The aim of this study was to determine liver and kidney concentrations of selenium in wild boars from the northwest part of Poland, depending on season of the year, age, sex, and body weight. Altogether, samples of livers and kidneys from 172 wild boars that were shot in 2005–2008 were investigated. Liver and kidney concentrations of selenium were determined using spectrofluorometric method. In all the animals studied, selenium concentration was several times lower in the liver than in the kidneys. Selenium concentration averaged 0.19 μg/g wet weight (w.w.) in the liver and 1.20 μg/g w.w. in kidneys. The present study showed that season (P ≤ 0.05), age (P ≤ 0.01), and body weight (P ≤ 0.01) have a significant effect on selenium concentration in the liver of wild boars. Liver selenium concentration was the highest in spring (0.23 μg/g w.w.) and the lowest in autumn (0.16 μg/g w.w). Young animals (up to 1 year of age) and those with the lowest body weight (up to 20 kg) were characterized by a slightly lower selenium concentration in the liver compared to older and heavier animals. No significant differences were found in organ selenium concentration between males and females. According to biochemical criteria for the diagnosis of selenium deficiency in pig liver, which were used to evaluate selenium concentration in the liver of wild boars, no individuals were found to have optimal levels. Considering that in Se deficiency higher selenium concentrations are found in kidneys than in the liver, it can be presumed that the wild boars had Se deficiency. However, this is difficult to state conclusively because there are no reference values for this species.  相似文献

The aim of the study was to compare selenium concentrations in different organs of roe deer from northwestern Poland. Samples of liver, kidneys, heart and lungs, collected from 74 roe deer shot during the hunting seasons of 2008–2009 in northwestern Poland, were studied. Selenium concentration in the organs was determined spectrofluorimetrically. Mean selenium concentration was 0.06 μg/g w.w. in the liver, 0.41 μg/g w.w. in the kidneys and 0.05 μg/g w.w. in the heart and lungs. Season had a significant effect on selenium concentration in the liver, kidneys, lungs and heart. In all the organs, the highest selenium concentration was found in spring and the lowest in autumn and winter. All animals studied were deficient in selenium. The low selenium concentration in the liver or heart can disturb their function, and in the future, it may be a factor contributing to the population decline of roe deer in the northwestern part of Poland.  相似文献

This study was conducted to determine the effects of dietary supplementation with Cr nicotinate and Cr chloride and their optimum inclusion rate on performance, carcass traits, meat oxidative stability, serum metabolites, hematological parameters, and liver chromium concentration in heat-stressed broilers. A total number of 420, 1-day-old male broiler chicks were randomly assigned to seven treatments with four replicates of 15 chicks. The dietary treatments consisted of the basal diet supplemented with 0 (control), 500, 1,000, and 1,500 μg/kg Cr in the form of Cr nicotinate and Cr chloride. Chicks were raised for 6 weeks in heat stress condition (33 ± 2°C). Supplements of organic and inorganic Cr particularly at 1,500 μg/kg incorporation increased feed consumption (P < 0.05) and body mass gain of broilers (P < 0.01). Cr supplementation increased carcass yield and decreased abdominal fat (P < 0.01). Supplementation of 1,500 μg/kg Cr nicotinate (P < 0.05) enhanced liver Cr concentration. Storage time increased lipid oxidation of meat (P < 0.01). Cr decreased lipid oxidation of breast and thigh muscles over 2 (P < 0.01) or 6 (P < 0.05) days of storage time. Birds fed 1,500 μg/kg Cr nicotinate, had lower concentration of serum glucose and triglyceride at 21 days (P < 0.05). Hematological parameters tested at 21 and 42 days, were not influenced. The results suggested that dietary Cr supplementation regardless of its source have a positive effect on productive, and carcass traits, also enhances oxidative stability of refrigerated meat in broilers reared under heat stress conditions.  相似文献

