Endothelin-3 (ET-3) elicited a concentration-dependent positive inotropic effect on rabbit papillary muscle, the maximal response being approximately 65% of the maximal response to isoproterenol. ET-1 induced a positive inotropic effect over the concentration range below 10–9 M, at which ET-3 did not produce a positive inotropic effect, but the maximal response to ET-1 was equivalent to or slightly lower than that of ET-3. The nonselective ET receptor antagonist PD 145065 effectively antagonized the positive inotropic effect of ET-3 in a concentration-dependent manner and abolished it at 10–5 M. PD 145065 decreased the positive inotropic effect induced by ET 1 at lower concentrations (< 10–9 M) but it did not affect the main portion of the concentration-response curve for the positive inotropic effect, i.e., the effect induced by high concentrations (> 10–9 M) of ET-1. PD 145065 antagonized also the positive inotropic effect of sarafotoxin S6c. PD 145065 inhibited the specific binding of [125I]ET-1 and of [125I]ET-3 with a high- and a low-affinity site for competition. ETB selective ligands, RES-701-1 and sarafotoxin S6c, displaced [125Iuc]ET-3 with high affinity but they scarcely affected the [1251]ET-1 binding. These findings indicate that different subtypes of the ET receptor are responsible for the induction of the positive inotropic effect of ET-3 and ET-1. ET receptors involved in the production of the positive inotropic effect in the rabbit ventricular myocardium have pharmacological characteristics that are different from those of conventional ET receptors originally classified based on the pharmacological findings in noncardiac tissues. The positive inotropic effect of ET-3 in the rabbit ventricular muscle may be mediated predominantly by ETA1 receptors that are susceptible to PD 145065 as well as BQ-123 and FR139317, and partially mediated by ETB receptors that are inhibitable with RES-701-1. ETA2 receptors that are resistant to ETA selective as well as nonselective antagonists may mainly be responsible for the positive inotropic effect of ET-1 in the rabbit ventricular muscle.  相似文献

The cell-growth-inhibitory and phase-specific effects of D-penicillamine on cell-cycle progression were investigated using cell-proliferation patterns, quantitative cell-cycle analysis by flow cytometry, and determination of the mitotic index and binucleate cell fraction of normal (rabbit articular chondrocytes, L 809, rabbit fibroblasts) and transformed (HeLa, L 929) cells. D-penicillamine treatment resulted in an inhibition of growth within a dose range of 5 × 10?4 M to 7.5 × 10?3 M. Examination of DNA by flow cytometric analysis revealed that rabbit articular chondrocytes were preferentially arrested in the G0/1 phase of the cell cycle, whereas the other cell lines were blocked in the G2 + M phase; the increase in the proportion of cells with G2 + M DNA content was partially due to an enhancement of binucleate cells, resulting in a cytokinesis perturbation for HeLa and L 929 cells. These results showed that D-penicillamine affects cell proliferation through different events according to cell type.  相似文献

Cerebral microvessels isolated from cat forebrain contain a specific β-adrenergic-sensitive adenylate cyclase. Among various compounds tested, the most potent activator of enzyme activity is isoproterenol (ka = 1.4 × 10?7M), followed in order by epinephrine (ka= 1.5 × 10?6M), norepinephrine (ka= 1.4 × 10?5M) and phenylephrine (ka> 3 × 10?4M). Isoproterenol-stimulated enzyme activity is blocked by propranolol (ki= 2.4 × 10?9M, IPS 339 (ki= 4 × 10?9M), H35/25 (ki = 1.2 × 10?7M), atenolol (ki= 5.9 × 10?6M) and practolol (ki= 1.8 × 10?5M). These agonist and antagonist properties are quite similar to those demonstrated by β2-adrenergic receptors and β2-stimulated adenylate cyclase present in other tissues and indicate that the majority of adenylate cyclase-associated adrenergic receptors in cerebral microvessels are β2. The findings are relevant to physiological studies of cerebral blood flow and vascular permeability.  相似文献

