The degradation of an azo dye mixture by an aerobic bacterial consortium was studied in a rotating biological reactor. Laterite pebbles of particle size 850 microm to 1.44 mm were fixed on gramophone records using an epoxy resin on which the developed consortium was immobilized. Rate of degradation, BOD, biomass determination, enzymes involved, and fish bioassay were studied. The RBC has a high efficiency for dye degradation even at high dye concentrations (100 microg/mL) and high flow rate (36 L/h) at alkaline pH and salinity conditions normally encountered in the textile effluents. Bioassays (LD-50) using Thilapia fish in treated effluent showed that the percentage mortality was zero over a period of 96 h, whereas the mortality was 100% in untreated dye water within 26 h. Fish bioassay confirms that the effluent from RBC can be discharged safely to the environment.  相似文献

Bacterial consortium-AIE2 with a capability of contemporaneous Cr(VI) reduction and azo dye RV5 decolourization was developed from industrial wastewaters by enrichment culture technique. The 16S rRNA gene based molecular analyses revealed that the consortium bacterial community structure consisted of four bacterial strains namely, Alcaligenes sp. DMA, Bacillus sp. DMB, Stenotrophomonas sp. DMS and Enterococcus sp. DME. Cumulative mechanism of Cr(VI) reduction by the consortium was determined using in vitro Cr(VI) reduction assays. Similarly, the complete degradation of Reactive Violet 5 (RV5) dye was confirmed by FTIR spectroscopic analysis. Consortium-AIE2 exhibited simultaneous bioremediation efficiencies of (97.8 ± 1.4) % and (74.1 ± 1.2) % in treatment of both 50 mg l−1 Cr(VI) and RV5 dye concentrations within 48 h of incubation at pH 7 and 37°C in batch systems. Continuous bioreactor systems achieved simultaneous bioremediation efficiencies of (98.4 ± 1.5) % and (97.5 ± 1.4) % after the onset of steady-state at 50 mg l−1 input Cr(VI) and 25 mg l−1 input RV5 concentrations, respectively, at medium dilution rate (D) of 0.014 h−1. The 16S rRNA gene copy numbers in the continuous bioreactor as determined by real-time PCR assay indicated that Alcaligenes sp. DMA and Bacillus sp. DMB dominated consortium bacterial community during the active continuous bioremediation process.  相似文献

Decolorization and degradation of polyazo dye Direct Black 22 was carried out by distillery spent wash degrading mixed bacterial consortium, DMC. Response surface methodology (RSM) involving a central composite design (CCD) in four factors was successfully employed for the study and optimization of decolorization process. The hyper activities and interactions between glucose concentration, yeast extract concentration, dye concentration and inoculum size on dye decolorization were investigated and modeled. Under optimized conditions the bacterial consortium was able to decolorize the dye almost completely (>91%) within 12h. Bacterial consortium was able to decolorize 10 different azo dyes. The optimum combination of the four variables predicted through RSM was confirmed through confirmatory experiments and hence this bacterial consortium holds potential for the treatment of industrial waste water. Dye degradation products obtained during the course of decolorization were analyzed by HPTLC.  相似文献

Bacteria capable of degrading the sulfonated azo dye Red HE7B were isolated from textile mill effluent contaminated soil. The most efficient isolate was identified as Bacillus sp. Azo1 and the isolate could successfully decolorize up to 89 % of the dye. The decolorized cultural extract analyzed by HPLC confirmed degradation. Enzymatic analysis showed twofold and fourfold increase in the activity of azoreductase and laccase enzymes, respectively, indicating involvement of both reductive and oxidative enzymes in biodegradation of Red HE7B. Degraded products which were identified by GC/MS analysis included various metabolites like 8-nitroso 1-naphthol, 2-diazonium naphthalene. Mono azo dye intermediate was initially generated from the parent molecule. This mono azo dye was further degraded by the organism, into additional products, depending on the site of cleavage of R–N=N–R molecule. Based on the degradation products identified, three different pathways have been proposed. The mechanism of degradation in two of these pathways is different from that of the previously reported pathway for azo dye degradation. This is the first report of a microbial isolate following multiple pathways for azo dye degradation. Azo dye Red HE7B was observed to be phytotoxic, leading to decrease in root development, shoot length and seedling fresh weight. However, after biotreatment the resulting degradation products were non-phytotoxic.  相似文献

