With applied to the petioles of detached Begonia x cheimantha leaves before planting, Gibberellic acid (GA3) inhibited the formation of adventitious buds and roots ill an apparently irreversible manner. Bud formation was entirely suppressed by 10?6M and higher concentrations and a significant inhibition was still present at 10?9M the lowest concentration tested. Root formation was not affected by GA3 below 10?7M and was possible even at 10?4 M GA3. Petiole elongation was stimulated by GA3 with an optimum at 10?5M. GA3 also blocked the action of 6-benzyiamino-purine (BAP) and 1-naphthaleneacetic acid (NAA), compounds which are potent stimulators of bud and root formation, respectively. When applied simultaneously with GA3 they were, at their optimal concentrations, devoid of any effect in counteracting or reversing the gibberellin-induced inhibitions. Abscisic acid and the growth retardants CCC and Phosfon also were unable to restore bud and root formation. In leaves initially treated with water or 10?5M BAP, endogenous bud and root formation as well as BAP-induced bud formation were entirety suppressed when 10?5M GA3 was applied 8 days after the initial treatments. Even when delayed for 14 days GA3 treatment inhibited BAP-induced bud formation, while treatment after 21 days bad little effect on bud and root formation. Development of pre-existing, visible bud primordia was not inhibited by GA3. BAP and NAA competitively inhibited the action of GA3 in petiole extension growth. The results are discussed in relation to results obtained in other plant systems. It is suggested that GA3 acts by blocking of the organized cell divisions initiating the formation of bud and root primordia.  相似文献

The elongation growth of the Avena first internode segments was studied in the presence of one or several of the following growth substances: indoleacetic acid (IAA), 6-fur-furylamino purine (FAP, kinetin), 6-benzylamino purine (BAP), gibberellin A3 (GA3) and A4+7 (GA4+7), and abscisic acid (ABA). The cytokinins at concentrations of 10?7 to 10?6M stimulated growth with 4 to 6 per cent but this effect was not statistically significant. Concentrations higher than 5 × 10?6M inhibited growth. FAP and BAP (from 10?8M to 10?6M) had no significant interaction with any other growth substance used. The two-factor interactions of IAA × ABA, IAA × GA3, and GA3× ABA, as well as the three-factor interaction IAA × ABA × GA3 were significant. However, the IAA × ABA interaction was significant only when high concentration (10?6M) of ABA was used. The growth inhibition produced by 10?7 and 10?6M ABA was overcome by about equimolar concentrations of IAA. The stimulation of growth by GA3 and GA4+7 (10?9 to 10?7M) was prevented by simultaneous application of ABA, and it was reduced significantly by application of IAA (10?7 to 10?8M). GA3 at 10?8M combined with different concentrations of IAA gave slightly higher elongation than IAA alone but the observed values were significantly lower than expected assuming independent additive action.  相似文献

Pea plants (Pisum sativum L. cv. Alaska) were grown from seeds for eleven days at different irradiances. Cuttings were then excised and rooted at 16 W × m?2. Gibberellic acid (GA3, 10?11 to 10?3M) was applied to the cuttings immediately after excision. Cuttings from stock plants grown at the highest level of irradiance (38 W × m?2) formed the lowest number of roots. An increasing number of roots per cutting was obtained by decreasing the irradiance to the stock plants. In cuttings from stock plants grown at low irradiances, low concentrations of GA3 (10?8 and 10?7M) promoted root formation further. No effect on rooting by these GA3 concentrations was observed when applied to cuttings originating from stock plants grown at the high irradiances. Root formation in all cuttings was inhibited by GA3 at concentrations higher than 10?6M. The degree of inhibition by GA3 was influenced by the irradiance pretreatment and was increased with an increase in the irradiance during the stock plant growth. Seeds from different years produced cuttings with different response patterns regarding the irradiance and GA3 effects on rooting.  相似文献

