The brainstem premotor neurons of the facial nucleus (VII) and hypoglossal (XII) nucleus can integrate orofacial nociceptive input from the caudal spinal trigeminal nucleus (Vc) and coordinate orofacial nociceptive reflex (ONR) responses. However, the synaptoarchitectures of the ONR pathways are still unknown. In the current study, we examined the distribution of GABAergic premotor neurons in the brainstem local ONR pathways, their connections with the Vc projections joining the brainstem ONR pathways and the neurochemical properties of these connections. Retrograde tracer fluoro-gold (FG) was injected into the VII or XII, and anterograde tracer biotinylated dextran amine (BDA) was injected into the Vc. Immunofluorescence histochemical labeling for inhibitory/excitatory neurotransmitters combined with BDA/FG tracing showed that GABAergic premotor neurons were mainly distributed bilaterally in the ponto-medullary reticular formation with an ipsilateral dominance. Some GABAergic premotor neurons made close appositions to the BDA-labeled fibers coming from the Vc, and these appostions were mainly distributed in the parvicellular reticular formation (PCRt), dorsal medullary reticular formation (MdD), and supratrigeminal nucleus (Vsup). We further examined the synaptic relationships between the Vc projecting fibers and premotor neurons in the VII or XII under the confocal laser-scanning microscope and electron microscope, and found that the BDA-labeled axonal terminals that made asymmetric synapses on premotor neurons showed vesicular glutamate transporter 2 (VGluT2) like immunoreactivity. These results indicate that the GABAergic premotor neurons receive excitatory neurotransmission from the Vc and may contribute to modulating the generation of the tonic ONR.  相似文献

三叉神经脊束间质核(interstitial nucleus of the spinal trigeminal tract,INV)为位于三叉神经脊束内的一些灰质团块,经三叉神经和舌咽及迷走神经接受口面部的三叉神经躯体传入与上消化道的内脏伤害性传入。INV内含有大量含calbinding D-28k(CB)神经元,但尚不清楚支配口面部的三叉神经躯体传入与支配上消化道的内脏伤害性传入是否汇聚于INV内含CB的神经元。本文应用跨节追踪法并结合CB和Fos免疫组织化学的激光共聚焦显微镜和电镜技术,研究了下牙槽神经(interior alveolarnerve.IAN)的初级传入和上消化道伤害性信息向INV内含CB神经元的汇聚。结果如下:(1)将生物素化的葡聚糖胺(biotinylated dextran amine,BDA)和甲醛分别注入IAN和上消化道后,BDA跨节标记的浓密初级传入纤维和末梢分布于同侧INV内,在其膨大部较为集中;大量的CB和Fos免疫反应阳性神经元分布于双侧INV内,并与BDA注射侧的BDA标记末梢区相重叠:共聚焦显微镜观察显示,约半数CB免疫反应阳性的神经元同时呈Fos阳性的双标记神经元(74/153),其中部分双标神经元与IAN末梢形成紧密接触状。(2)辣根过氧化物酶(horseradish peroxidase,HRP)注射到IAN后,HRP跨节标记的纤维和末梢的分布与BDA标记的分布相似;电镜下观察到,INV内有大量CB免疫反应阳性神经元的树突和少量胞体,以及HRP标记的传入末梢,其中一些HRP标记的轴突终末分别与CB免疫反应阳性树突和胞体形成非对称型轴-树或轴-体突触。结果提示口面部躯体初级传入信息和内脏伤害性信息汇聚于INV内含CB的神经元上,可能在躯体传入信息对内脏伤害性信息的调制和内脏心血管活动中发挥重要作用。  相似文献

应用荧光金(FG)逆行束路追踪结合Fos和calbindin D-28k(CB)免疫荧光组织化学三重标记法,观察了大鼠三叉神经脊束间质核(INV)接受口面部皮肤和上消化道伤害性信息的CB神经元向臂旁核(PB)的投射。结果显示,口周刺激组FG逆标细胞和Fos免疫反应阳性细胞主要分布于注射和刺激同侧INV的背侧边缘旁核(PaMd)和三叉旁核(PaV);大量的CB免疫阳性细胞分布于双侧INV。同侧INV内FG逆标细胞中有77.3%呈CB免疫反应阳性,40.7%呈Fos免疫反应阳性。在FG和CB双标记的神经元中,又有一部分(约38.5%)为FG/CB/Fos三标细胞。上消化道刺激组的FG逆标细胞、CB免疫阳性细胞和FG/CB双标细胞的数量和分布与口周刺激组相似,但Fos免疫阳性细胞分布于双侧的INV。在同侧INV,FG/Fos双标细胞占FG逆标细胞总数的41.9%,FG/CB/Fos三标细胞占FG/CB双标细胞的52.0%。以上结果提示,INV直接投射到PB的CB神经元接受口面部皮肤和上消化道的伤害性信息,CB神经元可能参与经INV中继的外周伤害性信息向PB的传递。  相似文献

