松嫩平原盐碱地中耐(嗜)盐菌的生物多样性   总被引:3,自引:0,他引:3  
【目的】分离纯化松嫩平原盐碱地中可培养的耐盐菌和嗜盐菌,并分析其生物多样性。【方法】采用纯培养法和定向富集法从该地区盐碱土样中分离耐盐菌和嗜盐菌,然后通过16S rRNA基因同源性比对鉴定所分离细菌的系统发育学地位,从而获取松嫩平原盐碱地中耐盐菌和嗜盐菌的多样性信息。【结果】共分离到细菌40株,分属于细菌域中3个门(Actinobacteria,Firmicutes,γ-Proteobacteria)、8个科、16个属、34个种。其中多数菌株属于厚壁菌门(Firmicutes),最优势属为葡球菌属(Staphylococcus)(8株,占总菌株的20%),其次依次为盐单胞菌属(Halomonas)(5株,12.5%)、芽胞杆菌属(Bacillus)(4株,10%)、大洋芽胞杆菌属(Oceanbacillus)(4株,10%)、库克菌属(Kocuria)(4株,10%)和假单胞菌属(Pseudomonas)(3株,7.5%)等。其中9株细菌的16S rRNA基因序列与最近缘种的同源性在97.2%-99.0%之间,可能为新种。菌株耐盐能力主要在5%-10%之间,其中62.5%的菌株为耐盐菌,其余则为中度嗜盐菌。所有菌株的耐碱能力在pH 9-12之间,其中60%的菌株耐碱能力则高达pH 12,除两株为嗜碱菌,其余均为耐碱菌。【结论】研究结果表明,松嫩平原盐碱地中耐盐菌与嗜盐菌种群丰富,主要以葡萄球菌和盐单胞菌为主,菌株不仅耐盐能力高而且耐碱能力也高,并且该地区可能含有丰富的耐盐菌和嗜盐菌的新物种。  相似文献

【目的】研究海水池塘养殖条件下的脊尾白虾肠道微生物菌群组成及多样性。【方法】采用PCR-RFLP技术,以直接提取的脊尾白虾肠道细菌总DNA为模板进行16S rRNA 基因扩增,产物与T载体连接建立质粒文库,从RFLP建立的文库中筛选出不同细菌来源的克隆子,将测定的差异克隆子16S rRNA片段序列与GenBank数据库进行比对。【结果】从脊尾白虾肠道的菌群文库中共获得114个克隆子,Hae Ⅲ和Msp I双酶切得到11种差异克隆子;对差异克隆子测序后,其中8个差异序列与已知细菌具有较高的同源性,分别是假单胞菌属(Pseudomonas)、肠杆菌属(Enterobater)、冰冻小杆菌属(Frigoribacterium)、褐杆菌属(Phaeobacter)、弧菌属(Vibrio)、赤杆菌属(Erythrobacter)、气单胞菌属(Aeromonas)、葡萄球菌属(Staphylococcus)和其他未培养细菌。【结论】初步揭示了脊尾白虾肠道微生物菌群结构的组成,为开发脊尾白虾专用微生态制剂提供基础资料。  相似文献

【目的】研究腹泻犊牛直肠细菌多样性,以及与健康犊牛直肠细菌多样性的差异。【方法】通过建立直肠菌群16S rRNA基因克隆文库,分别用限制性内切酶MspⅠ和HhaⅠ对阳性克隆的PCR产物进行限制性酶切片段长度多态性(RFLP)分析,通过测定16S rRNA基因序列,绘制系统发育树,确定犊牛直肠菌群的组成。【结果】腹泻组克隆阳性率达98.75%(474/480),优势菌群以乳杆菌属(14%)、肠球菌属(10%)和埃希菌属(8%)等需氧和兼性厌氧菌为主,健康组克隆阳性率达96.45%(488/506),优势菌群以梭菌属(13%)、双歧杆菌属(8%)和巨型球菌属(5%)等专性厌氧菌为主。【结论】2周龄犊牛直肠菌群复杂多样,并且具有自己的独特性菌群,且腹泻时乳杆菌属、肠球菌属、埃希氏菌属等显著增加。  相似文献

