用40条多态引物对乌羊、小香羊、南江黄羊、黑山羊、川东白山羊、波尔山羊和马头山羊7个品种(或群体)进行RAPD分析,其中28条引物扩增出多态性谱带,并用于进一步对12只乌羊个体和12只小香羊个体基因组进行扩增.扩增产物以1.5%琼脂糖凝胶(含0.5μg/ml溴化乙锭)电泳分离.Nei氏公式计算品种间的遗传距离指数和品种内的遗传相似指数,NJ法构建系统聚类图.结果表明:乌羊和川东白山羊间的遗传距离最小,亲缘关系较近,而小香羊与各品种间的遗传距离都较大,亲缘关系较远.乌羊群体及小香羊品种都具有一定的遗传稳定性。 Abstract:A total of 40 primers generated polymorphic loci were used in random amplified polymorphism DNA(RAPD)analysis in seven goat breeds(or populations)including Wu goat,Small-Xiang goat,Nanjiang Brown goat,Black goat,Chuandong white goat,Boer goat,Matou goat.28 of them were selected for further amplification in individuals of 12 Wu goats and 12 Small-Xiang goats because of their different amplified patterns among seven goat breeds.Amplification products were separated by 1.5% agarose gel(contain 0.5μg/ml ethidium bromide)electrophoresis.Genetic distance indexes among breeds and genetic similarity indexes within breed were calculated by Nei,and the phylogenetic tree was constructed by NJ method.The results indicate that the genetic distance index between Wu goat population and Chuandong white goat breed is smallest,therefore Wu goat population is closely related to Chuandong white goat breed.While the genetic distance indexes between Small-Xiang goat breed and any other goat breeds are all big,so Small-Xiang goat breed is far related to any other goat breeds.The results also indicate that the genetic stability either among the Wu goat population or the Small-Xiang goat breed reaches some extent.  相似文献

贵州地方山羊品种的RAPD分析   总被引:12,自引:0,他引:12  
用180条引物对黔东南小香羊、贵州白山羊、贵州黑山羊和黔北麻羊4个贵州地方山羊品种(种群),以及南江黄羊和波尔山羊进行RAPD分析,其中27条引物扩增出多态性图谱。这27条引物共扩增出281条带,多态带为115条,平均多态频率为40.92%(范围20%~80%);每条引物平均扩增条带为10.41条(范围4~16条);扩增带分子量在210~2800bp。贵州白山羊与贵州黑山羊之间的遗传距离指数(0.0605)最小,而波尔山羊与其他品种之间的遗传距离指数(0.1059~0.1488)最大。NJ法聚类结果显示,贵州白山羊与贵州黑山羊间的亲缘关系最近,其次为黔北麻羊,而黔东南小香羊与其他3个贵州地方品种的亲缘关系较南江黄羊还远。分析表明,黔东南小香羊在遗传上为一独立的品种;而贵州地方山羊品种间具有较近的亲缘关系,遗传变异较小,具有较高的遗传稳定性。  相似文献

研究了AFLP标记在研究小香羊遗传多态性方面应用的可行性和该山羊个体基因组DNA的AFLP扩增结果。实验应用10条AFLP引物,用PstI酶切,对15只小香羊基因组DNA进行AFLP反应,共获得113个AFLP标记,单引物获得的标记数在2~19之间,小香羊群体相似系数AFLP研究结果为0.913(0.814~0.980)。该研究为评价小香羊的遗传稳定性提供了相关的参数,准确评价尚待和其它品种对比研究后确定。  相似文献

采用15种限制性内切酶,研究贵州省4个山羊品种共93只个体的线粒体DNA多态性。其中BomHI、HindⅢ和SalⅠ3种酶的酶切类型存在多态。共检测到18种限制性态型,归结为3种mtDNA单倍型。单倍型Ⅰ、Ⅱ在贵州山羊4个品种分布频率较高,分别为77.42%和21.50%,单倍型Ⅲ分布频率较低(1.08%);品种间亲缘关系聚类分析表明白山羊和黑山羊亲缘关系最近,其次为黔林羊,而与小香羊的亲缘关系最  相似文献

