Abstract The combined effects of organic matter additions and temperature on short chain fatty acid (SCFA) turnover, sulfate reduction and nutrient accumulation were examined in an organic-rich fish farm sediment. Fish food pellets, which contribute significantly to the organic matter loss from fish farms, were added to surface sediment at three loadings (2.8; 14.0; 28.0 mg ww g−1 ww sediment; equivalent to organic matter loadings measured during fish farming) and incubated for 30 days in anaerobic bags at 5°C and 15°C. SCFA accumulated to high levels (acetate up to 85 mM, propionate up to 17 mM, butyrate up to 25 mM) in sediments amended with food pellets, and sulfate reduction was stimulated up to 30 times relative to unamended sediments. Sulfate reducers appeared saturated with substrates (SCFA) even in the lowest additions. A low C/N ratio (0.4–1.8) of the major mineralization products (TCO2 and NH4+) indicated preferential nitrogen mineralization in amended sediment compared with the total particulate pool (C/N = 8.8–11.9) and added food pellets (C/N = 8.4).  相似文献

Abstract— Partly purified chromaffin granules were incubated in vitro with Ca2+ (with trace amounts of 45Ca2+) in concentrations ranging from 4 μm to 1 mm. After incubation the granules were washed with media containing EDTA and then subjected to density gradient centrifugation (1.3 to 2.0 m-sucrose solutions) in order to characterize the particles which had taken up 45Ca2+. By using marker enzymes and various inhibitors of Ca2+ uptake into such cell particles as mitochondria it was established that under the conditions of the experiments chromaffin granules took up Ca2+ from the incubation medium. To characterize this uptake a simplified density gradient procedure was tested and found to be suitable. The uptake of Ca2+ into chromaffin granules was strongly dependent on temperature. It was not activated by ATP. The uptake was linear up to 10 min. At high calcium concentrations (above 200 μm) the rate of uptake levelled off. The uptake at 37°C was 1 nmol Ca2+/mg protein/min at a Ca2+ concentration of 500 μm. Mg2+ had no influence on Ca2+ uptake, whereas Sr2+ (1 mm) inhibited it. The methods established in this study should prove useful for a further characterization of this Ca2+ uptake into chromaffin granules which is likely to represent a useful model for the Ca2+ uptake occurring in the intact gland.  相似文献

Uptake of amino acids and peptides by developing barley embryos   总被引:1,自引:0,他引:1  
Developing embryos of barley ( Hordeum vulgare L. cv. Bomi) detached 21–27 days after anthesis took up 1 mM [14C]-glutamine at pH 5 and 30°C at a rate of about 20 nmol embryo−l h−1 (5 μol g−1h−1). The uptake was inhibited by about 50% by di-nitrophenol and by about 80% by 300 m M unlabelled glutamine or alanine. The bulk of the uptake appeared, therefore, to be due to carrier-mediated active transport. The pH optimum of the uptake was 4.5. Leucine, proline, lysine, arginine and as-paragine were taken up at approximately similar rates as glutamine, and they also inhibited the uptake of glutamine. This, suggests that the uptake of glutamine was at least partly due to an unspecific carrier(s) also shared by other amino acids. The embryos also took up the dipepti.de glycykarcosine; the rate was about 6 nmol embryo−1h−1 (1.5 μol g−1h−1) (2 mM glycylsarcosine, pH 4.5, 30°C). The uptake was inhibited by about 70% by dinitrophenol or by 300 m M glycylglycine. This indicates that the bulk of the uptake was due to carrier-mediated active transport. The pH optimum of the uptake was about 4.5.
The rates of glutamine and glycylsarcosine uptake increased during the early and middle stages of embryo development (until day 28 after anthesis), but decreased towards the end of the maturation of the grain. These changes, as well as the relatively high activities, suggest that carrier-mediated active uptake of amino acids, and possibly also that of peptides, plays a role in the nutrition of the developing embryo.  相似文献