In oilseeds, storage lipids provide the respiratory fuel for seedling growth. The enzyme responsible for their initial hydrolysis is lipase (triacylglycerol acylhydrolase; EC 3.1.1.3). We investigated the possibility that lipase is regulated by gibberellins (GAs). In four oilseed rape cultivars of Brassica napus and B. rapa, seed imbibition in 10?6 to 10?3M GA3 increased lipase activity 1.5- to 7-fold over control levels. Conversely, imbibition in 10?7 to 10?5M abscisic acid or 10?6 to 10?4M paclobutrazol, an inhibitor of GA biosynthesis, markedly decreased lipase activity. While lipase activity in B. napus cv. Parkland increased during the first 5 days following imbibition, concentrations of endogenous GA1, GA8 and GA19 (as measured by GC-selected ion monitoring using [2H2]GA internal standards) were relatively constant and GA20, a precursor of GA1, decreased. Levels of endogenous GA3 were apparently variable. Thus, lipase activity was not correlated with GA1 concentration, but the inverse correlation with GA20 concentration suggests that GA turnover could be positively correlated with lipase activity. Lipase activity was also examined in three genotypes of rapid cycling B. rapa that vary in endogenous GA content: rosette, a GA-deficient dwarf, a normal line and elongated internode, a tall mutant with high GA content. The three genotypes showed similar patterns of lipase activity during the first 4 days following imbibition and the subcellular distribution of lipase activity was also similar in the three genotypes. Although GA may be involved in the regulation of lipase in oilseed rape germinants, it is not the sole regulatory factor.  相似文献

In Schizaeaceae ferns, including Anemia phyllitidis, formation of antheridia is known to be induced by exogenously applied gibberellic acid. Also present studies show that GA3 (10−5 mol·dm−3) modifies the development of gametophytes of Anemia phyllitidis. Simultaneously with formation of antheridia, they exhibit lower number of cells but only slightly lowered profile areas and lengths of prothalli. Growth in size of individual cells compensates for lowered division frequency. Cytophotometric measurements reveal no essential changes in the DNA content in vegetative cells of the control and GA3-stimulated gametophytes. It remains at haploid level and therefore it is assumed that cell cycle is blocked at G1 phase. Application of GA3 increases the total amount of proteins. CZE (Capillary Zone Electrophoresis) separation of peptides extracted from control and GA3-treated prothalli indicates the differences in the ratio of their particular forms. In GA3-treated gametophytes the activities of acid and basic phosphatases, contents of carbohydrates (glucose, starch), chlorophyll, the number of chloroplasts and dry mass of prothalli are increased. GA3-intensified metabolism, evidenced in gametophytes of A. phyllitidis, may be interpreted as a stimulatory mechanism which influences metabolic pathways involved in forming, developing and maturing of male sex organs.  相似文献

BA at 10–5 M, GA3 at 3×10–4 M or GA4+7 at 3×10–5 M partially or largely reversed the inhibition of Amaranthus caudatus seed germination due to JA-Me. BA or GA3 did not affect ethylene production and ACC oxidase activity in vivo in the presence of JA-Me before radicle protrusion. However, both increased ethylene production after 72 h of incubation, when the reversal of the JA-Me inhibition of seed germination was observed. AVG at 3×10–4 M decreased ethylene production when it was applied simultaneously with BA and JA-Me or GA3 and JA-Me, but it had no effect on seed germination. NBD almost completely reversed the stimulatory effect of BA, GA3 or GA4+7 on the germination of seeds in the presence of JA-Me. Exogenous ethylene reversed the inhibitory effect of NBD. The results indicate that action of endogenous ethylene is involved in the response of JA-Me inhibited seeds to BA or GAs.  相似文献

