Methane (CH4) and nitrous oxide (N2O) dynamics were studied in a boreal Sphagnum fuscum pine bog receiving annually (from 1991 to 1996) 30 or 100 kg NH4NO3-N ha–1. The gas emissions were measured during the last three growing seasons of the experiment. Nitrogen treatment did not affect the CH4 fluxes in the microsites where S. fuscum and S. angustifolium dominated. However, addition of 100 kg NH4NO3-N ha–1 yr–1 increased the CH4 emission from those microsites dominated by S. fuscum. This increase was associated with the increase in coverage of cotton grass (Eriophorum vaginatum) induced by the nitrogen treatment. The differences in the CH4 emissions were not related to the CH4 oxidation and production potentials in the peat profiles. The N2O fluxes were negligible from all microsites. Only minor short-term increases occurred after the nitrogen addition.  相似文献

Anabaena doliolum and Chlorella vulgaris immobilized on chitosan were more efficient at removing NO3 , NO2 p–, PO4 3– and CR2O7 2– from wastewaters than cells immobilized on agar, alginate, carrageenan or even free cells. Carrageenan-immobilized cells, however, were better at removing NH4 + and Ni2+. The PO4 3– uptake capacity was significantly increased in cells starved of PO4 3– for 24 h. Agar-immobilized cells, though having good metal and nutrient uptake efficiency, had only a slow growth rate. The best culture medium composition for the production of bikaverin by Gibberella fujikuroi in shake-flasks, i.e. 100 g glucose l–1; 1 g NH4Cl l–1; 2 g rice flour l–1; 5 g KH2PO4 l–1 and 2.5 g MgSO4 l–1, was obtained through a fractional factorial design and then scaled-up to a fluidized bioreactor. The effects of carbon and nitrogen concentrations, inoculum size, aeration, flow rate and bead sizes on batch bikaverin production using immobilized G. fujikuroi in a fluidized bioreactor were determined by an orthogonal experimental design. Concentrations of up to 6.83 g bikaverin l–1 were obtained when the medium contained 100 g glucose l–1 and 1 g NH4Cl l–1 with an inoculum ratio of 10% v/v, an aeration rate of 3 volumes of air per volume of medium min–1, and a bead size of 3 mm. Based on dry weight, the bikaverin production was 30–100 times larger than found in submerged culture and approximately three times larger than reported for solid substrate fermentation.  相似文献

The dynamics of inorganic N are important in soil, and this applies particularly to the saline–alkaline soils of the former lake Texcoco in Mexico with high pH and salinity where a forestation program was started in the 1970s. In soils of lake Texcoco, in Mexico, more than 50% of applied N could not be accounted for one day after application of 200 mg kg–1 soil along with glucose amendment. It was not clear whether this was due to abiotic or biotic processes, the form of inorganic N applied or the result of applying an easily decomposable substrate. We investigated this by adding glucose and 200 mg kg–1 soil as (NH4)2SO4-N or KNO3-N to sterilized and unsterilized soil. The changes in inorganic and ninhydrin N, microbial biomass C and production of CO2 were then monitored. Between the time of applying N and extraction with 0.5 M K2SO4, i.e., after ca 2 h, approximately 110 mg NH4 +-N kg–1 dry soil could not be accounted for in the unsterilized and sterilized soil and that remained so for the entire incubation in the sterilized soil. After 1 day this increased to 140 mg NH4 +-N kg–1 dry soil in the unsterilized control and 170 mg NH4 +-N kg–1 dry soil in C amended soil. Volatilization of NH3 accounted for 56 mg NH4 +-N kg–1 so the rest appeared to be adsorbed on the soil matrix. The NH3 volatilization and NH4 + fixed in the soil matrix remained constant over time and no oxidation to NO2 or NO3 had occurred, so unaccounted N in unsterilized soil was probably incorporated into the microbial biomass in excess of what was required for metabolic activity. The unaccounted N was ca 70 mg NO3 –N in nitrate amended soil after 3 days and 138 NO3 –N when glucose was additionally added. Losses through abiotic processes were absent as inferred from changes in sterilized soil and the aerobic incubation inhibited possible losses through denitrification. It was inferred that NO3 that could not be accounted for was taken up by micro-organisms in excess of what was required for metabolic activity.  相似文献

