Summary In contrast to all filamentous fungi examined to date, vegetative hyphae ofAllomyces macrogynus, whether extending or not, produced an outward flow of positive electrical current, at a maximum of 0.16 A cm–2 around 40 m behind the apex, as measured with a vibrating probe. Inward currents of up to 0.55 A cm–2 were recorded around the rhizoids. Increases in outward current were observed in hyphae pre-grown under oxygen deficiency and then allowed to widen backwards to the hyphal base in sufficient oxygen. When spores were germinated in an applied electrical field they produced rhizoids predominantly towards the anode. Hyphae were produced initially towards the cathode but later bent around towards the anode. Experiments with a range of chemicals provided no evidence for the involvement of calcium in vegetative growth and development inA. macrogynus. Polyoxin and nikkomycin, inhibitors of chitin synthesis, had no effect on swimming zoospores, but inhibited wall formation of cysts, rhizoids and forward and backward growing hyphae.  相似文献

The characteristics of a subjective sound field formed under conditions of dichotic stimulation were studied in healthy subjects (six females and seven males) with normal auditory sensitivity upon movement of a sound image (SI) in different directions. The character and the trajectory values of the emerging subjective sound image (SSI) were determined depending on the direction of its motion and the initial interaural delay (700, 400, and 200 s). Certain differences in the assessment of the parameters of moving sound images between the groups of male and female subjects were revealed. In female subjects, the averaged trajectory values in the right and left hemispheres were the same when the SSI moved in both directions and shortened uniformly with a decrease in the initial interaural delay. With a 700-s delay, the trajectory values of the male subjects for all directions of motion of the SSI were the same as those of female subjects. With initial 400- and 200-s delays, the trajectory values were significantly greater in the group of male subjects if a SI moved from the right or left ear to the median line of the head. With the method used, no interhemispheric asymmetry was revealed in the process of lateralization of moving sound images, which, under certain conditions, may be of importance for increasing the accuracy of localization of sound sources in the environment.  相似文献

Electron microscopic examination of nucleic acids requires the use of special spreading techniques. The classical method was developed by Kleinschmidt and Zahn in 1959. Modifications of this method increased sensitivity to allow detection of a total amount of about 1×10-3 g of single-stranded DNA and 1×10-5 g of double-stranded DNA. Here we describe two rapid and simple procedures increasing sensitivity by 1–2 orders of magnitude to visualize at least 1×10-5 g of single- and/or double-stranded DNA.  相似文献

A procedure for adventitious shoot regeneration from leaf explants of quince (Cydonia oblonga Mill.) using thidiazuron (TDZ) was developed. Excised leaves of cultures grown on Murashige and Skoog (MS) medium containing 5 M benzyladenine (BA) and 0.9% Gibco Phytagar were used. Several experiments were conducted to determine optimum concentrations of thidiazuron, -naphthaleneacetic acid (NAA) and sucrose. When the medium contained 1.5 M TDZ and 2.5 M NAA, 85% of the discs regenerated shoots with an average of eight shoots per leaf disc. An incubation period of three weeks in the dark was necessary for optimum shoot regeneration. Leaves excised from four to six-week-old cultures gave a higher percent shoot regeneration than leaves from cultures older than six weeks. Regeneration percentages were significantly reduced when sucrose concentration in the medium was less than 3%. A significantly higher percentage of shoots regenerated when leaf discs were placed on the regeneration medium abaxial side down as compared to the adaxial side.Regenerated shoots were cultured on MS medium containing 5 M BA and rooted on half-strength MS medium containing 10 M NAA. Rooted plantlets were acclimatized to greenhouse conditions for evaluation of any somaclonal variation. Protoplasts were isolated from young leaves or etiolated shoot apices. For initiation of divisions the protoplasts were embedded in sodium alginate and cultivated in MS or MI medium supplemented with 2.2 M BA, 2.6 M NAA and 2.2 M 2,4-dichlorophenoxyacetic acid. The protoplasts of all seven lines tested developed to protocalluses at high frequencies. No genotypic differences were observed. When BA was used in combination with NAA in the regeneration experiments, only a few protocalluses (highest frequency 3%) exhibited shoot organogenesis. When BA was replaced with thidiazuron, the percentage of protocalluses that developed shoots increased in two of three tested lines to 7% and 56%, respectively. Shoot development was achieved under light conditions. Perception of signals simulating directional movement of a sound source was studied in two groups of patients with cortical temporal lobe epilepsy and epileptic activity foci in the right or left temporal area of the cortex. On dichotic stimulation, the character and length of the trajectories of subjective auditory images (SAIs) were determined as dependent on the direction of SAI movement and the initial interaural delay (700, 400, and 200 s). For any delay or direction examined, SAI trajectories were shorter in the patients of both groups than in healthy subjects. Regardless of the side of an epileptic focus, the shortest trajectories were detected in the hemisphere where SAI movement ended, especially at an interaural delay of 200 s. The narrowest subjective acoustic field was observed in patients with epileptic foci in the right temporal cortex. Possible mechanisms of the changes in spatial hearing are discussed. The changes in SAI perception are assumed to result from distorted binaural interactions, which manifest themselves in functional asymmetry of the two auditory centers and may be caused by a convulsive activity focus present in one temporal lobe.  相似文献

