The structure of the α-keratin microfibril   总被引:3,自引:0,他引:3  
Quantitative measurements of the intensity of the meridional reflections in the X-ray-diffraction pattern of alpha-keratin are shown to be consistent with a microfibril structure in which a surface lattice with an axially projected period around 200 A is subject to a periodic interruption with an axially projected period of 470 A. Taken in conjunction with recent evidence on the chemical structure of alpha-keratin and other intermediate filaments this finding enables an elaboration to be made of a model proposed earlier by RDB Fraser, TP MacRae, & E Suzuki (J. Mol. Biol. 108, 435-452, 1976) for the alpha-helical framework of the microfibril. The disposition and connectivity of the helical segments suggested here provides a straightforward explanation of a number of recent physicochemical and electron-microscopical observations on intermediate filaments and provides a starting point for the development of models for the framework of other intermediate filaments.  相似文献

Broadening of the infrared amide A, amide I and amide II bands of α-helical polypeptides has been observed for thermodynamically unstable α-helices. This spectroscopic fact can be explained now by the geometrical distortions of the backbone of the helical structure. Two models for distorted helices which include regular or irregular distortions of the angles of internal rotation of the main polypeptide chain have been considered. It is pointed out that the instability of α-helix is associated with irregular distortions of the polypeptide backbone.  相似文献

Nicotinic acetylcholine receptors (nAChRs) are involved in fast synaptic transmission in the central and peripheral nervous system. Among the many different types of subunits in nAChRs, the β2 subunit often combines with the α4 subunit to form α4β2 pentameric channels, the most abundant subtype of nAChRs in the brain. Besides computational predictions, there is limited experimental data available on the structure of the β2 subunit. Using high-resolution NMR spectroscopy, we solved the structure of the entire transmembrane domain (TM1234) of the β2 subunit. We found that TM1234 formed a four-helix bundle in the absence of the extracellular and intracellular domains. The structure exhibited many similarities to those previously determined for the Torpedo nAChR and the bacterial ion channel GLIC. We also assessed the influence of the fourth transmembrane helix (TM4) on the rest of the domain. Although secondary structures and tertiary arrangements were similar, the addition of TM4 caused dramatic changes in TM3 dynamics and subtle changes in TM1 and TM2. Taken together, this study suggests that the structures of the transmembrane domains of these proteins are largely shaped by determinants inherent in their sequence, but their dynamics may be sensitive to modulation by tertiary and quaternary contacts.  相似文献

Morphological structure and optical properties of the wings of Morphidae   总被引:2,自引:0,他引:2  
The morphological structure and optical properties of the wings of 14 species of Morphidae have been investigated. Most of the scales of the iridescent species of Morphidae (Lepidoptera) present a very particular structure. The ground scales, responsible for the major part of the optical properties, are covered by a very regular set of longitudinal ridges. The ridges themselves are constituted by a superposition of lamellae that act locally as a multilayered structure. This very specific morphology leads to both interferences and diffraction effects. The first one is responsible of the brilliant blue coloration of the males, while the second one diffracts this colored light at a very large angle. These two phenomena give to the butterfly a very effective long-range communication system. The morphological characteristics of the scales of the various species are presented in detail. Two types of optical measurement were performed on the iridescent wings of 14 different species of Morphidae: spectroscopic measurements under various incidences and gonioscopic measurements for a given incidence angle and wavelength. The first allows a determination of the index of refraction of the cuticular material. The second leads to the drawing of spatial diffraction maps. It shows that most of the reflected light is diffracted laterally over a very large angle (90° 〈 0 〈 120°, according to the different species) and that this repartition depends of the polarization of incident light. As predicted by previous calculations, the dissymmetric structure of the ridge is responsible for the separation of the polarization modes in the various diffraction orders.  相似文献

The amino acid sequence of the major β-chain, IIβ, from rat haemoglobins was established with an automated sequencer. Amino acid heterogeneities were found that appear to result from allelic variation at particular residues. We applied several new or unusual techniques in determining the sequence: (1) reaction of the polypeptide with dansylaziridine for detection of cysteine; (2) blockage of the N-terminal residue and the ε-amino group of lysine residues with 1-fluoro-2-nitro-4-trimethylammoniobenzene iodide and subsequent identification of the modified lysine phenylthiohydantoin by absorbance at 420nm; (3) identification of histidine phenylthiohydantoin by its blue fluorescence under long-wave u.v. light; (4) cleavage of the chain into two or three fragments and subsequent sequencing without purification [a detailed statement giving the major phenylthiohydantoins assigned at each step for each sequence run before their alignment in individual sequences has been deposited as Supplementary Publication SUP 50084 (10 pages) at the British Library Lending Division, Boston Spa, Wetherby, West Yorkshire LS23 7BQ, U.K., from whom copies can be obtained on the terms indicated in Biochem. J. (1978) 169, 5]; (5) separation of fragments produced by CNBr cleavage by cation-exchange chromatography; (6) peptide sequencing after attachment of the peptide to cytochrome c. The amino acid sequence was confirmed by amino acid compositions of the complete chain, of CNBr fragments 1 and 3, and of 11 purified tryptic peptides.  相似文献