In short-term water culture experiments with different 15N labeled ammonium or nitrate concentrations, citrus seedlings absorbed NH4 + at a higher rate than NO3 . Maximum NO3 uptake by the whole plant occurred at 120 mg L–1 NO3 -N, whereas NH4 + absorption was saturated at 240 mg L–1 NH4 +-N. 15NH4 + accumulated in roots and to a lesser degree in both leaves and stems. However, 15NO3 was mostly partitioned between leaves and roots.Adding increasing amounts of unlabeled NH4 + (15–60 mg L–1 N) to nutrient solutions containing 120 mg L–1 N as 15N labeled nitrate reduced 15NO3 uptake. Maximum inhibition of 15NO3 uptake was about 55% at 2.14 mM NH4 + (30 mg L–1 NH4 +-N) and it did not increase any further at higher NH4 + proportions.In a long-term experiment, the effects of concentration and source of added N (NO3 or NH4 +) on nutrient concentrations in leaves from plants grown in sand were evaluated. Leaf concentration of N, P, Mg, Fe and Cu were increased by NH4 + versus NO3 nutrition, whereas the reverse was true for Ca, K, Zn and Mn.The effects of different NO3 -N:NH4 +-N ratios (100:0, 75:25, 50:50, 25:75 and 0:100) at 120 mg L–1 total N on leaf nutrient concentrations, fruit yield and fruit characteristics were investigated in another long-term experiment with plants grown in sand cultures. Nitrogen concentrations in leaves were highest when plants were provided with either NO3 or NH4 + as a sole source of N. Lowest N concentration in leaves was found with a 75:25 NO3 -N/NH4 +-N ratio. With increasing proportions of NH4 + in the N supply, leaf nutrients such as P, Mg, Fe and Cu increased, whereas Ca, K, Mn and Zn decreased. Yield in number of fruits per tree was increased significantly by supplying all N as NH4 +, although fruit weight was reduced. The number of fruits per tree was lowest with the 75:25 NO3 -N:NH4 +-N ratio, but in this treatment fruits reached their highest weight. Rind thickness, juice acidity, and colour index of fruits decreased with increasing NH4 + in the N supply, whereas the % pulp and maturity index increased. Percent of juice in fruits and total soluble solids were only slightly affected by NO3 :NH4 + ratio.  相似文献

A laboratory microcosm experiment was established to study whether the role of Cognettia sphagnetorum (Enchytraeidae) in affecting Scots pine (Pinus sylvestris) seedling growth is influenced by wood ash-amendment, i.e., neutralisation of the raw humus soil. Coniferous forest soil, inoculated with soil microbes and nematodes, was either treated with wood ash or left as ash-free control. Wood ash (corresponding to an amount of 5000 kg ha–1) was either spread on the soil surface or mixed into the soil. Enchytraeid and pine seedling biomass, abundance of nematodes, and water leachable NH4 +-N and NO3 -N were measured 26 and 51 weeks after initiation of the experiment and root length and N concentration of needles were measured 51 weeks after initiation of the experiment. Wood ash when mixed into the soil, reduced the biomass of C. sphagnetorum. Nematodes were unaffected by the treatments. In the ash-free soils C. sphagnetorum had little influence on pine growth, but it did decrease root length and root to shoot ratio. In the absence of enchytraeids wood ash decreased pine biomass production and root length. However, the presence of enchytraeids in the ash-treated soil compensated the ash-induced negative effects on the pine performance. Enchytraeids increased and wood ash decreased water leachable NH4 +-N in the presence but not in the absence of enchytraeids, while water leachable NO3 -N was not affected by the treatments. It was concluded that C. sphagnetorum can be important in ensuring nutrient cycling and plant growth in situations when an ecosystem encounters disturbances.  相似文献

