To achieve nitritation from complete-nitrification seed sludge at room temperature of 19 ± 1 °C, a lab-scale sequencing batch reactor (SBR) treating domestic wastewater with low C/N ratios was operated to investigate the control and optimization of nitrifying communities. Ammonia oxidizing bacteria (AOB) dominance was enhanced through the combination of low DO concentrations (<1.0 mg/L) and preset short-cycle control of aeration time. Nitritation was successfully established with NO2?-N/NOx?-N over 95%. To avoid the adverse impact of low DO concentrations on AOB activities, DO concentrations were increased to 1–2 mg/L. At the normal DO levels and temperatures, on-line control strategy of aerobic durations maintained the stability of nitritation with nitrite accumulation rate over 95% and ammonia removal above 97%. Fluorescence in-situ hybridization (FISH) analysis presented that the maximal percentage of AOB in biomass reached 10.9% and nitrite oxidizing bacteria (NOB) were washed out.  相似文献

Activated sludge obtained from two municipal wastewater treatment facilities (WWTF) was used as seed sludge for enriched nitrifiers, which were later entrapped in polyvinyl alcohol. Seed sludge from one WWTF was acclimated to high ammonia level (1813 mg NH3-N l?1) through the return of sludge digester supernatant back to primary clarifier while seed sludge from the other WWTF was un-acclimated. To elucidate on how to control partial nitrification by entrapped cells, which could be different from suspended cells, kinetics of entrapped enriched nitrifiers were studied using a respirometric assay. The community of nitrifiers within the entrapment matrix, which was observed by fluorescence in situ hybridization (FISH) technique, was related to the nitritation and nitratation kinetics based on oxygen uptake rate. Maximum oxygen uptake rate, and substrate and oxygen affinities of both ammonia oxidizing bacteria (AOB) for nitritation and nitrite oxidizing bacteria (NOB) for nitratation in entrapped cells were lower than those of corresponding suspended cells. Under dissolved oxygen (DO) limiting conditions, nitratation was more suppressed than nitritation for suspended cells, while for the entrapped cells, the results were the contrary. A free ammonia (FA) inhibition affected only the un-acclimated sludge. Either FA inhibition or DO limitation might not be a sole effective control parameter to achieve partial nitrification by entrapped cells. FISH results revealed that Nitrosomonas europaea was the dominant AOB while Nitrobacter species was the dominant NOB in all cases. Heterotrophs were also present in the entrapment at 22.8 ± 18.6% and 41.5 ± 4.3% of total bacteria for acclimated and un-acclimated originated sludge. The availability of substrate and oxygen governed the distributions of AOB, NOB and heterotrophs within the entrapment and nitritation kinetics of entrapped nitrifiers.  相似文献

The coupled system of partial nitrification and anaerobic ammonium oxidation (Anammox) is efficient in nitrogen removal from wastewater. In this study, the effect of different oxygen concentrations on partial nitrification performance with a sequencing batch reactor (SBR) was investigated. Results indicate that, partial nitrification of landfill leachate could be successfully achieved under the 1.0–2.0 mg L−1 dissolved oxygen (DO) condition after 118 d long-term operation, and that the effluent is suitable for an Anammox reactor. Further decreasing or increasing the DO concentration, however, would lead to a decay of nitrification performance. Additionally, the MLSS concentration in the reactor increased with increasing DO concentration. Respirometric assays suggest that low DO conditions (<2 mg L−1) favor the ammonia-oxidizing bacteria (AOB) and significantly inhibit nitrite oxidizing bacteria (NOB) and aerobic heterotrophic bacteria (AHB); whereas high DO conditions (>3 mg L−1) allow AHB to dominate and significantly inhibit AOB. Therefore, the optimal condition for partial nitrification of landfill leachate is 1.0–2.0 mg L−1 DO concentration.  相似文献