对蒙古羊、湖羊、滩羊、小尾寒羊、乌珠穆沁羊、藏绵羊、阿勒泰羊7个地方绵羊品种和无角陶赛特羊、德国美利奴羊、萨福克羊3个引入品种基因组DNA进行了RAPD分析。结果表明:(1)RAPD可作为一种有效的标记用于绵羊品种之间遗传亲缘关系的分析。(2)在所使用的43种随机引物中,有35种引物扩增出多态谱带,多态频率为66.24%,说明RAPD技术用于研究绵羊核DNA的遗传变异具有较高的检出率和灵敏度。(3)总群体平均遗传多样性指数(HSP)为0.9139,说明绵羊群体具有较为丰富的遗传多样性。(4)我国地方绵羊品种间的分子聚类关系与其所处的地理位置、考古学结果,以及细胞遗传学研究结果基本,引入品种间的分子聚类关系也与其育成史基本一致。  相似文献

采用18个微卫星位点对河南省5个地方山羊品种(牛腿山羊、槐山羊、河南奶山羊、太行黑山羊和伏牛白山羊)的遗传多样性进行了评价.结果表明:18个微卫星位点在5个山羊品种均为高度多态;5个山羊品种的多态信息含量、群体杂合度、有效等位基因数值较高,说明其遗传多样性和各品种内的遗传变异比较丰富;聚类分析显示,牛腿山羊与太行黑山羊的关系较近,与河南奶山羊的关系较远.群体遗传分化系数和群体遗传距离表明,河南省地方山羊的变异主要存在于品种内,品种间的变异相对较小.结合5个山羊品种的实际生态地理分布,提出了避免近交,并有选择地进行品种间杂交的保种模式.  相似文献

波尔山羊杂交后代及其亲本随机扩增多态DNA研究   总被引:24,自引:4,他引:20  
利用随机扩增多态DNA技术研究了波尔山羊、唐山奶山羊、青龙本地山羊以及波尔山羊与这两个山羊群体杂交后代共计128个山羊个体的随机扩增多态DNA。结果表明:(1)总群体平均遗传多样性指数(Hsp)为0.6974,群体遗传分化指数为0.9706,山羊群体间平均遗传距离指数(0.1314~0.2052)明显大于群体内的相应值(0.0582~0.1440),上述结果说明,所研究山羊群体不仅具有较为丰富的遗传多样性,而且其核基因组遗传变异主要存在于群体间。(2)山羊群体间的分子聚类关系与各群体间的亲缘关系基本一致。 Abstract:The random amplified polymorphic DNA(RAPD)of 128 individuals was studied,which were from Boer goat, Tangshan dairy goat,Qinglong native goat and their hybrids crossbred with Boer goat.The average index of genetic polymorphism for whole population(Hsp)and the index of genetic differentiation were 0.6974 and 0.9706,respectively.The average index of genetic distance between populations(0.1314~0.2052)was significantly higher than that within populations(0.0582~0.1440).All of these indicated that the genetic polymorphism was not only abundant,but also the genetic variation was mainly existed between goat populations.The molecular dendrogram among goat populations was in accord with their genetic relationship.  相似文献

报道了AFLP标记在研究贵州小型香猪遗传多性方面的应用和该品系猪个体基因组DNA的AFLP扩增结果,分析了贵州小型香猪的群体遗传结构。实验应用10条AFLP引物,用PstⅠ酶切,对17头猪基因组DNA进行AFLP反应,共获得116个AFLP标记,单引物获得的标记数在2-22间,贵州小型香猪群体相似系数AFLP研究结果为0.866(0.760-0.967),该研究为贵州小型香猪的遗传稳定性提供了相关的参数,准确评价尚待和其它品种猪对研究后确定。  相似文献

小尾寒羊是世界上具有非季节性发情和多胎特性的高繁殖率绵羊品种之一。选择4个位于绵羊6号染色体上且与FecB基因紧密连锁的微卫星标记,对小尾寒羊基因组进行PCR扩增后,采用最小二乘法估计各等位基因片段对产羔数的影响。分析结果表明,等位基因OarJIA-5、OarJIA-10、BM143-12和OarHH55-11可以作为小尾寒羊多胎位点的分子标记。从100条随机引物中筛选出18条引物,对小尾寒羊、大尾寒羊、洼地绵羊、滩羊等4个绵羊品种和鲁北山羊的基因组进行扩增,共扩增出146条带,其中94条表现出多态性,占64.60%,同时扩增出每个品种的特异性条带。采用Nei氏标准距离和NJ聚类分析对不同品种的遗传关系进行分析,结果表明,4个绵羊品种亲缘关系很近,提示它们可能起源于共同的原始祖先。  相似文献

