The digestive system of the sea hare, Aplysia depilans , includes a pair of ribbon-shaped salivary glands. A central duct and a large blood vessel run close to each other along the length of these glands and both are surrounded by a layer of muscle cells. Three cell types form the glandular epithelium: granular cells, vacuolated cells and mucocytes. The granular cells possess cilia and spherical secretion granules, located primarily in the apical region. The granules of immature cells have a low electron density and are mainly formed by neutral polysaccharides with small amounts of proteins. The granules of mature cells are larger, have a high electron density and are mainly formed by proteins with lower amounts of neutral polysaccharides. Transition stages between immature and mature granular cells are observed. The vacuolated cells are large and frequently pyramidal in shape, but after the application of histochemical techniques almost all vacuoles remain uncoloured. The numerous vacuoles contain flocculent material in a clear background and the mitochondria possess large crystalline structures in the matrix. A pyramidal shape is also typical of the mucocytes, which are filled with vesicles containing granular masses surrounded by a network of secretion material. These large cells are strongly stained by Alcian blue, revealing the presence of acidic mucopolysaccharides. This is the first ultrastructural study of the salivary glands in opisthobranch gastropods.  相似文献

The amounts of phosphate, zinc, copper and calcium in gonadmaterial from Nucella lapillus (L.) was higher in males comparedwith females. Water insoluble extracts from the digestive glandwere found to contain granules high in concentrations of phosphate,magnesium, zinc, copper and calcium when compared with watersoluble extracts and both types of extract from gonad tissueof male and female dog whelks. Male N. lapillus contained thehighest amounts of zinc and copper in isolated digestive glandgranules. The changes in the ultrastructure of cambial cells of easternwhite pine (Pinus strobus L.) during an annual cycle are observedand recorded as are relationships of cambial cells during dormancyand at resumption of cambial activity. Cambial activity wasresumed late in March or early in April, when a few cells dividedpericlinally. Cambial activity reached a maximum during thelatter part of May with 15 to 20 undifferentiated cells present.In July it declined markedly, and the number of undifferentiatedcells equalled that of the dormant period. The xylem and phloemtissue cells produced late in the annual cycle overwinteredat varying developmental stages. In October cambial cells structurallyresembled dormant cells. The number of dormant cells in easternwhite pine cambium varied from 6 to 10. Active cells were characterizedby a large central vacuole, by an abundance of all cell organelles,and by thin cell walls. The U-shaped alimentary tract of Cephalodiscus is of exclusively epithelial structure; on the basis of fine structural criteria the entire tract can be divided into two large subdivisions: an anterior one with mouth, mouth cavity, pharynx and oesophagus, and a posterior one with stomach and intestine. The anterior subdivision is built up of a relatively uniform, innervated, pseudostratified, ciliated epithelium with mucus cells which are concentrated in the initial parts of the mouth cavity. Cilia and mucus presumably constitute a mechanism transporting food particles into the stomach. In the area of the gill slits specific vacuolated cells occur which may lend rigidity to the walls of the slits. The gastric epithelium consists of prismatic cells characterized by, among others, large inclusion bodies, which may represent digestive vacuoles, small dense rod-shaped granules and an elaborate system of microridges, at the base of which abundant endocytotic vesicles occur. The dorsal gastric pouch contains cells rich in rough ER and secretory granules, probably containing digestive enzymes. Thus morphological evidence points both to intra- and extracellular digestion. The intestinal epithelium resembles that of the stomach, however, it is lower, its organelles are fewer and it bears, beside cilia, mainly microridges, which towards its distal end become sparse. Both in the gastric and intestinal epithelium small granulated cells have been found, which presumably represent endocrine cells.  相似文献

