Films based on TiO2 nanoparticles (NPs) have been successfully used as sensing elements in chemical sensors. TiO2 colloidal suspensions can be obtained by spontaneous hydrolysis in acidic solutions of Ti(IV) compounds. The obtained TiO2 NPs can be employed to build up nanostructured films. With the purpose of preparing TiO2-based nanostructured, imprinted materials as sensing elements for piezoelectric sensors, we obtained TiO2 NP dispersions by hydrolyzing potassium titanyl oxalate in the presence of a target analyte (tyrosine). Since morphological properties of the synthesized NPs are known to influence the nanostructured film characteristics, an analytical strategy to characterize such colloidal systems can combine a size-based separation method with spectroscopic analysis to correlate the particle size distribution (PSD) with the particle-target interaction properties able to determine the sensing efficiency.In this work, we present the characterization of colloidal tyrosine-TiO2 NP systems by flow field-flow fractionation (FlFFF) with online, UV/Vis absorption detection and offline fluorescence analysis. FlFFF eliminates the possible contribution of free tyrosine to the absorption and fluorescence properties of the NPs. FlFFF also fractionates NPs on a size basis. Particle size distribution (PSD) profiles of the fractionated NPs are then obtained by conversion of the multi-wavelength UV/Vis fractograms. Size of the fractionated NPs is finally related to fluorescence properties of the collected NPs fractions. Good correlation between the fluorescence intensity, which is proportional to the tyrosine uptake, and the FlFFF-based, NP mass-size frequency distribution finally confirms the existence of tyrosine-TiO2 NP interaction.  相似文献

Listeria monocytogenes is the agent of listeriosis, a food-borne disease. It represents a serious problem for the food industry because of its environmental persistence mainly due to its ability to form biofilm on a variety of surfaces. Microrganisms attached on the surfaces are a potential source of contamination for environment and animals and humans. Titanium dioxide nanoparticles (TiO2 NPs) are used in food industry in a variety of products and it was reported that daily exposure to these nanomaterials is very high. Anti-listerial activity of TiO2 NPs was investigated only with UV-irradiated nanomaterials, based on generation of reactive oxigen species (ROS) with antibacterial effect after UV exposure. Since both Listeria monocytogenes and TiO2 NPs are veicolated with foods, this study explores the interaction between Listeria monocytogenes and non UV-irradiated TiO2 NPs, with special focus on biofilm formation and intestinal cell interaction. Scanning electron microscopy and quantitative measurements of biofilm mass indicate that NPs influence both production and structural architecture of listerial biofilm. Moreover, TiO2 NPs show to interfere with bacterial interaction to intestinal cells. Increased biofilm production due to TiO2 NPs exposure may favour bacterial survival in environment and its transmission to animal and human hosts.  相似文献

Nanotechnology is becoming increasingly important for products used in our daily lives, such as the masses of titanium dioxide nanoparticle agglomerates (TiO2 NPs) used in the pharmaceutical industry, for cosmetic products, or for pigments. Meanwhile, a serious lack of detailed information concerning the interaction between the nanomaterials and cells limits their biological and medical applications. Sensing technology is very important for understanding these interactions. We have shown that TiO2 NPs induce heat shock protein 70B' (HSP70B') mRNA [Okuda‐Shimazaki et al., 2010. Int J Mol Sci 11:2383–2392]. In the current work, sensor cells for detection of cellular responses to NPs were prepared by transfecting an HSP70B' promoter–reporter plasmid. First, to find suitable cells for detection, five different mammalian cell lines were chosen as potential sensor cells. The results showed TiO2 NP response in some cell lines, although different sensor cells had different TiO2 NP response levels, as heat shock response ability is important for the detection. Then, we studied the TiO2 NP time‐course response and dose response. The results indicated that our sensor cells can detect TiO2 NP cellular responses. Titanium dioxide (TiO2) anatase nanoparticles (NPs) are metal oxide NPs commercialized for several uses of everyday life. However their toxicity has been poorly investigated. Cellular internalization of NPs has been shown to activate macrophages and neutrophils that contribute to superoxide anion production by the NADPH oxidase complex. Transmission electron micrososcopy images showed that the membrane fractions were close to the NPs while fluorescence indicated an interaction between NPs and cytosolic proteins. Using a cell-free system, we have investigated the influence of TiO2 NPs on the behavior of the NADPH oxidase. In the absence of the classical activator molecules of the enzyme (arachidonic acid) but in the presence of TiO2 NPs, no production of superoxide ions could be detected indicating that TiO2 NPs were unable to activate by themselves the complex. However once the NADPH oxidase was activated (i.e., by arachidonic acid), the rate of superoxide anion production went up to 140% of its value without NPs, this effect being dependent on their concentration. In the presence of TiO2 nanoparticles, the NADPH oxidase produces more superoxide ions, hence induces higher oxidative stress. This hyper-activation and the subsequent increase in ROS production by TiO2 NPs could participate to the oxidative stress development.  相似文献