A survey was conducted over a range of 17 Dutch heathland locations, subdivided into 41 sites dominated by either dwarf-shrubs (Calluna vulgaris or Erica tetralix) or grass species (Deschampsia flexuosa or Molinia caerulea). Among the habitats of the dominant plant species relatively little differences in general soil properties were observed. The P status of Deschampsia sites was relatively high as well as the NO3 -N concentrations in the 0–10 cm layer (FH included) at the grass-dominated sites. At sites with a dead or degenerating dwarf-shrub vegetation, NH4 +-N concentrations reached very high levels.Net production of nitrate was observed during incubation of intact 0–10 cm soil cores (FH-layer included) in the laboratory for all sites, even though in some instances, particularly at Calluna and Erica sites, no nitrate was initially measured. Generally, a higher nitrification rate was found for the grass-dominated sites, and for Deschampsia in particular. The net production of nitrate was highly significantly correlated with net N mineralization, being a reasonable predictor of nitrification in a simple regression model (R2=0.47; P<0.001). Net nitrification was also significantly correlated with the NO3 -N initially present at the start of the growing season (R=0.65; P<0.001) and with the labile organic P content of the soil (R=0.65; P<0.001). By including initial NO3 -N and labile organic P, together with net N mineralization and pH, in a multiple regression model, net nitrate production could be predicted with a much higher precision (R2=0.75; P<0.001). Although apparent nitrification was not significantly correlated with pH, the latter contributed significantly to the multiple regression equation for the prediction of the former.The influence of the labile organic P pool may act via its positive correlation with microbial biomass, thus more or less reflecting the potential mineralization/nitrifying capacity of a particular site.  相似文献

There is continuous interest in many countries in maintaining and manipulating the rich ecological value of hypersaline ecosystems for aquaculture. The Megalon Embolon solar saltworks (northern Greece) were studied in sites of increasing salinity of 60–144 ppt to evaluate Dunaliella salina abundance and microalgal composition, in relation to physical and chemical parameters. Cluster and ordination analyses were performed based on the biotic and abiotic data matrices. Using fresh aliquots from 60 and 140 ppt salinity waters, phytoplankton performance was appraised with flask cultures in the laboratory by varying the inorganic PO4-P concentration at 23 °C and 30 °C. At the saltworks, among the most abundant microalgae identified were species of the genera Dunaliella, Chlamydomonas, Amphora, Navicula, and Nitzschia. Dunaliella salina populations were predominant comprising 5–22% of the total microalgal assemblages during spring, but only 0.3–1.0% during the summer, when grazing by Artemia parthenogenetica and Fabrea salina was intense. D. salina cell density in April–July was in the range of 0.4–12.5 × 106 L−1 with typical densities of 1.5–4.5 × 106 L−1. Overall, microalgal densities were high in salinities of ≥100 ppt when inorganic-P concentrations were ≥0.20 mg L−1 within saltworks waters. Multivariate analysis of species abundance showed that algal growth responses were primarily related to variation in salinity and inorganic-P concentrations, but also to NO3-N concentration. In the laboratory, experiments indicated effective fertilization and denser microalgal growth under high inorganic PO4-P applications (4.0 and 8.0 mg L−1) at 60 ppt salinity and 23 °C. The lower PO4-P applications (0.6–2.0 mg L−1) were more effective at 60 ppt salinity and 30 °C. At 140 ppt salinity, microalgal growth response was less obvious at any of the corresponding phosphorus concentrations or temperatures. In both salinity experiments, Dunaliella salina bloomed easily and was predominant among the microalgae. Our observations indicate that Dunaliella salina populations and the overall rich microalgal profile of the saltworks, along with their performance in laboratory mono–and mixed cultures hold promise for mass cultivation within the M. Direct ammonium fumarate production from glucose-based media with Rhizopus arrhizus NRRL 1526 was obtained using 2-kmol (NH4)2CO3 per-m3 as neutralising agent and controlling mycelial growth by phosphorous (P) limitation. As the P level in the production medium was increased from 0 to 0.3-kg of KH2PO4 per m3, the fumarate yield decreased from 0.32 to 0.13-g per-g of glucose consumed; maximum ammonium fumarate productivity (0.46-kg-m–3-h–1) was obtained when using 0.1-kg phosphate-m–3.  相似文献

