Here, we investigated the patterns of microbial nitrogen cycling communities along a chronosequence of soil development in a salt marsh. The focus was on the abundance and structure of genes involved in N fixation (nifH), bacterial and archaeal ammonium oxidation (amoA; AOB and AOA), and the abundances of genes involved in denitrification (nirS, nirK, nosZ). Potential nitrification and denitrification activities were also measured, and increases in nitrification were found in soils towards the end of succession, whereas denitrification became maximal in soils at the intermediate stages. The nifH, nirK and nirS gene markers revealed increases in the sizes of the respective functional groups towards the intermediate stage (35 years), remaining either constant (for nifH) or slightly declining towards the latest stage of succession (for nirK and nirS). Moreover, whereas the AOB abundance peaked in soils at the intermediate stage, that of AOA increased linearly along the chronosequence. The abundance of nosZ was roughly constant, with no significant regression. The drivers of changes in abundance and structure were identified using path analysis; whereas the ammonia oxidizers (AOA and AOB) showed patterns that followed mainly N availability, those of the nitrogen fixers followed plant diversity and soil structure. The patterns of denitrifiers were group-dependent, following the patterns of plant diversity (nirK and nirS) and belowground shifts (nosZ). The variation observed for the microbial groups associated with the same function highlights their differential contribution at different stages of soil development, revealing an interplay of changes in terms of niche complementarity and adaptation to the local environment.  相似文献

Burrowing benthic animals belonging to the same functional group may produce species-specific effects on microbially mediated nitrogen (N) processes depending upon different ecological traits. We investigated the effects of two tube-dwelling organisms, amphipods (Corophium insidiosum) and chironomid larvae (Chironomus plumosus), on benthic N cycling in bioturbated estuarine sediments. Aims of this work were to analyze the interactions among burrowers and N-related microbial processes in two distinct sedimentary environments colonized by benthic animals with different ecological traits. We hypothesized higher rates of nitrification and higher coupled nitrification–denitrification in sediments with C. insidiosum due to continuous ventilation rates. We expected higher denitrification of water column nitrate in sediments with C. plumosus due to lower and intermittent ventilation activity and lower oxygen levels in burrows. To this purpose, we combined process–specific (nitrification and denitrification) with net N flux measurements in intact and reconstructed sediments. Sediments with C. insidiosum had higher rates of oxygen demand and of potential nitrification and higher concentration of pore water NH4+ as compared to sediments with C. plumosus. Sediments with both species displayed comparable net N2 fluxes, mostly sustained by respiration of water column NO3? in sediments with chironomid larvae and by NO3? produced within sediments in sediments with corophiid amphipods. Corophium insidiosum stimulated nitrification nearly 15-fold more as compared to C. plumosus. Overall, our results demonstrate that sediments with burrowing fauna may display similar rates of denitrification, but underlying mechanisms may deeply vary and be species-specific.  相似文献

In this study, we analysed metagenomes along with biogeochemical profiles from Skagerrak (SK) and Bothnian Bay (BB) sediments, to trace the prevailing nitrogen pathways. NO3 ? was present in the top 5 cm below the sediment-water interface at both sites. NH4 + increased with depth below 5 cm where it overlapped with the NO3 ? zone. Steady-state modelling of NO3 ? and NH4 + porewater profiles indicates zones of net nitrogen species transformations. Bacterial protease and hydratase genes appeared to make up the bulk of total ammonification genes. Genes involved in ammonia oxidation (amo, hao), denitrification (nir, nor), dissimilatory NO3 ? reduction to NH4 + (nfr and otr) and in both of the latter two pathways (nar, nap) were also present. Results show ammonia-oxidizing bacteria (AOB) and ammonia-oxidizing archaea (AOA) are similarly abundant in both sediments. Also, denitrification genes appeared more abundant than DNRA genes. 16S rRNA gene analysis showed that the relative abundance of the nitrifying group Nitrosopumilales and other groups involved in nitrification and denitrification (Nitrobacter, Nitrosomonas, Nitrospira, Nitrosococcus and Nitrosomonas) appeared less abundant in SK sediments compared to BB sediments. Beggiatoa and Thiothrix 16S rRNA genes were also present, suggesting chemolithoautotrophic NO3 ? reduction to NO2 ? or NH4 + as a possible pathway. Our results show the metabolic potential for ammonification, nitrification, DNRA and denitrification activities in North Sea and Baltic Sea sediments.  相似文献