Abstract The sulfur cycle in a microbial mat was studied by determining viable counts of sulfate-reducing bacteria, chemolithoautotrophic sulfur bacteria and anoxygenic phototrophic bacteria. All three functional groups of sulfur bacteria revealed a maximum population density in the uppermost 5 mm of the mat: 1.1 × 108 cells of sulfate reducers cm−3 sediment, 2.0 × 109 cells of chemolithoautotrophs cm−3 sediment, and 4.0 × 107 cells of anoxygenic phototrophs cm−3 sediment. Bacterial dynamics were studied by sulfate reduction rate measurements, both under anoxic conditions (dark incubation) and oxic conditions (incubation in the light), and determination of the vertical distribution of the potential rate of thiosulfate consumption under oxic conditions. Sulfate reduction rates in the top 5 mm of the sediment were 566 nmol cm−3 d−1 in the absence of oxygen, and 123 nmol cm−3 d−1 in the presence of oxygen. In the latter case, the maximum rate was found in the 5–10-mm depth horizon (361 nmol cm−3 d−1). Biological consumption of amended thiosulfate was rapid and decreased with depth, while in the presence of molybdate, thiosulfate consumption decreased to 10–30% of the original rate.  相似文献

In Escherichia coli , lacZ operon fusions were isolated that were derepressed under iron repletion and repressed under iron depletion. Two fusions were localized in genes that formed an operon whose gene products had characteristics of a binding protein-dependent transport system. The growth defect of these mutants on TY medium containing 5 mM EGTA was compensated for by the addition of Zn2+. In the presence of 0.5 mM EGTA, only the parental strain was able to take up 65Zn2+. This high-affinity transport was energized by ATP. The genes were named znuACB (for zinc uptake; former name yebLMI ) and localized at 42 min on the genetic map of E. coli . At high Zn2+ concentrations, the znu mutants took up more 65Zn2+ than the parental strain. The high-affinity 65Zn2+ uptake was repressed by growth in the presence of 10 μM Zn2+. A znuA–lacZ operon fusion was repressed by 5 μM Zn2+ and showed a more than 20-fold increase in β-galactosidase activity when Zn2+ was bound to 1.5 μM TPEN [tetrakis-(2-pyridylmethyl) ethylenediamine]. To identify the Zn2+-dependent regulator, constitutive mutants were isolated and tested for complementation by a gene bank of E. coli . A complementing gene, yjbK of the E. coli genome, was identified and named zur (for zinc uptake regulation). The Zur protein showed 27% sequence identity with the iron regulator Fur. High-affinity 65Zn2+ transport of the constitutive zur mutant was 10-fold higher than that of the uninduced parental strain. An in vivo titration assay suggested that Zur binds to the bidirectional promoter region of znuA and znuCB .  相似文献

Abstract Thermoanaerobacter thermohydrosulfuricus Rt8.B1 catabolized xylose by the pentose phosphate pathway, and xylose isomerase and xylulokinase were inducible. The uptake of xylose was by two low-affinity, inducible systems. Both systems were resistant to the protonophore, tetrachlorosalicylanilide, the F1F0-ATPase inhibitor, N , N -dicyclohexylcarboiimide, and the sodium/proton antiporter, monensin. The high capacity system (100 nmol min−1 (mg protein)−1) was only expressed when the bacterium was grown with a high concentration of xylose (50 mM). It took more than 60 mM xylose to saturate the high capacity system. When T. thermohydrosulfuricus was grown with a low concentration of xylose (5 mM), xylose uptake was saturated by as little as 10 mM xylose (18 nmol min−1 (mg protein)−1). Cells grown with 50 mM xylose could not transport glucose, and high capacity xylose transport was not inhibited by glucose or non-metabolizable glucose analogues. Cells grown with 5 mM xylose transported glucose at a rapid rate (30 nmol min−1 (mg protein)−1), and low capacity xylose uptake was competitively inhibited by either glucose or 2-deoxy-glucose. Because the glucose uptake of cells grown on 5 mM xylose was competitively inhibited by xylose, it appeared that the low capacity xylose uptake system was a glucose/xylose carrier.  相似文献