The purpose of this study was to analyze the nature of the interaction between gibberellic acid (GA3) and abscisic acid (ABA) in the regulation of growth in excised Avena (oat) stem segments. Growth, compared to sucrose controls, was inhibited by ABA in the range of 10?4 to 10?6M. GA3-promoted growth was also inhibited by ABA in the same concentration range. A Lineweaver-Burk analysis of the interaction between GA3 and ABA indicated that ABA acts in a non-competitive fashion with GA3. This same result was obtained previously with GA3-indoleacetic acid (IAA) and GA3-kinetin interactions with Avena stem sections. Our results indicate that ABA can inhibit GA3-promoted growth within physiological concentrations, and that it is probably acting at a different physiological site from that for GA3.  相似文献

金毛狗[Cibotium barometz(L.)J.Sm.]是珍稀观赏蕨类的重要类群,为国家二级重点保护野生植物。该研究以金毛狗孢子为试验材料,探究培养密度、外源赤霉素以及光质等不同环境因子对金毛狗配子体性别分化的影响,为金毛狗人工繁育和蕨类植物配子体性别决定机制研究提供技术支持。结果表明:(1)低配子体培养密度(1个/cm2和5个/cm2)有利于颈卵器和雌配子体形成,随着配子体培养密度增加,颈卵器平均数量及雌配子体比率下降,精子器平均数量以及雄配子体和两性配子体比例增加,但配子体培养密度过高(80个/cm2)会导致大量无性配子体产生。(2)不同配子体培养密度下,随着培养时间延长,两性配子体比率均有增加,且增加幅度基本一致。(3)外源GA4显著抑制颈卵器和雌配子体形成,并显著促进精子器和雄配子体形成;外源GA3对金毛狗配子体性别分化没有显著影响。(4)白光、红光、蓝光等不同光质对金毛狗配子体性别分化未产生显著影响,但会影响配子体的发育和形态建成。  相似文献

The growth response (increase in weight) of cultured explants from seedling date (Phoenix dactylifera L.) and mature coconut (Cocos nucifera L. cv. Malayan Dwarf) palms to source and concentration of organic nitrogen. carbohydrate, auxins, cytokinins and gibberellins was examined. Growth was strongly stimulated by the presence of auxins (10?7 to 10?6M), cytokinins (10?6 to 10?5M), high concentrations of sucrose (0.2 M), and in the absence of NH4Cl, by organic sources of reduced nitrogen. Higher concentrations of auxin (2,4-D or NAA at 10?6 to 10?5M) which still stimulated growth of Phoenix tissue, proved inhibitory to growth of freshly excised Cocos tissues. Explants from both palms initiated roots when subcultured on a medium with increased levels of auxin (NAA, 2.5 × 10?6 to 2.5 × 10?5M) and reduced levels of cytokinin (6-BAP, 5 × 10?8M). Isolated roots excised from these explants continued growth and produced new laterals when subcultured on media with GA3 (5 × 10?7M) and reduced levels of auxin, cytokinin, and either minerals or sucrose.  相似文献

InEquisetum arvense, apogamous sporophytes were produced on medium containing 5×10?6–5×10?8 g/ml kinetin. NAA, IAA, GA3, glucose and saccharose were ineffective for the induction of apogamy. On medium containing 5×10?7–5×10?8 g/ml kinetin, the gametophytes passed into sporophytic structures directly. On medium containing 5×10?6 g/ml kinetin, some gametophytes passed into sporophytic structures directly, and others became a callus-like cell mass from which an apogamoun shoot arose. The results of the morphological observations on them were reported and compared with the sexually produced sporophyes. The apogamous sporophytes induced by 5×10?7 g/ml kinetin were haploid in their nuclear phase and some of those induced by 5×10?6 g/ml kinetin had a tendency to become diploid.  相似文献

