The extracellular polysaccharides of seven strains of Rhizobium japonicum were investigated by using a gas-chromatographic scheme developed for determination of the various sugars present. These polysaccharides were more heterogeneous in their composition than those of any other species of Rhizobium yet examined. Five strains (1809, 110, 123, 127, and 709) produced polysaccharides containing the same constituents, although in varying relative amounts: glucose (36–44%), galactose (7–25%), mannose (18–20%), 4-O-methylgalactose (5–13%), galacturonic acid (12–16%), and acetyl groups (4–8%). The sugars of the polysaccharide of strain 1809 were all of the d series. These are the first bacterial polysaccharides reported to contain 4-O-methylgalactose and the first Rhizobium polysaccharides in which galacturonic acid has been found. In contrast to this, the polysaccharide of strain 129 consisted of glucose (7%), galactose (51%), mannose (5%), xylose (5%), glucuronic acid (5%), and pyruvic acid (2%). The released polysaccharide from the halophilic cyanobacterium Aphanothece halophytica GR02 was separated into two main fractions byanion-exchange chromatography. The major fraction consisted of glucose,fucose, mannose, arabinose and glucuronic acid. Judging from thechromatography on Sepharose 2B, the major fraction was not furtherfractionated, and its apparent molecular weight was above 2.0 × 106 Da.The minor fraction consisted of rhamnose, mannose, fucose,glucose, galactose and glucuronic acid, with traces of arabinose.Methylation and GC-MS spectrometry analyses of the major fractionrevealed the presence of 1-linked glucose, 1,3-linked glucose, 1,3-linkedfucose, 1,4-linked fucose, 1,3-linked arabinose, 1,2,4-linked mannose,1,3,6-linked mannose, 1-linked glucuronic acid and 1,3-linked glucuronicacid residues. The major fraction was thought to originate from capsularpolysaccharide. The released polysaccharides, obtained from cultures atdifferent age of culture, showed no striking variations in themonosaccharide composition and the relative proportions of themonosaccharides. However, the proportions of galactose and rhamnose inthe released polysaccharides, obtained from cultures under different salinity,were significantly different. The Hindak strain of a Cryptomonas species (Cryptophyceae) produces extracellular polysaccharides. Because there is no information on the structure of these compounds in the Cryptophyceae we conducted structural studies. Gas–liquid chromatographic analyses showed that the polysaccharide is composed of fucose, rhamnose, xylose, mannose, glucose, galactose, galacturonic acid, glucuronic acid, and traces of 3-O-methyl galactose. The polysaccharide was separated into two subtractions by ion-exchange chromatography. Fraction A consisted mainly of 1,3-linked galactose units and 1,4-linked galacturonic acid. Unlike fraction B, fraction A did not have xylose, 3-O-methyl galactose, or glucuronic acid. Also, its degree of branching was low compared to that of fraction B. Only traces of sulfate were present infraction A, but fraction B was 10–15% sulfated. Protein was approximately 1% in both fractions. These polysaccharides appear to be a novel type of polymer in algae.  相似文献

Hot water-soluble polysaccharides were extracted from field colonies and suspension cultures of Nostoc commune Vaucher, Nostoc flagelliforme Berkeley et Curtis, and Nostoc sphaeroides Kützing. Excreted extracellular polymeric substances (EPS) were isolated from the media in which the suspension cultures were grown. The main monosaccharides of the field colony polysaccharides from the three species were glucose, xylose, and galactose, with an approximate ratio of 2:1:1. Mannose was also present, but the levels varied among the species, and arabinose appeared only in N. flagelliforme. The compositions of the cellular polysaccharides and EPS from suspension cultures were more complicated than those of the field samples and varied among the different species. The polysaccharides from the cultures of N. flagelliforme had a relatively simple composition consisting of mannose, galactose, glucose, and glucuronic acid, but no xylose, as was found in the field colony polysaccharides. The polysaccharides from cultures of N. sphaeroides contained glucose (the major component), rhamnose, fucose, xylose, mannose, and galactose. These same sugars were present in the polysaccharides from cultures of N. commune, with xylose as the major component. Combined nitrogen in the media had no qualitative influence on the compositions of the cellular polysaccharides but affected those of the EPS of N. commune and N. flagelliforme. The EPS of N. sphaeroides had a very low total carbohydrate content and thus was not considered to be polysaccharide in nature. The field colony polysaccharides could be separated by anion exchange chromatography into neutral and acidic fractions having similar sugar compositions. Preliminary linkage analysis showed that 1) xylose, glucose, and galactose were 1→4 linked, 2) mannose, galactose, and xylose occurred as terminal residues, and 3) branch points occurred in glucose as 1→3,4 and 1→3,6 linkages and in xylose as a 1→3,4 linkage. The polymer preparations from field colonies had higher kinematic viscosities than those from correspondingsuspension cultures. The high viscosities of the polymers suggested that they might be suitable for industrial uses.  相似文献

