsequently cultured in medium containing 1 μg/ml 20-hydroxyecdysone for 24 hr and maintained in hormone-free media for 6 additional days. The degree of switchover induction was measured as the ratio of the number of tissue fragments showing pupal characteristics to the total number of fragments used. The degree of switchover increased with the duration of culture, as well as with the concentration of the hormone (up to 0.1 μg/ml), in the first hormonal treatment. Above this concentration, apolysis and new cuticle formation were induced without change in the epidermal commitment. Cultured integument fragments from larvae in the diapause stage, 40–50 days after hatching, and from those in the penultimate stage, showed the switchover under almost the same hormonal conditions as those used with tissue from the early last-instar larvae. After the first hormone treatment, culture in hormone-free medium was unnecessary for cuticle tanning. Juvenile hormone II added to the medium (3 ng/ml) in the first hormonal treatment completely inhibited the switchover induced by 20-hydroxyecdysone. The potential use of the C. suppressalis integument as a bioassay system for juvenoids is discussed.  相似文献

Treatment of Chironomus larvae with dilute (0.5%–1.0%) ethanol results in puffing changes similar to those obtained with galactose in the Balbiani rings (BRs) of the salivary gland chromosomes. A shift in the relative size of BR1 and BR2 in chromosome 4 of C. pallidivittatus or C. tentans was observed within 1–2 days after ethanol treatment. The exceptional Balbiani ring, BR6 in chromosome 3, began to appear within 1 day after ethanol treatment of C. pallidivittatus and was fully developed after 3–4 days. Prepupae appeared to be refractory to the treatment. To localize possible controls of BR puffing in Chironomus, ligatures were made at various positions along the thorax and the anterior abdominal segments of the ethanoltreated larvae. In surviving larvae, ligated anterior to the brain or posterior to the salivary glands, induction of BR6 could be detected. In contrast, little or no BR6 puff induction was found in animals ligated in the middle of the second segment approximately between the brain and the salivary glands. No shift in the BR1/BR2 relation occurred with any of the ligations combined with ethanol treatment.  相似文献

Incubation of root tips in cycloheximide (CHM) at concentrations of 0.3–50 μg/ml inhibits the incorporation of [14C]leucine by 40–100% within 2 h. A depression in the incorporation of [3C]thymidine was observed after a 2-h incubation in CHM solution at 1 μg/ml.In root tips exposed for 2 h to CHM at 1 μ/ml the mitotic activity of cells was severely depressed within 15 h of recovery. Metaphases appearing after 20 h carried infrequent aberrations of the chromatid type. CHM at this concentration had no effect on the yield of aberrations induced by the alkylating agents diepoxybutane (DEB) and N-ethyl-N-nitrosourea (ENU) when applied as post-treatment.  相似文献

Incubation of Chironomus pallidivittatus salivary glands in the presence of cordycepin induces the collapse of Balbiani rings. In spite of the great reduction of the Balbiani ring size, 3H-uridine incorporation takes place in these structures and also in puffs and nucleolus. Modifications in the nucleolar pattern of labelling have been observed. Differences were found between the effect of cordycepin and those of actinomycin and adenosine both of which induce the collapse of Balbiani rings.  相似文献

Thionins are small basic peptides found in different plant species, which are known to exert cytotoxic properties. In addition, previous data indicated an activation of human granulocytes by thionins from European mistletoe (viscotoxins, VT). To extend these latter findings, we investigated the influence of VT and from thionins from wheat flour (purothionin) on human granulocytes by flow cytometry and tried to characterise the involved molecular structures and mechanisms. Phagocytosis was determined by incorporation of FITC-labelled Escherichia coli and respiratory burst by oxidation of dihydrorhodamine 123 to rhodamine 123. VT and purothionin significantly enhanced E. coli-stimulated phagocytosis and respiratory burst at 25 and 250 μg/ml. Phagocytosis of damaged lymphocytes by granulocytes was detected by electron microscopy in the VT-stimulated (100 μg/ml) but not in the control cultures. The poly-cationic structure of the intact molecule seems to be crucial, as evidenced by comparison of the burst and phagocytosis-enhancing effects induced by other poly-cationic (protamine sulphate, histone, poly-l-arginine, poly-l-lysine) and poly-anionic (poly-l-glutamic acid) peptides, while pore forming due to amphipathic properties seems to be less important. Ca2+ and Mg2+ could not inhibit VT-enhanced phagocytosis and, thus, could not inhibit binding of VT to granulocytes. In addition, verapamil at low concentrations inhibited VT activity, suggesting the involvement of Ca2+ channels for granulocyte activation by the VT. Similarly, thionins and histones in contrast to protamine sulphate induced cell death of granulocytes at 250 μg/ml as demonstrated by an enhanced release of reactive oxygen intermediates in unstimulated granulocytes. From these data one may suggest that activity of VT is induced by strong unspecific ionic binding, probably followed by specific receptor binding, and thionins exhibit stimulatory and cytotoxic effects on immune cells, which have to be further characterised.  相似文献

A method for assaying cortisol and cortisone using chromatography on either paper or Sephadex LH-20 columns for isolation, followed by competitive protein binding, has been applied to umbilical cord and maternal plasma samples. In mixed cord plasma the mean cortisol concentration was 6.0 ± 0.8 μg/100 ml (n = 9) and the mean cortisone concentration was 13.5 ± 2.9 μg/100 ml (n = 9). In cord arterial plasma the mean cortisol concentration was 6.3 ± 2.9 μg/100 ml (n = 6) and the mean cortisone level was 10.1 ± 2.5 μg/100 ml (n = 6). For cord venous plasma, the mean level of cortisol was 5.6 ± 1.5 μg/100 ml (n = 6) and of cortisone was 13.5 ± 2.4 μg/100 ml (n = 6). Maternal plasma gave a mean value of cortisol of 42.3 ± 4.5 μg/100 ml (n = 6) and of cortisone of 6.2 ± 0.9 μg/100 ml. The results of this study suggest that the fetus at term-gestation produces cortisol. The significance of this production compared with placental transfer of maternal cortisol into the fetal circulation however is uncertain.  相似文献

Rapidly growing thalli of Zonaria have been treated with colchicine (10,000, 5,000, and 1,250 μg/ml) and phenobarbital (10,000 and 5,000 μg/ml). Responses of the meristems to drug treatment have been described at the ultrastructural, cellular, and organismic levels. With colchicine treatment microtubule disruption, nuclear envelope abnormalities, nuclear lobing, and increased nuclear sizes were observed. Apical cells ceased dividing but continued to elongate until cessation of treatment when abnormal cytokineses, irregular cell shapes, and cell deaths were observed. The coordinated growth of the meristem margin was disrupted, regions recovering left a zone line on the thalli, and cell deaths resulted in thallus branching. With phenobarbital treatment ER elaboration, inactive-appearing dictyosomes, pyrenoid-like structures, and deeply cup-shaped mitochondria aggregated with ER were observed. Apical cell elongation ceased and the mitotic cycle appeared to be arrested just after cytokinesis. Recovery from 10,000 μg/ml phenobarbital was low, with subapical cells and occasional apical cells starting new thalli. Recovery from 5,000 μg/ml was good, with occasional cell death dividing the meristem and zone lines resulting from the temporary synchrony. Correlations of responses at the ultrastructural, cellular, and organismic levels are discussed.  相似文献