Passive fluxes of K+ (86Rb) into roots of sunflower ( Helianthus annuus L. cv. Uniflorus) were determined at low K+ concentration (0.1 and 1.0 mM K+) in the ambient solution. Metabolic uptake of K+ was inhibited by 10−4M 2,4-dinitrophenol (DNP). K+ (86Rb) fluxes were studied both continuously and by time differentiation of uptake. In high K+ roots passive uptake was directly proportional to the K+ concentration of the uptake solution, indicating free diffusion. This assumption was supported by the fact that passive Rb+ uptake was not affected by high K+ concentrations. In low K+ roots the passive uptake of K+ was higher than in high K+ roots. The increase was possibly due to carrier-mediated K+ transport. As K+ effluxes were quantitatively similar to influxes, it is suggested that passive K+ fluxes represent exchange diffusion without relation to net K+ transport.  相似文献

Reed plants ( Phragmites communis Trinius) were grown at NaCl concentrations up to 500 m M and their growth, mineral contents and leaf blade osmotic potential were determined. Addition of NaCl up to 300 m M did not affect growth significantly. Sucrose, Cl-and Na+ concentrations in the shoots increased with the salinity of the medium and the shoot water content decreased. K+ always contributed most to the leaf osmotic potential. Even in the presence of 250 m M NaCl in the rooting medium, the leaf blade contained only 50 mM Na+, suggesting that the plants have an efficient mechanism for Na+ exclusion. 22Na+ uptake experiments suggested that the retranslo-cation of absorbed Na+ from shoots to the rooting medium lowered the uptake of Na+.  相似文献

Abstract Bacterial sulfate reduction and transformations of thiosulfate were studied with radiotracers in a Microcoleus chthonoplastes -dominated microbial mat growing in a hypersaline pond at the Red Sea. The study showed how a diel cycle of oxygen evolution affected respiration by sulfate-reducing bacteria and the metabolism of thiosulfate through oxidative and reductive pathways. Sulfate reduction occurred in both oxic and anoxic layers of the mat and varied diurnally, apparently according to temperature rather than to oxygen. Time course experiments showed that the radiotracer method underestimated sulfate reduction in the oxic zone due to rapid reoxidation of the produced sulfide. Extremely high reduction rates of up to 10 μmol cm−3 d−1 were measured just below the euphotic zone. Although thiosulfate was simultaneously oxidized, reduced and disproportionated by bacteria in all layers of the mat, there was a shift from predominant oxidation in the oxic zone to predominant reduction below. Concurrent disproportionation of thiosulfate to sulfate and sulfide occurred in all zones and was an important pathway of the sulfur cycle in the mat.  相似文献

Taurine entered the alga Chlorella fusca Shihira et Krauss strain 21l-8b via a pH and energy-dependent system ("permease"). Transport followed triphasic kinetics from 10−6 to 10−2 M with Km values for taurine of 5.4 × 10−5, 4.1 × l0−4 and l.5 × 10−3 M. This uptake system was specific for sulfonic acids and showed no affinity for α- and β -amino acids or Na+; thus the permease of C. fusca is different from all known taurine transport systems with respect to structural specificity and lack of Na+ -dependence. Uptake was not observed in sulfate-grown algae but developed as a response to sulfate limitation within 2 h. Sulfate addition caused a rapid decline in taurine transport capacity. Labeled taurine was rapidly metabolized in C. fusca to sulfate and ethanolamine, suggesting oxidative hydrolysis as the mechanism of C-S bond cleavage. Further incorporation of these catabolic products in C - and S -metabolism was demonstrated. Taurine catabolism was also detected in other green algae and some cyanobacteria.  相似文献

