Polyphenol oxidase (PPO) activity in potato (Solanum tuberosum) plants was high in stolons, tubers, roots, and flowers but low in leaves and stems. PPO activity per tuber continued to increase throughout tuber development but was highest on a fresh weight basis in developing tubers. PPO activity was greatest at the tuber exterior, including the skin and cortex tissue 1 to 2 mm beneath the skin. Flowers had high PPO activity throughout development, particularly in the anthers and ovary. Five distinct cDNA clones encoding PPO were isolated from developing tuber RNA. POT32 was the major form expressed in tubers and was found in all parts of the tuber and at all stages of tuber development. It was also expressed in roots but not in photosynthetic tissues. POT33 was expressed in tubers but mainly in the tissue near the skin. POT72 was detected in roots and at low levels in developing tubers. NOR333 was identical with the P2 PPO clone previously isolated from potato leaves (M.D. Hunt, N.T. Eannetta, Y. Haifeng, S.M. Newman, J.C. Steffens [1993] Plant Mol Biol 21: 59-68) and was detected in young leaves and in tissue near the tuber skin but was highly expressed in flowers. The results indicate that PPO is present as a small multigene family in potato and that each gene has a specific temporal and spatial pattern of expression.  相似文献

Cytokinin-like substances were produced in higher amounts by soil isolates than by those isolated from the rhizosphere and mycorrhizosphere. However more bacterial strains isolated from the rhizosphere and mycorrhizosphere were capable of synthesizing cytokinin-like compounds. A distinct effect of pH on the production of these substances was found. It is suggested that zeatin and zeatin riboside are likely to be among the cytokinin-like substances detected.  相似文献

Trans-zeatin, trans-zeatin riboside and N6-(2-isopentenyl)adenosine(i6Ado) were found to be the major cytokinins by high performanceliquid chromatographic separation and gas chromatography-massspectrometric analysis in the small tubers of sweet potato (Ipomoeabatatas Lam. cv. Minamiyutaka) with a diameter of about 5 mm.During tuber development cytokinin levels were high in tubershaving a diameter below 12 mm, minimal in tubers with a diameterof 22.5 mm, and then gradually increased as the tuber developed. The role of endogenous gibberellins (GAs) in the regulation of potato (Solanum tuberosum) tuber dormancy was examined by determining: 1. changes in endogenous GA levels during natural dormancy progression, 2. the effects of GA biosynthesis inhibitors on tuber dormancy duration and 3. the dormancy status and tuber GA levels in a dwarf mutant of potato. The tubers (cv. Russet Burbank) used in these studies were still completely dormant after 98 days of storage. Between 98 and 134 days of storage, dormancy began to end and tubers exhibited limited (< 2 mm) sprout growth. Tuber dormancy weakened with further storage and tubers exhibited greater rates of sprout growth after 187 days of storage. Tubers stored for 212 days or longer were completely non-dormant and exhibited vigorous sprout growth. Immediately after harvest, the endogenous contents of GA19, GA20, and GA1 were relatively high (0.48-0.62 ng g fresh weight(-1)). The content of these GAs declined between 33 and 93 days of storage. Internal levels of GA19, GA20, and GA, rose slightly between 93 and 135 days of storage reaching levels comparable to those found in highly dormant tubers immediately after harvest. Levels of GA19, GA20, and GA1 continued to increase as sprout growth became more vigorous. Neither GA4 nor GA8 was detected in any tuber sample regardless of dormancy status. Dormant tubers exhibited a time-dependent increase in apparent GA sensitivity. Freshly harvested tubers were completely insensitive to exogenous GAs. As postharvest storage continued, exogenous GAs promoted premature dormancy release with GA1 and GA20 eliciting the greatest response. Injection of up to 5 microg tuber(-1) of kaurene, GA12, GA19 or GA8 had no effect on dormancy release. Sprout growth from non-dormant tubers was also promoted by exogenous GA in the following sequence of activity: GA1 = GA20 > GA19. Kaurene, GA12, and GA8 were inactive. Continuous exposure of developing tubers to inhibitors of GA biosynthesis (AMO-1618, ancymidol, or tetcyclasis) did not extend tuber dormancy but rather hastened dormancy release. Comparison of tuber dormancy and GA1 content in tubers of a wild-type and dwarf mutant of S. tuberosum ssp. andigena revealed a near-identical pattern of dormancy progression in spite of the absence of detectable levels of GA1 in tubers of the dwarf sibling at any time during dormancy progression. Collectively, these results do not support a role for endogenous GA in potato tuber dormancy release but are consistent with a role for GAs in the regulation of subsequent sprout growth.  相似文献