High-throughput screening requires simple assays that give reliable quantitative results. A microplate assay was developed for reducing sugar analysis that uses a 2,2-bicinchoninic-based protein reagent. Endo-1,4--d-xylanase activity against oat spelt xylan was detected at activities of 0.002 to 0.011 IU ml–1. The assay is linear for sugar concentrations from 0 to 86 g ml–1 and can also be used to assay protein concentrations (0 to 143 g ml–1) on the same plate. A variety of temperatures and pH conditions can be used and, after incubation, the assay requires only one detection reagent and one heating step.  相似文献

The perceived trajectory length of the movement of a fused auditory image (FAI) against the background of interaural intensity differences was estimated in eight subjects upon dichotic stimulation through headphones. FAI movement was created by gradually changing the interaural delay (ΔT) between clicks in binaurally presented series of clicks from 0 to ±630 μs. When the FAI moved from the left to the right, an increase in interaural intensity difference (ΔI) from 0 to +13 dB (in the right channel of stimulation) significantly shortened the FAI trajectory because of a greater lateralization of the start point of FAI movement as compared to its end point. In the case of an oppositely directed movement (from the right to the left), the movement trajectory significantly decreased when ΔI increased from 0 to −13 dB (i.e., in the left stimulation channel), which was also explained by the greater lateralization of the start point of FAI movement as compared to its end point.__________Translated from Fiziologiya Cheloveka, Vol. 31, No. 3, 2005, pp. 32–38.Original Russian Text Copyright © 2005 by Varyagina.  相似文献

The binaural masking level difference (BMLD) is a psychophysical effect whereby signals masked by a noise at one ear become unmasked by sounds reaching the other. BMLD effects are largest at low frequencies where they depend on signal phase, suggesting that part of the physiological mechanism responsible for the BMLD resides in cells that are sensitive to interaural time disparities. We have investigated a physiological basis for unmasking in the responses of delay-sensitive cells in the central nucleus of the inferior colliculus in anaesthetized guinea pigs. The masking effects of a binaurally presented noise, as a function of the masker delay, were quantified by measuring the number of discharges synchronized to the signal, and by measuring the masked threshold. The noise level for masking was lowest at the best delay for the noise. The mean magnitude of the unmasking across our neural population was similar to the human psychophysical BMLD under the same signal and masker conditions.  相似文献

Stable diatom populations in earthen ponds for fishpond effluent treatment supported fast rates of bivalve growth in integrated mariculture systems. However, when these ponds were lined with PVC plastic sheets to prevent seepage, the populations of benthic diatoms dwindled, and did not support, as before, a commercially acceptable rate of growth by oysters and clams. Experiments were undertaken to understand the problem and restore the diatom productivity of such ponds. Clones of Amphora luciae, A,cftenerrima, Cylindrotheca closterium, Navicula cf lineola, N.cf lenzii, N. salinicola, N. cf viminoides, and Nitzschia laevis were isolated in axenic culture from an earthen sedimentation pond. Their N, P, Si, and trace element requirements for growth in fully defined media, and in media formulated with mariculture effluent, were studied in axenic batch culture. In fully defined batch culture tests, most of the isolates achieved their highest density in media with 32 M P, 0.7 mM N, 20 M Fe, 10 nM–20 M Mn, 10–20 M Zn and Co, and 17.5 M Si. Enrichment by trace elements and Si stimulated the populations of these diatoms even in media based on nutrient-enriched mariculture effluents. However, in large flow-through agnotobiotic mesocosms (700 L), only Si enrichment was needed. Si concentration >100 M was required to promote the sustained blooms of diatoms in full-sized and commercial PVC-lined fishpond effluent treatment ponds (300 m2, 1 m depth). Except for Si, the requirements of the diatoms for micronutrients were apparently fully satisfied by the fishpond effluents (uneaten food and fish-waste). A molar ratio of 1:1 between Si and N is necessary to sustained dense diatom populations in the pond water. It is therefore recommended to enrich plastic lined mariculture effluent treatment/sedimentation ponds with Si, if the goal is to raise bivalves as a secondary crop.  相似文献

