Lettuce mosaic virus (LMV) Cucumber mosaic virus (CMV) and Tomato spotted wilt virus (TSWV) were identified in fields of Tehran province. In this study 452 leaf samples were collected from the fields throughout the Tehran province during 2002 and 2003. Distribution of Lettuce mosaic virus (LMV), Cucumber mosaic virus (CMV), Tomato spotted wilt virus (TSWV)and Arabis mosaic virus (ArMV) was determined with DAS-ELISA. Percentage of single Infection to LMV. CMV or TSWV was 20.58, 15.93 and 9.96% respectively. Also 15.28% of samples were co- infected with LMV+CMV, 8.19% with LMV+TSMV and 7.74% with CMV+TSWV. 4.65% of samples were Infected to all of these three viruses. LMV was found in 48.69%, CMV in 43.59% and TSWV in 30.54% of samples totally. Therefore LMV is major dominant agent of lettuce mosaic disease in Tehran province. This is the first report of occurrence of TSWV on lettuce in Iran and first report of CMV and LMV in Tehran province.  相似文献

Using enzyme‐linked immunosorbent assays, the frequency of occurrence of six viruses was determined in Brassica nigra collected from five coastal sites in Dorset, spanning approximately 24 km. During 1998–2000, the viruses detected were: Turnip mosaic virus (genus Potyvirus) (TuMV), Turnip yellow mosaic virus (genus Tymovirus) (TYMV), Turnip crinkle virus (genus Carmovirus) (TCV), Turnip rosette virus (genus Sobemovirus) (TRoV), Beet western yellows virus (genus Polerovirus) (BWYV) and Cauliflower mosaic virus (genus Caulimovirus) (CaMV). Multiple infections were detected in some individuals (48/447). TuMV was detected infrequently over the three‐year period (5/597). A representative isolate of each virus was tested for its effects on glasshouse‐grown individuals from different half‐sib families of B. nigra from four of the sites. Whether inoculated manually or via aphids (Myzus persicae), TuMV caused a rapid (within 10 days) lethal systemic necrosis in the B. nigra seedlings except when they were near flowering at the time of inoculation. Each of the other viruses invaded systemically but were not lethal. Indeed, BWYV systemically invaded 13/19 glasshouse‐grown B. nigra seedlings but did not produce any visible symptoms. Otherwise, the isolates tested differed in their pathogenicity and in the symptoms they produced in infected B. nigra. With TYMV or TCV viral antigen concentration was closely linked to pathogenicity; for TRoV or CaMV, there was little or no difference in virus concentration between plants with and without symptoms. Substantial and reproducible differences were observed in sensitivity/susceptibility among B. nigra genotypes from different sites in Dorset challenged with the same virus isolate.  相似文献

Abstract

The presence of latent infections was studied in five cowpeas varieties. Seeds of the varieties were planted and the seedlings inoculated with antigens from Cucumber mosaic virus (CMV) genus Cucumovirus, Bean common mosaic virus (BCMV) genus Potyvirus (Blackeye cowpea mosaic virus strain), Southern bean mosaic virus (SBMV) genus Sobemovirus and Cowpea mottle virus (CPMoV) genus Carmovirus seven days after planting. Seedlings expressing symptoms were rouged at two weeks after inoculation, while asymptomatic ones were subjected to serological indexing to detect the presence/absence of latent infection. Protein A-sandwich enzyme-linked immunosorbent assay (PAS ELISA) was employed for the serological detection of CMV, SBMV and CPMoV, while antigen-coated plate (ACP) ELISA was used to detect BCMV in the asymptomatic plants. Cowpea seedlings without virus symptoms but with positive serological reactions were considered as being latently infected. All of the inoculated TVu 1272 and SuVita-2 plants showed symptoms consistent with CMV and CPMoV infections, respectively. The rate of CMV latent infection was high in TVu 1179 (14.5%), low in SuVita-2 (1.3%) but not recorded in TVu 1272.  相似文献