The present research investigates the biological profile of eight symmetrical diheteroarylureas and phenylheteroarylureas, testing their hypothetical cytokinin-like activity and rooting activity. Cytokinin-like activity was assayed by the betacyanin (so-called amaranthin) accumulation test and by the tomato regeneration test. The rooting activity was assessed using the mung bean rooting test, the apple stem slice test and the rooting of apple microcuttings. Three compounds, 1,3-di(pyrazin-2-yl)urea (3a), 1,3-di(benzo[d]oxazol-5-yl)urea (3b) and 1,3-di(benzo[d]oxazol-6-yl)urea (3c), enhanced adventitious root formation in apple stem slice test, but only 3b and 3c were active in the mung bean rooting test. Compound 3b, that showed the best rooting activity, was also able to enhance the adventitious root formation in apple microcuttings. None of the compounds showed cytokinin-like activity.  相似文献

The activity of enzymes involved in the conversion of sucrose to starch together with the distribution of 14C-labelled photosynthate and 4C-sucrose was studied in potato tubers showing a range of growth rates and growth patterns. Within a particular tuber the uptake of 14C from labelled photosynthate and the conversion to ethanol-insoluble 14C was greatest in the apical tissue where both the rate of production of new storage cells and starch synthesis were likely to be greatest. Uptake and conversion of 14C was lowest in the older tissue of the tuber base. Pre-treatment of tubers with gibberellic acid reduced the total input of 14C from labelled photosynthate, reversed the gradient in 14C uptake between apical and basal tuber tissue, increased the amount of 14C per g fresh weight in the basal tissue and decreased the conversion of labelled sugars to starch. For tubers with different growth rates both the total uptake of 14C from labelled photosynthate and the ratio ethanol-insoluble 14C/ethanol-soluble 14C appeared to be correlated with growth rate. In contrast when tubers were fed directly with 14C-sucrose via the tuber surface, total uptake was independent of growth rate but the correlation between growth rate and the ratio ethanol-insoluble 14C/ethanol-soluble 14C persisted. Within a particular tuber there was a decreasing gradient in sucrose synthetase activity between youngest tissue of the tuber apex and the older tissue at the tuber base but there was no clear correlation between mean enzyme activity and tuber growth rate. ADPG-pyrophosphorylase and the ratio ADPG-pyrophosphorylase/starch phosphorylase showed some correlation with tuber growth rate. Starch synthase, starch phosphorylase and UDPG-pyro-phosphorylase activities per g fresh weight of tuber tissue appeared to be relatively constant. The results suggest that the transport of sugar from the phloem sieve tubes to the tuber storage parenchyma cells, in particular the phloem unloading step, and the conversion of sugar into starch are subject to separate regulation in the potato tuber.  相似文献

Oxalic acid (as oxalate) was detected in four tubers commonly used for food in Nigeria-Dioscorea rotundata (White yam), Solanum tuberosum (Irish potato), Ipomoea batatas (Sweet potato), and Manihot esculenta (cassava). Whereas healthy I. batata had the highest oxalic acid content, healthy M. esculenta contained the lowest. When all tubers were artifically inoculated with four fungi-Penicillium oxalicum CURIE and THOM, Aspergillus niger VAN TIEGH, A. flavus and A. tamarii KITA, there was an increase in oxalate content/g of tuber tissue. The greatest amount of oxalate was produced by P. oxalicum in D. rotundata tuber. Consistently higher amounts of oxalate were produced by the four fungi in infected sweet potato tuber than in any other tuber and consistently lower amounts of oxalate were produced by the four fungi in Irish potato tuber. Differences in the carbohydrate type present in the tubers and in the biosynthesis pathway are thought to be responsible for variation in the production of oxalate in the different tubers by the four fungi used.  相似文献

