利用解剖镜和扫描电镜,对我国9种石斛属(Dendrobium)植物的花药帽形态和超微特征进行观察。结果显示,花药帽的颜色、形态、大小和外壁纹饰特征在种间差异明显。9种植物的花药帽按颜色可分为:黄药帽、白药帽、紫药帽;按极轴和赤道轴比值可分为:长球形和半球形。扫描电镜观察结果发现,花药帽外壁纹饰有4种类型:扁平-光滑、扁平-条纹、乳突-条纹和乳突-光滑。本研究初步探讨了9种石斛花药帽形态的分类学意义,支持以下观点:(1)石斛组是一个多系和并系;(2)球花石斛(D. thyrsiflorum Rchb. F.)和密花石斛(D. densiflorum Lindl. ex Wall)是一对近缘种;(3)杯鞘石斛(D. gratiosissimum Rchb. F.)和大苞鞘石斛(D.wardianum Warner)是一对近缘种,且与喇叭唇石斛(D. lituiflorum Lindl.)亲缘关系较近。 相似文献

石斛属(Dendrobium)隶属于兰科(Orchidaceae)树兰亚科(Epidendroideae)石斛兰族(Dendrobiinae),是兰科最大的属之一,终生附生于树上或岩石上。石斛属很多种类具有很高的药用价值与观赏价值。由于人为过度采挖和野生生境的破坏,使得野生石斛资源濒临灭绝。石斛属植物为典型的兰科菌根植物,在自然条件下需要与真菌共生,才能完成生活史。菌根真菌对于石斛属植物的种子萌发和植株生长具有重要的作用。对石斛属植物菌根的形成、菌根真菌的作用、菌根真菌多样性及菌根技术在石斛属植物中的应用做了评述,并对今后的研究内容和重点提出了一些思路。 相似文献

About 63 species of Dendrobium are identified in China, making the identification of the origin of a particular Dendrobium species on the consumer market very difficult. We report evaluation of multiple species-specific probes screened from genomic DNA for closely related Dendrobium species identification, based on DNA array hybridization. Fourteen species-specific probes were screened from five closely related Dendrobium species, D. aurantiacum Kerr, D. officinale Kimura et Migo, D. nobile Lindl., D. chrysotoxum Lindl. and D. fimbriatum Hook., based on the SSH-Array technology we developed. Various commercial Dendrobium samples and unrelated samples were definitely identified. The specificity and accuracy of the multiple species-specific probes for species identification was assessed by identifying various commercial Dendrobium samples (Herba Dendrobii). Hybridization patterns of these multiple probes on digested genomic DNAs of Dendrobium species indicated that there are distinct polymorphic sequence fragment in the higher eukaryotes. This is the first report on detection and utilization of multiple species-specific probes of Dendrobium in whole genomic DNA, and this could be useful tools not only for a new technical platform for the closely related species identification but also for epidemiological studies on higher eukaryotes. 相似文献

Genomlc DNA polymorphlsms are very useful for tracing genetic traits end studying biological diversity among species. Here, we present a method we call the "diversity suppresslon-subtractlve hybridization array" for effectively profiling genomlc DNA polymorphisms. The method first obtains the subtracted gDNA fragments between any two species by suppression subtraction hybridization (SSH) to establish e subtracted gDNA library, from which diversity SSH arrays are created with the selected subtracted clones. The diversity SSH array hybridizes with the DIG-labeled genomlc DNA of the organism to be assayed. Six closely related Dendrobium species were studied as model samples. Four Dendrobium species as testers were used to perform SSH. A total of 617 subtracted positive clones were obtained from four Dendrobium species, and the average ratio of positive clones was 80.3%. We demonstrated that the average percentage of polymorphlc fragments of palrwlse comparisons of four Dendrobium species was up to 42.4%. A dendrogram of the relatedness of six Dendrobium species was produced according to their polymorphic profiles. The results revealed that the diversity SSH array Is a highly effective platform for profiling genomlc DNA polymorphlsms and dendrograms. 相似文献

Plant phylogeny constrains orchid mycorrhizal(OrM) fungal community composition in some orchids. Here, we investigated the structures of the OrM fungal communities of eight Dendrobium species in one niche to determine whether similarities in the OrM fungal communities correlated with the phylogeny of the host plants and whether the Dendrobium-OrM fungal interactions are phylogenetically conserved. A phylogeny based on DNA data was constructed for the eight coexisting Dendrobium species,and the OrM fungal communities were characterized by their roots. There were 31 different fungal lineages associated with the eight Dendrobium species. In total, 82.98% of the identified associations belonging to Tulasnellaceae, and a smaller proportion involved members of the unknown Basidiomycota(9.67%). Community analyses revealed that phylogenetically related Dendrobium tended to interact with a similar set of Tulasnellaceae fungi. The interactions between Dendrobium and Tulasnellaceae fungi were significantly influenced by the phylogenetic relationships among the Dendrobium species. Our results provide evidence that the mycorrhizal specificity in the eight coexisting Dendrobium species was phylogenetically conserved. 相似文献

当前已开发的石斛(Dendrobium)SSR标记仅100余对,难以满足研究的需要。为开发更多石斛分子标记,本研究通过生物信息学方法从公共核酸数据库搜索石斛SSR。结果表明,GenBank收录的3599条石斛DNA序列经拼接获得1343条Uni—DNA序列。经搜索,共检测出283个SSR,分布于205条Uni—DNA序列,平均每2815bp含一个SSR。通过序列比对,剔除86条已开发引物的SSR—DNA序列,从剩余的119条序列设计76对引物,并在石斛属32个种间进行可转移性分析,结果显示47对引物得到有效扩增,种间可转移率为51.1%N95.7%,平均为75.9%。有效扩增引物中有46对在石斛种间具多态性,检测出等位基因数2~8个,平均4.0个。选取10对多态性引物扩增60份铁皮石斛资源,每对引物检测出等位基因数2~5个,平均3.4个。根据SSR扩增带型将60份铁皮石斛资源聚为5大类,同一类型内的表型较类群间接近。对DMl21扩增产物测序表明铁皮石斛种内的SSR等位变异由SSR重复次数差异造成,而石斛种间的SSR等位变异还包含SSR位点侧翼序列的插入缺失以及替换。 相似文献

对兰科植物的兰属、兜兰属及石斛属16个种折叶片及其横断面进行了扫描电镜的观察。兰属各种叶片上表皮细胞均为矩形,上表皮细胞表面具小乳突或不明显突起。石斛属及兜兰属的各个种上下表皮细胞均为多边形,但石斛属表皮细胞表面无坦无纹饰,而兜兰属花叶类上表皮细胞表面明显呈乳突状,绿叶类呈龟背状隆起。兰属及石斛属叶片叶肉组织没有栅栏组织及海绵组织的分化,而兜兰属的绿叶类叶肉不分化;花叶类叶肉有分化。 相似文献