目的为了定位向咬肌运动神经元投射的最后一级运动前神经元在脑干内的分布。方法注射麦芽凝集素结合的辣根过氧化物酶(WGA-HRP)至咬肌神经逆行跨突触追踪,然后通过免疫组织化学方法显示了该类神经元。结果这类神经元分布在双侧三叉上核(Vsup)、三叉神经感觉主核背侧部(Vpdm)、小细胞网状结构(PCR)和三叉神经脊束核吻侧亚核背侧部(Vodm),以及对侧三叉神经运动核(Vmo)。数量上,Vsup,特别是注射侧Vsup中,标记的神经元数量最多;其他核团内,双侧标记的神经元的数量无明显差别。结论一侧咬肌运动神经元直接接受脑干双侧多个区域调控。  相似文献

The distribution of GABAergic neurons in the rat trigeminal nuclei was studied using a highly specific monoclonal antibody (mAb3A12) to gamma-aminobutyric acid (GABA). Immunopositive cells were relatively abundant in the marginal and gelatinosa beds of the caudal part of the trigeminal spinal tract nucleus, and in the dorsomedial areas of the oral subnucleus and the principal nucleus. A high density of GABA-immunoreactive somata was also found in the rostral part of the oral subnucleus and in the adjacent parvicellular reticular formation as well as in the supratrigeminal and intertrigeminal regions. Thus, the distribution of the GABAergic cells showed a relatively high density in areas related to the convergence of sensory stimuli, and in zones that contain interneurons inhibiting masticatory motorneurons. 研究用荧光金(FG)逆行追踪与免疫荧光组化染色相结合的双标技术对大鼠脑干向延髓网状背侧亚核(SRD)的5┐羟色胺(5┐HT)能、P物质(SP)能和亮氨酸┐脑啡肽(L┐ENK)能投射进行了观察。将FG注入SRD后,FG逆标神经元主要见于中脑导水管周围灰质、脑干中缝核簇(中缝背核、中缝正中核、中缝桥核、中缝大核、中缝隐核和中缝苍白核)、巨细胞网状核α部、延髓网状结构的内侧部和外侧部、延髓外侧网状核、三叉神经脊束核尾侧亚核和孤束核。5┐羟色胺(5┐HT)样、P物质(SP)样和亮氨酸脑啡肽(L┐ENK)样阳性神经元主要见于中脑导水管周围灰质、脑干中缝核簇和巨细胞网状核α部;此外,SP样和L┐ENK样阳性神经元还见于臂旁核、背外侧被盖核和孤束核。FG逆标并呈5┐HT样、SP样或L┐ENK样阳性的双标神经元也主要见于中脑导水管周围灰质、脑干中缝核簇和巨细胞网状核α部,尤其是位于延髓中缝核团内的双标神经元数量较多。本研究的结果说明SRD内的5┐HT样、SP样和L┐ENK样阳性终末主要来自中脑导水管周围灰质、脑干中缝核簇和巨细胞网状核α部,向SRD发出5┐HT能、SP能和L┐ENK能投射的上述核团对SRD发挥“弥漫性伤害抑  相似文献

实验在6只成年猫上进行,将WGA-HRP微量注入C5膈神经核内,通过逆行追踪及5-HT免疫组织化学FITC荧光双重标记方法,研究了中缝核5-HT能神经元向脊髓膈神经核的投射,同时观察了延髓膈肌产运动神经元接受5-HT能纤维投射的情况,结果在中缝苍白核观察到较多的HRP-5-HT双标记神经元,在中缝大核,中缝隐核观察到少数散在的双标记神经元,在延髓疑核,孤束核腹外侧区域的HRP单核记神经元(即膈肌前运动神经元)周围观察到5-HT能轴突末梢,结构表明:发自中缝苍白核5-HT能神经元的传出纤维可投射到脊髓膈神经核,延髓膈肌前运动神经元接受5-HT能纤维的传入投射。  相似文献

Background

The descending noradrenergic (NAergic) system is one of the important endogenous analgesia systems. It has been suggested that noxious stimuli could activate descending NAergic system; nevertheless, the underlying neuronal circuit remains unclear. As NAergic neurons in the A7 catecholamine cell group (A7) are a part of the descending NAergic system and the lateral parabrachial nucleus (LPB) is an important brainstem structure that relays ascending nociceptive signal, we aimed to test whether LPB neurons have direct synaptic contact with NAergic A7 neurons.

