A rabbit lung preparation, perfused in vitro, was used to examine pulmonary metabolism of prostaglandin A1 (PGA1) and to compare the vasoconstrictor actions of PGA1, prostaglandin F (PGF) and angiotensin II. PGF caused significantly more, and angiotensin II significantly less, vasoconstriction than did an equimolar concentration of PGA1. Of three likely PGA1 metabolites only 15-keto-PGA1 had any significant vasoconstrictor action. Furosemide and aminophylline (10?3 M) reduced PGA1, PGF or angiotensin II-induced vasoconstruction. Diphloretin phosphate potentiated the vascular effect of angiotensin II. Furosemide (10?3 M) and DPP (9.5 × 10?6 M) significantly reduced pulmonary metabolism of PGA1 while aminophylline (10?3 M) had no effect on this process. Perfusion of the lungs with a hypoxic medium had no effect on PGA1 metabolism.  相似文献

The contracting and relaxing potencies of anf interactions between a number of prostaglandins (PGs) were studied in vitro on spiral strips of small canine mesenteric arteries (outside diameter < mm). PGF2α and PGE2, the most potent contracting PGs, were nearly equal in potency (EC50 4 × 10?7M) and did not cause relaxation under our experimental conditions. PGI2 and PGE1 were equal and the most potent relaxing PGs (EC50 3 × 10?9M). PGE1 also caused contraction, but this effect was not consistent. PGI2 did not cause contraction in concentrations up to 3 × 10?6M. In higher concentrations, however, it caused abrupt and near maximal contraction. PGD2 was weak in both respect, causing incomplete relaxation and contraction or biphasic effects. Interaction studies showed that PGE1 and PGI2 mutually excluded the relaxing effects of each other. PGE1 also reversed the relaxing effect of isoproterenol. However, pre-exposure to PGD2 did not attenuate the relaxing effect of PGE1 or PGI2 nor was the relaxing effect of PGD2 changed by pre-exposure to PGE1. Two different orders of potency of PGs suggest two PG receptors subserving contraction and relaxation, respectively. Further, it appears that several PGs can act upon both receptors which may explain unusual interactions between the PGs and some of their atypical effects. Finally, the data also suggest that there may be subtypes of the PG receptors subserving contraction and relaxation.  相似文献

The ATPase enzyme system from two-spotted spider mites, Tetranychusurticae (Koch) was sensitive, in vitro, to four acaricides. Tricyclohexylhydroxytin (PlictranR) was an outstanding inhibitor of oligomycin-sensitive (mitochondrial) Mg2+ATPase from fish brain and spider mite homogenates. The I50 values were 6.6×10?11M and 6.2×10?10M, respectively. Less effective were chlorbenside, chlorfenethol and ovotran. Plictran at a higher concentration (2×10?7M) was also more effective on Na+-K+ATPase both in mites and fish brain homogenates as compared to chlorfenethol, chlorbenside and ovotran. Plictran inhibited oligomycin-insensitive Mg2+ATPase at concentrations of 10?8M but stimulated at high concentrations (5×10?6M and higher).  相似文献

The spectral changes associated with the addition of prostaglandins (PGs) to hepatic microsomes from guinea pigs and rats were examined. PGA1, PGA2, PGE1, PGE2, PGF and PGF when added to guinea pig liver microsomes exhibited type I spectra. The binding affinities as determined from spectral dissociation constants (Ks) were highest with PGA1 and PGA2. With liver microsomes from control or 3-methyl-cholanthrene (MC)-treated rats, PGs did not yield type I spectra; however, in this case a weak spectrum, designated here as type “II” was at times observed, With microsomes from phenobarbital (Pb)-treated rats only PGA1 and PGA2 yielded type I spectra; again in absence of type I spectrum, a weak type “II” was occasionally observed. The addition of PGA1 and PGA2 to liver microsomes from Pb-treated rats inhibited the microcomal mediated hydroxylation of hexobarbital. The inhibition by PGA1 was competitive; the Ki = 8.2 × 10?4 M was found to be similar in magnitude to the Ks = 7.3 × 10?4 M of PGA1 observed with rat liver microsomes. These observations suggested that PGs particularly of the A series interact with the hepatic microsomal cytochrome P-450 monooxygenase system.  相似文献