In the last few years, three laboratories have reported three entirely different crystallographic models for the L photointermediate of bacteriorhodopsin. All are from X-ray diffraction of illuminated crystals that contain L in photostationary states created at similar cryogenic temperatures. This article compares the models and their implications, the crystallographic statistics and the methods used to derive them, as well as their agreement with non-crystallographic information.  相似文献

The lifetime of the O intermediate of bacteriorhodopsin (BR) is extended by a factor of ~250 in the Leu93‐to‐Ala mutant (BR_L93A). To clarify the structural changes occurring in the last stage of the proton pumping cycle of BR, we crystallized BR_L93A into a hexagonal P622 crystal. Diffraction data from the unphotolyzed state showed that the deletion of three carbon atoms from Leu93 is compensated by the insertion of four water molecules in the cytoplasmic vicinity of retinal. This insertion of water is suggested to be responsible for the blue‐shifted λmax (540 nm) of the mutant. A long‐lived substate of O with a red‐shifted λmax (~565 nm) was trapped when the crystal of BR_L93A was flash‐cooled after illumination with green light. This substate (Oslow) bears considerable similarity to the M intermediate of native BR; that is, it commonly shows deformation of helix C and the FG loop, downward orientation of the side chain of Arg82, and disruption of the Glu194/Glu204 pair. In Oslow, however, the main chain of Lys216 is less distorted and retinal takes on the 13‐cis/15‐syn configuration. Another significant difference is seen in the pH dependence of the structure of the proton release group, the pKa value of which is suggested to be much lower in Oslow than in M. The molecular conformation of proteins is sensitive to the nature of the aqueous environment. In particular, the presence of ions can stabilize or destabilize (denature) protein secondary structure. The underlying mechanisms of these actions are still not fully understood. Here, we combine circular dichroism (CD), single-molecule Förster resonance energy transfer, and atomistic computer simulations to elucidate salt-specific effects on the structure of three peptides with large α-helical propensity. CD indicates a complex ion-specific destabilization of the α-helix that can be rationalized by using a single salt-free computer simulation in combination with the recently introduced scheme of ion-partitioning between nonpolar and polar peptide surfaces. Simulations including salt provide a molecular underpinning of this partitioning concept. Furthermore, our single-molecule Förster resonance energy transfer measurements reveal highly compressed peptide conformations in molar concentrations of NaClO4 in contrast to strong swelling in the presence of GdmCl. The compacted states observed in the presence of NaClO4 originate from a tight ion-backbone network that leads to a highly heterogeneous secondary structure distribution and an overall lower α-helical content that would be estimated from CD. Thus, NaClO4 denatures by inducing a molten globule-like structure that seems completely off-pathway between a fully folded helix and a coil state.  相似文献

Surnames were obtained for the second half of the 20th century from civil and religious marriage registers on fifteen Proven?al-Italian and five Italian villages of Cuneo Province, Italy. To insert in the analysis an outward comparison, surnames from two Italian villages of Turin Province, one parish of Turin, one village of Alessandria Province and one village of Asti Province were also collected. Ethnicity does not seem to be the main factor affecting the present genetic structure of the Proven?al-Italians. They are an open community, and evidence the end of the genetic isolation of the alpine populations.  相似文献

We describe the complete nucleotide sequence of the human ?-globin gene including 387 nucleotides of 5′ flanking sequence and 301 nucleotides of 3′ flanking sequence. The arrangement of coding, noncoding and intervening sequences in this gene is entirely consistent with its identification as the embryonic β-like globin gene.  相似文献