The purpose of this study was to determine glutathione peroxidase (GSHPx) activity in the liver of red deer in relation to selenium concentrations in the liver, as well as to evaluate changes in GSHPx activity according to sex, body weight and season of the year. Total selenium concentration in the liver of red deer averaged 0.095?±?0.018?μg/g of wet weight. GSHPx activity in the liver of red deer ranged widely from 4.4 to 45.8?U/g of protein. Females were characterized by higher GSHPx activity compared to males (21.2 vs. 17.0?U/g protein). The highest GSHPx activity was recorded in autumn and the lowest in summer. The lowest GSHPx activity in the liver was found in the heaviest animals (>100?kg body weight), averaging 14.0?U/g protein. Animals weighing <66?kg and 66-100?kg were characterized by similar activity of 25.1 and 24.5?U/g, respectively. Despite the differences in GSHPx activity according to sex, body weight and season of the year, these factors had no significant effect on the activity of this enzyme. The main factor regulating GSHPx activity in the liver of examined red deer was selenium concentration.  相似文献

To clarify how tributyltin (TBT) and triphenyltin (TPT) interact with the retinoid X receptor (RXR) to induce growth of male sex organs in female gastropods, we treated female rock shells (Thais clavigera) with three different concentrations (0.1, 1, or 5 μg/g wet wt) of 9-cis-retinoic acid (9CRA) or with a single concentration (1 μg/g wet wt) of TBT, TPT, or fetal bovine serum (as a control). The effects of each treatment were measured as the incidence of imposex, the length of the penis-like structure, and the vas deferens sequence (VDS) index. 9CRA induced imposex in a dose-dependent manner; imposex incidence was significantly higher in the rock shells that received 1 (P < 0.05) or 5 μg (P < 0.001) 9CRA than in the controls. After 1 month, the rock shells treated with 5 μg 9CRA exhibited substantial growth of the penis-like structure that was not as evident in the other treated shells. The length of the structure differed between the 0.1- and 5-μg 9CRA treatment groups (P < 0.05) but not between the 1- and 5-μg 9CRA treatment groups (P > 0.05). Compared with the control, the VDS index increased significantly in the 1- (P < 0.05) and 5-μg (P < 0.001) 9CRA groups. The penis-like structures behind the right tentacle in female rock shells treated with 5 μg 9CRA were essentially the same as the penises and vasa deferentia of normal males and of TBT-treated or TPT-treated imposexed females. These results further support the hypothesis that imposex in gastropods could be mediated by RXR.  相似文献

This longitudinal study addresses the relationship of different husbandry systems to fecal cortisol metabolites (FCM) concentrations in captive brown brocket deer and marsh deer in order to ascertain a less stressful captive condition for these species. Thus, three pairs from both species were submitted to three different husbandry systems (10 days per system), and fecal samples were collected in the last 5 days of each management. A cortisol enzyme immunoassay was validated and used to measure FCM. The physiological significance of these measurements was verified in brown brocket deer by an adrenocorticotropic hormone challenge test, which induced a fourfold (above baseline) increase of FCM within 24 to 28 h. In marsh deer, wild-capture individuals (2,802 ± 115 ng/g) had significantly higher concentrations (P < 0.05) than captive-born ones (122 ± 32 ng/g). Higher values of FCM (P < 0.05) were measured in brown brocket deer kept as pairs in outdoor exhibits all day (150 ± 15 ng/g) compared to animals kept integrally in individual stalls (71 ± 7 ng/g) or in outdoor exhibits during daytime and individual stalls during nighttime (78 ± 4 ng/g). In contrast, no differences in FCM were found in marsh deer submitted to any husbandry system (P > 0.05). These findings suggest that brown brocket deer may benefit from husbandry systems, which keep animals at least for one period in individual stalls, while marsh deer apparently exhibit a high individual variability to different husbandry conditions with no generic pattern to its better maintenance in captivity.  相似文献