Vegetation in grasslands is changing at an unprecedented rate. In the Nebraska Sandhills, this shift is attributed in part to encroachment of the woody species Juniperus virginiana. We investigated changes in resource availability and their feedback on seasonal trends in photosynthetic characteristics of J. virginiana trees scattered in open grasslands vs. a dense 57-year-old stand. Dense stand exhibited lower volumetric soil water content, NH4 +, NO3 , and δ13C, as well as foliage δ13C, δ15N, and N content, compared to grasslands. Water potential was higher in trees in grasslands compared to dense stand. J. virginiana in dense stand exhibited similar trends to trees in grasslands for net photosynthetic rate (P N), stomatal conductance, transpiration, maximum photochemical efficiency of PSII, maximum carboxylation velocity, and maximum rate of electron transport. P N peaked early summer and declined in the fall, with trees in open grasslands lagging behind those in dense stand. Plasticity of this species may place it at a competitive advantage in the Sandhills, further altering grasslands vegetation and ecosystem processes.  相似文献

To enhance the startup and efficient simultaneous nitrification and denitrification for sewage treatment, sequencing batch biofilm reactors (SBBRs) partially coupled with rice husk were established and operated under various intermittent micro-aeration cycles (IMCs) and COD/N ratios under oxygen-limiting intermittent aeration conditions. Experimental results showed that the increase of IMCs with non-aeration/micro-aeration mode of (8 h/4 h)1 to (2 h/1 h)4 in a 12 h-cycle accelerated the startup performance and improved NH4+–N and COD removal. NH4+–N, TN and COD removal efficiencies were 98.7?±?0.9, 89.2?±?5.2 and 82.9?±?6.7% at COD/N ratio of 7.6 with the highest IMCs in SBBR, respectively. Higher TN removal efficiencies of 87.2?±?4.0 and 58.1?±?3.5% were also achieved at lower COD/N ratio of 5.6 and 2.8, respectively. In SBBRs with various IMCs, facultative denitrifier like genus Acinetobacter and solid-phase denitrifier belonging to Comamonadaceae family were enriched. However, aerobic denitrifiers with function of heterotrophic nitrification like Paracoccus were favored to enrich under higher IMCs condition, and more anoxic denitrifiers like sulfur-based autotrophic denitrifier Thiothrix and heterotrophic denitrifiers like Pseudomonas and Methyloversatilis were observed at lower IMCs condition. Autotrophic nitrifier (Nitrosomonas and Nitrosipra) and heterotrophic nitrifiers both contributed to the efficient nitrification.  相似文献

Goatgrass (Aegilops) species are some of the most aggressive invasive plants in the Western U.S. Despite intense management efforts, goatgrass continues to reduce the ecological and economic integrity of natural and agroecological systems. The mismatch between current research outcomes and practical needs of land managers is likely a result of limitations associated with generalizing from single location, treatment, or season studies. We conducted a systematic review of experiments testing control of two dominant goatgrass invaders (A. cylindrica and A. triuncialis) to identify general patterns in treatment efficiency. Using data from 391 separate experiments, we found that experimental treatments were more successful at controlling the dominance of A. cylindrica compared to A. triuncialis. For A. cylindrica, no treatment demonstrated particular utility for control. Treatment of A. cylindrica in the vegetative stage was more effective for control than treatment at other stages. For A. triuncialis, burning and grazing demonstrated effective overall control among all treatments, although grazing produced variable results. Treatment in the fruiting stage of A. triuncialis was more effective for control than treatment at other stages. For both species, multiple applications of a management treatment within a year resulted in no better control than a single application within a year. Additionally, treatments deployed in two consecutive years resulted in better control of both species, than a treatment deployed in a single year. This work highlights promising avenues for more intensive research on goatgrass weed control and suggests that management funding is most effectively utilized when employed across years rather than focused on a single year.  相似文献

Background and aims

Competition from the annual grass Bromus tectorum threatens aridland perennial bunchgrass communities. Unlike annuals, perennials must allocate part of their first year nitrogen (N) budget to storage rather than growth, potentially placing them at a competitive disadvantage.

