A major problem for communication research is to provide standardized stimuli and to disentangle the relative contribution of different sensory channels to the compound signal and its effectiveness. Video techniques have frequently been used for this purpose. Fifty Hertz cathode ray tube (CRT) screens cannot be used for birds because their flicker frequency is lower than the time resolution of at least songbirds. Thin film transistor (TFT) screens have successfully been used to present video clips of behaving conspecifics. We show here that they are indeed transmitting video images in a way that enables zebra finches to react appropriately to live video images of conspecifics and that the test birds are able to detect small differences in the behaviour of the stimulus animals. Adding acoustic information strongly enhanced the reaction to the video clips.  相似文献

This paper compared two general methods for assessing critical fusion frequency in hens (gallus gallus domesticus). The first method was a conditional discrimination procedure with the stimuli presented successively. The second was a two-alternative forced-choice procedure with the stimuli presented simultaneously. It was found that both methods of stimulus presentation gave comparable results suggesting that either method is useful when investigating psychophysics in animals. The results also show that hens’ critical fusion frequency is considerably higher than that of humans which may account for hens’ inability to recognise images presented on standard computer or television monitors.  相似文献

While some lower vertebrates, such as zebrafish, do not appear to possess anatomically separate pathways of processing visual information (such as M-pathways and P-pathways), it is believed that separate processing of the visual stimulus (such as luminance and chromatic processing) is a basic requirement of vertebrate vision. In this study, spectral sensitivity functions were obtained from electroretinogram responses to heterochromatic flicker photometry stimuli at several flicker rates, including a low flicker rate (2 Hz), in an attempt to predominantly stimulate chromatic processes and a high flicker rate (16 Hz), in an attempt to predominantly stimulate luminance processes. In addition, chromatic adaptation was used to isolate and examine the temporal properties of the different cone-type contributions to the electroretinogram response. Spectral sensitivity functions based on responses to heterochromatic stimuli of a low flicker rate appeared to receive both opponent and nonopponent contributions; however, when the stimulus flicker rate was high, spectral sensitivity appeared to be a function of only nonopponent mechanisms. Also, the differences in cone contributions to the spectral sensitivity functions across the different flicker rates appear to be related to the temporal properties of the cone contributions to the electroretinogram response.  相似文献

We trained jungle crows to discriminate among photographs of human face according to their sex in a simultaneous two-alternative task to study their categorical learning ability. Once the crows reached a discrimination criterion (greater than or equal to 80% correct choices in two consecutive sessions; binomial probability test, p < .05), they next received generalization and transfer tests (i.e., greyscale, contour, and ‘full’ occlusion) in Experiment 1 followed by a ‘partial’ occlusion test in Experiment 2 and random stimuli pair test in Experiment 3. Jungle crows learned the discrimination task in a few trials and successfully generalized to novel stimuli sets. However, all crows failed the greyscale test and half of them the contour test. Neither occlusion of internal features of the face, nor randomly pairing of exemplars affected discrimination performance of most, if not all crows. We suggest that jungle crows categorize human face photographs based on perceptual similarities as other non-human animals do, and colour appears to be the most salient feature controlling discriminative behaviour. However, the variability in the use of facial contours among individuals suggests an exploitation of multiple features and individual differences in visual information processing among jungle crows.  相似文献

Background

Applications of cell electropermeabilization are rapidly growing but basic concepts are still unclear. In particular, the impact of electric pulse repetition rate in the efficiency of permeabilization has not yet been understood.

Methods

The impact of electric pulse repetition rate in the efficiency of permeabilization was analyzed in experiments performed on potato tissue and partially transposed on mice liver. On potato tissue, pulses with durations of 100 μs or 10 ns are applied. The intensity of permeabilization was quantified by means of bioimpedance changes and electric current measurements and a new index was defined.

Results

For the two pulse durations tested, very low repetition rates (below 0.1 Hz) are much more efficient to achieve cell permeabilization in potato tissue. In mice liver, using 100 μs pulses, the influence of the repetition rate is more complex. Indeed, repetition rates of 1 Hz and 10 Hz are more efficient than 100 Hz or 1 kHz, but not the repetition rate of 0.1 Hz for which there is an impact of the living mice organism response.