The effect of various supplementary feeds on bark browsing was studied in red deer under controlled conditions. Fifteen female red deer were randomly assigned to five dietary treatments (n = 3 animals each): meadow hay as a control (C), combinations of meadow hay with grass silage (GS), maize silage (MS), maize silage with oats (MSO) or a pelleted concentrate (PC). The deer were kept in enclosures where we offered fresh spruce stems daily. The experiment consisted of two periods. In the first period (A), the animals had free access to food during 24 h, while in the second period (B), the access was restricted to night hours only. In both periods, the supplementary feeds were available ad libitum. Mean daily consumption of browsed bark in the control group in period A was 1,053 g while in the remaining groups it ranged from six to 20 g. In period B, simulating the conditions in disturbed hunting areas, bark consumption in all groups increased considerably, the highest increase being found in groups GS (430 vs. 19 g/day) and MS (101 vs. 6 g/day). In period A, the highest dry matter (DM) intake including browse was found in group MS (2,816 g/day) and the lowest one in group GS (2,307 g/day). In period B, the highest (2,763 g/day) and the lowest (2,153 g/day) DM intake was observed in groups PC and MSO, respectively. If red deer is fed supplementary, meadow hay should be combined with maize silage, grass silage or concentrate to reduce bark browsing in forests. The bark browsing rate may increase considerably if red deer are disturbed or their grazing cycle is impaired.  相似文献

The trace element selenium is an essential micronutrient for human health and its low levels in serum are implicated in the pathogenesis of several chronic diseases. Therefore, the determination of total selenium in serum may contribute to the assessment of the health and nutritional status of certain populations. The objective of the present work was to determine total selenium in the serum of 506 healthy volunteers that participated in the ATTICA study. Selenium was determined in serum by using the technique of inductively coupled plasma mass spectrometry. The mean serum selenium concentration was determined to be 91.8 ± 33.7 μg/L (N = 506); 87.6% of women and 88.5% of men had serum selenium concentration below 125 μg/L, the cutoff considered to be required for optimal glutathione peroxidase activity. No association was found between serum selenium levels and the gender of the participants while a significant decline of selenium with age (p < 0.0001) was observed. According to our results, no anthropometric, lifestyle, nutritional, or biochemical indices were able to affect the association between serum selenium and age. This result may indicate that other factors such as selenium distribution as well as retention may be affecting the relationship between serum selenium and age.  相似文献

Temporal and spatial distributions of dissolved organic carbon (DOC), dissolved organic nitrogen (DON), chlorophyll-a and inorganic nitrogen were investigated in two small mountainous lakes (Lake Hongfeng and Baihua), on the Southwestern China Plateau, based on almost 2 years’ field observation. DOC concentrations ranged from 163 μM to 248 μM in Lake Hongfeng and from 143 μM to 308 μM in Lake Baihua, respectively, during the study period. DON concentrations ranged from 7 μM to 26 μM in Lake Hongfeng and from 14 μM to 47 μM in Lake Baihua. DOC showed vertical heterogeneity with higher concentrations in the epilimnion than in the hypolimnion during the stratification period. The DON concentration profiles appeared to be more variable than the DOC profiles. Apparent DON maxima occurred in the upper layer of water. In Lake Hongfeng, DOC concentration in the surface water was highest at the end of spring and early summer. DON concentration was 2–5 μM higher in May 2003 and in June 2004 than in adjacent months. DOC and chlorophyll-a concentrations were significantly correlated (r = 0.79, P < 0.05). The period of highest concentrations of DOC in Lake Hongfeng was also the season of concentrated rainfall. Algae activity and allochthonous input might result in an increase of DOC and DON concentrations together. In Lake Baihua, the maximum concentrations of DOC and DON in the surface water occurred simultaneously in May 2003 and February 2004. DOC concentrations were significantly correlated with DON (r = 0.90, P < 0.01), indicating the common sources. Allochthonous input, biological processes, stratification and mixing were the most important factors controlling the distributions and cycling of dissolved organic matter (DOM) and inorganic nitrogen in these two lakes. Inference from the corresponding vertical distributions of DOM and inorganic nitrogen indicated that DOM played potential roles in the internal loading of nitrogen and metabolism in the water body in these small lakes. The carbon/nitrogen (C/N) ratio showed a potential significance for tracing the source and biogeochemical processes of DOM in the lakes. These results are of significance in the further understanding of biogeochemical cycling and environmental effects of DOM and nitrogen in lake ecosystems.  相似文献