Methods

We evaluated N acquisition and conservation for two perennial bunchgrasses, Agropyron desertorum and Pseudoroegneria spicata, at the seedling stage to investigate potential trade-offs between storage and growth when grown with and without B. tectorum under two levels of soil N.

Results

Agropyron desertorum had higher growth rates, N uptake, and N productivity than P. spicata when grown without B. tectorum, but trait values were similarly low for both species under competition. Without competition, N resorption was poor under high soil N, but it was equally proficient among species under competition.

Conclusions

A. desertorum had higher growth rates and N productivity than P. spicata without competition, suggesting these traits may in part promote its greater success in restoration programs. However, B. tectorum neighbors reduced its trait advantage. As plant traits become more integral to restoration ecology, understanding how N capture and conservation traits vary across candidate species and under competition may improve our ability to select species with the highest likelihood of establishing in arid, nutrient-limited systems.

The N2-fixing shrub Alnus viridis is currently encroaching on montane grasslands in the Alps as a result of reduced land management and complete abandonment. Alnus introduces large amounts of nitrogen (N) into these formerly N-poor grasslands and restricts the succession to montane forests. We studied pools and fluxes of N and the associated C pools in pastures (controls) and adjacent Alnus shrublands at two elevations (1650 versus 1950 m a.s.l.) in three valleys in the Swiss central Alps. The total N and C pools stored in 50-year-old Alnus shrubland did not exceed those in adjacent pastures with a total of approximately 610 g N m?2 in phytomass plus soil (down to 30 cm) at both elevations. In Alnus stands, reduced soil N pools balanced the gain in phytomass N pools, a likely result of a faster turnover of soil N. The soil solution under Alnus was continuously enriched with nitrate, with a total N leaching of 0.79 g N m?2 season?1 (June–October) under 50-year-old stands at both elevations and the highest flux of 1.76 g N m?2 season?1 in 25-year-old shrubland at low elevation, clearly indicating an excess of available N in Alnus shrubland. In contrast, N leaching across all pastures was close to zero (0.08 g N m?2) throughout the season. At the catchment scale, streamlet water showed increased nitrate concentrations with typical flushing peaks in spring and autumn, provided more than one fifth of the catchment area was covered by Alnus shrubs. We conclude that the expansion of Alnus rapidly converts centuries-old, N-poor grassland into N saturated shrubland, irrespective of elevation, and it reduces the C storage potential of the landscape because the Alnus dominance constrains re-establishment of a natural montane forest.  相似文献

Abstract

Production of unreduced gametes is common among interspecific hybrids, and may be affected by parental genotypes and genomic similarity. In the present study, five cultivars of Triticum aestivum and two tetraploid Aegilops species (i.e. Ae. triuncialis and Ae. cylindrica) were reciprocally crossed to produce 20 interspecific hybrid combinations. These hybrids comprised two different types: T. aestivum × Aegilops triuncialis; 2n = ABDUtCt (which lack a common subgenome) and T. aestivum × Ae. cylindrica; 2n = ABDDcCc (which share a common subgenome). The frequency of unreduced gametes in F1 hybrids was estimated in sporads from the frequency of dyads, and the frequency of viable pollen, germinated pollen and seed set were recorded. Different meiotic abnormalities recorded in the hybrids included precocious chromosome migration to the poles at metaphase I and II, laggards in anaphase I and II, micronuclei and chromosome stickiness, failure in cell wall formation, premature cytokinesis and microspore fusion. The mean frequency of restitution meiosis was 10.1 %, and the mean frequency of unreduced viable pollen was 4.84 % in T. aestivum × Ae. triuncialis hybrids. By contrast, in T. aestivum × Ae. cylindrica hybrids no meiotic restitution was observed, and a low rate of viable gametes (0.3 %) was recorded. This study present evidence that high levels of homologous pairing between the D and Dc subgenomes may interfere with meiotic restitution and the formation of unreduced gametes. Variation in unreduced gamete production was also observed between T. aestivum × Ae. triuncialis hybrid plants, suggesting genetic control of this trait.