Conclusions

We propose that the effects reported here might be caused by an electroporation-induced cell membrane ‘electro-desensitization’ which requires seconds to dissipate due to membrane resealing.

General significance

This study not only reinforces previous observations, but moreover it sustains a new concept of ‘electro-desensitization’ which is the first unifying mechanism enabling to explain all the results obtained until now both in vitro and in vivo, with long and short pulses.  相似文献

How do humans perceive the passage of time and the duration of events without a dedicated sensory system for timing? Previous studies have demonstrated that when a stimulus changes over time, its duration is subjectively dilated, indicating that duration judgments are based on the number of changes within an interval. In this study, we tested predictions derived from three different accounts describing the relation between a changing stimulus and its subjective duration as either based on (1) the objective rate of changes of the stimulus, (2) the perceived saliency of the changes, or (3) the neural energy expended in processing the stimulus. We used visual stimuli flickering at different frequencies (4–166 Hz) to study how the number of changes affects subjective duration. To this end, we assessed the subjective duration of these stimuli and measured participants'' behavioral flicker fusion threshold (the highest frequency perceived as flicker), as well as their threshold for a frequency-specific neural response to the flicker using EEG. We found that only consciously perceived flicker dilated perceived duration, such that a 2 s long stimulus flickering at 4 Hz was perceived as lasting as long as a 2.7 s steady stimulus. This effect was most pronounced at the slowest flicker frequencies, at which participants reported the most consistent flicker perception. Flicker frequencies higher than the flicker fusion threshold did not affect perceived duration at all, even if they evoked a significant frequency-specific neural response. In sum, our findings indicate that time perception in the peri-second range is driven by the subjective saliency of the stimulus'' temporal features rather than the objective rate of stimulus changes or the neural response to the changes.  相似文献

Aotus monkeys were tested in a forced-choice discrimination task to determine their ability to discriminate sinusoidally flickering lights varying in temporal frequency and luminance contrast. Under conditions of moderate light adaptation this primate is maximally sensitive to lights flickering at 10 Hz while the highest frequency they can discriminate is about 42 Hz. At very low light levels (10(-5) ft L) maximum sensitivity is for 2.2--5 Hz flicker. The highest flicker rate that could be discriminated under these conditons was about 29 Hz. In comparison to humans tested in the same situation. Aotus monkeys show relatively lower sensitivity to temporal flicker under conditions of light adaptation but relatively higher sensitivity at very low light levels.  相似文献

An irregular ventricular response during atrial fibrillation (AF) has been shown to mediate an increase in sympathetic nerve activity in human subjects. The molecular mechanisms remain unclear. This study aimed to investigate the impact of rate and irregularity on nerve growth factor (NGF) expression in cardiomyocytes, since NGF is known to be the main contributor to cardiac sympathetic innervation density. Cell cultures of neonatal rat ventricular myocytes were electrically stimulated for 48 h with increasing rates (0, 5 and 50 Hz) and irregularity (standard deviation (SD) = 5%, 25% and 50% of mean cycle length). Furthermore, we analyzed the calcineurin-NFAT and the endothelin-1 signalling pathways as possible contributors to NGF regulation during arrhythmic stimulation. We found that the increase of NGF expression reached its maximum at the irregularity of 25% SD by 5 Hz (NGF: 5 Hz 0% SD = 1 vs. 5 Hz 25% SD = 1.57, P < 0.05). Specific blockade of the ET-A receptor by BQ123 could abolish this NGF increase (NGF: 5 Hz 25% SD + BQ123 = 0.66, P < 0.05). High frequency electrical field stimulation (HFES) with 50 Hz decreased the NGF expression in a significant manner (NGF: 50 Hz = 0.55, P < 0.05). Inhibition of calcineurin-NFAT signalling with cyclosporine-A or 11R-VIVIT abolished the HFES induced NGF down-regulation (NGF: 50 Hz + CsA = 1.14, P < 0.05). In summary, this study reveals different signalling routes of NGF expression in cardiomyocytes exposed to increasing rates and irregularity. Whether this translates into different degrees of NGF expression and possibly neural sympathetic growth in various forms of ventricular rate control during AF remains to be elucidated in further studies.  相似文献