A microbial consortium capable of aerobic degradation of a mixture of azo dyes consisting of two isolated strains (RRL,TVM) and one known strain of Pseudomonas putida (MTCC 1194) was immobilized on laterite stones. The amount of bacterial biomass attached to the laterite stones was 8.64 g per 100 g of the stone on a dry weight basis. The packed bed reactor was filled with these stones and had a total capacity of 850 mL and a void volume of 210 mL. The feed consisted of an equal mixture of seven azo dyes both in water as well as in a simulated textile effluent, at a pH of 9.0 and a salinity of 900 mg/L. The dye concentrations of influent were 25, 50, and 100 microg/mL.The residence time was varied between 0.78 and 6.23 h. It was found that at the lowest residence time 23.55, 45.73, and 79.95 microg of dye was degraded per hour at an initial dye concentration of 25, 50, and 100 microg, respectively. The pH was reduced from 9.0 to 7.0. Simulated textile effluent containing 50 microg/mL dye was degraded by 61.7%. Analysis of degradation products by TLC and HPLC showed that the dye mixture was degraded to nontoxic smaller molecules. The bacteria-coated pebbles were stable, there was no washout even after 2 months, and the reactor was found to be suitable for the aerobic degradation of azo dyes.  相似文献

Summary Soil samples collected from contaminated sites of Vatva, Gujarat, India were studied for screening and isolation of organisms capable of decolourizing textile dyes. A bacterial consortium RVM 11.1 was selected on the basis of rapid dye decolourization. Reactive Violet 5 (RV 5) was used as model dye. The consortium exhibited 94% decolourization ability within 37 h under a wide pH range from 6.5 to 8.5 and temperature ranging from 25 to 40 °C. The bacterial consortium was able to grow and decolourize RV5 under static conditions in the presence of glucose and yeast extract and also showed an ability to decolourize in the presence of starch in place of glucose. Maximum decolourization efficiency was observed at 200 ppm (mg/l) concentration of RV 5. Bacterial consortium RVM11.1 had the ability to decolourize 10 different dyes tested. The transformation and degradation products after decolourization were examined by HPTLC.  相似文献

Combination of anaerobic–aerobic sequencing processes result in both anaerobic color removal and aerobic aromatic amine removal during the treatment of dye-containing wastewaters. The aim of the present study was to gain more insight into the competitive biochemical reactions between sulfate and azo dye in the presence of glucose as electron donor source. For this aim, anaerobic–aerobic sequencing batch reactor fed with a simulated textile effluent including Remazol Brilliant Violet 5R (RBV 5R) azo dye was operated with a total cycle time of 12 h including anaerobic (6 h) and aerobic cycles (6 h). Microorganism grown under anaerobic phase of the reactor was exposed to different amounts of competitive electron acceptor (sulfate). Performance of the anaerobic phase was determined by monitoring color removal efficiency, oxidation reduction potential, color removal rate, chemical oxygen demand (COD), color, specific anaerobic enzyme (azo reductase) and aerobic enzyme (catechol 1,2-dioxygenase), and formation of aromatic amines. The presence of sulfate was not found to significantly affect dye decolorization. Sulfate and azo dye reductions took place simultaneously in all operational conditions and increase in the sulfate concentration generally stimulated the reduction of RBV 5R. However, sulfate accumulation under anaerobic conditions was observed proportional to increasing sulfate concentration.  相似文献

Treatment of textile wastewater is a big challenge because of diverse chemical composition, high chemical strength and color of the wastewater. In the present study, treatment of wastewater containing reactive black-5 azo dye was studied in anaerobic sequencing batch bioreactor (SBBR) using mixed liquor suspended solids (MLSS) from suspended and attach growth bioreactors. MLSS at concentration of 1000 mg/L and reactive black-5 azo dye at 100 mg/L were used. A culture (108–109 CFU/ml) of pre-isolated bacterial strains (Psychrobacter alimentarius KS23 and Staphylococcus equorum KS26)) capable of degrading azo dyes in mineral salt medium was used to accelerate the treatment process in bioreactor. Different combinations of sludge, culture and dye were used for treatment using different co-substrates. About 85% COD removal was achieved by consortium (MLSS + KS23 + KS26) after 24 h in attach growth bioreactor. Similarly, 92% color removal was observed with consortium in attach growth bioreactor compared to 85% color removal in suspended bioreactor. Addition of bacterial culture (20%, v/v) to the bioreactor could enhance the rate of color removal. This study suggests that biotreatment of wastewater containing textile dyes can be achieved more efficiently in the attach growth bioreactor using yeast extract as a co-substrate and MLSS augmented with dye-degrading bacterial strains.  相似文献