Artemisinin, an anti-malarial drug isolated from the annual wormwood Artemisia annua L., has a marked activity against chloroquine-resistant and chloroquine-sensitive strains of Plasmodium falciparum, and is useful in treatment of cerebral malaria. Shoot cultures of Artemisia annua L. were established on Murashige and Skoog basal medium which contained (per litre) 30 g sucrose, 0.5 mg 6-benzyladenine and 0.05 mg naphthaleneacetic acid. Using an optimized combination of sucrose (30 g/l), nitrate (45 mM), inorganic phosphate (200 mg/l), gibberellic acid (7 mg/l) and the ratio of NH4 +-N to NO3 -N of 1:3, artemisinin production reached 26.7 mg/l after 30 days. This procedure provides a potential alternative for production of artemisinin from in vitro tissue cultures.  相似文献

The High Arctic glacial ecosystem: new insights from nutrient budgets   总被引:3,自引:2,他引:1  
This paper describes detailed budgets of water, Cl, dissolved Si and both inorganic and organic forms of nitrogen and phosphorus for two small glacier basins in Arctic Svalbard (Midre Lovénbreen and AustreBrøggerbreen). Rates of nutrient deposition are modest, dominated by inorganic nitrogen and episodically enhanced by extreme events. Hence deposition rates are also variable, ranging from 20 to 72 kg NO3-N km−2 a−1 and 10–37 kg NH4-N km−2 a−1 over just two consecutive years. Deposition of dissolved organic and particulate forms of nitrogen (DONand PN respectively) also appears significant and therefore requires further investigation (3–8 kg DON-N km−2 and 7–26 kg PN-N km−2 during winter – no summer data are available). Evidence for microbially mediated nutrient cycling within the glacial system is clear in the nutrient budgets, as is the release of large phosphorus, Si and organic/particulate nitrogen fluxes by subglacial erosion. The latter is entirely dependent upon the presence of subglacial drainage, promoting silicate mineral dissolution and the erosion of largely unweathered apatite. The large DON and PN fluxes are surprising and may relate to young organic nitrogen associated with microbial life within the glaciers. This is because wide spread assimilation of NH4+ and perhaps even nitrification occurs on the glacier surface, most likely within abundant cryoconite holes. Further microbial activity also occurs at the glacier bed, where denitrification and sulphate reduction is now known to take place. Thus a two component ‘glacial ecosystem’ is proposed that is highly sensitive to climate change.  相似文献

Alcaligenes faecalis sp. No. 4, that has the ability of heterotrophic nitrification and aerobic denitrification in high-strength ammonium at about 1200 mg-N/l, converted about one-half of removed NH 4+-N to intracellular nitrogen and nitrified only 3% of the removed NH4+. From the nitrogen balance, 40–50% of removed NH4+-N was estimated to be denitrified. Production of N2 was confirmed by GC-MS and 90% of denitrified products was N2. The maximum ammonium removal rate, 29 mg-N/l h and its denitrification rate in aerated batch experiments, were 5–40 times higher than those of other bacteria with the same ability.  相似文献

An evaluation was made of the annual productivity of Spirulina (Arthrospira) and its ability to remove nutrients in outdoor raceways treating anaerobic effluents from pig wastewater under tropical conditions. The study was based at a pilot plant at La Mancha beach, State of Veracruz, Mexico. Batch or semi-continuous cultures were established at different seasons during four consecutive years. The protein content of the harvested biomass and the N and P removal from the ponds were also evaluated. Anaerobic effluents from digested pig waste were added in a proportion of 2% (v/v) to untreated sea-water diluted 1:4 with fresh water supplemented with 2 g L–1 sodium bicarbonate, at days 0, 3 and 5. A straight filament strain of Spirulina adapted to grow in this complex medium was utilized. A pH value 9.5 ± 0.2 was maintained. The productivity of batch cultures during summer 1998 was significantly more with a pond depth of 0.10 m than with a depth 0.065 m. The average productivity of semi-continuous cultures during summer 1999 was 14.4 g m–2 d–1 with a pond depth of 0.15 m and 15.1 g m–2 d–1 with a depth of 0.20 m. The average annual productivity for semi-continuous cultures operating with depths of 0.10 m for winter and 0.15 and 0.25 m for the rest of the year, was 11.8 g m–2 d–1. This is the highest value reported for a Spirulina cultivation system utilising sea-water. The average protein content of the semi-continuous cultures was 48.9% ash-free dry weight. NH4-N removal was in the range 84–96% and P removal in the range of 72–87%, depending on the depth of the culture and the season.  相似文献