目的建立树鼩RAPD(random amplified polymorphic DNA)遗传标记分析方法,了解中缅树鼩种群的遗传多样性。方法采用PCR技术对40条随机引物进行优化,筛选出能有效用于树鼩群体遗传分析的RAPD位点,对48只树鼩个体的基因组DNA进行PCR扩增,并应用Popgene 1.32与RAPDistance Package Version 1.04等软件进行数据处理,分析树鼩的群体遗传特性。结果20个RAPD引物共检测到113个位点,平均每个引物可扩增出5.65个位点,其中多态位点数为69个(占61.1%)。个体间的遗传相似系数平均为0.8307,个体间遗传距离在0.09-0.27之间,平均遗传距离为0.1693。雄性群体的遗传多态度(H0)(0.1864)略高于雌性群体(0.1470),平均遗传多态度(Hpop)为0.1667;树鼩遗传多态度所占的比例在群体内为48.29%,而雌、雄群体间为51.71%。NJ法进行聚类分析得出T15、T33和T47号树鼩个体聚类成一大类,其余45只树鼩个体聚成另一大类,雌、雄个体呈相互交叉现象。结论实验所筛选的随机引物可有效用于中缅树鼩种群的遗传结构分析。本树鼩群体具有较好的遗传多样性。  相似文献

Abstract: I investigated seasonal altitudinal movements of 42 mountain goats (Oreamnos americanus) in the Cascade Range of Washington, USA. Because mountain goats typically move to lower elevations during the winter, I partitioned locations from Global Positioning System collars into summer and winter seasons based on elevation. Using an iterative narrowing search, I identified summer and winter start dates for each individual and year and derived several measures of altitudinal movements from these, and examined differences in these measures on the basis of sex and year and their interrelationship. Generally, female mountain goats started summer about 2 weeks earlier than nondispersing males; winter start dates varied among years. Horizontal distance moved between seasons was unrelated to measures of altitudinal movement. Based on elevation, winters were generally longer than summers for mountain goats I studied, suggesting that the common perception of mountain goats as inhabitants of alpine and subalpine terrain is biased, because they spent the greater part of the year at lower elevations. Mountain goats showed a wide range of responses to seasonal environmental changes and individuals cannot be easily classified as migratory or nonmigratory. Because ecological conditions in mountain environments are closely related to elevation and horizontal and altitudinal movements were unrelated, studies of seasonal movements of mountain animals based on horizontal movement may be misleading. Because seasonal altitudinal movements of mountain goats are highly variable, the management needs of each population must be considered separately.  相似文献

本文报道2例两性畸形的山羊,它们均具雌雄两套生殖器官,染色体组型分析显示它们均为性染色体嵌合体,核型为60,XX/XY,两头畸形山羊的XX细胞系的比例分别为42%和54%,XY细胞系的比例分别为58%和46%。 Abstract:We herein report two cases of goats with sexual deformity.They had two systems of reproductive organs and the karyotype analysis showed mosaicism for their sex chromosomes,i.e.,60,XX / XY.The proportion of XX cell line was 42% and 54%,while XY cell line was 58% and 46%,respectively,in the two abnormal goats.  相似文献

用10对山羊微卫星DNA多态性引物对2只体细胞克隆济宁青山羊、青山羊供体细胞、受体奶山羊母羊以及具有亲缘关系的3只对照济宁青山羊进行微卫星DNA分析.结果表明有5对山羊微卫星DNA多态性引物,即SR-CRSP1, SR-CRSP5, SR-CRSP6, SR-CRSP7和SR-CRSP24,扩增产物有明显的多态性.扩增产物经过6%变性聚丙烯酰胺凝胶电泳后银染,结果2只体细胞克隆山羊的微卫星DNA指纹与供体细胞完全相同,而且不同于其受体母亲也不同于其他所有同品种不同个体的对照青山羊.证明体细胞克隆山羊基因组来源于供体细胞.  相似文献