Results

Stereotaxic injections of an anterograde tracer, biotinylated dextran-amine (BDA), were administered to LPB in rats. The BDA-labeled axonal terminals that have physical contacts with tyrosine hydroxylase-positive (presumed noadrenergic) neurons were identified in A7. Consistent with these morphological observations, the excitatory synaptic currents (EPSCs) were readily evoked in NAergic A7 neurons by extracellular stimulation of LPB. The EPSCs evoked by LPB stimulation were blocked by CNQX, a non-NMDA receptor blocker, and AP5, a selective NMDA receptor blocker, showing that LPB-A7 synaptic transmission is glutamatergic. Moreover, the amplitude of LPB-A7 EPSCs was significantly attenuated by DAMGO, a selective μ-opioid receptor agonist, which was associated with an increase in paired-pulse ratio.

Conclusions

Taken together, the above results showed direct synaptic connections between LPB and A7 catecholamine cell group, the function of which is subject to presynaptic modulation by μ-opioid receptors.  相似文献

Background

Rapid eye movement sleep (REMS) is characterized by activation of the cortical and hippocampal electroencephalogram (EEG) and atonia of non-respiratory muscles with superimposed phasic activity or twitching, particularly of cranial muscles such as those of the eye, tongue, face and jaw. While phasic activity is a characteristic feature of REMS, the neural substrates driving this activity remain unresolved. Here we investigated the neural circuits underlying masseter (jaw) phasic activity during REMS. The trigeminal motor nucleus (Mo5), which controls masseter motor function, receives glutamatergic inputs mainly from the parvocellular reticular formation (PCRt), but also from the adjacent paramedian reticular area (PMnR). On the other hand, the Mo5 and PCRt do not receive direct input from the sublaterodorsal (SLD) nucleus, a brainstem region critical for REMS atonia of postural muscles. We hypothesized that the PCRt-PMnR, but not the SLD, regulates masseter phasic activity during REMS.

Methodology/Principal Findings

To test our hypothesis, we measured masseter electromyogram (EMG), neck muscle EMG, electrooculogram (EOG) and EEG in rats with cell-body specific lesions of the SLD, PMnR, and PCRt. Bilateral lesions of the PMnR and rostral PCRt (rPCRt), but not the caudal PCRt or SLD, reduced and eliminated REMS phasic activity of the masseter, respectively. Lesions of the PMnR and rPCRt did not, however, alter the neck EMG or EOG. To determine if rPCRt neurons use glutamate to control masseter phasic movements, we selectively blocked glutamate release by rPCRt neurons using a Cre-lox mouse system. Genetic disruption of glutamate neurotransmission by rPCRt neurons blocked masseter phasic activity during REMS.

Conclusions/Significance

These results indicate that (1) premotor glutamatergic neurons in the medullary rPCRt and PMnR are involved in generating phasic activity in the masseter muscles, but not phasic eye movements, during REMS; and (2) separate brainstem neural circuits control postural and cranial muscle phasic activity during REMS.  相似文献

An anterograde biocytin and a retrograde WGA-colloidal gold study in the rat can provide information about reciprocal communication pathways between the red nucleus and the trigeminal sensory complex. No terminals were found within the trigeminal motor nucleus, in contrast with the facial motor nucleus. A dense terminal field was observed in the parvicellular reticular formation ventrally to the trigeminal motor nucleus. The parvicellular area may be important for the control of jaw movements by rubrotrigeminal inputs. On the other hand, the contralateral rostral parvicellular part of the red nucleus receives terminals from the same zone in the rostral part of the trigeminal sensory complex, where retrogradely labelled neurones were found after tracer injections into the red nucleus. Such relationships could be part of a control loop for somatosensory information from the orofacial area.  相似文献

Cells of origin of the spinal projections from the brainstem of the cat have been studied by means of retrograde axonal transport of horseradish peroxidase (HRP). Following injections of HRP into various levels of the spinal cord, many labeled cells were found in several structures in the brainstem. The labeled cells occurred in the raphe nuclei, reticular formation, vestibular complex, and nuclei of the dorsolateral pontine tegmentum. In the dorsolateral pontine tegmentum, many labeled cells were found in the nuclei of locus coeruleus, subcoeruleus and K?lliker-Fuse. In the coeruleus and subcoeruleus, the greatest number of labeled cells were found, when HRP was injected into the sacral cord. No difference emerged, however, in the number of labeled cells appearing in the K?lliker-Fuse nucleus after injection of the enzyme into different levels of the spinal cord. It appears that neurons in the lateral vestibular nucleus which project to different levels of the spinal cord are located in different parts of this nucleus.  相似文献

