为了建立能有效分离 1kDa的特小分子肽的Tricine SDS PAGE方法 ,调整分离胶中聚丙烯酰胺凝胶的分子组成 ,使致密胶中丙烯酰胺和双丙烯酰胺的交联度为 5 %,并加入 36 5 %的尿素 ,用于分离小至 1kDa的小分子肽 ,并与常规的SDS PAGE和先前报道的Tricine SDS PAGE相比较。结果改进的致密胶可以有效分离分子量小至 1kDa的小肽 ,明显优于常规的SDS PAGE和现有的Tricine SDS PAGE方法。  相似文献

BackgroundSmall bowel adenocarcinoma (SBA), neuroendocrine tumors (NET) and gastrointestinal stromal tumors (GIST) are neoplastic lesions of the small bowel while small bowel adenomas are precursors of SBA.AimTo examine mortality in patients diagnosed with SBA, small bowel adenomas, NET and GIST.MethodsWe performed a population-based matched cohort study encompassing all individuals with SBA (n = 2289), adenomas (n = 3700), NET (n = 1884) and GIST (n = 509) in the small bowel diagnosed at any of Sweden’s 28 pathology departments between 2000 and 2016 (the “ESPRESSO study”). Each case was matched by sex, age, calendar year and county of residence to up to 5 comparators from the general population. Through Cox regression we estimated hazard ratios (HRs) and 95% confidence intervals (95%CIs) for death and cause-specific death adjusting for education.ResultsDuring follow-up until December 31, 2017, 1836 (80%) deaths occurred in SBA patients, 1615 (44%) in adenoma, 866 (46%) in NET and 162 (32%) in GIST patients. This corresponded to incidence rates of 295, 74, 80 and 62/1000 person-years respectively and adjusted HRs of 7.60 (95%CI=6.95–8.31), 2.21 (2.07–2.36), 2.74 (2.50–3.01) and 2.33 (1.90–2.87). Adjustment for education had a substantial impact on the HR for death in SBA but not for other neoplasias. The predominant cause of excess death was cancer in all groups.ConclusionThis study confirms earlier findings of increased death rates in patients with SBA and NET in a modern study population. We also demonstrate a more than 2-fold increased risk of death in both GIST and the SBA precursor adenoma.  相似文献

Growth and population density of the larvae, Hynobius nebulosus tokyoensisTago , were estimated in a small pond within the study site settled in Habu village of Hinodemachi, a suburb of Tokyo City, during the period from 1975 to 1980. The mortality factors which influenced the survival rate of larvae were also evaluated from the ecological point of view. Laboratory experiments on the growth of larvae and predation by newts were conducted in pararell with the field survey. The results showed that growth rate of larvae under the natural condition was very slow, as compared with that under the laboratory condition with sufficient food supply, and mean body size at metamorphosis was negatively correlated with the density at that time. This suggested that food resources were in short supply in the pond, and there occurred a severe intraspecific competition for food among larvae. The mortality rate of larvae was so high, 80–99% in each year, and the density of larvae survived until metamorphosis varied so greatly from year to year that the larval stage was the most important stage throughout the life cycle to the maintenance of a population for this salamander. The most important factors which contributed to this high mortality were the predation by the newt, Triturus pyrrhogaster pyrrhogasterBoie , and cannibalism. From the laboratory experiment, it was found that predators could attack only small larvae successfully, and successful attack rate decreased sharply as larvae grew larger. This relationship resulted in the characteristic L-shaped pattern of survivorship curve of larvae; that is, heavy mortality just after hatching period.  相似文献

So far as the population data thus far amassed go, commensal rats in residential habitats are of type II; house mice may be of type I or III in fields but might show type II in residences, and fieral species or subspecies may range from type I to III. Altogether alloresponsive populations seem to be much commoner than the isoresponsive.

It is as yet little disclosed to what degree the differentiation of the type is correlated with speciation or subspeciation, even if only a proof that a subspecific population would hold type III as its fixed feature is given. The response types are supposed to be associated with the levels of population shyness. Hence some characters reflected in those may possibly be connected with sexual isolation provoking speciation.
  3. Some awkward respects due to the heterogeneous trappability, depending upon the trap-response type, among a mixed population of marked and unmarked animals have been confirmed regarding the methodology for estimating parameters.