Abstract: We identified and characterized 125I-endothelin-1 (125I-ET-1) binding sites in tumor capillaries isolated from human glioblastomas, using the quantitative receptor autoradiographic technique with pellet sections. Quantification was done using the computerized radioluminographic imaging plate system. High-affinity ET receptors were localized in capillaries from glioblastomas and the surrounding brain tissues (KD = 4.7 ± 1.0 × 10?10 and 1.6 ± 0.3 × 10?10M, respectively; Bmax = 161 ± 38 and 140 ± 37 fmol/mg, respectively; mean ± SEM, n = 5). BQ-123, a selective antagonist for the ETA receptor, potently competed for 125I-ET-1 binding to sections of the microvessels with IC50 values of 5.1 ± 0.3 and 5.1 ± 1.5 nM, and 10?6M BQ-123 displaced 84 and 58% of ET binding to capillaries from tumors and brains, respectively. In addition, competition curves obtained in the presence of increasing concentrations of ET-3 showed two components (IC50 = 5.7 ± 2.5 × 10?10 and 1.4 ± 0.2 × 10?6M for tumor microvessels, 1.8 ± 0.6 × 10?10 and 1.1 ± 0.3 × 10?6M for brain microvessels, respectively). Our results indicate that (a) the method we used is simple and highly sensitive for detecting and characterizing various receptors in tumor capillaries, especially in the case of a sparse specimen, and (b) capillaries in glioblastomas express specific high-affinity ET binding sites, candidates for biologically active ET receptors, which predominantly belong to the ETA subtype.  相似文献

Membrane receptors for Vicia graminea (Vg) lectin on human red cells were analyzed using deoxycholate lysates obtained from 125I-erythrocyte membranes incubated with a purified lectin immobilized on Sepharose 4B. The glycoproteins (GP) specifically bound to the gel were eluted and analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and autoradiography. Using native erythrocytes the results obtained demonstrate that N red cells have exposed Vg receptors located on GPα (synonym glycophorin A) and GPδ (synonym glycophorin B) whereas on M erythrocytes the Vg receptors are restricted to GPδ. The presence of Vg receptors was also found on the hybrid glycoprotein (made of the N-ter of GPδ and C-ter of GPα) carried by St(a+) erythrocytes. A similar amount of radioactivity was bound to Vg-Sepharose incubated with neuraminidase-treated N or M membranes. The material eluted was tentatively identified as asialo GPα and asialo GPδ, suggesting that numerous receptors have been uncovered mainly on asialo GPα species from M erythrocytes. No glycoprotein component could be identified from the material eluted from Vg Sepharose incubated with native or neuraminidase-treated membrane from a Tn(+) individual. Scatchard plot analysis obtained from binding experiments at equilibrium with M, N, and St(a+) cells revealed the existence of at least two classes of receptors both on native and neuraminidase-treated erythrocytes. Desialylation of the M, N, and St(a+) erythrocytes resulted in an increase in the number of low- and high-affinity binding sites but had no significant effect on the association constants. However, high-affinity binding constants were about six times higher with N (7.07 × 107 and 6.61 × 107m?1 for native and neuraminidase-treated N cells, respectively) as compared to M erythrocytes (1.13 × 107 and 1.17 × 107m?1 for native and neuraminidase-treated M cells, respectively) whereas the low-affinity binding constants were similar for all types of cells (in the range of 0.1 to 0.3 × 107m?1). The number of Vg binding sites increases from 0.085 × 105 to 0.8 × 105 (high affinity) and from 2.10 × 105 to 6.25 × 105 (low affinity) per native and neuraminidase-treated N cell, respectively. On native and neuraminidase-treated M cells the number of Vg receptors increases from 0.011 × 105 to 0.51 × 105 (high affinity) and 0.13 × 105 (low affinity), respectively. The large increase in the number of Vg receptors on neuraminidase-treated M cells is correlated with a large increase in agglutinability. Under similar treatment St(a+) cells behave like N erythrocytes whereas only 0.16 × 105 Vg receptors of low affinity could be detected on neuraminidase-treated Tn erythrocytes. The results demonstrate that sialic acid is not required for binding and favor the view that the binding site of V. graminea lectin accommodates with two types of erythrocyte membrane receptors, one including both a contribution of polypeptide and oligosaccharide chains and a second which involves a simple interaction with sugar sequence Galβ1–3GalNAc available only when sialic acids are removed. The latter disaccharide is recognized by the Arachis hypogea lectin which therefore inhibits further binding of the V. graminea to neuraminidase-treated erythrocytes.  相似文献