Abstract. The ribbon‐shaped salivary glands in Bulla striata were studied with light microscopy and transmission electron microscopy (TEM). Secretion is produced in tubules formed by two types of secretory cells, namely granular mucocytes and vacuolated cells, intercalated with ciliated cells. A central longitudinal duct lined by the same cell types collects the secretion and conducts it to the buccal cavity. In granular mucocytes, the nucleus is usually central and the secretory vesicles contain oval‐shaped granular masses attached to the vesicle membrane. Glycogen granules can be very abundant, filling the space around the secretory vesicles. These cells are strongly stained by PAS reaction for polysaccharides. Their secretory vesicles are also stained by Alcian blue, revealing acidic mucopolysaccharides, and the tetrazonium reaction detects proteins in minute spots at the edge of the vesicles, corresponding to the granular masses observed in TEM. Colloidal iron staining for acidic mucopolysaccharides in TEM reveals iron particles in the electron‐lucent region of the vesicles, while the granular masses are free of particles. In vacuolated cells, which are thinner and less abundant than the granular mucocytes, the nucleus is basal and the cytoplasm contains large electron‐lucent vesicles. These vesicles are very weakly colored by light microscopy techniques, but colloidal iron particles could be observed within them. The golf tee‐shaped ciliated cells contain some electron‐dense lysosomes in the apical region. In these cells, the elongated nucleus is subapically located, and bundles of microfibrils are common in the slender cytoplasmic stalk that reaches the basal lamina. The morphological, histochemical, and cytochemical data showed some similarities between salivary glands in B. striata and Aplysia depilans. These similarities could reflect the phylogenetic relationship between cephalaspidean and anaspidean opisthobranchs or result from a convergent adaptation to an identical herbivorous diet.  相似文献

Ultrastructural studies (SEM and TEM) were performed on cotyledonsof pineapple guava ( Feijoa sellowiana Berg, Myrtaceae) inducedto form embryos on medium containing 1.0 mg l-1(4.5µM2,4-D) and 0.3M sucrose. At the time of culture, the cells werefilled with protein and lipid bodies. Microbodies and poorlydifferentiated organelles could also be seen. In contrast togerminating cotyledons, where lipid and protein reserves werequickly metabolized, cells of the embryogenically induced cotyledonsshowed evidence of reserve consumption only after 5 d of culture.Subepidermal cells of the upper cotyledonary surface underwentseveral divisions giving rise to a meristematic layer of severalcells thickness from which somatic embryos developed. Embryoscould also be formed directly by successive divisions of epidermalcells. Cells involved in somatic embryo formation containeda large nucleus with a conspicuous nucleolus and dense cytoplasmwhere numerous ribosomes, mitochondria, plastids with starchand short profiles of rough endoplasmic reticulum were present.Plasmodesmata were present both in cell walls of the meristematiccells and in few celled embryos whereas in degenerating embryosor in more advanced stages of somatic embryo development noplasmodesmata could be found. Although oil bodies were not observedin the meristematic cells they were identified in very youngembryos, being the first reserve compounds to appear. Cellsnot involved in somatic embryo differentiation were characterizedby the presence of several microbodies containing a crystalloidinclusion and elongated mitochondria. The possible involvement of enzymes in the penetration of intrusivecells of the parasitic angiospermOrobancheinto host root tissueswas studied using cytochemical and immunocytochemical methods.Pectin methyl esterase (PME) was detected, with specific antibodies,in the cytoplasm and cell walls ofOrobancheintrusive cells andin adjacent host apoplast. Depletion and chemical changes ofpectins in host cell walls were shown by histochemical stainingwith PATAg, which detects carbohydrates that are sensitive toperiodic acid, especially pectins, and with the monoclonal antibodiesJIM 5 and JIM 7 that label pectins with low and high rates ofesterification, respectively. Galacturonic sequences with lowrates of esterification were more abundant in host cell wallsadjacent to the parasite, which is consistent with pectin de-methylationby PME release from the parasite. Pectins were absent in middlelamellae and in host cell walls neighbouring mature intrusivecells of the parasite, consistent with further degradation ofpectins by other enzymes. In the glume of Phalaris canariensis L. silicon deposition takesplace in the macrohairs, papillae, prickle hairs and silicacells of the abaxial epidermis before panicle emergence. Early in their