Potato plants (Solanum tuberosum L. cv. Ostara) were grown in aerated water culture in a controlled environment. When the tubers had reached a diameter of 1–3 cm. 14C-labelled or unlabelled gibberellic acid (GA3) was applied to the surface of the stolons at points approximately 1 crn from the developing tubers, and treatment continued for 10 days. - Significant quantities of GA3 moved into tuber tissue within 2–4 days of hormone application. This influx of GA3 was accompanied by a marked reduction in both the activity of ADPG-pyrophospharylase and the ratio ADPG-pyrophosphorylase/starch phosphorylase and an increase in the activity of UDPG-pyrophosphorylase. Starch phosphorylase activity initially increased slightly but then fell, whereas the activity of starch synthase remained constant throughout the experiment. The soluble sugar composition of the tubers changed qualitatively towards a pattern characteristic of growing stolon tips prior to tuber initiation, but there was no clear evidence of net starch degradation. Changes in the activities of the enzymes were observed prior to noticeable effects of the hormone on tuber growth rate or the development of new stolons at the tuber eyes. - GA3- treated tubers imported more 14C from labelled photosynthate than expected on the basis of growth rate. However, the capacity to convert solub#e-14C to ethaTiol-insoluble-14C (predominantly starch) was reduced in comparison with non-treated tubers. - The observed changes in carbohydrate composition and enzyme activities indicate that GA3 induces a drastic change in potato tuber metabolism towards a pattern characteristic for the termination of the storage process.  相似文献

Methods are described whereby the prevalence of pathogenic fungi on seed potato tubers can be measured quickly and quantitatively. Eye-bearing plugs of tuber tissue, 8 mm diameter by 10 mm long, are excised, incubated over water at 15 °C for 5 days and then examined microscopically. The occurrence of fungi and macroscopic tuber symptoms are recorded directly on punched tape and tabulated by computer.  相似文献

Green islands were observed on mustard leaves beneath the infection drops containing germinating conidia of Alternaria brassicicola, when the surrounding uninfected tissue had yellowed due to senescence. Green island formation has been correlated with the secretion of cytokinin-like substances by the pathogens. A. brassicicola secreted cytokinin-like substances in a liquid synthetic medium and their application to detached host leaves evoked the formation of green islands in the dark. 14C studies confirmed that green islands act as metabolic sinks in which photosynthates are retained or accumulated. Cytokinin-like substances appear to be actively involved in infection and pathogenesis of A. brassicicola.  相似文献