TheRhizobium sp. isolated from the root nodules of the leguminous climbing shrubDerris scandens produced a large amount of extracellular polysaccharides in a yeast extract—mannitol medium in the stationary phase of growth. The production was maximum when the medium was supplemented with mannitol (3%), (+)-biotin (3 mg/L) and KNO3 (0.3%). The extracellular polysaccharides contained glucose, galactose and mannose. The possible role of the rhizobial extracellular polysaccharide is discussed.  相似文献

Two extracellular polysaccharides, ENP1 and ENP2, were isolated from the fermentation liquid of the marine fungus Epicoccum nigrum JJY-40 by anion-exchange chromatography and gel-filtration chromatography, and their structures were investigated using chemical and spectroscopic methods including methylation analysis and NMR spectroscopy. The results demonstrated that ENP1 was composed of mannose, glucose, and galactose in the molar ratio of 5.0:2.1:1.0, and the main chain of the polysaccharide consisted of (1?→?2)-linked mannose, (1?→?3)-linked mannose, terminal mannose, (1?→?6)-linked glucose, (1?→?4)-linked glucose, and (1?→?4)-linked galactose. ENP2 was composed of mannose, galactose, glucose, and glucuronic acid in a molar ratio of 12.4:11.2:8.3:1.0, and its glycosidic linkage patterns included terminal mannose, (1?→?6)-linked glucose, (1?→?4)-linked galactose, and (1?→?3)-linked mannose. The two polysaccharides had a partially branched structure with branch point located at C-3 position of (1?→?6)-linked glucose residue. The molecular weights of ENP1 and ENP2 were 19.2 kDa and 32.7 kDa, respectively. Antioxidant properties of the two polysaccharides were evaluated with hydroxyl, superoxide, and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities and lipid peroxidation inhibition in vitro, and results showed that ENP2 and ENP1 had good antioxidant activities, especially ENP2. An organism producing extracellular polysaccharide was isolated from soil and identified as Aeromonas hydrophila (Chester) Stanier. The effects of medium components and cultural conditions on production of the polysaccharide were studied. The optimal concentrations of carbon and nitrogen sources were 5% and 0.3%, respectively, for production of the polysaccharide. The optimal initial pH was 7~9. The maximum polysaccharide yield was obtained at 4~8 days of fermentation. From sucrose and raffinose as carbon source, the organism produced levan and acidic polysac-charide in the ratio of 7:3 and 4:6, respectively. From glucose, galactose, fructose, mannose, maltose and lactose, mainly acidic polysaccharide was produced. The acidic polysaccharide was found to contain galactose, mannose and glucuronic acid in a ratio of 5:4:2. The acidic polysaccharides obtained from sucrose and lactose seemed to be the same polysaccharide.  相似文献

The extracellular polysaccharides (EPS) of six strains of cowpea rhizobia were examined. The strains (MI50A, M6-7B, IRC253) produced polysaccharides containing glucose, galactose and mannose in a molar ratio of 2:1.1:1, 1:1.3:3.1 and 1:1.3:3.5 respectively. Two strains (513-B and Ez-Aesch) produced polysaccharides containing galactose and mannose in a molar ratio of 2:3. Mannose was the only sugar detected in the EPS of strain IRC291. Summary An extracellular polysaccharide producing bacterium Zoogloea sp. was isolated from marine environments. This strain could produce two different polysaccharides. One (water-soluble polysaccharide : WSP) was from cell-free liquid medium, the other (cell-bound polysaccharide : CBP) was obtained from cell surface. Both polysaccharides contained glucose, galactose and mannose as sugar components, but their molar ratios were different (WSP : 2:2:3, CBP : 1:2:2) and half of the sugar components existed as uronic acid form. Both polysaccharide productions started at the early stage of the logarithmic growth phase. The amount of WSP and CBP was influenced by culture conditions such as additional carbon and nitrogen sources. Isolated Zoogloea sp. showed a high product yield without the increase of cell mass.  相似文献