Cytotoxic actions of various prostaglandins were examined on L1210 mouse leukemia and several human leukemia cell lines, and prostaglandin D2 (PGD2) was found most active. PGD2 exerted a dose dependent inhibition of L1210 cell growth over 3.6 μM. At 14.3 μM growth was completely inhibited, and the number of viable cells remarkably decreased during culture. Microscopically the remaining cells showed degenerative changes with many vacuoles in their cytoplasm. The IC50 value of PGD2 on L1210 cell growth was calculated to be 6.9 μM (2.4 μg/ml), and at this concentration the DNA synthesis in 24 hr cultured cells was also decreased to a half of the level in the control cells. Such growth inhibition by PGD2 was also found at similar concentrations with several human leukemia cell lines such as NALL-1, RPMI-8226, RPMI-8402, and Sk-Ly-16. Among other prostaglandins tested, PGA2 showed a comparable, and PGE2 a less but significant growth inhibitory activity, while PGB2, PGF and PGI2 had no such effects on cell proliferation at 14.3 μM concentration. These results suggest a potential antineoplastic activity of PGD2.  相似文献

In vitro cultures of Crithidia sp. were exposed to various concentrations of hydroxyurea (HU) during the logarithmic phase. In the presence of 5 × 10?2M HU, cell division was completely blocked after an initial increase in cell numbers by about 20%. Inhibition of incorporation of 3H-thymidine into acid-insoluble material was effective within 1 hr of exposure to the drug (5 × 10?2M) and it reached a level of 80% after 8 hr. At lower concentrations (5 × 10?4M ? 1 × 10?3M), however, incorporation of 3H-thymidine was remarkably increased while cell division remained unaffected indicating that the increase in incorporation was not due to increased DNA synthesis in preparation for cell division.  相似文献

2-Deoxy-2,3-dehydro-N-acetylneuraminic acid and its methyl ester are competitive inhibitors of Arthrobacter sialophilus neuraminidase with Ki = 1.4 × 10?6M and 4.8 × 10?5M, respectively. The Km for the substrate, N-acetylneuraminlactose, is 1.0 × 10?3M. These data, taken together with the conformation of these compounds, indicate that these compounds are transition-state analogs of the enzyme. These results also suggest that the substrate upon binding to neuraminidase is distorted to a conformation approaching that of a half-chair.  相似文献

Growth rates of two lines of tobacco (Nicotianatabacum) cell suspension cultures were measured in the presence or absence of G418, a new 2-deoxystreptamine antibiotic related to Gentamycin. Cell growth rates of N. tabacum cv. Burley were inhibited at drug concentrations as low as 1.65 × 10?7 M. At 4 × 10?7 M, the doubling time was increased from 1.5 days (control) to 2.3 days (treatment). The drug was lethal to cells at 4 × 10?6 M, and inhibition was irreversible. Cells of N. tabacum cv. Wisconsin 38 also were inhibited by the drug, although at slightly higher concentrations (ca. 2–5 fold).In view of our findings, G418 and its associated resistance factors could be of great value in plant genetic engineering.  相似文献

The stoichiometric reaction between d-TpGpGpCpCpA (d(T-G-G-C-C-A)) and cis-[Pt(NH3)2(H2O)2](NO3)2 (8.4 × 10?6 to 1.3 × 10?4M in water at pH 5.5–6) gives a single complex. High pressure gel permeation chromatography and pH-dependent 1H NMR analyses of the nonexchangeable base protons, show that it is a platinum chelate with the cis-PtII(NH3)2 moiety bound to the two N7 atoms of the adjacent guanines. A 3 × 10?3M reaction gives the same platinum chelate, via the formation of intermediate complexes, together with unsoluble adducts.  相似文献

S-(p-azidophenacyl)-glutathione, l, is a linear competitive inhibitor at pH 7.40 of beef liver glyoxalase II with Ki = 7.96 × 10?4 M. On irradiation at 340 nm it covalently inhibits glyoxalase II to a level of 42 ± 5% inhibition. This photoaffinity labelling is prevented by the presence of a glyoxalase II competitive inhibitor (the hemimercaptal of glutathione and methylglyoxal). Vasoactive intestinal peptide (VIP), secretin, catecholamines and prostaglandin E1 (PGE1) in the presence of a cyclic nucleotide phosphodiesterase inhibitor stimulate the accumulation of cyclic AMP in two colorectal carcinoma cell lines (HT 29 and HRT 18) with subsequent activation of the cyclic AMP-dependent protein kinases. In HT 29 cells incubated without phosphodiesterase inhibitor, 10?9 M VIP promotes a rapid and specific activation of the low Km cyclic AMP phosphodiesterase (1.7-fold); at 25°C the effect is maintained for more than 15 min, while at 37°C the activity returns to basal value within 15 min. As shown by dose-response studies, VIP is by far the most effective inducer (Ka = 4 · 10?10M) of the cyclic AMP phosphodiesterase activity; partial activation of the enzyme is obtained by 3 · 10?7 M secretin, 10?5 M isoproterenol and 10?5 M PGE1; PGE2 and epinephrine are without effect. In HRT 18 cells VIP is less active (Ka = 2 · 10?9M) whereas 10?6 M PGE1, 10?6 M PGE2 and 10?5 M epinephrine are potent inducers of the phosphodiesterase activity. The positive cell response to dibutyryl-cyclic AMP further indicates that cyclic AMP is a mediator in the phosphodiesterase activation process. The incubation kinetics and dose response effects of the various agonists on the cyclic AMP-dependent protein kinase activity determined for both cell types in the same conditions show a striking similarity to those of phosphodiesterase. Thus coordinate regulation of both enzymes by cyclic AMP was observed in all incubation conditions.  相似文献