A comparative study to produce the correct influent for Anammox process from anaerobic sludge reject water (700–800 mg NH4+-N L−1) was considered here. The influent for the Anammox process must be composed of NH4+-N and NO2-N in a ratio 1:1 and therefore only a partial nitrification of ammonium to nitrite is required. The modifications of parameters (temperature, ammonium concentration, pH and solid retention time) allows to achieve this partial nitrification with a final effluent only composed by NH4+-N and NO2-N at the right stoichiometric ratio. The equal ratio of HCO3/NH4+ in reject water results in a natural pH decrease when approximately 50% of NH4+ is oxidised. A Sequencing batch reactor (SBR) and a chemostat type of reactor (single-reactor high activity ammonia removal over nitrite (SHARON) process) were studied to obtain the required Anammox influent. At steady state conditions, both systems had a specific conversion rate around 40 mg NH4+-N g−1 volatile suspended solids (VSS) h−1, but in terms of absolute nitrogen removal the SBR conversion was 1.1 kg N day−1 m−3, whereas in the SHARON chemostat was 0.35 kg N day−1 m−3 due to the different hydraulic retention time (HRT) used. Both systems are compared from operational (including starvation experiments) and kinetic point of view and their advantages/disadvantages are discussed.  相似文献

The effect of the changes of culturing environments of Aspergillus terreus on lovastatin production was investigated in the study. A relatively low supplement of dissolved O2 (DO) by the fungus almost stopped performing product formation. With the DO controlled at 20%, lovastatin production using a 5-l fermenter enhanced by 38%, biomass production decreased by 25% and sugar utilization increased by 18%, as compared with the shaking-flask culture. Meanwhile, an average diameter 0.95 mm of compact pellets was found. We thus concluded that pellet formation with a narrow size distribution dominated lovastatin production by A. terreus, which was closely affected by the relatively saturated level of DO. Nevertheless, manipulating the broth pH at 5.5–7.5 starting from 48 h provided no benefit to product formation although biomass production was reduced largely. In the part of work, a pH/DO interaction was also confirmed.A simple temperature-shift method (28–23 °C) was proved surprisingly valuable to the fermentation process. Such experiments showed that the maximum of lovastatin production was further enhanced by 25% (572 mg/l at day 10) in comparison with that when the fungus was cultured at 28 °C. The timing to initiate the temperature-shift (96 h) corresponded to that of pellet formation and the subsequent core compactness. Hence, it was found that lovastatin production by A. terreus favored sub-optimal growth conditions.  相似文献

Bacterial strain QZ1 was isolated from sludge of anoxic sulfide-oxidizing (ASO) reactor. Based on 16S rDNA sequence analysis and morphological characteristics, the isolate was identified as Pseudomonas stutzeri. The isolate was found to be a facultative chemolithotroph, using sulfide as electron donor and nitrite as electron acceptor. The strain QZ1 produced sulfate as the major product of sulfide oxidation, depending on the initial sulfide and nitrite concentrations. The isolate was capable of growth under strictly autotrophic conditions. The growth and substrate removal of Pseudomonas stutzeri QZ1 were optimal at an initial pH of 7.5–8.0 at 30 °C. The specific growth rate (μ) was found as 0.035 h−1 with a doubling time of 21.5 h. For isolate QZ1, the EC50 values both for sulfide and nitrite were found to be 335.95 mg S L−1 and 512.38 mg N L−1, respectively, showing that the sulfide oxidation into sulfate by Pseudomonas stutzeri QZ1 was badly affected beyond these substrate concentrations.  相似文献

The objective of this study was to investigate nitrification rates in algal–bacterial biofilms of waste stabilization ponds (WSP) under different conditions of light, oxygen and pH. Biofilms were grown on wooden plates of 6.0 cm by 8.0 cm by 0.4 cm in a PVC tray continuously fed with synthetic wastewater with initial NH4-N and Chemical Oxygen Demand (COD) concentrations of 40 mg l?1 and 100 mg l?1, respectively, under light intensity of 85–95 μE m?2 s?1. Batch activity tests were carried out by exposure of the plates to light conditions as above (to simulate day time), dim light of 1.8–2.2 μE m?2 s?1 (to simulate reduced light as in deeper locations in WSP) and dark conditions (to simulate night time). Dissolved oxygen (DO) concentration and pH were controlled. At some experiments, both parameters were kept constant, and at others they were left to vary as in WSP. Results show biofilm nitrification rates of 945–1817 mg-N m?2 d?1 and 1124–1615 mg-N m?2 d?1 for light and dark experiments. When the minimum DO was 4.1 mg l?1, the biofilm nitrification rates under light and dark conditions did not differ significantly at 95% confidence. When the minimum DO in the dim light experiment was 3.2 mg l?1, the nitrification rates under light and dim light conditions were 945 mg-N m?2 d?1 and 563 mg-N m?2 d?1 and these significantly differed. Further decrease of DO to 1.1 mg l?1 under dark conditions resulted in more decrease of the nitrification rates to 156 mg-N m?2 d?1. It therefore seems that under these experimental conditions, biofilm nitrification rates are significantly reduced at a certain point when bulk water DO is between 3.2 mg l?1 and 4.1 mg l?1. As long as bulk water DO under dark is high, light is not important in influencing the process of nitrification.  相似文献