Estradiol withdrawal after pregnancy is hypothesized to precipitate depressive symptoms in vulnerable women. A hormone-simulated pregnancy was induced in female rats and the effects of a ‘postpartum’ drop in estradiol on hippocampal cell proliferation were examined. All groups were ovariectomized or given sham surgery prior to treatment. Rats were randomly assigned to ‘postpartum’, ‘postpartum’ + EB (estradiol benzoate), ‘postpartum’ + DPN (diarylpropionitrile; an ERβ agonist), ‘postpartum’ + IMI (imipramine; a tricyclic antidepressant), sham, ovariectomized (OVX), sham + IMI or OVX + IMI groups. All ‘postpartum’ groups received hormone injections (estradiol and progesterone) over 23 days to simulate pregnancy, while IMI groups also received daily imipramine injections. After day 23, ‘postpartum’ rats were withdrawn from the hormone-simulated pregnancy (mimicking the postpartum drop in gonadal hormones), while other ‘postpartum’ treatment groups received daily injections of DPN, EB or IMI. On day 3 ‘postpartum’ all rats were injected with bromodeoxyuridine (BrdU; a DNA synthesis marker) and perfused 24 h later to assess cell proliferation and cell death in the dentate gyrus. ‘Postpartum’ hormone withdrawal decreased hippocampal cell proliferation in the ‘postpartum’ and ‘postpartum’ + EB groups only. Chronic imipramine significantly increased hippocampal cell proliferation in sham + IMI, but not OVX + IMI rats suggesting that imipramine's effects to increase hippocampal cell proliferation in female rats is related to reproductive status. Cell death (pyknotic cells) was decreased only in the ‘postpartum’ group. Together, these results suggest an important, though complex, role for gonadal hormones in the cellular changes accompanying this model of postpartum depression.  相似文献

The winner of an international contest to find the world’s fastest drummer (WFD) can perform repetitive wrist tapping movements with one hand using a handheld drumstick at 10 Hz, much faster than the maximum tapping frequency of 5-7 Hz in the general population. The muscle activity facilitating this improved performance, however, has only recently been explored. The present study investigated the rise rate and timing variability of surface electromyographic (EMG) activity of wrist flexor/extensor muscles in the WFD, and compared them with those in two control groups: non-drummers (NDs) and ordinary drummers (ODs). The WFD showed more rapid EMG amplitude rise, earlier decline of EMG activity, and more stable muscle activation time than the NDs and ODs. In addition, there was a significant correlation between the EMG rise rate and the duration of drum training in the group of drummers (i.e., ODs and WFD). These results indicate that the 10-Hz performance of the WFD was achieved by a ‘sharper’ and ‘less noisy’ burst pattern of wrist muscles, and that drum training would have the effect to increase the speed of development of muscle tension.  相似文献

Background

Calreticulin (CRT), a Ca2+-binding chaperone of the endoplasmic reticulum, can also be found in several other locations including the cytosol, nucleus, secretory granules, the outer side of the plasma membrane, and the extracellular matrix. Whether CRT is localized at mitochondria of cardiomyocytes and whether such localization is affected under DCM are still unclear.