The response of seven lettuce cultivars to two geographically different Lettuce mosaic virus (LMV) isolates (LMV‐A, LMV‐T) was statistically evaluated based on infection rate, virus accumulation and symptom severity in different time trials. LMV‐A is characterized by the ability to systemically infect cv. Salinas 88 (mo12‐carrying resistant cultivar), and inducing mild mosaic symptoms. Among lettuce cultivars, Varamin (a native cultivar) similar to cv. Salinas showed the most susceptibility to both LMV isolates, whereas another native cultivar, Varesh, was tolerant to the virus with minimal viral accumulation and symptom scores, significantly different from other cultivars at P < 0.05. LMV‐A systemically infects all susceptible lettuce cultivars more rapidly and at a higher rate than LMV‐T. This isolate accumulated in lettuce cultivars at a significantly higher level, determined by semiquantitative ELISA and induced more severe symptoms than LMV‐T isolate at 21 dpi. This is the first evidence for a LMV isolate with ability to systemically infect mo12‐carrying resistant cultivar of lettuce from Iran. In this study, accumulation level of LMV showed statistically meaningful positive correlation with symptom severity on lettuce plants. Based on the results, three evaluated parameters differed considerably by lettuce cultivar and virus isolate.  相似文献

Using enzyme‐linked immunosorbent assays, the frequency of occurrence of six viruses was determined in Brassica rapa ssp. sylvestris collected from two Thameside sites (Abingdon and Culham) in Oxfordshire and one near the Avon (Claverton) in Bath & North East Somerset. During 2000–2001, the viruses detected were: Beet western yellows virus (genus Polerovirus) (BWYV), Cauliflower mosaic virus (genus Caulimovirus) (CaMV), Turnip crinkle virus (genus Carmovirus) (TCV), Turnip rosette virus (genus Sobemovirus) (TRoV), and Turnip yellow mosaic virus (genus Tymovirus) (TYMV). BWYV and TYMV were the most frequently detected viruses at the Oxford shire sites, both as single infections (20/1743 and 66/1743 respectively) and as dual infections (7/1743). Turnip mosaic virus (genus Potyvirus) (TuMV) was not detected in the field‐grown plants assayed from any of the sites. There was a highly significant (x2[1]=30.07, P<0.001) difference in the proportion of plants at each Oxfordshire site in which one or more viruses were detected, and essentially the same pattern of virus infection was observed in tests on B. rapa from the site near Claverton. At least one representative isolate of each detected virus was tested for its morphological and serological effects on glasshouse‐grown individuals from different half‐sib families of B. rapa from both Oxfordshire sites. Except for TRoV, where there was a large difference in the frequency of successful infection in B. rapa from the two locations (1/15 vs 11/15), no clear evidence of resistance or immunity to challenge was observed, although tolerance (virus invasion without symptoms) was frequent. Fewer of the plants from Abingdon were infected than those from Culham, when mechanically challenged with TRoV, but the two B. rapa populations were not otherwise consistently different, either in their infectibility by this virus or in their responses to challenge. However, with TCV, viral antigen concentration was closely linked to the severity of disease and the B. rapa from both Oxfordshire sites segregated into two classes: those with symptoms and most viral antigen, and those without symptoms and least viral antigen. These results suggest that generic risk assessments cannot be made due to differences in the way distinct B. rapa populations react to virus challenge.  相似文献

Lettuce mosaic virus (LMV) is transmitted by aphid vectors in a nonpersistent manner as well as by seeds. The virus causes severe disease outbreaks in commercial lettuce crops in several regions of Spain. The temporal and spatial patterns of spread of LMV were studied in autumn 2002 in the central region of Spain. Symptomatic lettuce (var. Cazorla) plant samples were collected weekly, first at the seedling stage from the greenhouse nursery and later outdoors after transplantation. The exact position of symptomatic plants sampled in the field was recorded and then material was tested by enzyme‐linked immunosorbent assay to assess virus infection. Cumulative spatial data for infected plants at different growth stages were analysed using spatial analysis by distance indices. For temporal analysis, the monomolecular, Gompertz, logistic and exponential models were evaluated for goodness of fit to the entire set of disease progress data obtained. The results indicated that the disease progress curve of LMV epidemics in the selected area is best described by a Gompertz model and that the epidemic follows a polycyclic disease progression. Our data suggest that secondary cycle of spread occurs when noncolonising aphid species land on the primary infected plants (probably coming from infected seed) and move to adjacent plants before leaving the crop. The role of weeds growing close to lettuce fields as potential inoculum sources of virus and the aphid species most likely involved in the transmission of LMV were also identified.  相似文献