development the macrohairs have large vacuolesand thin walls. At maturity the walls become thickened and aremajor sites of silica deposition. Dry ashing reveals a helicalpattern within the hair walls. Distinct papillae and prickle hairs were first observed oneweek before panicle emergence. Here silicification was initiallyconfined to outer tangential walls, but by two weeks after emergencetheir cytoplasmic contents had broken down, and the lumina werefilled with siliceous granules. Cork-silica twin cells werealso present in the abaxial epidermis. By panicle emergencethe silica cells were infilled, but the cork cells retainedtheir cytoplasmic contents. The long cells of the abaxial epidermiswere initially thin walled, but thickening occurred in the outertangential wall, this being complete by one week after emergence.These cells remained relatively unsilicified throughout. After panicle emergence the adaxial epidermal cells, and theirassociated parenchyma cell layers began to lose their cellularcontents and collapse. This process was complete two weeks afteremergence when the collapsed walls formed a thin internal layerbetween the two epidermi. Electron opaque granular material,containing several elements, but predominantly calcium, waspresent between the collapsed cell walls. The endophyte, that is, the haustorial part within the tissues of the host plant Impatiens balsamina, of the parasitic angiosperm Cuscuta japonica was studied with light and electron microscopy. The endophyte consisted mainly of vacuolated parenchymatous axial cells and elongate, superficial (epidermal) cells. Then the elongate, epidermal cells separated from each other and transformed into filamentous cells, called searching hyphae. The hyphae grew independently either intercellularly or intracellularly in the host parenchyma. The apical end of the hyphal cells was characterized by conspicuous, large nuclei with enlarged nucleoli and very dense cytoplasm with abundant organelles, suggesting that the hyphal cells penetrating host tissue were metabolically very active. Numerous osmiophilic particles and chloroplasts were noted in the hyphae. The osmiophilic particles were assumed to be associated with elongation of the growing hyphe. Plasmodemata connections between the searching hyphal cells of the parasite and the host parenchyma cells were not detected. Hyphal cells that reached the host xylem differentiated into water-conducting xylic hyphae by thickening of the secondary walls. A xylem bridge connecting the parasite and the host was confirmed from serial sections. Some hyphal cells that reached the host phloem differentiated into nutrient-conducting phloic hyphae. Phloic hyphae had a thin layer of peripheral cytoplasm with typical features of sieve-tube members in autotrophic angiosperms, i.e., parallel arrays of smooth endoplasmic reticulum, mitochondria, and plastids with starch granules. Interspecific open connections via the sieve pores of the host sieve elements and plasmodesmata of the parasite phloic hyphae were very rarely observed, indicating that the symplastic translocation of assimilate to the parasite from the host occurred.  相似文献

The ventriculus and the midgut caeca of the fed females of Anystis baccarum (L.) were investigated by using light and electron microscopy. In addition to the main type of polyfunctional digestive cells, special secretory cells were detected in the anterior region of the ventriculus. The shape and the ultrastructure of the digestive cells vary depending on their physiological state. Intracellular digestion, absorption or excretion processes prevail at different stages of the cell cycle. The secretory cells are characterized by the presence of extensive rough endoplasmic reticulum, filling whole space of the cell. These cells do not contain the apical network of pinocytotic canals, which are typical for the digestive cells. Three types of secretory granules were found in the cytoplasm of the secretory cells that probably correspond to three sequential stages of granulogenesis. The primary secretory granules are formed by the fusion of Golgi vesicles. The primary granules fuse to form complex vesicles with heterogeneous contents. These secondary granules aggregate to form very large inclusions of high electron density (tertiary secretory granules), which probably represent the storage of the secretory product. All types of secretory granules were observed close to the apical plasmalemma.  相似文献