Abstract Cell suspensions of Methanobacterium thermoautotrophicum took up 45Ca2+ in a temperature-dependent, Ca2+-saturable and Co2+-sensitive process. The accumulation of 45Ca2+ was lower in the cells energized by CO2+ H2 than in those under non-energized conditions. The accumulated Ca2+ were, in part, released by the divalent cations ionophore A23187 in the presence of EGTA while the uptake of Ca2+ was accelerated by the addition of A23187 to the medium containing Ca2+. The results indicate the presence of a carrier-mediated Ca2+ uptake in the Methanobacterium thermoautotrophicum membrane which is compensated by an energy-dependent and outward-directed Ca2+ transport.  相似文献

Abstract Salterns in the Bretagne (France), exhibiting different stages of salinity of 5% to 33% evolved up to 0.7 mmol methane per m2 per day. Methane concentrations of up to 0.4 mM were found. High methane evolution rates and increased methane concetration were restricted to basins of up to 7% and more than 15% salinity, and to the upper 10 cm of the sediments, where high sulfate concentrations (50 to 100 mM) occurred as well. Basins of 10–15% salinity exhibited only low methane evolution rates (less than 0.05 mmol methane per m2 per day) and low methane concentrations (less than 0.02 mM). Gas bubbles arising during times of increased photosynthetic activity from the microbial mats covering the sediments of the saltern basins contained up to 2% methane. Addition of methylated amines and methanol, but not of H2/CO2, formate or acetate, to sediments slurries from basins of up to 12% salinity resulted in a rapid enrichment of methanogenic populations. Enriched methanogenic bacteria did not grow at salinities exceeding 15% or temperatures exceeding 45°C, and showed characteristics similar to those documented for Methanococcus halophilus and strain SF1 (DSM 3243). No enrichments were obtained from basins of more than 20% salinity in spite of methane being produced and evolved from those basins.  相似文献

Uptake of Rb+ from a complete nutrient solution with 2.0 mM Rb+ was studied in roots of spring wheat seedlings ( Triticum aestivum L. cv. Svenno) with different K+ levels. The relationship between Rb+ uptake and concentration of K+ in the roots indicated a negative feedback mechanism operating through allosteric control. The Rb+ uptake process in root cells was divided into two steps: (1) binding of the ion in the free space, and (ii) transmembrane transport into the cytoplasm. Metabolic and non-metabolic components of uptake were separated by addition of the metabolic inhibitor 2,4-dinitrophenol (DNP) to the nutrient solution. It is suggested that metabolic Rb+ uptake requires energy in two uptake steps (for binding to the carrier entity in the free space and for transmembrane transport) or in one step only (for transmembrane transport), dependent on the K+ status of the roots. The change from metabolic to non-metabolic binding in the free space is accomplished by changing the conformational state of the carrier (slow/fast transitions). There may be a hysteretic effect on metabolic Rb+ uptake through a slow transition between carrier states. This is superimposed on the negative cooperativity, strengthening further cooperativity at intermediate K+ levels in the roots. Non-metabolic Rb+ uptake probably consists of two components, a carrier-mediated (facilitated diffusion) and a parallel diffusive component.  相似文献