Australian species of the genus Acacia are amongst the most invasive trees. As nitrogen fixers, they are able to invade oligotrophic ecosystems and alter ecosystem functioning to their benefit. We aimed to answer three questions: How does early Acacia invasion influence nitrogen and light in a sandy savanna? How does early Acacia invasion impact biodiversity? Does early invasion alter ecosystem functioning towards the dominance of Acacia? We analyzed (using generalized linear mixed models and richness estimators) paired plots focused on plants of Acacia mangium (Fabaceae) and plants of Marcetia taxifolia (Melastomataceae) by taking hemispherical photos and sampling plants, leaves and soil for measurements of light, richness, leaf nitrogen, leaf δ15N, soil nitrogen and soil coarse sand. The results suggest that early Acacia invasion alters the control of soil and of leaf nitrogen and increases shading, enabling a much wider range of light variation. The δ15N results suggest that the nitrogen taken up by Acacia is transferred to neighboring plants and influences the light environment, suggesting facilitation. The enrichment of plant species observed during early Acacia invasion is consistent with the wider range of light variation, but the forecasted leaf nitrogen conditions during the established phase of Acacia invasion might cause loss of light-demanding species because of increased shading. If early Acacia invasion turns into an established phase with highly increased shading, Acacia seedlings might be favored and ecosystem functioning might change towards its dominance.  相似文献

Cyanobacterial blooms may stimulate epiphytic nitrification and denitrification in the water column. To validate this hypothesis, a 4-week floating mesocosms experiment that involved a cyanobacterial decay–growth–decay period was conducted at Lake Taihu. In addition to conventional methods for detecting the physical and chemical properties, quantitative real-time PCR was used to identify the nitrification and denitrification genes (archaeal and bacterial amoA, nirS and nirK). Treatment with cyanobacteria led to removal of about 3.62 mg N L?1 total nitrogen, 40% of which was organic nitrogen, indicating a nitrogen transformation and removal mechanism was present in the system. Variations in the biogeochemical properties suggested that remineralization and coupling nitrification and denitrification by epiphytic and pelagic microorganisms was the primary pathway through which organic nitrogen was removed. The results of this study revealed that algal blooms can accelerate nitrogen removal efficiency, which may be the primary reason that nitrogen is limited in summer in Lake Taihu.  相似文献

Creeping bentgrass (Agrostis stolonifera) and redtop (A. gigantea) are introduced turfgrasses that are naturalized throughout the northern U.S. Interest in creeping bentgrass has risen following the 2003 escape of a genetically modified (GM), herbicide-resistant cultivar near Madras, Oregon. The objectives of this study were to characterize the floristic attributes of the plant communities associated with naturalized Agrostis populations in the Madras area, and to identify plant communities at risk of invasion by transgenic Agrostis. Vegetation data collected from 62 stratified random vegetation plots with and without A. stolonifera and A. gigantea identified 11 distinct plant communities. Community composition was strongly correlated with an indirect soil moisture index based on the wetland status of individual species. Results indicate that wetland plant communities are at the highest risk of invasion by transgenic A. stolonifera. Also, inter-specific gene flow to A. gigantea could affect additional habitats and plant communities where A. stolonifera is not found. Both A. stolonifera and A. gigantea were invasive in wetland and riparian settings in the Madras study area, and introducing glyphosate (e.g., Roundup®, Rodeo®) herbicide tolerance into these populations would eliminate the primary means of control for these species.  相似文献