【目的】沿海滩涂耐盐植物重金属抗性内生细菌的筛选及其促生长潜在能力的研究有助于我们获得一些能够耐受并促进耐盐植物在被Cd2+、Pb2+、Hg2+、Cu2+,Zn2+等重金属离子污染的贫瘠的沿海滩涂上正常生长的菌株,达到既能够利用广袤的滩涂生物资源产生经济价值又能够净化生态环境的目的。【方法】以江苏南通沿海滩涂地区的4种耐盐植物为材料,采用稀释平板涂布法从中分离得到45株内生细菌,从中挑取23株代表性的菌株,对其进行抗重金属Cu2+、Pb2+、Cd2+、Zn2+,Hg2+的活性筛选;固氮、解磷、吲哚乙酸(IAA)的产生、1-氨基环丙烷-1-羧酸(ACC)脱氨酶活性等促生指标以及NaCl耐受能力的筛选。【结果】发现分离所得的大多数细菌能够耐受高浓度的Cu2+以及Pb2+,但是对Cd2+、Zn2+,Hg2+的耐受能力则较弱;26.1%的细菌具有固氮能力,21.7%的细菌具有解磷能力,60.9%的细菌能够产生IAA,39.1%的细菌含有ACC脱氨酶。对他们进行16S rRNA基因鉴定后发现,他们分属于芽胞杆菌属(Bacillus)、喜盐芽胞杆菌属(Halobacillus)、海洋芽胞杆菌属(Oceanobacillus)、微小杆菌属(Exiguobacterium)、沙雷氏菌属(Serratia)、短波单胞菌属(Brevundimonas)、弧菌属(Vibrio)、葡萄球菌属(Staphylococcus)共8个属,显示了丰富的多样性。其中菌株KLBMP 2432以及菌株KLBMP 2447为潜在的新种。【结论】沿海滩涂地区的耐盐植物内生细菌具有丰富多样的生物多样性以及促生长能力,且存在潜在的新种资源,并对重金属Cu2+,Pb2+具有较强的抗性。  相似文献

降雨对秦皇岛西浴场细菌总数和可培养菌群组成的影响   总被引:3,自引:0,他引:3  
【目的】研究降雨条件对浴场细菌总数和优势菌群组成的影响。【方法】2014年8月强降雨前后采集秦皇岛西浴场3个站位的海水样品,采用荧光显微镜计数法和平板计数法分别对细菌总数和可培养细菌总数进行计数;对群落结构组成进行分析,并对可培养细菌进行鉴定。【结果】雨前3个站位细菌总数和可培养细菌总数平均值分别为5.6×10~9 CFU/L和8.3×10~7 CFU/L,雨后分别为9.2×109 CFU/L和2.1×10~8 CFU/L。在可培养菌群中,变形菌门(Proteobacteria,雨前占80%,雨后占73%)是主要的微生物类群,其次为拟杆菌门(Bacteroides,雨前占12%,雨后占13%)、厚壁菌门(Firmicutes,雨前占7%,雨后占11%)等;肠杆菌属(Enterobacter spp.,21株)、海杆菌属(Marinobacter spp.,13株)、弓形菌属(Arcobacter spp.,13株)、假单胞菌属(Pseudomonas spp.,10株)、芽孢杆菌属(Bacillus spp.,10株)和弧菌属(Vibrio spp.,6株)为雨前可培养细菌优势属,而雨后可培养细菌优势属为肠杆菌属(22株)、海杆菌属(21株)、芽孢杆菌属(14株)、不动杆菌属(Acinetobacter spp.,11株)、假单胞菌属(9株)和弓形菌属(5株)等。【结论】降雨对细菌总数有显著的影响,同时降雨后浴场微生物群落结构发生了改变。  相似文献