The Uptake of Carnitine by Slices of Rat Cerebral Cortex   总被引:5,自引:3,他引:2  
Abstract: The properties of carnitine transport were studied in rat brain slices. A rapid uptake system for carnitine was observed, with tissue-medium gradients of 38 ± 3 for L-[14CH3]carnitine and 27 ± 3 for D-[14CH3]carnitine after 180 min incubation at 37°C in 0.64 mM substrate. Uptake of L- and D-carnitine showed saturability. The estimated values of K m for L- and D-carnitine were 2.85 mM and 10.0 mM, respectively; but values of V max (1 μmol/min/ml in-tracellular fluid) were the same for the two isomers. The transport system showed stereospecificity for L-carnitine. Carnitine uptake was inhibited by structurally related compounds with a four-carbon backbone containing a terminal carboxyl group. L-Carnitine uptake was competitively inhibited by γ-butyrobetaine ( K i= 3.22 mM), acetylcarnitine ( K i= 6.36 mM), and γ-aminobutyric acid ( K i= 0.63 mM). The data suggest that carnitine and γ-aminobutyric acid interact at a common carrier site. Transport was not significantly reduced by choline or lysine. Carnitine uptake was inhibited by an N2 atmosphere, 2,4-dinitrophenol, carbonylcyanide- N -chlorophenylhydrazone, potassium cyanide, n-ethylmaleimide, and ouabain. Transport was abolished by low temperature (4°C) and absence of glucose from the medium. Carnitine uptake was Na+-dependent, but did not require K+ or Ca2+.  相似文献

Uptake of 35S-labelled sulfate was studied with a new isolate of Desulfovibrio desulfuricans, strain CSN. Micromolar additions of sulfate (1–10 M or nmol/mg protein) to cell suspensions incubated in 150 mM KCl at-1°C were almost completely taken up and accumulated about 5,000-fold. Accumulation was not influenced by incubation in NaCl instead of KCl, by acidic pH (5.5) or by incubation under air for 10 min. In alkaline milieu (pH 8.5), after prolonged contact with air (2 h), or after growth with excess sulfate or thiosulfate as electron acceptor, the amount taken up was diminished approximately by half. Pasteurization inhibited sulfate uptake completely. With increasing concentrations of added sulfate (0.1 to 2.5 mM) the intracellular concentration increased only slowly up to 25 mM, and the accumulation factor decreased down to 8. Sulfate transport was reversible. Accumulated sulfate was rapidly lost from the cells after addition of excess non-labelled sulfate or after addition of the uncoupler carbonyl cyanide m-chlorophenylhydrazone (CCCP). The ATPase inhibitor dicyclohexylcarbodiimide (DCCD) specifically inhibited sulfate reduction but had no immediate influence on sulfate accumulation. Addition of the phosphate analogue arsenate (5 mM) was without effect. These results were not in favour of an ATP-dependent transport system. The K+-H+-antiporter nigericin (in 150 mM KCl) and the Na+-H+-antiporter monensin (in 150 mM NaCl) caused partial inhibition of sulfate accumulation, whereas the K+-transporter valinomycin (in 150 mM KCl) and the Na+-H+ exchange inhibitor amiloride (2 mM) were without effect. The permeant thiocyanate anion (150 mM) inhibited sulfate uptake by 60% at pH 7, and completely at pH 8.5. Although the effects of the different ionophores on the chemiosmotic gradients have not been studied so far, the results indicated that probably both, pH and drive sulfate accumulation and that sulfate is taken up electrogenically in symport with more than 2 protons. The structural sulfate analogues tungstate and molybdate (0.1 mM, each) did not affect sulfate accumulation, although molybdate inhibited sulfate reduction. Chromate completely blocked both of these activities. The Mg2+ requirement in fertilization was investigated in sea urchins. It was found that when sea urchin eggs were inseminated in sea water free of Mg2+, little fertilization took place. Even when spermatozoa pre-treated with dissolved egg-jelly to induce the acrosome reaction, which needs Ca2+, were used, the fertilization rate remained quite low in the absence of Mg2+. In Strongylo-centrotus intermedius , the lowest concentration of Mg2+ required for 50% fertilization was 0.05 mM in the presence of 10 mM Ca2+, whereas that of calcium was 3 mM in the presence of 49 mM Mg2+. These critical concentrations increased when the concentration of the other ion decreased. Removal of Mg2+ or Ca2+ or both from the suspending medium had little adverse effect on sperm motility. The elevation of the fertilization membrane was also induced by butyric acid independent of the presence or absence of Mg2+ and/or Ca2+. These results indicate that Mg2+ are required at least in some process(es) between acrosome reaction and fertilization membrane elevation, such as sperm penetration or membrane fusion.  相似文献