The study was conducted to investigate the effects of dietary maternal selenomethionine or sodium selenite supplementation on performance and selenium status of broiler breeders and their next generation. Two hundred and forty 39-week-old Lingnan yellow broiler breeders were allocated randomly into two treatments, each of which included three replicates of 40 birds. Pretreatment period was 2 weeks, and the experiment lasted 8 weeks. The groups were fed the same basal diet supplemented with 0.30 mg selenium/kg of sodium selenite or selenomethionine. After incubation, 180 chicks from the same parental treatment group were randomly divided into three replicates, with 60 birds per replicate. All the offspring were fed the same diet containing 0.04 mg selenium/kg, and the experiment also lasted 8 weeks. Birth rate was greater (p < 0.05) in hens fed with selenomethionine than that in hens fed with sodium selenite. The selenium concentration in serum, liver, kidney, and breast muscle of broiler breeders, selenium deposition in the yolk, and albumen and tissues' (liver, kidney, breast muscle) selenium concentrations of 1-day-old chicks were significantly (p < 0.01) increased by maternal selenomethionine supplementation compared with maternal sodium selenite supplementation. The antioxidant status of 1-day-old chicks was greatly improved by maternal selenomethionine intake in comparison with maternal sodium selenite intake and was evidenced by the increased glutathione peroxidase activity in breast muscle (p < 0.05), superoxide dismutase activity in breast muscle and kidney (p < 0.05), glutathione concentration in kidney (p < 0.01), total antioxidant capability in breast muscle and liver (p < 0.05), and decreased malondialdehyde concentration in liver and pancreas (p < 0.05) of 1-day-old chicks. Feed utilization was better (p < 0.05), and mortality was lower (p < 0.05) in the progeny from hens fed with selenomethionine throughout the 8-week growing period compared with those from hens fed with sodium selenite. In summary, we concluded that maternal selenomethionine supplementation increased birth rate and Se deposition in serum and tissues of broiler breeders as well as in egg yolk and egg albumen more than maternal sodium selenite supplementation. Furthermore, maternal selenomethionine intake was also superior to maternal sodium selenite intake in improving the tissues Se deposition and antioxidant status of 1-day-old chicks and increasing the performance of the progeny during 8 weeks of post-hatch life.  相似文献

This experiment was carried out to investigate the effects of different levels of organic and inorganic chromium on the performance, immune function and some serum mineral concentrations of broilers under heat stress condition (23.9–37°C cycling). A total of 150 one-day-old broiler chicks according to a completely randomized design were assigned into five treatment groups. Each treatment consisted of three replicates and each replicate contained ten chicks. Chicks were fed on corn–soybean meal basal diets with added different concentrations of chromium (0, 600 and 1,200 μg kg−1 chromium chloride or 600 and 1,200 μg/kg chromium l-methionine) from 1 to 49 days of age. Humoral immunity was assessed by intravenous injection of 7% sheep red blood cell (SRBC) followed by evaluation of serum for antibody titers in primary and secondary responses. Cell-mediated immunity was assessed by the cutaneous basophil hypersensitivity (CBH) test to phytohemagglutinin (PHA)-P at day 32 and PHA-M at day 48. Heterophil/lymphocyte (H/L) ratio was also measured as a reliable indicator of stress. The body mass, feed intake and conversion ratio were not influenced by dietary chromium (P > 0.05). Dietary supplementation of both organic and inorganic chromium significantly increased primary and secondary antibody responses (P < 0.01), and also improved H/L ratio (P < 0.05), CBH response (P < 0.01) as well as relative weights of thymus (P < 0.05) and spleen (P < 0.01). Both dietary organic and inorganic chromium caused an increase in serum concentrations of Cr and Zn (P < 0.01), but decreased the serum concentration of Cu (P < 0.01). These results suggest that supplemental chromium especially in organic form offers a good management practice to reduce heat stress-related depression in immunocompetence of broiler chicks.  相似文献