Uncertainty about controls on long-term carbon (C) and nitrogen (N) balance, turnover, and isotopic composition currently limits our ability to predict ecosystem response to disturbance and landscape change. We used a two-century, postglacial chronosequence in Glacier Bay, Alaska, to explore the influence of C and N dynamics on soil and leaf stable isotopes. C dynamics were closely linked to soil hydrology, with increasing soil water retention during ecosystem development resulting in a linear decrease in foliar and soil δ13C, independent of shifts in vegetation cover and despite constant precipitation across sites. N dynamics responded to interactions among soil development, vegetation type, microbial activity, and topography. Contrary to the predictions of nutrient retention theory, potential nitrification and denitrification were high, relative to inorganic N stocks, from the beginning of the chronosequence, and gaseous and hydrological N losses were highest at mid-successional sites, 140–165 years since deglaciation. Though leaching of dissolved N is considered the predominant pathway of N loss at high latitudes, we found that gaseous N loss was more tightly correlated with δ15N enrichment. These results suggest that δ13C in leaves and soil can depend as much on soil development and associated water availability as on climate and that N availability and export depend on interactions between physical and biological state factors.  相似文献

Alteration of soil nutrient dynamics has recently garnered more attention as both a cause and an effect of plant invasion. This project examines how nutrient dynamics are affected by native (Elymus elymoides, Pseudoroegneria spicata, and Vulpia microstachys) and invasive (Aegilops triuncialis, Agropyron cristatum, Bromus tectorum, and Taeniatherum caput-medusae) grass species. This research questions whether natives and invasives differ in their effects on nutrient dynamics. A greenhouse study was conducted using two field-collected soils. Effects on nutrient dynamics were compared using an integrated index that evaluates the total nutrients in soil and in plant tissue compared to an unplanted control. With this index, we evaluated whether soil nutrients increased or decreased as a result of plant growth, controlling for plant uptake. We found no consistent support for our hypothesis that invasive grass species as a group influence nutrient dynamics differently than native grass species as a group. Our results indicate species-specific effects on nutrient dynamics. Alteration of nutrient dynamics is not a trait shared by all of the invasive grass species in our study. However, alteration of nutrient dynamics may be a mechanism by which some individual species increase their invasive potential.  相似文献

Abstract

Rust, anthracnose, and angular leaf spot are major diseases of common bean in the Americas and Africa. The cultivar Ouro Negro has the Ur-14 gene that confers broad spectrum resistance to rust and the gene cluster Co-3 4 /Phg-3 containing two tightly linked genes conferring resistance to anthracnose and angular leaf spot, respectively. We used co-segregation analysis and high-throughput genotyping of 179 F2:3 families from the Rudá (susceptible) × Ouro Negro (resistant) cross-phenotyped separately with races of the rust and anthracnose pathogens. The results confirmed that Ur-14 and Co-3 4 /Phg-3 cluster in Ouro Negro conferred resistance to rust and anthracnose, respectively, and that Ur-14 and the Co-3 4 /Phg-3 cluster were closely linked. Genotyping the F2:3 families, first with 5398 SNPs on the Illumina BeadChip BARCBEAN6K_3 and with 15 SSR, and eight KASP markers, specifically designed for the candidate region containing Ur-14 and Co-3 4 /Phg-3, permitted the creation of a high-resolution genetic linkage map which revealed that Ur-14 was positioned at 2.2 cM from Co-3 4 /Phg-3 on the short arm of chromosome Pv04 of the common bean genome. Five flanking SSR markers were tightly linked at 0.1 and 0.2 cM from Ur-14, and two flanking KASP markers were tightly linked at 0.1 and 0.3 cM from Co-3 4 /Phg-3. Many other SSR, SNP, and KASP markers were also linked to these genes. These markers will be useful for the development of common bean cultivars combining the important Ur-14 and Co-3 4 /Phg-3 genes conferring resistance to three of the most destructive diseases of common bean.