基于高通量测序研究青藏高原茶卡盐湖微生物多样性   总被引:6,自引:0,他引:6  
【目的】茶卡盐湖(Chaka Salt Lake,CSL)是青藏高原有名的天然结晶盐湖,具有独特的石盐盐湖矿床,盛产青盐。盐湖卤水环境中存在丰富的嗜盐菌资源和潜在的新种,细菌和古菌的群落结构特征和物种多样性尚不明确。【方法】采用Illumina高通量测序平台对茶卡盐湖水样和底泥混合物中的细菌和古菌群落进行16S r RNA基因(V3-V5区)高通量测序,检测4个样本的群落结构差异和微生物多样性。【结果】获得细菌和古菌总有效序列分别为117 192和110 571条。结果分析表明细菌和古菌的物种注释(Operational taxonomic unit,OTU)数目分别为421和317,获得分类地位明确的细菌种类为14门28纲170属,古菌为5门4纲34属。细菌的优势类群是厚壁菌门(Firmicutes),所占比例为68.37%,其次为变形菌门Proteobacteria(20.49%);优势种属依次为芽孢杆菌属Bacillus(41.94%)、海洋芽孢杆菌属Oceanobacillus(8.03%)、假单胞菌属Pseudomonas(7.67%)、盐厌氧菌属Halanaerobium(7.42%)和乳球菌属Lactococcus(7.38%);古菌的优势类群以广古菌门(Euryarchaeota)盐杆菌纲(Halobacteria)为主,优势菌是Halonotius(17.21%)和盐红菌属Halorubrum(16.23%)。【结论】揭示了茶卡盐湖中细菌和古菌的群落结构及物种多样性,为嗜盐菌的开发及后续微生物资源的挖掘提供了理论依据。  相似文献

【背景】烟用香精是卷烟生产中的重要辅料,有些香精,特别是剩余的加料香精在运输和储存等过程中可能污染微生物,引起腐败变质发生。【目的】分析变质烟用香精中腐败微生物的群落多样性,确定引起加料香精变质的主要腐败微生物。【方法】采用稀释平板法,对我国典型湿热地区云南、福建和广西三地变质加料香精样品中的污染细菌、霉菌或酵母进行分离,分析其16S rRNA、ITS或26S rRNA基因序列,并通过反证试验确定引起变质的主要腐败微生物。【结果】所考察3个典型湿热地区变质加料香精存在不同程度的细菌、霉菌或酵母污染,其中细菌污染最为普遍。分离的76株污染细菌分布在2门4纲5目10科15属,细菌的群落多样性比较丰富,其中雷尔氏菌属(Ralstonia)、柠檬酸杆菌属(Citrobacter)、肠杆菌属(Enterobacter)及芽孢杆菌属(Bacillus)是优势菌群;9株霉菌分布在子囊菌门(Ascomycota)的3纲6属,每个属分离到1-2株霉菌;42株酵母分布在子囊菌门的6个属,其中毕赤酵母属(Pichia)、细枝钩酵母菌属(Wickerhamomyces)及酵母菌属(Saccharomyces)是优势菌群。云南和福建两地样品的细菌菌群组成明显多于广西,广西样品的霉菌菌群组成以及福建样品的酵母菌群组成均分别多于其余两地。反证试验结果显示,柠檬酸杆菌属、肠杆菌属、芽孢杆菌属、雷尔氏菌属和毕赤酵母属是引起烟用加料香精变质的主要腐败微生物。【结论】变质加料香精中污染微生物菌群组成及腐败微生物确定的研究将有助于我国湿热地区香精香料中污染微生物的控制和腐败变质的预防。  相似文献