The season dependent changes in growth response to treatment with auxin or gibberellin were studied in the aquatic macrophyte Ceratophyllum demersum. Control plants show, under experimental conditions, a maximum growth in length in February. In the same period most of the lateral buds appear. Growth of the lateral buds occurs later. IAA causes a stimulation of growth in length from late November or December until February, in concentrations of 10?9M and 10?6M. There is almost no stimulation of lateral bud formation by IAA, only a slight increase from late November until December occurs by the lowest concentrations used. The highest concentration used, 10?4M, is in most cases supraoptimal for lateral bud formation; only when plants become dormant (August), this high dose may stimulate the process. GA3, in concentrations of 10?9, 10?6 or 10?4M, exhibits a dose dependent increase of the response with respect to growth in length and lateral bud formation. The response occurs earlier than that for IAA: already early in November, or December, until February. Growth of the lateral buds may show only a slight stimulation by IAA as well as GA in winter. From February until April all GA concentrations used could cause a small increase of the growth of sprouts. In the case of IAA, however, only the lowest concentration could cause a small increase.  相似文献

Summary The effect of exogenous applications of gibberellins (GAs) or the growth retardant -chloroethyltrimethylammonium chloride (CCC) on root nodule formation and activity (C2H2-reduction) in soya was studied. Daily foliar application of GA3 (2.89×10–6 M) delayed the formation of nodule initials and reduced the numbers mass nodule–1 and specific activity of nodules by 43%, 31% and 47% respectively, without affecting plant growth. Similar effects on nodulation were produced by foliar application of GA4 (3.01×10–5 M) or GA7 (3.03×10–5 M), or by the addition of GA3 (2.89×10–6 M) to the rooting medium. GA effectiveness in reducing nodule numbers was decreased by delaying its application until after the initial infection process had occurred, but the nodules formed were smaller and less active than those of the untreated control plants. The GA effect on nodulation and nodule activity was not associated with alterations in root exudate or due to a direct inhibitory effect of the hormone on the nitrogenase system. When the endogenous root content of GA-like substances was reduced (86% decrease) by foliar application of CCC (6.30×10–5 M), nodule numbers were increased by 56%, but nodule size and total nodule activity were similar to those of control plants. The GA and CCC treatments had no effect on rhizobial growth in liquid culture nor on root colonisation by rhizobia.The results suggest that the endogenous content of root GA may have a regulatory role in both the infection process and in subsequent nodule morphogenesis, thus controlling both the number and effectiveness of the root nodules formed.  相似文献

The effects of a plant-derived smoke extract, BA and GA3 on the thermodormancy of Grand Rapids lettuce seeds were studied. Thermodormant lettuce seeds treated either with BA, GA3 or smoke extract alone did not germinate. Combinations of BA with smoke extract and BA with GA3 were most effective in overcoming induced thermodormancy. GA3 plus smoke did not break the induced thermodormancy. The effects of the different treatments on germination were concentration dependent. Narrow concentration intervals were used, covering 10?6– 10?4M desaspidin. The interaction with glycolysis involves three steps, the inhibitor constants (Ki:s) being in turn 2.7 × 10?5M, 1.3 × 10?4M, and high. About 18% of total glycolysis is inhibited in each of the two first steps, and 65% left for the third reaction. After compensation for glycolysis, oxidative phosphorylation may show a sudden jump to about 10% inhibition at 1.5 × 10?5M desaspidin, the possible Ki of the reaction starting here being very high. Correcting for glycolysis, desaspidin affects total Photophosphorylation in two steps, with the Ki values of 7.8 × 10?5M and 4.6 × 10?4M respectively. Inhibition in the first step is about 27% of the total photophosphorylation. By applying 10?6M DCMU[/3-(3, 4-dichlorophenyl)-l, l-dimethy lurea], one can abolish non-cyclic photophosphorylation. Desaspidin then reacts in a single step with a Ki of 1.4 × 10?4M. At 5 × 10?5M DCMU, also the pseudocyclic photophosphorylation is abolished. The remaining, true cyclic photophosphorylation has a single Ki of 2.3 × 10?5M for desaspidin. Under non-cyclic conditions, the true cyclic process contributes about 25% to total Photophosphorylation. Under pseudocyclic conditions, no cyclic photophosphorylation occurs. Under true cyclic conditions, the non-cyclic and pseudocyclic processes are inoperative. This indicates a regulative system, so that either (1) the (non-cyclic + true cyclic), (2) only the pseudocyclic, or (3) only the true cyclic systems can be traced, dependent on the level of DCMU applied. There are two sites for non-cyclic Photophosphorylation, one of them common to the pseudocyclic pathway. Cyclic photophosphorylation has a third site, different from the other two.  相似文献

