Three new C19-diterpenoid alkaloids vilmotenitines A-C (1-3), together with seven known ones, vilmorine D (4), talatizidine (5), isotalatizidine (6), vilmorrianine B (7), vilmorrianine D (8), talatizamine (9), 8-deacetyl-yunaconitine (10), were isolated from Aconitum vilmorinianum var. patentipilum. Vilmotenitines A (1) and B (2) are the second natural occurrences of C19-diterpenoid alkaloids with a unique rearranged six-membered B ring formed by the C(8)–C(10) linkage.  相似文献

Shawurenine C ( 1a ) and D ( 1b ), a new pair of regioisomeric C19-diterpenoid alkaloids, and five known C19-diterpenoid alkaloids ( 2 – 6 ) were isolated from the aerial part of Delphinium shawurense W. T. Wang. The chemical structures of new compounds were established based on spectroscopic analyses: HR-ESI-MS, and 1D, 2D NMR spectroscopic data. The anti-inflammatory and cytotoxic activities of these diterpenoid alkaloids were also evaluated.  相似文献

Five new aconitine-type C19-diterpenoid alkaloids, namely, carmichaenine A–E (15), and six known diterpenoid alkaloids, namely, 14-benzoylneoline (6), neoline (7), 10-hydroxyneoline (8), neolinine (9), songoramine (10), and songorine (11), were isolated from the aerial parts of Aconitum carmichaeli. Their structures were determined by extensive spectroscopic methods, especially 2D NMR analyses. Compounds 8 and 9 were isolated for the first time from A. carmichaeli.  相似文献

Three new aconitine-type C19-diterpenoid alkaloids, richardsonines A-C (1–3), were isolated from the roots of Aconitum richardsonianum Lauener, together with seventeen known compounds (4–20). Their structures were elucidated by extensive spectroscopic analysis, and the absolute stereochemistry of 1 was confirmed by X-ray crystallography. Compounds 2, 5, 8 and 9 were rare C19-diterpenoid alkaloids bearing an OH group at C-16. Isolated compounds were evaluated for inhibition of nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated neuroinflammatory responses in BV-2 microglial cells. Compounds 3 and 7 showed weak antineuroinflammatory effects with inhibition rates of 35.32 ± 2.29% and 39.94 ± 2.50%, respectively, at a concentration of 100 µM.  相似文献

Twenty-one alkaloids, including five new ones, acoapetaldines A–E (15), were isolated from the whole plants of Aconitum apetalum. Among them, 1 is an aconitine-type diterpenoid alkaloid, 2 and 3 are aporphine alkaloids and 4 and 5 are napelline-type diterpenoid alkaloids. The structures of the new alkaloids were elucidated by spectroscopic data analysis and that of acoapetaldine D (4) was confirmed by single crystal X-ray crystallography. Acoapetaldine A (1) and aconorine (12) exhibited moderate anti-tobacco mosaic virus (anti-TMV) activity, while corydine (15) displayed moderate antimicrobial activity against Pseudomonas aeruginosa.  相似文献

Diterpenoid alkaloids exhibit remarkable chemical properties and biological activities. Such compounds are frequently found in plants of the genera Aconitum, Delphinium, and Garrya. Several diterpenoid alkaloid components from Delphinium elatum cv. Pacific Giant and their derivatives exhibited cytotoxic activity against lung, prostate, nasopharyngeal, and vincristine-resistant nasopharyngeal cancer cell lines. Phytochemical investigations on the seeds of D. elatum cv. Pacific Giant led to the isolation of four new C19-diterpenoid alkaloids, melpheline (1), 19-oxoisodelpheline (2), N-deethyl-19-oxoisodelpheline (3), and N-deethyl-19-oxodelpheline (4). The isolated alkaloids were elucidated by extensive spectroscopic methods including NMR (1D and 2D), IR, and MS (HRMS).  相似文献

Incubation of [5,15,20-14C3]uro'gen III (1) with a cell-free system from P. shermanii has revealed that during the formation of cobyrinic acid (3) from this substrate C-20 is lost to the medium and can be trapped as the dimedone adduct of formaldehyde. Similarly, [5,15,20-14C3]uro'gen heptacarboxylic acid (5) is converted with loss of C-20 to cobyrinic acid although much less efficiently. The significance of these results for the mechanism of corrin biosynthesis is discussed.  相似文献