为了阐明纳米二氧化钛颗粒(TiO2NPs)对生菜(Lactuca sativa)生长的影响,采用自行设计的水培装置探究不同浓度TiO2NPs (300~1 200 mg/L)下,生菜生长和生理生化指标的变化。结果表明,300 mg/L TiO2NPs能促进生菜幼苗的根长、茎长、叶表面积、鲜重和干重;随着TiO2 NPs浓度增大,生菜的生长指标呈现下降趋势,但仍优于对照组。生菜体内的抗氧化酶(SOD、POD)在低TiO2 NPs浓度(300 mg/L)时,活性明显下降;随着TiO2 NPs浓度增大,这两种抗氧化酶活性逐渐增强。因此,生菜对TiO2NPs胁迫具有浓度依赖性,表现为“低促高抑”,且能够通过抗氧化酶系统来减轻TiO2NPs伤害。  相似文献

In the present work, a bionanocomposite for plant crop protection was prepared by non-toxic biocompatible & biodegradable nanomaterials (Cellulose & TiO2) to utilize its synergistic effects against antimicrobial pathogens. The commercially available microcrystalline cellulose has been reduced to a nanometric scale regime using acid hydrolysis, while the standard TiO2 nano-powder of particle size ~20 nm has been used to prepare their nanocomposite (NC). The antibacterial studies via agar well diffusion method demonstrated that after 72 h of incubation, parent nanomaterials Ncell and TiO2 were not showing any activity against phytopathogens X. campestris pv. campestris, and Clavibacter while the nanocomposite's NC's were still effective depicting both bacteriostatic and bactericidal actions. However, the bacterial growth of biocontrol P. fluorescence was not affected by Ncell, TiO2 NPs and NC after 72 h of incubation. The antifungal testing results via poison food agar assay method suggest that the nanocomposite, along with Ncell and TiO2 NPs, exhibited strong inhibition of fungal growth of Phytophthora Spp at 0.125 mg/ml concentration while for F. graminearum, similar effect was observed at 0.25 mg/ml concentration. The nanocomposite has proved its potential by exhibiting longer & stronger synergistic effects against plant pathogens as a good antimicrobial agent for protection of agricultural crops.  相似文献

Polycyclic aromatic hydrocarbons (PAH), like Benzo[alpha]Pyrene (BaP) are known to cause a number of toxic manifestations including lung cancer. As Titanium dioxide Nanoparticles (TiO2 NPs) have recently been shown to adsorb a number of PAHs from soil and water, we investigated whether TiO2 NPs could provide protection against the BaP induced toxicity in biological system. A549 cells when co-exposed with BaP (25 µM, 50 µM and 75 µM) along with 0.1 µg/ml,0.5 µg/ml and 1 µg/ml of TiO2 NPs, showed significant reduction in the toxic effects of BaP, as measured by Micronucleus Assay, MTT Assay and ROS Assay. In order to explore the mechanism of protection by TiO2 NP against BaP, we performed in silico studies. BaP and other PAHs are known to enter the cell via aromatic hydrocarbon receptor (AHR). TiO2 NP showed a much higher docking score with AHR (12074) as compared to the docking score of BaP with AHR (4600). This indicates a preferential binding of TiO2 NP with the AHR, in case if both the TiO2 NP and BaP are present. Further, we have done the docking of BaP with the TiO2 NP bound AHR-complex (score 4710), and observed that BaP showed strong adsorption on TiO2 NP itself, and not at its original binding site (at AHR). TiO2 NPs thereby prevent the entry of BaP in to the cell via AHR and hence protect cells against the deleterious effects induced by BaP.  相似文献