Anthers from Frumoasa alba (White beauty), Otilia, Valerien, Mission and Siegfried Rebe (FS4) cultivars were cultured at the uninucleate stage of the microspore on Murashige and Skoog (1962) and Nitsch and Nitsch (1969) media supplemented with 2,4-dichlorophenoxyacetic acid (4.9 M) and benzyladenine (4.4 M). The primary calli were subcultured on MS medium with 6.6 M BA and 1.1 M indolylacetic acid, in order to induce their growth and plant regeneration. After seven months, vegetative buds were obtained with Frumoasa alba (2.7%), Otilia (0.3%), Valerien (4.5%), embryogenic callus was obtained with Mission and plant regeneration with Siegfried Rebe. Long term embryogenesis was maintained in Mission cv. for four years, by selection and regular transfer of the embryogenic areas of anther-derived calli. The embryogenic calli have the ability to generate abnormal somatic embryos with one, two or three cotyledons and cup or trumpet-shaped with fused cotyledons. In parallel with the embryogenic process, organogenesis with buds, leaf and shoot differentiation was regularly observed.  相似文献

Evidence is presented that the high levels of internal l-glutamic and l-aspartic acid in frog Rana esculenta red blood cells are due to the existence of a specific carrier for acidic amino acids of high affinity K m = 3 m and low capacity (Vmax) 0.4 mol l-Glu · Kg–1 dry cell mass · 10 min–1. It is Na+ dependent and the incorporation of l-glutamic acid can be inhibited by l and d-aspartate and l-cysteic acid, while d-glutamic does not inhibit. Moreover, this glutamic uptake shows a bell-shaped dependence on the external pH. All these properties show that this carrier belongs to the system X AG family. Besides the incorporation through this system, l-glutamic acid is also taken up through the ASC system, although, under physiological conditions, this transport is far less important, since it has relatively low affinity K m 39 m but high capacity (V max) 1.8 mol l-Glu · Kg–1 dry cell mass · 10 min–1.  相似文献

The effect of anti-regucalcin monoclonal antibody on neutral phoshatase activity in rat liver cytosol was investigated. Phosphotyrosine, phosphoserine, and phosphothreonine were used as the substrate toward phosphatase asssy. Liver cytosolic phosphatase activity with three phosphoaminoacids was significantly increased in the presence of anti-regucalcin antibody (100 and 200 ng/ml) in the enzyme reaction mixture with calcium chloride (0.1 mM) or EGTA (1.0 mM). The effect of anti-regucalcin antibody was completely abolished in the presence of exogenous regucalcin (1.0 M), indicating the involvement of endogenous regucalcin. The anti-regucalcin anti body- increased phosphatase activity was not significantly altered in the presence of trifluoperazine (20 M), an antagonist of calmodulin, or akadaic acid (10 M), an inhibitor of protein phosphatase, although these inihibitors caused a slight decrease in liver cytosolic phosphatase activity. The effect of endogenous regucalcin might be not related to calmodulin, and it was insensitive to okadaic acid. The present findings suggest that endogenous regucalcin is involved in the regulation of protein phasphatase in rat liver cytoplasm.  相似文献