Methods and results

The DCM model was generated in rats by the daily oral administration of furazolidone for thirty weeks. Echocardiographic and hemodynamic studies demonstrated enlarged left ventricular dimensions and reduced systolic and diastolic function in DCM rats. Immuno-electron microscopy and Western blot showed that CRT was present in cardiomyocyte mitochondria and the mitochondrial content of CRT was increased in DCM hearts (< 0.05). Morphometric analysis showed notable myocardial apoptosis and mitochondrial swelling with fractured or dissolved cristae in the DCM hearts. Compared with the control group, the mitochondrial membrane potential level of the freshly isolated cardiac mitochondria and the enzyme activities of cytochrome c oxidase and succinate dehydrogenase in the model group were significantly decreased (< 0.05), and the myocardial apoptosis index and the caspase activities of caspase-9 and caspase-3 were significantly increased (< 0.05). Pearson linear correlation analysis showed that the mitochondrial content of CRT had negative correlations with the mitochondrial function, and a positive correlation with myocardial apoptosis index (< 0.001). The protein expression level of cytochrome c and the phosphorylation activity of STAT3 in the mitochondrial fraction were significantly decreased in the model group compared with the control group (< 0.05).

Conclusions

These data demonstrate that CRT is localized at cardiomyocyte mitochondria and its mitochondrial content is increased in DCM hearts.  相似文献

In the tropical butterfly Bicyclus anynana (Nymphalidae) essential components of fitness (such as fecundity and longevity) depend to a large degree on exogenous adult-derived nutrients, particularly carbohydrates. We investigated which of the nutrients/compounds found in the adult diet act as feeding stimuli, and whether butterflies show preferences for particular nutrients or combinations. Only sugars and alcohols acted as feeding stimuli, the highest responses being found for sucrose, glucose, ethanol, butanol and propanol. Various other compounds (e.g. amino acids, acetic acid, vitamins, lipids, salts, and yeast) did not elicit any probing or feeding responses. Behavioural tests revealed a clear preference hierarchy for sugars (sucrose > glucose > fructose > maltose), but not for alcohols. Butterflies did not discriminate between sucrose solutions enriched with different nutrients and plain sucrose solutions, although they showed a preference for acetic acid and an aversion to salts and ascorbic acid when offered in combination with sucrose. Throughout, both sexes showed very similar patterns. We conclude that locating carbohydrate sources seems sufficient to cover all the butterflies’ nutritional needs, while alcohols function primarily as long range signals, guiding the butterflies to food sources. Thus, fruit-feeding butterflies, in contrast to nectar-feeding butterflies, appear not to have distinctive preferences for e.g. amino acids or salts, but do share a common primary preference for sucrose.  相似文献

The growth characteristics of microalgae under different light conditions (continuous or intermittent) are essential information for photobioreactor design and operation. In this study, we constructed a thin-layer (10 mm) flat plate photobioreactor device with a light/dark (L/D) alternation system to investigate the growth of Spirulina platensis under two different light regimes: (1) continuous illumination in a wide range of light intensities (1.00-77.16 mW cm−2); (2) intermittent illumination in medium frequency (0.01-20 Hz). Specific growth rate and light efficiency based on biomass production were determined for each round of experiment. Four regions (light limited region, intermediate region, light saturated region and light inhibition region) were recognized according to the results under continuous illumination. Under intermittent illumination, when L/D frequency increased from 0.01 Hz to 20 Hz, specific growth rate and light efficiency were enhanced. However, the enhancement was different, depending on the applied light intensity and light fraction. The higher the light intensity, the greater the enhancement would be when L/D frequency increased from 0.01 Hz to 20 Hz; and the higher the light intensity, the lower the light fractions is needed to maintain light efficiency as high as that under continuous illumination in light limited region.  相似文献

Efficient dynamic interactions among cofactor, enzymes and substrate molecules are of primary importance for multi-step enzymatic reactions with in situ cofactor regeneration. Here we showed for the first time that the above dynamic interactions could be significantly intensified by exerting an external alternating magnetic field on magnetic nanoparticles-supported multi-enzymatic system so that the inter-particle collisions due to Brownian motion of nanoparticles could be improved. To that end, a multienzyme system including glutamate dehydrogenase (GluDH), glucose dehydrogenase (GDH) and cofactor NAD(H) were separately immobilized on silica coated Fe3O4 magnetic nanoparticles with an average diameter of 105 nm, and the effect of magnetic field strength and frequency on the kinetics of the coupled bi-enzyme reaction was investigated. It was found that at low magnetic field frequency (25 Hz and 100 Hz), increasing magnetic field strength from 9.8 to 161.1 Gs led to only very slight increase in reaction rate of the coupled bi-enzyme reaction expressed by glucose consumption rate. At higher magnetic field of 200 Hz and 500 Hz, reaction rate increased significantly with increase of magnetic field strength. When the magnetic field frequency was kept at 500 Hz, the reaction rate increased from 3.89 μM/min to 8.11 μM/min by increasing magnetic field strength from 1.3 to 14.2 Gs. The immobilized bi-enzyme system also showed good reusability and stability in the magnetic field (500 Hz, 14.2 Gs), that about 46% of original activity could be retained after 33 repeated uses, accounting for totally 34 days continuous operation. These results demonstrated the feasibility in intensifying molecular interactions among magnetic nanoparticle-supported multienzymes by using nano-magnetic stirrer for efficient multi-step transformations.  相似文献