A record of simulated removal trapping of 50 samples drawn from N=50 under a design has been considered as to how it does contribute to practical censuses of small mammals. It follows that, for ordinary-density populations so far experienced, the removal census based on the theory may frequently lead to quite wrong estimates from single-plot data by the simplest method, and that the maximum likelihood method is doubtless better, but even this method may produce right estimates in about half of the cases, their standard error being of limited availability. Therefore pooling of data from many plots are desirous for such populations.  相似文献

Neuronal loss characterizes many of the most intractable nervous system diseases that deprive our ageing population of their quality of life. Neuroprotective pharmacological modalities are urgently needed to address this burgeoning population. Small ubiquitin-like modifier (SUMO) conjugation has been established as an endogenous neuroprotective response, and we have discovered several classes of small molecules that enhance SUMO conjugation. Herein we describe the hit to lead campaign that enabled the discovery of 3 diverse classes of drug-like SUMOylation activators. Optimized compounds were ultimately validated in cell-based models of neuronal loss and provide a foundation for establishing systemically active SUMO activators to treat degenerative diseases such as Parkinson’s disease, Alzheimer’s disease, and stroke.  相似文献

The endosomal compartment and the plasma membrane form a complex partnership that controls signal transduction and trafficking of different molecules. The specificity and functionality of the early endocytic pathway are regulated by a growing number of Rab GTPases, particularly Rab5. In this study, we demonstrate that IL4 (a Th-2 cytokine) and prostaglandin E2 (PGE2) synergistically induce Rab5 and several Rab effector proteins, including Rin1 and EEA1, and promote the formation of an enlarged early endocytic (EEE) compartment. Endosome enlargement is linked to a substantial induction of the mannose receptor (MR), a well-characterized macrophage endocytic receptor. Both MR levels and MR-mediated endocytosis are enhanced approximately 7-fold. Fluid-phase endocytosis is also elevated in treated cells. Light microscopy and fractionation studies reveal that MR colocalizes predominantly with Rab5a and partially with Rab11, an endosomal recycling pathway marker. Using retroviral expression of Rab5a:S34N, a dominant negative mutant, and siRNA Rab5a silencing, we demonstrate that Rab5a is essential for the large endosome phenotype and for localization of MR in these structures. We speculate that the EEE is maintained by activated Rab5, and that the EEE phenotype is part of some macrophage developmental program such as cell fusion, a characteristic of IL4-stimulated cells.  相似文献

ExoU is a potent virulence factor of Pseudomonas aeruginosa and is considered a potential therapeutic target. In order to discover novel ExoU inhibitors, we screened an in-house chemical library utilizing a yeast-based screening system. Some sulfonamides displayed significant activity without nonspecific cytotoxicity. 目的:探讨腹腔镜下小切口胆囊切除术治疗胆囊结石的临床疗效。方法:选择2009年6月至2014年6月在我院接受胆囊切除术的190例胆囊结石患者,随机分为小切口组和对照组组,分别接受小切口胆囊切除术和常规胆囊切除术。观察并比较两组患者的手术时间、术中出血量、肠道功能恢复时间、下床活动时间、住院时间、住院总费用以及术后并发症的发生情况。结果:小切口组患者的手术时间、术中出血量、肠功能恢复时间、下床活动时间以及住院时间均优于对照组组(t=18.890、16.748、24.650、11.750、36.409、14.084,P=0.000),但住院费用高于对照组组(t=-8.185,P=0.000);小切口组患者术后并发症的发生率显著低于对照组组,差异具有统计学意义(x2=8.360,P=0.004)。结论:腹腔镜下小切口胆囊切除术治疗胆囊结石的临床疗效显著,不仅手术时间短、术中出血量少,而且利于患者术后恢复。  相似文献

