A strain 5-1-2T was isolated from a root nodule of Hedysarum multijugum collected from Zhangye city, Gansu province, north-west China. Phylogenetic analysis based on the 16S rRNA gene sequence and other housekeeping genes (recA and atpD) indicated that the strain represents a novel species in the genus Rhizobium close to the strain Rhizobium subbaraonis JC85T with similarities of 98.27, 88.92 and 89.62%, respectively. Strain 5-1-2T contained Q-10 as the predominant ubiquinone. Our results showed that the major fatty acids were feature 8 (C18:1 ω7c and/or C18:1 ω6c; 38.90%). In addition, the DNA–DNA hybridizations with the type strains R. subbaraonis JC85T and Rhizobium halophytocola YC6881T were 39.2 ± 2.1 and 44.3 ± 1.9, respectively. Therefore, a novel species Rhizobium hedysari sp. nov. is proposed, and 5-1-2T (=CGMCC1.15677T = NBRC112532T) is designated as the type strain. 相似文献

Studying rhizobia in the root nodules of Sphaerophysa salsula (Pall.) DC in the northwest of China, we obtained five strains classified as genus Rhizobium on the basis of their 16S rRNA gene sequences. The sequence similarity of strain CCNWQTX14T with the most related species was 99.0%. Further phylogenetic analysis of housekeeping genes (recA and atpD) suggested the five strains comprised a novel lineage within Rhizobium. The nifH and nodD gene sequences of CCNWQTX14T were phylogenetically closely related with those of Sinorhizobium kummerowiae and R. sphaerophysae, respectively. The five strains isolated from different places were also distinct from related Rhizobium species using ERIC fingerprint profiles. The DNA–DNA hybridization value was 41.8% between CCNWQTX14T and Rhizobium sphaerophysae CCNWGS0238T. Our novel strains were only able to form effective nodules on its original host Sphaerophysa salsula. Our data showed that the five Rhizobium strains formed a unique genomic species, for which a novel species Rhizobium

A Gram-negative, yellow-pigmented bacterial strain, designated IPC6T, was isolated from soil in an arid region of Goyang-si (Gyeonggi-do, South Korea). Cells were strictly aerobic, non-spore-forming, rod-shaped. The strain grew within a temperature range of 10–42°C (optimum, 30°C) and pH of 5.0–11.0 (optimum, pH 8.0) in the presence of 0–2% (w/v) NaCl. Phylogenetically, the novel strain was closely related to members of the Lysobacter genus based on 16S rRNA sequence similarity, and showed the highest sequence similarity to Lysobacter niastensis KACC 11588T (98.5%). The predominant fatty acids were iso-C15:0, iso-C16:0, and summed feature 9 (iso-C17:1ω9c), with Q-8 identified as the major ubiquinone. The polar lipid content included diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, an unknown aminophospholipid, and an unidentified phospholipid. DNA-DNA hybridization results indicated that the strain IPC6T was distinct from Lysobacter niastensis KACC 11588T (37.9 ± 0.14%), Lysobacter panacisoli KACC 17502T (56.4 ± 0.13%), Lysobacter soli KCTC 22011T (8.1 ± 0.04%), Lysobacter gummosus KCTC 12132T (9.6 ± 0.03%), and Lysobacter cavernae KCTC 42875T (37.5 ± 0.14%), respectively. The DNA G + C content of the novel strain was 71.1 mol%. Based on the collective phenotypic, genotypic and chemotaxonomic data, the IPC6T strain is considered to represent a novel species in the genus Lysobacter, for which the name Lysobacter pedocola sp. nov. A novel actinomycete strain, designated PAL84, was isolated from a Saharan soil sample collected from Béni-Isguen, Ghardaïa (South of Algeria). This strain was studied for its taxonomic position using a polyphasic approach and was identified as a member of the genus Actinokineospora. Phylogenetic analysis showed that strain PAL84 had 16S rRNA gene sequence similarities with members of the genus Actinokineospora ranging from 96.2 % (Actinokineospora inagensis DSM 44258T) to 97.8 % (Actinokineospora baliensis NBRC 104211T). The strain was observed to produce pinkish-purple aerial mycelium and purplish red substrate mycelium, which fragmented readily into chains of non-motile elements. The optimum growth temperature and pH were found to be 25–30 °C and 5.0–7.0, respectively. The cell-wall hydrolysate of strain PAL84 was found to contain meso-diaminopimelic acid and the diagnostic whole-cell sugars were identified as arabinose and galactose. The predominant menaquinone was identified as MK-9 (H4). The major fatty acids were found to be iso-C16:0, iso-C15:0, iso-C16:1 H and iso-C16:0 2OH. The diagnostic phospholipid detected was phosphatidylethanolamine. The genotypic and phenotypic data show that the strain represents a novel species of the genus Actinokineospora, for which the name Actinokineospora mzabensis sp. nov. is proposed, with the type strain PAL84T (=DSM 45961T = CECT 8578T). 相似文献