The frequency of occurrence of nearest neighbour residue pairs on adjacent antiparallel (βA) and parallel (βP) strands is obtained from 30 known protein structures. The specificity of interstrand recognition due to such pairing as a factor in the folding of β-sheets is studied by statistical methods. Residues of sufficiently high count for statistical analysis are treated individually while the rest are combined into small groups of similar size, polarity, and/or genetic exchangeability. The hypothesis of specific recognition between individuals and small groups is contrasted with the alternative hypothesis of non-specific recognition between broad classes (hydrophobia, neutral, polar) of residues. A χ2 test of pair correlations favours specific recognition against non-specific recognition with a high level of confidence. The largest and most significant correlations are: Ser/Thr (1.9 ± 0.3), Ile/Val (1.7 ± 0.3) and Lys-Arg/Asp-Gln (1.8 ± 0.3) in βA, and Ile/Leu (1.9 ± 0.4) in βP. The pair Gly/Gly never occurs in any β-sheet. The specific residue-pair correlations derived here may be useful in statistical prediction methods of protein tertiary structure.  相似文献

In order to study the influence of Ser and Thr on the structure of transmembrane helices we have analyzed a database of helix stretches extracted from crystal structures of membrane proteins and an ensemble of model helices generated by molecular dynamics simulations. Both complementary analyses show that Ser and Thr in the g? conformation induce and/or stabilize a structural distortion in the helix backbone. Using quantum mechanical calculations, we have attributed this effect to the electrostatic repulsion between the side chain Oγ atom of Ser and Thr and the backbone carbonyl oxygen at position i ? 3. In order to minimize the repulsive force between these negatively charged oxygens, there is a modest increase of the helix bend angle as well as a local opening of the helix turn preceding Ser/Thr. This small distortion can be amplified through the helix, resulting in a significant displacement of the residues located at the other side of the helix. The crystal structures of aquaporin Z and the β2-adrenergic receptor are used to illustrate these effects. Ser/Thr-induced structural distortions can be implicated in processes as diverse as ligand recognition, protein function and protein folding.  相似文献

Summary The commonest intracellular organelle characteristic of the Phylum Cnidaria or Coelenterata (Subclass Zoantharia) is the spirocyst. Based on scanning and transmission electron microscopy of the tentacles of sea anemones and corals, it appears that the tip of the spirocyst is either exposed to the environment or covered by a thin plasma membrane and often has a pebbled or knobby appearance. Surrounding the spirocyst tip is a ring-like structure which seems to be formed by the junction of the enclosing cell (the spirocyte) and the tip of the spirocyst. The spirocyst thread is continuous with the capsule wall and emerges from within the apical ring during discharge. No ciliary structures appear to be associated with spirocysts. Instead, two different types of microvilli have been found: short microvilli on the spirocyte itself and long microvilli furnished by the cell or cells surrounding the spirocyte. Taxonomic composition and variations in density and biomass of the plankton community in the Öregrundsgrepen, a shallow coastal area, were investigated from June 1972 to November 1973. The phytoplankton biomass was large in spring but small during the rest of the year. The spring bloom was dominated by diatoms and dinoflagellates, especially byThalassiosira spp. which were also important during other seasons. Small forms, such asCryptomonas spp.,Rhodomonas spp. and monads, dominated during summer. Blue-green algae were never of any major importance. During the summer, the trophogenic layer exceeded 10 m in thickness. The metazoan fauna was of lower diversity than the plankton flora. The dominating species, the copepodsAcartia bifilosa andEurytemora affinis, constituted on the average 83% of the standing crop. The low salinities, 5–6 S, were regarded as the principal pertinent limiting factor. The metazoan fauna reached large biomass values from July to October. The protozoan fauna (in the case of ciliates), obtained biomass maxima during the spring bloom. It is suggested that the Öregrundsgrepen represents an area of elevated productivity within a region of low overall production, presumably due to local upwelling. From June 1972 to May 1973, the average biomasses were: phytoplankton 0.464 g C m–2, ciliates 0.040 g C m–2, copepod nauplii 0.010 g C m–2, micro-rotifers 0.004 g C m–2, and mesozooplankton (larger than 0.2 mm) 0.312 g C m–2. It is estimated that about than 60% of the phytoplankton production is consumed by the microzooplankton (<0.2 mm).  相似文献

In this paper, we develop the mathematical structure of the Wright–Fisher model for evolution of the relative frequencies of two alleles at a diploid locus under random genetic drift in a population of fixed size in its simplest form, that is, without mutation or selection. We establish a new concept of a global solution for the diffusion approximation (Fokker–Planck equation), prove its existence and uniqueness and then show how one can easily derive all the essential properties of this random genetic drift process from our solution. Thus, our solution turns out to be superior to the local solution constructed by Kimura.  相似文献