Our previous studies have shown that selenium (Se) is protective against dimethylhydrazine (DMH)-induced preneoplastic colon cancer lesions, and protection against DNA damage has been hypothesized to be one mechanism for the anticancer effect of Se. The present study was designed to determine whether dietary selenite affects somatic mutation frequency in vivo. We used the Big Blue transgenic model to evaluate the in vivo mutation frequency of the cII gene in rats fed either a Se-deficient (0 μg Se/g diet) or Se-supplemented diet (0.2 or 2 μg Se/g diet; n = 3 rats/diet in experiment 1 and n = 5 rats/group in experiment 2) and injected with DMH (25 mg/kg body weight, i.p.). There were no significant differences in body weight between the Se-deficient and Se-supplemented (0.2 or 2 μg Se/g diet) rats, but the activities of liver glutathione peroxidase and thioredoxin reductase and concentration of liver Se were significantly lower (p < 0.0001) in Se-deficient rats compared to rats supplemented with Se. We found no effect of dietary Se on liver 8-hydroxy-2′-deoxyguanosine. Gene mutation frequency was significantly lower in liver (p < 0.001) than that of colon regardless of dietary Se. However, there were no differences in gene mutation frequency in DNA from colon mucosa or liver from rats fed the Se-deficient diet compared to those fed the Se-supplemented (0.2 or 2 μg Se/g diet) diet. Although gene mutations have been implicated in the etiology of cancer, our data suggest that decreasing gene mutation is not likely a key mechanism through which dietary selenite exerts its anticancer action against DMH-induced preneoplastic colon cancer lesions in a Big Blue transgenic rat model. A total of 32 wild Hokkaido sika deer (Cervus nippon yesoensis) were shot (13 in summer, nine in autumn and 10 in winter) in the Syari district, Shiretoko Peninsula of Hokkaido Island, Japan. The ingested foods, rumen fermentation parameters and microbes were determined to evaluate digestive strategy and food availability in each season. Ingested foods and ruminal characteristics greatly varied by season. Rumen digesta mainly comprised of graminoids in summer, graminoids and agricultural products in autumn, and bark and twigs in winter. Rumen pH showed seasonal differences (P<0.05) and was lowest in summer, highest in winter, and intermediate in autumn, reflecting the seasonal differences in ruminal concentration of total volatile fatty acids which were significantly lower (P<0.05) in winter than in summer and autumn. Acetate proportions were significantly higher in winter than in other seasons (P<0.05), while the opposite trend was seen in proportions of propionate and butyrate. Rumen ammonia levels showed significant seasonal differences (P<0.05), decreasing from summer to autumn to winter. Rumen protozoa levels in autumn and winter decreased to 28 and 10% of the levels observed in summer, respectively (P<0.05 for both). The rumen bacteria level in winter was lower (P<0.05) than that in autumn, but no difference was seen for the other seasonal comparisons. Gram negative cocci were present in significantly higher proportions in winter than in other seasons (P<0.05), while Gram negative curved rods were less frequently observed in winter (P<0.05). Based on these results, wild sika deer in this area are shown to survive with rumen microbial populations altered with the dietary conditions that vary greatly by season.  相似文献

The essential nutrient selenium is required in microgram amounts [recommended dietary allowance (RDA) = 55 μg/day, 699 nmol/day] and has a narrow margin of safety (upper tolerable intake limit = 400 μg/day, 5 μmol/day). We conducted a randomized placebo-controlled study of high-selenium yeast, the form used in most supplements (300 μg/day, 3.8 μmol/day), administered to 42 free-living healthy men for 48 weeks. Dietary intakes of selenium, macronutrients, and micronutrients were not different between groups and did not change during the study. Supplementation more than doubled urinary selenium excretion from 69 to 160 μg/day (876 to 2,032 nmol/day). Urinary excretion was correlated with recent selenium intake estimated from 3-day diet records: urinary selenium excretion = 42 μg/day (533 nmol/day) + 0.132 × dietary selenium intake, p < 0.001. Dietary selenium intake was not significantly correlated with the other indicators of selenium status, presumably because urinary selenium excretion reflected recent intake, and tissue selenium was homeostatically controlled. After 48 weeks of supplementation, plasma selenium was increased 60% from 142 to 228 μg/l (1.8 to 2.9 μmol/l), and erythrocyte selenium was approximately doubled from 261 to 524 μg/l (3.3 to 6.6 μmol/l). Selenium concentrations increased more modestly in hair (56%) and platelets (42%). Platelets were the only blood component in which glutathione peroxidase activity was significantly related to selenium content. Selenium levels decreased rapidly after the end of supplementation, and there were no significant differences in selenium status indicators between groups by week 96. The absorption, distribution, and excretion of selenium from high-Se yeast were similar to selenium in foods.  相似文献