Endosperm cells of lettuce (Lactuca sativa L.) are characterizedby thick cell walls and dense cytoplasm which contains numerousprotein bodies. Other organelles such as nucleus, mitochondria,plastids and dictyosomes are typical of plant cells. Light andelectron microscopy reveal that before radicle emergence micropylarcells of endosperm tissue undergo drastic protoplast alterations.These alterations seem to be the only structural modificationsbefore rupturing of the tissue since the walls of the endospermcells seem to degrade only after radicle emergence. The differentialbehaviour of the micropylar area of the endosperm before radicleemergence and the observation that the micropylar cells remainmetabolically active long after radicle emergence while therest of the tissue is almost completely disintegrated, suggeststhat the endosperm cells of the micropylar area may have a roleother than being a main reserve site like the rest of the endosperm. The anatomy and histology of the digestive tract of the suspensionfeeding bivalve Lyonsia hyalina were examined using microdissectionand conventional light microscopy. Lyonsia hyalina has a typeIV stomach which contains a major typhlosole that does not penetratethe left pouch, as in other members of the Pandoroidea. Theventral and posterior sorting areas of the stomach are sitesof vigorous ciliary activity. The gastric shield is locatedon the left and posterior stomach walls, underlain by tall basophiliccells with microvilli that project into the gastric shield.The style sac and midgut are combined, and contain the morphologicalcell types A-D seen in other bivalves. Many ciliated cells ofthe digestive tract appear to have high densities of apicalmitochondria. The ducts within the digestive diverticula arelined by epithelia containing a conspicuous brush border. Bothcrypt cells and digestive cells exist in the digestive tubules.The presence of numerous fragmentation spherules throughoutthe digestive diverticula indicates that intracellular digestionoccurs there. The effects of thyroidectomy, adrenalectomy, and castration on the pars distalis of male Japanese quail, and of injection of LH-RH on sexually inactive females, were investigated by light and electron microscopy. Correlation between light and electron microscopy was attained by use of alternate thin and thick sections. Six types of secretory cells were identified and the ultrastructural characteristics described. Putative endocrine functions have been designated on the basis of responses to experimental interventions and on other criteria. The putative STH cells are characterized by the presence of large dense secretory granules (250-300 nm) that are stained with orange-G by the trichrome method. They occur only in the caudal lobe and appear to be unchanged by castration, thyroidectomy, adrenalectomy and LH-RH injection. The putative prolactin cells are characterized by large (400-600 nm), spherical or polmorphic, dense secretory granules stainable with acid fuchsin and aniline blue; prominent Golgi apparatus and well developed endoplasmic reticulum with densely packed, regularly parallel lamellae. They are found mainly in the cephalic lobe. The prolactin cells develop some vacuolization after adrenalectomy and undergo some degeneration after castration. The ACTH cells, which are restricted to the cephalic lobe, are identified by the dense, spherical granules (250-300 nm) that are stained with acid fuchsin. After adrenalectomy, they lose their secretory granules and are transformed into large, chromophobic adrenalectomy cells. TSH cells are so designated by their response to thyroidectomy including loss of their fine secretory granules and transformation to large, vacuolated thyroidectomy cells. We have found TSH cells and thyroidectomy cells only in the cephalic lobe. Basophilic cells, considered to be gonadotropes, occur in both the cephalic and caudal lobes. The gonadotropes of the cephalic lobe appear to have slightly larger (120-200 nm) granules than the caudal lobe (120-150 nm). However, after castration, the gonadotropes in both lobes become hypertrophied and vacuolated and are transformed into mutually indistinguishable castration cells. Twenty minutes after injection with LH-RH, the gonadotropes of both lobes increase in size and number, degranulate, develop vacuoles in the cytoplasm, and appear very similar to castration cells.  相似文献