Pond cultivation of the subtropical, euryhaline macroscopic red algaGracilaria tenuisipitata var.liui Zhanget Xia was carried out in brackish seawater (6–7) in the Gryt archipelago on the east coast of Sweden, using four outdoor tanks of 30–40 m3. Growth rate and nutrient uptake in batch culture were measured with the aim of estimating the water purification capacity ofG. tenuisipitata in outdoor conditions. Its ability to withstand epiphytic infections was also studied. An average growth rate of 4 biomass increase per day was recorded during two seasons with a maximum growth rate of 9 d–1. The initial biomass was usually 1 kgFW m–3 (FW, fresh weight). The nutrient uptake capacity was on average ca. 1 g Ni kgFW–1 d–1 and 0.08 g Pi kgFW–1 d–1 and the uptake rates for NH4 +-N were higher than those for NO3 -N. Both the growth rate and the nutrient uptake rate were highest at the highest water temperature. Co-cultivation with rainbow trout (Oncorhynchus mykiss) was tested: with trout fodder as the only nutrient inputG. tenuistipitata could grow and maintain low levels of Ni and Pi with optimum efficiency at a trout: alga ratio of 1:1 (w:w). Epiphytic growth of filamentous green and brown algae was limited, probably as a result of the high pH values caused by inorganic carbon uptake byG. tenuistipitata. The growth ofEnteromorpha intestinalis, the only significant epiphyte, was completely inhibited and the majority of plants died by a few days treatment with 100 µg 1–1 Cu2+, a concentration that did not severely affectG. tenuistipitata. We conclude thatG. tenuistipitata can be cultivated in outdoor ponds in southern Sweden during 5–6 months of the year using aerated or unaerated batch cultures and that wastewater from trout cultivation may be used as a nutrient source, resulting in purification with respect to N and P.  相似文献

The transformed root culture of Polygonum tinctorium Lour. was established by infecting leaf explants with Agrobacterium rhizogenes A4. These cultures were examined for their growth and indigo content under various culture conditions. Among the four different culture media tested, SH medium showed the highest yield for root growth (28 mg dry wt/30 ml) and indigo production (152 g/dry wt). In SH medium, 30 g sucrose l–1, 2500 mg KNO3 l–1, 300 mg NH4H2PO4 l–1 were the best conditions for indigo production at pH 5.7. The production of indigo in hairy roots slightly increased with the addition of 200 mg chitosan l–1 (186 g/dry wt) and 20 U pectinase l–1 (181 g/dry wt).  相似文献

The growth rate ofGracilaria gracilis maintained in tanks at an abalone farm near Port Elizabeth, South Africa, was examined under various tank conditions and NH4-N pulse frequencies and concentrations. This was accompanied by analyses of the components of the internal nitrogen pool. A maximum growth rate of ca. 35% wk–1 was obtained at 1200 M NH4-N. The alga was able to grow at non-nitrogen limited rates using only internal stored nitrogen to sustain growth for one week before the growth rate decreased to ca. 17% per week. NH4-N pulse frequency did not affect growth rate but one pulse per week led to a marked decrease in total-N, protein, phycoerythrin and chlorophyll-a content. An increase in pulse frequency to two pulses per week doubled the protein content from 2.351 ± 0.143 to 4.453 ± 0.090% (per unit dry mass). Carbohydrate content was inversely related to nitrogen storage. The growth rate in fouled tanks was always lower than in clean tanks. It seems likely that seaweeds and diatoms colonising the tank sides reduced light reflected off the inside of a tank, thereby reducing the growth rate.  相似文献