Titanium dioxide nanoparticles are massively produced and widely used in daily life, which has posed potential risk to human health. However, the molecular mechanism of TiO2 nanoparticles (NPs) with different crystal phases is not clear. In this study, the characterization of two crystalline phases of TiO2 NPs is evaluated by transmission electron microscopy and X-ray absorption fine structure spectrum; an interaction of these TiO2 NPs with HaCaT cells is studied in vitro using transmission electron microscopy, chemical precipitation method, and X-ray absorption fine structure spectrometry. The coordination and surface properties indicate that only the anatase–TiO2 NPs allow spontaneous reactive oxygen species (ROS) generation, but rutile–TiO2 NPs do not after dispersion. The interaction between TiO2 NPs and cellular components might also generate ROS for both anatase–TiO2 NPs and rutile–TiO2 NPs. The ROS generation could lead to cellular toxicity if the level of ROS production overwhelms the antioxidant defense of the cell or induces the mitochondrial apoptotic mechanisms. Furthermore, Ti had a direct combination with some protein or DNA after NPs enter the cell, which could also lead to cellular toxicity.  相似文献

Bombyx mori nucleopolyhedrovirus (BmNPV) causes infection in the silkworm that is often lethal. The infection is hard to prevent, partly because of the nature of the virus particles and partly because of the different strains of B. mori. Titanium dioxide nanoparticles (TiO2 NPs) have been demonstrated to have antimicrobial properties. The present study investigated whether TiO2 NPs added to an artificial diet can increase the resistance of B. mori larvae to BmNPV and examined the molecular mechanism behind any resistance shown. The results indicated that ingested TiO2 NPs decreased reactive oxygen species and NO accumulation in B. mori larvae under BmNPV infection, which in turn led to a decrease in their growth inhibition and mortality. In addition, the TiO2 NPs significantly promoted the expression of resistance-related genes, including those encoding superoxide dismutase, catalase, glutathione peroxidase, acetylcholine esterase, carboxylesterase, heat shock protein 21, glutathione S transferase o1, P53, and transferring and of genes encoding cytochrome p302 and nitric oxide synthase. These findings are a useful addition to the understanding of the mechanism of BmNPV resistance of B. mori larvae in response to TiO2 NPs addition. Such information also provides a theoretical basis for the use of TiO2 NPs in sericulture.  相似文献

Superparamagnetic Fe3O4 nanoparticles (NPs) based nanomaterials have been exploited in various biotechnology fields including biomolecule separation. However, slow accumulation of Fe3O4 NPs by magnets may limit broad applications of Fe3O4 NP-based nanomaterials. In this study, we report fabrication of Fe3O4 NPs double-layered silica nanoparticles (DL MNPs) with a silica core and highly packed Fe3O4 NPs layers. The DL MNPs had a superparamagnetic property and efficient accumulation kinetics under an external magnetic field. Moreover, the magnetic field-exposed DL MNPs show quantitative accumulation, whereas Fe3O4 NPs single-layered silica nanoparticles (SL MNPs) and silica-coated Fe3O4 NPs produced a saturated plateau under full recovery of the NPs. DL MNPs are promising nanomaterials with great potential to separate and analyze biomolecules.  相似文献

Several authors have highlighted the potential risks of nanoparticles (NPs). Still, little is known about the magnitude of emissions of NPs from society. Here, the method of explorative particle flow analysis (PFA), a modification of the more well‐known substance flow analysis (SFA), is suggested. In explorative PFA, particle number instead of mass is used as flow and stock metric and explorative scenarios are used to account for potential technology diffusion and, consequently, potentially higher emissions. The method has been applied in a case study of the use phase of titanium dioxide (TiO2) NPs in paint, sunscreen and self‐cleaning cement. The results indicate that the current largest emissions of TiO2 NPs originate from the use of sunscreen. One scenario implies that, in the future, the largest flows and stocks of TiO2 NPs could be related to self‐cleaning cement. Gaps in current knowledge are identified and suggestions for future research are given.  相似文献