Plant virus diseases cause major losses in agricultural and horticultural products, especially in tropical and subtropical regions. The first step to manage these diseases is detecting, identifying and determining the pathogen characteristics. Cucumber mosaic disease is one of the most prevalent and damaging plant diseases in the world which is caused by Cucumber mosaic virus (CMV). Each year, this virus causes yield decreasing and substantial economic damages in its host plants worldwide including the north of Iran. In order to study and identify CMV, 935 leaf samples were collected based on typical symptoms from 10 crops (tomato, pea, tobacco, soybean, watermelon, broad bean, squash, cucumber, eggplant and lettuce) in different regions of Golestan and Mazandaran provinces (north of Iran) during 2009–2010. Suspicious samples were analysed by DAS-ELISA and polyclonal antibodies. The results showed that 275 samples (29.4%) were infected by CMV. Between these hosts, the highest and the lowest CMV infection was associated to watermelon (62.44%) and lettuce (0%), respectively. Among sampling locations, Behshahr (100%) and Minoodasht (3.47%) showed the maximum and minimum infection, respectively.  相似文献

Cowpea plays a key nutritional role in the diet of the Nigerian people. Viral diseases are a major limitation to cowpea production worldwide, and thus, constant viral surveillance is crucial for monitoring and management purposes. In this study, cowpea leaf samples from fields in three northern Nigeria states, Kano, Kaduna and Niger, were tested to determine the status of six common viruses previously reported in these cowpea-producing states following the release of virus-resistant varieties. Cowpea aphid-borne mosaic virus (CABMV), Blackeye cowpea mosaic virus (BICMV), Cowpea mottle virus, Southern bean mosaic virus and Cucumber mosaic virus (CMV) were detected. Cowpea yellow mosaic virus, which was previously reported in all three states, was not detected in any of the samples tested, while CMV that was previously regarded as unimportant to cowpea production in Nigeria had the highest incidence in all three states, and the overall highest incidence of 58.8%, while CABMV had the lowest incidence (7.5%). CMV was also present in seven of the ten mixed infection combinations detected. Dual infection of CMV and BICMV, which often results in cowpea stunt, the most devastating cowpea disease in the USA, was the most frequently detected mixed infection (28.1%) and was detected in all three states. This observed elevation in CMV infection in cowpea must be closely monitored and swiftly managed to avert possible devastating crop yield losses.  相似文献

A field survey was conducted in eight local government areas (LGA) of Ogun state, Nigeria to assess the incidence of viral diseases of yams in the areas. Leaf samples were collected from 90 yam plants which were either symptomatic or asymptomatic. These were bulked into 45 during serological tests and the viruses indexed include yam mosaic virus (YMV); Dioscorea alata bacilliform virus (DaBV) and cucumber mosaic virus (CMV). DaBV was the most prevalent virus on the field with incidence of 48.9% (22/45) followed by YMV which occurred in 42.2% (19/45). CMV had the lowest percentage of incidence; 2.2% (1/45). Of all the LGAs visited, Abeokuta north and Abeokuta south had the highest incidence of YMV and DaBV, respectively. Mixed virus infections were also detected.  相似文献