The reaction of almond β-glucosidase with p-nitrophenyl-β-D-glucoside has been investigated over the temperature range +25° to ?45° using 50% aqueous dimethyl sulfoxide (DMSO) as solvent. At temperatures below those at which turnover occurs a “burst” of p-nitrophenol proportional to the enzyme concentration is observed. Such a “burst” suggests the existence of a glucosyl-enzyme intermediate whose breakdown is rate-limiting, and provides a method for measuring the active-site normality. At pH 5.9, 25°, the presence of 50% DMSO causes an increase in Km from 1.7×10?3M (0%) to 1.7×10?2M, whereas Vmax is unchanged. The DMSO thus apparently acts as a competitive inhibitor with Ki = 0.7M. The Arrhenius plot for turnover is linear over the accessible temperature range with Ea = 23.0 ± 2.0 kcal/mole.  相似文献

The inhibitory effect of various prostaglandin analogues on the anchorage independent growth of murine and human melanoma cells was measured. PGA analogues (which were modified at C-16 and C-18) did not demonstrate any major improvement in activity over PGA alone. These included 16, 16-dimethyl PGA1, 16,16-dimethyl-PGA2, 16,16-dimethyl-18-oxa-PGA2 and trans-δ-2-15-α acetoxy-16,16-dimethyl-18-oxa-11-deoxy-PGE1-methylester. The thromboxane synthetase inhibitor, U51605, demonstrated weak anti-proliferative activity. PGD2 (with a ketone at C-11 versus C-9 for PGA and PGE) was the most potent prostaglandin tested. Cells from melanoma lines displayed species differences in their sensitivities. PGA1 and PGE1 were the most potent inhibitors of the anchorage independent growth of murine melanoma cells. On human melanoma cells PGD2 was the most active prostaglandin, 2–3 times more potent than PGA1; PGE1 was a very weak inhibitor.  相似文献

Precipitation of human fibrinogen in 0.15 m NaCl occurred at pH 7.4 (Tris-HCl buffer) when ZnCl2, CuCl2, NiCl2, or CoCl2 were added beyond their respective critical concentrations. The critical concentrations were about 4 × 10?5m ZnCl2, 6 × 10?5m CuCl2, 3 × 10?4m NiCl2 and 1 × 10?3m CoCl2. At pH 5.8 2-(N-morpholino)-ethane sulphonic acid buffer, the critical concentrations were found only for CuCl2 and ZnCl2, and were about 3 × 10?5and 3 × 10?4m, respectively. CaCl2 and MgCl2 were not effective up to 1 × 10?2and 2 × 10?2m at pH 7.4 and 5.8, respectively. At pH 7.4, precipitation was better in 0.015 m NaCl than in 0.15 m NaCl for both CuCl2 and ZnCl2. Little or no conformational change was indicated on binding Cu2+ ions. The fluorescence of tryptophan was quenched only by CuCl2, while other metal ions (ZnCl2, NiCl2, CoCl2 and CaCl2) were ineffective as quenchers.  相似文献

4′-Iodo-, 4′-bromo-, 4′-chloro- and 4′-fluoro-2,3,4,5-tetrachlorobiphenyl were administered to immature male Wistar rats and the effects of this homologous series of 4′-halo-2,3,4,5-tetrachlorobiphenyls on the microsomal drug-metabolizing enzymes were determined. All the halogenated biphenyls increased microsomal benzo[a]pyrene hydroxylase (or aryl hydrocarbon hydroxylase, AHH), ethoxyresorufin (ER) O-deethylase and dimethylaminoantipyrine (DMAP) N-demethylase. The effects of the 4′-halo-2,3,4,5-tetrachlorobiphenyls on the microsomal enzyme activities and on the relative peak intensities and spectral shifts of the reduced cytochrome P-450:CO and ethylisocyanide (EIC) binding difference spectra were similar to those observed after coadministration of phenobarbitone (PB) and 3-methylcholanthrene (MC). The relative activities of the halogenated biphenyls were determined using two invitro assays; namely cytochrome P-448 associated induction in rat hepatoma H-4-II E cells in culture and competitive binding to the hepatic cytosolic Ah receptor protein from male Wistar rats. Dose-response experiments for the iodo, bromo, chloro and fluoro analogs gave EC50(M) values of 8.5×10?9, 6.6×10?8, 5.7×10?7, and 3.3×10?5, and 1.5×10?6, 2.5×10?6, 4.1×10?6 and 2.5×10?5 for the ER O-deethylase induction and receptor binding assays respectively. The relative potencies of the 4′-halo-2,3,4,5-tetrachlorobiphenyls followed the order I>Br>Cl>F for both assays and differences in the EC50 values for the iodo and fluoro analogs were greater than three orders of magnitude for ER O-deethylase induction in rat hepatoma cells in culture. One possible explanation for these effects may be associated with differences in the polarizability of the laterally substituted halogen groups. However, other differences in the physico-chemical properties of the halogen atoms may also be important.  相似文献