Potato plants (Solanum tuberosum L.) were grown in water culture.About 14 d after tuber initiation no significant differenceswere found between apical and basal tuber parts in 14C-uptakeand partitioning into various fractions from 14C-labelled photosynthate.Thus, the fresh weight of these tubers could be used as a parameterfor the sink size. The 14C-content per tuber (sink strength)20 h after 14CO2-supply to the foliage was significantly correlatedwith the tuber fresh weight. No correlation was found betweenthe 14C-concentration of the tuber (sink activity; ct. ming fr. wt.) and tuber fresh weight. Consequently, tuberfresh weight (sink size) per se must have been a factor whichinfluenced sink strength. Stolon parameters characterizing theirgrowth prior to tuber initiation (e.g. stolon volume) and theircapacity for photosynthate transport (diameter, length) weremeasured at the time of tuber initiation. Significant correlationswere found between these stolon parameters and subsequent growthof individual tubers. Anatomical studies on the proportion ofvarious tissues in the cross sectional area of stolons supportthe idea of a negative relation between growth of individualtubers and transport resistance in the phloem of the stolons.It is concluded that in the initial phase of tuber growth, mainlyfactors outside of the tuber determine its growth rate. In laterstages of tuber growth, when the sink strength increases, thecompeting strength of individual tubers for photosynthate isdominated mainly by factors within the tuber itself, such astheir sink size and sink activity. The relationships between rain and blight (Phytophthora infestans) were studied in unsprayed crops of cultivars differing widely in foliage and tuber susceptibility. The occasions when tubers were infected depended on rain and not cultivar, but numbers of tubers infected after rain was affected by the blight susceptibility of the cultivar. Infected tubers were first found when less than 5 % (BMS key) of the potato foliage was infected but few fresh infections occurred when 50–75% of the foliage had been destroyed. Some tubers were infected after 8 mm rain (tubers near the surface with even less) but large increases in numbers of tubers infected usually occurred only after 25 mm or more had increased soil moisture to above ‘field capacity’ around the tuber for at least 24 h. The most susceptible cultivars Ulster Ensign and Arran Banner had all plants with some tuber blight, and some plants with all tubers affected and often many lesions per tuber. Cultivars of intermediate susceptibility, King Edward and Up-to-Date, had some plants without blighted tubers, many with a few and very few with all. The more resistant cultivars Majestic and Arran Viking had many plants without infected tubers and many lesions that aborted while still necrotic threads, so that the fungus did not spread. Most infections occurred through tuber eyes, lenticels or sometimes growth cracks. The distribution of blight lesions on tubers differed in the different seasons, for example, lenticels were most commonly infected on Arran Banner and Ulster Ensign and eyes on King Edward, Majestic and Arran Viking. In late or slowly developing attacks, lesions on stems became more numerous and larger than in fast, early attacks and were prolific sources of spores on King Edward and Up-to-Date but not on Majestic and Arran Viking. Because much rain water runs down the stems of Up-to-Date and King Edward, stem lesions can provide an important source of inoculum for tubers.  相似文献

Potato (Solanum tuberosum L.) plants were grown in a greenhouse using zinc- and boron-deficient soil. The effects of seed-tuber treatment with 3 mM zinc sulfate and 8 mM boric acid on the content and ratio of phytohormones in the leaves and mature tubers, the indices of photosynthetic activity, the rate and NaF-sensitivity of respiration, and the tuber growth were studied. Zinc-sulfate treatment shifted the hormonal balance toward a substantial increase in the cytokinin content and the cytokinin/ABA ratio, as well as a decrease in the IAA/cytokinin ratio. Boric-acid treatment resulted in an increase in the IAA content and IAA/cytokinin ratio. Zinc-sulfate treatment abolished the apical dominance and increased the tuber weight due to their increased number and the number of phellem (cork) cell layers. Boric-acid treatment increased cell diameter in the tuber perimedullary zone; an increase in tuber weight per plant was related to tuber growth. A relationship between changes in the plant hormonal status induced by zinc-sulfate and boric-acid treatments and the activity of physiological processes is discussed.  相似文献

To explain the preference of wintering greylag geese Anser anser for small Scirpus maritimus tubers (<10mm) over larger ones, our hypothesis was that the former would provide higher intake rates. This 'consumption rate hypothesis' was tested experimentally by deriving the functional responses of geese feeding on tubers of three contrasting sizes. Goose consumption rates were measured as: (i) feeding rate (tubers/min) and (ii) instantaneous intake rate (g fresh weight/min) on various tuber densities (5-200 tubers/tray). Geese had linear functional responses over the range of tuber densities offered, and tuber size affected their consumption rate. The results were then used to re-examine intake rates according to relative biomass of tubers found in the wild. In support of our hypothesis, the larger tubers allowed the lower intake rates. Foraging mechanisms that possibly control the tuber ingestion rate of geese were explored. After controlling (through chemical analyses) that nutritional components of tubers do not differ between sizes, it was concluded that geese preference is mainly due to size-related constraints (i.e. handling time) on their ingestion rates. Additional limiting factors (tuber extraction, digestion capacity) which are likely to constrain food intake of wild geese are also discussed.  相似文献