Diabetes is a chronic health problem and major cause of death in most of the countries. Diet management plays an important role in controlling diabetes and its complications along with insulin and drugs. We have examined the effect of banana (Musa sp. var. elakki bale) flower and pseudostem on hyperglycemia and advanced glycation end-products (AGEs) in streptozotocin-induced diabetic rats. Our results indicated that banana flower and pseudostem have low glycemic index and have a high content of dietary fiber and antioxidants. Diabetic symptoms like hyperglycemia, polyuria, polyphagia, polydipsia, urine sugar, and body weight were ameliorated in banana flower- and pseudostem-treated rats. Increased glomerular filtration rate in the diabetic group (5.1 ± 0.22 ml/min) was decreased in banana flower-fed (2.5 ± 0.37 ml/min) and pseudostem-fed (3.0 ± 0.45 ml/min) groups and were significant at P < 0.001 and P < 0.01, respectively. Fructosamine and AGEs formed during diabetes were inhibited in treated groups when compared with the diabetic group. The diabetic group showed 11.5 ± 0.64 μg of AGEs/mg protein in kidney, whereas, in banana flower- and pseudostem-fed groups, it was reduced to 9.21 ± 0.32 and 9.29 ± 0.24 μg/mg protein, respectively, and were significant at P < 0.01. These findings suggest that banana flower and pseudostem have anti-diabetic and anti-AGEs properties and are beneficial as food supplements for diabetics.  相似文献

Premature graying of hair with unclear etiology, which is known as premature canities, is a common cause of referrals to the dermatologists. We assessed the relationship between serum iron, copper, and zinc concentrations with premature canities. This study was conducted on patients under 20 years old suffering from premature canities, having a minimum of ten gray hair fibers, and referring to university hospitals of Isfahan (Iran). The results were compared with age–sex-matched controls. Demographic data and disease characteristics were recorded for two groups. We studied serum iron, copper, and zinc concentrations of 66 patients and 66 controls using atomic absorption and Ferrozine methods. The mean age of studied cases was 17.8 ± 2.0 years, and the mean age of the onset of canities was 15.5 ± 3.2 years with no significant difference between males and females (P > 0.05). Serum copper concentration was significantly lower in patients compared with controls (90.7 ± 37.4 vs. 105.3 ± 50.2 μg/dL, P = 0.048), but serum iron concentration was significantly lower in controls compared to patients (88.8 ± 39.5 vs. 108.3 ± 48.4 μg/dL, P = 0.008). Also, there was no significant difference between patients and controls in serum zinc concentration (114.8 ± 67.8 vs. 108.2 ± 49.9 μg/dL, P = 0.285). According to these results, among copper, zinc, and iron, a low serum copper concentration may play a role in premature graying of hairs in our society. Further studies are needed to find the underlying mechanism of this relationship.  相似文献

Patients with chronic kidney disease (CKD) have an increased incidence of cancer. It is well known that long periods of hemodialysis (HD) treatment are linked to DNA damage due to oxidative stress. In this study, we examined the effect of selenium (Se) supplementation to CKD patients on HD on the prevention of oxidative DNA damage in white blood cells. Blood samples were drawn from 42 CKD patients on HD (at the beginning of the study and after 1 and 3 months) and from 30 healthy controls. Twenty-two patients were supplemented with 200 μg Se (as Se-rich yeast) per day and 20 with placebo (baker's yeast) for 3 months. Se concentration in plasma and DNA damage in white blood cells expressed as the tail moment, including single-strand breaks (SSB) and oxidative bases lesion in DNA, using formamidopyrimidine glycosylase (FPG), were measured. Se concentration in patients was significantly lower than in healthy subjects (P < 0.0001) and increased significantly after 3 months of Se supplementation (P < 0.0001). Tail moment (SSB) in patients before the study was three times higher than in healthy subjects (P < 0.01). After 3 months of Se supplementation, it decreased significantly (P < 0.01) and was about 16% lower than in healthy subjects. The oxidative bases lesion in DNA (tail moment, FPG) of HD patients at the beginning of the study was significantly higher (P < 0.01) compared with controls, and 3 months after Se supplementation it was 2.6 times lower than in controls (P < 0.01). No changes in tail moment was observed in the placebo group. In conclusion, our study shows that in CKD patients on HD, DNA damage in white blood cells is higher than in healthy controls, and Se supplementation prevents the damage of DNA.  相似文献