Ten different seaweed species were compared on the basis of lead uptake at different pH conditions. The brown seaweed, Turbinaria conoides, exhibited maximum lead uptake (at pH 4.5) and hence was selected for further studies. Sorption isotherms, obtained at different pH (4–5) and temperature (25–35 °C) conditions were fitted using Langmuir and Sips models. According to the Langmuir model, the maximum lead uptake of 439.4 mg/g was obtained at optimum pH (4.5) and temperature (30 °C). The Sips model better described the sorption isotherms with high correlation coefficients at all conditions examined. Various thermodynamic parameters such as ΔG°, ΔH° and ΔS° were calculated indicating that the present system was a spontaneous and endothermic process. Through potentiometric titrations, number of binding sites (carboxyl groups) and pK1 were determined as 4.1 mmol/g and 4.4, respectively. The influence of co-ions (Na+, K+, Mg2+ and Ca2+) on lead uptake was well pronounced in the case of divalent ions compared to monovalent ions. The solution of 0.1 M HCl successfully eluted all lead ions from lead-loaded T. conoides biomass. The regeneration experiments revealed that the alga could be successfully reused for five cycles without any loss in lead biosorption capacity. A glass column (2 cm i.d. and 35 cm height) was used to study the continuous lead biosorption performance of T. conoides. At 25 cm (bed height), 5 ml/min (flow rate) and 100 mg/l (initial lead concentration), T. conoides exhibited lead uptake of 220.1 mg/g. The column was successfully eluted using 0.1 M HCl, with elution efficiency of 99.7%.  相似文献

In recent decades, β-xylosidases have been used in many processing industries. In this work, the study of xylosidase production by Penicillium sclerotiorum and its characterization are reported. Optimal production was obtained in medium supplemented with oat spelts xylan, pH 5.0, at 30 °C, under stationary condition for six days. The optimum activity temperature was 60 °C and unusual optimum pH 2.5. The enzyme was stable at 50 and 55 °C, with half-life of 240 and 232 min, respectively. High pH stability was verified from pH 2.0 to 4.0 and 7.5. The β-xylosidase was strongly inhibited by divalent cations, sensitive to denaturing agents SDS, EDTA and activated by thiol-containing reducing agents. The apparent Vmax and Km values was 0.48 μmol PNXP min?1 mg?1 protein and 0.75 mM, respectively. The enzyme was xylose tolerant with a Ki of 28.7. This enzyme presented interesting characteristics for biotechnological process such as animal feed, juice and wine industries.  相似文献

A combined ammonia gas absorption and nitrification was conducted in a single bioscrubber. The reactor was consisted of a bubble column (gas absorption) and a packed bed (nitrification) which contained poly-urethane foams with immobilized nitrifying activated sludge. The entering gas and scrubbing liquid were contacted countercurrently. The bubble column elimination capacity (EC) was 26.74 g NH3/m3 h at >99% ammonia gas removal and effluent gas concentration lower than 2 ppmv. Without ammonium supplement, EC can reach 35.66 g NH3/m3 h which is equivalently the highest tolerable ammonia loading rate of 700 g N/m3 day (1650 mg N/L) at the packed bed. At this level, 593 g N/m3-day ammonia removal rate was achieved via nitrification, dominated by ammonia oxidation. Partial recycling (R/Q = 0.5) of scrubbing solution reduced the secondary wastewater volume by producing 233% more concentrated nitrified products. Hydraulic retention time (HRT) of 24 h was found optimal for both processes (gas absorption and nitrification).  相似文献