A novel actinobacterial strain, designated TRM SA0054T, was isolated from the roots of Sophora alopecuroides grown in Alar, Xinjiang, north-west China, and characterised by a polyphasic taxonomic approach. Phylogenetic analysis showed that strain TRM SA0054T has 16S rRNA gene sequence similarity of 98.22% with Streptomyces barkulensis RC 1831T. Whole cell hydrolysates of strain TRM SA0054T were found to contain ll-diaminopimelic acid as the diagnostic diamino acid and ribose, glucose and xylose as the major whole cell sugars. The major fatty acids were identified as iso-C16:0, iso-C16:1 G, anteiso-C17:0, anteiso-C15:0, C16:0, anteiso-C17:1 ω9c. The main menaquinones were determined to be MK-8 (H4), MK-9 (H6) and MK-9 (H8). The polar lipids were identified as diphosphatidylglycerol, phosphatidylmethylethanolamine, phosphatidylethanolamine, phosphatidylcholine, phosphatidylinositol, phosphatidylinositol mannoside and an unidentified lipid. The G?+?C content of the genomic DNA was determined to be 73.04%. Strain TRM SA0054T has a relatively low DNA–DNA relatedness value with Streptomyces barkulensis RC 1831T as determined by calculating the average nucleotide identity value (ANI?=?84.1%). Based on the phenotypic, chemotaxonomic and phylogenetic data, it is concluded that strain TRM SA0054T should be designated as a novel species of the genus Streptomyces, for which the name Streptomyces carminius sp. nov. is proposed, with type strain TRM SA0054T (=?CCTCC AA 2016041T?=?KCTC39903T). 相似文献

A nitrogen-fixing, endospore-forming bacterium, designated strain L201T was isolated from the leaves of Bryophyllum pinnatum growing in South China Agricultural University. Phylogenetic analysis of the 16S rRNA gene sequence indicated that strain L201T is affiliated with the genus Paenibacillus, and closely related to Paenibacillus albidus Q4-3T (97.4%), Paenibacillus odorifer DSM 15391T (97.3%) and Paenibacillus borealis DSM 13188T (97.2%). The main fatty acids components was anteiso-C15:0 (48.1%). The predominant isoprenoid quinone was MK-7. The major polar lipids were found to be diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylglycerol. The G+C content of strain L201T was 43.9%. DNA–DNA relatedness between L201T and the reference strain was 29.8%. Biological and biochemical tests, protein patterns, genomic DNA fingerprinting and comparison of cellular fatty acids distinguished strain L201T from the closely related Paenibacillus species. Based on these data, the novel species Paenibacillus bryophyllum sp. nov. is proposed, with the type strain L201T(=?KCTC 33951 T?=?GDMCC 1.1251 T). 相似文献

Two Gram-stain positive, aerobic actinomycete strains, designated NEAU-JGR1T and NEAU-JGC41, were isolated from soil collected from Fairy Lake Botanical Garden in Shenzhen, Guangdong Province, south of China. The 16S rRNA gene sequences analysis showed that the two strains exhibited 99.5% 16S rRNA gene sequence similarity with each other and were closely related to Promicromonospora thailandica JCM 17130T (99.4, 99.3%) and Promicromonospora citrea DSM 43110T (99.2, 99.2%). Phylogenetic analysis based on the 16S rRNA gene sequences indicated that the two strains clustered together and formed a cluster with P. thailandica JCM 17130T and P. citrea DSM 43110T. Both strains were observed to contain MK-9(H4) and MK-9(H2) as predominant menaquinones. Their whole cell sugar profiles were found to main contained rhamnose, ribose, glucose and galactose. The phospholipid profile contained diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, glycophosphatidylinositol, phosphatidylinositol mannoside, an unidentified glycolipid and an unidentified phospholipid. The predominant cellular fatty acids for the two strains were identified as anteiso-C15:0, iso-C15:0 and anteiso-C17:0. The DNA–DNA hybridization value between strains NEAU-JGR1T and NEAU-JGC41 was 85.1?