We previously reported that cyclic compressive force (CCF) induced interleukin-6 mRNA expression in osteocyte-like MLO-Y4 cells. But little is known about how the stimuli are converted into the biochemical signals in MLO-Y4 cells. The aim of this research was to study the effect of CCF on the IL-6 secretion and the role of extracellular signal-regulated kinases 1/2 (ERK1/2) in this process. The cells were exposed to CCF with different magnitudes (1000, 2000 and 4000 μstrain), frequencies (0.5, 1.0 and 2.0 Hz) and durations (10 min, 30 min, 1 h, 3 h and 6 h) by a four-point bending system. The IL-6 secretion and ERK1/2 phosphorylation of the cells were determined by ELISA and Western blotting, respectively. The results showed that IL-6 protein secretion was significantly up-regulated in response to CCF in a magnitude-, frequency- and duration-dependent fashion. The phosphorylation of ERK1/2 also increased in all cases but not depended on the magnitude, frequency or duration of CCF. Furthermore, the inhibition of the ERK1/2 pathway by its specific inhibitor PD098059 decreased but not completely abrogated the IL-6 secretion from stressed MLO-Y4 cells. These findings demonstrate that CCF-induced IL-6 secretion occurs via a mechanism that involves ERK1/2 signaling pathway and suggest that modulation of this event contributes to the pathogenesis of osteoporosis and stress-induced pathological bone resorption as well.  相似文献

Recent evidence suggests that non-muscle myosin IIB (NMIIB) contributes to smooth muscle contraction. This study was designed to determine the effects of NMIIB on the cross-bridge cycling rate. The cross-bridge cycling rate was investigated using sinusoidal analysis. Frequency analysis revealed two asymptotes in the Bode plot of the data; and the intersection of the asymptotes (corner frequency) was higher for the B+/− strain (8.73 ± 1.10 Hz vs 16.56 ± 1.26 Hz, P < 0.05), consistent with a higher overall cross-bridge cycling rate in heterozygous NMIIB KO (B+/−) vs WT mice. These results demonstrate that because of their long duty cycle, NMIIB cross-bridges act as an internal load on smooth muscle myosin to decrease the overall cross-bridge cycling rate and muscle Vmax during force maintenance.  相似文献

We present FLEX (Functional evaluation of experimental perturbations), a pipeline that leverages several functional annotation resources to establish reference standards for benchmarking human genome‐wide CRISPR screen data and methods for analyzing them. FLEX provides a quantitative measurement of the functional information captured by a given gene‐pair dataset and a means to explore the diversity of functions captured by the input dataset. We apply FLEX to analyze data from the diverse cell line screens generated by the DepMap project. We identify a predominant mitochondria‐associated signal within co‐essentiality networks derived from these data and explore the basis of this signal. Our analysis and time‐resolved CRISPR screens in a single cell line suggest that the variable phenotypes associated with mitochondria genes across cells may reflect screen dynamics and protein stability effects rather than genetic dependencies. We characterize this functional bias and demonstrate its relevance for interpreting differential hits in any CRISPR screening context. More generally, we demonstrate the utility of the FLEX pipeline for performing robust comparative evaluations of CRISPR screens or methods for processing them.  相似文献