新疆两盐湖可培养极端嗜盐菌组成及功能多样性研究   总被引:1,自引:0,他引:1  
【目的】通过分析不同成盐类型盐湖中的极端嗜盐菌群落组成差异,探究可培养极端嗜盐菌的功能特性。【方法】采集新疆硫酸盐型盐湖七角井和碳酸盐型盐湖南湖的土壤样品,通过平板稀释涂布法分离极端嗜盐菌,经过形态学观察、特征分析获取代表菌株,通过耐盐性测定和16S rRNA基因序列测序等对代表菌株进行鉴定,并对极端嗜盐菌的蛋白酶、淀粉酶、纤维素酶和酯酶活性进行筛选,同时检测苯酚降解能力。【结果】本研究共获得1 679株极端嗜盐菌,代表菌株45株,隶属于5门14个属,古菌数量(70.58%)明显多于细菌,最优盐浓度生长范围为18.4%–20.0%。在属水平上,盐湖中优势类群为古菌的Haloterrigena属(32.94%)和Natrialba属(26.03%),以及细菌的Aquisalimonas属(9.85%)和Aliifodinibius属(8.10%)。两盐湖中,盐度较低的南湖物种丰富度高于七角井盐湖,古菌物种组成相似,均以Haloterrigena属为主;细菌群落组成有差异,南湖以Aquisalimonas属为主,而七角井以Aliifodinibius属为主。功能筛选表明,盐湖中80%的嗜盐...  相似文献

【背景】锡林河-河滨湿地-阶地草原是蒙古高原典型草原区代表性的水生-湿生-陆生生境,但不同生境中反硝化菌群的空间分布特征尚不明晰。【目的】阐明典型草原区不同生境反硝化菌群的组成、丰度、空间分布特征及异质性成因。【方法】利用16S rRNA基因测序研究锡林河流域水生、湿生、陆生生境6个样带沉积物/土壤细菌群落组成及相对丰度。基于2014年及以前文献报道的反硝化细菌及16S rRNA基因信息构建参比菌库,筛选生境关联的反硝化菌属。通过典范对应分析等探究反硝化菌群空间异质性成因。【结果】参比菌库包含80种反硝化细菌(65个属),6个样带测序获得的469个细菌属中36个为反硝化细菌属。3种生境共存的反硝化细菌有14个属,其中黄杆菌属(1.65%-14.17%)和噬氢菌属(1.56%-1.69%)是水生和湿生生境共有的优势菌,假单胞菌属(1.85%)是低河漫滩样带的优势菌。空间分布特征显示反硝化菌群沿水生-湿生-陆生生境呈现先升后降的分布趋势,在低河漫滩湿地达到最高值。典范对应分析表明:黄杆菌属、噬氢菌属、气单胞菌属、鞘氨醇单胞菌属等与pH值、水分及沙粒含量呈正相关关系,而芽孢杆菌属、链霉菌属、马杜拉放线菌属等与粘粒、粉粒、有机质、总氮含量等呈正相关关系。【结论】典型草原区反硝化菌群组成及丰度具有明显的生境异质性,低河漫滩湿地是反硝化细菌生长繁殖的最佳生境,由颗粒组成、水分含量和pH等环境因子共同驱动。  相似文献

【目的】揭示陕北花马盐湖沉积物原核微生物群落组成,并分析其潜在的耐盐功能基因。【方法】构建盐湖沉积物宏基因组16S r RNA文库和fosmid文库,利用Illumina HiSeq高通量测序及生物信息技术分析细菌古菌群落组成和耐盐菌株(5-5)外源宏基因组的潜在耐盐基因。【结果】获得18978条有效的16Sr RNA序列,共5221个OTUs,包括23个门,155个属,其中广古菌门(Euryarchaeota)和变形菌门(Proteobacteria)为优势菌门,盐杆状菌属(Halorhabdus)、盐红菌属(Halorubrum)及假单胞菌属(Pseudomonas)等16个属为优势属,以及嗜盐单胞菌属(Halomonas)、冷弯菌属(Psychroflexus)及不动细菌属(Acinetobacter)等139个属为非优势属。从4126个fosmid文库菌株中筛选出37株耐盐菌株,其中菌株5-5、2E4和2F4对不同浓度的NaCl、CuSO_4、ZnSO_4及CdSO_4具有耐受性,从5-5的外源宏基因组序列中获得61个Unigene,其中12个Unigene的同源基因编码的蛋白质如无机焦磷酸酶、转座酶、亚碲酸钾抗性蛋白及钙调蛋白等广泛参与其他生物的耐盐逆境。【结论】盐湖沉积物中蕴藏着丰富多样的细菌古菌类群以及潜在耐盐功能基因资源。  相似文献