Abstract: Adult rat cerebral cortex slices when incubated in media of increasing K+ concentrations, from 3 to 20 mM ("physiological" conditions), exhibited an increase of Ki+ and a decrease of Nai+ and Cli-. This phenomenon appeared at 30 days postnatally, which is fairly late during development. Over 20 mM-K+ ("pathological" conditions) an intense water uptake was observed together with an increase of Nai+ and Cli-. This was observed in both adult and young animals. The results are discussed in relation to the well known properties of glial (Na+,K+)-ATPase and carbonic anhydrase.  相似文献

Iron uptake by clover plants ( Trifoliuin pratense L. cv. Hruszowska) was studied using radioactive ferric iron (55FeCl2). As shown by autoradiography of non-infected plants, purified rhizobial siderophore isolated from Rhizobium leguminosarum by, trifolii , stimulated the uptake and shoot transport of iron. Addition of rhizobial siderophore into the growth medium of nodulated clover did not affect the iron transport to the shoots. In the absence of the rhizobial siderophore, clover infected by either nitrogen-fixing (Nod Fix+) or nonfixing (Nod+ Fix) R. L. trifolii strains took up and transported into the shoots more iron than the non-infected control plants. Nodulated clover reduced Fe(III) more efficiently than the non-infected control plants.  相似文献

Abstract: To study mechanisms of K+ transport in peripheral nerve, uptake of rubidium (Rb+), a K+ tracer, was characterized in rat tibial nerve myelinated axons and glia. Isolated nerve segments were perfused with zero-K+ Ringer's solutions containing Rb+ (1–20 m M ) and x-ray microanalysis was used to measure water content and concentrations of Rb, Na, K, and Cl in internodal axoplasm, mitochondria, and Schwann cell cytoplasm and myelin. Both axons and Schwann cells were capable of removing extracellular Rb+ (Rb+o) and exchanging it for internal K+. Uptake into axoplasm, Schwann cytoplasm, and myelin was a saturable process over the 1–10 m M Rb+o concentration range, although corresponding axoplasmic uptake rates were higher than respective glial velocities. Mitochondrial accumulation was a linear function of axoplasmic Rb+ concentrations, which suggests involvement of a nonenzymatic process. At 20 m M Rb+o, a differential stimulatory response was observed; i.e., axoplasmic Rb+ uptake velocities increased more than fivefold relative to the 10 m M rate, and glial cytoplasmic uptake rose almost threefold. Finally, Rb+o uptake rate into axons and glia was completely inhibited by ouabain (2–4 m M ) exposure or incubation at 4°C. These results suggest that Rb+ uptake into peripheral nerve internodal axons and Schwann cells is mediated by Na+,K+-ATPase activity and implicate the presence of axonal- and glial-specific Na+ pump isozymes.  相似文献

Abstract: The Na+ and K+ concentrations in isolated Torpedo marmorata synaptosomes were determined. Synaptosomes made according to the method of Israël et al. have high internal Na+ (290 MM) and low internal K+ (30 mM) concentrations. Modification of the homogenisation media permitted the isolation of synaptosomes which could maintain transmembrane ion gradients (internal Na+, 96 mM; K+, 81 mM); 0.1 mM-ouabain abolished these gradients. The trans-membrane Na+ gradient started to dissipate after 15 min at 20°C. Inclusion of ATP in the homogenisation medium enabled the synaptosomes to maintain the Na+ gradient for about 90 min. The presence of these transmembrane ion gradients stimulated choline uptake sevenfold. It is concluded that (a) by selecting the isolation media, Torpedo synaptosomes can be prepared with transmembrane ion gradients; (b) these gradients are ouabain-sensitive and stimulate choline uptake: (c) the synaptosomes require additional ATP to maintain the ion gradients.  相似文献