1. The number and distribution pattern of -adrenergic receptors in the brain have been reported to be species specific. The aim of the present study was to describe binding of the -adrenoceptor ligand [125I]iodocyanopindolol in the brain of the tree shrew (Tupaia belangeri), a species which provides an appropriate model for studies of psychosocial stress and its consequences on central nervous processes.2. 125I-Iodocyanopindolol (125ICYP) labeling revealed a high degree of nonspecific binding, which was due mainly to interactions of this ligand with serotonin binding sites. For a quantitative evaluation of 1- and 2-adrenoceptors, serotonin binding sites had to be blocked by 100 M 5HT.3. Binding of the radioligand to 1- and 2-adrenoceptors was characterized using the 1-specific antagonist CGP20712A and the 2-specific antagonist ICI118.551. 1-adrenoceptor binding is present in the whole brain, revealing low receptor numbers in most brain regions (up to 1.5 to 2.7 fmol/mg). A slight enrichment was observed in cortical areas (lateral orbital cortex: 4.0±0.7 fmol/mg) and in the cerebellar molecular layer (8.7±1.0 fmol/mg).4. Competition experiments demonstrated high- and low-affinity binding sites with considerable variations in K i values for CGP20712A, showing that various affinity states of 1-adrenoceptors are present in the brain (K i: 0.61 nM to 67.1 M). In the hippocampus, only low-affinity 1-adrenoceptors were detected (K i: 1.3±0.2 M). Since it is known that 125ICYP labels not only membrane bound but also internalized -adrenoceptors, it can be assumed that the large population of the low-affinity sites represents internalized receptors which may be abundant due to a high sequestration rate.5. High numbers of 2-adrenoceptors are present in only a few brain structures of tree shrews (external layer of the olfactory bulb, 15.8±2.0 fmol/mg; claustrum, 19.3±1.5 fmol/mg; anteroventral thalamic nucleus, 19.4±1.5 fmol/mg; cerebellar molecular layer, 55.0±4.3 fmol/mg). Also for this class of -adrenoceptors, high- and low-affinity binding sites for the 2-selective antagonist ICI118.551 were observed, indicating that 125ICYP labels membrane bound and internalized 2-adrenoceptors. Only in the cerebellar molecular layer was a high percentage of high-affinity 2-adrenoceptors detected (K i for ICI118.551 was 1.8±0.3 nM for 90% of the receptors).6. In conclusion, 1- and 2-adrenoceptor binding can be localized and quantified by in vitro receptor autoradiography in the brains of tree shrews when serotonergic binding sites are blocked. The amount of masking of sounds from one source (signals) by sounds from a competing source (maskers) heavily depends on the sound characteristics of the masker and the signal and on their relative spatial location. Numerous studies investigated the ability to detect a signal in a speech or a noise masker or the effect of spatial separation of signal and masker on the amount of masking, but there is a lack of studies investigating the combined effects of many cues on the masking as is typical for natural listening situations. The current study using free-field listening systematically evaluates the combined effects of harmonicity and inharmonicity cues in multi-tone maskers and cues resulting from spatial separation of target signal and masker on the detection of a pure tone in a multi-tone or a noise masker. A linear binaural processing model was implemented to predict the masked thresholds in order to estimate whether the observed thresholds can be accounted for by energetic masking in the auditory periphery or whether other effects are involved. Thresholds were determined for combinations of two target frequencies (1 and 8 kHz), two spatial configurations (masker and target either co-located or spatially separated by 90 degrees azimuth), and five different masker types (four complex multi-tone stimuli, one noise masker). A spatial separation of target and masker resulted in a release from masking for all masker types. The amount of masking significantly depended on the masker type and frequency range. The various harmonic and inharmonic relations between target and masker or between components of the masker resulted in a complex pattern of increased or decreased masked thresholds in comparison to the predicted energetic masking. The results indicate that harmonicity cues affect the detectability of a tonal target in a complex masker.  相似文献

Thresholds of the event-related potentials (ERPs) appearance were measured for one stationary and four moving auditory images presented in silence or under forward masking conditions. The difference between thresholds in silence and after noise masker was considered as masking level. Under the forward masking, the amplitude of the ERP to the first click in the test series decreased in guinea pig auditory cortex. Masking level decreased with the time lag between signal and masker and didn't depend on the fused auditory image localization that corresponded to the first click in different test signals. This fact can support the hypothesis that for the long test signals the initial part can be masked more than the final one. The ERPs amplitude to next clicks in test series depended on interaction of two factors: forward masking in the "masker-signal" system and interaction of separate ERPs in the series evoked by the test signal.  相似文献