九龙江河口及厦门污水处理设施抗生素抗性基因污染分析   总被引:3,自引:0,他引:3  
【目的】近年来由于抗生素的滥用,导致了多药物抗性超级细菌的产生,有关抗生素抗性基因(Antibiotic resistance genes,ARGs)在环境介质中分布、迁移和扩散已经引起人们的广泛关注。针对九龙江河口及厦门污水处理设施抗生素抗性基因污染情况开展研究。【方法】通过定性PCR研究九龙江河口水体、沉积物和厦门污水处理设施活性污泥中4种磺胺类、13种四环素类ARGs及2种整合子基因的污染情况,并选择四环素类tet(W)基因进行克隆文库测序分析。【结果】除tet(O)和tet(S)外,其他基因均被检出。不同环境介质中的ARGs及整合子基因检出率为活性污泥(0.86)>沉积物(0.57)>水体(0.24)。在淡水和淡盐水中,sul(l)、int(1)、tet(A)、tet(C)、tet(E)、tet(M)和tet(W)的检出率要高于海水,表明九龙江上游可能是ARGs的污染源之一。【结论】主成分分析表明污水处理设施是ARGs的高发载体;沉积物是ARGs的稳定载体;而水体中的ARGs易于分解。此外,tet(W)基因克隆文库分析表明,厦门污水处理设施也可能是九龙江河口及厦门沿岸的ARG污染源。  相似文献

九龙江河口区nirS型反硝化细菌多样性及系统发育学分析   总被引:2,自引:0,他引:2  
【目的】结合16S rRNA基因克隆文库和nirS基因克隆文库的分析,揭示九龙江河口区nirS型反硝化细菌多样性。【方法】选取九龙江河口区一富营养化采样点,分别采集水样及沉积物样品,进行理化因子的测定并提取细菌总DNA。以水样DNA构建16S rRNA基因克隆文库,以沉积物DNA构建nirS基因克隆文库,分析微生物群落结构的多样性并构建系统发育树。【结果】从16S rRNA基因克隆文库中获得86条有效序列,按97%的序列相似性划分为53个OTU,分别属于Proteobacteria门、Planctomycetes门、Bacteroidetes门、Actinobacteria门、Firmicutes门和Chloroflexi门。其中属于Proteobacteria门OTU的克隆子占克隆数的62.9%,是最优势的类群,分属于Alphaproteobacteria、Betaproteobacteria、Gammaproteobacteria和Deltaproteobacteria纲等。从nirS基因克隆文库中获得190条有效序列,翻译为氨基酸序列后,按82%的序列相似性划分为60个OTU,并定位到属的水平。其中Proteobacteria门是最优势的类群,占文库克隆子总数的71.6%,包括Alphaproteobacteria纲(5.8%)、Betaproteobacteria纲(49.0%)和Gammaproteobacteria纲(16.9%)。nirS基因克隆文库中丰度最高的OTU与GenBank中的一株可培养反硝化菌Thauera sp. R-26906具有100%的序列相似性。【结论】九龙江河口区的微生物以及亚硝酸盐还原酶基因(nirS)具有丰富的多样性。大部分NirS序列在GenBank中的最相似序列来源于河口、海湾等相似的环境。  相似文献

Vibrio and Vibrio-like bacteria were isolated from water, sediments, plants and faeces from eight riverine sites in South East Queensland, Australia, using filtrations, enrichments and selective growth on thiosulphate-citrate-bile salts-sucrose (TCBS) and Simidu agars. Isolates (402 in toto) were classified by numerical analysis of phenotypic characteristics and comparison with fifteen reference cultures. Of the isolates 41 (10.2%) were identified as Vibrio cholerae, 33 (8.2%) as V. fluvialis and 118 (29.4%) as motile Aeromonas species. Other isolates resembled V. parahaemolyticus, V. alginolyticus, V. vulnificus and V. hollisae. No isolates were positively identified as V. damsela or Plesiomonas shigelloides. Vibrio species tolerant of low salt levels and aeromonads were widely distributed in riverine locations but more halophilic species were restricted to more saline areas. Simidu agar failed to select for vibrios as effectively as TCBS agar. Enrichment for 18 h produced more isolates than for 6 h.  相似文献