The aim of this review was to give a general aspect of the sensorial function of the striatum related to pain modulation, which was intensively studied in our laboratory. We analyse the effect of electrical and chemical stimulation of the striatum on the orofacial pain, especially that produced by tooth pulp stimulation of the lower incisors. We demonstrated specific sites within the nucleus which electrical or chemical stimulation produced inhibition of the nociceptive jaw opening reflex. This analgesic action of the striatum was mediated by activation of its dopamine D2 receptors and transmitted through the indirect pathways of the basal ganglia and the medullary dorsal reticular nucleus (RVM) to the sensorial nuclei of the trigeminal nerve. Its mechanism of action was by inhibition of the nociceptive response of the second order neurons of the nucleus caudalis of the V par.  相似文献

Trigeminothalamic projection neurons are important components of the pathways for conscious perception of pain, temperature, and tactile sensation from the orofacial region. The neurotransmitters utilized by trigeminal neurons projecting to the thalamus are unknown. By use of a monoclonal antibody specific for fixative-modified glutamate and a polyclonal antiserum against glutaminase, we recently identified neurons in the trigeminal sensory complex that cocontain glutamate-like immunoreactivity (Glu-LI) and glutaminase-like immunoreactivity. In the present study, we utilized combined retrograde transport-immunohistochemical techniques to localize putative glutamatergic trigeminothalamic neurons.

Following injection of the retrograde tracer, wheatgerm agglutinin conjugated to horseradish peroxidase (WGA:HRP), into the ventroposterior medial thalamus (VPM), the number of neuronal profiles that were double-labeled with WGA:HRP and Glu-LI was greatest in principal sensory nucleus (Pr5), followed by subnuclei interpolaris (Sp51) and caudalis (Sp5C). The average percentages of projection neurons double-labeled with Glu-LI were approximately 60-70% in Pr5 and Sp51 and 40% in Sp5C. The majority of double-labeled profiles in Sp5C were located in the magnocellular layer, as opposed to the marginal and substantia gelatinosa layers. A large injection site that spread into the intralaminar thalamic nuclei and nucleus submedius—areas implicated in the processing of nociceptive information—resulted in an increase in the ratio of single-labeled to double-labeled projection profiles in Sp5C.

These results suggest that glutamate may be the neurotransmitter for a majority of trigeminothalamic projection neurons located in Sp51 and Pr5. However, on the basis of anatomical association, glutamate does not appear to be the major transmitter for neurons in Sp5C that forward nociceptive information to the thalamus.  相似文献

Ulex europaeus agglutinin I (UEA-I) is a plant lectin with an affinity for L-fucosyl residues in the chains of lactoseries oligosaccharides associated with medium- and smaller-diameter dorsal root ganglion neurons and their axonal processes. These enter Lissauer's tract and terminate within the superficial laminae of the spinal cord overlapping projections known to have a nociceptive function. This implies that the surface coatings of neuronal membranes may have a relationship with functional modalities. The present investigation further examined this concept by studying a neuronal projection with a nociceptive function to determine whether fucosyl-lactoseries residues were incorporated in its primary afferent terminals. Transganglionic transport of horseradish peroxidase (HRP) following injection into tooth pulp chambers was employed to demonstrate dental pulp terminals in the trigeminal spinal complex, while peroxidase and fluorescent tags were used concomitantly to stain for UEA-I. Double immunolabeling for substance P (SP) and gamma-aminobutyric acid (GABA) using peroxidase and colloidal gold allowed a comparison of the distribution of a known excitatory nociceptive transmitter with that of UEA-I binding in specific subnuclei. Synaptic interrelationships between UEA-I positive dental pulp primary afferent inputs and specific inhibitory terminals were also examined. SP immunoreactivity occurred in laminae I and outer lamina II (IIo) of subnucleus caudalis (Vc) and in the ventrolateral and lateral marginal region of the caudal half of subnucleus interpolaris (Vi), including the periobex area in which Vi is slightly overlapped on its lateral aspect by cellular elements of Vc. The adjacent interstitial nucleus (IN) also showed an intense immunoreactivity for this peptide antibody. UEA-I binding displayed a similar distribution pattern in both Vc and Vi, but extended into lamina IIi and the superficial part of Lamina III in Vc. Dental pulp terminals were found to have a comparable distribution; however, many extended into the dorsal portion of the caudal half of Vi and the ventromedial quadrant of rostral Vi. Electron-microscopic analysis showed that transganglionically labeled dental pulp terminals contained ovoid, complex membrane-bound vacuoles laden with transported HRP. The preterminal axon and synaptic membranes of those dental pulp terminals located in zones of Vc and Vi displaying an affinity for UEA-I were usually characterized by a patchy, electron-dense coating of the peroxidase tag. The expression of the immediate early gene, c-fos, was used to determine the distribution of brainstem neurons activated by stimulation of the distal hypoglossal nerve (XIIn) trunk. The traditional view of the XIIn is one of purely motor function; however, stimulation of XIIn excites neurons in the trigeminal spinal nucleus. The rationale for this study was to use c-fos expression as a marker for postsynaptic activity to define the pattern of brainstem neurons excited by XIIn stimulation. It was further hypothesized that if the afferent fibers that course within XIIn supply deep lingual tissues, then c-fos expression after direct stimulation of XIIn should display a pattern similar to that seen after chemical irritant stimulation of the deep tongue muscle. In barbiturate-anesthetized male rats electrical stimulation of XIIn produced a significant increase in Fos-positive neurons in the dorsal paratrigeminal nucleus (dPa5) and laminae I-II of caudal subnucleus caudalis (Vc) and upper cervical dorsal horn. Mustard oil injection into the deep tongue muscle also produced an increase in c-fos expression in dPa5; however, the highest density of expression occurred in laminae I-II at the dorsomedial aspect of rostral Vc. Both electrical stimulation of XIIn and mustard oil stimulation of the deep tongue increased c-fos expression in the caudal ventrolateral medulla, an autonomic relay nucleus. These results suggest that one site of innervation for afferent fibers that travel within the distal trunk of XIIn is to supply the deep tongue muscle and to terminate in the dPa5. A second group of postsynaptic neurons activated only by XIIn stimulation was located in lamina I-II in caudal portions of Vc and upper cervical dorsal horn, a laminar distribution consistent with a role for XIIn afferents in sensory or autonomic aspects of lingual function.  相似文献