Integration of partial nitrification (nitritation) and anaerobic ammonium oxidation (anammox) in constructed wetlands creates a sustainable design for nitrogen removal. Three wetland treatment systems were operated with synthetic wastewater (60 mg NH3–N L?1) in a batch mode of fill – 1-week reaction – drain. Each treatment system had a surface flow wetland (unplanted, planted, and planted plus aerated, respectively) with a rooting substrate of sandy loam and limestone pellets, followed by an unplanted subsurface flow wetland. Meanwhile, three surface flow wetlands with a substrate of sandy loam and pavestone were operated in parallel to the former surface flow wetlands. Influent and effluent were monitored weekly for five cycles. Aeration reduced nitrogen removal due to hindered nitrate reduction. Vegetation maintained pH near neutral and moderate dissolved oxygen, significantly improved ammonia removal by anammox, and had higher TN removal due to coexistence of anammox and denitrification in anaerobic biofilm layers. Nitrite production was at a peak at the residence time of 4–5 d. Relative to pavestone, limestone increased the nitrite mass production peak by 97%. The subsurface flow wetlands removed nitrogen via nitritation and anammox, having an anammox activity of up to 2.4 g N m?3 d?1 over a startup operation of two months.  相似文献

Mesoporous activated carbon (MAC) derived from rice husk is used for the immobilization of acidic lipase (ALIP) produced from Pseudomonas gessardii. The purified acidic lipase had the specific activity and molecular weight of 1473 U/mg and 94 kDa respectively. To determine the optimum conditions for the immobilization of lipase onto MAC, the experiments were carried out by varying the time (10–180 min), pH (2–8), temperature (10–50 °C) and the initial lipase activity (49 × 103, 98 × 103, 147 × 103 and 196 × 103 U/l in acetate buffer). The optimum conditions for immobilization of acidic lipase were found to be: time—120 min; pH 3.5; temperature—30 °C, which resulted in achieving a maximum immobilization of 1834 U/g. The thermal stability of the immobilized lipase was comparatively higher than that in its free form. The free and immobilized enzyme kinetic parameters (Km and Vmax) were found using Michaelis–Menten enzyme kinetics. The Km values for free enzyme and immobilized one were 0.655 and 0.243 mM respectively. The immobilization of acidic lipase onto MAC was confirmed using Fourier Transform-Infrared Spectroscopy, X-ray diffraction analysis and scanning electron microscopy.  相似文献

The performance of up-flow anaerobic sludge blanket (UASB) in combination with down-flow hanging sponge (DHS) system for sewage treatment at an average wastewater temperature of 15 °C has been investigated for 6 months. The results showed that a combined system operated at a total HRT of 10.7 h and total SRT of 88 days represents a cost effective sewage treatment process. The average CODtotal and BOD5 total concentrations measured in the final effluent of the total system (UASB + DHS) amounted to 43 and 3.0 mg/l, respectively, corresponding to the overall removal efficiency of 90% for CODtotal and 98% for BOD5 total. The total process provided a final effluent containing a low concentration of 12 mg/l for TSS.Eighty-six percent of ammonia was eliminated at space loading rate of 1.6 kg COD/m3 d and HRT of 2.7 h. The calculated nitrification rate of the DHS system according to the nitrate and nitrite production amounted to 0.18 kg/m3 d. The removal of F. coliform in the UASB reactor only amounted to 0.86 log10. On the other hand, the F. coliform concentration dropped substantially, i.e. by 2.6 log10 in the DHS system resulting only 2.7 × 103/100 ml in the final effluent.The calculated average sludge production for UASB operated at an HRT of 8.0 h amounted to 30 g TSS/d, corresponding to sludge yield coefficient of 0.2 g TSS/g total COD removed, while it was indeed very low only 6.0 g TSS/d corresponding to sludge yield coefficient of 0.09 g TSS/g total COD removed, for DHS system.The DHS profile results revealed that in the first and second segment of DHS system, the CODtotal, BOD5 total and TSS was eliminated, followed by the oxidation of ammonia in the next segments.  相似文献