Small GTP-binding proteins exist in eukaryotes from yeast to animals to plants and constitute a superfamily whose members function as molecular switches that cycle between “active” and “inactive” states. They regulate a wide variety of cell functions such as signal transduction, cell proliferation, cytoskeletal organization, intracellular membrane trafficking, and gene expression. In yeast and animals, this superfamily is structurally classified into at least five families: the Ras, Rho, Rab, Arf/Sar1, and Ran families. However, plants contain Rab, Rho, Arf, and Ran homologs, but no Ras. Small GTP-binding proteins have become an intensively studied group of regulators not only in yeast and animals but also in plants in recent years. In this article we briefly review the class and structure of small GTP-binding proteins. Their working modes and functions in animals and yeast are listed, and the functions of individual members of these families in plants are discussed, with the emphasis on the recently revealed plant-specific roles of these proteins, including their cross-talk with plant hormones and other signals, regulation of organogenesis (leaf, root, and embryo), polar growth, cell division, and involvement in various stress and defense responses.  相似文献

Background

Ontology-based enrichment analysis aids in the interpretation and understanding of large-scale biological data. Ontologies are hierarchies of biologically relevant groupings. Using ontology annotations, which link ontology classes to biological entities, enrichment analysis methods assess whether there is a significant over or under representation of entities for ontology classes. While many tools exist that run enrichment analysis for protein sets annotated with the Gene Ontology, there are only a few that can be used for small molecules enrichment analysis.

Results

We describe BiNChE, an enrichment analysis tool for small molecules based on the ChEBI Ontology. BiNChE displays an interactive graph that can be exported as a high-resolution image or in network formats. The tool provides plain, weighted and fragment analysis based on either the ChEBI Role Ontology or the ChEBI Structural Ontology.

Conclusions

BiNChE aids in the exploration of large sets of small molecules produced within Metabolomics or other Systems Biology research contexts. The open-source tool provides easy and highly interactive web access to enrichment analysis with the ChEBI ontology tool and is additionally available as a standalone library.

Spleen tyrosine kinase (SYK) is a non-receptor cytosolic kinase. Due to its pivotal role in B cell receptor and Fc-receptor signaling, inhibition of SYK has been targeted in a variety of disease areas. Herein, we report the optimization of a series of potent and selective SYK inhibitors, focusing on improving metabolic stability, pharmacokinetics and hERG inhibition. As a result, we identified 30, which exhibited no hERG activity but unfortunately was poorly absorbed in rats and mice. We also identified a SYK chemical probe, 17, which exhibits excellent potency at SYK, and an adequate rodent PK profile to support in vivo efficacy/PD studies.  相似文献

 本文以日本滋贺县大津市桐生自然保护区的日本赤松(Pinus densiflora)、黑松(Pinus thunbergii)、扁柏(Chamaecypari sobtusa)混交林小流域为研究地,以若女自然保护区内荒山小流域为对照地。通过对两个小流域为期一年的定位观测研究,测定了林外降水、流出水、穿透水、树干流中的pH值、电导率、钾、钠等14种金属元素以及硅、硼非金属元素。研究结果表明,松柏混交林生态系统和花岗岩发育形成的土壤对雨水的pH值和金属元素等均具有较大的影响,但松柏混交林生态系统对其影响大于荒山。此外,系统内大多数元素的收入大于支出,呈积累趋势。  相似文献

利用多年工作积累的观察资料 ,讨论几种植食性小哺乳动物的越冬对策。其中 ,高原鼢鼠、甘肃鼠兔和根田鼠均贮存食物 ,以减少寒冷条件下的取食暴露。高原鼢鼠以个体为单位贮存和利用贮存食物 ,相互之间不协作 ;而两种地面活动的种类则可能以家庭为单位贮存和分享越冬食物。喜马拉雅旱獭体型较大 ,不贮存食物 ,它以冬眠方式越冬 ,这是一种对食物依赖最小的方式。高原鼠兔 ,既不贮存食物 ,也不进入冬眠 ,而是主要靠增加身体产热能力来保持体温 ,抵御严寒。作者认为 ,动物自身的生理限制、生活方式、环境条件以及捕食风险等诸多因素的综合作用决定动物的越冬对策。  相似文献