The roles of gibberellic acid (GA3) and ethylenediaminetetraacetic acid (EDTA) in phytoremediation of cadmium (Cd)-contaminated soil by Parthenium hysterophorus plant was investigated. GA3 (10?9, 10?7, and 10?5M) was applied as a foliar spray. EDTA was added to soil in a single dose (160 mg/kg soil) and split doses (40 mg/kg soil, four split doses). GA3 and EDTA were used separately and in various combinations. P. hysterophorus was selected due to its fast growth and unpalatable nature to herbivores to reduce the entrance of metal into the food chain. The Cd phytoextraction potential of the P. hysterophorus plant was evaluated for the first time. Cd significantly reduced plant growth and dry biomass (DBM). GA3 alone increased the plant growth and biomass in Cd-contaminated soil, whereas EDTA reduced it. GA3 in combination with EDTA significantly increased the growth and biomass. The highest significant DBM was found in treatment T3 (10?5M GA3). All treatments of GA3 or EDTA significantly enhanced the plant Cd uptake and accumulation compared with control (C1). The highest significant root and stem Cd concentrations were found in the combination treatment T11 (GA3 10?5M + EDTA split doses), whereas in leaves it was found in the EDTA treatments. Cd concentration in plant parts increased in the order of stem < leaves < roots. The combination treatment T9 (GA3 10?7M + EDTA split doses) showed the significantly highest total Cd accumulation (8 times greater than control C1, i.e., only Cd used). The GA3 treatments accumulated more than 50% of the total Cd in the roots, whereas the EDTA treatments showed more than 50% in the leaves. Root dry biomass showed a positive and significant correlation with Cd accumulation. GA3 is environment friendly as compared with EDTA. Therefore, further investigation of GA3 is recommended for phytoremediation research for the remediation of metal-contaminated soil.  相似文献

Physiological (metabolite analysis) and molecular (gene expression) approaches were used to understand the mechanism underlying russet formation in response to the application of GA3 and CPPU (Forchlorfenuron) in a Japanese table grape cultivar ‘Shine Muscat’. Several different concentrations of GA3 and GA3?+?CPPU [25?mg?L?1 GA3 (A), 25?mg?L?1 GA3?+?5?mg?L?1 CPPU (B), 25?mg?L?1 GA3?+?10?mg?L?1 CPPU (C), and 25?mg?L?1 GA3?+?15?mg?L?1 CPPU (D)] were applied to grape berry clusters at two weeks after flowering (WAF). No russet was observed on the berries treated with the ‘C’ combination. Lower levels of phenylalanine ammonia-lyase (PAL) activity was observed in the treated samples, relative to the untreated material. Reduced peroxide (POD) activity was also observed in response to different treatments, while the expression of Peroxidase 17 and Phenylalanine ammonia-lyase G1 genes mirrored lignin content. Increased activity of 4-coenzyme A ligase (4CL) may contribute to decreasing the level of russet and help to improve grape berry quality.  相似文献