Rat striatal slices were continuously superfused with 3H-tyrosine to study the effect of a low concentration of nicotine on the spontaneous release of newly synthesized 3H-dopamine (3H-DA). Nicotine (10?6M) stimulated the calcium-dependent spontaneous release of 3H-DA. This effect was prevented by nicotinic blockers such as pempidine (10?5M) and d-tubocurarine (5×10?6M). The stimulatory effect of nicotine and its blockade by pempidine were still observed in the presence of tetrodotoxin (5×10?7M). This demonstrates for the first time that a low concentration of nicotine is effective in releasing DA by acting on presynaptic nicotinic receptors located on terminals of the nigro-striatal dopaminergic neurons.  相似文献

Picrotoxin does not by itself affect [3H] diazepam binding to synaptosomal membranes of rat cerebellum; however, picrotoxin stimulated the binding in the presence of Cl? ion or Cl? ion plus low concentrations of GABA. On the other hand, in the presence of GABA at concentrations higher than 1 × 10?6 M, picrotoxin inhibited [3H]diazepam binding. This inhibition seems to be the result of reduced GABA binding, which occurred in the presence of picrotoxin and Cl? ion. These results may indicate that benzodiazepine receptors, GABA receptors, and the Cl? ionophore are closely associated with each other.  相似文献

The formaldehyde method was used to examine the interaction of PGE1 with morphine, β-endorphin and Met-enkephalin on rat mast cells by their effects on IgE-mediated 14C-serotonin release. PGE1 (2×10?8?2×10?5 M) caused a dose-related inhibition of the mediator release 1 min after an antigen challenge, and morphine (3×10?7?3×10?5 M) reversed this PGE1 effect dose-dependently and stereospecifically; naloxone (2×10?4 M) antagonized this action of morphine. β-Endorphin (3×10?7?10?5 M) and Met-enkephalin (3×10?6?10?4 M) mimicked this morphine action dose-dependently and were antagonized by naloxone (2×10?4 M). These results suggest that morphine and endorphins modulate immunological mediator release from rat mast cells through opioid receptors.  相似文献

Papaverine inhibited the basal renin secretion of rat kidney slices incubated in a physiological salt solution at 37°C. Inhibition was concentration-dependent; secretion was 99 ± 0.2 % inhibited by 5 × 10?4 M papaverine, and 8 × 10?5 M was the estimated ED50. In contrast, 2 × 10?4 M IBMx (3-isobutyl-1-methyl-xanthine) increased the rate of secretion from 215 ± 17 to 366 ± 30 ng hr?1mg?1/20 min (p < 0.001). Isoprotenol (4 × 10?7, 8 × 10?7, and 5 × 10?6 M) stimulated renin secretion in a concentration-dependent manner; the stimulatory effects were antagonized by papaverine but unaffected by IBMx. Thus, two known inhibitors of phosphodiesterase--IBMx and papaverine--produce sharply contrasting effects on basal and on isoproterenol-stimulated renin secretion from rat kidney slices.  相似文献

Epidermal growth factor (EG factor) and insulin stimulate the incorporation of thymidine into contact-inhibited rabbit lens epithelial cells in culture. The maximal stimulation observed with EG factor is greater than with insulin. Half-maximal stimulation by EG factor is observed at 6 × 10?10m and for insulin at 1 × 10?9m. [125I]-labeled EG factor (2000 Ci/mmol, about 1 g atom of iodine per mol) is equipotent with native EG factor in stimulating DNA synthesis. Both insulin and EG factor bind to distinct high-affinity sites in intact lens cell monolayers; half-maximal binding is observed at about 10?9m for both polypeptides. A maximum of approximately 8 × 104 insulin molecules and 4 × 104 EG factor molecules are bound per cell. These observations indicate that cultured rabbit lens cells possess receptors for insulin and EG factor by biological and physicochemical criteria and raise the possibility that both peptides may play a role in lens growth and development.  相似文献