PurposeTitanium dioxide nanoparticles (TiO2 NPs) have been investigated for their role as radiosensitisers for radiation therapy. The study aims to increase the efficiency of these NPs by synthesising them with samarium.MethodsSamarium-doped TiO2 NPs (Ti(Sm)O2 NPs) were synthesised using a solvothermal method. Transmission electron microscopy (TEM), X-ray diffraction (XRD), and energy-dispersive X-ray spectroscopy (EDS) were performed for characterising of the Ti(Sm)O2 NPs. The intracellular uptake and cytotoxicity were assessed in vitro using A549 and DU145 cancer cell lines. Furthermore, the effect of dose enhancement and generation of reactive oxygen species (ROS) in response to 6 MV X-rays was evaluated. Additionally, the image contrast properties were investigated using computed tomography (CT) images.ResultsThe synthesised Ti(Sm)O2 NPs were about 13 nm in diameter as determined by TEM. The XRD pattern of Ti(Sm)O2 NPs was consistent with that of anatase-type TiO2. EDS confirmed the presence of samarium in the nanoparticles. At 200 μg/ml concentration, no differences in cellular uptake and cytotoxicity were observed between TiO2 NPs and Ti(Sm)O2 NPs in both A549 and DU145 cells. However, the combination of Ti(Sm)O2 NPs and X-rays elicited higher cytotoxic effect and ROS generation in the cells than that with TiO2 NPs and X-rays. The CT numbers of Ti(Sm)O2 NPs were systematically higher than that of TiO2 NPs.ConclusionsThe Ti(Sm)O2 NPs increased the dose enhancement of MV X-ray beams than that elicited by TiO2 NPs. Samarium improved the efficiency of TiO2 NPs as potential radiosensitising agent.  相似文献

Rapid commercialization, industrialization and the use of nanotechnology has led to an increase in the distribution of nanoparticles (NPs) in the environment. The most common metal oxide NPs which is present within products is Titanium dioxide (TiO2). TiO2 NPs have photocatalytic nature and can affect plant growth. The current study investigated the morphological, anatomical and biochemical features of Baby sun rose (Aptenia cordifolia) after exposure to different concentrations of TiO2 nanoparticles (0, 1, 5, 10 and 20 mg L−1). Treatment with TiO2 NPs showed changes in the morphological features and increased photosynthetic pigmentation within the plant. An increase in the level of phenolics (12%) and flavonoid compounds (13%) was observed when plants were treated with moderate levels of TiO2 NPs. A reduction in the diameter of the vascular bundles and increased thickening of the transverse wall were observed in several samples. The number of scattered vascular bundles in the stems increased. The morphological, biochemical, and anatomical responses of Baby sun rose indicates that plants can adapt to environments contaminated with up to 20 mg L−1 TiO2 NPs. The cultivation of Baby sun rose plants in environments polluted with TiO2 NPs is recommended. This study enhances the knowledge of the effect of TiO2 NPs on the growth of Baby sun rose which is an ornamental plant, widely cultivated in different regions of Iran. The results of this study suggest that contaminated environments up to 20 mg L−1 TiO2 NPs can be managed by phytoremediation. Further studies are needed to investigate this plant''s tolerance strategies against stress caused by TiO2 NPs and bulk TiO2 as well as the effect of other nanoparticles on plant.  相似文献

近年来,二氧化钛纳米颗粒(TiO2NPs)环境释放量不断增加,并通过多种途径进入湿地生态系统,不可避免地影响到湿地生态系统环境和功能。然而,关于TiO2NPs对沼泽土壤反硝化作用和氧化亚氮(N2O)排放的影响机及制尚不明确。选择典型沼泽土壤,通过室内培养实验研究土壤理化性质、反硝化酶活性、反硝化速率(DNR)和N2O排放对不同剂量TiO2NPs 0 mg/kg (CK)、10 mg/kg (A10)、100 mg/kg (A100)、1000 mg/kg (A1000)输入的响应,探讨TiO2NPs输入对沼泽土壤反硝化作用和N2O排放影响的内在机制。结果表明:不同剂量TiO2NPs处理显著降低了土壤pH (P<0.05),A10处理显著降低土壤总有机碳(TOC)含量(P<0.01),A1000处理显著降低硝态氮(NO3--N)和亚硝态氮(NO2--N)含量(P<0.05)。TiO2NPs处理抑制硝酸盐还原酶(NAR)活性,促进一氧化氮还原酶(NOR)和氧化亚氮还原酶(NOS)活性(P<0.01),A1000处理先促进后抑制了亚硝酸盐还原酶(NIR)活性(P<0.05)。不同剂量TiO2NPs处理抑制了土壤DNR,促进了N2O排放,TiO2NPs处理通过抑制NIR活性,降低土壤DNR,同时通过促进NOR活性,提高N2O排放。综上,TiO2NPs输入通过影响反硝化还原酶活性改变沼泽土壤反硝化过程,导致沼泽土壤N2O排放增加,改变湿地氮的源、汇功能,影响全球气候变化。为TiO2NPs输入的湿地环境风险评估研究提供理论基础。  相似文献