To determine the distribution of reticulospinal (RS) neurons in the chicken, WGA-HRP was injected into the cervical or lumbosacral enlargement either unilaterally or bilaterally. The brainstem reticular nuclei sent largely descending fibers to both the spinal enlargements. The mesencephalon (medial and lateral mesencephalic reticular formation) and the rostral pons (nucleus reticularis [n.r.] pontis oralis) project mainly to the cervical enlargement. RS neurons were mainly distributed from the pontomedullary junction to the rostral medulla including n. r. pontis caudalis and pars gigantocellularis, n. r. gigantocellularis, n. r. parvocellularis, n. r. paragigantocellularis, and n. r. subtrigeminalis. It is suggested that the majority of these neurons send axons at least as far as the lumbosacral enlargement. In the lower medulla, RS neurons were distributed in the dorsal and ventral parts of the central nucleus of the medulla.  相似文献

Connections of the neurons of the spinal cord ventral horn with the structures, situating above have been investigated. After injection of uranyl acetate into the TIII segment of the spinal cord, labelled neurons are found in various reticular nuclei of the medulla oblongata. At the level of the roots of the XII pair of the cranial nerves they are revealed in the reticular paramedian, ventral, parvocellular and lateral nuclei. The formations mentioned participate in regulation of the cardio-vascular system. More rostral (2 and 4 mm relatively to the roots of the XII pair of the cranial nerves) the neurons are observed in the reticular giant cellular nucleus, in nuclei of the raphe and in the group of the P-substance reactive neurons. Besides, labelled neurons are revealed in the posterior, lateral fields and in the dorso- and ventromedial nuclei of the hypothalamus.  相似文献

The location of the trigeminal motoneurons of the jaw muscles has been determined in the brainstem of the mallard utilizing retrograde axonal transport of horseradish peroxidase (HRP). Injections with HRP into the jaw muscles or application of HRP to the mandibular nerve showed that the trigeminal motor nucleus can be subdivided into five subnuclei, mV1-mV5. Three functional groups of jaw muscles are represented in separate subnuclei. The most lateral subnucleus mV2 innervates all but one adductor muscles, the intermediate mV1 innervates the pterygoid muscles + one adductor and the medial mV4 the two protractor muscles. The most ventral subnucleus mV3 contains the neurons innervating two extrinsic tongue muscles as well as some perikarya of adductor muscles. Subnucleus mV5 lies dorsomedial to mV4 and contains the motoneurons of the depressor muscle of the lower eye lid. Elements of the proprioceptive system, viz. presumptive gamma-neurons and mesencephalic trigeminal nucleus cells, could also be visualized. The topological and functional aspects of the subdivision of the motor nucleus are discussed.  相似文献