This study examined the effect of different elicitors (seven, different concentrations) and environmental factors (water activity (aw), pH) on taxol production by strains of two endophytic fungi, Paraconiothyrium variabile and Epicoccum nigrum, isolated from temperate yew trees. A defined liquid broth medium was modified with elicitors, solute aw depressors at different pH values. For P. variabile, the best elicitor was salicylic acid at 50 mg/l which gave a taxol yield of 14.7 ± 4.8 μg/l. The study of synergistic effects between elicitor, aw and pH on taxol production showed that the highest yield of taxol (68.9 ± 11.9 μg/l) was produced under modified ionic stress of 0.98 aw (KCl) at pH 5 when supplemented with 20 mg/l of salicylic acid. For E. nigrum, serine was the best elicitor which increased yield significantly (29.6 fold) when KCL was used as the aw depressor (0.98 aw) at pH 5.0 with 30 mg/l of serine. The maximum taxol yield produced by E. nigrum was 57.1 ± 11.8 μg/l. Surface response models were used to build contour maps to determine the conditions for maximum and marginal conditions for taxol yield in relation to the best elicitor and aw, and the best pH for the first time. This will be beneficial for identifying key parameters for improvement of taxol yields by endophytic fungi.  相似文献

Nitrifying bacteria are characterized by low biomass yields and slow activity rates, the enhancement of nitrifying biomass retention by promoting the formation of granules would allow improving nitrification process. The use of a pulsing sequencing batch reactor (SBR) was tested to produce nitrifying granules minimizing the air consumption. After 400 days of operation, 1.07 g VSS/L of granular biomass with a mean diameter of 0.9 mm was accumulated in the system. The maximal nitrate production rate reached a value of 0.3 kg N/(m3 d).Sporadic nitrite accumulations were registered and attributed to oxygen diffusion limitations which were solved improving the granule-liquid mass transfer with a liquid recirculation system. FISH technique was used in order to identify the main populations in the nitrifying granules; Nitrosomonas spp. and Nitrospira spp. were identified as the main ammonia and nitrite oxidizing population, respectively. With the introduction of liquid recirculation in the reactor the ammonia oxidation efficiencies improved and the proportion of Nitrospira spp. increased from 1% to 15%.  相似文献

This study investigated the characteristics of nitrous oxide (N2O) emission from intermittently aerated sequencing batch reactors (IASBRs) treating high strength slaughterhouse wastewater at 11 °C, where partial nitrification followed by denitrification (PND) was achieved. N2O generation and emission was examined at three aeration rates of 0.4, 0.6, and 0.8 L air/min in three IASBRs (SBR1, SBR2, and SBR3, respectively). The slaughterhouse wastewater contained chemical oxygen demand (COD) of 6057 ± 172.6 mg/L, total nitrogen (TN) of 576 ± 15.1 mg/L, total phosphorus (TP) of 52 ± 2.7 mg/L and suspended solids (SS) of 1843 ± 280.5 g/L. In the pseudo-steady state, the amount of N2O emission was up to 5.7–11.0% of incoming TN. The aeration rate negatively affected N2O emission and the ratio of N2O emission to incoming TN was reduced by 48.2% when the aeration rate was increased from 0.4 to 0.8 L air/min. Results showed that more N2O was generated in non-aeration periods than in aeration periods. Lower DO concentrations enhanced N2O generation in the aeration periods (probably via nitrifier denitrification) while low DO concentrations (lower than 0.2 mg/L) did not affect N2O generation in the non-aeration periods (probably via heterotrophic denitrification). When PHB was utilized as the organic substrate for denitrification, there was a high N2O generation potential. It was estimated that 1.8 mg N2O-N was generated accompanying per mg PHB consumed.  相似文献

Inhibition of ammonium oxidation and nitrite oxidation by free ammonia (FA) and free nitrous acid (FNA) was studied using three different sludges. An uncompetitive inhibition model fit the experimental data well when the reactions were under FA inhibition, whereas a noncompetitive model fit well under FNA inhibition. The estimates of the inhibition constant (KI) of nitrite oxidation were 46 μM for FA and 1.7–6.8 μM for FNA, each of which was significantly smaller than that of ammonium oxidation, which were 290–1600 μM for FA and 12 μM for FNA. The much smaller values of KI for nitrite oxidation reflected the susceptibility of that reaction to inhibition by FA and FNA, which could lead to accumulation of nitrite during nitrification. A kinetic model for simultaneous inhibition by FA and FNA was derived. The model predicted that nitrite oxidation should be affected more seriously than ammonium oxidation by the simultaneous inhibition, which would accelerate the accumulation of nitrite in a strong nitrogenous wastewater treatment. It also indicated that a complete removal of ammonia could be achieved with high accumulation of nitrite in a sequencing batch reactor, which is impossible in a continuous-flow reactor.  相似文献