Various nanoparticles, such as silver nanoparticles (AgNPs) and titanium nanoparticles (TiO2NPs) are increasingly used in industrial processes. Because they are released into the environment, research into their influence on the biosphere is necessary. Among its other effects, dietary TiO2NPs promotes silk protein synthesis in silkworms, which prompted our hypothesis that TiO2NPs influence protein kinase B (Akt)/Target of rapamycin (Tor) signaling pathway (Akt/Tor) signaling in their silk glands. The Akt/Tor signaling pathway is a principle connector integrating cellular reactions to growth factors, metabolites, nutrients, protein synthesis, and stress. We tested our hypothesis by determining the influence of dietary TiO2NPs (for 72 h) and, separately, of two Akt/Tor pathway inhibitors (LY294002 and rapamycin) on expression of Akt/Tor signaling pathway genes and proteins in the silk glands. TiO2NPs treatments led to increased accumulation of mRNAs for Akt, Tor1 and Tor2 by 1.6‐, 12.1‐, and 4.8‐fold. Dietary inhibitors led to 2.6‐ to 4‐fold increases in mRNAs encoding Akt and substantial decreases in mRNAs encoding Tor1 and Tor2. Western blot analysis showed that dietary TiO2NPs increased the phosphorylation of Akt and its downstream proteins. LY294002 treatments led to inhibition of Akt phosphorylation and its downstream proteins and rapamycin treatments similarly inhibited the phosphorylation of Tor‐linked downstream proteins. These findings support our hypothesis that TiO2NPs influence Akt/Tor signaling in silk glands. The significance of this work is identification of specific sites of TiO2NPs actions.  相似文献

Mesoporous Ln(III)-TiO2 (Ln = Tb, Eu, Sm) nanomaterials composites have been successfully synthesized by using sol-gel technique.XRD pattern, FT-IR, Raman spectra, and SEM were used to characterize the Ln(III)-TiO2 nanomaterials. The prepared lanthanide doped TiO2 nanomaterials have anatase phase and exhibit Ti-O-Ln bond. The absorption spectra of all prepared samples reflect the increasing photoresponse of doped samples to visible light over pure TiO2. Surface area is remarkably increased due to lanthanide ion-doping.Two newly prepared Ln(III)-TiO2 (Ln = Eu, Sm) luminescent nanomaterials exhibit enhanced pure red or orange light emission due to energy transfer from host TiO2 to guest Eu(III) or Sm(III), respectively.In addition, the commercially available textile dye Remazol Red RB-133 degradation was used as a probe reaction to determine the efficiency of the Ln(III)-TiO2 photocatalysts. The Ln(III) doping brought about remarkable improvement in the photoactivity over pure TiO2.  相似文献

Controversial and inconsistent results on the eco-toxicity of TiO2 nanoparticles (NPs) are commonly found in recorded studies and more experimental works are therefore warranted to elucidate the nanotoxicity and its underlying precise mechanisms. Toxicities of five types of TiO2 NPs with different particle sizes (10∼50 nm) and crystal phases were investigated using Escherichia coli as a test organism. The effect of water chemistry on the nanotoxicity was also examined. The antibacterial effects of TiO2 NPs as revealed by dose-effect experiments decreased with increasing particle size and rutile content of the TiO2 NPs. More bacteria could survive at higher solution pH (5.0–10.0) and ionic strength (50–200 mg L−1 NaCl) as affected by the anatase TiO2 NPs. The TiO2 NPs with anatase crystal structure and smaller particle size produced higher content of intracellular reactive oxygen species and malondialdehyde, in line with their greater antibacterial effect. Transmission electron microscopic observations showed the concentration buildup of the anatase TiO2 NPs especially those with smaller particle sizes on the cell surfaces, leading to membrane damage and internalization. These research results will shed new light on the understanding of ecological effects of TiO2 NPs.  相似文献