【目的】为了获得能够在高盐环境下脱色偶氮染料的嗜盐菌群及其降解机理。【方法】采用富集驯化的方法获得一个嗜盐菌群,采用Illumina HiSeq2500测序平台对其群落结构进行测定;采用分光光度法测定了其降解特性;采用GC-MS和红外图谱分析了其降解机理;采用微核实验的方法比较了偶氮染料降解前后的毒性。【结果】该菌群在10%的盐度下,使100mg/L的酸性金黄G在8h内脱色。菌群主要由Zobellella、Rheinheimera、Exiguobacterium和Marinobacterium组成。最适宜的脱色条件是:pH=6,酵母粉为碳源,蛋白胨或硝酸钾作为氮源,盐度为1%–10%。酸性金黄G降解产物的毒性比降解前降低。酸性金黄G主要的降解产物是对氨基二苯胺和二苯胺。此外,该菌群还能使酸性大红GR和直接湖蓝5B等多种偶氮染料脱色,具有较好的脱色广谱性。【结论】获得了快速降解偶氮染料的嗜盐菌群及降解机理,为该嗜盐菌群应用于高盐印染废水的处理提供菌种资源和理论支持。  相似文献

Mixed anaerobic bacterial consortia have been show to reduce azo dyes and batch decolourisation tests have also demonstrated that predominantly methanogenic cultures also perform azo bond cleavage. The anaerobic treatment of wool dyeing effluents, which contain acetic acid, could thus be improved with a better knowledge of methanogenic dye degradation. Therefore, the decolourisation of two azo textile dyes, a monoazo dye (Acid Orange 7, AO7) and a diazo dye (Direct Red 254, DR254), was investigated in a methanogenic laboratory-scale Upflow Anaerobic Sludge Blanket (UASB), fed with acetate as primary carbon source. As dye concentration was increased a decrease in total COD removal was observed, but the acetate load removal (90%) remained almost constant. A colour removal level higher than 88% was achieved for both dyes at a HRT of 24h. The identification by HPLC analysis of sulfanilic acid, a dye reduction metabolite, in the treated effluent, confirmed that the decolourisation process was due mainly to azo bond reduction. Although, HPLC chromatograms showed that 1-amino-2-naphthol, the other AO7 cleavage metabolite, was removed, aeration batch assays demonstrated that this could be due to auto-oxidation and not biological mineralization. At a HRT of 8h, a more extensive reductive biotransformation was observed for DR254 (82%) than for AO7 (56%). In order to explain this behaviour, the influence of the dye aggregation process and chemical structure of the dye molecules are discussed in the present work.  相似文献

A bacterial consortium (consortium GR) consisting of Proteus vulgaris NCIM-2027 and Micrococcus glutamicus NCIM-2168 could rapidly decolorize and degrade commonly-used sulfonated reactive dye Green HE4BD and many other reactive dyes. Consortium GR shows markedly higher decolorization activity than that of the individual strains. The preferable physicochemical parameters were identified to achieve higher dye degradation and decolorization efficiency. The supplementation of cheap co-substrates (e.g., extracts of agricultural wastes) could enhance the decolorization performance of consortium GR. Extent of mineralization was determined with TOC and COD measurements, showing nearly complete mineralization of Green HE4BD by consortium GR (up to 90% TOC and COD reduction) within 24 h. Oxidoreductive enzymes seemed to be involved in fast decolorization/degradation process with the evidence of enzymes induction in the bacterial consortium. Phytotoxicity and microbial toxicity studies confirm that the biodegraded products of Green HE4BD by consortium GR are non-toxic. Consortium GR also shows significant biodegradation and decolorization activities for mixture of reactive dyes as well as the effluent from actual dye manufacturing industry. This confers the possibility of applying consortium GR for the treatment of industrial wastewaters containing dye pollutants.  相似文献