Renal and small intestinal (re-)absorption contribute to overall phosphate(Pi)-homeostasis. In both epithelia, apical sodium (Na+)/Pi-cotransport across the luminal (brush border) memi brane is rate limiting and the target for physiological/pathophysiological alterations. Three different Na/Pi-cotransporters have been identified: (i) type I cotransporter(s) - present in the proximal tubule - also show anion channel function and may play a role in secretion of organic anions; in the brain, it may serve vesicular glutamate uptake functions; (ii) type II cotransporter(s) seem to serve rather specific epithelial functions; in the renal proximal tubule (type IIa)and in the small intestine (type IIb), isoform determines Na+-dependent transcellular Pi-movements; (iii) type III cotransporters are expressed in many different cells/tissues where they could serve housekeeping functions. In the small intestine, alterations in Pi-absorption and, thus, apical expression of IIb protein are mostly in response to longer term (days) situations (altered Pi-intake, levels of 1.25 (OH2) vitamin D3, growth, etc), whereas in renal proximal tubule, in addition, hormonal effects (e.g. Parathyroid Hormone, PTH) acutely control (minutes/hours) the expression of the IIa cotransporter. The type II Na/Pi-cotransporters operate (as functional monomers) in a 3 Na+:1 Pi stoichiometry, including transfer of negatively charged (-1) empty carriers and electroneutral transfers of partially loaded carriers (1 Na+, slippage)and of the fully loaded carriers (3 Na+, 1 Pi). By a chimera (IIa/IIb) approach, and by site-directed mutagenesis (including cysteine-scanning), specific sequences have been identified contributing to either apical expression, PTH-induced membrane retrieval, Na+-interaction or specific pH-dependence of the IIa and IIb cotransporters. For the COOH-terminal tail of the IIa Na/Pi -cotransporter, several interacting PDZ-domain proteins have been identified which may contribute to either its apical expression (NaPi-Cap1) or to its subapical/lysosomal traffic (NaPi-Cap2).  相似文献

RNA interference (RNAi) is the most exciting insight in biology in past decades, which provided new perspectives into the genome-wide surveys of gene function by targeted degradation of mRNA with the introduction of small interfering RNAs (siRNAs) or small hairpin RNAs (shRNAs) in a large variety of organisms, and turned out to be a more efficient and convenient method compared with the traditional knockout pathway. What's more, as the enhancement of its stability and improvement of its delivery vehicles, RNAi is bound to be a practical tool in determine gene function first in vitro and then in vivo. In this paper, we will focus on the recent achievements of RNAi and also depict the development of RNAi as a potentially powerful tool in studying bone-related diseases.  相似文献

Antibody immobilization on a solid surface is inevitable in the preparation of immunochips/sensors. Antibody-binding proteins such as proteins A and G have been extensively employed to capture antibodies on sensor surfaces with right orientations, maintaining their full functionality. Because of their synthetic versatility and stability, in general, small molecules have more advantages than proteins. Nevertheless, no small molecule has been used for oriented and specific antibody immobilization. Here is described a novel strategy to immobilize an antibody on various sensor surfaces by using a small antibody-binding peptide. The peptide binds specifically to the Fc domain of immunoglobulin G (IgG) and, therefore, affords a properly oriented antibody surface. Surface plasmon resonance analysis indicated that a peptide linked to a gold chip surface through a hydrophilic linker efficiently captured human and rabbit IgGs. Moreover, antibodies captured by the peptide exhibited higher antigen binding capacity compared with randomly immobilized antibodies. Peptide-mediated antibody immobilization was successfully applied on the surfaces of biosensor substrates such as magnetic particles and glass slides. The antibody-binding peptide conjugate introduced in this work is the first small molecule linker that offers a highly stable and specific surface platform for antibody immobilization in immunoassays.  相似文献

Encephalitozoon spp. is an obligate intracellular microsporidian parasite that infects a wide range of mammalian hosts, including humans. This study was conducted to determine the prevalence of Encephalitozoon spp. in wild living rodents from Poland, the Czech Republic and Slovakia. Faecal and spleen samples were collected from individuals of Apodemus agrarius, Apodemus flavicollis, Apodemus sylvaticus, and Myodes glareolus (n = 465) and used for DNA extraction. PCR, targeting the ITS region of the rRNA gene was performed. The overall prevalence of microsporidia was 15.1%. The occurrence of Encephalitozoon cuniculi in the abovementioned host species of rodents has been presented for the first time, with the highest infection rate recorded for A. flavicollis. Sequence analysis showed that the most frequent species was E. cuniculi genotype II (92.5%). E. cuniculi genotypes I (1.5%) and III (6.0%) were also identified.  相似文献