On isolated rabbit mesenteric arteries pretreated with phenoxybenzamine (10-5M) and contracted with prostaglandin F (PGF) dopamine (10?6M to 3×10?4M) and isoprenaline (10-9M to 10-5M) caused a dose-related relaxation. Pindolol (10?7M) significantly suppressed the effects evoked by isoprenaline, but did not affect those produced by dopamine. The dopamine receptor antagonist metoclopramide (5×10?5 and 10?4M), however, shifted the dose-response curve for dopamine-induced relaxation significantly to the right in a concentration dependent manner without affecting relaxations caused by isoprenaline or papaverine. These results demonstrate for the first time a specific antagonism to dopamine-induced relaxation on rabbit mesenteric arteries in vitro. They support the hypothesis of the existence of specific dopamine receptors in vascular smooth muscles.  相似文献

Results from this study indicate that adult rat brain posesses guanylate cyclase activity sensitive to serotonin (5-HT) and localized in the synaptic plasma membrane. The enzyme appears to have multiple activation sites for 5-HT with specific activity maxima at the 5-HT concentrations of 5 × 10?10M and 7 × 10?8M respectively. The rates of guanosine-3′:5′-monophosphate (cyclic GMP) formation at these concentrations of 5-HT are, respectively, 170% and 307% above the endogenous or basal production rate of 2.7±0.3picomoles/minute/milligram of synaptosomal membrane protein. We have also been able to identify four distinct types (Type #1, #2, #3, and #4) of high affinity, specific binding sites for 5-HT on isolated synaptosomal membranes from rat brain. Dissociation constants of 2.6 × 10?10M, 2.5 × 10?9M, 7.0 × 10?9M, and 4.6 × 10?8M, characterize the binding of 5-HT to our sites of Type #1 through Type #4 respectively. The specific, high affinity binding was saturated at 5-HT concentrations of 5 × 10?10M for the Type #1 sites, 5 × 10?9M for our Type #2 sites, 1 × 10?8M for our Type #3 sites, and 7 × 10?8M for our Type #4 sites. The 5-HT concentrations producing saturation of our specific binding sites of Type #1 and Type #4 are virtually identical to those that elicit the two maxima of 5-HT stimulated cyclic GMP production, indicating that a membrane-bound guanylase cyclase may be closely associated with certain 5-HT receptors and/or re-uptake sites.  相似文献

A high affinity (Kd=2.7 × 10?10M at 0°) dexamethasone binding macro-molecule has been identified in the cytosol fraction of rabbit uteri. Competition studies show high specificity for glucocorticoids since binding of labeled dexamethasone is inhibited by cortisol and corticosterone but not by progesterone, testosterone, or estradiol 17β. The binding component has a sedimentation coefficient of 8S and its concentration in uterine cytosol is about 0.2 pmoles per mg protein. Uptake of labeled dexamethasone by isolated uterine nuclei requires the presence of cytosol and is temperature dependent. The KCl-extractable nuclear complex sediments at 4S. Thus the dexamethasone binding components of the rabbit uterus have properties similar to those described for steroid hormone receptors present in target tissues. Specific dexamethasone binding could not be demonstrated in rat uterine cytosol.  相似文献

The true KDs of [3H] (?) QNB binding to muscarinic receptors were found to be 4.13 and 6.43 × 10?11 M in pancreas of 21 day fetal and adult rats. The competition curves of specific [3H] (?) QNB bound by two antagonists have shown that the affinity of these drugs did not change with age with Hill coefficients near unity. However, with the agonist carbamylcholine as the competitive drug, a more flat curve was obtained with a Hill coefficient below unity. At least two populations of carbamylcholine binding sites were found with different KDs: a KH around 7 × 10?7 M and a KL around 3 × 10?5 M. These two populations were present during all the developmental periods studied. The ED50 of bethanechol stimulated amylase secretion did not change within the age limit studied (from 11 to 365 days). The high affinity sites for carbamylcholine would seem to be the receptor implicated in the physiological response of the pancreas.  相似文献