In this study control of spread by insect vectors of non‐persistent Lily symptomless virus and Lily mottle virus in lily, Tulip breaking virus in tulip, Iris mild mosaic virus, Narcissus latent virus and Iris severe mosaic virus in bulbous iris, and semi‐persistent Dahlia mosaic virus and persistent Tomato spotted wilt virus in dahlia has been evaluated with weekly sprays of mineral oil, beta‐pinene emulsion, polydimethylsiloxane emulsions and pyrethroid insecticide. In lily, beta‐pinene in ‘Wilt Prufgave’ 40% reduction of virus spread. In 1995–97 deltamethrin in ‘Decisgave’ 22–58% reduction. Deltamethrin added to sprays of mineral oil ‘Luxan oil H’ and polydimethylsiloxane (PDMS), e.g. in ‘Dow Corning 36’, efficiently improved control efficacy. The latter was also observed in tulip and dahlia. Mineral oil and deltamethrin gave best control by 81–97% reduction of virus spread at standard spray volumes (6.25 litre ha?1+0.4 litre ha1). ‘Luxan oil H’ at 3.125 litre ha?1 with deltamethrin gave 69–91% control. Efficacy of control by polydimethylsiloxane in ‘Dow Corning 36’ was superior to ‘Luxan anti‐foam’. ‘Dow Corning 36’ with deltamethrin (7+0.4 litre ha?1) gave satisfactory control (68–87%). In tulip, the control by ‘Dow Corning 36’/deltamethrin sprays proved satisfactory compared with ‘Luxan oil H’/‘Decis’‐sprays. In bulbous iris the efficacy of tested PDMS‐brands was clearly different in favour of ‘Dow Corning 36’. In dahlia mineral‐oil and PDMS‐sprays gave some control of semi‐persistent DaMV (16–24%). This ranged at higher level (65–80%) when deltamethrin was added to the spray mixture. Similar trends were observed in the control of persistent TSWV. The effect of polydimethylsiloxane emulsions in the spectrum of virus‐control agents is described for the first time. The effect of PDMS compared with that of mineral oils and synthetic pyrethroid insecticides is discussed with respect to efficacy, mode of action to prevent virus transmission and possible reduction of bulb weights in vegetatively propagated bulb crops.  相似文献

本文报道了高产量提取芜菁花叶病毒(TuMV)、莴苣花叶病毒(LMV)、芋花叶病毒(DMV)和大豆花叶病毒(SMV)的提取方法。本方法通过使用高盐浓度的磷酸盐缓冲液以及在缓冲液中加入氯化镁和脲,并用TritonX-100作为澄清剂,替代常规使用的氯仿和正丁醇,成功的提取到了大量病毒粒子,上述四种病毒提取的得率分别是TuMV为173.3mg/kg病叶,LMV为96mg/kg病叶,SMV为199.2mg/kg病叶,DMV为176.6mg/kg病叶。  相似文献

The 3′‐terminal sequences (c. 1700 nt) of the RNA genome of 10 Turnip mosaic virus (TuMV) isolates from different hosts in Zhejiang province, China, were determined. Phylogenetic analysis of the coat protein nucleotide sequences revealed that most TuMV sequences fell into two distinct clusters. The Chinese isolates B1‐B4 (from Brassica spp.) were similar and placed in the largest group (Group 1), while the isolates R1‐R6 (from Raphanus) were usually placed in a distinct but smaller group (Group 2). There were only approximately 90% identical nucleotides between the two groups. However, one isolate (R5) showed evidence of recombination in that the region between nucleotides 430 and 450, from the start of the coat protein gene and its 3′‐terminus, was a Group 1 type.  相似文献

Fields surveys were conducted to assess the incidence of commonly known legume viruses on cowpeas and weed hosts within and around the cowpeas farms in nine locations across the three agro-ecological zones of Nigeria. Of 315 cowpea leaf samples collected and tested for eight viruses, 69.5% were found to be infected. Bean common mosaic virus-blackeye mosaic (BCMV-BlCM), genus Potyvirus had the highest incidence (70%) and was also the most prevalent (78%). Cowpea aphid-borne mosaic virus (CABMV, genus Potyvirus) had 64% as incidence, incidence of Southern bean mosaic virus (SBMV, genus Sobemovirus) was 21%. Bean pod mosaic virus (BPMV, genus Comovirus) was detected in 1% of the samples tested. Cowpea mosaic virus (CPMV, genus Comovirus) was undetected. Other viruses tested included Cowpea mottle virus (CPMoV, genus Carmovirus), Cucumber mosaic virus (CMV, genus Cucumovirus), and Cowpea mild mottle virus (CPMMV, genus Carlavirus). Multiple virus infections were detected in 68.0% of the infected cowpea leaf samples. The combination of BCMV-BlCM and CABMV was the most common, occurring in 76.4% of all samples. Virus incidence in weeds around the cowpea plots was 2.5% (9 out of 356) whereas 1.5% (5 out of 332) of the weeds collected within the cowpea plots were infected. Some of the weeds infected were Chromoleana odorata, Centrosema sp., Thithonia diversifolia and Talinum triangulare.  相似文献