【目的】了解黑龙江省大豆田大豆胞囊线虫胞囊可培养细菌的多样性。【方法】运用稀释平板法和16SrDNA基因序列的系统发育分析对胞囊可培养细菌多样性进行研究。【结果】用NA培养基从胞囊上分离90株具有不同菌落形态的细菌。16S rDNA序列分析结果表明:90株菌株分属于7个属22个种。46株属于变形菌门γ亚群(Gammaproteobacteria),32株属于厚壁菌门(Firmicutes),10株属于变形菌门β亚群(Betaproteobacteria),2株属于变形菌门ɑ亚群(Alphaproteobacteria)。假单胞菌属(Pseudomonas)和芽孢杆菌属(Bacillus)为优势菌属。【结果】黑龙江省大豆胞囊线虫胞囊中存在丰富的细菌物种多样性,这些细菌对大豆胞囊线虫可能具有一定的生理生态作用。  相似文献

This study characterises the bacteria associated with a marine hatchery in Tunisian coastal marine waters. Presumptive vibrios (TCBS agar) and heterotrophic aerobic microflora (CFU) were studied at different stages within the hatchery: seawater, batches of algal cultures, rotifers andArtemia culture tanks. The bacterial strains were isolated on TCBS Agar plates and described using different bacteriological tests (standardised micromethods “API 20 E Strips”, exoenzymes production, growth at different temperatures, pH and salinity, vibriostatic agent O/129 and antibiotics susceptibility). Two dominant genera of bacteria were found (Vibrio andAeromonas) associated with some strains of thePseudomonadaceae family.Vibrio alginolyticus was the dominant bacteria (75% of total isolates) found in rotifers (Brachionus plicatilis) andArtemia cultures (Artemia salina). In larvae rearing tanks, an increase ofVibrionaceae was noted after larvae were fed withArtemia. Most of the studied bacteria used the skin mucus ofSparus aurata larvae as their sole source of carbon. All theV. alginolyticus strains were β-haemolytic, hydrolyse the DNA and were susceptible to several tested antibiotics.  相似文献

A specific search for Vibrio vulnificus in natural marine samples from the Spanish Mediterranean Sea was carried out by nested PCR and cultural approaches using thiosulphate-citrate-bile salts-sucrose agar (TCBS) and cellobiose-polymixin B-colistin agar (CPC), incubated at 40 degrees C, as selective media. Presumptive colonies were identified by PCR using specific primers against 23S rRNA sequences. This species was isolated from sea water and edible bivalves, mainly after preenrichment in alkaline peptone water (APW) at 40 degrees C followed by CPC agar. None of the V. vulnificus isolates identified corresponded to serovar E. Dominant Vibrio species on directly inoculated TCBS plates incubated at 25 degrees C were V. splendidus below 20 degrees C and V. harveyi and V. mediterranei above that temperature. Low percentages of several pathogenic vibrios were recorded but V. vulnificus was never recovered at this incubation temperature. The incidence of this species in the samples studied was lower than that described for other geographical areas, probably due to the high salinity values of the Mediterranean Sea.  相似文献

摘要:【目的】为了分析厦门近海原位海水中多环芳烃降解菌的多样性。【方法】将涂有菲的聚氯乙烯(PVC)板悬挂在厦门国际邮轮码头的海水中,进行菲降解菌的原位富集。利用变性梯度凝胶电泳(Denaturing gradient gel electrophoresis,DGGE)和16S rRNA基因文库两种方法分析了在PVC板表面富集微生物的菌群结构。之后,在实验室模拟原位条件下,对PVC板表面富集的菲降解菌群进行进一步富集、分离和初步鉴定。【结果】PVC板在海水中浸没6 d后,16S rRNA基因文库分析表明,在涂菲的PVC板表面富集的菌群中解环菌属(Cycloclasticus)对应的克隆子占文库总克隆子的50%;在未涂菲的PVC板表面吸附的菌群中红杆菌科(Rhodobacteraceae)为优势菌,其对应的克隆子占文库总克隆子的47%;而解环菌属的克隆子只占文库总克隆子的2%。DGGE的分析结果也证明解环菌是菲原位富集降解菌群中的优势菌。实验室进一步富集后,从该菌群中分离鉴定出14株细菌,其中一株新鞘氨醇杆菌B14(Novosphingobium sp.B14)具有菲降解能力。但是,解环菌未能获得纯培养。【结论】菲原位富集发现,厦门近海水体中解环菌是多环芳烃的主要降解菌。  相似文献