Drainage ditches are ubiquitous yet understudied features of the agricultural landscape. Nitrogen pollution disrupts the nutrient balance of drainage ditch ecosystems, as well as the waterbodies in which they drain. Denitrification can help ameliorate the impact of N-fertilization by converting reactive nitrogen into dinitrogen gas. However, factors affecting denitrification in drainage ditches are still poorly understood. In this study, we tested how within-ditch and regional environmental conditions affect denitrifier activity, abundance, and community structure, to understand controls on denitrification at multiple scales. To this end, we quantified in situ denitrification rates and denitrifier abundance in 13 drainage ditches characterized by different types of sediment, vegetation and land-use. We determined how denitrification rates relate to denitrifier abundance and community structure, using the presence of nirS, nirK and nosZ genes as a proxy. Denitrification rates varied widely between the ditches, ranging from 0.006 to 24 mmol N m?2 h?1. Ditches covered by duckweed, which contained high nitrate concentrations and had fine, sandy sediments, were denitrification hotspots. We found highest rates in ditches next to arable land, followed by those in grasslands; lowest rates were observed in peatlands and nature reserves. Denitrification correlated to nitrate concentrations, but not to nirK, nirS and nosZ abundance, whereas denitrifier-gene abundance correlated to organic matter content of the sediment, but not to nitrate concentrations. Our results show a mismatch in denitrification regulators at its different organizational scales. Denitrifier abundance is mostly regulated at within-ditch scales, whereas N-loads, regulated by landscape factors, are most important determinants of instantaneous denitrification rates.  相似文献

To examine the effects of soil fertility on biomass production, plant species diversity, and early vegetation development, we performed a mesocosm experiment using soil seed bank under three soil fertility levels (ombrotrophic, mesotrophic, and eutrophic). Biomass production linearly increased (P < 0.01), whereas plant species diversity significantly decreased (P < 0.001) as soil fertility increased. Soil fertility seemed to play a role as an environmental sieve in early vegetation development, in turn, lead the patterns of biomass production and plant species diversity. Several Poaceae species which are forming tall and dense canopy, such as Phalaris arundinacea, Miscanthus sacchariflorus, and Zizania latifolia, showed higher importance values under fertilized condition, whereas relatively small macrophytes of Eleocharis acicularis, Carex dickinsii, and Isachne globosa mainly survived under ombrotrophic condition. Although plant species richness decreased as soil fertility increased, the percentages of perennials and exotics increased rather than annuals and natives. Predominance of perennials and exotics under eutrophic condition in early vegetation development seemed to lead a continuous decrease in plant species diversity as a result of competitive effect. A significant negative relationship between plant species diversity and biomass production (P < 0.0001) was obtained when all experimental plots are included in correlation analysis.  相似文献

Abstract

Zhou8425B is an elite wheat (Triticum aestivum L.) line widely used as a parent in Chinese wheat breeding programs. Identification of genes for adult-plant resistance (APR) to powdery mildew in Zhou8425B is of high importance for continued controlling the disease. In the current study, the high-density Illumina iSelect 90K single-nucleotide polymorphism (SNP) array was used to map quantitative trait loci (QTL) for APR to powdery mildew in 244 recombinant inbred lines derived from the cross Zhou8425B/Chinese Spring. Inclusive composite interval mapping identified QTL on chromosomes 1B, 3B, 4B, and 7D, designated as QPm.caas-1BL.1, QPm.caas-3BS, QPm.caas-4BL.2, and QPm.caas-7DS, respectively. Resistance alleles at the QPm.caas-1BL.1, QPm.caas-3BS, and QPm.caas-4BL.2 loci were contributed by Zhou8425B, whereas that at QPm.caas-7DS was from Chinese Spring. QPm.caas-3BS, likely to be a new APR gene for powdery mildew resistance, was detected in all four environments. One SNP marker closely linked to QPm.caas-3BS was transferred into a semi-thermal asymmetric reverse PCR (STARP) marker and tested on 103 commercial wheat cultivars derived from Zhou8425B. Cultivars with the resistance allele at the QPm.caas-3BS locus had averaged maximum disease severity reduced by 5.3%. This STARP marker can be used for marker-assisted selection in improvement of the level of powdery mildew resistance in wheat breeding.