To ensure the safe use of nanoparticles (NPs) in modern society, it is necessary and urgent to assess the potential toxicity of NPs. Cardiovascular system is required for the systemic distribution of NPs entering circulation. Therefore, the adverse cardiovascular effects of NPs have gained extensive research interests. Metal based NPs, such as TiO2, ZnO and Ag NPs, are among the most popular NPs found in commercially available products. They may also have potential applications in biomedicine, which could increase their contact with cardiovascular systems. This review aimed at providing an overview about the adverse cardiovascular effects of TiO2, ZnO and Ag NPs. We discussed about the bio-distribution of NPs following different exposure routes. We also discussed about the cardiovascular toxicity of TiO2, ZnO and Ag NPs as assessed by in vivo and in vitro models. The possible mechanisms and contribution of physicochemical properties of metal based NPs were also discussed.  相似文献

BackgroundTitanium is widely used in biomedicine. Due to biotribocorrosion, titanium dioxide (TiO2) nanoparticles (NPs) can be released from the titanium implant surface, enter the systemic circulation, and migrate to various organs and tissues including the brain. A previous study showed that 5 nm TiO2 NPs reached the highest concentration in the brain. Even though TiO2 NPs are believed to possess low toxicity, little is known about their neurotoxic effects. The aim of the study was to evaluate in vitro the effects of 5 nm TiO2 NPs on a human neuroblastoma (SH-SY5Y) cell line.MethodsCell cultures were divided into non-exposed and exposed to TiO2 NPs for 24 h. The following were evaluated: reactive oxygen species (ROS) generation, apoptosis, cellular antioxidant response, endoplasmic reticulum stress and autophagy.ResultsExposure to TiO2 NPs induced ROS generation in a dose dependent manner, with values reaching up to 10 fold those of controls (p < 0.001). Nrf2 nuclear localization and autophagy, also increased in a dose dependent manner. Apoptosis increased by 4- to 10-fold compared to the control group, depending on the dose employed.ConclusionsOur results show that TiO2 NPs cause ROS increase, induction of ER stress, Nrf2 cytoplasmic translocation to the nucleus and apoptosis. Thus, neuroblastoma cell response to TiO2 NPs may be associated with an imbalance of the oxidative metabolism where endoplasmic reticulum-mediated signal pathway seems to be the main neurotoxic mechanism.  相似文献

This study is focused on the fabrication and characterization of titanium oxide (TiO2) NPs. Afterwards; the interaction of TiO2 NPs with human hemoglobin (Hb) was investigated by FTIR spectroscopy, fluorescence spectroscopy, and molecular docking studies. Also, the cytotoxic effect of fabricated TiO2 NPs against human white blood cells (WBCs) was considered by MTT assay. The antibacterial effect of synthesized NPs was examined on Pseudomonas aeruginosa (ATCC 27853); Escherichia coli (ATCC 25922) and Staphylococcus aureus (ATCC 25923). TEM and DLS investigations showed that the synthesized TiO2 NPs have a narrow nano-sized distribution. XRD pattern of the fabricated NPs exhibited that the TiO2 NPs contain anatase phase. Similarity in amide I and II signal intensities showed that secondary structure of the adsorbed Hb is preserved. The intrinsic fluorescence study revealed that the fluorescence quenching of Hb was done by complex formation between Hb and TiO2 NPs trough the hydrogen bond and van der Waals interactions. Synchronous fluorescence spectroscopy determined that interaction of TiO2 NPs with Hb did not unfold the Hb structure in the vicinity of the Tyr and Trp residues. Molecular docking study depicted that Glu-95, Thr-134 and Tyr-140 are involved in the formation of hydrophilic bonds. MTT data and antibacterial assays indicated that TiO2 NPs endow distinguished antibacterial activities against Gram-negative and Gram positive strains at safe concentrations. This study may reveal that fabricated TiO2 NP can be used as a safe and potent antibacterial agent.