Propoxyphene (Darvon) was compared to SKF 525-A, a prototypical inhibitor of hepatic microsomal mixed function oxidases, to assess propoxyphene's potential to inhibit drug metabolism in morphine tolerant/dependent mice. In vitro, both propoxyphene (Ki = 3.5 × 10?5M) and SKF 525-A (Ki = 4.3 × 10?6M) inhibited the activity of aminopyrine N-demethylase competitively in hepatic microsomes from tolerant/dependent animals. Propoxyphene and SKF 525-A were weaker, noncompetitive inhibitors of aniline hydroxylase activity. In vivo, equimolar doses (0.24 mmoles/kg, i.p.) of each compound inhibited both of the above monooxygenases in the 10,000g supernatant fractions of livers from the tolerant/ dependent animals. Propoxyphene was 40–50% as potent an inhibitor of these activities as SKF 525-A. A dose (300 mg/kg) of propoxyphene napsylate, shown to prevent narcotic abstinence signs with no observable toxicity in withdrawing mice, significantly prolonged the blood levels of injected pentobarbital and tripled pentobarbital sleeping time in these animals. When administered at 300 mg/kg chronically, propoxyphene napsylate acted as an inducer of its own metabolism. Propoxyphene napsylate, then, given acutely to narcotic tolerant/dependent mice, is a potent inhibitor of microsomal drug metabolizing capacity. Experiments were designed to determine whether or not various antiestrogens in direct competition with estradiol-17β (E2) would inhibit the translocation of the estrogen receptor complex from the cytoplasm to nuclei in rat uterine tissue. Incubation of the antiestrogens CI-628, cis-clomiphene, U-11,100A and MER-25 with rat uteri caused the nuclear uptake of the antiestrogen receptor complex which was greatest for most antiestrogens at concentrations of 1 × 10?6 to 1 × 10?5M. At higher concentrations of CI-628, cis-clomiphene, and U-11,100A the nuclear binding of the antiestrogen receptor complex was greatly decreased. Incubation of the antiestrogens with E2 resulted in a dramatic inhibition of the nuclear uptake of the estrogen receptor. Trans-clomiphene, a weak estrogen, did not inhibit the movement of the uterine cytoplasmic receptor into the nuclear fraction.  相似文献

Kinetic studies of the reduction of ferrioxamine B (Fe(Hdesf)+) by Cr(H2O)62+, V(H2O)62+, and dithionite have been performed. For Cr(H2O)62+ and V(H2O)62+, the rate is ?d[Fe(Hdesf)+]/dt = k[Fe(Hdesf)+][M2+]. For Cr(H2O)62+, k = 1.19 × 104 M?1 sec?1 at 25°C and μ = 0.4 M, and k is independent of pH from 2.6 to 3.5. For V(H2O)62+, k = 6.30 × 102 M?1 sec?1 at 25°C, μ = 1.0 M, and pH = 2.2. The rate is nearly independent of pH from 2.2 to 4.0. For Cr(H2O)62+ and V(H2O)62+, the activation parameters are ΔH = 8.2 kcal mol?1, ΔS ?12 eu and ΔH = 1.7 kcal mol?1, ΔS = ?40 eu (at pH 2.2) respectively. Reduction by Cr(H2O)62+ is inner-sphere, while reduction by V(H2O)62+ is outer-sphere. Reduction by dithionite follows the rate law ?d[Fe(Hdesf)+]/dt =kK12[Fe(Hdesf)+][S2O42?]12 where K is the equilibrium constant for dissociation of S2O42? into SO2? radicals. The value of k at 25°C and μ = 0.5 is 2.7 × 103 M?1 sec?1 at pH 5.8, 3.5 × 103 M?1 sec?1 at pH 6.8, and 4.6 × 103 M?1 sec?1 at pH 7.8, and ΔH = 6.8 kcal mol?1 and ΔS = ?19 eu at pH 7.8.  相似文献