±?0.3%, and the values between the two strains and their close phylogenetic relatives were well below 70%, supporting the conclusion that they represent a distinct genomic species. An array of phenotypic characteristics also differentiated the isolates from closely related species. On the basis of the genetic and phenotypic properties, strains NEAU-JGR1T and NEAU-JGC41 can be classified as representatives of a novel species of the genus Promicromonospora, for which the name Promicromonospora viridis sp. nov., is proposed. A novel actinomycete, designated strain NEAU-LA29T, was isolated from soil collected from Xianglu Mountain and subjected to a polyphasic taxonomic study. Based on a polyphasic taxonomic approach comprising chemotaxonomic, phylogenetic, morphological and physiological characterisation, the isolate has been affiliated to the genus Streptomyces. 16S rRNA gene sequence analysis showed that the isolate is closely related to Streptomyces vastus JCM4524T (98.8% identity) and Streptomyces cinereus DSM43033T (97.9%). However, multilocus sequence analysis based on five other house-keeping genes (atpD, gyrB, rpoB, recA and trpB) and low DNA–DNA relatedness values enabled the strain to be differentiated from these closely related species of the genus Streptomyces. Thus, strain NEAU-LA29T is concluded to represent a novel species of the genus Streptomyces, for which the name Streptomyces xiangluensis sp. nov. is proposed. A novel actinomycete strain designated S2T was isolated from Tunisian rhizosphere soil of Lavandula officinalis. This isolate exhibited broad spectrum antibacterial activity against several Gram-positive and Gram-negative bacteria and also antifungal activity against yeast and filamentous fungi. The isolate S2T presents morphological and chemotaxonomic characteristics typical of the members of the genus Streptomyces. Whole cell hydrolysates of S2T were found to contain LL-diaminopimelic acid. The major fatty acids were identified as C16:0, anteiso-C15:0 and iso-C16:0 whereas the predominant menaquinones were found to be MK-9(H6) and MK-9(H8). The polar lipids were identified as diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, phosphatidylinositol, phosphatidylinositol mannoside and three unidentified compounds. The G+C content of the genomic DNA was determined to be 71.8 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain S2T belongs to the genus Streptomyces and is closely related to Streptomyces netropsis DSM 40259T with 99.86% sequence similarity. Multi-locus sequence analysis (MLSA) based on four house-keeping gene alleles (gyrB, recA, trpB, rpoB) showed that isolate S2T is closely related to S. netropsis, with an MLSA distance greater than 0.007. The DNA–DNA relatedness between strain S2T and its near phylogenetic neighbour was 63.6 ± 2.3%, which is lower than the 70% threshold value for delineation of genomic prokaryotic species. This isolate was also distinguished from the type strain S. netropsis DSM 40259T, using a combination of morphological and physiological features. Based on its phenotypic and molecular properties, strain S2T is considered to represent a novel species of the genus Streptomyces, for which the name Streptomyces tunisialbus sp. nov. is proposed. A novel strain, DCY108T was isolated from soil of a Panax ginseng field, Yeoncheon province (38°04′N 126°57′E), Republic of Korea. Strain DCY108T is Gram-negative, non-motile, non-flagellate, rod-shaped, and aerobic. The bacterium grows optimally at 25–30 °C, pH 6.5–7.0 and 1 % NaCl. Phylogenetically, strain DCY108T is closely related to Pedobacter jejuensis JCM 18824T, Pedobacter aquatilis JCM 13454T, Pedobacter kyungheensis LMG 26577T and the type strain of the genus Pedobacter heparinus DSM 2366T. The DNA–DNA relatedness values between strain DCY108T and its close phylogenetic neighbors were below 30.0 %. The DNA G+C content of strain DCY108T was determined to be 45.1 mol%. The predominant quinone was menaquinone 7 (MK-7). The major polar lipids were identified as phosphatidylethanolamine and three unidentified aminolipids AL1, AL13 and AL17. Iso-C15:00, iso-C17:03OH and summed feature 3 (C16:1 ω7c/C16:1 ω6c) were identified as the major fatty acids present in strain DCY108T. The results of physiological and biochemical tests allowed strain DCY108T to be differentiated phenotypically from other recognized species belonging to the genus Pedobacter. Therefore, it is suggested that the newly isolated organism represents a novel species, for which the name Pedobacter panacis sp. nov is proposed with the type strain designated as DCY108T (=CCTCCAB 2015196T = KCTC 42748T). 相似文献