Three new picraline-type alkaloids, alstiphyllanines E–G (13) and a new ajmaline-type alkaloid, alstiphyllanine H (4) were isolated from the leaves of Alstonia macrophylla together with 16 related alkaloids (520). Structures and stereochemistry of 14 were fully elucidated and characterized by 2D NMR analysis. Alstiphyllanines E and F (1 and 2) showed moderate Na+-glucose cotransporter (SGLT1 and SGLT2) inhibitory activity. A series of a hydroxy substituted derivatives 2128 at C-17 of the picraline-type alkaloids have been derived as having potent SGLT inhibitory activity. 10-Methoxy-N(1)-methylburnamine-17-O-veratrate (6) exhibited potent inhibitory activity, suggesting that the presence of an ester side chain at C-17 may be important to show SGLT inhibitory activity. Structure activity relationship of alstiphyllanines on inhibitory activity of SGLT was discussed.  相似文献

Six new cassane diterpenoids (1–6) and 15 known compounds were isolated and identified from the seeds of Caesalpinia sappan. The structures of the new compounds were determined based on the spectroscopic methods, including 1D and 2D NMR techniques. Compound 1 is a rare cassane diterpenoid featuring a nitrogen containing bridge between C-19 and C-20. Compounds 1 and 2 exhibited moderate cytotoxicity against HCT116, AGS, and HepG2 cell lines with IC50 values ranging from 6.36 to 27.86 μM.  相似文献

The following primary sulphonates have been converted into the corresponding deoxyfluoro derivatives by reaction with potassium fluoride in ethylene glycol:1,2:3,4-di-O-isopropylidene-6-O-tosyl α-D-galactopyranose (1), methyl 2,3-O2-isopropyliden-5-O-tosyl-α,β-D-ribofuranoside (2), 1,2:3,4-di-O-methylene-6-O-tosyl-α-D-glucofuranose (3), 3,5-di-O-benzylidene-1,2-O-isopropylidene-6-O-tosyl-α-D-glucofuranose (4), and 1,2:3,5-di-O-isopropylidene-6-O-tosyl-α-D-glucofuranose (5). The yields were generally poor; in the reaction of 1, a major by-product was 6-O-(2-hydroxyethyl)-1,2:3,4-di-O-isopropylidene-α-D-galactopyranose (11). The reaction of the primary hydroxyl precursor of each of the above tosylates with N2-(2-chloro- 1,1,2-trifluoroethyl)-N,N-diethylamine generally yielded the O-chlorofluoroacetyl derivative; however, 1,2:3,5-di-O-methylene-α-D-glucofuranose (12) was converted into the 6-deoxy-6-fluoro derivative (8). The 19F resonances of compounds containing the CH2F moiety fall between φC +213 and φC +235 p.p.m. The differences between the vicinal19F-1H couplings of compounds having the D-gluco and D-galacto configurations clearly reflect the influence of the C-4O-4 substitutents on the populations of the C-5C-6 rotamers. A novel type of noise-modulated, heteronuclear decoupling experiment is described.  相似文献

3-C-(Acetamidomethyl)-1,2-O-isopropylidene-β-l-threofuranose (4) and the 3-acetate (5) have been prepared in high yields from mono-O-isopropylidene-d-apiose [3-C-(hydroxymethyl)-1,2-O-isopropylidene-β-l-threofuranose] (1). Acid-catalyzed methanolysis of 4 caused migration of the isopropylidene group and the formation of methyl 4-acetamido-4-deoxy-3-C-(hydroxymethyl)-2,3-O-isopropylidene-β-d-erythrofuranoside (8) in 25% yield. The major product (45%) from the acetolysis of 4 was also a pyrrolidine derivative, namely, 4-acetamido-3-C-(acetoxymethyl)-1-O-acetyl-4-deoxy-2,3-O-isopropylidene-β-d-erythrofuranose (10). Acetolysis of 5 removed the isopropylidene group and gave four acetylated pyrrolidines (isomeric at C-1 and C-2). Conditions which resulted in minimal epimerization at C-2 were established, and the major isomers 12 and 13 were isolated in reasonable yields. 1H- and 13C-n.m.r. data for equilibrium solutions of the pyrrolidines, and for intermediates 1-5, are given.  相似文献

Four new hetisine‐type C20‐diterpenoid alkaloids, named as coreanines A–D ( 1 – 4 ), were isolated from the roots of Aconitum coreanum, together with thirteen known alkaloids ( 5 – 17 ). Phytochemical investigation of the leaves and twigs of Tabernaemontana bovina led to the isolation of 10 monoterpenoid indole alkaloids, including two new taberbovinines A (1) and B (2) along with eight known analogs: mehranine (3), 14α,15β-dihydroxy-N-methylaspidospermidine (4), (16S*)− 15-epi-E-isositsirikine (5), (16R*)− 15-epi-E-isositsirikine (6), 16 R*-19,20-E-isositsirikine acetate (7), hecubine (8), voafinidine (9), and voacangarine (10). Taberbovinine B (2) represents the first case of an unusual ring C/D cleavage among the natural Corynanthe-type alkaloids. Compounds 2 and 8 exhibited weak cytotoxicity against five human cancer cell lines, including SK-LU-1, HepG2, MCF-7, SK-Mel-2, and LNCaP, with IC50 values ranging from 42.9 to 66.3 μM, whereas compounds 4 − 6 and 9 were cytotoxic toward MCF-7, SK-LU-1 and LNCaP cells, with IC50 values in a range of 51.6–93.3 μM.  相似文献