Summary In small (<300 m diameter) pulmonary arterial (PA) rings isolated from the cat, hypoxia induced a transient contraction (250±120 mg, n=7), whereas in rings of rabbit PA of the same size, hypoxia had no significant effect (n=19). Precontraction by 40 mmol KCl · l-1, noradrenaline (NA) 10-6 mol · l-1, or histamine (His 10-5 mol · l-1) did not modify this difference between the two species and did not potentiate the hypoxic contraction of small rings of the cat PA. Large rabbit pulmonary arterial segments (300–2000 m) exhibited no response to hypoxia before precontraction (n=15). In the presence of procaine (2%) rabbit PA rings (n=6, small) exhibited no hypoxic contraction. Summary Isogenic diploid plasmodia of different ages were produced by crossing clonal gamete populations of the myxomycete Didymium iridis. These plasmodia were then used to produce age heterokaryons to study the timing of death in comparison to the senescence of the two original homokaryons. A majority of the age heterokaryons died concurrently with the oldest nuclear component which indicates that the control of aging may be a dominant or active process in the true slime molds. Age heteroplasmons using transfilter contact between two plasmodia across 1 pore size Nucleopore membrane filters were also produced which indicates that senescence may not be a cytoplasmic infectious particle.  相似文献

Summary When cell suspensions of Saccharomyces cerevisiae NRRL-Y132 and Kluyveromyces marxianus IGC-2771 were incubated in the presence of different concentrations of ethanol, the final stable pH values (pH f ) reached in these suspensions increased with increasing ethanol concentration, indicating that ethanol enhanced passive proton diffusion into the cells. The plots of pH f as a function of ethanol concentration were linear but biphasic, displaying different slopes below and above the transition ethanol concentrations. When S. cerevisiae NRRL-Y132 and K. marxianus IGC-2771 were grown in the presence of different concentrations of ethanol, the specific growth rate () similarly depended upon ethanol concentration in a linear, biphasic way. Plots of at each ethanol concentration against pH f reached in cell suspensions at that ethanol concentration were linear and monophasic for S. cerevisiae NRRL-Y132 but biphasic for K. marxianus IGC-2771. Ethanol inhibition of growth and enhancement of proton diffusion are therefore correlated in these yeasts. Whereas ethanol inhibition of growth and enhancement of transmembrane proton diffusion were affected to the same degree by ethanol below and above the transition ethanol concentration in S. cerevisiae NRRL-Y132, these two parameters of ethanol inhibition were affected to different degrees below and above the transition in K. marxianus IGC-2771 as indicated by the inflection point in the plot of vs pH f . Attempts to extent these findings to other yeasts showed that the correlation between the effects of ethanol on pH f and is not a universal phenomenon among yeasts.Offprint requests to: S. G. Summary Cytochalasins B and D in the concentration range 0.5–10 g ml–1 produced similar effects on growth, movement and cytoplasmic structure in the pollen tubes of Iris spp. cultured in vitro. Continuous video recording showed that at 5 g ml–1, CB was capable of stopping organelle circulation in as short a period as 20 s. The usually elongated vegetative nuclei were also arrested, and subsequently contracted irregularly. Generative cells were not radically changed in shape, but occasionally moved erratically before being halted. Detailed examination of CD- and CB-treated tubes regarded as being capable of recovering growth upon transfer to normal medium revealed several characteristic effects on cytoplasmic structure. Fibrils presumed to consist of, or contain, microfilament bundles are readily visible in the older parts of the living tube where they form the pathways of organelle movement; these were either condensed into amorphous columns or fragmented by treatment. In the distal parts of the tube, the cytoplasm had contracted into amorphous masses which continued to show very slow shape changes. With the arrest of extension growth, pectin accumulated over the tube tip and in patches along the flanks. In a medium containing 1 mM ATP, recovery from treatment was achieved in some instances within l min. Organelle circulation in the younger tubes was resumed, and fresh adventive tube tips were formed. The fibrillar system of the older tubes was not restored, however; instead, the cytoplasm in these zones formed aggregates which underwent continuous amoeboid movement, the organelles within moving rapidly in irregular trajectories with no indication of the resumption of the original long-range cyclotic flux.