Biodegradation of Reactive Blue 59 by isolated bacterial consortium PMB11   总被引:2,自引:0,他引:2  
Morphologically different, three bacterial strains, capable of decolorizing Reactive Blue 59 were isolated from dye effluent contaminated soil sample, collected from Ichalkaranji, India. The individual bacterial strains viz. Bacillus odysseyi SUK3, Morganella morganii SUK5 and Proteus sp. SUK7 decolorized Reactive Blue 59 (50 mg l(-1)) completely within 60, 30, 24 h, respectively, while the bacterial consortium PMB11 of these strains required 3 h for the complete decolorization. The decolorization was confirmed by UV-Vis spectroscopy. Further, the biodegradation of Reactive Blue 59 in to different metabolites was confirmed by High performance liquid chromatography and Fourier transform infrared spectroscopy analysis. Significant increase in the activity of aminopyrine N-demethylase (AND) in the individual as well consortium cells, obtained after decolorization showed involvement of AND in the decolorization process. Phytotoxicity studies, revealed the nontoxic nature of the degraded metabolites of Reactive Blue 59 indicating effectiveness of bacterial consortium PMB11 for the treatment of textile effluent containing Reactive Blue 59.  相似文献

Textile industry uses azo dyes in its processes, which are complex organic molecules that are not easy to be degraded. Reactive dyes are especially difficult to remove from wastewater because of the characteristics of the molecule: one or more azo bonds, naphthalene‐disulfonate, triazine or chloro‐triazine, and phenyl‐amine groups. The degradation of the azo dye reactive red 272 was studied under anaerobic conditions in a hybrid Upflow Anaerobic Sludge Bed reactor (UASB) with an activated carbon bed. An adapted consortium of microorganisms was used in the kinetic study (batch) and to inoculate the UASB reactor. The experimental design identified the main factors determining the dye reduction efficiency are the initial concentration of dye and dextrose (as electron donor) and the residence time in the reactor. Dye reduction rate was decreased as the concentration increases in the wastewater; as a result, a kinetic model with a change from first to second order is proposed. The kinetic study showed that the process is first abiotic (adsorption) and then biotic (biodegradation).  相似文献

【目的】获得能够在高盐环境下脱色偶氮染料的高效脱色菌群,应用于印染废水的生物处理。【方法】采用在5%盐度培养基富集的方法,从印染废水的活性污泥中富集能够在5%盐度下脱色酸性大红GR的嗜盐混合菌群,利用高通量测序方法研究其群落结构,利用静置培养的方法研究其脱色性能。【结果】该菌群可以在5%盐度、静置培养下15 h内将100 mg/L的酸性大红GR几乎完全脱色,主要由Halomonas、Salinicoccus、Nitratireductor和Aequorivita等4个属组成,Halomonas是主要的脱色菌。高浓度的Na NO_3、Na_2SO_4和Na Cl抑制菌群的脱色,其中Na NO_3抑制作用最强。该菌群的最佳脱色条件是在p H 7.0、盐度5%、30°C脱色效果最好,可脱色直接耐黑G和分散深蓝S-3BG等偶氮染料,并且具有连续脱色的能力。【结论】嗜盐菌群在处理偶氮染料废水中具有良好的应用价值。  相似文献

Stoichiometric amounts of the aromatic amines 6-aminonaphthalene-2-sulfonate and 5-aminosalicylate were generated and excreted into the medium. After re-aeration of the culture, these amines were mineralized by different members of the bacterial culture. Thus, total degradation of a sulfonated azo dye was achieved by using an alternating anaerobic-aerobic treatment. The ability of the mixed bacterial culture to reduce the azo dye was correlated with the presence of strain BN6, which possessed the ability to oxidize various naphthalenesulfonic acids. It is suggested that strain BN6 has a transport system for naphthalenesulfonic acids which also catalyzes uptake of sulfonated azo dyes. Anaerobic acclimatization of activated sludge from a textile effluent treatment plant to high concentration of RB5 could effectively decolorize RB5 dye solution. The strains viz. Pseudomonas aeruginosa and Bacillus circulans and other unidentified laboratory isolates (NAD1 and NAD6) were predominantly present in the microbial consortium. The conditions for efficient decolorization, biostimulation to increase effectiveness of microbial consortium, its tolerance to high salt concentration and non-specific ability towards decolorization of eight azo dyes, are reported. The optimum inoculums concentration for maximum decolorization were found to be 1-5 ml of 1800+/-50 mg l(-1) MLSS and 37 degrees C, respectively. The decolorization efficiency was 70-90% during 48 h. The biomass showed efficient decolorization even in the presence of 10% NaCl, as tested with RB5. In the presence of flavin adenine dinucleotide (FAD) more than 99% decolorization occurred in 8h. The decolorization of RB5 was traced to extracellular enzymes. The effectiveness of acclimatized biomass under optimized conditions towards decolorization of two types of synthetic dye bath wastewaters that were prepared using chosen azo dyes is reported.  相似文献