Specific antibodies were used to localize both pectic structuresand pectinmethylesterases (PME) along the mung bean hypocotyl.Calcium ions were also detected and estimated in both young,plastic and mature, stiffened cell walls. Highly methylesterifiedpectins were present in all cell walls but decreased from thehypocotyl hook downwards. Expanded cell walls were characterizedby a high content of calcium ions and acidic pectins, althoughthe latter's cross-reactivity to JIM 5 antibodies was partlylost. Co-localization of acidic homogalacturonan and calciumions suggests the presence of egg-box structures that mightparticipate in the cell wall stiffening process which developsalong the hypocotyl. Acidic polymers could originate from theactivity of the pectinmethylesterases present in precise wallareas but direct export of acidic polygalacturonan through Golgivesicles was also observed. Abstract The blood cells of a solitary ascidian, Halocynthia roretzi, were examined by electron microscopy (EM) with reference to their appearance by light microscopy (LM). In addition, their movement and stainability by vital dyes was observed by phase-contrast microscopy, and their stainability by Giemsa was also examined. Nine cell types were recognized: vacuolated cells, hyaline amoebocytes, small amoebocytes, granular amoebocytes, macrogranular cells, globular cells, lymphocyte-like cells, large basophilic cells and large granular cells. Vacuolated cells were found to possess various numbers of vacuoles containing strongly electron-dense materials and could be divided into at least three subgroups. Granular amoebocytes contained microfilaments and many granules of uniform size. Hyaline amoebocytes and small amoebocytes seemed to be specialized as phagocytes. Macrogranular cells and globular cells were not well characterized. In the blood of adult individuals, hemoblasts were rarely found, although lymphocyte-like cells were present. Each of two large cells, large basophilic cells and large granular cells, possessed novel granules or vacuoles, whose functions remain to be elucidated. The possible functions and relationships of these cells among various ascidian species are discussed.  相似文献

At maturity, Torenia fournieri(Lind.) has an embryo sac whichprotrudes through the micropyle placing the synergids, egg celland part of the central cell within the ovary locule adjacentto the placenta. The present study utilized this unique attributein combination with confocal and light microscopy to characterizethe timing and associated structural changes during pollinationevents leading to double fertilization. The observation of spermnuclei in living gametophyte tissue is an important advancein the identification, in real time, of stages leading to fertilizationin angiosperms. A continuum of fertilization occurred between12 and 16 h after pollination (hap), with peak frequency ofegg and sperm fusion at 14 hap (43%). Movement of the spermcells through the degenerated synergid took several hours andfusion between sperm and their respective female nuclei occurredsimultaneously. Changes in embryo sac structure were also documented.Cell walls in the region between the synergids and egg cellwere poorly developed prior to pollen tube penetration. Thickenedcell walls were observed around the periphery of the synergidsand egg cell following pollination, and in the central cellwhere it lay within the body of the ovule. Starch was observedin the cells of the embryo sac, although the number and distributionof granules varied before and after pollination. Membranous organelles, acid glycoconjugates and lipids were characterized in the digestive tract mucosa of Hemisorubim platyrhynchos by cytochemistry techniques. Two types of mucous‐secreting cells were observed in the digestive tract epithelium: goblet cells in the oesophagus and intestine and epithelial cells in the stomach. These cells had a Golgi apparatus more developed than the other cell types. The cytochemical analysis revealed that secretory granules are reactive to acid glycoconjugates, varying in reaction intensity according to the region of the digestive tract. Acid glycoconjugate reactions were also observed in oesophageal epithelial cell microridges and in enterocyte microvilli. In the digestive tract, acid glycoconjugates act to protect the epithelial surface, increasing mucous viscosity, which facilitates the passage of food, prevents the binding of parasites and facilitates their removal. Through lipid staining, a coated membrane was observed around each secretory granule of the oesophageal and intestinal goblet cells, while gastric epithelial cells granules were fully reactive. Oxynticopeptic cells of the gastric glands showed lipid droplets in the cytoplasm and also in the mitochondrial matrix, which act as an energy reserve for these cells that have a high energy demand. Enterocytes showed a well‐developed smooth endoplasmic reticulum, especially in the apical region of the cell, being related to absorption and resynthesis of lipids.  相似文献