Two complementary field experiments at Rothamsted in 1965, with the potato variety Majestic, related the incidence of common scab (Streptomyces scabies) to rainfall, soil moisture and time of tuber formation. In plots where the soil was maintained at field capacity (less than 10 cm. Hg moisture tension) by watering, tubers had little or no scab; infection increased in amount on plots allowed to dry to 30 cm., 50 cm. or more Hg moisture tension during late June. The time tubers formed was varied by planting sprouted (chitted) and non-sprouted seed tubers, and tuber development and scab incidence were observed on sample plants lifted at frequent intervals from unwatered plots. Scab lesions were first seen on 12 July when the distribution of lesions on the surface of tubers was affected by size of tuber both within and between the different seed-tuber treatments. These differences were correlated with estimates of tuber size on 28 June, the beginning of the first dry period. Tubers 1.0 cm. or more in diameter on 28 June had few or no lesions at the stolon attachment (heel) end of tubers on 12 July, whereas tubers smaller than this on 28 June had many lesions in this region on 12 July. The larger the tuber was on 28 June, the greater was the area free from scab lesions on 12 July.  相似文献

Pithiness in radish tubers (Raphanus sativus L.) is a syndromewhere xylem parenchyma cells die and are filled with air. Featuresassociated with rapid tuber growth such as large cells, greatdistances between strands of vascular tissue and rupture ofxylem vessels have been suggested to induce pithiness. The objectiveof this study was to analyse the extent to which pithiness inradish was related to tuber growth. Growth rates were variedby altering irradiance and plant density. In addition, the relationshipbetween pithiness and tuber growth (length, diameter, freshmass) was investigated in batches of plants grown under identicalconditions. The growth rates of shoot and tuber strongly increased withincreasing inter-plant distance and irradiance. Our resultssuggest a ‘true’ effect of assimilate supply ondry matter partitioning, as not only the ratio between shootand root dry mass, but also the slope of this relationship decreasedwith increasing irradiance and inter-plant distance. Withina treatment the ratio between tuber growth and shoot growthremained constant with plant age. Increase in length of thetuber did not appear to be an important factor in inducing pithiness.Within a batch of plants, a positive correlation between tuberdiameter and pithiness was observed only for tubers smallerthan 14 mm. An increase in both irradiance and inter-plant distanceincreased tuber size (diameter, mass) and pithiness, but theeffects on pithiness could not be ascribed only to differenttuber sizes. Pithiness strongly increased with plant age andit was concluded that pithiness occurred, to a great extent,according to genetic programming. The data showed that for treatmentswhich affect tuber growth, conclusions about their effects onpithiness depend on whether comparisons were made at the sametuber size or the same time, explaining apparent contradictionsin the literature. 通过大田试验研究了集雨限灌对同薯23号和大西洋两个马铃薯品种的补灌效应.结果表明:集蓄雨水在苗期补灌能显著提高旱作马铃薯的产量,且大西洋增产幅度高于同薯23号;在薯块膨大期补灌同薯23号的产量增加不显著,而大西洋减产.低额补灌(补灌45 mm)有利于同薯23号水分利用效率(WUE)和灌水利用效率(IWUE)的提高.大西洋苗期补灌的WUE和IWUE均高于薯块膨大期.补灌提高了同薯23号的大薯率、中薯率和单株薯产量,降低了小薯率, 但绿薯率、烂薯率有所增加;大西洋的大薯率和小薯率增加,中薯率稍有下降,烂薯率增加.  相似文献