The aim of this study was to exploit the bacterial biofilms to remove dyes from industrial effluents. Biofilms of strains AK1, AK2, VKY1 and a consortium on sheep bone chips were examined in batch, repeated batch and continuous packed bed bioreactor. Biofilms are more efficient for decolourization of Amaranth dye at three different dye concentrations (200, 400, and 600 mg l−1). 100% decolourization of Amaranth dye was observed even at higher concentrations (400 and 600 mg l−1) by all the tested biofilms in 24 h than that of corresponding free cells. The biofilms were superior over those of free cells and could be reused for more than 18 repeated cycles. In a packed bed bioreactor, biofilms could be operated with much higher dilution rates and at lower hydraulic retention time. Further, the decolourization of dye was confirmed by UV–visible spectrophotometer, TLC and HPLC analysis of Amaranth dye degradation products from packed bed bioreactor effluent.  相似文献

In batch toxicity assays, azo dye compounds were found to be many times more toxic than their cleavage products (aromatic amines) towards methanogenic activity in anaerobic granular sludge. Considering the ability of anaerobic microorganisms to reduce azo groups, detoxification of azo compounds towards methanogens can be expected to occur during anaerobic wastewater treatment. In order to test this hypothesis, the anaerobic degradation of one azo dye compound, Mordant orange 1 (MO1), by granular sludge was investigated in three separate continuous upflow anaerobic sludge-blanket reactors. One reactor, receiving no cosubstrate, failed after 50 days presumably because of a lack of reducing equivalents. However, the two reactors receiving either glucose or a volatile fatty acids (acetate, propionate, butyrate) mixture, could eliminate the dye during operation for 217 days. The azo dye was reductively cleaved to less toxic aromatic amines (1,4-phenylenediamine and 5-aminosalicylic acid) making the treatment of MO1 feasible at influent concentrations that were over 25 times higher than their 50% inhibitory concentrations. In the reactor receiving glucose as cosubstrate, 5-aminosalicylic acid could only be detected at trace levels in the effluent after day 189 of operation. Batch biodegradability assays with the sludge sampled from this reactor confirmed the mineralization of 5-aminosalicylic acid to methane. In this work, the anaerobic period of an anaerobic–aerobic sequencing batch reactor was found to allow the reductive decolourisation of azo dyes. 1-l reactors were operated in 24-h cycles comprising anaerobic and aerobic reaction phases, fed with a simulated textile effluent including a reactive type (Remazol Brilliant Violet 5R) or an acid type (Acid Orange 7) azo dye. The aim was to assess the role of different redox phenomena in the anaerobic decolourisation process. Selective inhibition of sulphate reducing bacteria was carried out in the sulphate-containing, reactive dye fed reactor, resulting in nearly complete, though reversible and inhibition of decolourisation. The acid dye fed reactor's supplementation with sulphate, though resulting in sulphate reduction, did not improve decolourisation. Other redox mediators, namely quinones, were more effective in promoting electron transfer to the azo bond. Bio-augmentation of the acid dye fed reactor with a pure sulphate reducer strain known to decolourise azo dyes, Desulfovibrio alaskensis, was also carried out. Decolourisation was improved, but apparently as a result of the carbon source change required to support D. alaskensis growth. A chemically mediated reduction of the azo bond coupled to biological sulphate reduction, thus seemed to account for the high decolourisation yields of both dyes.  相似文献