Endopeptidase activity in cotyledons of 5-day seedlings of Pisum sativum increased rapidly during germination. However, the increase of the activity in detached cotyledons was depressed. We examined whether a growth regulator can be substituted for the embryonic axis on the development of endopeptidase activity. As monitored by an assay with azoalbumin, the development of endopeptidase activity from crude extracts of detached cotyledons appeared to be slightly accelerated by incubation with 10–5 M GA3. However, the pattern after gelatin-polyacrylamide gel suggested that the activity induced in detached cotyledons during a 5-d incubation at 10–7 M GA3 was the same as that in attached ones during germination for 5 days and an even greater increase in activity was obtained with 10–5 M GA3. The effect of application of GA3 on hypocotyl growth, the endogenous GAs, and the metabolism of applied 3H-GA1 were investigated in relation to dwarfism and light-mediated growth inhibition in the normal (tall) strain Violet and the dwarf strain Kidachi of Japanese morning glory (Pharbitis nil). GA3 applied in a wide concentration range (10−9 to 10−3m) to 4-day-old seedlings caused great extension of the hypocotyls in light-grown plants of both the normal and the dwarf strain. However, the dwarf strain did not attain the same length as the normal one at any given GA3 concentration, even when saturation was reached. Dark-grown plants of the dwarf strain responded to GA3, although relatively much less than light-grown ones; dark-grown plants of the normal strain showed no GA3 response at all.  相似文献

Effects of the ethylene precursor 1-aminocyclopropane-1-carboxylic acid (ACC) on the development and expression of male sex were tested using the model of the three-zonal structure of 12-day-old (15-celled) Anemia phyllitidis gametophyte. ACC at 10 M concentration enhanced the number of antheridia induced by gibberellic acid. Cytomorphological measurements showed that this effect was limited to only the antheridial region of gametophytes and depended on transverse expansion of antheridial mother cells. Abstract: The muscarinic receptors involved in phosphoinositide (PI) hydrolysis have been pharmacologically characterized in cat cerebral blood vessels. Carbachol elicited a concentration-dependent increase in inositol phosphate accumulation [inositol monophosphate, bisphosphate, trisphosphate (IP3) and tetrakisphosphate] in both major cerebral arteries and small pial vessels, which reached 140–280% of baseline at 10?3M carbachol (referred to as maximal effect). However, the inositol phosphate accumulation response was found to be biphasic with a submaximal effect (30–50% of the maximal stimulation) obtained at low carbachol concentrations (<10?5M). Endothelial denudation induced a virtual disappearance of the submaximal PI response without affecting that elicited by high concentrations of carbachol. The pharmacology of the two carbachol-induced PI responses was investigated by comparing the potency of selected muscarinic antagonists to block the IP3 accumulation induced by 10?7M (endothelium-dependent submaximal effect) and 10?4M (endothelium-independent near-maximal effect) carbachol. In both major arteries and pial vessels, the activation of IP3 production by 10?4M carbachol was similarly inhibited by muscarinic antagonists with the following averaged rank order of potency (in -log IC50): 4-diphenylacetoxy-N-methylpiperidine methiodide (4-DAMP; 8.65) > pirenzepine (8.28) > 6-chloro-5,10-dihydro-5-[(1-methyl-4-piperidinyl)acetyl]-11H-dibenzo[b,e][1,4]diazepine-11-one (UH-AH 371; 7.87) > 11-[[2-[(diethylamino)methyl]-1-piperidinyl]acetyl]-5,-11-dihydro-6H-pyridol[2,3-b][1,4]benzodiazepine-6-one (AF-DX 116; 6.62), a pharmacological profile compatible with an M1 receptor subtype. In contrast, the submaximal stimulation of the PI metabolism elicited by 10?7M carbachol in major arteries was blocked by the same antagonists with the following order of potency (in -log IC50): 4-DAMP (8.38) > pirenzepine (7.25) > UH-AH 371 (6.25) > AF-DX 116 (5.72), which was reminiscent of an M3 pharmacological profile. These findings indicate that stimulation of cerebrovascular muscarinic receptors is accompanied by PI hydrolysis via two distinct receptors, most probably the M1 and M3 subtypes that have been associated with constriction and dilatation, respectively, of cat cerebral arteries. Furthermore, these results provide strong evidence for an endothelial localization of the M3 dilatatory receptors within the vessel wall.  相似文献