The Genetic Diversity of mtDNA D-loop and the Origin of Chinese Goats   总被引:5,自引:0,他引:5  
The complete sequences of mitochondrial DNA D-loop of 128 individuals in nine Chinese goat (Capra hircu) breeds were analyzed by DNA sequencing technology. The results show that the length of mtDNA D-loop in Chinese goats is 1 212-1 213 bp. There are 102 polymorphic sites, accounting for 8.42% of 1 212 bp sequence. Ninety-two mtDNA haplotypes were determined. The haplotype diversity and nucleotide diversity are 0.9333-1 .0000 and 0.7062%-1.8265%, respectively. The results indicate that the genetic diversity of Chinese goats is very abundant. The NJ tree indicates that Chinese goats have two types of maternal origins from lineage A and lineage B. The possibility of lineage B originating from China is also discussed.  相似文献

在松嫩平原羊草草地,通过控制放牧实验对山羊的时限采食量和食性选择进行了研究。结果表明:(1)5-9月份,山羊的时限(1h)采食量平均为0.42kg干物质,其季节动态为5月份最低,随季节推移不断增大,8月份达到最大,9月份又有所减小;时限采食量基本上随放牧率减小而增大,但在最低放牧率小区有所减小。(2)山羊的食性选择随季节推移和放牧率不同而变化。(3)山羊对20-25cm高度草层的选择性最高;各高度草层的食性选择指数随季节推移和放牧率不同而变化;山羊对不同植物的高度选择性存在差异,但高度选择指数的最大值都在15-30cm范围内。  相似文献

本研究以辽宁绒山羊、柴达木绒山羊和柴达木山羊3个群体共147只山羊为研究对象,运用PAGE和RAPD技术对山羊的体重、绒产量和绒细度3个性状进行了与标记基因关系的遗传分析,结果表明:EsD2-2型、LAPBB型和PA-32-2型分别为体重、绒产量和绒细度性状的优势标记基因型;可利用标记辅助预测的方法充分利用多基因座标记基因间的互作效应;在体重上,寻找到有显著选择效应的RAPD条带11个,在绒产量和绒细度上分别为9和6个;在多目标性状选择中,CY0818/A0型和OPW19/C1型为体重和绒产量的双重优势RAPD标记,CY0818/G1型为体重和绒细度的双重优势RAPD标记。Abstract: The genetic relationships between economic traits and genetic markers were studied in 147 goats including Chaidamu goat (CS), Chaidamu Cashmere goat (CRS) and Liaoning Cashmere goat (LRS) in Qinghai province, China. CRS was the population of CS×LRS crossbred. The results showed as follows: the selection reaction of these blood protein polymorphisic loci were great, such as EsD, LAP and PA-3; and EsD2-2, LAPBB and PA-32-2 were the superior marker genotypes on body weight ,Cashmere yield and Cashmere fineness respectively by Least Square method. The interaction between marker genotypes at double loci was found frequently, and their ratio between interaction variance component and genetic variance was higher. With the method of marker assisted prediction( MAP), some interaction effect could be used effectively in the crossbreed population. On the aspect of random amplified polymorphic DNA (RAPD), the number of the superior RAPD marker bands were 11 on body weight trait, 9 and 6 RAPD marker bands on Cashmere yield and Cashmere fineness. For multi-goal traits, CY0818/A0 type and OPW19/C1 type were superior RAPD markers of body weight and Cashmere yield, CY0818/G1 type was superior one of body weight and Cashmere fineness.  相似文献

Viable Transgenic Goats Derived from Skin Cells   总被引:3,自引:0,他引:3  
The current study was undertaken to evaluate the possibility of expanding transgenic goat herds by means of somatic cell nuclear transfer (NT) using transgenic goat cells as nucleus donors. Skin cells from adult, transgenic goats were first synchronized at quiescent stage (G0) by serum starvation and then induced to exit G0 and proceed into G1. Oocytes collected from superovulated donors were enucleated, karyoplast-cytoplast couplets were constructed, and then fused and activated simultaneously by a single electrical pulse. Fused couplets were either co-cultured with oviductal cells in TCM-199 medium (in vitro culture) or transferred to intermediate recipient goat oviducts (in vivo culture) until final transfer. The resulting morulae and blastocysts were transferred to the final recipients. Pregnancies were confirmed by ultrasonography 25-30 days after embryo transfer. In vitro cultured NT embryos developed to morulae and blastocyst stages but did not produce any pregnancies while 30% (6/20) of the in vivo derived morulae and blastocysts produced pregnancies. Two of these pregnancies were resorbed early in gestation. Of the four recipients that maintained pregnancies to term, two delivered dead fetuses 2-3 days after their due dates, and two recipients gave birth to healthy kids at term. Fluorescence in situ hybridization (FISH) analysis confirmed that both kids were transgenic and had integration sites consistent with those observed in the adult cell line.  相似文献