Membranes from homogenates of corpus striatum bound 3H-dihydromorphine in a saturable fashion with a Km value of 1 × 10?9M. The binding of 3H-dihydromorphine to the membranes was reduced to about 10% by 10?7M levorphanol but not by 10?7M dextrorphan. The binding of 3H-dihydromorphine became less sensitive to 10?7M levorphanol when the concentration of 3H-dihydromorphine was greater than 2 × 10?9M. Other opiate narcotics, e.g. morphine and l-methadone, were as effective as levorphanol in competition for the binding 3H-dihydromorphine with ED50 values of 2–4 × 10?9M. d-Methadone and dextrorphan were about 1/50 and 1/2000 as effective as their respective levo-isomers. The opiate antagonist, naloxone, also competed effectively for the binding sites with an ED50 value of 3.3 × 10?9M. Substances like acetylcholine, choline, serotonin, norepinephrine and dopamine were ineffective. Only ionophores specific for divalent cations stimulated the binding of 3H-dihydromorphine suggesting that some endogenous divalent cations may be inhibitory to the binding of the opiate narcotic. The receptors of 3H-dihydromorphine probably exist in the membranes of nerve endings and have a density of 6 × 1012 sites per g in corpus striatum. We conclude that the described technique can successfully detect the opiate narcotic receptors in the central nervous system without the usual method of displacement.  相似文献

Cytoplasmic receptors for 1α, 25-dihydroxyvitamin D3 from human parathyroid adenoma tissue and rachitic chick parathyroid glands have been characterized with regard to a number of physical, chemical, and ligand binding properties. Both receptors are 3.6–3.7 S proteins with molecular weights of approximately 75,000 and Stoke's molecular radii of 36 Å. It was found that the receptors possess a cysteine residue in or near the 1α, 25-dihydroxyvitamin D3 binding site which is critical for ligand binding activity. The receptors both have equilibrium dissociation constants for 1α, 25-dihydroxyvitamin D3 in the range of 2 to 5 × 10?10m at 4 °C and second-order association rate constants for their seco-steroid ligand of 1 × 107, m?1 min?1 (0 °C). The dissociation rate constants were found to be 5.3 × 10?4 min?1 (4 °C) for the human receptor and 1.3 × 10?5 min?1 (4 °C) for the chick receptor. The great deal of similarity which exists between the cytoplasmic 1α, 25-dihydroxyvitamin D3 receptors from avian and mammalian parathyroid glands suggests a homologous function for these molecules in the two tissues.  相似文献