A novel Gram-negative and red-pinkish bacterium designated DG5BT was isolated from a dry soil. Cells were rods that were catalase- and oxidase-positive, and non-motile. The strain was found to grow at temperatures from 10 to 30°C (optimum 25°C) and pH 6.0–8.0, (optimum pH 7) on R2A broth. 16S rRNA gene sequence (1,452 bp) analysis of this strain identified it as a member of the genus Hymenobacter that belongs to the class Cytophagia. The highest gene sequence similarities were with Hymenobacter arizonensis OR362-8T (98.3%), Hymenobacter humi DG31AT (97.6%), and Hymenobacter glaciei VUG-A130T (96.6%). Strain DG5BT exhibited <70% DNA-DNA relatedness with H. arizonensis (34.7 ± 7.0%; reciprocally, 29.7 ± 1.2%) and H. humi (39.4 ± 4.3%; reciprocally, 39.5 ± 3.3%) as a different genomic species, and its genomic DNA G+C content was 59.8%. Strain DG5BT had the following chemotaxonomic characteristics: the major fatty acids are iso-C15:0, anteiso-C15:0, C16:1ω5c, and summed feature 3 (C16:1ω7c / C16:1ω6c); polar lipid profile contained phosphatidylethanolamine (PE), unknown aminophospholipid (APL), unknown glycolipids (GL), unknown phospholipids (PL), and unknown polar lipids (L); the major quinone is MK-7. The absorbance peak of pigment is at 481.0 nm. Strain DG5BT showed low-level resistance to gamma-ray irradiation. Phenotypic, chemotaxonomic, and genotypic properties indicated that isolate DG5BT represents a novel species within the genus Hymenobacter for which the name Hymenobacter sedentarius sp. nov. is proposed. A basidiomyetous yeast strain RO-203, which formed orange-red colored colonies, was isolated from a sample of paddy rice crops at the ripe stage in Japan. Morphological, physiological and biochemical characterization indicated that this strain belonged to the genus Rhodotorula. Molecular taxonomic analysis based on the 26S rDNA D1/D2 domain and internal transcribed spacer (ITS) region sequences showed that RO-203 represents an undescribed yeast species, for which the name Rhodotorula oryzae sp. nov. is proposed (type strain: AS 2.2363T = MAFF 516128T). The new species clustered in a branch together with Sakaguchia dacryoidea in phylogenetic trees based on the D1/D2 and ITS sequences. A Gram-negative, motile, aerobic and rod-shaped bacterial strain designated 119BY6-57T was isolated from spongin. The taxonomic position of the novel isolate was confirmed using the polyphasic approach. Strain 119BY6-57T grew well at 25–30°C on marine agar. On the basis of 16S rRNA gene sequence similarity, strain 119BY6-57T belongs to the family Xanthomonadaceae and is related to Lysobacter aestuarii S2-CT (99.8% sequence similarity), L. maris KMU-14T (97.5%), and L. daejeonensis GH1-9T (97.3%). Lower sequence similarities (97.0%) were found with all of the other recognized members of the genus Lysobacter. The G + C content of the genomic DNA was 69.9 mol%. The major respiratory quinone was Q-8 and the major fatty acids were C16:0 iso, C15:0 iso, summed feature 9 (comprising C17:1 iso ω9c and/or C16:0 10-methyl), summed feature 3 (comprising C16:1ω7c and/or C16:1ω6c), and C11:0 iso 3-OH. The polar lipids were phosphatidylglycerol, phosphatidylethanolamine, diphosphatidylglycerol, three unidentified phospholipids, and an unidentified polar lipid. DNADNA relatedness values between strain 119BY6-57T and its closest phylogenetically neighbors were below 48.0 ± 2.1%. Based on genotypic and phenotypic characteristics, it is concluded that strain 119BY6-57T is a new member within the genus Lysobacter, for which the name Lysobacter spongiae sp. nov. is proposed. A non-motile, pink-pigmented bacterial strain designated IMCC25679T, was isolated from freshwater Lake Chungju of Korea. Phylogenetic trees based on 16S rRNA gene sequences showed that the strain IMCC25679T formed a lineage within the genus Pedobacter. The strain IMCC25679T was closely related to Pedobacter daechungensis Dae 13T (96.4% sequence similarity), Pedobacter rivuli HME8457T (95.3%) and Pedobacter lentus DS-40T (94.3%). The major fatty acids of IMCC- 25679T were iso-C15:0, iso-C16:0 and summed feature 3 (comprising C16:1ω6c and/or C16:1ω7c). The major respiratory quinone was MK-7. The major polar lipids were phosphatidylethanolamine (PE), an unidentified sphingolipid (SL), an unidentified aminolipid (AL) and three unidentified polar lipids (PL). The DNA G + C content of IMCC25679T was 32.2 mol%. Based on the evidence presented in this study, the strain IMCC25679T represents a novel species within the genus Pedobacter, with the proposed name Pedobacter aquicola, sp. nov. A Gram-staining-negative, non-motile, curved rod-shaped, aerobic bacterium, designated S1-2-4T, was isolated from soil in Jeollabuk-do province, Republic of Korea, and was characterized taxonomically using a polyphasic approach. Comparative 16S rRNA gene sequence analysis showed that strain S1-2-4T was a member of the family Cytophagaceae and most closely related to ‘Spirosoma radiotolerans’ DG5A (97.2%), Spirosoma fluviale MSd3T (96.4%), and Spirosoma linguale DSM 74T (96.3%). The genomic DNA G + C content of strain S1-2-4T was 49.7 mol%. The major fatty acids were summed feature 3 (C16:1ω7c/C16:1ω6c), C16:1ω5c, and C16:0, and the major polar lipid was phosphatidylethanolamine. MK-7 was the predominant respiratory quinone. Phenotypic and chemotaxonomic data supported the affiliation of strain S1-2-4T with the genus Spirosoma. DNA-DNA hybridization between strain S1-2-4T and ‘Spirosoma radiotolerans’ showed relatively low DNA-DNA relatedness (31%). Strain S1-2-4T could be distinguished from its closest phylogenetic neighbors based on its phenotypic, genotypic, and chemotaxonomic features. A Gram-stain-negative, non-motile, non-spore-forming, rod-shaped, aerobic bacterium, designated 15J9-8T, was isolated from soil on Jeju Island, Republic of Korea. The isolate was able to grow between 10 and 30°C, pH 6.5–8.5, and in presence of 0–1% (w/v) NaCl. The results of comparative 16S rRNA gene sequence analysis indicated that strain 15J9-8T represented a member of the family Cytophagaceae, phylum Bacteroidetes, and was most closely related to Spirosoma aerophilum 5516J-17T (96.1% similarity), Spirosoma pulveris JSH5-14T (95.6%), and Spirosoma linguale DSM 74T (95.2%). The G + C content of the genomic DNA of the isolate was 47.0 mol%. Strain 15J9-8T contained summed feature 3 (C16:1 ω7c/C16:1 ω6c), C16:1 ω5c, and iso-C15:0 as the major fatty acids, phosphatidylethanolamine and an unidentified aminophospholipid as the main polar lipids, and menaquinone MK-7 as the predominant respiratory quinone. On the basis of its phenotypic and genotypic properties, and phylogenetic distinctiveness, strain 15J9-8T should be classified as a representative of a novel species of the genus Spirosoma, for which the name Spirosoma migulaei sp. nov. is proposed. Four ascosporulating strains of an undescribed methanol-assimilating yeast species were isolated from forest habitats in Hungary.
Three were recovered from rotten wood and one from leaves of a sessile oak (Quercus petraea). An additional isolate of the undescribed species sharing similar phenotypic characters with the above-noted strains was
recovered from the gut of an unidentified beetle collected from under the bark of a coniferous tree in Bulgaria. A closely
related, but somewhat divergent strain was recovered from insect frass in a Ponderosa pine (Pinus ponderosa) collected in New Mexico, USA. Analysis of the D1/D2 sequences of the LSU rRNA gene placed the new species in the Ogataea clade. The ITS and the D1/D2 LSU sequences of the rRNA gene repeats were compared for the above-noted strains and that of
the type strain of Ogataea zsoltii, the closest neighbour among currently recognized Ogataea species. Their relatedness was investigated by parsimony network analysis as well. As a result of the sequence analysis,
it was concluded that the six strains isolated from tree associated habitats represent a single new yeast species. Ogataea saltuana sp. nov. is proposed to accommodate these strains. The type strain NCAIM Y.01833T (CBS 10795T, NRRL Y-48448T) was recovered from rotten wood of Scotch pine (Pinus silvestris) in Hungary. The GenBank accession number for the D1/D2 domain nuclear large subunit rRNA gene sequence of strain NCAIM Y.01833T (CBS 10795T, NRRL Y-48448T) is EU327033.