选用5只年龄为3~4岁的波尔山羊公羊研究在稀释液中添加海藻糖对山羊精子功能和膜完整性的影响。山羊精子分别用含6.6 mmol/L、13.2 mmol/L、19.8 mmol/L、26.4 mmol/L、39.6 mmol/L、52.9 mmol/L、66.1mmol/L、79.3 mmol/L的不同海藻糖的Tris-柠檬酸-葡糖糖(TCG)稀释液(卵黄:18%;甘油:6%)稀释和冷冻。结果表明:39.6 mmol/L、52.9 mmol/L、66.1 mmol/L、79.3 mmol/L组降温后的精子活率显著(P<0.05)降低;52.9 mmol/L、66.1 mmol/L、79.3 mmol/L组降温后的精子畸形率和39.6 mmol/L组降温后的膨胀精子率显著(P<0.05)提高。26.4 mmol/L组和39.6 mmol/L组冻融后的精子活率显著(P<0.05)高于对照组;66.1mmol/L和79.3 mmol/L组冻融后的精子活率、畸形率分别显著(P<0.05)低于和高于对照组。19.8 mmol/L、26.4 mmol/L、39.6 mmol/L组冻融后精子获能率显著(P<0.05)低于对照组。39.6 mmol/L组冻融后顶体完整率和膨胀精子率显著(P<0.05)高于对照组,而66.1 mmol/L组和79.3 mmol/L组显著(P<0.05)低于对照组。39.6 mmol/L组的受胎率显著(P<0.05)高于对照组,而66.1mmol/L组和79.3 mmol/L组的受胎率显著(P<0.05)低于对照组。结果表明,在含18%的卵黄(v/v)、6%甘油(v/v)的TCG稀释液中,添加适宜浓度(26.4mmol/L和39.6 mmol/L)海藻糖,可显著提高山羊精子功能和膜的完整性。  相似文献

在已经获得的乳腺特异性表达人促红细胞生成素 (rhEPO)成年转基因山羊 (Caprahircus)的基础上 ,取其耳尖成纤维细胞和卵巢颗粒细胞 ,进行体外传代培养 ,然后将这种培养的转基因山羊的体细胞移入去核的处于第Ⅱ次减数分裂中期的卵母细胞中 ,并进行电融合 ,构建重构胚胎 ,重构胚胎在体内培养 6d ,再将发育至囊胚或桑椹胚的重构胚胎移入同步情期的寄母羊子宫内。结果 ,有 2只寄母羊妊娠并最终产下 2只成活的克隆山羊。她们分别来自同一成年母羊的耳尖成纤维细胞和卵巢颗粒细胞。克隆羊经PCR RFLP图谱分析显示 :以克隆羊组织DNA为模板的PCR产物与相应的提供体细胞的基因羊的PCR产物的酶切图谱完全一致 ;并且经PCR对外源hEPO基因检测表明 2只克隆山羊均携带hEPO外源基因。由此证明获得了转基因成年体细胞的克隆山羊  相似文献

生长激素基因多态性与山羊体重性状的关系   总被引:20,自引:0,他引:20  
以鲁北白山羊、波尔山羊以及波尔山羊与鲁北白山羊的杂交一代、回交一代共计224只山羊为材料,根据山羊生长激素基因5′调控区的序列(GenBank登录号:D00476)设计两对引物,用PCRSSCP法进行多态性分析。多态性片段的纯合基因型经克隆测序,发现共有5处突变。对突变位点产生的不同基因型与体重、体尺性状进行分析表明:第一对引物扩增片段波尔山羊AA型个体的初生重、周岁重显著高于BB型和AB型(P<0.05);杂交一代中断奶重、周岁重也以AA型偏高,但不同基因型间的差异均不显著(P>0.05);而鲁北白山羊BB基因型的体重相对于另外两种基因型的偏低,且断奶重的差异达到显著水平(P<0.05)。第二对引物扩增片段不同基因型对体重、体尺没有显著影响。由此初步推断生长激素基因可能是影响山羊体重性状的主基因或与主基因相连锁,可以用该位点对山羊体重性状进行标记辅助选择。  相似文献