对10株灵芝菌株发酵菌丝体生物量、胞内多糖含量、胞外粗多糖得率及其多糖含量和单糖组成特征进行了系统分析。结果表明,随着培养时间的延长,10株菌株的生物量均有不同程度增加,但不同菌株胞内多糖含量和胞外粗多糖得率变化趋势有所不同。在培养至7d时,G0023和G0160菌株胞外粗多糖得率均较高,分别为3.02g/L和3.14g/L,其多糖含量分别达到了84.11%和91.03%,可作为发酵高产胞外多糖的优良菌株。分级醇沉胞外多糖分析结果表明,各菌株胞外液20%乙醇沉淀所得20E组分的得率和多糖含量均高于50%乙醇沉淀所得的50E组分,说明胞外液中主要以大分子量多糖为主。20E主要由葡萄糖组成,含有少量木糖和甘露糖;50E主要由葡萄糖和甘露糖组成,含有少量木糖和半乳糖。胞外液表观粘度随剪切速率变化曲线分析结果显示,各菌株胞外液均呈现剪切变稀非牛顿流体特性,其表观粘度大小与菌株对应20E组分的得率及多糖含量呈正相关。  相似文献

Summary Two types of polysaccharides were separated from the extracellular polysaccharide produced by Anabaena flos-aquae A-37 by ion exchange chromatography. The neutral polysaccharide is composed of mainly glucose with minor amounts of xylose in a molar ratio of 8:1. Glucose is believed to constitute the polysaccharide core to which xylose is attached. The acidic polysaccharide is composed of glucose and uronic acid as the major monomers with equal amounts of xylose and ribose as the minor constituents. The molar ratio of the monomers found in the acidic polymer is 6:1:1:10 as glucose: xylose: ribose: uronic acid. Chemical analyses showed that the extracellular polysaccharide consists of more neutral polymer (62%) than the acidic polymer (38%).  相似文献

本文探讨了气升环流反应器培养小刺猴头菌过程中菌体生物量、胞内外真菌多糖的积累规律,与同一菌株复元的子实体多糖的含量、组成分别进行分析对比,得到液体深层培养菌丝体多糖的含量为0.0918g/g(干),而子实体仅为外0.378g/g(干),发酵液中多糖含量达0.285g/l,它们的单糖构成比例均相同,即葡萄糖:甘露糖:半乳糖=20.2:5.2:1,但液体深层培养的菌丝体多糖与人工栽培的子实体多糖的电荷性能不同。  相似文献

This paper reports an investigation of the extracellular polysaccharides produced by 26 strains ofRhizobium andAgrobacterium. Strains ofRhizobium leguminosarum andR. phaseoli produced a water-soluble polysaccharide containing glucose, glucuronic acid and 4-0-methylglucuronic acid. These substances were also identified in the polysaccharide of a single strain fromLotus uliginosus. Glucose was the only detectable component in the polysaccharide produced by strains ofAgrobacterium radiobacter andA. tumefaciens. The polysaccharides obtained from slow-growing rhizobia were not freely water-soluble. Glucose, mannose, rhamnose, galactose and 4-0-methylglucuronic acid were identified as components of this extracellular material.These results are related to previous studies on rhizobial taxonomy and to the infection process in legumes.  相似文献

A bacterium that was isolated from soil and identified as Bacillus circulans was found to produce a highly viscous extracellular polysaccharide when it was grown aerobically in a medium containing glucose as a sole source of carbon. The product was characterized by TLC and GC analyses as a novel heteropolysaccharide consisted of rhamnose, mannose, galactose, and mannuronic acid as sugar components. A maximal yield of polysaccharide reached about 2 g/liter by jar-fermentor culture at 30°C for 48 hr with a medium containing 1% glucose, 0.05% asparagine, 0.005% yeast extract, and small amounts of inorganic salts. Some culture conditions for the production of polysaccharide were investigated with flask culture; an optimal production was attained with a medium containing 0.1–1 % glucose and 0.01–0.05% asparagine, pH 7–8, at 30°C under aerobic conditions.  相似文献