To study the impact of high atmospheric nitrogen deposition on the leaching of NO3 and NH4+ beneath forest and heathland vegetation, investigations were carried out in adjacent forest and heathland ecosystems in Northwest Germany. The study area is subjected to high deposition of nitrogen ranging from 15.9 kg ha–1 yr–1 in bulk precipitation to 65.3 kg ha–1 yr–1 beneath a stand of Pinus sylvestris L. with NH4–N accounting for 70–80% of the nitrogen deposited. Considerable leaching of nitrogen compounds from the upper horizons of the soil, mostly as nitrate, occurred at most of the forest sites and below a mixed stand of Calluna vulgaris (L.) Hull. and Erica tetralix, but was low in a Betula pubescens Ehrh. swamp forest as well as beneath Erica tetralix L. wet heath and heath dominated by Molinia caerulea(L.) Moench. Ground water concentrations of both NO3–N and NH4–N did not exceed 1 mg L–1 at most of the sites investigated.  相似文献

The potential of three estuarine macroalgae (Ulvarotundata, Enteromorpa intestinalis andGracilaria gracilis) as biofilters for phosphate ineffluents of a sea bass (Dicentrarchus labrax) cultivationtank was studied. These seaweeds thrive in Cádiz Bay and were alsoselected because of their economic potential, so that environmental andeconomicadvantages may be achieved by future integrated aquaculture practices in thelocal fish farms. The study was designed to investigate the functioning of Pnutrition of the selected species. Maximum velocity of phosphate uptake (2.86mol PO4 g–1 dry wth–1) was found in U. rotundata.This species also showed the highest affinity for this nutrient. At low flowrates (< 2 volumes d–1), the three species efficientlyfiltered the phosphate dissolved in the waste water, with a minimum efficiencyof 60.7% in U. rotundata. Net phosphate uptake rate wassignificantly affected by the water flow, being greatest at the highest rateassayed (2 volumes d–1). The marked decrease in tissue P shownby the three species during a flow-through experiment suggested that growth wasP limited. However, due to the increase in biomass, total P biomass increasedinthe cultures. A significant correlation was found between growth rates and thenet P biomass gained in the cultures. A three-stage design under low water flow(0.5 volumes d–1) showed that the highest growth rates (up to0.14 d–1) and integrated phosphate uptake rates(up to 5.8 mol PO4 3– g–1dry wt d–1) were found in E.intestinalis in the first stage, with decreasing rates in thefollowing ones. As a result, phosphate become limiting and low increments oreven losses of total P biomass in these stages were found suggesting thatphosphate was excreted from the algae. The results show the potential abilityofthe three species to reduce substantially, at low water flow, the phosphateconcentration in waste waters from a D. labrax cultivationtank, and thus the quality of effluents from intensive aquaculture practices.  相似文献

A mutant strain of Penicillium citrinum grown in a chemically-defined production medium, yielded 145 mg compactin l–1. The medium also facilitated spectrophotometric analysis of compactin. Addition of KH2PO4in the production medium did not increase the compactin production, while addition of a surfactant, Tween 80, increased compactin to 175 mg l–1. Inoculation with 107 spores ml–1 and initial pH of 6.5–7 were the most suitable for compactin production.  相似文献

Ulva rigida was cultivated in 7501 tanks at different densities with direct and continuous inflow (at 2, 4, 8 and 12 volumes d–1) of the effluents from a commercial marine fishpond (40 metric tonnes, Tm, of Sparus aurata, water exchange rate of 16 m3 Tm–1) in order to assess the maximum and optimum dissolved inorganic nitrogen (DIN) uptake rate and the annual stability of the Ulva tank biofiltering system. Maximum yields (40 g DW m–2 d–1) were obtained at a density of 2.5 g FW 1–1 and at a DIN inflow rate of 1.7 g DIN m–2 d–1. Maximum DIN uptake rates were obtained during summer (2.2 g DIN M–2 d–1), and minimum in winter (1.1 g DIN m–2 d–1) with a yearly average DIN uptake rate of 1.77 g DIN m–2 d–1 At yearly average DIN removal efficiency (2.0 g DIN m–2 d–1, if winter period is excluded), 153 m2 of Ulva tank surface would be needed to recover 100% of the DIN produced by 1 Tm of fish.Abbreviations DIN= dissolved inorganic nitrogen (NH inf4 sup+ + NO inf3 sup– + NO inf2 sup– ); - FW= fresh weight; - DW= dry weight; - PFD= photon flux density; - V= DIN uptake rate  相似文献