An outbreak of serious mortality among the cultivated juvenile cobia Rachycentron canadum L. (weighing 8–10 g) characterized by a swollen intestine containing transparent yellow fluid (ascites and gastroenteritis) occurred in August 2001 in Taiwan. Ten motile bacterial strains, C3d1–C3d10, were isolated from head kidney (an organ located near the head of the fish) and/or the intestinal yellow fluid on tryptic soy agar supplemented with 1% NaCl (TSA1) and/or thiosulphate citrate bile salt sucrose (TCBS) agar plates. These strains were characterized and identified as Vibrio harveyi(V. carchariae) on the basis of biochemical characteristics, and comparisons with those of three reference strains, originally identified as V. harveyior V. carchariae. The strain C3d1 was selected as a representative strain for virulence tests and was found lethal to the cobia with an LD50 value of 7.48 × 104 colony forming units g–1 fish body weight. All the moribund/dead fish exhibited gastroenteritis as that observed in natural outbreak. The same bacteria could be reisolated from kidney and the transparent yellow fluid of swollen intestine of fish after bacterial challenge using TSA1 and TCBS plates. This is a first report showing that V. harveyi(V. carchariae) is the causative agent of gastroenteritis in the cobia.  相似文献

自2002年后半年开始,汕尾出现大规模九孔鲍鲍苗脱板问题。为了解养殖环境中的细菌(主要是弧菌)对鲍苗的影响,于汕尾一养殖场跟踪观察多次鲍鱼育苗过程,取鲍苗正处于脱板过程中的水样和采苗板样用TCBS平板分离出25株菌,测定其分泌酪蛋白酶、胰蛋白酶和明胶酶3种胞外酶的能力,并进行其溶血现象分析。结果表明,产酶菌株达到17株,其中10株菌兼具分泌溶血素的能力。对这10株菌进行了种类鉴定,表明它们是副溶血弧菌(4株)、溶藻弧菌(2株)、腐败希瓦菌(3株)和杀鲑气单胞菌(1株)。  相似文献

Bacteria inhibitory to fish larval pathogenic bacteria were isolated from two turbot larva rearing farms over a 1-year period. Samples were taken from the rearing site, e.g., tank walls, water, and feed for larvae, and bacteria with antagonistic activity against Vibrio anguillarum were isolated using a replica plating assay. Approximately 19,000 colonies were replica plated from marine agar plates, and 341 strains were isolated from colonies causing clearing zones in a layer of V. anguillarum. When tested in a well diffusion agar assay, 173 strains retained the antibacterial activity against V. anguillarum and Vibrio splendidus. Biochemical tests identified 132 strains as Roseobacter spp. and 31 as Vibrionaceae strains. Partial sequencing of the 16S rRNA gene of three strains confirmed the identification as Roseobacter gallaeciensis. Roseobacter spp. were especially isolated in the spring and early summer months. Subtyping of the 132 Roseobacter spp. strains by randomly amplified polymorphic DNA with two primers revealed that the strains formed a very homogeneous group. Hence, it appears that the same subtype was present at both fish farms and persisted during the 1-year survey. This indicates either a common, regular source of the subtype or the possibility that a particular subtype has established itself in some areas of the fish farm. Thirty-one antagonists were identified as Vibrio spp., and 18 of these were V. anguillarum but not serotype O1 or O2. Roseobacter spp. strains were, in particular, isolated from the larval tank walls, and it may be possible to establish an antagonistic, beneficial microflora in the rearing environment of turbot larvae and thereby limit survival of pathogenic bacteria.  相似文献