To investigate whether sodium selenate treatment would impact on the onset of diabetic nephropathy, we examined blood glucose, serum biochemical components, and interrelationship between oxidative stress, TGF-β1, and apoptosis in streptozotocin (STZ) induced diabetic rats. Sixty male Wistar rats were divided into six groups. Group I (n = 10), normal control; Group II (n = 10), diabetic control; Group III (n = 10), sodium selenate (16 μmoles/kg) + diabetic; Group IV (n = 10), sodium selenate (32 μmoles/kg) + diabetic; Group V (n = 10), sodium selenate (16 μmoles/kg) control; and Group VI (n = 10), sodium selenate (32 μmoles/kg) control. Sodium selenate was administered via orogastric route for 10 weeks. In the diabetic group, diabetes was induced by single intraperitoneal injection of STZ (50 mg/kg). The levels of blood glucose were estimated and total cholesterol, high-density lipoprotein (HDL) cholesterol, triglycerides, creatinine, urea, and albumin were detected in serum. Antioxidant status was examined by measuring the superoxide dismutase (SOD), catalase, glutathione, and lipid peroxidation in kidney tissues. Histopathological studies were performed in the kidney tissue sections. The expression of TGF-β1 was estimated by the immunohistochemical analysis in kidneys. Apoptotic study in kidney was performed using the TdT-mediated dUTP nick end labeling technique. It was observed that blood glucose, serum, total cholesterol, HDL cholesterol, triglycerides, creatinine, urea, and albumin were significantly higher in diabetic control groups. Diabetic + sodium selenate (16 and 32 μmoles/kg) significantly reduced blood glucose, serum, total cholesterol, HDL cholesterol, triglycerides, creatinine, urea, and albumin levels. Selenium-treated groups significantly increased antioxidant enzyme activities (SOD, catalase, and glutathione) in kidneys of diabetic rats. All enzyme activities of selenium control groups did not differ compared with the normal control. Sodium selenate reduces significantly lipid peroxidation in diabetic rats. Cellular architecture of the diabetic rats was altered whereas sodium selenate administration rectifies the degenerative changes of the kidney. Profound immunopositivity of TGF-β1 was observed in the glomerular and tubulointerstitial cells of diabetic rat kidney. Immunopositivity of TGF-β1 was significantly reduced in both low and high dose of sodium-selenate-treated rats (P < 0.05, P < 0.01). High numbers of apoptotic cells were observed in diabetic rats whereas sodium selenate in both doses significantly reduces the incidence of apoptosis (P < 0.05, P < 0.01). We conclude herein that sodium selenate has the potential to play a significant role in limiting the renal impairment by altering the apoptosis and TGF-β1 in experimental diabetic rats.  相似文献

The experiment was conducted to study the effects of different selenium (Se) sources on productive performance, serum and milk Se concentrations, and antioxidant status of sows. A total of 12 sows (Landrace×Yorkshire) with same pregnancy were randomly divided into two groups; each group was replicated six times. These two groups received the same basal gestation and lactation diets containing 0.042 mg Se/kg, supplemented with 0.3 mg Se/kg sodium selenite or selenomethionine (i.e., seneno-dl-methylseleno), respectively. The feeding trial lasted for 60 days, with 32 and 28 days for gestation and lactation period, respectively. Compared with sodium selenite, maternal selenomethionine intake significantly increased (P < 0.05) the weaning litter weight and average weight of piglet. The Se concentration in the serum, colostrum, and milk of sows were significantly higher (p < 0.05) in the selenomethionine-treated group. The antioxidant status was greatly improved in sows of selenomethionine-treated group and was illuminated by the increased total antioxidant capability (T-AOC; P < 0.05) and decreased malondialdehyde (MDA; P < 0.01) level in the serum of sows, increased T-AOC (P < 0.05), glutathione (GSH) peroxidase (P < 0.05), superoxide dismutase (P < 0.05) and GSH (P < 0.05), and MDA (P < 0.05) level in the colostrum and milk of sows. These results suggested that maternal selenomethionine intake improved Se concentration and antioxidant status of sows, thus maintain maternal health and increase productive performance after Se was transferred to its offspring.  相似文献