Significant stimulation of the number of appressoria, penetration and colonization by conidia ofHelminthosporium carbonum occurred on decolorized maize leaves when exogenous carbohydrates and leaf leachates were added. Germination and germ tube length, however, did not exhibit appreciable differences on decolorized or non-decolorized maize leaves. Lower germination was recorded by leached conidia on decolorized leaves; while appressoria, penetration and colonization were absent. Addition of exogenous nutrients (sucrose>leaf leachates>yeast extract>glucose) enabled conidia to accomplish appressoria, penetration and colonization. Optimum levels for various nutrients observed were 2% (w/v) sucrose/glucose or 0.1% (w/v) yeast extract. Higher concentrations inhibited the infection stages of the pathogen. Depletion of host carbohydrates from green islands/infection sites adversely affect appressoria formation, penetration and colonization; and the loss of carbohydrates from the spore affects germination. Cytokinin-like activity at the infection site/green islands increased with the period of incubation of the host as compared to the surrounding tissue or tissue under water drops. The culture filtrate extracts ofH. carbonum recorded cytokinin-like activity which increased with growth of the fungus. TLC (thin layer chromatography) of cytokinin-like substances (tissue extract and culture filtrate) revealed major activity was confined to Rf zones 0.6 to 0.8 which co-chromatographed with zeatin and zeatin riboside. These substances increase at infection sites by virtue of which carbohydrates accumulate at these sites ensuring a continuous supply to the growing pathogen.  相似文献

Wound healing of potato tubers involves the concerted action of several enzymes that facilitate polymerization of phenolics into suberin at the wound site. A decline in the efficiency of healing and resistance to pathogens with advancing tuber age was associated with reduced ability of older tubers to produce superoxide radicals (FRs) in response to wounding. Autophotographs of luminol‐treated longitudinal sections of tissue from 6‐, 18‐ and 30‐month‐old tubers revealed a substantial decline in superoxide production at the wound surface with advancing age. Older tubers were less able to respond to wounding by increasing phenylalanine ammonia lyase (PAL) activity. This enzyme produces t‐cinnamic acid, which constitutes a component of the phenolic domain of suberin, and is normally induced by wounding and/or ethylene. Interestingly, the ability of wounded tissue to oxidize exogenous 1‐aminocyclopropane‐1‐carboxylic acid (ACC) to C2H4 also decreased with advancing tuber age. The oxidation of ACC was inhibited by the FR scavenger, n‐propyl gallate (PG), and inhibition was greatest in tissue from younger tubers, reflecting their greater ability to produce superoxide radicals upon wounding. Regardless of tuber age, 1‐aminocyclobutane‐1‐carboxylic acid, an ACC oxidase inhibitor, did not inhibit C2H4 generation from exogenous ACC. Hence, C2H4 production from ACC by wounded tuber tissue is largely non‐enzymatic and FR‐driven, and thus serves as an indicator of the ability of wounded tissue to produce superoxide. Age‐induced reduction in PAL activity and FR production at the wound surface probably limited the oxidative polymerization of phenolics into suberin during wound periderm formation. The age‐induced loss in ability of wounded tissue to heal and resist pathogens is thus consistent with reduced synthesis and polymerization of phenolic adducts into suberin, a consequence of reduced FR and PAL activity at the wound surface.  相似文献

Transgenic potato tubers that overexpressed either a cytosolic or an apoplastic invertase in the wild type or AGPase antisense background were used to analyse the effect of invertase activity on cell expansion, starch granule formation and turgor pressure during tuber development. Although the transgenic plants did not develop a visible phenotype in aerial regions the size and number of tubers were significantly modified in the various lines. Transmission electron and light microscopy were performed to monitor starch grain size and number, cell size and cell wall thickness. Water potential, osmotic pressure, and indirectly, turgor pressure were determined during the final stages of tuber development. Glucose levels were high in transgenic tubers that overexpressed a yeast-derived invertase. The number of starch grains per cell was almost identical in all transgenic lines. However, the amount of starch was modified in the transgenics as compared to the wild type. As expected, the size of starch grains was reduced in all lines that expressed an AGPase antisense mRNA. These results indicate that invertase activity and glucose levels do not affect initiation of starch grain formation during the early stages of tuber development, but growth of starch corns in the later stages of tuber maturation.  相似文献