This study details on cloning and characterization of Cu,Zn superoxide dismutase (Ca–Cu,Zn SOD) from a medicinally important plant species Curcuma aromatica. Ca–Cu,Zn SOD was 692 bp with an open reading frame of 459 bp. Expression of the gene in Escherichia coli cells followed by purification yielded the enzyme with Km of 0.047 ± 0.008 μM and Vmax of 1250 ± 24 units/mg of protein. The enzyme functioned (i) across a temperature range of −10 to +80 °C with temperature optima at 20 °C; and (ii) at pH range of 6–9 with optimum activity at pH 7.8. Ca–Cu,Zn SOD retained 50% of the maximum activity after autoclaving, and was stable at a wide storage pH ranging from 3 to 10. The enzyme tolerated varying concentrations of denaturating agent, reductants, inhibitors, trypsin, was fairly resistant to inactivation at 80 °C for 180 min (kd, 6.54 ± 0.17 × 10−3 min−1; t1/2, 106.07 ± 2.68 min), and had midpoint of thermal transition (Tm) of 70.45 °C. The results suggested Ca–Cu,Zn SOD to be a kinetically stable protein that could be used for various industrial applications.  相似文献

The enzymatic route for biodiesel production has been noted to be cost ineffective due to the high cost of biocatalysts. Reusing the biocatalyst for successive transesterification cycles is a potential solution to address such cost inefficiency. However, when organic solvent like methanol is used as acyl-acceptor in the reaction, the biocatalyst (lipase) gets severely inactivated due to the inhibitory effect of undissolved methanol in the reaction medium. Thus, organic solvent–tolerant lipase is highly desirable for enzymatic transesterification. In response to such desirability, a lipase (LS133) possessing aforesaid characteristic was extracted from Streptomyces sp. CS133. Relative molecular mass of the purified LS133 was estimated to be 39.8 kDa by SDS-PAGE. Lipase LS133 was stable in pH range 5.0–9.0 and at temperature lower than 50 °C while its optimum lipolytic activity was achieved at pH 7.5 and 40 °C. It showed the highest hydrolytic activity towards long chain p-nitrophenyl palmitate with Km and Vmax values of 0.152 mM and 270.2 mmol min?1 mg?1, respectively. It showed non-position specificity for triolein hydrolysis. The first 15 amino acid residues of its N-terminal sequence, AIPLRQTLNFQAXYQ, were noted to have partial similarity with some of the previously reported microbial lipases. Its catalytic involvement in biodiesel production process was confirmed by performing enzymatic transesterification of vegetable oils with methanol.  相似文献

Bovine liver catalase was covalently immobilized onto Eupergit C. Optimum conditions of immobilization: pH, buffer concentration, temperature, coupling time and initial catalase amount per gram of carrier were determined as 7.5, 1.0 M, 25 °C, 24 h and 4.0 mg/g, respectively. Vmax and Km were determined as 1.4(±0.2) × 105 U/mg protein and 28.6 ± 3.6 mM, respectively, for free catalase, and as 3.7(±0.4) × 103 U/mg protein and 95.9 ± 0.6 mM, respectively, for immobilized catalase. The thermal stability of the immobilized catalase in terms of half-life time (29.1 h) was comparably higher than that of the free catalase (9.0 h) at 40 °C. Comparison of storage stabilities showed that the free catalase completely lost its activity at the end of 11 days both at room temperature and 5 °C. However, immobilized catalase retained 68% of its initial activity when stored at room temperature and 79% of its initial activity when stored at 5 °C at the end of 28 days. The highest reuse number of immobilized catalase was 22 cycles of batch operation when 40 mg of immobilized catalase loaded into the reactor retaining about 50% of its original activity. In the plug flow type reactor, the longest operation time was found as 82 min at a substrate flow rate of 2.3 mL/min when the remaining activity of 40 mg immobilized catalase was about 50% of its original activity. The resulting immobilized catalase onto Eupergit C has good reusability, thermal stability and long-term storage stability.  相似文献