Aconitum carmichaelii Debeaux is a widely used traditional Chinese medicine and an important source of clinical drugs, of which the parent and lateral roots are known as ‘Chuanwu’ and ‘Fuzi’, respectively. Four new C19‐diterpenoid alkaloids, carmichasines A – D ( 1 – 4 ), were isolated from the roots of Aconitum carmichaelii, together with twelve known compounds ( 5 – 16 ). Their structures were elucidated via spectroscopic analyses, including HR‐ESI‐MS, IR, and NMR. Carmichasine A ( 1 ) is the first natural C19‐diterpenoid alkaloid possessing a cyano group. Most of the diterpenoid alkaloids isolated were C19‐category, which might provide further clues for understanding the chemotaxonomic significance of this plant. The cytotoxicity of the new compounds was also investigated against several human cancer cell lines, including MCF‐7, HCT116, A549, and 786‐0, and none of them showed considerable cytotoxic activity.  相似文献

A pair of new alkaloid enantiomers [(+)- and (−)-1] as well as a pair of known enantiomeric analogues [(+)- and (−)-2] were isolated from the fruiting bodies of Ganoderma luteomarginatum. Their planar structures were determined by extensive spectroscopic analyses. The absolute configurations were established by comparison of the experimental and calculated electronic circular dichroism (ECD) or comparison of the experimental and reported specific optical rotation ([α]D). These rare Ganoderma alkaloids have a phenyl-substituted 6,7-dihydro-5H-cyclopenta[c]pyridine skeleton that has only been reported from the genus Ganoderma. The (+)- and (−)-1 were new Ganoderma alkaloids, while (+)- and (−)-2 were isolated from G. luteomarginatum for the first time. Thus, these four isolates could be tentatively determined as chemotaxonomic constituents of G. luteomarginatum.  相似文献

The syntheses of three classes of C-3 perfluoroalkyl substituted steroids are described. They are the 3β-hydroxy-3α-perfluoroalkylandrost-4-en-17-ones (5a-c), 3-perfluoroalkylandrosta-3,5-dien-3-ones (8a-c) and 3β-hydroxy-3α-perfluoroalkylandrost-5-en-17-ones (12a-c). Addition of a series of perfluroalkylorganometallic reagents (RFLi; RF = C2F5, C3F7, or C4F9) to the 3 position of silylated testosterone 2b afforded Δ4 perfluoroalkyl carbinols 3. In Scheme 1, deprotection with HF and oxidation at the C-17 carbon with PCC produced Δ4 ketones 5. In Scheme 2 dehydration of 3 with 1,2-phenylenephosphorochloridite and iodine afforded Δ3,5 dienes 6 which were deprotected and oxidized as above to the C-17 ketones 8. In Scheme 3 isomerization of the double bond of 3 from the C-4 to the C-5 position using the allylic halogenation followed by treatment with lithium aluminum hydride led to the synthesis of the double bond isomer series 12. A new method for dehydration was developed. On average and within experimental error, 3β-hydroxy-3α-perfluoroalkylandrost-5-en-17 ones (12a-c) were better than the 3-perfluoroalkylandrosta-3,5-dien-17-ones (8a-c) and 3β-hydroxy-3α-perfluoroalkylandrost-4-en-17-ones (5a-c) at inhibiting glucose-6-phosphate dehydrogenase.  相似文献

Twenty-nine compounds, including five acetophenone derivatives (1–5), three phenanthroindolizidine alkaloids (12–14), seven pentacyclic triterpenoids (15–21) and five C21 steroidal sapogenins (22–26), were isolated from the root of Cynanchum paniculatum (Bunge) Kitag. Their structures were determined by spectroscopic methods and comparison with reported data. Moreover, the chemotaxonomic relationships were also discussed. As a result, acetophenone derivatives, pentacyclic triterpenoids and C21 pregnane sapogenins can be recognized as chemotaxonomic markers for Cynanchum genus, and C. paniculatum has close relationships with some species of genus Cynanchum.  相似文献