Soil application of CCC reduced stem and leaf growth in Begonia plants. This effect was evident with all concentrations tested at 18°C, whereas at 21 and 24°C no growth–retarding effect was observed with 2 × 10?2 M CCC, and with 5 × 10?3 M growth was even stimulated. Flowering was promoted by CCC in long day and neur–critical temperature, particularly under low light intensity in the winter. The formation of adventitious buds in leaves of plants grown at 21 and 24°C was stimulated when the plants received 5 × 10?2 and 2 × 10?2 M CCC, while 8 7times; 10?2 M was inhibitory. In plants grown at 18°C bud formation was inhibited by all CCC concentrations. Root formation in the the leaves was usually stimulated by high CCC concentrations, while root elongation was reduced. The level of ether–extractable. acidic auxin (presumably IAA) in the leaves was lowered by CCC treatment of the plants, hut this required higher CCC concentrations at higt than at low temperature. When applied to detached leaves CCC stimulated bud formation at concentrations ranging from 10?4 to 10?2 M in leaves planted at 18 and 21°C. At 24°C budding was inhibited by 10?2 M CCC, the lower concentrations being stimulatory also at this temperature. Root formation and growth were not much affected by CCC treatment of the leaves, but increased with the temperature. Soil application of Phosfon (4 × 10?4 M) had no effect on growth and flowering, nov did it affect the subsequent regeneration of buds and roots in the leaves. In detached leaves Phosfon stimulated bud formation with au optimum at 10?6 M. Root formation was stimulated by Phosfon at all temperatures, the optimal concentration being 10?5 M, whereas root length was conversely affected. Foliar application of B-995 to intact plants and treatment of detached leaves greatly inhibited the formation of buds and had little effect on root formation. The pattern of morphologic and functional development of lung during intrauterine period is influenced by several endogenous compounds. Recently Epidermal Growth Factor (EGF), when administered in vivo, has been shown to accelerate pulmonary maturation in fetal rabbit and sheep. We sought evidence for EGF receptor occurrence in fetal and maternal rabbit lung plasma membranes. The percent specific binding (mean ± S.E.M.) (125-I) EGF to LPM in the mother (n=5) and the fetus at term (n=7) was 1.08 ± 0.08 and 2.25 ± 0.12 per 175 μg of LPM protein respectively. The number of receptor sites per mg of LPM protein in the mother were significantly less than that in the fetus (44 ± 11 and 250 ± 24 × 10?10, p < 0.001) with no apparent differences in Kd (2.10 ± 0.39 and 2.47 ± 0.24 × 109). Presence of high affinity receptors for EGF in fetal and maternal lung plasma membranes suggests a direct role of EGF in fetal lung maturation.  相似文献

Rainbow trout ovarian follicles at four different stages of maturation (very early vitellogenesis, early vitellogenesis, peak vitellogenesis, and pre-ovulatory) were incubated either in Cortland's medium alone, or medium containing melatonin at one of five concentrations (1 × 10-10, 1 × 10-8, 1 × 10-6, 1 × 10-4 or 1 × 10-2M) to assess the direct actions of melatonin on basal (non-gonadotrophin stimulated) secretion of 17β-estradiol and testosterone. Testosterone secretion by peak vitellogenic phase follicles was significantly (p &lt; 0.01) reduced by melatonin at 1 × 10-4 or 1 × 10-2 M, but there were no other significant effects of melatonin on T secretion. There were no significant effects of melatonin at concentrations of 1 × 10-10 or 1 × 10-8 M on 17β-estradiol secretion for any stage of follicular maturation, although for follicles at the peak vitellogenesis stage, melatonin at 1 × 10-6 M had a significant (p &lt; 0.05) stimulatory effect compared with the controls. Melatonin also stimulated 17β-estradiol secretion by very early vitellogenic stage follicles (1 × 10-2 M concentration; p &lt; 0.05). At concentrations of 1 × 10-4 and 1 × 10-2 M, melatonin significantly (p &lt; 0.01) inhibited 17β-estradiol secretion of peak vitellogenic and preovulatory stage follicles. The findings suggest a stimulatory action of melatonin on steroidogenesis of early stage ovarian follicles, a shift to an inhibitory action of the higher concentrations of melatonin in later stages of development, but with a bimodal action of melatonin (related to hormone concentration) being present during the time of maximum follicular growth.  相似文献

The effects of endothelins (ETs) on the production of stromelysins, a sub-family of matrix metalloproteinases, were examined in cultured astrocytes. The treatment of cultured rat astrocytes with ET-1 increased stromelysin-1 mRNA levels, while stromelysin-2 and -3 mRNAs were not affected. Immunocytochemical observations showed that cultured astrocytes produced stromelysin-1 protein. ET-1 and Ala1,3,11,15-ET-1, an ETB receptor selective agonist, stimulated the release of stromelysin-1 from cultured astrocytes. Accompanying the increase in protein release, the peptidase activity of stromelysin-1 in the medium was also increased by ET-1. The effects of ET-1 on astrocytic stromelysin-1 expression were inhibited by PD98059, staurosporine, and Ca2+ chelation, but not by SB203580 or pyrrolidine dithiocarbamate. These results show that activation of astrocytic ET receptors stimulates the production of stromelysin-1, suggesting a role for ETs in stromelysin production in brain pathologies.  相似文献