We report a survey of four viruses (beet western yellows luteovirus (BWYV), cauliflower mosaic caulimovirus (CaMV), turnip mosaic potyvirus (TuMV), turnip yellow mosaic tymovirus (TYMV)) in five natural populations of Brassica oleracea in Dorset (UK). All four viruses were common; 43% of plants were infected with BWYV, 60% with CaMV, 43% with TuMV and 18% with TYMV. For each virus there were significant differences in the proportion of infected plants among populations, which were not completely explained by differences in the age of plants. Multiple virus infections were prevalent, with 54% of plants having two or more virus types. There were statistically significant associations between pairs of viruses. The CaMV was positively associated with the other three viruses, and BWYV was also positively associated with TuMV. There was no detectable association between BWYV and TYMV, whereas TuMV and TYMV were negatively associated. We suggest these associations result from BWYV, CaMV and TuMV having aphid vectors in common, as aphids are attracted to plants that already have a virus infection. Infected plants were distributed randomly or were very weakly aggregated within populations. The implications of widespread multiple virus infections in natural plant populations are discussed with respect to the release of transgenic plants expressing virus-derived genes.  相似文献

Sunflower is one of the most important oilseed crops, which is grown in many countries. Increasing demand to use sunflower oil has expanded the under cultivation area throughout the world. Sunflower viruses have been reported as one of the pathogens that reduce the quantity and the quality of this product in a number of countries. In our research to study the occurrence and distribution of viruses in sunflower fields of Iran, 562 samples were collected from different fields in Kerman and Isfahan provinces during growing seasons from 2009 to 2011. Potato virus Y (PVY), Cucumber mosaic virus (CMV) and Tomato spotted wilt virus (TSWV) were detected by double antibody sandwich enzyme-linked immunosorbent assay. Also, numbers of samples were positive for infection to potyviruses (except PVY) in antigen-coated-plate enzyme-linked immunosorbent assay test using potyviruses general antiserum. Infected plants had symptoms like mosaic, yellowing, deformation, necrotic and chlorotic lesions and mottling on leaves and stunting. The infection rates of potyviruses, PVY, CMV and TSWV in Isfahan province were 33, 22, 4.18 and 3.25% of collected samples, respectively. The corresponding rates for samples from Kerman province were 15, 5, 0.8 and 0.4% of collected samples. According to these results, viral infection in Isfahan province was more than surveyed in Kerman province during the mentioned period of three years. Furthermore, generally there was a decrease in the percentage of viral infection during this three growing seasons in both provinces. This is the first report of the detection of CMV and TSWV in sunflower fields of Isfahan and Kerman and first report of PVY in Isfahan.  相似文献

Different viral diseases infect common bean crops in Iran. A total of 248 symptomatic samples were collected from common bean fields throughout main growing fields of Guilan province in Iran during the summer of 2006. Eight viruses were detected using double antibody-sandwich – enzyme-linked immunosorbent assay (DAS-ELISA). Bean common mosaic virus – BCMV (1%), Bean leaf roll virus – BLRV (9%), Cowpea mild mottle virus – CpMMV (6%), Southern bean mosaic virus – SBMV (3%), Cucumber mosaic virus – CMV (15%), Bean golden mosaic virus – BGMV (2%), Bean common mosaic necrosis virus – BCMNV (1%) and Bean yellow mosaic virus – BYMV (1%) were detected. Comparatively CMV (15%) was found to be more prevalent in Guilan province. Multiple infections of viruses were recorded in many samples. Weed species belonging to Chenopodiaceae, Solanaceae, Malvaceae and Amaranthaceae families were also found to be infected with the viruses.  相似文献