The structure of the extracellular polysaccharide gum from nitrogen-fixing Rhizobium sp. strain CB744 (a member of the slow-growing Cowpea group) has been investigated. Gas-chromatographic analysis of the alditol acetates of the acid hydrolysate showed the gum to be composed of galactose, 4-O-methylgalactose, mannose, and glucose in the molar ratio of 1:2.5:3.5:7.0. The polysaccharide is unusual in that it contains no carbonyl substituent, although such substituents are common amongst polysaccharides produced by the slow-growing group. The native and de-branched polysaccharides were examined by methylation analysis. The anomeric configurations were determined by 13C-n.m.r. and oxidation by chromium trioxide. It is concluded that there are two β-(1→4)-linked glycopyranosyl residues for each α-(1→4)-linked mannopyranosyl residue, and that each mannose is substituted at O-6 by a β-galactopyranosyl residue, with 71% of the galactose groups being present as 4-O-methylgalactose.  相似文献

Analysis showed that the polysaccharide consisted of glucose, galactose, and mannose in the ratio of 60:14:20. Chemical analysis of grapefruit (Citrus paradisi) pectic polysaccharides demonstrated that galacturonic acid constitutes 78% by weight of the total carbohydrates found. The remaining 22% was accounted for by a number of sugars which include galactose, glucose, arabinose, xylose, and mannose and, by weight, galactose accounted for almost 50% of the total neutral sugar components found in these pectic polysaccharides. Treatment of pectic polysaccharides with galactose oxidase followed by reduction of oxidized galactose residues with tritiated potassium borohydride resulted in the labeling of pectic polysaccharides. Analysis of the labeled polysaccharides demonstrated that of the total radioactivity incorporated more than 90% was recovered in the galactose residues. These results clearly demonstrate the successful utilization of the galactose oxidase/tritiated potassium borohydride method in labeling plant pectic polysaccharide.  相似文献

Summary The effects of components of the medium on the production of extracellular polysaccharide (EPS) by cultured cells of Polianthes tuberosa (tuberose) were studied. Optimization of media components culturing in flask resulted in increasing EPS production from 1.4 to 4.1 g/l. In particular, relatively high concentration (10\s-5M) of 2,4-dichlorophenoxyacetic acid (2,4-D) markedly stimulated the production of EPS. Based on these results, EPS production by a 30-1 jar fermenter was attempted and the final rate of Production was 4.6 g/l at 30th day of culture. The EPS consisted mainly of acidic polysaccharides with glucuronic acid, mannose, arabinose, galactose, glucose and xylose.  相似文献

Extracellular polymeric substances (EPS) play an important role in the formation and activity of biofilms in wastewater treatment (WWT). The EPS of the denitrifying biomarker Comamonas denitrificans strain 110, produced in different culture media and growth modes, were characterized. The EPS mainly contained protein (3–37%), nucleic acids (9–50%), and carbohydrates (3–21%). The extracellular DNA was found to be important for initial biofilm formation since biofilm, but not planktonic growth, was inhibited in the presence of DNase. The polysaccharide fraction appeared to consist of at least two distinct polymers, one branched fraction (A) made up of glucose and mannose with a molecular weight around 100 kDa. The other fraction (B) was larger and consisted of ribose, mannose, glucose, rhamnose, arabinose, galactose, and N-acetylglucosamine. Fraction B polysaccharides were mainly found in capsular EPS which was the dominant type in biofilms and agar-grown colonies. Fraction A was abundant in the released EPS, the dominant type in planktonic cultures. Biofilm and agar-grown EPS displayed similar overall properties while planktonic EPS showed clear compositional disparity. This study presents results on the physiology of a key WWT organism, which may be useful in the future development of improved biofilm techniques for WWT purposes.  相似文献

A suspension culture of Mentha was established from callus which formed on the tips of young shoots of a Mentha hybrid (M. arvenis × M. spicata). Changes in growth parameters during a culture cycle were recorded. The general appearance of cells during division and growth, including the changes in cell form, was also represented.Suspension-cultured cells of Mentha hybrid released a large amount of extracellular polysaccharides (ECP) mainly at the logarithmic phase of the growth cycle. The ECP contained galacturonic acid as major components and arabinose, galactose, glucose, xylose, rhamnose and mannose as minor components. The ratio of the uronic acid content to total sugar content in the ECP was below 40% at day 7, but increased up to 90% at day 21. The relative contents of xylose and glucose in the ECP decreased during the culture period, while the arabinose content increased and those of rhamnose, mannose and galactose remained constant.The IR spectrum suggested that the ECP were low-methoxylated pectic polysaccharides. The presence of lignin and related compounds in the ECP was not detected. The protein content of the ECP was about 10% and the main amino acids were alanine, proline, hydroxyproline, valine, asparticacid and serine, in that order.  相似文献
